Fractionation of soil organic carbon (SOC) is crucial for mechanistic understanding and modeling of soil organic matter decomposition and stabilization processes. It is often aimed at separating the bulk SOC into fractions with varying turnover rates, but a comprehensive comparison of methods to achieve this is lacking. In this study, a total of 20 different SOC fractionation methods were tested by participating laboratories for their suitability to isolate fractions with varying turnover rates, using agricultural soils from three experimental sites with vegetation change from C3 to C4 22–36 years ago. Enrichment of C4-derived carbon was traced and used as a proxy for turnover rates in the fractions. Methods that apply a combination of physical (density, size) and chemical (oxidation, extraction) fractionation were identified as most effective in separating SOC into fractions with distinct turnover rates. Coarse light SOC separated by density fractionation was the most C4-carbon enriched fraction, while oxidation-resistant SOC left after extraction with NaOCl was the least C4-carbon enriched fraction. Surprisingly, even after 36 years of C4 crop cultivation in a temperate climate, no method was able to isolate a fraction with more than 76% turnover, which challenges the link to the most active plant-derived carbon pools in models. This study examines the distribution of soil organic carbon and carbon-isotopes with depth and among particle size fractions in 2 forest soil profiles of contrasting texture from Cape York Peninsula, Queensland, Australia. The profile on sand has a comparatively low inventory of carbon (557 mg/c[m.sup.2] from 0-100 cm) and exhibits comparatively small variations in [δ.sup.13]C value. In contrast, the clay-rich profile has a much larger inventory of soil organic carbon (1725 mg/c[m.sup.2] from 0-100 cm) and large variations in [δ.sup.13]C value occur both with depth in the profile and between different particle size fractions. The considerable differences in carbon inventories and [δ.sup.13]C values between the sites appear to be largely due to soil textural differences. In the absence of fine minerals the trend in [δ.sup.13]C value with decreasing particle size is to similar or lower [δ.sup.13]C values, due to an increase in the relative abundance of low [δ.sup.13]C compounds in the residue left by microbial decomposition. In the presence of fine minerals, the trend is to higher [δ.sup.13]C values due to the stabilisation of the products of microbial decomposition by the fine minerals. Thus, the bulk [δ.sup.13]C value of soil organic carbon appears to be determined as much by the abundance of fine minerals in a soil profile as by isotope fractionation effects accompanying degradation. It is further postulated that an initial rapid rise in [δ.sup.13]C value in the upper soil layers is due to an increase in the relative importance of higher [sup.13]C, root-derived carbon immediately below the soil surface. Climate change is particularly strong in Northern Eurasia and substantial ecological changes are expected in this wide region. The reshaping and the migration northward of bioclimatic zones may offer opportunities for agriculture development in western and central Siberia. However, the bioclimatic vegetation models currently employed for projections still do not consider soil fertility whereas it is highly critical for plant growth. In the present study, we surveyed the phosphorus (P) status in the south-west of Siberia where soils are developed on loess parent material. We selected six sites differing by pedoclimate conditions and sampled the soil at different depths down to one meter in aspen (Populus tremula L.) forest as well as in grassland areas. The P status was assessed by conventional methods and by isotope dilution kinetics. We found that P concentrations and stocks, as well as their distribution through the soil profile, were rather homogeneous at the studied regional scale, although there were some differences among sites (particularly in organic P). The young age of the soils, together with slow kinetics of soil forming processes, have probably not yet conducted to a sufficiently wide range of soil physico-chemical conditions to observe more diverging P status. The comparison of our dataset to similar vegetation contexts on the global scale revealed that the soils of south-western Siberia, and more generally of Northern Eurasia, has often (very) high levels of total, organic and inorganic P. The amount of plant-available P in topsoils, estimated by the isotopically exchangeable phosphate ions, was not particularly high, but intermediate at the global scale. However, large stocks of plant-available P are stored in subsurface layers which have currently low fine root exploration intensities. Compound-specific isotopic analyses were used to assess the dynamics and origin of organic matter in soils across a 30 year chronosequence where native savanna (C) had been replaced with eucalyptus (C). Apolar lipid fractions were recovered from plants and soils planted with Eucalyptus 0, 7.5, 17 and 30 years ago. The molecular composition of lipids in plants and soils identified three major pentacyclic triterpene methyl esters (PTMEs) specific to savanna, and three odd n-alkanes common to both vegetation types. Savanna-derived