Purpose: To evaluate changes in color and translucency of stained and glazed monolithic lithium disilicates and zirconia after toothbrushing with conventional or whitening dentifrices followed by thermocycling., Materials and Methods: Samples (n &#61; 20/material) from CAD/CAM (IPS e.max CAD [EC]) and pressable (IPS e.max Press [EP]) lithium disilicates and 5Y-PSZ zirconia (Prettau 4 Anterior [PA]) were prepared and received staining and glazing procedures. Aging was performed by toothbrushing with conventional (n &#61; 10) or whitening (n &#61; 10) dentifrices, followed by thermocycling simulating 5, 10, and 15 years. Color coordinates L*, a*, b*, , and C* and h° values were assessed with a spectrophotometer at baseline and after each aging period to calculate color changes (ΔE00), lightness (ΔL*), and translucency parameter (TP). Data were analyzed using three-way repeated measures ANOVA's followed by Sidak post-hoc test (α &#61; .05)., Results: When comparing the dentifrices, EC and EP produced the greatest ΔE00 after 15 years with the whitening dentifrice (P < .05). Regardless of the dentifrice, PA showed greater ΔE00 than EC after 10 years (P < .05). When comparing the aging periods, EC and PA did not show differences in ΔE00 for both dentifrices (P ≥ .05). EC and EP produced an increased ΔL* after 15 years in the whitening group (P < .05). In the whitening groups, the ΔL* values of PA differ (P < .005) from those of lithium disilicates. Regardless of the experimental group, the TP values followed the inequality: PA < EC < EP (P < .05)., Conclusions: Overall, the color of stained and glazed monolithic lithium disilicates and zirconia changed under aging with greater effects for whitening dentifrice. Aging did not influence translucency.

Objectives: This laboratory study assessed the performance of a novel fluoride dentifrice containing micro-fibrillated cellulose (MFC) and entrapped silica., Methods: Removal of extrinsic stains was assessed using the pellicle cleaning ratio (PCR) method, and radioactive dentin abrasivity (RDA) was measured, to calculate a cleaning efficiency index (CEI). Fluoride efficacy was evaluated using widely used remineralization and fluoride uptake methods. The test product (Protegera™) was compared to common dentifrices (Crest - Cavity Protection™ and ProHealth™, Sensodyne Pronamel™, Arm & Hammer™ Advanced Whitening, Crest ProHealth™, and Colgate Optic White™)., Results: The PCR for the MFC dentifrice (141) was comparable to three known marketed stain-removing dentifrices (Arm & Hammer™ Advanced Whitening, Crest ProHealth™, and Colgate Optic White™) but it had a significantly lower RDA (88 ± 6) than 5 other products. This gave it the highest CEI of the tested products (2.0). In a 10-day pH cycling study, the fluoride efficacy of the MFC product was comparable to Sensodyne Pronamel and Crest Cavity Protection. The MFC dentifrice was superior for promoting fluoride uptake into incipient enamel lesions compared to the USP reference dentifrice., Conclusion: The MFC dentifrice has low abrasion, but despite this, it is highly effective in removing stained pellicle. It also is an efficacious fluoride source when compared to relevant commercially available fluoride dentifrices with high dentin abrasivity., Clinical Significance: The addition of micro-fibrillated cellulose to a fluoride dentifrice gives a low abrasive product that can effectively remove external stains, and serve as an effective fluoride source. Specific products containing natural resources can contribute to the innovation of complete denture hygiene., Objective: To conduct an in vitro evaluation of experimental dentifrices containing essential oils of Bowdichia virgilioides Kunth (BvK), Copaifera officinalis (Co), Eucalyptus citriodora (Ec), Melaleuca alternifolia (Ma) and Pinus strobus (Ps) at 1%., Methodology: The variables evaluated were organoleptic and physicochemical characteristics, abrasiveness (mechanical brushing machine) simulating 2.5 years, and microbial load (Colony Forming Units - CFU/mL), metabolic activity (XTT assay) and cell viability (Live/Dead® BacLight™ kit) of the multispecies biofilm (Streptococcus mutans: Sm, Staphylococcus aureus: Sa, Candida albicans: Ca and Candida glabrata: Cg). Specimens of heat-polymerized acrylic resins (n=256) (n=96 specimens for abrasiveness, n=72 for microbial load count, n=72 for biofilm metabolic activity, n=16 for cell viability and total biofilm quantification) with formed biofilm were divided into eight groups for manual brushing (20 seconds) with a dental brush and distilled water (NC: negative control), Trihydral (PC: positive control), placebo (Pl), BvK, Co, Ec, Ma or Ps. After brushing, the specimens were washed with PBS and immersed in Letheen Broth medium, and the suspension was sown in solid specific medium. The organoleptic characteristics were presented by descriptive analysis. The values of density, pH, consistency and viscosity were presented in a table. The data were analyzed with the Wald test in a generalized linear model, followed by the Kruskal-Wallis test, Dunn's test (mass change) and the Bonferroni test (UFC and XTT). The Wald test in Generalized Estimating Equations and the Bonferroni test were used to analyze cell viability., Results: All dentifrices showed stable organoleptic characteristics and adequate physicochemical properties. CN, Ec, Ps, Pl and PC showed low abrasiveness. There was a significant difference between the groups (p<0.001) for microbial load, metabolic activity and biofilm viability., Conclusions: It was concluded that the BvK, Ec and Ps dentifrices are useful for cleaning complete dentures, as they have antimicrobial activity against biofilm. The dentifrices containing Bowdichia virgilioides Kunth showed medium abrasiveness and should be used with caution.

Aim: The purpose of the current study was to evaluate the impact of three various mouthwashes on the effectiveness of fluoride dentifrices in preventing enamel erosion., Materials and Methods: A total of 120 sound intact human premolar teeth which were extracted for orthodontic treatment were selected for the study. A 3 × 3 mm window section was positioned in the middle of the coronal surface of the tooth in order to define the study area. Each sample was placed in a solution of 1% citric acid (pH 3.5) for 10 minutes in order to produce an eroded surface. All samples were divided into two main groups (60 samples each) as follows: Group A for sodium fluoride dentifrices and group B for stannous fluoride dentifrices, again it is subdivided into: CHX: Chlohex ADS®, EO: Listerine®, CPC: Colgate® Plax (20 samples in each subgroup). After that, samples underwent the pH cycling model for 5 days. Samples were examined for surface loss using a scanning electron microscope., Results: In sodium fluoride dentifrices group, before intervention, the surface loss was 3.12 ± 1.03 in CHX group, 3.08 ± 1.20 in EO group, and 3.09 ± 0.96 in CPC group. After intervention, the less surface loss found with CHX group (2.18 ± 0.84), followed by CPC (2.34 ± 0.74) and EO group (2.46 ± 0.97). In stannous fluoride dentifrices group, before intervention, the surface loss in CHX group was 3.26 ± 1.19, in EO group, it was 3.18 ± 1.31, and in CPC group, it was 3.22 ± 1.06. After intervention, the less surface loss found with CHX: group (1.90 ± 0.54), followed by CPC (2.24 ± 0.28) and EO group (2.38 ± 0.20)., Conclusion: The present study concluded that the fluoride dentifrices' preventive effects against tooth surface loss were unaffected by a different mouthwashes with varying compositions and major constituents. In terms of erosion, fluoridated toothpaste containing stannous fluoride was found to provide better surface loss protection than sodium fluoride., Clinical Significance: Primary prevention and the eradication of contributing causes are the greatest strategies for preventing erosion. Simultaneously, antibacterial agent in the mouthwashes may help in enhancing the effect of fluoride in the enamel, owing to their high affinity for teeth structures. Therefore, in addition to cause-related treatment, further efforts to reduce tooth tissue loss are also necessary.

The present study evaluated the effect of high-fluoride dentifrice on dentine demineralization and bacterial composition in a multispecies biofilm model in vitro . A seven-organism bacterial consortium was grown on bovine dentine discs in a high-throughput active attachment model. The biofilms were submitted twice per day to the following dentifrices treatments: 5,000 ppm F, 1,100 ppm F, with placebo as a negative control. After 5 days of biofilm growth, dentine samples were assessed by transversal microradiography, the biofilm was collected for bacterial counts and the pH of the media was determined. Lower integrated mineral loss values were observed when 5,000 ppm F-treatment was used compared to the other treatments. Overall microbiological counts decreased with increasing F-concentration as well the pH of the media throughout the experiment. The 5,000 ppm F-treatment caused a shift in microbial composition and reduced dentine demineralization in the in-vitro experimental model.

