Previous resting-state functional magnetic resonance imaging (rs-fMRI) studies have widely explored the temporal connection changes in the human brain following long-term sleep deprivation (SD). However, the frequency-specific topological properties of sleep-deprived functional networks remain virtually unclear. In this study, thirty-seven healthy male subjects underwent resting-state fMRI during rested wakefulness (RW) and after 36 hours of SD, and we examined frequency-specific spectral connection changes (0.01–0.08 Hz, interval = 0.01 Hz) caused by SD. First, we conducted a multivariate pattern analysis combining linear SVM classifiers with a robust feature selection algorithm, and the results revealed that accuracies of 74.29%-84.29% could be achieved in the classification between RW and SD states in leave-one-out cross-validation at different frequency bands, moreover, the spectral connection at the lowest and highest frequency bands exhibited higher discriminative power. Connection involving the cingulo-opercular network increased most, while connection involving the default-mode network decreased most following SD. Then we performed a graph-theoretic analysis and observed reduced low-frequency modularity and high-frequency global efficiency in the SD state. Moreover, hub regions, which were primarily situated in the cerebellum and the cingulo-opercular network after SD, exhibited high discriminative power in the aforementioned classification consistently. The findings may indicate the frequency-dependent effects of SD on the functional network topology and its efficiency of information exchange, providing new insights into the impact of SD on the human brain.

Summary: The cognitive and behavioral functions of the human brain are supported by its frequency multiplexing mechanism. However, there is limited understanding of the dynamics of the functional network topology. This study aims to investigate the frequency-specific topology of the functional human brain using 7T rs-fMRI data. Frequency-specific parcellations were first performed, revealing frequency-dependent dynamics within the frontoparietal control, parietal memory, and visual networks. An intrinsic functional atlas containing 456 parcels was proposed and validated using stereo-EEG. Graph theory analysis suggested that, in addition to the task-positive vs. task-negative organization observed in static networks, there was a cognitive control system additionally from a frequency perspective. The reproducibility and plausibility of the identified hub sets were confirmed through 3T fMRI analysis, and their artificial removal had distinct effects on network topology. These results indicate a more intricate and subtle dynamics of the functional human brain and emphasize the significance of accurate topography.

IntroductionSpontaneous low-frequency oscillations play a key role in brain activity. However, the underlying mechanism and origin of low-frequency oscillations remain under debate.MethodsOptical imaging and an electrophysiological recording system were combined to investigate spontaneous oscillations in the hemodynamic parameters and neuronal activity of awake and anesthetized mice after Nω-nitro-L-arginine methyl ester (L-NAME) administration.ResultsThe spectrum of local field potential (LFP) signals was significantly changed by L-NAME, which was further corroborated by the increase in energy and spatial synchronization. The important finding was that L-NAME triggered regular oscillations in both LFP signals and hemodynamic signals. Notably, the frequency peak of hemodynamic signals can be different from that of LFP oscillations in awake mice.DiscussionA model of the neurovascular system was proposed to interpret this mismatch of peak frequencies, supporting the view that spontaneous low-frequency oscillations arise from multiple sources.

Comparative analysis of human, macaque and mouse function and genetic heterogeneity in the brain reveals links between gene expression and orderly topography of functional networks.

Much effort has been devoted towards the identification of brain areas recruited during driving—as one of the most common motor skills of human beings. However, how driving experience impacts on the brain’s intrinsic functional architecture has not been fully investigated. Using resting-state fMRI data collected from 20 taxi drivers and 20 nondrivers, this paper asks whether there exists specific brain network integration encoding driving behavior. First, to address this, we proposed a general framework combining whole-brain functional connectivity analysis with effective connectivity analysis based on spectral Dynamic Causal Modeling. The validation results indicated that the application of this framework could effectively discover the brain network that best explained the observed BOLD fluctuations. Second, by segmenting supplementary motor area (SMA) into pre-SMA and SMA proper sub-regions, we used the above framework and discovered a hierarchical architecture with pre-SMA located at the higher level in both driver and control groups. Third, we further evaluated the possibility that driving behavior could be encoded by directed connections among the hierarchy, and found that the effective connectivity from pre-SMA to left superior frontal gyrus could distinguish drivers from nondrivers with a sensitivity of 80%. Our findings provide a new paradigm for analyzing the brain’s intrinsic functional integration, and may shed new light on the theory of neuroplasticity that training and experience can remodel the patterns of correlated spontaneous brain activity between specific processing regions. Meanwhile, from a methodological advantage perspective, our proposed framework takes the functional connectivity results as a prior, enabling subsequent spectral DCM to efficiently assess functional integration at a whole-brain scale, which is not available by only using other DCM methods, such as stochastic DCM or the State-of-the-Art multimodal DCM.

Brain-controlled wheelchairs are one of the most promising applications that can help people gain mobility after their normal interaction pathways have been compromised by neuromuscular diseases. The feasibility of using brain signals to control wheelchairs has been well demonstrated by healthy people in previous studies. However, most potential users of brain-controlled wheelchairs are people suffering from severe physical disabilities or who are in a “locked-in” state. To further validate the clinical practicability of our previously proposed P300-based brain-controlled wheelchair, in this study, 10 subjects with severe spinal cord injuries participated in three experiments and completed ten predefined tasks in each experiment. The average accuracy and information transfer rate (ITR) were 94.8% and 4.2 bits/min, respectively. Moreover, we evaluated the physiological and cognitive burdens experienced by these individuals before and after the experiments. There were no significant changes in vital signs during the experiment, indicating minimal physiological and cognitive burden. The patients’ average systolic blood pressure before and after the experiment was 113±13.7 mmHg and 114±11.9 mmHg, respectively (P = 0.122). The patients’ average heart rates before and after the experiment were 79±8.4/min and 79±8.2/min, respectively (P = 0.147). The average task load, measured by the National Aeronautics and Space Administration task load index, ranged from 10.0 to 25.5. The results suggest that the proposed P300-based brain-controlled wheelchair is safe and reliable; additionally, it does not significantly increase the patient’s physical and mental task burden, demonstrating its potential value in clinical applications. Our study promotes the development of a more practical brain-controlled wheelchair system.

