Background Heat polymerized acrylic (HPA) resins are known to have high porosity that contributes to increased surface roughness and microcrack formation in stress areas. This facilitates the attachment and growth of polymicrobial biofilms contributing to increased antimicrobial resistance. Many research had been carried out on avocado seeds, but no research that studies the effect of avocado seeds on denture-plaque microorganism biofilm on HPA resin has been found. Methods This study used 144 samples (n=144), namely HPA resin discs covered with mono-species and polymicrobial biofilms consisting of Candida albicans, Candida glabrata, Actinomyces odontolyticus, Streptococcus gordonii, and Staphylococcus aureus. The discs were soaked for 8 hours in the 5%, 10%, 15%, 20% avocado seed extract, positive control (alkaline peroxide), and negative control (aquadest). Each disc was shaken with a vortex mixer for 1 minute, and 100 μL was added into 96-well microplates with three times repetition and incubated for 24 hours. The inhibition values were determined from the percentage inhibition value formula which required absorption values from a microplate reader (595 nm). Results In this research, it was found that the MBIC 50 of avocado seed extract against the mono-species of C. albicans (5%), C. glabrata (5%), A. odontolyticus (15%), S. gordonii (15%), S. aureus (10%), while against the biofilm was 20%. There was a significant effect of soaking HPA resin in avocado seed extract of 5%, 10%, 15%, 20% on the inhibition of mono-species and polymicrobial biofilms of denture-plaque microorganisms with a value of p Conclusion The MBIC 50 of avocado seed extract in polymicrobial biofilm group was higher than that in the mono-species biofilm groups. Although alkaline peroxide showed higher inhibition value than that of the MBIC 50 in polymicrobial biofilm group, 20% avocado seed extract was effective in inhibiting polymicrobial biofilm because it was able to inhibit more than 50% polymicrobial biofilm.

Denture plaque, a biofilm that develops on denture surfaces, could contribute to many oral and systemic afflictions. Hence, a quantitative assessment of denture plaque is important to evaluate the denture hygiene of denture wearers, particularly to prevent plaque biofilm-associated diseases. The aim of this systematic review, therefore, was to review and summarize the visual denture hygiene assessment methods using denture plaque indices and with planimetries published in the literature. English language studies published up to March 2022 in four electronic databases, PubMed, Medline, Embase, and Cochrane Library, were searched, followed by a manual search of Google Scholar by two assessors. The review followed Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) whenever possible. Details of the visual assessment methods, including the types of denture assessed, its materials and its surfaces, as well as the use of a disclosing agent, were the main outcomes. Of 492 screened studies, 74 were included per the inclusion and exclusion criteria. Of these, 60 studies utilized various denture plaque indices while 18 used planimetries. 43 out of 60 studies with indices and 17 out of 18 studies with planimetries used disclosing agents for visual evaluation of plaque. A total of 21 indices were described in the included studies, of which seven graded a divided denture surface, while the remainder graded entire denture surface. Of the 18 planimetric assessments, one study quantified squares of the disclosed plaques on denture images, 16 studies quantified such pixels with computer programs, and a single study quantified points, pixels, and contour of plaque areas. In summary, denture plaque indices appear to be popular in denture plaque assessment due to their simplicity. Computerized planimetric assessment, though more time-consuming, provides a more accurate assessment of plaque load as it is less prone to subjectivity and assessor errors.