Ethnopharmacological Relevance: Chewing sticks have served as the primary form of dental care for rural communities in resource-poor settings for millennia. They are one of the most important under-researched, non-timber forest products in Namibia. This review provides an overview of plants that are used as chewing sticks in Namibia and highlights pharmacological as well as phytochemical studies conducted on them., Aim of the Study: This review aims to present a summary of studies that have been done on the ethnomedicinal uses, phytochemistry, biological activity as well as evidence on the scientific validation and geographical distribution of chewing sticks in Namibia. It also highlights research gaps and provides an impetus for the scientific investigations of these plant species., Material and Methods: Literature searches using keywords including oral hygiene, chewing sticks, ethnomedicinal uses, phytochemistry, antimicrobial, antioxidants, anti-inflammatory activities and toxicity studies, chewing sticks, and distribution in Namibia on various electronic search engines was conducted., Results: Of the 41 plant species identified, Cordia sinensis Lam., Faidherbia albida (Delile) A.Chev. and Harpagophytum zeyheri Decne. are used for both gargling and as mouthwash. The plant families Fabacae, Ebenaceae, and Burseraceae account for 22.0%, 12.2%, and 7.30% of plant species recorded as chewing sticks in Namibia respectively. This study revealed a significant relationship between plant family and scientific validation. Species belonging to Burseraceae, Apocynaceae, Montiniaceae, and Cucurbitaceae families have only been partially validated. The Kunene region, home to the Ovahimba ethnic group, had the highest proportion (87.8%) of chewing sticks species compared to other regions., Conclusion: This review revealed that most of the plants used as chewing sticks in Namibia require an in-depth pharmacological and phytochemical investigation as deduced from the paucity of literature on the therapeutic methods, mechanisms of action, efficacy, toxicity, and clinical relevance of these species., (Copyright © 2021 Elsevier B.V. The physicochemical properties, especially pH value of dental medicines, have significant influence on the health of oral cavity tissues. The pH of formulations should correspond to the value of saliva pH (5.5-8.0). For carbomer-based gels, the required pH value is obtained by neutralizing them with alkaline components, which leads to their structuring (thickening). This affects the physical properties of the gel, its residence time at the application site and the rate of release of active pharmaceutical ingredient. Therefore, the main purpose of this study is to evaluate the rheological, textural, and biopharmaceutical properties of Carbomer Polacril ® 40P-based dental gel depending on the pH value. Evaluation of the rheological properties of gel preparations were performed by measuring the structural viscosity of the samples as a function of pH and temperature. The textural properties of the gel were evaluated by performing tests regarding back extrusion and spreadability. Carbomer Polacril ® 40P-based gels haven't shown noticeable thixotropic behavior, and were characterized by plastic flow in the whole studied pH range. The structural viscosity at the selected average pH value hasn't differed at storage (25 °C) and application (37 °C) temperature. Texture studies of dental gels have shown a strong correlation with rheoparameters. Their rheological behavior and textural properties haven't changed significantly between the pH range of 5.5-6.6. The relatively narrow range of working pH values does not affect the change in the viscosity of the preparation significantly and, consequently, does not affect the release of APIs from the developed Carbomer Polacril ® 40P-based dental gel.

Background: High-fluoride dentifrice is used to manage root caries, but there is no evidence whether its association with nanohydroxyapatite could provide an additional protection for root caries. Therefore, this study aimed to develop and evaluate the effect of an experimental dentifrice with high fluoride (F - ) concentration and nanohydroxyapatite (nano-HA) on root dentin demineralization., Materials and Methods: After formulation of dentifrices, root dentin specimens were randomly assigned to six groups (n = 10) using different dentifrice treatments: placebo; nano-HA without F - ; 1,100 µg F - /g; 1,100 µg F - /g + nano-HA; 5,000 µg F - /g; and 5,000 µg F - /g + nano-HA. A pH cycling model was performed for 10 days, in which treatments were performed twice a day. After that period, the longitudinal hardness was evaluated and the area of demineralization (ΔS) was calculated. The formulated dentifrices were evaluated for primary stability, cytotoxicity, and other technical parameters. Two-way ANOVA and Tukey's test with p set at 5% were used for data analysis., Results: The experimental dentifrices were stable and had no cytotoxicity. Regarding dentin demineralization, the placebo group significantly increased ΔS compared to all other treatment groups (p<0.001). The dentifrices containing 5,000 µg F - /g, regardless of the presence of nano-HA, led to a smaller lesion area in relation to the other treatments (p<0.001)., Conclusion: The findings of this study suggest that nano-HA reduced dentin demineralization, and dentifrice with 5,000 µg F - /g dentifrices, regardless of the presence of nano-HA, showed a greater reduction in root dentin demineralization., Competing Interests: The authors declare that they have no conflicts of interest for this work., (© 2020 Leal et al.)

BACKGROUND The purpose of this study is to evaluate and compare color stability of 3 resin based restorative materials when immersed into staining media at 0, 3, 30, 45, 60, and 75 days, and to assess the efficacy of surface polishing in reducing possible discoloration. MATERIAL AND METHODS Ninety composite discs were prepared in a custom-made mold (5 mm diameter and 2 mm thick) from 3 different light cured composites (Filtek™ Z250, Harmonize™, and G-aenial). Color differences of all specimens were measured by a spectrophotometer according to the CIE L*a*b* system. After baseline color measurements, 30 discs from each composite group were randomly divided into 3 subgroups of 10 specimens each. Subgroups (n=10) were immersed for 75 days into different staining solutions: coffee, tomato sauce, and distilled water (control). Solutions were changed every week and color measurements were repeated every 15 days. At day 75, color measurements of the specimens were performed before and after surface polishing with aluminum oxide discs. The experimental data were statistically evaluated using repeated measures one-way ANOVA test followed by Tukey's multiple pairwise comparison with a significance level of 5%. RESULTS Of the 3 composites, G-aenial reported the highest color change deviation when immersed into coffee (ΔE=8.674), and tomato sauce (ΔE=7.737) at day 75, followed by Harmonize that also exhibited a significant difference for coffee (ΔE=4.7) and tomato sauce (ΔE=3.8) when compared to distilled water. While Filtek™ Z250 did not show any significant difference between the 3 storage solutions (P>0.05). Only G-aenial had significant color change (P<0.05) after polishing with aluminum oxide discs for all tested samples, whereas Filtek™ Z250 and Harmonize presented no significant difference after surface polishing (P>0.05). CONCLUSIONS Under the tested experimental conditions, Filtek™ Z250, among the 3 resin-based composites, exhibited the highest color stability when subjected to coffee and tomato sauce as well. Whereas, G-aenial presented the highest color deviation when immersed in both staining media. Surface polishing effectively reduced coffee and tomato sauce discolorations for G-aenial's specimens and had no significant effect regarding Filtek™ Z250 and Harmonize.

Nontoxic and biodegradable chitosan is potentially useful in various applications. We prepared submicron chitosan particles with high dispersibility in aqueous solution utilizing the electrostatic interaction phase separation method described in a previous report, but using citric acid as the polyvalent anionic compound instead of sodium sulfate. The submicron chitosan particles showed significant antibacterial activity and anti-adhesive action against Streptococcus mutans , even at around neutral pH. However, chitosan granules showed no antibacterial activity under the same conditions. The addition of the chitosan particles to dental polishing paste provided stainless steel discs (the same hardness as dental enamel) with a smoother surface than polishing paste without additives. In view of their submicron size and antibacterial activity, chitosan particles could potentially be multifunctional components of oral and dental cleaning materials.

Two analytical methods were developed using electrochemical and spectrometric techniques for the simultaneous determination of endocrine disruptors triclosan and methylparaben in the monitoring of personal care products. For the electroanalytical analyses, a sensitive electrode based on graphene quantum dots supported in chitosan was employed. Under optimized conditions and a working potential of typically + 0.60 V for triclosan and + 0.81 V (vs. Ag/AgCl) for methylparaben, the calibration plots obtained by differential pulse voltammetry were linear in the range 0.10 to 10.0 μmol L -1 . The detection limits were 0.03 and 0.04 μmol L -1 for triclosan and methylparaben, respectively. For the spectrometric method, UV/VIS spectrometry was used with a mathematical processing of non-linear deconvolution. This processing was used to solve the problem of overlapping absorption bands of triclosan (282 nm) and methylparaben (257 nm), which enabled simultaneous determination. The calibration plots by UV/VIS spectrometry were linear in the range 1.0 to 14.0 μmol L -1 with detection limits of 0.42 and 0.37 μmol L -1 , respectively, for triclosan and methylparaben. Similar results obtained from the calibration plots of individual analytes suggest that the methods can be applied for individual or simultaneous determination of these species. Both methods were employed in the analysis of five samples of personal care products: toothpaste, antiseptic soap, antiseptic deodorant, shampoo, and a bath kit (soap and shampoo). The statistical tests indicated that there were no significant differences regarding the accuracy and precision of the data provided by the two methods described herein. Objectives: The aim of this study was to investigate the effect of a dentifrice that contains calcium silicate, sodium phosphate, and fluoride on erosive-abrasive enamel wear., Material and Methods: This randomized, single-blind in situ/ex vivo study was conducted with four crossover phases of 5 days (one group tested per phase). Bovine enamel blocks (n = 256) were allocated to 16 volunteers and 8 groups. The groups under study were test dentifrice, with calcium silicate, sodium phosphate, and 1450 ppm sodium monofluorophosphate; tin dentifrice, with 3500 ppm stannous chloride, 700 ppm amine fluoride, and 700 ppm sodium fluoride; conventional dentifrice, with 1450 ppm sodium monofluorophosphate; and control (deionized water). Half of the enamel blocks were subjected to erosion and the other half to erosion plus abrasion. The daily extraoral protocol consisted in four citric acid exposures (2 min) and two applications of dentifrice slurry on all blocks for 30 s; after, half of the blocks were brushed for 15 s. The response variable was enamel loss. Data were analyzed by two-way ANOVA and Fisher's test (p < 0.05)., Results: For erosion, the test dentifrice promoted less enamel loss than water (4.7 ± 3.1 and 5.8 ± 2.5 μm, respectively, p < 0.05), and did not differ from tin (4.8 ± 2.5 μm) and conventional (4.8 ± 1.4 μm) dentifrices (p > 0.05). However, the test dentifrice (7.7 ± 3.8 μm) promoted higher wear after erosive plus abrasive procedures than tin (5.4 ± 1.5 μm) and conventional (6.2 ± 1.7 μm, p < 0.05) dentifrices, and did not differ from water (6.9 ± 2.0 μm)., Conclusions: The investigated dentifrice reduced enamel loss against acid challenge but had no effect against acid and brushing challenge., Clinical Relevance: Little is known regarding the preventive effect of dentifrices indicated for dental erosion. The tested anti-erosive dentifrice was unable to protect enamel when erosion was associated to toothbrushing abrasion.

