The 5-HT3 receptor and indoleamine 2,3-dioxygenase 1 (IDO1) enzyme play a crucial role in the pathogenesis of depression as their activation reduces serotonin contents in the brain. Since molecular docking analysis revealed lycopene as a potent 5-HT3 receptor antagonist and IDO1 inhibitor, we hypothesized that lycopene might disrupt the interplay between the 5-HT3 receptor and IDO1 to mitigate depression. In mice, the depression-like phenotypes were induced by inoculating Bacillus Calmette-Guerin (BCG). Lycopene (intraperitoneal; i.p.) was administered alone or in combination with 5-HT3 receptor antagonist ondansetron (i.p.) or IDO1 inhibitor minocycline (i.p.), and the behavioral screening was performed by the sucrose preference test, open field test, tail suspension test, and splash test which are based on the different principles. Further, the brains were subjected to the biochemical analysis of serotonin and its precursor tryptophan by the HPLC. The results showed depression-like behavior in BCG-inoculated mice, which was reversed by lycopene administration. Moreover, prior treatment with ondansetron or minocycline potentiated the antidepressant action of lycopene. Minocycline pretreatment also enhanced the antidepressant effect of ondansetron indicating the regulation of IDO1 activity by 5-HT3 receptor-triggered signaling. Biochemical analysis of brain samples revealed a drastic reduction in the levels of tryptophan and serotonin in depressed animals, which were restored following treatment with lycopene and its combination with ondansetron or minocycline. Background: COVID-19 pandemic had disrupted postgraduate (PG) medical education in India. This study aimed to assess PG teaching, learning, evaluation (TLE), and innovative TLE methods used during the pandemic., Methods: We conducted a cross-sectional survey using Google Forms to seek anonymous responses from psychiatry teachers across India. Information about PG TLE, postings in COVID-19 care units, methods of teaching adopted in different settings, difficulties faced, and methods of evaluation used were sought in responses. Descriptive statistics methods were used for analysis., Results: The pandemic led to compromised TLE opportunities for the students. Posting of students and teachers in COVID-19 wards for an average of 15 months, students and teachers getting COVID-19, conversion of psychiatry wards to COVID-19 wards, and reduced number of outpatients and inpatients impaired the teaching-learning opportunities. For examination, the evaluation was done with limited standards, like using dummy patients or case vignettes. A few innovative TLE methods were also used during the pandemic., Conclusions: COVID-19 pandemic had led to sub-optimal PG TLE. The study investigated the suitability of three different sludge collected from diverse environments for anammox process establishment. Sludge was collected from SBR (S1) treating municipal wastewater, nitrification tank (S2), denitrification tank (S3) treating fertilizer industrial wastewater. The microbial community in the seed sludge was studied. The presence of anammox bacteria was detected only in seed sludge S2 treating high NH 4 + -N wastewater. Seed sludge S3 showed high abundance of denitrifiers due to NO 3 - -N and organic carbon rich environments in denitrification tank. The anammox start-up performances of sludge were assessed. S2 achieved start-up within 65 days whereas S1 and S3 showed longer start-up period of 79 and 93 days, respectively. S1, S2, S3 achieved nitrogen removal rate of 148.84 gN m -3 day -1 , 159.70 gNm -3 day - 1 and 120.90 gNm -3 day -1 , respectively. Influent NH 4 + -N, NO 3 - -N and organic carbon concentrations governed the abundance of anammox and denitrifying bacteria in seed sludge thereby impacting the anammox start-up., (Copyright © 2022 Elsevier Ltd. Introduction: This in-vitro study aims to evaluate the effect of acidic environment and intracanal medicament on push out bond strength of Biodentine and Mineral Trioxide Aggregate Plus (MTA Plus)., Method: Forty extracted single rooted teeth were sectioned below the cement-enamel junction. The root canals were instrumented using rotary files and then peeso reamer was used to obtain standardized root canal dimension. Specimens were randomly classified into following groups- Group 1: calcium hydroxide in the absence of acidic environment; Group 2: calcium hydroxide in the presence of acidic environment; Group 3: no intracanal medicament in the absence of acidic environment; Group 4: no intracanal medicament in the presence of acidic environment. Specimens were kept for 7 days at room temperature. Thereafter, specimens of each group were transversely sectioned into 1 mm thick slices and divided into 2 sub-groups according to the use of biodentine and MTA Plus. Using Universal Testing Machine, push out bond strength test was carried out and the data were analyzed statistically., Results: There was no statistically significant difference in the bond strength of biodentine and MTA Plus (P>0.05). For both MTA Plus and biodentine, with or without calcium hydroxide, the push out bond strength was less in acidic environment and this difference was more pronounced without calcium hydroxide. In all the four groups, MTA plus showed comparable bond strength to biodentine., Conclusion: MTA Plus is a viable option for apexification. The push out bond strength of Biodentine and MTA Plus is impaired by acidic environment. Prior application of calcium hydroxide slightly increased the bond strength, though the difference was statistically insignificant.

