Multiple sclerosis (MS) is a T cell-driven autoimmune disease that attacks the myelin of the central nervous system (CNS) and currently has no cure. MS etiology is linked to both the gut flora and external environmental factors but this connection is not well understood. One immune system regulator responsive to nonpathogenic external stimuli is the aryl hydrocarbon receptor (AHR). The AHR, which binds diverse molecules present in the environment in barrier tissues, is a therapeutic target for MS. However, AHR's precise function in T lymphocytes, the orchestrators of MS, has not been described. Here, we show that in a mouse model of MS, T cell-specific Ahr knockout leads to recovery driven by a decrease in T cell fitness. At the mechanistic level, we demonstrate that the absence of AHR changes the gut microenvironment composition to generate metabolites that impact T cell viability, such as bile salts and short chain fatty acids. Our study demonstrates a newly emerging role for AHR in mediating the interdependence between T lymphocytes and the microbiota, while simultaneously identifying new potential molecular targets for the treatment of MS and other autoimmune diseases.

Catharanthus roseus produces a diverse range of specialized metabolites of the monoterpenoid indole alkaloid (MIA) class in a heavily branched pathway. Recent great progress in identification of MIA biosynthesis genes revealed that the different pathway branch genes are expressed in a highly cell type- and organ-specific and stress-dependent manner. This implies a complex control by specific transcription factors (TFs), only partly revealed today. We generated and mined a comprehensive compendium of publicly available C. roseus transcriptome data for MIA pathway branch-specific TFs. Functional analysis was performed through extensive comparative gene expression analysis and profiling of over 40 MIA metabolites in the C. roseus flower petal expression system. We identified additional members of the known BIS and ORCA regulators. Further detailed study of the ORCA TFs suggests subfunctionalization of ORCA paralogs in terms of target gene-specific regulation and synergistic activity with the central jasmonate response regulator MYC2. Moreover, we identified specific amino acid residues within the ORCA DNA-binding domains that contribute to the differential regulation of some MIA pathway branches. Our results advance our understanding of TF paralog specificity for which, despite the common occurrence of closely related paralogs in many species, comparative studies are scarce.

- Abstract
Monitoring of minimal residual disease (MRD) by flow cytometry (FCM) is a powerful prognostic tool for predicting outcomes in acute lymphoblastic leukemia (ALL). To apply FCM-MRD in large, collaborative trials, dedicated laboratory staff must be educated to concordantly high levels of expertise and their performance quality should be continuously monitored. We sought to install a unique and comprehensive training and quality control (QC) program involving a large number of reference laboratories within the international Berlin-Frankfurt-Münster (I-BFM) consortium, in order to complement the standardization of the methodology with an educational component and persistent quality control measures. Our QC and quality assurance (QA) program is based on four major cornerstones: (i) a twinning maturation program, (ii) obligatory participation in external QA programs (spiked sample send around, United Kingdom National External Quality Assessment Service (UK NEQAS)), (iii) regular participation in list-mode-data (LMD) file ring trials (FCM data file send arounds), and (iv) surveys of independent data derived from trial results. We demonstrate that the training of laboratories using experienced twinning partners, along with continuous educational feedback significantly improves the performance of laboratories in detecting and quantifying MRD in pediatric ALL patients. Overall, our extensive education and quality control program improved inter-laboratory concordance rates of FCM-MRD assessments and ultimately led to a very high conformity of risk estimates in independent patient cohorts.

COVID-19 hastalığı kısa süre içerisinde tüm dünyaya yayılmıştır ve pandemi ilanedilmiştir. Hastalığın belirsizliği ve salgın niteliği taşıması olumsuz duygulara sebep olabilmektedir. Bu çalışmanın amacı COVID-19 salgın sürecinde bireylerin duygusal yeme davranışlarını çeşitli değişkenlere göre incelemektir. Duygusal yeme davranışı olumsuz duygulara karşılık geliştirilen, işlevsel olmayan bir baş etme biçimi olarak değerlendirildiğinde, bireylerin salgın sürecinde sürdürdükleri veya geliştirdikleri duygusal yeme davranışı bazı değişkenler açısından incelenmiştir. İlişkisel tarama modelindeki araştırmada veriler demografik bilgi formu, Türkçe Duygusal Yeme Ölçeği ve Beck Depresyon Ölçeği ile toplanmıştır. Toplanan veriler betimsel istatistiklerle birlikte Pearson Momentler Çarpımı Korelasyon, Spearman-Brown Sıra Farkları Korelasyon Katsayısı, Tek Yönlü Varyans Analizi (ANOVA) ile analiz edilmiştir. Yapılan analizler sonucunda katılımcıların duygusal yeme davranışlarının cinsiyet, medeni durum, iş sürdürme durumları, beden kitle indeksine göre farklılaştığı saptanmıştır. Aynı zamanda bireylerdeki duygusal yeme davranışının yaş ve depresyon düzeyi ile anlamlı bir ilişki içerisinde olduğu saptanmıştır. Ulaşılan bulgular literatürdeki diğer araştırmalarla birlikte tartışılmış ve ileriki araştırmalar için öneriler sunulmuştur.