PTMEs and δC remained at similar levels in soils after 30 years of eucalyptus growth. C content of PTMEs under savanna was lower than that of bulk soil and displayed the presence of older (1,000s of years) PTMEs in soils. However, the C content of soil PTMEs under eucalyptus was higher than that of the soil PTMEs under savanna. This suggests both recent (10s of years) and 'pre-bomb' mineralization of savanna-derived PTMEs. Compound-specific C analyses of soil n-alkanes showed a trend typical of a C/C vegetation shift, with more depleted δC values with increasing time under Eucalyptus. C analyses of n-alkanes suggest that n-alkanes have high turnover rates in the first few years after land conversion, and n-alkane composition thereafter is dominated by eucalyptus inputs. Compound-specific analyses reveal that lipids from past vegetations remained in soils. The Russian boreal forest, which mainly consists of extensive forests in Siberia, is the largest continuous forest region on Earth and represents 70 % of the world's boreal forest. Siberian forest is a tremendous repository of terrestrial organic carbon (C), which may increase owing to climate change, potential increases in ecosystem productivity and hence C sequestration. Phosphorus (P) availability could limit the C sequestration potential, but tree roots may mine the soil deeper to increase access to mineral P. Improved understanding and quantification of the processes controlling P availability in surface and deep soil layers of forest ecosystems are thus required. Relative contributions of organic and inorganic P and, consequently, P availability in forest ecosystems depend on decomposition processes, which could be strongly affected by vegetation composition, temperature, precipitation, and their changes due to a warming climate. The objectives of the present study were to (1) evaluate P status of surface and deep forest soil horizons from two contrasted biomes in Southwestern Siberia (i.e. forest steppe in the West Siberian plain and blackish ("chernevaya" in Russian) taiga in the low Salair mountains) and (2) assess the effects of vegetation (siberian fir stand, common aspen stand and herbs in a forest gap) and local climate on soil P fractions. Results revealed high contents in total P (645-1042 mgkg"1 in the surface mineral soils) and available inorganic P (diffusive phosphate ions in one week = 83-126 mgkg"1). In addition, there was an accumulation of diffusive phosphate ions in the subsoils resulting from differences between soil horizons in total inorganic P and soil properties. Consequently, deeper root systems may mine substantial amounts of available P for the trees and the potential enhanced growth and C sequestration due to climate change should thus a priori not be P-limited. High proportions of total organic P (47-56% of total P in the surface mineral soils) show that decomposition processes potentially play a significant role in P availability. Results show that decomposition processes are affected by vegetation (deciduous broadleaved trees, evergreen coniferous, herbs) and local climate (precipitations; snow cover with its isolating effect on soil). Results on the effects of plant species and local climate improved our understanding of the potential effects of climate change on P availability through warming and vegetation redistribution. Understanding the chemical composition of soil organic matter (SOM) requires the determination of the dynamics of each class of compounds. We measured the dynamics of carbon in neutral carbohydrates by use of natural 13C labelling in an experimental wheat and maize sequence extending over 23 years. The isotopic composition of individual neutral monosaccharides was determined in hydrolysed particle-size fractions by gas chromatography/combustion/isotope ratio mass spectrometry (GC/C/IRMS) of trimethylsilyl (TMS) derivatives. The sensitivity in terms of 13C/12C ratios ranged between 1 and 2‰ depending on the monosaccharide. The age distribution of neutral sugar carbon was very similar to that of total soil carbon. Particulate organic matter (POM) was characterized by the predominance of glucose and xylose of vegetal origin. In POM > 200 µm, the mean age of sugar-C (5 years) was slightly less than that of total carbon (7 years). Xylose was younger than glucose. The fine fraction 0–50 µm contained mainly glucose, arabinose, galactose, xylose, fucose and mannose, which had predominantly microbial origins. The mean age of carbohydrate carbon in the fraction 0–50 µm was between 60 and 100 years and was similar to that of total organic carbon (OC). No difference in the age of carbon between the individual monosaccharides was found. The POM fraction 50–200 µm had an intermediate signature and turnover. Considering the typical lability of carbohydrates, the relatively great age of carbohydrate carbon may be explained by physical or chemical protection from degradation, as well as by recycling of soil organic matter carbon by soil microbes. Rhizodeposition, i.e. the release of carbon into the soil by growing roots, is an important part of the terrestrial carbon cycle. However thein situnature and dynamics of root-derived carbon in the soil are still poorly understood. Here we made an investigation of the latter in laboratory experiments