Introduction Assessment of the denture plaque can provide a valuable report regarding the oral health of geriatric patients and the oral hygiene habits in this population group. Focusing on the current research gap on this topic in Romania, our research aims to highlight the microbial plaque on the different surfaces and types of removable poly(methyl methacrylate) (PMMA) prostheses to qualitatively evaluate the microbial species, and to assess the combined effect of mechanical and chemical cleaning of dentures on the prosthetic microbiome. Methods The prosthetic plaque from four denture wearers was identified with a UV activated fluorescent revealer and digitally photographed. Swab samples according to a certain hygiene protocol were cultured on three different growth media such as blood agar (BA), bromothymol blue lactose agar (AABTL) and Sabouraud dextrose agar (SDA). Results Denture plaque was variably distributed on the surface of the prostheses. Regardless of the denture type (conventional complete denture, tooth or implant supported overdenture, removable partial denture) the microbial plaque was identified at the retentive areas, especially at interdental spaces. The main plaque deposition areas were the vestibular incline of the labial flange of the maxillary denture and the lingual incline of the posterior lingual flange of the mandibular denture. The prosthetic microbiome consisted of pathogenic Gram-positive aerobes (Streptococcus spp. and coagulase-negative Staphylococcus spp.), Gram-positive anaerobes (Actinomyces spp. and Klebsiella spp.) as well as subspecies of fungi of the genus Candida. Conclusions Despite its poor variety, the denture microbiome in the investigated cases hosts colonies with high pathogenic potential. Some areas of the prostheses are more likely to accumulate dental plaque. Dentists should insist through more regular checks of the patients and their caregivers on ensuring the biocontrol of the dentures, especially in frail geriatric patients exposed to greater risks related to general health. Purpose: We aimed to evaluate the effectiveness of photoreactive 2-methacryloyloxyethyl phosphorylcholine (MPC) in inhibiting Candida albicans biofilm formation on polymethyl methacrylate (PMMA) and assess its mechanism and need for re-application by evaluating its interaction with salivary mucin and durability during temperature changes. Methods: PMMA discs were used as specimens. The MPC coating was applied using the spray and cure technique for the treatment groups, whereas no coating was applied to the control. The MPC treatment (MT) groups were further differentiated based on the number of thermal cycles involved (0, 1000, 2500, and 5000). The optical density was measured to assess mucin adsorption (MA). Contact angle (CA) was calculated to evaluate surface hydrophilicity. The presence of MPC components on the PMMA surface was assessed using X-ray photoelectron spectroscopy (XPS). C. albicans biofilms were evaluated qualitatively (scanning electron microscope images) and quantitatively (colony-forming units (CFUs)). Statistical analysis was conducted using two-way analysis of variance and Tukey's multiple comparison test. Results: MA rate and CA increased significantly in the MT groups, which exhibited significantly fewer CFUs and thinner biofilms than those of the control group. Based on the XPS, MA, and CFU evaluations, the durability and efficacy of the MPC coating were considered stable up to 2500 thermal cycles. Additionally, a significant interaction was observed between mucin concentration and MPC efficacy. Conclusions: The photoreactive MPC coating, which was resistant to temperature changes for approximately 3 months, effectively prevented C. albicans biofilm formation by modifying surface hydrophilicity and increasing mucin adsorption. We investigated whether irradiation with 405-nm blue LED light could inhibit the growth of not only single- but dual-species biofilms formed by Candida albicans and Streptococcus mutans on denture base resin and cause the alteration in gene expression related to adhesion and biofilm formation. C. albicans and S. mutans single-/dual-species biofilms were formed on the denture base specimens. The biofilms were irradiated with 405-nm blue LED light (power density output: 280 mW/cm2) for 0 (control) and 40 min. Dual-species biofilms were analyzed using CFU assay and fluorescence microscopy, and single-/dual-species biofilms were analyzed using alamarBlue assays and gene expression analysis. To assess the inhibitory effect of irradiation on dual-species biofilms, specimens after irradiation were aerobically incubated for 12 h. After incubation, the inhibition of growth was assessed using CFU assays and fluorescence microscopy. Data were analyzed using the Mann–Whitney U or Student's t test (p < 0.05). Irradiation produced a significant inhibitory effect on biofilms. Fluorescence microscopy revealed that almost all C. albicans and S. mutans cells were killed by irradiation, and there was no notable difference in biofilm thickness immediately after irradiation and after irradiation and incubation for 12 h. alamarBlue assays indicated the growth of the biofilms was inhibited for 12–13 h. The expression of genes associated with adhesion and biofilm formation—als1 in C. albicans and ftf, gtfC, and gtfB in S. mutans—significantly reduced by irradiation. This study investigated: (1) the microbicidal effect of 405-nm blue LED light irradiation on biofilm formed by Candida albicans hyphae and Streptococcus mutans under dual-species condition on denture base resin, (2) the generation of intracellular reactive oxygen species (ROS) induced by irradiation, and (3) the