- Abstract
Objectives: to estimate the prevalence of disability among Syrian refugees living in Sultanbeyli district, Istanbul and compare people with and without disabilities in terms of demographic and socio-economic characteristics. Methods: using the municipality refugee database as the sampling frame, 80 clusters of 50 people (aged 2+ years) were selected using probability proportionate to size sampling of clusters and random selection of households within clusters. Disability assessment included: i) self-reported difficulties in functioning (using the Washington Group Short Set-Enhanced tool and Child Functioning Modules), ii) Rapid Assessment of Musculoskeletal Impairment and iii) screening for symptoms of common mental disorders for children aged 8–17. Results: the overall prevalence of disability was 24.7% (95% CI 22.1–27.4), when including people self-reporting a lot of difficulty/cannot do in at least functional domain (15%, 95% CI 13.1–17.2), moderate/severe MSI (8.7%, 95% CI 7.6–9.9), and/or symptomatic anxiety, depression and PTSD among children 8–17 (21.0%, 95% CI 18.2–23.9). Men with disabilities were significantly less likely to be in paid work compared to their peers without disabilities (aOR 0.3 95% CI 0.2–0.5). Overall 60% of households included at least one person with a disability. Households with at least one person with a disability had a significantly higher dependency ratio, lower proportion of working-age adults in paid work, and were more likely to be female headed and in receipt of social protection schemes (p<0.05). Conclusion: disability is common among Syrian refugees in Sultanbeyli. People with disabilities in this setting experience greater vulnerability to poverty and exclusion from work, highlighting an urgent need for inclusive services, programmes and policies that are developed and implemented in partnership with people with disabilities., United States Department of State Relief International; LSHTM Grant; UK Aid AT2030 Programme; Global Disability Innovation Hub

- Abstract
Aims: research demonstrates elevated levels of common mental disorders among Syrian refugees, but the majority of studies have, to date, focused on adult populations. This study aims to estimate the prevalence of depression, anxiety and post-traumatic stress disorder (PTSD) among Syrian children and adolescents living in Sultanbeyli district of Istanbul, Turkey. Methods: a population-based survey among Syrian children and adolescents aged 8-17 years living in Sultanbeyli district was conducted in 2019, as part of an all-age survey of disability. 80 clusters of 50 participants (all-ages) were selected from the local municipality's refugee registration database using probability proportionate to size sampling. Children aged 8-17 years were assessed for symptoms of common mental disorders using the Child Revised Impact of Event Scale (CRIES-8) and abbreviated versions of the Center for Epidemiologic Studies Depression Scale for Children (CES-DC) and the Screen for Child Anxiety Related Disorders (SCARED). Results: of the 852 participants, 23.7% (95% CI 19.9-27.2) screened positive for symptomatic depression, PTSD and anxiety. The prevalence estimates for depression, PTSD and anxiety were 12.5% (95% CI 9.8-15.6), 11.5% (95% CI 9.1-14.4) and 9.2% (95% CI 6.8-12.1), respectively. Depression and PTSD were significantly more common in older adolescents, whilst anxiety and PTSD were significantly more common in girls. Depression was more common in children from poorer households and those who had received no education. Children coming from larger households were less likely to show symptoms of PTSD. Conclusions: Syrian refugee children and adolescents are vulnerable to common mental disorders, and culturally appropriate prevention and intervention support are needed for this population., United States Department of State to Relief International

These guidelines are a consensus work of a considerable number of members of the immunology and flow cytometry community. They provide the theory and key practical aspects of flow cytometry enabling immunologists to avoid the common errors that often undermine immunological data. Notably, there are comprehensive sections of all major immune cell types with helpful Tables detailing phenotypes in murine and human cells. The latest flow cytometry techniques and applications are also described, featuring examples of the data that can be generated and, importantly, how the data can be analysed. Furthermore, there are sections detailing tips, tricks and pitfalls to avoid, all written and peer-reviewed by leading experts in the field, making this an essential research companion.