Anti-caries activity of fluoride ions is due to the protection against demineralization and the enhancement of remineralization of tooth enamel. Dentifrices available on the market contain sodium fluoride, sodium monofluorophosphate, stannous fluoride, and amine fluoride as source of these ions. A new compound working both as fluoride ion source and as abrasive was projected. Hybrids based on F - ions intercalated between the lamellae of hydrotalcite-like compounds (HTlc-F), namely MgAl-HTlc-F and ZnAl-HTlc-F, were prepared and characterized. Then, three different percentages (2, 3, and 4%) of both HTlc-F compounds were assayed. After the rheological characterization, the dentifrices containing 3 and 4% of MgAl-HTlc-F and ZnAl-HTlc-F, respectively, resulted to be the most suitable ones. Two novel in vitro methods, "rotary toothbrush method" and "manual brushing method," were developed and used in order to study the F - ions release from the prepared dentifrices. The obtained results showed that the dentifrice containing ZnAl-HTlc-F (4%) was the most effective in releasing fluoride ions. The "rotary toothbrush method" resulted to be the most suitable as the simulation of the brushing movements is standardizable and reproducible.

This study aimed to evaluate the gastrointestinal absorption and renal excretion of fluoride after the ingestion of high-fluoride dentifrice. Twelve volunteers participated in this in vivo, crossover, and blinded study. In three experimental phases, the volunteers were randomly assigned to one of three treatment groups, who ingested either the following: distilled and deionized water (control), conventional dentifrice (1100 μg/g), or high-fluoride dentifrice (5000 μg/g). Both dentifrices contained fluoride in the form of NaF/SiO 2 . To determine the rate of fluoride absorption, non-stimulated saliva was collected for up to 120 min after ingestion and the area under the curve of the salivary fluoride concentration was plotted as a function of time and the maximum concentration determined. All urine produced during the 24 h before and after ingestion was collected, and urinary excretion was calculated from the difference between the urinary fluoride concentrations in the two periods. A specific ion electrode coupled to an ion analyzer was used to measure fluoride concentrations. Statistical analysis was performed by ANOVA followed by Tukey's test with p set at 5%. All measured parameters were highest after the ingestion of the dentifrice with 5000 μg/g (p < 0.001), confirming that this has an increased level of bioavailable fluoride compared with the conventional dentifrice. The high-fluoride dentifrice increases the concentration of salivary fluoride, which may explain its greater anticaries effect. However, it poses a potential risk of causing dental fluorosis and so should not be used by children.

Objectives: Bioactive glasses which degrade in aqueous solutions may release bioactive ions such as fluoride (F - ) and support fluoride bioavailability in saliva. We investigated how these effects would be apparent in an in vivo experimental trial after toothbrushing in comparison with sodium fluoride and amine fluoride., Material and Methods: In this single-center, randomized, parallel in vivo trial with a three strata block design, where healthy subjects were randomly assigned into three groups. Each group brushed their teeth either with fluoridated bioactive glass containing dentifrice, with a sodium fluoride (NaF) containing dentifrice or with amine fluoride (AmF) containing toothpaste. Saliva was collected time intervals before, immediately after, 30, 60 and 120 min after toothbrushing. Fluoride concentration was determined in supernatant saliva and salivary sediment using a fluoride ion selective electrode. The data were evaluated statistically using non-parametric tests., Results: The increase of bioactive fluoride in supernatant saliva was higher after application of NaF or AmF compared to fluoridated bioactive glass. In salivary sediment bioavailability of fluoride lasted longer after application of fluoridated bioactive glass., Conclusions: Toothbrushing with the fluoride containing bioactive glass dentifrices had positive effects on the fluoride bioavailability within two hours. Fluoride containing bioactive glass represent a new area for investigation in caries prophylaxis. The bioactive potential impact on the tooth remineralization should be examined further., Trial Registration: DRKS00016038 .

Context: The comparison of fluoride levels in saliva and plaque following the use of conventional, 2800 and 5000 ppm dentifrices for different time intervals up to 24 hours has not been explored., Aim: The aim of the present study was to assess salivary and plaque fluoride levels at different time intervals following the use of high fluoride dentifrices., Study Design: This randomised control trial was conducted on sixty adolescents between the age group of 16 and 18 years divided into three groups A, B, and C., Intervention: Subjects were asked to brush the occlusal surfaces of posterior teeth with one of the dentifrices for 2 min. Unstimulated whole saliva and plaque samples were collected at different time intervals. Fluoride levels were determined using SPADNS method. Data was collected and statistically analyzed using SPSS 20 Inc. by mixed repeated measure ANOVA., Results: A significant difference in fluoride levels was observed at different time intervals in both saliva (p=0.048) and plaque (p=0.03). The variance was low with time and concentration of the dentifrice used in saliva, whereas the magnitude of treatment was large for time (>0.25) but small for (<0.09) concentration in plaque., Conclusion: A significant difference in fluoride levels was observed at different time intervals in both saliva and plaque among the three groups. There was a positive correlation between fluoride levels in saliva and plaque. Both high fluoride dentifrices were effective in maintaining higher intraoral fluoride levels up to 12 hours and 5000 ppm up to 24 hours compared to conventional dentifrice.

Purpose: To evaluate the effect on dentin of chondroitin sulfate and L-arginine on dentin tubule occlusion., Methods: The dentin samples were activated by submersion in an aqueous ( aq. ) solution of chondroitin sulfate ( ChS) or L-arginine prior to application of a commercial or custom-made toothpaste. After rinsing with water and ultrasonication, adhesion to dentin and occlusion of dentin tubules were evaluated by scanning electron microscopy and the elemental composition of the deposits was evaluated by energy-dispersive x-ray spectroscopy., Results: Rinsing a dentin sample with a solution of ChS resulted in an increase in the adherence of dentifrices containing either titanium dioxide (TiO₂ ) or calcium-based nanoparticles [ hydroxyapatite ( HA\ or calcium carbonate( to the dentin surface. ChS does not appear to enhance the adherence of dentifrices lacking TiO₂. Pretreatment by L-arginine improved adherence of calcium carbonate nanoparticles, but less efficiently than ChS. Addition of nanoparticles of hydroxyapatite or calcium citrate to dentifrices improved their adherence to dentin without any pre-treatment., Clinical Significance: The significant increase in adherence to the dentin surface of dentifrices of either TiO₂ or calcium-supplying nanoparticles to the dentin surface following pre-treatment with ChS or L-arginine opens the door to the development of two-step dental treatments, which accomplish dentin tubule occlusion and help to deliver active dentifrice components to the dentin surface. Purpose: To evaluate the effect of experimental (Ricinus communis) and commercial dentifrices used for denture cleaning on abrasiveness (gravimetric method; roughness), hardness, and color stability of a resilient relining material., Materials and Methods: Sixty circular (15 × 3 mm) specimens were distributed into four groups: C (control; brushing with water); CO (brushing with Colgate - for natural teeth); CB (brushing with Corega Brite - for complete dentures); RC (brushing with experimental dentifrice). Brushing was performed in a toothbrushing machine with a soft brush and a dentifrice suspension for 50 minutes, calculated to correspond to 1 year of regular brushing. Variables were measured initially and after the trial period. For the gravimetric method, the difference in mass was considered. The surface roughness was measured by a rugosimeter, and the hardness test was performed by a Shore A durometer. Color changes (ΔE; CIE L*a*b* and NBS systems) were measured by a portable spectrophotometer. Results were analyzed by ANOVA/Tukey's test (α = 0.05)., Results: The largest mass variation (μg; p < 0.0001) occurred in C (-6.21 ± 3.18). Concerning roughness, CB (0.26 ± 0.04) showed the lowest value, followed by RC (0.29 ± 0.08) and CO (0.34 ± 0.24) (p < 0.0001). Group C produced the greatest surface roughness (0.72 ± 0.25). Hardness values decreased after brushing with water (p = 0.014). No significant differences were found among RC (50.31 ± 1.03), CO (49.11 ± 1.31), CB (49.17 ± 1.23), and C (48.02 ± 1.26). Color stability was similar in all groups (p = 0.135; C: 2.3 ± 0.77; CO: 2.6 ± 0.54; CB: 2.2 ± 0.44; RC: 2.9 ± 1.56)., Conclusions: The use of experimental dentifrice could be indicated, as it showed similar results to the specific dentifrice, keeping the resilient material properties within acceptable values., (© 2017 by the American College of Prosthodontists.)