existence of intracellular porphyrins, which act as a photosensitizer. Denture base resin specimens were prepared and C. albicans and S. mutans dual-species biofilms were allowed to form on the specimens. The biofilms were irradiated with 405-nm blue LED light and analyzed using the colony-forming unit assay, fluorescence microscopy, and scanning electron microscopy (SEM). Single-species biofilms of C. albicans and S. mutans formed on the specimens were irradiated with 405-nm blue LED light. After the irradiation, the intracellular ROS levels in C. albicans and S. mutans cells were measured. In addition, the level of intracellular porphyrins in C. albicans and S. mutans were measured. Irradiation for more than 30 min significantly inhibited the colony formation ability of C. albicans and S. mutans. Fluorescence microscopy revealed that almost all C. albicans and S. mutans cells were killed by irradiation. SEM images showed various cell damage patterns. Irradiation led to the generation of intracellular ROS and porphyrins were present in both C. albicans and S. mutans cells. In conclusion, irradiation with 405-nm blue light-emitting diode light for 40 min effectively disinfect C. albicans hyphae and S. mutans dual-species biofilms and possibly react with intracellular porphyrins resulting in generation of ROS in each microorganism. BACKGROUND: The adsorption of salivary pellicle proteins onto the material surface is key for denture plaque formation. OBJECTIVE: We aimed to investigate the adsorption of bovine serum albumin (BSA) and mucin (MCN) onto denture base metal materials using a 27-MHz quartz crystal microbalance (QCM) method. METHODS: A gold (Au), titanium (Ti), and cobalt chromium alloy (Co–Cr) sensors were employed for QCM measurements. Adsorbed amounts of BSA or MCN were calculated by observing the frequency decrease, and the apparent reaction rate, kobs, was obtained by the curve fitting of the frequency shift against the adsorption time. RESULTS: The adsorbed amounts of BSA on Ti were significantly lower than those on Au and Co–Cr. For MCN adsorption, Au showed significantly greater amounts of adsorption than Co–Cr. The kobs of Ti for BSA adsorption was significantly smaller than for the Co–Cr. The kobs of Ti, and Co–Cr for MCN adsorption were significantly smaller than for the Au. A clear correlation was not determined between adsorbed amounts of BSA or MCN onto each sensor and the surface topography or contact angles. The opportunistic fungus Candida albicans is a component of denture plaque and is associated with denture-related stomatitis. Inter-kingdom interactions between C. albicans and bacteria exist in such multi-species biofilms, which may affect the microbial composition of the plaque. This study was performed to investigate the bacterial composition of denture plaques, and the correlation between the relative abundance of these bacteria and C. albicans. Thirty denture plaque and 16 dental plaque samples were collected from 18 denture wearers (mean age, 80.3 years). After DNA extraction, a meta 16S rDNA amplicon library was constructed using PCR primers targeting the V3–V4 hypervariable region of bacteria. The amplicon was evaluated by high-throughput sequencing, followed by bacterial population analysis. The concentrations of both C. albicans DNA and total bacterial DNA were determined by real-time PCR. The correlation between the relative abundance of each bacterial genus and C. albicans was analyzed through Spearman's rank correlation. The genera Streptococcus , Lactobacillus , Rothia , and Corynebacterium were found to be more abundant in dentures than in dental plaques. The predominant bacteria in healthcare-associated pneumonia also inhabited denture surfaces. C. albicans was positively correlated with three acidogenic bacteria and negatively correlated with Leptotrichia and pathogens associated with periodontitis and endocarditis. Dentures may be significant reservoirs of pathogens causing aspiration pneumonia. Bacteria showing negative correlation with C. albicans , such as Leptotrichia, may be useful for controlling the growth of C. albicans in antifungal therapies. • We determined the microbial composition in denture and dental plaque. • Pathogens causing aspiration pneumonia were abundant on denture surfaces. • Bacterial genera positively or negatively correlated C. albicans were identified. Aim: A comparative study for plaque removing efficacy between toothpaste, liquid handwashing soap, and two chemical-soak denture cleansers (clinsodent denture cleansing powder and fittydent denture cleansing tablet). Materials and Methods: A total of 140 healthy complete denture patients were selected and checked for denture cleansing using toothpaste, liquid handwashing soap, and two chemical denture cleansers: Clinsodent powder and fittydent tablets. Cleansing with plain water was kept as control for twenty patients. Results: The data were analyzed using Wilcoxon signed ranks test for checking the efficacy. The Kruskal–Wallis test was used for comparing the plaque removing efficacies of each denture cleanser used in the study. All four denture cleansers were significantly effective in removing plaque when compared with plain water, but there was no significant difference among them. Conclusions: There was no significant difference among action of four denture cleansers used in this study. Thus we can infer that patients can use liquid handwashing soap or commercial products to overcome disadvantages of toothpastes. Recommendation from the results of this study is the use of liquid handwashing soap for cleansing after every meal and soaking of the denture in commercial denture cleanser during the night.

