We have grown Ge3Mn5 clusters by codepositing germanium and manganese atoms on Ge(001) substrates using low temperature molecular beam epitaxy and further annealing the films at high temperature. Clusters are spherical and randomly distributed in the germanium film in epitaxial relationship with the diamond lattice. They exhibit a broad size distribution. By performing a careful x-ray diffraction analysis, we could find that 97% of Ge3Mn5 clusters have their c-axis perpendicular to the film plane while 3% exhibit in-plane c-axis. We could also show a slight in-plane distortion of the Ge3Mn5 lattice leading to a reduction of uniaxial magnetic anisotropy. These observations are well confirmed by complementary superconducting quantum interference device and electron paramagnetic resonance measurements. [ABSTRACT FROM AUTHOR]

Magnetic properties of a doped linear polyarylamine (PA2), whose chain includes alternating para-phenylene and meta-phenylene groups, and of two cyclic and linear model compounds (C2 and D2) were explored by pulsed-EPR nutation spectroscopy, SQUID magnetometry and DFT calculations. Stoichiometrically doped PA2 samples exhibit a pure S = 1 state (exchange coupling constant J = 18 K) with a high spin concentration (0.65) corresponding to 65% of mers bearing holes. Such properties were already observed for doped reticulated polyarylamines but are quite unusual for doped linear polyarylamines. In order to better understand the properties of PA2, model compounds C2 and D2 were also investigated: pure S = 1 spin states could also be obtained, but with higher J (respectively 57 K and 35 K) and, surprisingly, with high but still limited spin concentrations (respectively 0.77 and 0.65).

Photoactivatable fluorescent proteins (FPs) are powerful fluorescent highlighters in live cell imaging and offer perspectives for optical nanoscopy and the development of biophotonic devices. Two types of photoactivation are currently being distinguished, reversible photoswitching between fluorescent and nonfluorescent forms and irreversible photoconversion. Here, we have combined crystallography and (in crystallo) spectroscopy to characterize the Phe-173-Ser mutant of the tetrameric variant of EosFP, named IrisFP, which incorporates both types of phototransformations. In its green fluorescent state, IrisFP displays reversible photoswitching, which involves cis-trans isomerization of the chromophore. Like its parent protein EosFP, IrisFP also photoconverts irreversibly to a red-emitting state under violet light because of an extension of the conjugated pi-electron system of the chromophore, accompanied by a cleavage of the polypeptide backbone. The red form of IrisFP exhibits a second reversible photoswitching process, which may also involve cis-trans isomerization of the chromophore. Therefore, IrisFP displays altogether 3 distinct photoactivation processes. The possibility to engineer and precisely control multiple phototransformations in photoactivatable FPs offers exciting perspectives for the extension of the fluorescent protein toolkit.

The photoreduction of aromatic nitro compounds by alcohols is a well-known reaction; however, the first stages of its mechanism remain controversial. This study aims at characterizing the "primary" radicalar transients involved in this reaction by EPR spectroscopy. Laser flash photolysis (lambda = 266 nm) of nitrobenzene, 5-nitrouracil, p-nitroacetophenone, o-propylnitrobenzene, and 2-nitroresorcinol in ethylene glycol was followed by time-resolved EPR spectroscopy. In all reported TR-EPR spectra, except those obtained from the photolysis of 2-nitroresorcinol, the key intermediate N-hydroxy-arylnitroxide radicals (ArNO*OH, 1-4) could be identified unambiguously. In 2-nitroresorcinol, the radical anion (ArNO*O(-), 5) and a sigma iminoxy radical (6) were observed, and a third radical (7) remains unidentified. These observations indicate that two radicalar mechanisms (by H* transfer and by electron transfer) are competing in the photoreduction mechanism. The attribution of the EPR spectra was helped by DFT calculations of the hyperfine coupling constants (hcc's).

Proteus mirabilis catalase (PMC) belongs to the family of NADPH binding catalases. The function of NADPH in these enzymes is still a matter of debate. This study presents the effects of two independent phenylalanine mutations (F194 and F215), located between NADPH and heme in the PMC structure. The phenylalanines were replaced with tyrosines which we predicted could carry radicals in a NADPH-heme electron transfer. The X-ray crystal structures of the two mutants indicated that neither the binding site of NADPH nor the immediate environment of the residues was affected by the mutations. Measurements using H2O2 as a substrate confirmed that the variants were as active as the native enzyme. With equivalent amounts of peroxoacetic acid, wild-type PMC, F215Y PMC, and beef liver catalase (BLC) formed a stable compound I, while the F194Y PMC variant produced a compound I which was rapidly transformed into compound II and a tyrosyl radical. EPR studies showed that this radical, generated by the oxidation of Y194, was not related to the previously observed radical in BLC, located on Y369. In the presence of excess NADPH, compound I was reduced to a resting enzyme (k(obs) = 1.7 min(-1)) in a two-electron process. This was independent of the enzyme's origin and did not require any thus far identified tyrosyl radicals. Conversely, the presence of a tyrosyl radical in F194Y PMC greatly enhanced the oxidation of reduced beta-nicotinamide mononucleotide under a steady-state H2O2 flow with observable compound II. This process could involve a one-electron reduction of compound I via Y194.