using13CO2 pulse chase labelling of wheat (Triticum aestivumL.). We analyzed the kinetics of13C-labelled carbon and more specially13C carbohydrates in the rhizosphere. Wheat seedlings-soil mesocosms were exposed to13CO2 for 5 hours in controlled chambers and sampled repeatedly during two weeks for13C/C analysis of organic carbon. After a two-step separation of the soil from the roots, the amount of total organic13C was determined by isotope ratio mass spectrometry as well as the amounts of13C in arabinose, fructose, fucose, glucose, galactose, mannose, rhamnose and xylose. The amount and isotopic ratio of monosaccharides were obtained by capillary gas chromatography coupled with isotope ratio mass spectrometry (GC/C/IRMS) after trimethyl-silyl derivatization. Two fractions were analyzed : total (hydrolysable) and soluble monomeric (water extractable) soil sugars. The amount of organic13C found in the soil, expressed as a percentage of the total photosynthetically fixed13C at the end of the labelling period, reached 16% in the day following labelling and stabilised at 9% after one week. We concluded that glucose under the form of polymers was the dominant moietie of rhizodeposits. Soluble glucose and fructose were also present. But after 2 days, these soluble sugars had disappeared. Forty percent of the root-derived carbon was in the form of neutral sugars, and exhibited a time-increasing signature of microbial sugars. Resistance to poly (ADP-ribose) polymerase (PARP) inhibitors (PARPis) is the major obstacle to their effectiveness in the treatment of homologous recombination (HR)-deficient (HRD) tumors. Hence, developing alternative treatments for HRD tumors is critical. Here, we show that targeting the uracil excision pathway kills HRD tumors, including those with PARPi resistance. We found that the interplay between the two major uracil DNA glycosylases UNG and SMUG1 is regulated by nuclear nicotinamide adenine dinucleotide (NAD + ), which maintains UNG at replication forks (RFs) and restrains SMUG1 chromatin binding. In the absence of UNG, SMUG1 retention on chromatin leads to persistent abasic sites, which incision by APE1 results in PARP1 hyperactivation, stalled RFs, and RAD51 foci. In HRD cells (i.e., BRCA1/2-deficient), this leads to under-replicated DNA that, when propagated throughout mitosis, results in chromosome fragmentation and cell death. In European forests, most tree species form symbioses with ectomycorrhizal (EM) and arbuscular mycorrhizal (AM) fungi. The EM fungi are classified into different morphological types based on the development and structure of their extraradical mycelium. These structures could be root extensions that help trees to acquire nutrients. However, the relationship between these morphological traits and functions involved in soil nutrient foraging is still under debate.We described the composition of mycorrhizal fungal communities under 23 tree species in a wide range of climates and humus forms in Europe and investigated the exploratory types of EM fungi. We assessed the response of this tree extended phenotype to humus forms, as an indicator of the functioning and quality of forest soils. We found a significant relationship between the relative proportion of the two broad categories of EM exploration types (short- or long-distance) and the humus form, showing a greater proportion of long-distance types in the least dynamic soils. As past land-use and host tree species are significant factors structuring fungal communities, we showed this relationship was modulated by host trait (gymnosperms versus angiosperms), soil depth and past land use (farmland or forest).We propose that this potential functional trait of EM fungi be used in future studies to improve predictive models of forest soil functioning and tree adaptation to environmental nutrient conditions., (© 2024. Redesigning agrosystems to include more ecological regulations can help feed a growing human population, preserve soils for future productivity, limit dependency on synthetic fertilizers, and reduce agriculture contribution to global changes such as eutrophication and warming. However, guidelines for redesigning cropping systems from natural systems to make them more sustainable remain limited. Synthetizing the knowledge on biogeochemical cycles in natural ecosystems, we outline four ecological systems that synchronize the supply of soluble nutrients by soil biota with the fluctuating nutrient demand of plants. This synchrony limits deficiencies and excesses of soluble nutrients, which usually penalize both production and regulating services of agrosystems such as nutrient retention and soil carbon storage. In the ecological systems outlined, synchrony emerges from plant-soil and plant-plant interactions, eco-physiological processes, soil physicochemical processes, and the dynamics of various nutrient reservoirs, including soil organic matter, soil minerals, atmosphere, and a common market. We discuss the relative importance of these ecological systems in regulating nutrient cycles depending on the pedoclimatic context and on the functional diversity of plants and microbes. We offer ideas about how these systems could be stimulated within agrosystems to improve their sustainability. A review of the latest advances in agronomy shows that some