Objectives:: To evaluate the effect of a remineralizing gel combining fluoride and calcium silicate/phosphate or a sodium fluoride gel on bleached enamel microhardness, color, and wear susceptibility., Methods and Materials:: Two hundred forty bovine enamel-dentin samples were prepared. Baseline analysis of Knoop microhardness, color coordinates (L*a*b*), and surface profile were performed. According to the baseline microhardness values, specimens were stratified into six groups (n=40): NC (negative control)-no treatment; BL (positive control)-bleaching with 40% hydrogen peroxide gel (Opalescence Boost, Ultradent); BL/Rs-bleaching + application of calcium silicate/phosphate gel (Regenerate Serum, Unilever - Rs); Rs/BL-Rs + bleaching; Rs/BL/Rs-Rs + bleaching + Rs; and BL/F-bleaching + 2% sodium fluoride gel. After the treatment described for each group, color change (ΔE) and microhardness were evaluated again. To evaluate abrasion susceptibility, samples were randomly divided into two subgroups, according to the toothpaste used (Cp-Close Up or Rt-Regenerate), and underwent 100,000 brushing strokes. The profile of each sample was evaluated and the mean wear calculated. The data were analyzed by ANOVA and Tukey tests., Results:: All bleached groups showed a significant reduction of microhardness in relation to the negative control. The groups treated with remineralizing gels showed a significantly higher microhardness and less wear than the positive control, although nonsignificant differences were observed among them. Nonsignificant differences in ΔE were found among bleached groups. The groups brushed with Regenerate toothpaste showed significantly less wear than those brushed with Close Up toothpaste., Conclusions:: The remineralizing gels did not interfere with bleaching efficacy. However, all the treatments minimized the surface hardness reduction caused by the bleaching procedure and enamel loss after abrasion. Regenerate toothpaste resulted in less enamel abrasion.

Objective: To evaluate retention of intraoral fluoride in biofilm and saliva, an experimental dentifrice containing hydrocolloid (tara gum) was used as a controlled-release system for fluoride (F)., Materials and Methods: In a triple-blind randomized crossover trial with washout, 18 individuals used the following different dentifrices for a week: 100-TGF (sodium fluoride NaF associated with tara gum, 1100 mg/L), 50-TGF (50% NaF associated with tara gum + 50% free NaF, 1100 mg/L), PC (free NaF, 1100 mg/L), TG (with tara gum and without F), and placebo (without F or tara gum). On the seventh day of dentifrice use, biofilm was collected at 1 and 12 h, and saliva was collected up to 60 min and 12 h after the last toothbrushing. F concentrations were determined by physico-chemical analysis of fluoride using the hexamethyldisiloxane-facilitated diffusion technique. Data were subjected to two-way analysis of variance (repeated measures) and Spearman's correlation coefficient (p < 0.05) testing., Results: No significant difference was observed with the same dentifrice regarding F retention in biofilm at 1 and 12 h after toothbrushing for the 100-TGF, placebo, and TG groups (p > 0.05). The highest area under the curve values in saliva were found for the 50-TGF, 100-TGF, and PC groups., Conclusion: The dentifrice containing hydrocolloid as a controlled-release system for F promoted F retention in the oral cavity, even at 12 h after brushing., Clinical Relevance: Hydrocolloid added to dentifrices as a controlled-release system for F might contribute to a higher anti-caries effect., Trial Registration: NCT02809014.

Objectives: To compare remineralization of enamel subsurface lesions by fluoride dentifrices with added calcium in a double-blind, randomized, cross-over, in situ study., Methods: Human enamel with subsurface lesions were prepared and inserted into intra-oral appliances worn by volunteers. A slurry (1 g toothpaste/4 ml H 2 O) was rinsed for 60 s, 4 times per day for 14 days. Seven toothpastes were tested: (i) 1450 ppm F (NaF), (ii) 5000 ppm F (NaF), (iii) 1450 ppm F (MFP) with calcium sodium phosphosilicate (CSP), (iv) 1450 ppm F (MFP) with CaCO 3 /Arg, (v) 1150 ppm F (SnF 2 ) with amorphous calcium phosphate (ACP), (vi) 1100 ppm F (NaF) with casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) and (vii) 5000 ppm F (NaF) with functionalized tri-calcium phosphate (TCP). Total (acid soluble) and bioavailable (water soluble) calcium, inorganic phosphate and fluoride levels of the dentifrices were measured using ion chromatography (F/MFP) and spectrophotometry (Ca and inorganic phosphate). Enamel lesion mineral content was measured using transverse microradiography. Data were statistically analysed using a linear mixed model., Results: All calcium and fluoride containing toothpastes released > 90% of bioavailable fluoride and were superior to the respective fluoride alone toothpastes in remineralization of enamel subsurface lesions. The level of remineralization followed the order: CPP-ACP/1l00 ppm F > ACP/1150 ppm F = TCP/5000 ppm F > 5000 ppm F = CaCO 3 /Arg/1450 ppm F = CSP/1450 ppm F > 1450 ppm F. This exploratory study investigated salivary concentrations of silicon, calcium, sodium, and phosphorous over a 60-min time period following the use of a calcium sodium phosphosilicate (CSPS)-containing dentifrice. Participants brushed with a dentifrice containing 5% (w/w) or 0% (w/w) CSPS or swilled with a slurry containing 5% (w/w) CSPS/glycerol. Saliva samples were collected before, and 2, 5, 15, and 60 min after, product use and were analysed using inductively coupled plasma optical emission spectroscopy. Intra-oral pH measurements were also taken. Primary analysis was of centrifuged saliva supernatant containing only dissolved material. At most time points, the CSPS-containing dentifrice and slurry generated significantly more salivary silicon than the dentifrice containing 0% CSPS. At 2-15 min after brushing there was significantly more salivary calcium after use of the CSPS-containing dentifrice and slurry, compared with the 0% CSPS dentifrice; a significant reduction, from baseline, in salivary calcium after use of dentifrice containing 0% CSPS; and an increase in salivary sodium after use of dentifrices containing either 5% or 0% CSPS, but no differences between them. Salivary phosphorous concentration decreased significantly with all treatments 2-5 min after use. There were no significant between-treatment differences in intra-oral pH. Products were generally well tolerated. This research aimed to determine the influence of fluoridated groundwater and 1,100 ppm fluoride dentifrice on biomarkers of exposure to fluoride in preschoolers. A cross-sectional study was performed on thirty preschoolers recruited from naturally fluoridated and non-fluoridated areas (n=15). Total Daily Fluoride Intake (TDFI) from diet and dentifrice, and Daily Urinary Fluoride Excretion (DUFE) was measured over 24 h. Nails samples were collected twice during 30 days. Fluoride analyses were performed using a fluoride-ion-specific electrode. Data were evaluated using the Student and paired t-test, Pearson correlation analysis, multiple linear regression analysis (α≤0.05). Fluoridated groundwater and dentifrice were the dominant sources (r2 > 0.83) of TDFI in children from a naturally fluoridated and a non-fluoridated area, respectively. A positive correlation between TDFI and DUFE (r=0.50), and between [F] in fingernails and toenails (r=0.60) were found in children from a naturally fluoridated area. The [F] in nails of finger and toe were not correlated to TDFI. The consumption of fluoridated groundwater influenced the fluoride concentration in urine. In addition, the use of 1,100 ppm fluoride dentifrice did not influence the fluoride concentration in urine and fingernails.