Objective: Dim target detection in remote sensing images is a significant and challenging problem. In this work, we seek to explore event-related brain responses of dim target detection tasks and extend the brain-computer interface (BCI) systems to this task for efficiency enhancement. Methods: We develop a BCI paradigm named Asynchronous Visual Evoked Paradigm (AVEP), in which subjects are required to search the dim targets within satellite images when their scalp electroencephalography (EEG) signals are simultaneously recorded. In the paradigm, stimulus onset time and target onset time are asynchronous because subjects need enough time to confirm whether there are targets of interest in the presented serial images. We further propose a Domain adaptive and Channel-wise attention-based Time-domain Convolutional Neural Network (DC-tCNN) to solve the single-trial EEG classification problem for the AVEP task. In this model, we design a multi-scale CNN module combined with a channel-wise attention module to effectively extract event-related brain responses underlying EEG signals. Meanwhile, domain adaptation is proposed to mitigate cross-subject distribution discrepancy. Results: The results demonstrate the superior performance and better generalizability of this model in classifying the single-trial EEG data of AVEP task in contrast to typical EEG deep learning networks. Visualization analyses of spatiotemporal features also illustrate the effectiveness and interpretability of our proposed paradigm and learning model. Conclusion: The proposed paradigm and model can effectively explore ambiguous event-related brain responses on EEG-based dim target detection tasks. Significance: Our work can provide a valuable reference for BCI-based image detection of dim targets.

In epidemiological studies, type 2 diabetes mellitus (T2DM) has been associated with cognitive impairment and dementia, but studies about functional network connectivity in T2DM without cognitive impairment are limited. This study aimed to explore network connectivity alterations that may help enhance our understanding of damage-associated processes in T2DM. MRI data were analyzed from 82 patients with T2DM and 66 normal controls. Clinical, biochemical, and neuropsychological assessments were conducted in parallel with resting-state functional magnetic resonance imaging, and the cognitive status of the patients was assessed using the Montreal Cognitive Assessment-B (MoCA-B) score. Independent component analysis revealed a positive correlation between the salience network and the visual network and a negative connection between the left executive control network and the default mode network in patients with T2DM. The differences in dynamic brain network connectivity were observed in the precuneus, visual, and executive control networks. Internal network connectivity was primarily affected in the thalamus, inferior parietal lobe, and left precuneus. Hemoglobin A1c level, body mass index, MoCA-B score, and grooved pegboard (R) assessments indicated significant differences between the two groups (p < 0.05). Our findings show that key changes in functional connectivity in diabetes occur in the precuneus and executive control networks that evolve before patients develop cognitive deficits. In addition, the current study provides useful information about the role of the thalamus, inferior parietal lobe, and precuneus, which might be potential biomarkers for predicting the clinical progression, assessing the cognitive function, and further understanding the neuropathology of T2DM.

Dividing a pre-defined brain region into several heterogenous subregions is crucial for understanding its functional segregation and integration. Due to the high dimensionality of brain functional features, clustering is often postponed until dimensionality reduction in traditional parcellation frameworks occurs. However, under such stepwise parcellation, it is very easy to fall into the dilemma of local optimum since dimensionality reduction could not take into account the requirement of clustering. In this study, we developed a new parcellation framework based on the discriminative embedded clustering (DEC), combining subspace learning and clustering in a common procedure with alternative minimization adopted to approach global optimum. We tested the proposed framework in functional connectivity-based parcellation of the hippocampus. The hippocampus was parcellated into three spatial coherent subregions along the anteroventral–posterodorsal axis; the three subregions exhibited distinct functional connectivity changes in taxi drivers relative to non-driver controls. Moreover, compared with traditional stepwise methods, the proposed DEC-based framework demonstrated higher parcellation consistency across different scans within individuals. The study proposed a new brain parcellation framework with joint dimensionality reduction and clustering; the findings might shed new light on the functional plasticity of hippocampal subregions related to long-term navigation experience.

Abstract Major depressive disorder (MDD) is a prevailing chronic mental disorder with lifetime recurring episodes. Recurrent depression (RD) has been reported to be associated with greater severity of depression, higher relapse rate and prominent functioning impairments than first-episode depression (FED), suggesting the progressive nature of depression. However, there is still little evidence regarding brain functional connectome. In this study, 95 medication-free MDD patients (35 with FED and 60 with RD) and 111 matched healthy controls (HCs) underwent resting-state functional magnetic resonance imaging (fMRI) scanning. After six months of treatment with paroxetine, 56 patients achieved clinical remission and finished their second scan. Network-based statistics analysis was used to explore the changes in functional connectivity. The results revealed that, compared with HCs, patients with FED exhibited hypoconnectivity in the somatomotor, default mode and dorsal attention networks, and RD exhibited hyperconnectivity in the somatomotor, salience, executive control, default mode and dorsal attention networks, as well as within and between salience and executive control networks. Moreover, the disrupted components in patients with current MDD did not change significantly when the patients achieved remission after treatment, and sub-hyperconnectivity and sub-hypoconnectivity were still found in those with remitted RD. Additionally, the hypoconnectivity in FED and hyperconnectivity in RD were associated with the number of episodes and total illness duration. This study provides initial evidence supporting that impairment of intrinsic functional connectivity across the course of depression is a progressive process.

Economic Model Predictive Control (EMPC) has recently been researched in many areas, especially in terms of energy. The development of advanced control algorithms has promoted the cost reduction of marine energy generation and improved the efficiency and reliability of marine current turbines. By reducing the loss on the marine turbine system, better economic indicators can be achieved. Based on this, the maximum marine power can be captured such that the entire marine current turbine system can obtain maximum economic benefits. In this paper, it is mainly based on the economic model predictive control framework to optimize the ocean current turbine power generation system. The system index is directly used for the objective function of the EMPC problem, as EMPC has the advantage of dealing with system constraints and cost functions. Compare the performance of the proposed control framework with the performance of commonly used basic MPC controllers, simulation results show that the proposed controller can reduce the loss of the turbine system and gearbox, indicating that more energy capture can be obtained for the system to achieve better economic effects.