of the practices suggested to promote synchrony (e.g., reduced tillage, rotation with perennial plant cover, crop diversification) have already been tested and shown to be effective in reducing nutrient losses, fertilizer use, and N 2 O emissions and/or improving biomass production and soil carbon storage. Our framework also highlights new management strategies and defines the conditions for the success of these nature-based practices allowing for site-specific modifications. There is currently an intense debate about the potential for additional organic carbon storage in soil, the strategies by which it may be accomplished and what the actual benefits might be for agriculture and the climate. Controversy forms an essential part of the scientific process, but on the topic of soil carbon storage, it may confuse the agricultural community and the general public and may delay actions to fight climate change. In an attempt to shed light on this topic, the originality of this article lies in its intention to provide a balanced description of contradictory scientific opinions on soil carbon storage and to examine how the scientific community can support decision-making despite the controversy. In the first part, we review and attempt to reconcile conflicting views on the mechanisms controlling organic carbon dynamics in soil. We discuss the divergent opinions about chemical recalcitrance, the microbial or plant origin of persistent soil organic matter, the contribution of particulate organic matter to additional organic carbon storage in soil, and the spatial and energetic inaccessibility of soil organic matter to decomposers. In the second part, we examine the advantages and limitations of big data management and modeling, which are essential tools to link the latest scientific theories with the actions taken by stakeholders. Finally, we show how the analysis and discussion of controversies can guide scientists in supporting stakeholders for the design of (i) appropriate trade-offs for biomass use in agriculture and forestry and (ii) climate-smart management practices, keeping in mind their still unresolved effects on soil carbon storage., Competing Interests: Conflict of interestThe authors declare no competing interests., (© The Author(s) 2023.)

Blindsight regroups the different manifestations of preserved discriminatory visual capacities following the damage to the primary visual cortex. Blindsight types differentially impact objective and subjective perception, patients can report having no visual awareness whilst their behaviour suggests visual processing still occurs at some cortical level. This phenomenon hence presents a unique opportunity to study consciousness and perceptual consciousness, and for this reason, it has had an historical importance for the development of this field of research. From these studies, two main opposing models of the underlying mechanisms have been established: (a) blindsight is perception without consciousness or (b) blindsight is in fact degraded vision, two views that mirror more general theoretical options about whether unconscious cognition truly exists or whether it is only a degraded form of conscious processing. In this article, we want to re-examine this debate in the light of recent advances in the characterization of blindsight and associated phenomena. We first provide an in-depth definition of blindsight and its subtypes, mainly blindsight type I, blindsight type II and the more recently described blindsense. We emphasize the necessity of sensitive and robust methodology to uncover the dissociations between perception and awareness that can be observed in brain-damaged patients with visual field defects at different cognitive levels. We discuss these different profiles of dissociation in the light of both contending models. We propose that the different types of dissociations reveal a pattern of relationship between perception, awareness and metacognition that is actually richer than what is proposed by either of the existing models. A deviated repertoire of the gut microbiome predicts resistance to cancer immunotherapy. Enterococcus hirae compensated cancer-associated dysbiosis in various tumor models. However, the mechanisms by which E. hirae restored the efficacy of cyclophosphamide administered with concomitant antibiotics remain ill defined. Here, we analyzed the multifaceted modes of action of this anticancer probiotic. Firstly, E. hirae elicited emigration of thymocytes and triggered systemic and intratumoral IFNγ-producing and CD137-expressing effector memory T cell responses. Secondly, E. hirae activated the autophagy machinery in enterocytes and mediated ATG4B-dependent anticancer effects, likely as a consequence of its ability to increase local delivery of polyamines. Thirdly, E. hirae shifted the host microbiome toward a Bifidobacteria-enriched ecosystem. In contrast to the live bacterium, its pasteurized cells or membrane vesicles were devoid of anticancer properties. These pleiotropic functions allow the design of optimal immunotherapies combining E. hirae with CD137 agonistic antibodies, spermidine, or Bifidobacterium animalis. We surmise that immunological, metabolic, epithelial, and microbial modes of action of the live E. hirae cooperate to circumvent primary resistance to therapy.