Objectives: To investigate the interplay among brushing frequency, dentifrice slurry abrasivity, and fluoride content on the surface loss (SL) of incipient enamel caries-like lesions., Methods: Lesions were created in 96 bovine enamel specimens (5 × 5 mm) using methylcellulose acid gel. Specimens were randomly allocated to 12 groups (n = 8), resulting from the association of three experimental factors: (1) slurry abrasive level [low: REA = 4/RDA = 69 and high: REA = 7/RDA = 208], (2) fluoride concentration [275 and 1250 ppm F as NaF], and (3) brushing frequency [1, 2 and 3× daily]. Specimens were kept in artificial saliva in between brushings and overnight. SL was determined by optical profilometry after lesion creation, 1, 3, 5, and 7 days. Data was analyzed with repeated measures ANOVA and Tukey's tests (α = 0.05)., Results: High abrasive slurry caused significantly more SL than low with 275 ppm (p < 0.001) but not with 1250 ppm fluoride (p = 0.34). Fluoride at 275 ppm had significantly more SL than 1250 ppm with high abrasive slurry after 7 days (p = 0.008). Brushing 1×/day had significantly less SL than 3×/day after 7 days with high abrasive slurry (p = 0.016), especially in the 275 ppm fluoride groups., Conclusions: Higher fluoride concentration increased protection against the deleterious effect of high abrasive slurry. SL was higher if brushing was performed more than twice daily especially in low fluoride groups., Clinical Significance: Highly abrasive toothpaste formulations might cause more surface wear to incipient caries lesions, especially at higher brushing frequencies. Objectives: The aim of this in vitro study was to evaluate the role of highly fluoridated dentifrice on remineralization characteristics of lowly and highly pre-demineralized enamel artificial caries lesions., Methods: Bovine enamel specimens were prepared (pH 4.95; 21 days) and discriminated in either lowly [L] or highly [H] pre-demineralized artificial caries lesions. Specimens with a mean ΔZ baseline,L (95% CI) of 5120 (4995; 5245) vol.% × μm and a mean ΔZ baseline,H of 8187 (8036; 8339) vol.% × μm were selected and randomly allocated to 12 groups (n = 20). Treatments during pH-cycling (28 days; 6 × 60 min demineralization/day) were brushing 2×/day with fluoride-free (0 ppm F - [L 0 /H 0 ]), 1100 ppm F - [L 1100 /H 1100 ], 2800 ppm F - [L 2800 /H 2800 ], 5000 ppm F - [L 5000 /H 5000 ], 5000 ppm F -  + glycerin [L 5000 + glycerin /H 5000 + glycerin ], and 5000 ppm F -  + TCP [L 5000 + TCP /H 5000 + TCP ] containing dentifrices. Dentifrice slurries were prepared with deionized water (1:3wt/wt). After cycling specimens presenting lesion surface loss were discarded and for the remaining 202 specimens, transversal microradiographic (TMR) analyses (ΔZ pH-cycle /LD pH-cycle ) were performed again. Changes in mineral loss (ΔΔZ = ΔZ baseline - ΔZ pH-cycle ) and lesion depth (ΔLD = LD baseline - LD pH-cycle ) were calculated., Results: Significant differences for ΔΔZ could be found between L 0 , L 1100 , and L 5000 as well as H 0 , H 1100 , and H 2800 /H 5000 (p ≤ 0.01; ANCOVA). Except for 0 ppm F - , higher ΔΔZ could be found in highly compared with lowly demineralized specimens (p ≤ 0.004; ANCOVA). After pH-cycling, a second lesion front could only be observed in H 5000 and H 5000 + TCP . The correlation between ΔΔZ and F - was moderate for lowly and highly demineralized lesions (r L  = 0.591; p L  < 0.001; r H  = 0.746; p H  < 0.001), indicating a fluoride dose response for both., Conclusion: For both baseline substrate conditions, a dose response for fluoride could be revealed., Clinical Significance: Remineralization characteristics of enamel directly depended on baseline mineral loss.

The oral cavity has its own significant micro-flora but under unhygienic conditions can cause infections or diseases like gingivitis, caries, plaque and gum bleeding. Out of more than 700 oral microbial species, some opportunistic pathogens such as Staphylococcus aureus, Streptococcus spp. and Candida albicans are more prevalent. In this study, the antimicrobial activities of various toothpastes (dilutions ranging from 1:1-1:128) against above mentioned pathogens were assessed. The pathogens were isolated from clinical samples using various differential and selective media and identified through microscopic examination, cultural characteristics and biochemical tests using both conventional and API kit system (Biomerieux, France). Antimicrobial activities of selected dentifrice formulations against identified microbes were determined using agar well diffusion and Minimum Inhibitory Concentration assays. Statistical analysis of the data on different variables has been performed by Analysis of Variance and Mean ±SD using SPSS software. From the collected samples Staphylococcus aureus, Streptococcus mutans, Streptococcus salivarius, Streptococcus intermedius and Candida albicans were isolated and identified. All the selected toothpastes showed significant (p<0.01) antimicrobial activity against the bacterial and fungal isolates. Variable results (inhibitory zone diameters ranging from 35.10±8.00 to 2.40±5.37) were found when mean of different dilutions were compared. Conventional dentifrices exhibited more inhibition as compared to herbal products.

Objective: To evaluate the effect of different fluoride- and calcium- and/or phosphate-containing products on their ability to prevent enamel demineralization under pH cycling conditions., Material and Methods: Enamel bovine specimens were assigned to the following groups: G1-MPP (MI Paste Plus, 0.2% NaF, Recaldent™, GC Corporation Tokyo, Japan); G2-FD (Crest™ Cavity Protection, 0.243% NaF, Procter & Gamble, USA); G3-CLP (Clinpro™ 5000, 1.1% NaF, 3M ESPE, USA); and G4-CO (Control without fluoride, Silica-based dentifrice; Daudt Ltda, Brazil). The specimens were soaked in demineralizing solution for 6 h and remineralizing solution for 18 h alternatively for 10 days. The toothpaste was prepared with deionized water in a 1:3 ratio (w/v) for three minutes daily. The solutions were renewed every 48 h. After cycling, enamel changes were analysed by percentage change of SMH (%SMH) and energy-dispersive X-ray spectroscopy (EDS). The %SMH value observed for G3-CLP (2.9 ± 39.2) was higher than that found in G4-CO (-13.0 ± 20.7), G1-MPP (-8.9 ± 20.9) and G2-FD (-3.9 ± 27.1). The %SMH was similar for all treatment groups (one-way ANOVA and Tukey's HSD; p < .05). The pH, Ca 2+ and P total in the remineralization solutions were not different among all groups (Kruskal-Wallis; p < .05). At 24 h, the Ca 2+ concentration in the demineralization solution was significantly lower in G1-MPP. Ca 2+ concentration increased in all groups after 48 h, except for G3-CLP. The EDX quantitative analysis showed that the atomic % of elements is lower level at G4-CO., Conclusions: The Clinpro™ 5000 demonstrated having the most protective effect against demineralization; however, the % SMH was similar for all groups.

Since the classical epidemiological studies by Dean, it has been known that there should be an optimum level of exposure to fluoride that would be able to provide the maximum protection against caries, with minimum dental fluorosis. The "optimal" daily intake of fluoride for children (0.05-0.07 mg per kilogram bodyweight) that is still accepted worldwide was empirically determined. In the present review, we discuss the appropriateness of the current guidance for fluoride intake, in light of the windows of susceptibility to caries and fluorosis, the modern trends of fluoride intake from multiple sources, individual variations in fluoride metabolism, and recent epidemiological data. The main conclusion is that it is very difficult to think about a strict recommendation for an "optimal" range of fluoride intake at the individual level in light of existing knowledge of 1) the mechanisms of action of fluoride to control caries, 2) the mechanisms involved in dental fluorosis development, 3) the distinct factors that interfere in the metabolism of fluoride, and 4) the windows of susceptibility to both dental caries and fluorosis development. An "optimal" range of fluoride intake is, however, desirable at the population level to guide programs of community fluoridation, but further research is necessary to provide additional support for future decisions on guidance in this area. This list includes the effect of factors affecting fluoride metabolism, clinical trials on the effectiveness of low-fluoride dentifrices to prevent caries in the primary dentition, and validation of biomarkers of exposure to fluoride.

The purpose of this report is to examine critically the appropriateness of the current guidance for fluoride intake in the population (0.05-0.07 mg F/kg bodyweight/d), consider whether changes to the current guidance are desirable, and suggest further research that will strengthen the evidence base for future decisions on guidance/advice in this area. The benefits and the risks of using fluoride particularly concern preschool children because it is at this age that excessive fluoride intake may result in dental fluorosis. Data from mostly cross-sectional studies show a wide variation in exposure and a considerable variation in the amount of fluoride ingested. Fluorosis, mostly mild, is commonly observed. For considering changes in current guidance, there is a need for more knowledge on the relationship between exposure to fluoride at an early age and the development of fluorosis. For that, prospective epidemiological studies with sufficiently large and representative samples of children are required. It is also important to study children in communities both with and without water fluoridation and to include populations where salt or milk fluoridation is used. There is also a need for professional agreement on acceptable levels of mild and moderate/severe fluorosis and a more comprehensive knowledge on the appreciation of mild fluorosis among the public.