Machine learning methods have been frequently applied in the field of cognitive neuroscience in the last decade. A great deal of attention has been attracted to introduce machine learning methods to study the autism spectrum disorder (ASD) in order to find out its neurophysiological underpinnings. In this paper, we presented a comprehensive review about the previous studies since 2011, which applied machine learning methods to analyze the functional magnetic resonance imaging (fMRI) data of autistic individuals and the typical controls (TCs). The all-round process was covered, including feature construction from raw fMRI data, feature selection methods, machine learning methods, factors for high classification accuracy, and critical conclusions. Applying different machine learning methods and fMRI data acquired from different sites, classification accuracies were obtained ranging from 48.3% up to 97%, and informative brain regions and networks were located. Through thorough analysis, high classification accuracies were found to usually occur in the studies which involved task-based fMRI data, single dataset for some selection principle, effective feature selection methods, or advanced machine learning methods. Advanced deep learning together with the multi-site Autism Brain Imaging Data Exchange (ABIDE) dataset became research trends especially in the recent 4 years. In the future, advanced feature selection and machine learning methods combined with multi-site dataset or easily operated task-based fMRI data may appear to have the potentiality to serve as a promising diagnostic tool for ASD.

In blind source separation (BSS) for multisubject functional magnetic resonance imaging (fMRI) data, dimensionality reduction is generally performed for multiple times. This leads to the challenge of determining the number of the retained dimensionality, i.e. the order of BSS models, which dramatically influences the validity and performance of BSS models. In this study, a multisubject analysis method robust to order selection is developed. This approach remains effective for slight dimensionality reduction and thus utilizes more information from original data. Inspired by the idea of signal-intensity-maximization technology, which can suppress the overfitting that occurs during insufficient dimensionality reduction, we rotate the reduced dimensions to the optimized direction so that rotated components have the most significant intensity and smoothness. Because the optimized dimensions contain more useful information, involving dimensional optimization stage can reduce the negative impact of dimensionality reduction in multisubject data analysis. The experiments on simulated data and real fMRI data showed that involving dimensional optimization improves the validity and performance of the BSS model in analyzing multisubject data. The proposed method works better across a wide range of dimensionality reduction levels, allows inaccurate order selection, maintains more useful information, and is suitable for multisubject fMRI analysis, which requires multiple dimensionality reduction.

Some brain abnormalities persist at the remission phase, that is, the state-independent abnormalities, which may be one of the reasons for the high recurrence of major depressive disorder (MDD). Hence, it is of great significance to identify state-independent abnormalities of MDD through longitudinal investigation. Ninety-nine MDD patients and 118 healthy controls (HCs) received diffusion tensor imaging scanning at baseline. After 6-month antidepressant treatment, 68 patients received a second scan, among which 59 patients achieved full clinical remission. Differences in whole-brain structural connectivity (SC) between patients with MDD at baseline and HCs were estimated by two-sample t-tests. Masked with significantly changed SCs in MDD, two-sample t-tests were conducted between the remitted MDD subgroup at follow-up and HCs, and paired t-tests were implemented to compare the differences of SC in the remitted MDD subgroup before and after treatment. Significantly decreased SC between the right insula and the anterior temporal cortex (ATC), between the right ATC and the posterior temporal cortex (PTC), between the left ATC and the auditory cortex as well as increased connectivity between the right posterior cingulate cortex (PCC) and the left medial parietal cortex (MPC) were observed in the MDD group compared with the HC group at baseline (p < 0.05, FDR corrected). The decreased connectivity between the right insula and the ATC and increased connectivity between the right PCC and the left MPC persisted in the remitted MDD subgroup at follow-up (p < 0.05, FDR corrected). The decreased SC between the right insula and the ATC and increased SC between the right PCC and left MPC showed state-independent characters, which may be implicated in the sustained negative attention bias and motor retardation in MDD. In contrast, the decreased SC between the right ATC and the PTC and between the left ATC and the auditory cortex seemed to be state-dependent.

Increasing evidence has suggested that the dynamic properties of functional brain networks are related to individual behaviors and cognition traits. However, current fMRI-based approaches mostly focus on statistical characteristics of the windowed correlation time course, potentially overlooking subtle time-varying patterns in dynamic functional connectivity (dFC). Here, we proposed the use of an end-to-end deep learning model that combines the convolutional neural network (CNN) and long short-term memory (LSTM) network to capture temporal and spatial features of functional connectivity sequences simultaneously. The results on a large cohort (Human Connectome Project, n = 1,050) demonstrated that our model could achieve a high classification accuracy of about 93% in a gender classification task and prediction accuracies of 0.31 and 0.49 (Pearson’s correlation coefficient) in fluid and crystallized intelligence prediction tasks, significantly outperforming previously reported models. Furthermore, we demonstrated that our model could effectively learn spatiotemporal dynamics underlying dFC with high statistical significance based on the null hypothesis estimated using surrogate data. Overall, this study suggests the advantages of a deep learning model in making full use of dynamic information in resting-state functional connectivity, and highlights the potential of time-varying connectivity patterns in improving the prediction of individualized characterization of demographics and cognition traits.

In the mammalian visual system, early stages of visual form perception begin with orientation selective neurons in primary visual cortex (V1). In many species (including humans, monkeys, tree shrews, cats, and ferrets), these neurons are organized in pinwheel-like orientation columns. To study the functional organization within orientation pinwheels, it is important to target pinwheel subdomains precisely. We therefore developed a technique to provide a quantitative determination of the location of pinwheel centers (PCs). Previous studies relied solely on blood vessel images of the cortical surface to guide electrode penetrations to PCs in orientation maps. However, considerable spatial error remained using this method. In the present study, we improved the accuracy of targeting PCs by ensuring perpendicularity of electrodes and by utilizing the orientation tuning of local field potentials (LFP) recorded at or near the optically determined positions.

BackgroundEven with continuous antidepressant treatment, residual symptoms and the risk of relapse can persist in remitted major depressive disorder (MDD) patients. Hence, having a clear recognition of the persistent abnormalities of the underlying neural substrate in MDD through a longitudinal investigation is of great importance.MethodsA total of 127 adult medication-free MDD patients with an acute depressive episode and 118 matched healthy controls (HCs) underwent diffusion tensor imaging. Over a 6-month treatment course, 62 remitted patients underwent a second scan. Remission was defined as a 24-item Hamilton Depression Rating Scale (HAMD24) score ≤7 for at least two weeks. Diffusion tensor imaging was performed with a 3.0 T scanner. Differences in whole-brain fractional anisotropy (FA) between MDD patients and HCs were assessed by an independent t-test using gender, age, and education as covariates.ResultsSignificant FA reductions in the left insula, left middle occipital gyrus, right thalamus, left pallidum and left precuneus were observed in current MDD (cMDD) patients compared with HCs. Moreover, significant FA reductions in the left insula were observed in remitted (rMDD) patients compared to HCs. However, no significant differences in FA values were found when comparing cMDD and rMDD patients.ConclusionsThe abnormalities in the insula showed state-independent characteristics, while the abnormalities in the middle occipital gyrus, thalamus, pallidum and precuneus seemed to be state-dependent impairments in MDD patients.