The prolonged accumulated plaque, may lead to microorganism multiplication, thus causing pathological changes in oral cavity. Dental plaque consists of 20%-30% intercelullar matrices and 70% microorganisms. Thymoquinone denotes bio-active agent extracted from plants that has antibacterial and antifungal activity. This study aimed to determine the effective concentration of thymoquinone to detached plaque from acrylic plate. This was an analytical laboratory study involving denture users as sample. The accumulated denture plaque was replicated in BHIB solution. Acrylic plates were immersed in BHIB, and divided into groups for hydrolysis assessment; control group, then were macerated in distilled water; as for treatment group, the plates were macerated in thymoquinone in varying concentration for a range of time periods. Optical density of hydrolysis result analysis was done at a wavelength of 600nm. Result. The lowest hydrolysis rate recorded was control group, with mean of 31.32%. Thymoquinone at 50% concentration, with 10 minutes maceration periods showed the highest hydrolysis rate (74.42%). Denture plaque is a biofilm composed of various microorganisms aggregated with saliva. Various denture cleansers and cleaning apparatuses have been developed and studied. However, the optimum water temperature for denture cleaning is unknown. Therefore, the present study investigated the effects of water temperature during ultrasonic denture cleaning. In vitro, resin disks with artificial Candida albicans biofilm were pressed onto Candida GE media after ultrasonic cleaning with water at different temperatures for 5 min. The media were subsequently cultured at 37°C for 24 h. The colonies formed were observed and colony areas were quantified using ImageJ software (US National Institutes of Health, Bethesda, MD, USA). In situ, the bacterial count and degree of cleanliness on the tissue surface of maxillary dentures were measured before and after ultrasonic cleaning with water at different temperatures for 5 min. Changes in bacterial counts and cleanliness were calculated for each temperature. The ratio of the area occupied by bacterial colonies in vitro and reduction rates in situ after cleaning with warm water were markedly less than those observed after cleaning with cold water. Background: Accumulation of denture plaque can lead to pathological changes in oral mucosa, such as denture stomatitis, halitosis, and caries. Plaque matrix is mostly formed by protein (30%) and polysaccharide complexes. Thus, an alternative enzyme solution as denture cleanser is required for hydrolysis of denture plaque. Papain is a proteolytic enzyme hydrolyzing proteins, while β-1,3-glucanase is a hydrolase enzyme hydrolyzing polysaccharides. Purpose: This study aimed to analyze the differences of effectiveness of ß-1,3- glucanase Vigna unguiculata enzyme and papain Carica papaya enzyme in hydrolysis of denture plaque. Method: This research was a laboratory experimental research with post test only control group design. After using denture for 24 hours, the denture was soaked in a solution of 100 ml PBS, papain enzyme, and β 1-3 glucanase enzyme at a concentration of 0.5 mg/ml, 1 mg/ml, and 2 mg/ml for 10 minutes. The solution from plaque hydrolysis was soaked in PBS and vortex enzyme for 2 minutes, then soaked in ice water for 15 minutes, and centrifuged at 3000 rpm 5-10º for 10 minutes. The supernatant was separated and analyzed. Turbidity readings then were performed in spectrofotometer with a wavelength of 480 nm. Result: 2 mg/ml of ß-1,3 glucanase enzyme generated the highest values of hydrolysis with a mean percentage of 68.77% compared to papain enzyme (44.86 %). The lowest values of hydrolysis weregenerated by PBS with a mean percentage of 3.24%. Conclusion: ß-1,3-glucanase enzyme is more effective in hydrolysis of denture plaque than papain enzyme.

An opportunistic pathogen, Candida is not only related to oral problems such as oral candidiasis and denture stomatitis, but also to systemic diseases such as aspiration pneumonia and fungemia. The carriage rate of Candida species in the oral cavity of individuals wearing dentures and with removable orthodontic appliances, has increased. Moreover, it is one of the causal pathogens in refractory infected root canals because of its resistance to antifungal drugs in root canal therapy and poses a great challenge during the treatment of patients. This problem has led to the search for alternative strategies for the treatment and management of C. albicans infections. In this mini review, recent preventive strategies against Candida infection in the oral mucosa with natural product-derived antifungal molecules were discussed. Inhibitory strategies by introducing competitive naturally-derived antifungal peptide molecules with Candida adhesion molecules were specifically introduced. In addition, novel sterilization methods for Candida-infected root canals and tooth structures in the oral cavity were considered, with focused attention on the activities of reactive oxygen species. The possibility of application of these novel strategies in clinical treatments and daily life was also proposed.