Besides the effects on the health of individuals, cigarette smoking can also interfere with the appearance of their teeth. To evaluate the effect of cigarette smoking-toothbrushing-cycling (CSTC) with whitening toothpastes on the roughness and optical behavior of bovine enamel for eight weeks. Thirty bovine dentin/enamel discs, 8.0 mm in diameter and 2.0 mm thick, were randomly divided into three groups according to the toothpastes: whitening (Colgate Luminous White - CW and Oral B 3D White - OW), and a non-whitening (Colgate - C). The roughness, color (CIE L*a*b* system), translucency and gloss were measured before and after the specimens were submitted to CSTC. The topography of the specimens was analyzed by scanning electron microscopy. During the first week, the specimens were daily subjected to the consumption of 20 cigarettes and brushed (40 strokes/100 g) with the toothpastes' slurries. Thereafter, the CSTC was weekly applied in an accumulated model (140 cigarettes/280 strokes) for seven weeks. The data were submitted to two-way ANOVA, Tukey's HSD test, and paired-t test (α=0.05). The three toothpastes produced significant changes in roughness, color, translucency and gloss (p<0.05). After eight weeks, the roughness and the gloss produced by the three toothpastes were similar (p>0.05), while OW produced the lowest color change and the translucency of C was lower than that of CW (p<0.05). The three toothpastes produced a significant decrease in L* values and a significant increase in a* values after eight weeks (p<0.05). No significant difference in the b* coordinate was found for OW (p=0.13) There were topographic changes in the enamel surfaces. The whitening toothpastes increased the roughness, changed the topography and were not able to maintain the optical stability of enamel exposed over eight weeks.

Maintaining healthy teeth and soft oral tissue is important. Clinical research has shown manual toothbrushes with end-rounded filaments to cause significantly less gingival abrasions than those without end-rounding. Tapered filaments are an alternative to end-rounded filaments, but there is no solid evidence for their effectiveness. For the interdental spaces that the toothbrush doesn't reach, the use of special oral hygiene aids is advised. Clinical studies of gingivitis patients have shown that both interdental brushes and plastic-rubber bristles reduce gingivitis after 4 weeks of use. Plastic-rubber bristles result in a significant improvement in the tendency for interdental bleeding compared with interdental brushes, and they cause less abrasion of the gingiva. Other factors that can potentially traumatise soft and hard oral tissue are oral piercings. Wearing oral piercings is not without risk. Tongue and lip piercings are associated with the risk of gingival recession, and tongue piercings are associated with tooth fractures. To prevent the risk of complications, patients should be discouraged from wearing oral piercings.

The study aimed to investigate the effects of bacterial biofilms on changes in the surface microhardness of enamel treated with casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) with and without fluoride. Human enamel blocks with incipient caries-like lesions were divided into four groups of 13: G1: Saliva (Control); G2: fluoride dentifrice (Crest™, 1100 ppm as NaF); G3: CPP-ACP (MI Paste; Recaldent™); and G4: CPP-ACPF (MI Paste Plus; Recaldent™ 900 ppm as NaF). The specimens were soaked in demineralizing solution for 6 h and remineralized in artificial saliva for 18 h alternately for 10 days. The dentifrice was prepared with deionized water in a 1 : 3 ratio (w/w) or applied undiluted in the case of the CPP-ACP group. The surface microhardness (SMH) was evaluated at baseline, after artificial caries, after pH cycling and treatment with dentifrices, and after incubation in media with Streptococcus mutans for biofilm formation. The biofilms were exposed once a day to 2% sucrose and the biofilm viability was measured by MTT reduction. The percentage of change in surface microhardness (%SMHC) was calculated for each block. The data were analyzed by nonparametric test comparisons (α = 0.05). The %SMHC values observed in G2 were different from those of G1, G3, and G4 (p < 0.05). After biofilm formation, %SMHC was positive in G2 and G4 when compared to G1 and G3, but resistance to demineralization after biofilm formation was similar in all groups. In conclusion, the presence of biofilms did not influence the treatment outcomes of anticaries products.

Aim: This study was undertaken to study the effect of three most commonly used dentifrices for denture cleaning and to find out any positive association between number of strokes by toothbrush and surface roughness of acrylic resin., Materials and Methods: About 40 acrylic rectangular blocks with high finish were polished until surface roughness was calculated as 0 pirn. The materials tested were distilled water as a control group, Patanjali Dant Kanti, Colgate Total Cream, and Cleansodent as dentifrices. Stroking was done with dentifrices, and surface roughness readings were recorded at 1,000, 9,000, and 18,000 strokes., Results: Patanjali Dant Kanti was found to have maximum abrasive potential followed by Colgate Total Cream, followed by Cleansodent and least by distilled water., Conclusion: Surface roughness was found to be directly proportional to the type, size, and fineness of the abrasive component used; the professional denture cleanser (Cleansodent) has less abrasive potential than the regular toothpastes. Among those tested, the highest abrasive potential was found in Patanjali Dant Kanti and lowest by Cleansodent (denture cleanser)., Clinical Significance: The patient as well as dentist should choose the dentifrice with utmost care noting the size and type of abrasives used. Washing the dentures normally with water without toothbrush daily and cleaning of dentures once in 10 to 15 days with a soft toothbrush and professional denture cleanser should be the line of maintenance of dentures.

Background: The focus in caries management has shifted to early detection of caries lesions and targeted noninvasive management of incipient lesions using novel remineralizing agents., Aim: This study aimed to compare and evaluate the remineralization potential of commercially available agents containing nano-hydroxyapatite (nano-HA), casein phosphopeptide-amorphous calcium phosphate fluoride (CPP-ACPF), and Tricalcium phosphate (TCP) on artificially induced white spot lesions in primary teeth., Settings and Design: This is an in vitro double-blind randomized study., Materials and Methods: Forty extracted or exfoliated primary teeth were selected and randomized as follows: Group I: FTCP, Group II: fluoridated dentifrice, Group III: CPP-ACPF, and Group IV: Nano-HA. DIAGNOdent readings and scanning electron microscope (SEM) energy dispersive X-ray (EDX) analysis were carried out at baseline. The samples were subjected to the test agents after inducing white spot lesions. The readings were repeated postdemineralization and postremineralization., Statistical Analysis: The data were analyzed using IBM SPSS version 20 software with one-way ANOVA, post hoc Tukey's HSD, and paired t-test., Results: SEM evaluation showed favorable surface changes in all the four study groups after remineralization therapy. Intragroup comparison of DIAGNOdent and EDX readings showed a highly significant difference between baseline, postdemineralization, and postremineralization values. However, the intergroup comparison was statistically nonsignificant., Conclusion: All test agents were comparable in their remineralization potential.

Objectives: The present study evaluated fluoride (F) and calcium (Ca) concentrations in the biofilm fluid formed in situ under cariogenic challenge after using F dentifrices supplemented or not with sodium trimetaphosphate (TMP) or calcium glycerophosphate (CaGP)., Methods: Volunteers (n = 12) were randomly divided into 5 groups according to the toothpastes used: placebo (without F, CaGP or TMP), 1100 ppm F (1100F) and low-fluoride dentifrice (LFD, 550 ppm F) with no supplementation (550F) or supplemented with 1 % TMP (550F-TMP) or 0.25 % CaGP (550F-CaGP). In each phase, volunteers wore palatal appliances containing 4 bovine enamel blocks. Cariogenic challenge was performed with 30 % sucrose solution, 6 times/day. On the morning of the eigth day, biofilm samples were collected 12 h and 1 h after brushing and cariogenic challenge. F and Ca analyses in the biofilm fluid were performed with the inverted electrode after buffering with TISAB III and using the Arsenazo III method, respectively. Data were submitted to two-way ANOVA (repeated measures) and Student-Newman-Keuls test (p < 0.05)., Results: A dose-response relationship was verified between F concentrations in the dentifrices and in the biofilm fluid. Significant differences were observed among placebo, 550F, and 1100F only 1 h after brushing, without statistical differences among 550F, 550F-TMP, and 550F-CaGP. No defined trend was observed among the groups regarding Ca concentrations, with the highest values seen for placebo and 550F-CaGP., Conclusion: The anticaries effect of LFDs supplemented with CaGP or TMP cannot be related to an increased availability of F and Ca in the biofilm fluid., Clinical Significance: The better performance of LFDs containing CaGP or TMP shown in previous studies should be attributed to their ability to interact with tooth enamel and with the biofilm, rather to their effect on the biofilm fluid.

Objectives: The aim of these investigations was to assess the ability of two fluoride dentifrices to protect against the initiation and progression of dental erosion using a predictive in vitro erosion cycling model and a human in situ erosion prevention clinical trial for verification of effectiveness., Materials and Methods: A stabilized stannous fluoride (SnF 2 ) dentifrice (0.454 % SnF 2  + 0.077 % sodium fluoride [NaF]; total F = 1450 ppm F) [dentifrice A] and a sodium monofluorophosphate [SMFP]/arginine dentifrice (1.1 % SMFP + 1.5 % arginine; total F = 1450 ppm F) [dentifrice B] were tested in a 5-day in vitro erosion cycling model and a 10-day randomized, controlled, double-blind, two-treatment, four-period crossover in situ clinical trial. In each study, human enamel specimens were exposed to repetitive product treatments using a standardized dilution of test products followed by erosive acid challenges in a systematic fashion., Results: Both studies demonstrated statistically significant differences between the two products, with dentifrice A providing significantly better enamel protection in each study. In vitro, dentifrice A provided a 75.8 % benefit over dentifrice B (p < 0.05, ANOVA), while after 10 days in the in situ model, dentifrice A provided 93.9 % greater protection versus dentifrice B (p < 0.0001, general linear mixed model)., Conclusion: These results support the superiority of stabilized SnF 2 dentifrices for protecting human teeth against the initiation and progression of dental erosion., Clinical Relevance: Stabilized SnF 2 dentifrices may provide more significant benefits to consumers than conventional fluoride dentifrices.