The understanding of soil organic matter (SOM) dynamics has considerably advanced in recent years. It was previously assumed that most SOM consisted of recalcitrant compounds, whereas the emerging view considers SOM as a range of polymers continuously processed into smaller molecules by decomposer enzymes. Mainstreaming this new paradigm in current models is challenging because of their ill-adapted framework. We propose the C-STABILITY model to resolve this issue. Its innovative framework combines compartmental and continuous modeling approaches to accurately reproduce SOM cycling processes. C-STABILITY emphasizes the influence of substrate accessibility on SOM turnover and makes enzymatic and microbial biotransformations of substrate explicit. Theoretical simulations provide new insights on how depolymerization and decomposers ecology impact organic matter chemistry and amount during decomposition and at steady state. The flexible mathematical structure of C-STABILITY offers a promising foundation for exploring new mechanistic hypotheses and supporting the design of future experiments.

Pain resulting from metastatic bone disease is a major unmet clinical need. Studying spinal processing in rodent models of cancer pain is desirable since the percept of pain is influenced in part by modulation at the level of the transmission system in the dorsal horn of the spinal cord. Here, a rodent model of cancer-induced bone pain (CIBP) was generated following syngeneic rat mammary gland adenocarcinoma cell injection in the tibia of male Sprague Dawley rats. Disease progression was classified as "early" or "late" stage according to bone destruction. Even though wakeful CIBP rats showed progressive mechanical hypersensitivity, subsequent in vivo electrophysiological measurement of mechanically evoked deep dorsal horn spinal neuronal responses revealed no change. Rather, a dynamic reorganization of spinal neuronal modulation by descending controls was observed, and this was maladaptive only in the early stage of CIBP. Interestingly, this latter observation corresponded with the degree of damage to the primary afferents innervating the cancerous tissue. Plasticity in the modulation of spinal neuronal activity by descending control pathways reveals a novel opportunity for targeting CIBP in a stage-specific manner. Finally, the data herein have translational potential since the descending control pathways measured are present also in humans.

Southwest Siberia encompasses the forest-steppe and sub-taiga climatic zones and has historically been utilized for agriculture. Coinciding with predicted changes in climate for the region is the pressure of agricultural development; however, a characterization of the soil water and carbon dynamics is lacking. We assessed current soil water properties and soil organic carbon turnover in forests and grasslands for two sites that span the forest steppe and sub-taiga bioclimatic zones. Soil evaporation was 0.62 ± 0.17 mm d -1 (mean ± standard error) in grasslands and 0.45 ± 0.08 mm d -1 in the forests of the forest-steppe site. Evaporation at the sub-taiga site was 1.80 ± 1.70 mm d -1 in grasslands and 0.96 ± 0.05 mm d -1 in forest plots. Evaporation was significantly greater at the sub-taiga site than the forest-steppe site. The density of fine roots explained the soil water isotopic patterns between vegetation types and sites. We found soil organic matter turnover to be three times faster in the sub-taiga site than in the forest-steppe site. Our results show that while climate factors, in particular snow levels, between the two sites are drivers for water and carbon cycles, site level hydrology, soil characteristics, and vegetation directly interact to influence the water and carbon dynamics.