Everyday denture cleaning is needed to prevent the mouth from denture stomatitis and esthetic concern. Enzyme cleansers are developed in attempt to break down the organic components of denture plaque. The aim of this research is to determine the efficient cleansing duration of papain solution to remove 24-hour denture plaque. The research was done on 16 patients with complete dentures. Plaque removal was determined by Lowry method and SDS PAGE. The result shows that papain dose required to hydrolyze 24-hour dentue plaque is 15.66 TU/mg enzyme activity for 10 minutes soaking. Soaking the denture in papain for 10 minutes makes all of the detected plaque protein bands remobe. In conclusion, efficient cleansing duration of papain solution to remove 24-hour denture plaque is papain activity of 15.66 TU/mg by soaking duration of 10 minutes. Further research is suggested to examine papain residue on the denture that may influence the denture wearer biocompatibility.

Plaque accumulation on internal surface of denture is a common problem among removable denture wearers. Poor denture cleanliness can increase colonization of Candida albicans and cause inflammatory reaction of denture-bearing mucosa, i.e. denture stomatitis. Objective: To find out the effect of denture cleanliness level on denture stomatitis on maxillary denture-bearing mucosa in a group of removable denture wearers who received prosthodontic treatment at Poliklinik Gigi RSMH Palembang and to investigate the denture hygiene habits of removable denture wearers. Methods: Thirty subjects participated in this study. Denture cleanliness level was assessed with disclosing solution to disclose denture plaque on internal surface of maxillary denture. Cleanliness level was graded according to Budtz-Jorgensen. Intraoral examination was done to determine any visible signs of denture stomatitis. Data referring to denture hygiene habits of removable denture wearers was collected from interview using questionnaire. Data were analyzed using the Komolgorov-Smirnov test. Results: Result of the study showed that 40% subjects had poor upper denture cleanliness. Denture stomatitis was observed on maxillary denture-bearing mucosa in 43.3% subjects. Aim: A comparative study for plaque removing efficacy between toothpaste, liquid handwashing soap, and two chemical-soak denture cleansers (clinsodent denture cleansing powder and fittydent denture cleansing tablet). Materials and Methods: A total of 140 healthy complete denture patients were selected and checked for denture cleansing using toothpaste, liquid handwashing soap, and two chemical denture cleansers: Clinsodent powder and fittydent tablets. Cleansing with plain water was kept as control for twenty patients. Results: The data were analyzed using Wilcoxon signed ranks test for checking the efficacy. The Kruskal--Wallis test was used for comparing the plaque removing efficacies of each denture cleanser used in the study. All four denture cleansers were significantly effective in removing plaque when compared with plain water, but there was no significant difference among them. Conclusions: There was no significant difference among action of four denture cleansers used in this study. Thus we can infer that patients can use liquid handwashing soap or commercial products to overcome disadvantages of toothpastes. Recommendation from the results of this study is the use of liquid handwashing soap for cleansing after every meal and soaking of the denture in commercial denture cleanser during the night. Introduction: The role of bacterial infections in acute exacerbations of chronic obstructive pulmonary disease (COPD) is widely examined. Denture plaque in patients with COPD is an example of bacterial and fungal biofilm, which is a reservoir of potentially pathogenic respiratory tract microorganisms. Poor denture hygiene might cause acute exacerbations of COPD.Objective: Assessment of prevalence of respiratory tract pathogens in denture plaque in stable patients with COPD and it influence on oral ontocenoses depending upon the therapy.Materials and Methods: The study was based on the clinical assessment of oral mucosa and denture hygiene in 53 patients with COPD with mean age of 70 ± 18 years and 14 generally healthy participants with mean age of 65 ± 14 years. Microbiological and mycological tests were performed by culturing direct denture swabs.Results: The study showcased the presence of potential pathogenic micro-organisms in denture plaque of 48 patients with COPD (90%) and nine healthy subjects (64.3%). Yeast-like fungi prevailed in denture surface swabs of 40 (75%) in patients with COPD and 8 (57%) in cases of control group. In 66% of patients, various degree of oral mucosa inflammation prevailed.Conclusions: Denture plaque could be a potential source of bacterial and fungal infections in patients with COPD. Purpose Recent studies have established a relationship between dental plaque and pulmonary infection, particularly in elderly individuals. Given that approximately one in five adults in the UK currently wears a denture, there remains a gap in our understanding of the direct implications of denture