Objectives: To compare the abrasive wear on human dentine in an in situ model associated with use of an experimental low abrasivity anti-sensitivity dentifrice containing 1% alumina and 5% sodium tripolyphosphate (STP) with an experimental ultra-low abrasivity non-alumina 5% STP dentifrice, a higher abrasivity daily-use whitening dentifrice, and water as controls., Methods: This was a single-centre, single-blind, randomised, split-mouth, four-treatment, two-period, crossover in situ study in 29 healthy subjects. Subjects wore bilateral lower buccal appliances, each fitted with four dentine specimens. Study treatments were applied ex vivo (three times daily). Dentine loss was measured by non-contact profilometry after 5, 10 and 15days' treatment., Results: All 29 subjects were included in the efficacy analysis. Significantly less dentine loss was associated with brushing with the low and ultra-low abrasivity dentifrices than with the higher abrasivity dentifrice at all timepoints (p<0.01). Brushing with ultra-low abrasivity dentifrice or water resulted in statistically significantly less dentine loss compared with brushing with the low abrasivity dentifrice at all timepoints (p<0.05). Dentine loss after brushing with ultra-low abrasivity dentifrice was not significantly different from brushing with water., Conclusions: The degree of dentine loss observed in this in situ model reflected the abrasivity of the study dentifrices. Brushing with low or ultra-low abrasivity STP-containing anti-sensitivity dentifrices resulted in significantly less dentine loss (equating to dentine wear) than with a higher abrasivity daily-use whitening dentifrice., (Copyright © 2016 Elsevier Ltd. Objectives: Child dentifrices vary in their composition, with possible differential impacts on cells in the oral soft tissue. While cytotoxicity studies have been performed on adult dentifrices, no respective studies have thus far been reported on child dentifrices., Material and Methods: Seventeen commercial dentifrices for children up to 12 years of age were evaluated with respect to their in vitro cytotoxicity on gingival fibroblasts, oral squamous cell carcinoma HSC-2 cells, and L929 mouse fibroblasts. Proliferation was analyzed and live-dead staining was performed., Results: Ten child dentifrices greatly reduced cell viability with LC50 values below 5 %. Four dentifrices showed a moderate cytotoxicity with LC50 values between 5 and 20 %. Three child dentifrices showed almost no cytotoxicity with LC50 values above 95 %. The results of the assays for proliferation and live-dead staining supported these findings., Conclusions: The different composition of the child dentifrices translated into a broad spectrum of in vitro cytotoxicity on cells of the oral cavity., Clinical Relevance: The in vitro data provide the scientific foundation for further in vivo research testing the clinical relevance of the present findings., Competing Interests: Compliance with Ethical Standards This article does not contain any studies with human participants or animals performed by any of the authors. The aim of this study was to compare the caries-preventive effect of a stabilized stannous fluoride/sodium fluoride dentifrice containing sodium hexametaphosphate with those of a regular, solely sodium fluoride-containing and amine fluoride-containing dentifrice on pre-demineralized bovine enamel specimens using a pH-cycling model. Bovine enamel specimens with two artificial lesions each were prepared. Baseline mineral loss of both lesions was analyzed using transversal microradiography (TMR). Eighty-five specimens with a mean (SD) baseline mineral loss of 3393 (683) vol% × µm were selected and randomly allocated to five groups (n = 13/15). Treatments during pH-cycling (28 days and 2 × 20 min demineralization/day) were: brushing twice daily with slurries of AmF (1400 ppm F - ), NaF (1450 ppm F - ), SnF 2 /NaF (1100 ppm F - /350 ppm F - ), and fluoride-free (FF) dentifrices or they were immersed in distilled water and remained unbrushed (NB). Subsequently, from each specimen one lesion was covered with acid-resistant varnish, while the remaining lesion was demineralized for another 14 days. Differences in integrated mineral loss (∆∆Z) were calculated between values before and after pH-cycling (∆∆Z E1 ) as well as before pH-cycling and after second demineralization (∆∆Z E2 ) using TMR. Treatments AmF and NaF induced a significantly higher mineral gain (∆∆Z E1 /∆∆Z E2 ) compared to treatments FF and NB (p < 0.05; ANOVA test). Except for treatments AmF and NaF no significant differences in mineral loss between before and after pH-cycling could be observed (p < 0.05; t test) [∆∆Z E1 : AmF:1563 (767); NaF:1222 (1246); SnF 2 /NaF:258 (1259); FF:-52 (1223); NB:-151 (834)]. Both dentifrices with either AmF or NaF promoted remineralization, whereas SnF 2 /NaF dentifrice did not promote remineralization in a biofilm-free pH-cycling model.

Objective: The daily removal of supragingival dental plaque is a key factor in the prevention of gingivitis. The aim of the study was to compare the gingival health benefits of a triclosan/copolymer/fluoride toothpaste (Colgate Total, a fluoride toothpaste containing an antiseptic) to a commercially available toothpaste containing 0.243% sodium fluoride in a silica base (Colgate Herbal, a conventional fluoride toothpaste with herbal extracts)., Method and Materials: A total of 50 patients with gingivitis and at least one sensitive tooth were included. The subjects were randomly stratified into two groups: Colgate Total toothpaste, and Colgate Herbal toothpaste. After a 4-week pre-experimental phase, baseline Plaque Index (Quigley-Hein Index) (PI), Gingival Index (GI), Gingival Bleeding Index (GBI), and Visual Analog Scale (VAS) were assessed. The PI, GI, GBI, and VAS were reexamined at weeks 4, 12, and 24 after the baseline., Results: Fifty subjects complied with the protocol and completed the study. The conventional fluoride toothpaste with herbal extracts group and the fluoride toothpaste containing an antiseptic group exhibited significant reductions in PI, GI, GBI, and VAS over time. The amount of reduction after 6 months of the treatment was higher in the Total group compared to Herbal group (1.82 vs 1.39, P = .015 for PI; 0.67 vs 0.37, P < .005 for GI; and 56.64% vs 34.26%, P < .005 for GBI). No significant difference was seen for VAS., Conclusion: Twice daily brushing with a toothpaste containing 0.3% triclosan and polyvinyl methyl ether and maleic acid copolymer provides a more effective level of plaque control and gingival health with no effect on decreasing dentin hypersensitivity compared to conventional fluoride toothpaste. Toothpastes containing triclosan/copolymer, in addition to fluoride, result in a higher reduction in plaque, gingival inflammation, and gingival bleeding when compared with fluoride toothpastes without triclosan/copolymer.

Objectives: This study aimed to investigate whether addition of an octadecene/maleic anhydride copolymer (O/MA) to a potassium nitrate (KNO 3 ) dentifrice could facilitate delivery of potassium to dentine and enhance its efficacy in dentine hypersensitivity relief., Methods: This was a randomised, examiner-blind, controlled, parallel group study in 139 healthy subjects with at ≥2 sensitive teeth. Assessment of dentine hypersensitivity to tactile (Yeaple probe) and evaporative (air) stimuli (Schiff Sensitivity Scale, visual analogue scale [VAS]) was carried out at baseline and after 1, 2, 4 and 8 weeks twice daily treatment with an experimental 5% KNO 3 /3% O/MA dentifrice, a comparator 5% KNO 3 dentifrice (active comparator), a 0% KNO 3 /3% O/MA dentifrice (placebo) and a regular fluoride dentifrice (negative control). This study was not powered to detect statistically significant differences between treatments., Results: Across the treatment period an improvement in sensitivity to evaporative air stimulus was observed for all products and to a tactile stimulus for the potassium-containing treatments, with the greatest reductions for the experimental dentifrice (5% KNO 3 /3% O/MA). Reductions in sensitivity observed for the potassium-containing dentifrices compared to the placebo and negative control dentifrices were statistically significantly for Schiff sensitivity score and tactile threshold at all time-points and for VAS at Weeks 4 and 8. Trends in the study data also favoured the experimental dentifrice, compared to the active comparator dentifrice, for all clinical measures. Study treatments were generally well tolerated., Conclusion: This study provides initial clinical evidence to suggest that addition of a polymer excipient may enhance the anti-sensitivity efficacy of potassium-containing dentifrices., Clinical Significance: Daily use potassium-containing dentifrices are established as efficacious for the relief of dentine hypersensitivity. Inclusion of a polymer excipient in such formulations may facilitate delivery of potassium to the dentine surface and so enhance clinical efficacy. Context: In India, teeth cleaning with tooth powder is common in rural and semi-urban areas. These dentifrices may contain low-quality abrasives, which may have a deleterious effect on dental hard tissues., Aims: This study aims to evaluate the tooth substance loss caused by different dentifrices and to correlate it with chemical composition, size, and shape of abrasives used., Settings and Design: An indigenously made automated machine was used for brushing the specimens., Materials and Methods: Sixty-four freshly extracted premolars were allocated to eight groups (n = 8). Colgate toothpaste was used as the control group. Each specimen was brushed in a vertical motion for 2½ h at 200 strokes/min with a constant applied load of 200 g corresponding to 6-month brushing. The difference in weight (pre- and post-brushing) was determined by an analytical weighing machine. Chemical analysis was done to determine the presence of iron oxide by Inductively Coupled Plasma Mass Spectrometry method. Shape and size of the abrasive particles was evaluated under scanning electron microscopy (SEM)., Statistical Analysis Used: One-way analysis of variance and Paired t-test were used to analyze the data., Results: Tooth substance loss was maximum in the group brushed with red tooth powder, which was shown to contain the highest amount of iron oxide and also exhibited large, irregularly shaped abrasive particles under SEM., Conclusions: Tooth substance loss was documented to be correlated with chemical composition (iron oxide) and the size and shape of abrasive particles used in dentifrices.