This paper asks whether integrating multimodal EEG and fMRI data offers a better characterisation of functional brain architectures than either modality alone. This evaluation rests upon a dynamic causal model that generates both EEG and fMRI data from the same neuronal dynamics. We introduce the use of Bayesian fusion to provide informative (empirical) neuronal priors – derived from dynamic causal modelling (DCM) of EEG data – for subsequent DCM of fMRI data. To illustrate this procedure, we generated synthetic EEG and fMRI timeseries for a mismatch negativity (or auditory oddball) paradigm, using biologically plausible model parameters (i.e., posterior expectations from a DCM of empirical, open access, EEG data). Using model inversion, we found that Bayesian fusion provided a substantial improvement in marginal likelihood or model evidence, indicating a more efficient estimation of model parameters, in relation to inverting fMRI data alone. We quantified the benefits of multimodal fusion with the information gain pertaining to neuronal and haemodynamic parameters – as measured by the Kullback-Leibler divergence between their prior and posterior densities. Remarkably, this analysis suggested that EEG data can improve estimates of haemodynamic parameters; thereby furnishing proof-of-principle that Bayesian fusion of EEG and fMRI is necessary to resolve conditional dependencies between neuronal and haemodynamic estimators. These results suggest that Bayesian fusion may offer a useful approach that exploits the complementary temporal (EEG) and spatial (fMRI) precision of different data modalities. We envisage the procedure could be applied to any multimodal dataset that can be explained by a DCM with a common neuronal parameterisation.

Abstract Background Electroencephalogram-based brain–computer interfaces (BCIs) represent novel human machine interactive technology that allows people to communicate and interact with the external world without relying on their peripheral muscles and nervous system. Among BCI systems, brain-actuated wheelchairs are promising systems for the rehabilitation of severely motor disabled individuals who are unable to control a wheelchair by conventional interfaces. Previous related studies realized the easy use of brain-actuated wheelchairs that enable people to navigate the wheelchair through simple commands; however, these systems rely on offline calibration of the environment. Other systems do not rely on any prior knowledge; however, the control of the system is time consuming. In this paper, we have proposed an improved mobile platform structure equipped with an omnidirectional wheelchair, a lightweight robotic arm, a target recognition module and an auto-control module. Based on the you only look once (YOLO) algorithm, our system can, in real time, recognize and locate the targets in the environment, and the users confirm one target through a P300-based BCI. An expert system plans a proper solution for a specific target; for example, the planned solution for a door is opening the door and then passing through it, and the auto-control system then jointly controls the wheelchair and robotic arm to complete the operation. During the task execution, the target is also tracked by using an image tracking technique. Thus, we have formed an easy-to-use system that can provide accurate services to satisfy user requirements, and this system can accommodate different environments. Results To validate and evaluate our system, an experiment simulating the daily application was performed. The tasks included the user driving the system closer to a walking man and having a conversation with him; going to another room through a door; and picking up a bottle of water on the desk and drinking water. Three patients (cerebral infarction; spinal injury; and stroke) and four healthy subjects participated in the test and all completed the tasks. Conclusion This article presents a brain-actuated smart wheelchair system. The system is intelligent in that it provides efficient and considerate services for users. To test the system, three patients and four healthy subjects were recruited to participate in a test. The results demonstrate that the system works smartly and efficiently; with this system, users only need to issue small commands to get considerate services. This system is of significance for accelerating the application of BCIs in the practical environment, especially for patients who will use a BCI for rehabilitation applications.

Background: A lack of a sufficiently large sample at single sites causes poor generalizability in automatic diagnosis classification of heterogeneous psychiatric disorders such as schizophrenia based on brain imaging scans. Advanced deep learning methods may be capable of learning subtle hidden patterns from high dimensional imaging data, overcome potential site-related variation, and achieve reproducible cross-site classification. However, deep learning-based cross-site transfer classification, despite less imaging site-specificity and more generalizability of diagnostic models, has not been investigated in schizophrenia. Methods: A large multi-site functional MRI sample (n = 734, including 357 schizophrenic patients from seven imaging resources) was collected, and a deep discriminant autoencoder network, aimed at learning imaging site-shared functional connectivity features, was developed to discriminate schizophrenic individuals from healthy controls. Findings: Accuracies of approximately 85·0% and 81·0% were obtained in multi-site pooling classification and leave-site-out transfer classification, respectively. The learned functional connectivity features revealed dysregulation of the cortical-striatal-cerebellar circuit in schizophrenia, and the most discriminating functional connections were primarily located within and across the default, salience, and control networks. Interpretation: The findings imply that dysfunctional integration of the cortical-striatal-cerebellar circuit across the default, salience, and control networks may play an important role in the “disconnectivity” model underlying the pathophysiology of schizophrenia. The proposed discriminant deep learning method may be capable of learning reliable connectome patterns and help in understanding the pathophysiology and achieving accurate prediction of schizophrenia across multiple independent imaging sites. With the development of brain imaging technology, increasing amounts of magnetic resonance imaging data are being acquired, and traditional computational analysis methods based on single sites and small samples are facing substantial challenges. Deep learning technology, which is born via artificial intelligence, has shown the powerful ability to solve the classification problem based on big data in many studies, while it has not been widely used in brain imaging classification. Herein, we utilized our proposed novel 3-D deep adding neural network to classify 6008 samples from the largest data sets in the brain imaging field collected from more than 61 centers. The proposed method utilizes multiple convolutional layers to extract gradient information in different orientations and combines spatial information at two scales via the adding operation. High accuracy (over 92.5%) was obtained with a standard fivefold cross-validation strategy, demonstrating that the proposed method can effectively handle big data classifications from multiple centers. Compared with some traditional classification methods and some deep learning architectures, the proposed method was more accurate, demonstrating its stronger power to classify data from multiple centers. Our cross-site classification results prove that the proposed method is robust when training on a data set and testing on another data set. To the best of our knowledge, this paper is the first to classify neuroimaging data on such a large scale from multiple centers with such high accuracy. With its improved performance in classification and transferable program codes, the proposed method can potentially be used in intelligent medical treatment strategies and clinical practices based on mobile terminal.