CD55 and CD59 are complement regulatory proteins suggested to be related with progression of diabetes and its complications. The stromal cell-derived factor 1 (SDF-1) and C-X-C chemokine receptor type 4 (CXCR-4) are chemokine proteins. We aimed to investigate the relation of CD55 and CD59 expression levels and polymorphisms of SDF-1 and CXCR-4 with type 2 diabetes mellitus (T2DM) and its complications. Seventy-five T2DM patients and 73 controls were enrolled. Expression levels of CD55 and CD59 were measured by FACS Calibur; qRT-PCR was used to determine SDF-1 and CXCR-4 gene polymorphisms. CD55 and CD59 expressions in patients with nephropathy, retinopathy and cardiovascular disease were significantly lower than controls. Frequency of CXCR-4 T allele carrying was high in patients and created 1.6 fold risk for the disease (p =.07). CXCR-4 a allele carriers had decreased nephropathy; although there was no statistical significance in carrying CXCR-4 T allele, presence of nephropathy was approximately 2 times higher (p =.254). The nephropathy risk increased 10-fold in CXCR-4 TT genotype carriers (p =.02). All SDF-1 CC genotype carriers had retinopathy, so, it was considered that the CC genotype was effective in retinopathy development (p =.031). For the presence of cardiovascular disease, significant difference was observed for SDF-1 genotypes. Increased cardiovascular risk of 5- and 1.9-fold in SDF-1 T (p =.007) and CXCR-4 T (p =.216) allele carriers, respectively, was observed. We suggest that CD55 and CD59 protein levels and SDF-1 and CXCR-4 have predictive importance in process, complications and tendency of T2DM. [ABSTRACT FROM AUTHOR]

Cystic fibrosis (CF) is an important genetic disorder that usually manifests in early childhood. Better understanding and improved care of CF patients in our current practice with pancreatic enzyme replacement therapy and nutritional support has enabled physicians to see them more frequently from any age groups. After the lungs, pancreas is the most important organ involved with CF. Its most frequently affected component is the exocrine pancreas while endocrine functions may also be deranged. Parenchymal damage as acute or chronic pancreatitis may also develop. Herein, we aimed to outline the types of the disease, clinical picture, diagnosis and genetic aspect of the CF linked with the pancreatic involvement.

In the recent years, economic performance in Turkey has been praised due to its sustainedgrowth trends, stability and capacity to weather the global financial crisis. However, a number of issues related to the labor market and unemployment trends continue to be the economic and political underbelly to these positive trends. The conspicuously low labor forceparticipation among women over the years deserves special attention among these problems. The objective of this report is to provide an institutional analysis of public policies and programstargeting women’s access to labor markets and entrepreneurial activities. The report seeks to provide a comprehensive inventory of public programs, targeting women’s employment and entrepreneurship and evaluate the strengths and weaknesses of these programs with respect to their sustainability, institutional commitment, and their ability to address barriers to women’s labor force participation. In Turkey, women’s participation in the labor force has historically been low. While 34 percentof the women were in the labor force in 1990, the rate declined to 26 percent over the courseof the following decade and recovered up to 29 percent by 2012. Cross-country comparisons show that women in Turkey participate in the labor force at a much smaller rate compared to countries in all income groups. This is the case even in comparison with the lower-middle income countries, where women’s labor participation rate is 40 percent. A final focus in this study has been on the initiatives that aim to encourage women’s entrepreneurship. Given that the percentages of women among entrepreneurs in Turkey are even lower than that of women in the labor force, if the barriers to women’s entrepreneurial activities are removed, it appears that there is substantial room for improvement. This report sets its goal as interrogating in a holistic manner, programs’ objectives, contents,and ability to address problems standing in the way of women’s labor force participation. The aim of the report is not to conduct an impact analysis of these public programs, but to provide an extensive inventory of institutions and the specific departments and the programs that are directly responsible to carry out the public policies on the issue of women’s employment and entrepreneurship. Programs evaluated in this report were chosen on the basis of their scope both in terms of their targets and funding schemes. Local programs with very narrow scopes were not taken into the inventory. Only the programs, which target to create new employment and entrepreneurship opportunities for women with a wide sphere of influence, were inc luded. The public programs covered in this report can be also assessed based on the sectors ofeconomic activity in which women’s employment and entrepreneurship is promoted: Our report shows that the programs carry the danger of repeating existing gendered divisions of labor and the ensuing capacity to access the labor market. For long-term positive results inchanging existing horizontal and vertical segregations in the market, there should be more focus on how to change gendered assumptions about divisions of labor within the programs’design.