plaque on systemic health. The aim of this study was to undertake a comprehensive evaluation of putative respiratory pathogens residing upon dentures using a targeted quantitative molecular approach. Materials and Methods One hundred and thirty patients' dentures were sonicated to remove denture plaque biofilm from the surface. DNA was extracted from the samples and was assessed for the presence of respiratory pathogens by qunatitative polymerase chain reaction (qPCR). Ct values were then used to approximate the number of corresponding colony forming equivalents (CFEs) based on standard curves. Results Of the dentures, 64.6% were colonized by known respiratory pathogens. Six species were identified: Streptococcus aureus, Streptococcus pneumoniae, Pseudomonas aeruginosa, Haemophilus influenzae B, Streptococcus pyogenes, and Moraxella catarrhalis. P. aeruginosa was the most abundant species followed by S. pneumoniae and S. aureus in terms of average CFE and overall proportion of denture plaque. Of the participants, 37% suffered from denture stomatitis; however, there were no significant differences in the prevalence of respiratory pathogens on dentures between healthy and inflamed mouths. Conclusions Our findings indicate that dentures can act as a reservoir for potential respiratory pathogens in the oral cavity, thus increasing the theoretical risk of developing aspiration pneumonia. PURPOSE. The aim of this study was to characterize and compare bacterial diversity on the removable partial denture (RPD) framework over time. MATERIALS AND METHODS. This descriptive pilot study included five women who were rehabilitated with free-end mandibular RPD. The biofilm on T-bar clasps were collected 1 week (t1) and 4 months (t2) after the RPD was inserted (t0). Bacterial 16S rDNA was extracted and PCR amplified. Amplicons were cloned; clones were submitted to cycle sequencing, and sequences were compared with GenBank (98% similarity). RESULTS. A total of 180 sequences with more than 499 bp were obtained. Two phylogenetic trees with 84 (t1) and 96 (t2) clones represented the bacteria biofilm at the RPD. About 93% of the obtained phylotypes fell into 25 known species for t1 and 17 for t2, which were grouped in 5 phyla: Firmicutes (t1=82%; t2=60%), Actinobacteria (t1=5%; t2=10%), Bacteroidetes (t1=2%; t2=6%), Proteobacteria (t1=10%; t2=15%) and Fusobacteria (t1=1%; t2=8%). The libraries also include 3 novel phylotypes for t1 and 11 for t2. Library t2 differs from t1 (P=.004); t1 is a subset of the t2 (P=.052). Periodontal pathogens, such as F. nucleatum, were more prevalent in t2. CONCLUSION. The biofilm composition of the RPD metal clasps changed along time after RPD wearing. The RPD framework may act as a reservoir for potentially pathogenic bacteria and the RPD wearers may benefit from regular follow-up visits and strategies on prosthesis-related oral health instructions. The purpose of this study was to perform a quantitative and qualitative evaluation of the biofilms formed on framework alloys invitro. The biofilms formed by unfiltered fresh human saliva or Streptococcus mutans and/or Candida albicans on commerciallypure titanium and gold-copper-platinum demonstrated higher amounts than other alloy and resin samples. In contrast, the silverpalladium-copper-gold showed a significantly decreased level of biofilm formation. Although the adhesion level of Streptococcusmutans on cobalt-chromium was high, that of Candida albicans was less extensive. A T-RFLP analysis and qualitative PCR of themicrobes in the biofilms were performed. In a cluster analysis of all T-RFLP profiles, the cobalt-chromium pattern was integratedinto one cluster. On qualitative PCR, the existence of microorganisms related to caries, preriodontitis and aspiration pneumonia wasobserved. The microbiota of the denture plaque biofilm colonizing the fitting surface of dentures in edentulous subjects with healthy palates ( n = 20) and in edentulous subjects with denture stomatitis ( n = 20) was studied. The numbers of bacteria colonizing the dentures of healthy subjects was significantly less than the numbers colonizing the dentures of stomatitis subjects. The proportions and frequency of isolation of mutans streptococci, lactobacilli, bifidobacteria and yeasts were significantly ( P < 0.05) greater in the subjects with denture stomatitis. The proportions of these organisms in the denture plaque biofilm of the subjects with denture stomatitis were similar to those found in carious lesions, indicating that the site is a low pH environment. The predominant bifidobacterial species in the mouths of dentate subjects is Bifidobacterium dentium but in the edentulous subjects wearing dentures B. dentium was isolated from only one of the 20 subjects with denture stomatitis and from none of the 20 subjects with healthy palates. Instead, Bifidobacterium breve, Bifidobacterium scardovii and Bifidobacterium longum subsp. longum were isolated. Only a single non-oral bifidobacterial species was isolated from each individual and repetitive extragenic palindromic- and BOX-polymerase chain reaction typing methods indicated that the same genotypes were