With the advent of Big Data Imaging Analytics applied to neuroimaging, datasets from multiple sites need to be pooled into larger samples. However, heterogeneity across different scanners, protocols and populations, renders the task of finding underlying disease signatures challenging. The current work investigates the value of multi-task learning in finding disease signatures that generalize across studies and populations. Herein, we present a multi-task learning type of formulation, in which different tasks are from different studies and populations being pooled together. We test this approach in an MRI study of the neuroanatomy of schizophrenia (SCZ) by pooling data from 3 different sites and populations: Philadelphia, Sao Paulo and Tianjin (50 controls and 50 patients from each site), which posed integration challenges due to variability in disease chronicity, treatment exposure, and data collection. Some existing methods are also tested for comparison purposes. Experiments show that classification accuracy of multi-site data outperformed that of single-site data and pooled data using multi-task feature learning, and also outperformed other comparison methods. Several anatomical regions were identified to be common discriminant features across sites. These included prefrontal, superior temporal, insular, anterior cingulate cortex, temporo-limbic and striatal regions consistently implicated in the pathophysiology of schizophrenia, as well as the cerebellum, precuneus, and fusiform, middle temporal, inferior parietal, postcentral, angular, lingual and middle occipital gyri. These results indicate that the proposed multi-task learning method is robust in finding consistent and reliable structural brain abnormalities associated with SCZ across different sites, in the presence of multiple sources of heterogeneity. Functional connectivity, representing a statistical coupling relationship between different brain regions or electrodes, is an influential concept in clinical medicine and cognitive neuroscience. Electroencephalography-derived functional connectivity (EEG-FC) provides relevant characteristic information about individual differences in cognitive tasks and personality traits. However, it remains unclear whether these individual-dependent EEG-FCs remain relatively permanent across long-term sessions. This manuscript utilizes machine learning algorithms to explore the individual specificity and permanence of resting-state EEG connectivity patterns. We performed six recordings at different intervals during a six-month period to examine the variation and permanence of resting-state EEG-FC over a long period. The results indicated that the EEG-FC networks are quite subject-specific with a high-precision identification accuracy of greater than 90%. Meanwhile, the individual specificity remained stable and only varied slightly after six months. Furthermore, the specificity is mainly derived from the internal connectivity of the frontal lobe. Our work demonstrates the existence of specific and permanent EEG-FC patterns in the brain, providing potential information for biometric applications.

Background and PurposePrevious neuroimaging studies have demonstrated type 2 diabetes (T2D)-related brain structural and functional changes are partly associated with cognitive decline. However, less is known about the underlying mechanisms. Chronic hyperglycemia and microvascular complications are the two of most important risk factors related to cognitive decline in diabetes. Cerebral small vessel diseases (CSVDs), such as those defined by lacunar infarcts, white matter hyperintensities (WMHs) and microhemorrhages, are also associated with an increased risk of cognitive decline and dementia. In this study, we examined brain magnetic resonance imaging (MRI) changes in patients in the early stages of T2D without CSVDs to focus on glucose metabolism factors and to avoid the interference of vascular risk factors on T2D-related brain damage.MethodsT2D patients with disease durations of less than 5 years and without any signs of CSVDs (n = 34) were compared with healthy control subjects (n = 24). Whole-brain region-based functional connectivity was analyzed with network-based statistics (NBS), and brain surface morphology was examined. In addition, the Montreal Cognitive Assessment (MoCA) was conducted for all subjects.ResultsAt the whole-brain region-based functional connectivity level, thirty-three functional connectivities were changed in T2D patients relative to those in controls, mostly manifested as pathological between-network positive connectivity and located mainly between the sensory-motor network and auditory network. Some of the connectivities were positively correlated with blood glucose level, insulin resistance, and MoCA scores in the T2D group. The network-level analysis showed between-network hyperconnectivity in T2D patients, but no significant changes in within-network connectivity. In addition, there were no significant differences in MoCA scores or brain morphology measures, including cortical thickness, surface area, mean curvature, and gray/white matter volume, between the two groups.ConclusionThe findings indicate that pathological between-network positive connectivity occurs in the early stages of T2D without CSVDs. The abnormal connectivity may indicate that the original balance of mutual antagonistic/cooperative relationships between the networks is broken, which may be a neuroimaging basis for predicting cognitive decline in early T2D patients.

Previous group-level neuroimaging studies have shown significant gender differences in the human brain. Research on sex-specific brain differences in healthy individuals is an important base for understanding sex-specific expression in psychiatric disorders. This study proposes a multi-layer 3D convolution extreme learning machine (MCN-ELM) to classify male and female brains based on structural MRI (sMRI) grey matter (GM) data scans from human connectome projects (HCP) of 876 healthy adults (491 females). First, the authors extracted multi-scale features by three-scale multi-layer 3D convolution neural networks (CNNs) without fine-tuning the parameters of convolution kernels. Then, they pulled the network output feature maps into a vector as separate ELMs. By voting on the three-scale networks, the MCN-ELM algorithm classifies male and female brains with an accuracy of 98.06% through a 10-fold cross-validation strategy, outperforming other state-of-the-art algorithms. The proposed method may be used to understand other brain diseases. Additionally, the results show that the human brain can be categorised into two distinct classes, male and female brains, suggesting it is better to treat men and women separately when researching psychiatric disorders.

Difference exists widely in cognition, behavior and psychopathology between males and females, while the underlying neurobiology is still unclear. As brain structure is the fundament of its function, getting insight into structural brain may help us to better understand the functional mechanism of gender difference. Previous structural studies of gender difference in Magnetic Resonance Imaging (MRI) usually focused on gray matter (GM) concentration and structural connectivity (SC), leaving cortical morphology not characterized properly. In this study a large dataset is used to explore whether cortical three-dimensional (3-D) morphology can offer enough discriminative morphological features to effectively identify gender. Data of all available healthy controls (N = 1113) from the Human Connectome Project (HCP) were utilized. We suggested a multivariate pattern analysis method called Hierarchical Sparse Representation Classifier (HSRC) and got an accuracy of 96.77% for gender identification. Permutation tests were used to testify the reliability of gender discrimination (p < 0.001). Cortical 3-D morphological features within the frontal lobe were found the most important contributors to gender difference of human brain morphology. Moreover, we investigated gender discriminative ability of cortical 3-D morphology in predefined Anatomical Automatic Labeling (AAL) and Resting-State Networks (RSN) templates, and found the superior frontal gyrus the most discriminative in AAL and the default mode network the most discriminative in RSN. Gender difference of surface-based morphology was also discussed. The frontal lobe, as well as the default mode network, was widely reported of gender difference in previous structural and functional MRI studies, which suggested that morphology indeed affect human brain function. Our study indicates that gender can be identified on individual level by using cortical 3-D morphology and offers a new approach for structural MRI research, as well as highlights the importance of gender balance in brain imaging studies.