A novel synthon 6-amino-3-(4-methoxyphenyl)-1-phenyl-1H-pyrazolo[3,4-b] pyridine-5-carbaldehyde 4 was synthesized by multistep process. Then Friedlander condensation of this synthon with reactive methylenes was performed to achieve interesting tricyclic 6a-b, tetracyclic 6d and pentacyclic 6c N-heterocycles in excellent yield. The absorption and emission spectra of compounds 1-4 and 6 were measured and observed that the absorption and emission depends on the substituent at C-6 and C-7 position of pyridine ring in naphthyridines skeleton 6.

The inbuilt 2-N-hydroxy-1-oxo-3-carboxylic acid of isoquinolone was designed as pyrophosphate mimic for hepatitis C NS5B polymerase. Various 2-hydroxy-1-oxo-1,2-dihydroisoquinoline-3-carboxylic acid derivatives 11a-p were synthesized and evaluated as HCV NS5B polymerase inhibitors. Compound 11c exhibited moderate inhibitory potency based on the inorganic pyrophosphate generation (IC₅₀ = 9.5 μM) and based on NTP incorporation by NS5B enzyme (IC₅₀ = 5.9 μM). Compound 11c demonstrated antiviral activity (EC₅₀ = 15.7 μM) and good selectivity in HCV genotype 1b replicon Ava.5 cells. Compound 11c reduced the interaction of NTP to NS5B polymerase. Docking model showed that 11c situated in similar orientation to the bound uridine triphosphate in the active site of NS5B polymerase. As a result, 2-hydroxy-1-oxo-1,2-dihydroisoquinoline-3-carboxylic acid was disclosed as a novel inbuilt β-N-Hydroxy-γ-keto-acid pharmacophore for HCV NS5B polymerase inhibitors.

Hepatitis C virus (HCV), a causative agent for non-A and non-B hepatitis, has infected approximately 3% of world's population. The current treatment option of ribavirin in combination with pegylated interferon possesses lower sustained virological response rates, and has serious disadvantages. Unfortunately, no prophylactic vaccine has been approved yet. Therefore, there is an unmet clinical need for more effective and safe anti-HCV drugs. HCV NS5B RNA dependent RNA polymerase is currently pursued as the most popular target to develop safe anti-HCV agents, as it is not expressed in uninfected cells. More than 25 pharmaceutical companies and some research groups have developed ≈50 structurally diverse scaffolds to inhibit NS5B. Here we provide comprehensive account of the drug development process of these scaffolds. NS5B polymerase inhibitors have been broadly classified in nucleoside and non nucleoside inhibitors and are sub classified according to their mechanism of action and structural diversities. With some additional considerations about the inhibitor bound NS5B enzyme X-ray crystal structure information and pharmacological aspects of the inhibitors, this review summarizes the lead identification, structure activity relationship (SAR) studies leading to the most potent NS5B inhibitors with subgenomic replicon activity.

We present a case of 22 year old female who had pulmonary tuberculosis followed by tuberculous meningitis and tuberculomas in past. This time she presented to us with right hemiparesis and altered sensorium. Diagnosis of tumefactive demyelination was made on the basis of typical MRI findings. Patient showed good response to steroids.