shared between subjects. Using deferred antagonism spot plate assays, interspecies inhibition was demonstrated between oral isolates of B. dentium, B. breve, B. scardovii and B. longum subsp. longum. Here we have shown that bifidobacteria and caries-associated microbiota are present in denture plaque at levels similar to those of carious lesions and B. dentium cannot be maintained in an edentulous mouth. The purpose of this study was to clarify which oral environmental factors affected number of microbes in saliva in an edentulous environment. We enrolled 68 edentulous subjects in the study. Numbers of total anaerobic bacteria and Candida species in saliva were determined. Age, sex, un-stimulated salivary flow rate, pH and viscosity of saliva, histatin level in saliva, tongue coating status, tongue pressure, denture plaque status, material of denture base, duration of edentulism, frequency of self oral health care and number of cigarettes per day were also investigated as oral environmental factors. Correlation between number of total anaerobic bacteria or Candida species and each oral environmental factor was determined with the Spearman rank correlation coefficient. Stepwise logistic regression analysis was used to identify which factors were significantly associated with level of total anaerobic bacteria and Candida species. Correlation and stepwise logistic regression analyses revealed associations between un-stimulated salivary flow rate, tongue coating status, denture plaque status or frequency of self oral health care and number of total anaerobic bacteria. The correlation analysis showed a significant correlation between age and number of total anaerobic bacteria. Stepwise logistic analysis revealed associations between pH of saliva or viscosity of saliva and level of anaerobic bacteria; it also revealed associations between histatin level in saliva or un-stimulated salivary flow rate and level of Candida species. Mechanical cleaning of dentures is effective in preventing infections such as aspiration pneumonia and denture stomatitis. For denture wearers with a physical handicap and the elderly, however, mechanical cleaning can present problems. The aim of this study was to investigate the effect of coating denture base acrylic resin with titanium dioxide (TiO2) in the inhibition of oral microbial adhesion. We prepared uniformly sized acrylic resin plates (10 mm × 10 mm × 0·5 mm), which were divided into two groups (a non-coated group and a TiO2-coated group). The plates were immersed in cultured Streptococcus sanguinis or Candida albicans and incubated for 24 h. After incubation, each plate was washed to remove loosely adherent microorganisms, and then incubated for a further 24 h. Adenosine triphosphate (ATP) content of the microorganisms was evaluated using a reagent containing benzalkonium, which extracts intra-cellular ATP. In addition, to determine biofilm formation, we also observed each plate by scanning electron microscopy (SEM). We found that the ATP content of both S. sanguinis and C. albicans was reduced by the TiO2 coating ( P = 0·000). Observation by SEM confirmed that the TiO2 coating inhibited biofilm formation. Objectives: Current methods available for denture plaque assessment utilise visual and planimetric techniques. This paper evaluates the use of the Quantitative Light-induced Fluorescence system (QLF) in image capture of denture plaque and the suitability of these images for planimetric plaque measurement. It is proposed that fluorescence imaging could provide a valuable and sensitive standardising method for plaque assessment in clinical trials for denture cleansing products and denture hygiene. Indeed, the detection of red fluorescent plaque using the QLF system is indicative of black-pigmented obligate anaerobes and mature plaque.Methods: The QLF system was evaluated in a clinical study for use in denture plaque assessment in comparison to white light based image capture.Results: Despite appearing as a promising system for denture plaque quantification, this study revealed numerous problems associated with the QLF system including small focal depth, thus large numbers of images and processing time were required. In addition, differential fluorescence of acrylic made images unsuitable for plaque quantification.Conclusion: QLF is unsuitable for in vivo denture plaque assessment. Abstract: This article reviews the findings of the original papers related to complete dentures published in the Journal of the Japan Prosthodontic Society (Nippon Hotetsu Shika Gakkai Zasshi), Vol. 52, 2008. A total of six articles focused on complete dentures or materials related to complete dentures were selected and summarized. A variety of subjects in relation to removable prosthodontics were discussed in the articles, including denture plaque control, resilient denture lining materials or denture adhesives, and assessment and/or anatomical analysis of prognosis of complete dentures. The aim of this study was to determine the microbiological origin of plaque fluorescence observed during quantitative light-induced fluorescence (QLF) analysis. Plaque was sampled from dentures, because