Objective: To explore the relationship between functional connectivity and presence of interictal epileptic discharges (IEDs) in different brain regions in intracranial EEG (iEEG). Methods: We studied 38 focal epilepsy patients who underwent simultaneous EEG/fMRI scanning and subsequent intracerebral stereo-EEG investigation. In EEG/fMRI analysis, IEDs with different spatial distributions were considered independent studies and IED-related maximal BOLD responses were evaluated. Studies with iEEG electrodes inside the maximal responses were selected and divided into three groups: Studies with 1. distinct maximal BOLD highly concordant with seizure-onset-zone (SOZ); 2. Moderate maximal BOLD concordant with SOZ; 3. maximal BOLD discordant with SOZ. Using maximal BOLD as seed, its functionally connected zone (FCZ) was determined. IED rates in iEEG channels inside and outside the FCZ were compared in the three groups. The effect of laterality and distance between channels and maximal BOLD, and correlation between functional connectivity values and IED rates were analyzed. Results: Thirty-six studies in 25 patients were included. IED rates of intracranial EEG channels inside the FCZ were significantly higher than outside in Group 1 (p = 2.6×10−6) and Group 2 (p = 1.2×10−3) and the inside-outside difference remained after regressing distance and laterality factors. The graph convolution network has received a lot of attention because it extends the convolution to non-Euclidean domains. However, the graph pooling method is still less concerned, which can learn coarse graph embedding to facilitate graph classification. Previous pooling methods were based on assigning a score to each node and then pooling only the highest-scoring nodes, which might throw away whole neighbourhoods of nodes and therefore information. Here, we proposed a novel pooling method UGPool with a new point-of-view on selecting nodes. UGPool learns node scores based on node features and uniformly pools neighboring nodes instead of top nodes in the score-space, resulting in a uniformly coarsened graph. In multiple graph classification tasks, including the protein graphs, the biological graphs and the brain connectivity graphs, we demonstrated that UGPool outperforms other graph pooling methods while maintaining high efficiency. Moreover, we also show that UGPool can be integrated with multiple graph convolution networks to effectively improve performance compared to no pooling.

Previous studies have demonstrated differences of clinical signs and functional brain network organizations between the left and right mesial temporal lobe epilepsy (mTLE), but the anatomical connectivity differences underlying functional variance between the left and right mTLE remain uncharacterized. We examined 43 (22 left, 21 right) mTLE patients with hippocampal sclerosis and 39 healthy controls using diffusion tensor imaging. After the whole-brain anatomical networks were constructed for each subject, multivariate pattern analysis was applied to classify the left mTLE from the right mTLE and extract the anatomical connectivity differences between the left and right mTLE patients. The classification results reveal 93.0% accuracy for the left mTLE versus the right mTLE, 93.4% accuracy for the left mTLE versus controls and 90.0% accuracy for the right mTLE versus controls. Compared with the right mTLE, the left mTLE exhibited a different connectivity pattern in the cortical-limbic network and cerebellum. The majority of the most discriminating anatomical connections were located within or across the cortical-limbic network and cerebellum, thereby indicating that these disease-related anatomical network alterations may give rise to a portion of the complex of emotional and memory deficit between the left and right mTLE. Moreover, the orbitofrontal gyrus, cingulate cortex, hippocampus and parahippocampal gyrus, which exhibit high discriminative power in classification, may play critical roles in the pathophysiology of mTLE. The current study demonstrated that anatomical connectivity differences between the left mTLE and the right mTLE may have the potential to serve as a neuroimaging biomarker to guide personalized diagnosis of the left and right mTLE.

Idiopathic generalized epilepsy (IGE) patients with generalized tonic-clonic seizures (GTCS) suffer long-term cognitive impairments, and present a higher incidence of psychosocial and psychiatric disturbances than healthy people. It is possible that the cognitive dysfunctions and higher psychopathological risk in IGE-GTCS derive from disturbed causal relationship among core neurocognitive brain networks. To test this hypothesis, we examined the effective connectivity across the salience network (SN), default mode network (DMN), and central executive network (CEN) using resting-state functional magnetic resonance imaging (fMRI) data collected from 27 IGE-GTCS patients and 29 healthy controls. In the study, a combination framework of time domain and frequency domain multivariate Granger causality analysis was firstly proposed, and proved to be valid and accurate by simulation experiments. Using this method, we then observed significant differences in the effective connectivity graphs between the patient and control groups. Specifically, between-group statistical analysis revealed that relative to the healthy controls, the patients established significantly enhanced Granger causal influence from the dorsolateral prefrontal cortex to the dorsal anterior cingulate cortex, which is coherent both in the time and frequency domains analyses. Meanwhile, time domain analysis also revealed decreased Granger causal influence from the right fronto-insular cortex to the posterior cingulate cortex in the patients. These findings may provide new evidence for functional brain organization disruption underlying cognitive dysfunctions and psychopathological risk in IGE-GTCS.

Imaging connectomics based on graph theory has become an effective and unique methodological framework for studying functional connectivity patterns of the developing and aging brain. Normal brain development is characterized by continuous and significant network evolution through infancy, childhood, and adolescence, following specific maturational patterns. Normal aging is related to some resting state brain networks disruption, which are associated with certain cognitive decline. It is a big challenge to design an integral metric to track connectome evolution patterns across the lifespan, which is to understand the principles of network organization in the human brain. In this study, we first defined a brain network eigen-entropy (NEE) based on the energy probability (EP) of each brain node. Next, we used the NEE to characterize the lifespan orderness trajectory of the whole-brain functional connectivity of 173 healthy individuals ranging in age from 7 to 85 years. The results revealed that during the lifespan, the whole-brain NEE exhibited a significant non-linear decrease and that the EP distribution shifted from concentration to wide dispersion, implying orderness enhancement of functional connectome over age. Furthermore, brain regions with significant EP changes from the flourishing (7–20 years) to the youth period (23–38 years) were mainly located in the right prefrontal cortex and basal ganglia, and were involved in emotion regulation and executive function in coordination with the action of the sensory system, implying that self-awareness and voluntary control performance significantly changed during neurodevelopment. However, the changes from the youth period to middle age (40–59 years) were located in the mesial temporal lobe and caudate, which are associated with long-term memory, implying that the memory of the human brain begins to decline with age during this period. Overall, the findings suggested that the human connectome shifted from a relatively anatomical driven state to an orderly organized state with lower entropy.