The exchange of carbon between soil organic carbon (SOC) and the atmosphere affects the climate 1,2 and-because of the importance of organic matter to soil fertility-agricultural productivity 3 . The dynamics of topsoil carbon has been relatively well quantified 4 , but half of the soil carbon is located in deeper soil layers (below 30 centimetres) 5-7 , and many questions remain regarding the exchange of this deep carbon with the atmosphere 8 . This knowledge gap restricts soil carbon management policies and limits global carbon models 1,9,10 . Here we quantify the recent incorporation of atmosphere-derived carbon atoms into whole-soil profiles, through a meta-analysis of changes in stable carbon isotope signatures at 112 grassland, forest and cropland sites, across different climatic zones, from 1965 to 2015. We find, in agreement with previous work 5,6 , that soil at a depth of 30-100 centimetres beneath the surface (the subsoil) contains on average 47 per cent of the topmost metre's SOC stocks. However, we show that this subsoil accounts for just 19 per cent of the SOC that has been recently incorporated (within the past 50 years) into the topmost metre. Globally, the median depth of recent carbon incorporation into mineral soil is 10 centimetres. Variations in the relative allocation of carbon to deep soil layers are better explained by the aridity index than by mean annual temperature. Land use for crops reduces the incorporation of carbon into the soil surface layer, but not into deeper layers. Our results suggest that SOC dynamics and its responses to climatic control or land use are strongly dependent on soil depth. We propose that using multilayer soil modules in global carbon models, tested with our data, could help to improve our understanding of soil-atmosphere carbon exchange.

The impacts of fresh organic matter (OM) incorporation in an industrial PAH-contaminated soil on its structure and contaminant concentrations (available and total) were monitored. A control soil and a soil amended with the equivalent of 10 years maize residue input were incubated in laboratory-controlled conditions over 15 months. The structure of the amended soil showed an aggregation process trend which is attributable to (i) the enhanced microbial activity resulting from fresh OM input itself and (ii) the fresh OM and its degradation products. Initially the added organic matter was evenly distributed among all granulodensimetric fractions, and then rapidly degraded in the sand fraction, while stabilizing and accumulating in the silts. PAH degradation remained slight, despite the enhanced microbial biomass activity, which was similar to kinetics of the turnover rate of OM in an uncontaminated soil. The silts stabilized the anthropogenic OM and associated PAH. The addition of fresh OM tended to contribute to this stabilization process. Rationale: Nano-scale secondary ion mass spectrometry (NanoSIMS) is still hampered by a lack of appropriate calibration method for the quantification of elemental and isotopic ratios in heterogeneous materials such as soil samples. The potential of (13)C-(15)N-labeled density fractions of soil to calibrate the C/N, (13)C/(12)C and (15)N/(14)N ratios provided by NanoSIMS was evaluated., Methods: The spatial organization of soil particles found at the macro- and micro-scales were compared. The C/N, (13)C/(12)C and (15)N/(14)N ratios measured at the macroscopic scale from different density fractions using an elemental analyzer coupled to an isotope ratio mass spectrometer (EA/IRMS) were compared with the corresponding micro-scale NanoSIMS measurements. When the macro- and micro-scales patterns were similar, macroscopic scale measurements obtained by EA/IRMS and the corresponding NanoSIMS C/N and (15)N/(14)N ratios averaged per fraction were used to obtain correction equations. The correction method using the internal calibration procedure was compared with the traditional one using a single organic standard., Results: It was demonstrated that the correction method using an internal calibration procedure was applicable for NanoSIMS images acquired on more than 500 µm(2) per fraction and provided more accurate C/N and (15)N/(14)N ratios than the traditional correction method., Conclusions: As long as the NanoSIMS sampling was representative of the macroscopic properties, the correction method using an internal calibration procedure allowed better quantification of the isotope tracers and characterization of the C/N ratios. Direct observations of processes occurring at the mineral-organic interface are increasingly seen as relevant for the cycling of both natural soil organic matter and organic contaminants in soils and sediments. Advanced analytical tools with the capability to visualize and characterize organic matter at the submicrometer scale, such as Nano Secondary Ion Mass Spectrometry (NanoSIMS) and Scanning Transmission X-ray Microscopy (STXM) coupled to Near Edge X-ray Absorption Fine Structure Spectroscopy (NEXAFS), may be combined to locate and characterize mineral-associated organic matter. Taking advantage of samples collected from a decadal (15)N litter labeling experiment in a temperate forest, we demonstrate the potential of NanoSIMS to image intact soil particles and to detect spots of isotopic enrichment even at low levels of (15)N application. We show how microsites of isotopic enrichment detected by NanoSIMS can be speciated by STXM-NEXAFS performed on the same particle. Finally, by showing how (15)N enrichment at one microsite could be linked to the presence of microbial metabolites, we emphasize the potential of this combined imaging and spectroscopic approach to link microenvironment with geochemical process and/or location with ecological function.