of easy accessibility and the homogeneous background provided by the denture tooth during imaging, and the acknowledged comparability to occlusal plaque. Forty removable poly(methyl methacrylate) dentures were screened for the presence of fluorescent plaque deposits during QLF analysis. Dentures were photographed, QLF images were recorded and samples of fluorescent plaque were taken. Plaque samples were cultured on fastidious anaerobe agar, Wilkins Chalgren agar and Sabourauds dextrose agar. Plates were screened under QLF and fluorescent colonies were subcultured and identified. Areas of red, orange and green fluorescence were detected on the fitting and non-fitting surfaces of dentures. The red and orange fluorescing species were Prevotella melaninogenica, Actinomyces israelii and Candida albicans, which are generally acknowledged to be secondary colonisers, present in more mature plaque. Green fluorescence was observed in streptococcal species (early colonisers) and Fusobacterium nucleatum (important organism in plaque development). Non-fluorescent colonies were also cultured. Plaque which accumulates on susceptible surfaces tends to be associated with caries, but it may be its maturity, rather than the presence of cariogenic streptococci, that is more likely to provide a microbiological link between red fluorescence and caries. Oral Microbiol Immunol 2005: 20: 206–210. Ozone is known to act as a strong antimicrobial agent against bacteria, fungi, and viruses. We examined the effect of ozonated water on Candida albicans on acrylic denture plate. The heat-cured acrylic resins were cultured with C. albicans. After treatment of flowing ozonated water, the number of attached C. albicans was counted. In some experiments, the test samples were treated with ozonated water in combination with ultrasonication. After exposure to flowing ozonated water (2 or 4 mg/l) for 1 min, viable C. albicans cells were nearly nonexistent. The combination of ozonated water and ultrasonication had a strong effect on the viability of C. albicans adhering to the acrylic resin plates. There were no significant differences in antimicrobial activity against C. albicans between plates immersed in ozonated water with ultrasonication and those treated with commercially available denture cleaners. In addition, electron microscopic analysis revealed that small amounts of C. albicans remained on the plate after exposure to flowing ozonated water or immersion in ozonated water with ultrasonication. Denture plaque has not been studied to the same extent as dental plaque, and although there are many similarities in microbial composition, there are some significant differences. Denture-induced stomatitis is associated with poor denture hygiene, a more acidogenic plaque and the presence ofCandida albicans. Obligate Gram-negative anaerobic micro-organisms, although present in denture plaque, have rarely been specifically investigated. Opportunist pathogens including coliforms and staphylococci have been isolated from dentures. Teeth adjacent to partial dentures are more susceptible to caries and periodontal diseases, perhaps due to an increased plaque buildup at the prosthesis/tooth interface. Little work has been published on malodour associated with dentures. The inert material provides a substratum for the plaque biofilm, which encompasses a range of odour-producing species. The microbiology of the tongue in denture wearers has not been specifically studied. Thus the nature, origin and extent of malodour in denture wearers is ill-defined, but many species capable of producing malodorous compounds are present. The wide age and health range presented by denture wearers further confounds investigation. Objectives: The morbidity and mortality of the dependent elderly that result from aspiration pneumonia are recognized as a major geriatric health problem. Most cases of bacterial pneumonia are initiated following colonization or superinfection of the pharynx by pathogenic bacteria, followed by aspiration of pharyngeal contents. A recent study revealed that bacteria, that commonly cause respiratory infection, colonized the dentures of dependent elderly. This suggests that denture plaque may function as a reservoir of potential respiratory pathogens to facilitate colonization on the pharynx. The purpose of this study was to determine the possible correlation between denture and pharyngeal microflora.Study Design: The denture and pharyngeal bacterial flora of 50 dependent elderly were examined, and the microorganisms identified by culturing. The agreement between the bacterial species in denture plaque and pharyngeal microflora was investigated using the Kappa method.Results: The microorganism species on the dentures and pharyngeal mucosa of the subjects had an agreement rate of 68.5%. The agreement rate for each of the bacterial species of the dentures and pharynx was also demonstrated to be high.Conclusions: Dentures should be considered an important reservoir of organisations which could colonise the pharynx, and the importance of controlling denture plaque for the prevention of aspiration pneumonia cannot be overemphasized.
