Microglia are glial cells centrally related to pathophysiology and neuroimmunological regulation of pain through microglia–neuron crosstalk mechanisms. In contrast, anti-inflammatory mechanisms guided by immunological effectors such as IL-10 trigger the secretion of analgesic substances, culminating in the differential expression of genes encoding endogenous opioid peptides, especially β-endorphin. Thus, when β-endorphin binds to the µ-opioid receptor, it generates neuronal hyperpolarization, inhibiting nociceptive stimuli. This review aimed to summarize the recent advances in understanding the mechanism by which IL-10/β-endorphin can reduce pain. For this, databases were searched for articles from their inception up until November 2022. Two independent reviewers extracted the data and assessed the methodological quality of the included studies, and seventeen studies were considered eligible for this review. Several studies have demonstrated the impact of IL-10/β-endorphin in reducing pain, where IL-10 can stimulate GLP-1R, GRP40, and α7nAChR receptors, as well as intracellular signaling pathways, such as STAT3, resulting in increased β-endorphin expression and secretion. In addition, molecules such as gabapentinoids, thalidomide, cynandione A, morroniside, lemairamin, and cinobufagin, as well as non-pharmacological treatments such as electroacupuncture, reduce pain through IL-10 mediated mechanisms, reflecting a microglia-dependent β-endorphin differential increase. This process represents a cornerstone in pain neuroimmunology knowledge, and the results obtained by different studies about the theme are presented in this review.

Ceratozamia Brongn. is one of the species-rich genera of Cycadales comprising 38 species that are mainly distributed in Mexico, with a few species reported from neighboring regions. Phylogenetic relationships within the genus need detailed investigation based on extensive datasets and reliable systematic approaches. Therefore, we used 30 of the known 38 species to reconstruct the phylogeny based on transcriptome data of 3954 single-copy nuclear genes (SCGs) via coalescent and concatenated approaches and three comparative datasets (nt/nt12/aa). Based on all these methods, Ceratozamia is divided into six phylogenetic subclades within three major clades. There were a few discrepancies regarding phylogenetic position of some species within these subclades. Using these phylogenetic trees, biogeographic history and morphological diversity of the genus are explored. Ceratozamia originated from ancestors in southern Mexico since the mid-Miocene. There is a distinct distribution pattern of species through the Trans-Mexican Volcanic Belt (TMVB), that act as a barrier for the species dispersal at TMVB and its southern and northern part. Limited dispersal events occurred during the late Miocene, and maximum diversification happened during the Pliocene epoch. Our study provides a new insight into phylogenetic relationships, the origin and dispersal routes, and morphological diversity of the genus Ceratozamia. We also explain how past climatic changes affected the diversification of this Mesoamerica-native genus.

Potassium niobate ( KNbO 3) is an important ferroelectric and photorefractive material that finds multiple nonlinear optical and photorefractive applications such as two-beam coupling and dynamic holography. We employ standard Density Functional Theory to examine two defect complexes in KNbO 3 doped with 3-d transition metals. These defect complexes involve the substitution of a Nb with a 3-d transition metal element and (1) a coordinating oxygen vacancy induced in the nearest-neighbor oxygen shell or (2) a non-coordinating oxygen vacancy induced in the supercell as far away as possible from substitution impurity. With the exception of Ti and V, the 3-d transition metal dopants studied here may represent viable dopants for purposes of extending the photorefractive spectral response of KNbO 3. Our results indicate that each of these complexes introduces defect states into the gap of KNbO 3. Furthermore, we notice that the distribution of impurity states changes quite systematically across the 3-d transition metal row, and we note the effects of the disruption of the crystal field in the case of the coordinating O vacancy on the position and ordering of defect states within the gap. Substituting KNbO3 with transition metal ions can, therefore, significantly affect its optical and infrared properties, exemplifying how a detailed understanding of the electronic structure can be an important tool in tailoring the properties of this material for ferroelectric and photorefractive applications. The case study presented in this paper examines the work of one high school chemistry teacher who has integrated guided inquiry into a yearlong, traditional curricular framework in ways that take into account the constraints and realities of her classroom. The study's findings suggest (1) the extent and frequency to which teachers can realistically integrate guided inquiry into existing science curricula, (2) the manner in which teachers can make effective transitions into and out of a guided inquiry, and (3) the way in which teachers can set an overarching goal to bring a driving purpose to all of the guided inquiries that the students experience. Based on the insights gained from the current case study, an argument is made that researchers should examine the practice of other teachers (especially in the common subjects of biology, chemistry, and physics) who have integrated guided inquiry into the fabric of their curricula. By doing so, a library of case studies can be built that will provide science teachers with new ideas on how inquiry can be holistically and productively integrated into yearlong traditional curricular frameworks. Objetivo do estudo foi investigar os efeitos de três repetições de 10 segundos de permanência do método estático sobre os ganhos de amplitude de movimento em adultos jovens não atletas. Os 59 sujeitos foram divididos em um grupo experimental (GE; n=41), que foi submetido a exercícios de alongamento estático por 16 semanas, com aplicação de três repetições de 10 segundos de tempo de permanência na posição de cada movimento e um grupo controle (GC; n=18), que não foi submetido a nenhum treino. Foram avaliados os movimentos de abdução do ombro (AO) e flexão de quadril (FQ) através de goniometria. A ANOVA de medidas repetidas revelou aumentos significativos no GE do pré para o pós-teste no movimento AO (Δ% = 8,9; p = 0,00) e FQ (Δ% = 18,7; p= 0,00). No pós-teste não houve diferenças intergrupos. Conclui-se que o método proposto foi suficiente para produzir aumento significativo nos níveis de flexibilidade nos movimentos analisados.