Purpose: This study evaluated fluoride (F) concentrations in saliva of children after brushing with dentifrices containing different F concentrations (zero, 550, and 1,100 ppm F) in different quantities (full bristles, transversal technique, and pea-sized)., Methods: Eight- to 10-year-old volunteers (N=24) were randomly assigned into nine experimental groups (dentifrice type versus amounts) following a double-blind, crossover protocol. After a one-week washout period (use of placebo toothpaste), stimulated saliva was collected prior to toothbrushing (baseline) and at five, 15, 30, 60, and 120 minutes after toothbrushing with one of the possible combinations. Centrifuged saliva was used for F analysis after buffering with TISAB III. Data were submitted to repeated-measures ANOVA followed by Student-Newman-Keuls' test (P<0.05)., Results: Salivary F peaked at five minutes after brushing, decreasing exponentially afterward. A clear dose-response relationship was seen between F concentration/amount of dentifrice applied and the mean area under the curve (AUC) of salivary F concentrations (P<0.001). The low-fluoride toothpaste applied using transversal technique or full bristles led to a significantly higher AUC than the conventional toothpaste using a pea-sized amount., Conclusions: Brushing with a low-fluoride toothpaste applied using the transversal technique delivers more fluoride to saliva compared to a conventional toothpaste in a pea-sized amount.

The objective of this study was to evaluate the effects of diode laser and a desensitizing dentifrice on dentin permeability. Fifty-two root dentin fragments were obtained (5 × 5mm) and treated with 24% EDTA gel. The samples were divided into 4 groups (n = 13): G1, control (no treatment); G2, diode laser (λ = 908 nm, 1.5 W, continuous mode, 20s); G3, application of abrasive dentifrice for 1 minute (Elmex Sensitive Professional (International Gaba); and G4, application of abrasive dentifrice for 1 minute followed by irradiation with diode laser. Ten samples per group were immersed in 2% methylene blue solution for 4h. The specimens were washed, longitudinally sectioned, observed under optical microscopy, photographed and assessed based on the degree of dye leakage. The remaining samples were observed under scanning electron microscopy (SEM). The leakage data were subjected to ANOVA test, followed by Tukey's t-test (α = 5%). Groups 2, 3 and 4 showed less dye penetration than the control group (p < 0.05), but were similar among each other. SEM images showed that dentinal tubules were open in G1, and fused and occluded in G2. Group 3 showed dentinal tubules that were occluded by the metal ions from the toothpaste. G4 presented similar characteristics to G3, and the presence of fused dentin. The diode laser and the dentifrice were effective in reducing dentinal permeability, and the combination of the two treatments did not show better results than either one used alone.

Aim: To evaluate the effect of dentifrice with varying concentrations of fluoride on surface microhardness of fluorosed enamel as compared to a non-fluoridated dentifrice., Study Design: In vitro, comparative type., Method: 60 enamel blocks with score-3 enamel fluorosis were randomly divided into three groups: Group I non-fluoridated dentifrice, Group II 1000-ppm fluoridated dentifrice and Group III 1400-ppm fluoridated dentifrice. After evaluating the baseline surface microhardness, samples from each group were brushed for 2 min, twice daily for a period of 21 days with 0.02 g of respective interventional agents as per protocol. Microhardness of the samples was re-evaluated at the end of 21 days. Statistical analysis was performed using paired t test, ANOVA and Kruskal-Wallis test., Results: The surface microhardness within the group from the baseline to 21st day post intervention, showed a statistically significant increase in all three groups, hence indicating that all the three interventional agents used were effective against the fluorosed enamel surface. Intergroup comparison of the surface microhardness of fluorosed enamel from baseline to 21st day post intervention showed a statistically significant difference (p < 0.001) amongst the groups with Group III proving most effective followed by Group I and Group II., Conclusion: 1400-ppm fluoridated dentifrice is more effective as compared to 1000-ppm fluoridated dentifrice and non-fluoridated dentifrice in increasing the surface microhardness of fluorosed enamel.

Introduction: Acidic dentifrices have been shown to be more effective than neutral ones against dental caries using in vitro, in situ, and clinical protocols., Objectives: Given the scarcity of studies assessing intraoral fluoride (F) retention after using such formulations, the present study evaluated the influence of pH and F concentration of dentifrices on F uptake by saliva, biofilm, and biofilm fluid., Methods: Volunteers (n = 22) were randomly assigned to dentifrices containing 0 (placebo), 550 (LFD, low-fluoride dentifrice), and 1100 ppm F (CD, conventional dentifrice) at pH 4.5 and 7.0 and brushed their teeth 3 times/day following a double-blind, crossover protocol. Saliva and biofilm samples were collected after 7 days of using the dentifrices, 1 and approximately 12 h after last brushing. F and calcium (Ca) analyses were performed with the inverted electrode after buffering with TISAB III and the Arsenazo III method, respectively. Data were analyzed by 3-way ANOVA, Tukey's HSD test, and Pearson's correlation coefficient (p < 0.05)., Results: F concentrations in biofilm fluid and whole biofilm 1 h after brushing with acidic F-toothpastes were higher than those related to neutral counterparts, although the differences were small and not significant; no increases were observed in salivary F concentrations influenced by dentifrice pH. Moreover, no definite trend was observed for Ca concentrations in these compartments., Conclusion: Dentifrice pH had some influence on F uptake by the biofilm fluid, having lesser or no impact on F uptake by the biofilm and saliva, respectively., Clinical Significance: Toothbrushing with acidic toothpastes leads to slight increases in F concentrations in the biofilm fluid when compared to neutral formulations, which may contribute to the higher anticaries effect of acidic formulations.

Objective: To study the influence of particle size and morphology on zinc cation adsorption by hydroxyapatite (HA) and dentifrice containing HA., Methods: Four HAs with different particle sizes and morphologies, HA-containing dentifrice and blank dentifrice were prepared into suspensions of serial concentrations. Zinc ion solutions with an initial concentration of 10 mg/L was mixed with the suspensions and kept for 24 h for adsorption reaction. The zinc ion concentration in the supernatant was measured by inductively coupled plasma emission spectrometer and the sorption rate of zinc ion was calculated., Results: HA and HA-containing dentifrice with various particle sizes and morphologies were all capable of absorbing zinc ions from simulated waste water, and the adsorption rate of HA-containing dentifrice was 3%-10% higher than that of HA. HA with a particle size of 12 µm and a spherical morphology showed the strongest adsorption ability, followed by short bar-shaped HA with a particle size of 30 µm. Both Langmuir and Freundlich equation could simulate the sorption processes of HA dentifrice, while only Langmuir equation could simulate the sorption processes of HA., Conclusion: Incorporation of HA in dentifrice can enhance zinc ion adsorption capacity of the material. The particle size and morphology of HA both affect the adsorption of zinc ions, and 12-µm HA particle with a spherical morphology has the best adsorption ability.

Low-fluoride (F) dentifrices (<600 µg F/g) are widely available worldwide, but evidence to recommend the use of such dentifrices, with either regular or improved formulations, is still lacking. Therefore, the aim of this study was to evaluate the anticaries potential of low-F dentifrices found in the Brazilian market, using a validated and tested pH-cycling model. Enamel blocks were selected by surface hardness (SH) and randomized into four treatment groups (n=12): non-F dentifrice (negative control), low-F dentifrice (500 μg F/g), low-F acidulated dentifrice (550 μg F/g) and 1,100 μg F/g dentifrice (positive control). The blocks were subjected to pH-cycling regimen for 8 days and were treated 2x/day with dentifrice slurries prepared in water (1:3, w/v). The pH of the slurries was checked, and only the acidulated one had low pH. After the pH cycling, SH was again determined and the percentage of surface hardness loss was calculated as indicator of demineralization. Loosely- and firmly-bound F concentrations in enamel were also determined. The 1,100 μg F/g dentifrice was more effective than the low-F ones to reduce enamel demineralization and was the only one that differed from the non-F (p<0.05). All F dentifrices formed higher concentration of loosely-bound F on enamel than the non-F (p<0.05), but the 1,100 μg F/g was the only one that differed from the non-F in the ability to form firmly-bound F. The findings suggest that the low-F dentifrices available in the Brazilian market, irrespective of their formulation, do not have anticaries potential.