Introduction/ Purpose: The aim of this study is to review the pediatric ovarian torsions and to determine causing lesions, as well as the role of reticulum network to discern underlying tumor in a fully necrotic material. Material and methods: This study included 22 females who underwent unilateral salpingo-oophorectomy procedure because of ovarian torsion in a children’s research hospital between 1989- 2003. The hematoxylin-eosin stained slides (mean 4 slides) and one Gomori’s reticulum stained slide of each case were retrospectively reviewed. The reticulum network was evaluated as rich, poor or absent. Results: The mean age of 22 cases was 8 years (1 month- 14 years). Twelve were right ovary and 10 were left. The mean macroscopic diameter of the torsed ovary was 7.5 cm. Three cases had underlying solid tumor: dysgerminoma, endodermal sinus tumor and mixed germ cell tumor. In 10 cases cysts were detected and in 5 cases mature cystic teratoma was diagnosed. And in the other 4 cases no underlying lesion was detected. Conclusion: Torsion and hemorrhagic necrosis of ovary occurring from ovarian mass is very rare. The main reason of this is prior pelvic surgery in adults. But in pediatric age group, ovarian torsion is the most frequent cause of an ovarian mass requiring surgery. Because of reticulum rich parenchyma of ovary, reticulum network detection is useful of the differential diagnosis of torsion’s reason. We found that the reticulum network was rich in torsed ovarian parenchyma without solid tumors or mature cystic teratoma. In pediatric patients with torsed ovary underlying a solid tumor reticulum network was poor or absent. Finally we conclude that an underlying lesion more commonly occurs in children than adults in torsed ovary and reticulum network might help in differential diagnosis, Giriş ve amaç: Çalışmamızın amacı pediatrik olgularda, torsiyone ovaryuma ait tümüyle nekrotik örneklerde; altta yatan hazırlayıcı etkenin belirlenmesi için retiküler çatıyı incelemenin yararını irdelemektir. Olgular ve yöntem: Hastanemizde 1989- 2003 yılları arasında unilateral salpingo-ooforektomi yapılmış 22 olgu yeniden değerlendirildi. Demonstratif bloklardan yapılan kesitlere Gomori retikulum boyası uygulandı ve örnekler retikulumdan zengin, fakir ve damar duvarı dışında retikulum ağı yok şeklinde semi-kantitatif olarak sınıflandırıldı. Bulgular: Ortalama yaş 8 (1 ay-14 yıl), ortalama çap 7.5 cm bulundu. Materyal 12 olguda sağ, 10 olguda sol ovaryuma aitti. 3 olguda solid tümör varlığı (disgerminom, mikst germ hücreli tümör ve endodermal sinus tumörü), 5 olguda ise matür kistik teratom saptandı. 10 olguda folikül kistleri belirlendi. 4 olguda ise hazırlayıcı bir etken bulunamadı. Sonuçlar: Erişkinde ovaryum tümörüne bağlı torsiyon ve hemorajik nekroz oldukça ender olup; hazırlayıcı başlıca etken daha önce uygulanmış cerrahi girişimlerdir. Çocuklarda ise jinekolojik cerrahi girişim gerektiren en sık neden ovaryumda kitle varlığıdır. Çalışmamızda tümördışı torsiyonların, olağan ovaryum stroması gibi retikulumdan zengin olduğunu; solid tümör ve matür kistik teratom bulunan olgularda bu zengin retikuler çatının yok olduğunu gözledik. Sonuç olarak çocuklarda altta yatan bir lezyona bağlı ovaryum torsiyonlarının erişkine göre daha sık olduğunu; bu nedenle özellikle pediatrik olgularda nekroz nedeniyle dokunun doğası seçilemediği zaman retikulum boyamanın yardımcı olabileceği sonucuna ulaştık