Plant phototropism, the ability to bend toward or away from light, is predominantly controlled by blue-light photoreceptors, the phototropins. Although phototropins have been well-characterized in Arabidopsis thaliana, their evolutionary history is largely unknown. In this study, we complete an in-depth survey of phototropin homologs across land plants and algae using newly available transcriptomic and genomic data. We show that phototropins originated in an ancestor of Viridiplantae (land plants + green algae). Phototropins repeatedly underwent independent duplications in most major land-plant lineages (mosses, lycophytes, ferns, and seed plants), but remained single-copy genes in liverworts and hornworts—an evolutionary pattern shared with another family of photoreceptors, the phytochromes. Following each major duplication event, the phototropins differentiated in parallel, resulting in two specialized, yet partially overlapping, functional forms that primarily mediate either low- or high-light responses. Our detailed phylogeny enables us to not only uncover new phototropin lineages, but also link our understanding of phototropin function in Arabidopsis with what is known in Adiantum and Physcomitrella (the major model organisms outside of flowering plants). We propose that the convergent functional divergences of phototropin paralogs likely contributed to the success of plants through time in adapting to habitats with diverse and heterogeneous light conditions.

Background and Aims Cycads are regarded as an ancient lineage of living seed plants, and hold important clues to understand the early evolutionary trends of seed plants. The molecular phylogeny and spatio-temporal diversification of one of the species-rich genera of cycads, Macrozamia , have not been well reconstructed. Methods We analysed a transcriptome dataset of 4740 single-copy nuclear genes (SCGs) of 39 Macrozamia species and two outgroup taxa. Based on concatenated (maximum parsimony, maximum likelihood) and multispecies coalescent analyses, we first establish a well-resolved phylogenetic tree of Macrozamia. To identify cyto-nuclear incongruence, the plastid protein coding genes (PCGs) from transcriptome data are extracted using the software HybPiper. Furthermore, we explore the biogeographical history of the genus and shed light on the pattern of floristic exchange between three distinct areas of Australia. Six key diagnostic characters are traced on the phylogenetic framework using two comparative methods, and infra-generic classification is investigated. Key Results The tree topologies of concatenated and multi-species coalescent analyses of SCGs are mostly congruent with a few conflicting nodes, while those from plastid PCGs show poorly supported relationships. The genus contains three major clades that correspond to their distinct distributional areas in Australia. The crown group of Macrozamia is estimated to around 11.80 Ma, with a major expansion in the last 5–6 Myr. Six morphological characters show homoplasy, and the traditional phenetic sectional division of the genus is inconsistent with this current phylogeny. Conclusions This first detailed phylogenetic investigation of Macrozamia demonstrates promising prospects of SCGs in resolving phylogenetic relationships within cycads. Our study suggests that Macrozamia , once widely distributed in Australia, underwent major extinctions because of fluctuating climatic conditions such as cooling and mesic biome disappearance in the past. The current close placement of morphologically distinct species in the phylogenetic tree may be related to neotenic events that occurred in the genus. [ABSTRACT FROM AUTHOR]