Biometric recognition (also known as biometrics) refers to the automated recognition of individuals based on their biological or behavioral traits. Examples of biometric traits include fingerprint, palmprint, iris, and face. The brain is the most important and complex organ in the human body. Can it be used as a biometric trait? In this study, we analyze the uniqueness of the brain and try to use the brain for identity authentication. The proposed brain-based verification system operates in two stages: gray matter extraction and gray matter matching. A modified brain segmentation algorithm is implemented for extracting gray matter from an input brain image. Then, an alignment-based matching algorithm is developed for brain matching. Experimental results on two data sets show that the proposed brain recognition system meets the high accuracy requirement of identity authentication. Though currently the acquisition of the brain is still time consuming and expensive, brain images are highly unique and have the potential possibility for authentication in view of pattern recognition.

During the development of neuroimaging, numerous analyses were performed to identify population differences, such as studies on age, gender, and diseases. Researchers first normalized the brain image and then identified features that represent key differences between groups. In these studies, the question of whether normalization (a pre-processing step widely used in neuroimaging studies) reduces the diversity of brains was largely ignored. There are a few studies that identify the differences between individuals after normalization. In the current study, we analyzed brain diversity on an individual level, both qualitatively and quantitatively. The main idea was to utilize brain images for identity authentication. First, the brain images were normalized and registered. Then, a pixel-level matching method was developed to compute the identity difference between different images for matching. Finally, by analyzing the performance of the proposed brain recognition strategy, the individual differences in brain images were evaluated. Experimental results on a 150-subject database showed that the proposed approach could achieve a 100% identification ratio, which indicated distinct differences between individuals after normalization. Thus, the results proved that after the normalization stage, brain images retain their main distinguishing information and features. Based on this result, we suggest that diversity (individual differences) should be considered when conducting group analysis, and that this approach may facilitate group pattern classification.

Major depression and schizophrenia are two of the most serious psychiatric disorders and share similar behavioral symptoms. Whether these similar behavioral symptoms underlie any convergent psychiatric pathological mechanisms is not yet clear. To address this issue, this study sought to investigate the whole-brain resting-state functional magnetic resonance imaging (MRI) of major depression and schizophrenia by using multivariate pattern analysis. Thirty-two schizophrenic patients, 19 major depressive disorder patients and 38 healthy controls underwent resting-state functional MRI scanning. A support vector machine in conjunction with intrinsic discriminant analysis was used to solve the multi-classification problem, resulting in a correct classification rate of 80.9% via leave-one-out cross-validation. The depression and schizophrenia groups both showed altered functional connections associated with the medial prefrontal cortex, anterior cingulate cortex, thalamus, hippocampus, and cerebellum. However, the prefrontal cortex, amygdala, and temporal poles were found to be affected differently by major depression and schizophrenia. Our preliminary study suggests that altered connections within or across the default mode network and the cerebellum may account for the common behavioral symptoms between major depression and schizophrenia. In addition, connections associated with the prefrontal cortex and the affective network showed promise as biomarkers for discriminating between the two disorders.

OBJECTIVES: The thalamus and cerebral cortex are connected via topographically organized, reciprocal connections, which hold a key function in segregating internally and externally directed awareness information. Previous task-related studies have revealed altered activities of the thalamus after total sleep deprivation (TSD). However, it is still unclear how TSD impacts on the communication between the thalamus and cerebral cortex. In this study, we examined changes of thalamocortical functional connectivity after 36 hours of total sleep deprivation by using resting state function MRI (fMRI). MATERIALS AND METHODS: Fourteen healthy volunteers were recruited and performed fMRI scans before and after 36 hours of TSD. Seed-based functional connectivity analysis was employed and differences of thalamocortical functional connectivity were tested between the rested wakefulness (RW) and TSD conditions. RESULTS: We found that the right thalamus showed decreased functional connectivity with the right parahippocampal gyrus, right middle temporal gyrus and right superior frontal gyrus in the resting brain after TSD when compared with that after normal sleep. As to the left thalamus, decreased connectivity was found with the right medial frontal gyrus, bilateral middle temporal gyri and left superior frontal gyrus. CONCLUSION: These findings suggest disruptive changes of the thalamocortical functional connectivity after TSD, which may lead to the decline of the arousal level and information integration, and subsequently, influence the human cognitive functions.

Magnetic resonance imaging studies have reported significant functional and structural differences between depressed patients and controls. Little attention has been given, however, to the abnormalities in anatomical connectivity in depressed patients. In the present study, we aim to investigate the alterations in connectivity of whole-brain anatomical networks in those suffering from major depression by using machine learning approaches. Brain anatomical networks were extracted from diffusion magnetic resonance images obtained from both 22 first-episode, treatment-naive adults with major depressive disorder and 26 matched healthy controls. Using machine learning approaches, we differentiated depressed patients from healthy controls based on their whole-brain anatomical connectivity patterns and identified the most discriminating features that represent between-group differences. An enhanced understanding of how normal aging alters brain structure is urgently needed for the early diagnosis and treatment of age-related mental diseases. Structural magnetic resonance imaging (MRI) is a reliable technique used to detect age-related changes in the human brain. Currently, multivariate pattern analysis (MVPA) enables the exploration of subtle and distributed changes of data obtained from structural MRI images. In this study, a new MVPA approach based on sparse representation has been employed to investigate the anatomical covariance patterns of normal aging. Two groups of participants (group 1:290 participants; group 2:56 participants) were evaluated in this study. These two groups were scanned with two 1.5 T MRI machines. In the first group, we obtained the discriminative patterns using a t-test filter and sparse representation step. We were able to distinguish the young from old cohort with a very high accuracy using only a few voxels of the discriminative patterns (group 1:98.4%; group 2:96.4%). The experimental results showed that the selected voxels may be categorized into two components according to the two steps in the proposed method. The first component focuses on the precentral and postcentral gyri, and the caudate nucleus, which play an important role in sensorimotor tasks. The strongest volume reduction with age was observed in these clusters. The second component is mainly distributed over the cerebellum, thalamus, and right inferior frontal gyrus. These regions are not only critical nodes of the sensorimotor circuitry but also the cognitive circuitry although their volume shows a relative resilience against aging. Considering the voxels selection procedure, we suggest that the aging of the sensorimotor and cognitive brain regions identified in this study has a covarying relationship with each other.