The aim of this work was to develop a method for the clean-up of a mycotoxin, i.e. Ochratoxin A (OTA), from cereal extracts employing a new molecularly imprinted polymer (MIP) as selective sorbent for solid-phase extraction (SPE) and to compare with an immunoaffinity column. A first series of experiments was carried out in pure solvents to estimate the potential of the imprinted sorbent in terms of selectivity studying the retention of OTA on the MIP and on a non-imprinted polymer using conventional crushed monolith. The selectivity of the MIP was also checked by its application to wheat extracts. Then, after this feasibility study, two different formats of MIP: crushed monolith and micro-beads were evaluated and compared. Therefore an optimization procedure was applied to the selective extraction from wheat using the MIP beads. The whole procedure was validated by applying it to wheat extract spiked by OTA at different concentration levels and then to a certified contaminated wheat sample. Recoveries close to 100% were obtained. The high selectivity brought by the MIP was compared to the selectivity by an immunoaffinity cartridge for the clean-up of the same wheat sample. The aim of this work was to develop a method for the clean-up and preconcentration of zearalenone from corn and wheat samples employing molecularly imprinted polymer (MIP) as selective sorbent for solid-phase extraction (SPE). Cereal samples were extracted with acetonitrile/water (75:25, v/v) and the extract was diluted with water and applied to an AFFINIMIP ZON MIP-SPE column. The column was then washed to eliminate the interferences and zearalenone was eluted with methanol and quantified using HPLC with fluorescence detection (lambda(exc)=275/lambda(em)=450 nm). The precision and accuracy of the method were satisfactory for both cereals at the different fortification levels tested and it gave recoveries between 82 and 87% (RSDr 2.5-6.2%, n=3) and 86 and 90% (RSDr 0.9-6.8%, n=3) for wheat and maize, respectively. MIP-SPE column capacity was determined to be not less than 6.6 microg of zearalenone and to be at least four times higher than that of immunoaffinity column (IAC). The application of AFFINIMIP ZON molecularly imprinted polymer as a selective sorbent material for detection of zearalenone fulfilled the method performance criteria required by the Commission Regulation (EC) No. 401/2006, demonstrating the suitability of the technique for the control of zearalenone in cereal samples., (Copyright 2010 Elsevier B.V. Vibrio parahaemolyticus is one of the principal bacterial causes for seafood-borne gastroenteritis in the world. In the present study, three sites located on the French Atlantic coast were monitored monthly for environmental parameters over 1 year. The presence of total and pathogenic V. parahaemolyticus in sediment, water and mussel samples was detected following enrichment by culture and real-time PCR (toxR gene, tdh, trh1 and trh2 virulence genes). Using generalized linear models, we showed that the presence of V. parahaemolyticus in water could be explained by a combination of mean temperature over the 7 days before the day of sampling (P < 0.001) and turbidity (P = 0.058). In mussels, an effect of chlorophyll a (P = 0.005) was detected when an effect of the mean salinity over the 7 days before sampling was significant for the sediment (P < 0.001). We did not detect any significant effect of phytoplanktonic blooms or of the number of culturable bacteria on V. parahaemolyticus presence. No sample was revealed positive for tdh. The presence of trh1 and trh2 was positively influenced by the mean temperature during the 2 days before the day of sampling (P < 0.001 and P = 0.032). The importance of these ecological parameters is discussed in relation to the biology of V. parahaemolyticus.