BACKGROUND: Numerous studies have demonstrated the higher-order functions of the cerebellum, including emotion regulation and cognitive processing, and have indicated that the cerebellum should therefore be included in the pathophysiological models of major depressive disorder. The aim of this study was to compare the resting-state functional connectivity of the cerebellum in adults with major depression and healthy controls. METHODS: Twenty adults with major depression and 20 gender-, age-, and education-matched controls were investigated using seed-based resting-state functional connectivity magnetic resonance imaging. RESULTS: Compared with the controls, depressed patients showed significantly increased functional connectivity between the cerebellum and the temporal poles. However, significantly reduced cerebellar functional connectivity was observed in the patient group in relation to both the default-mode network, mainly including the ventromedial prefrontal cortex and the posterior cingulate cortex/precuneus, and the executive control network, mainly including the superior frontal cortex and orbitofrontal cortex. Moreover, the Hamilton Depression Rating Scale score was negatively correlated with the functional connectivity between the bilateral Lobule VIIb and the right superior frontal gyrus in depressed patients. CONCLUSIONS: This study demonstrated increased cerebellar coupling with the temporal poles and reduced coupling with the regions in the default-mode and executive control networks in adults with major depression. These differences between patients and controls could be associated with the emotional disturbances and cognitive control function deficits that accompany major depression. Aberrant cerebellar connectivity during major depression may also imply a substantial role for the cerebellum in the pathophysiological models of depression.

OBJECTIVE: To investigate white matter volume abnormalities in patients with major depression and the effects of antidepressant treatment on white matter volume. METHOD: Magnetic resonance imaging (MRI) was performed on 32 treatment-naïve depressed patients, 17 recovered patients who had received antidepressant treatment and subsequently achieved clinical recovery and 34 matched controls. RESULTS: Relative to the healthy controls, the treatment-naïve depressed patients showed increased white matter volumes in the left dorsolateral prefrontal cortex (DLPFC) and left putamen and reduced white matter volumes in the left cerebellum posterior lobe and left inferior parietal lobule. For the treatment-naïve patients, the length in months of the current depressive episode was positively correlated with the white matter volumes in both the left DLPFC and left putamen. In the recovered patients, the differences in white matter volume were no longer statistically significant relative to healthy controls. No significant difference was found in the total white matter volume among the three groups. CONCLUSIONS: This study demonstrates that there were alterations in the white matter volumes of depressed patients, which might disrupt the neural circuits that are involved in emotional and cognitive function and thus contribute to the pathophysiology of depression. The finding of the significant correlations between refractoriness and the white matter volumes in the left DLPFC and left putamen combined with the finding that antidepressant treatment normalized the white matter volume of recovered patients, suggests that a quantitative, structural MRI measurement could act as a potential biomarker in depression therapy for individual subjects.

The development of large-scale functional brain networks is a complex, lifelong process that can be investigated using resting-state functional connectivity MRI (rs-fcMRI). In this study, we aimed to decode the developmental dynamics of the whole-brain functional network in seven decades (8-79 years) of the human lifespan. We first used parametric curve fitting to examine linear and nonlinear age effect on the resting human brain, and then combined manifold learning and support vector machine methods to predict individuals' "brain ages" from rs-fcMRI data. We found that age-related changes in interregional functional connectivity exhibited spatially and temporally specific patterns. During brain development from childhood to senescence, functional connections tended to linearly increase in the emotion system and decrease in the sensorimotor system; while quadratic trajectories were observed in functional connections related to higher-order cognitive functions. The complex patterns of age effect on the whole-brain functional network could be effectively represented by a low-dimensional, nonlinear manifold embedded in the functional connectivity space, which uncovered the inherent structure of brain maturation and aging. Regression of manifold coordinates with age further showed that the manifold representation extracted sufficient information from rs-fcMRI data to make prediction about individual brains' functional development levels. Our study not only gives insights into the neural substrates that underlie behavioral and cognitive changes over age, but also provides a possible way to quantitatively describe the typical and atypical developmental progression of human brain function using rs-fcMRI.

In this work, we investigated the sustained negative blood oxygen level-dependent (BOLD) response (sNBR) using functional magnetic resonance imaging during a finger tapping task. We observed that the sNBR for this task was more extensive than has previously been reported. The cortical regions involved in sNBR are divided into the following three groups: frontal, somatosensory and occipital. By investigating the spatial structure, area, amplitude, and dynamics of the sNBR in comparison with those of its positive BOLD response (PBR) counterpart, we made the following observations. First, among the three groups, the somatosensory group contained the greatest number of activated voxels and the fewest deactivated voxels. In addition, the amplitude of the sNBR in this group was the smallest among the three groups. Second, the onset and peak time of the sNBR are both larger than those of the PBR, whereas the falling edge time of the sNBR is less than that of the PBR. Third, the long distance between most sNBR foci and their corresponding PBR foci makes it unlikely that they share the same blood supply artery. Fourth, the couplings between the sNBR and its PBR counterpart are distinct among different regions and thus should be investigated separately. These findings imply that the origin of most sNBR foci in the finger-tapping task is much more likely to be neuronal activity suppression rather than "blood steal."

We combined measures of resting-state functional magnetic resonance imaging and diffusion tensor imaging (DTI) to investigate alterations of function-structure relationships in patients with early-onset schizophrenia. DTI analysis revealed reduced fractional anisotropy in right frontal white matter. Corresponding gray matter regions showed reduced functional connectivity with other regions in the brain in patients with early-onset schizophrenia, compared with healthy controls. Our results demonstrated abnormal function-structure relationships in early-onset schizophrenia, and supported the view that white matter lesions might disrupt the neural circuits between frontal regions and other brain regions, and affect the functional connectivity in the frontal cortex.
