Lactobacillus delbrueckii subsp. bulgaricus (L. bulgaricus) is one of the most commonly employed Lactobacillus in the food industry. Exopolysaccharides (EPS) of Lactobacillus, which are known to exhibit probiotic properties, are secondary metabolites produced during the growth of Lactobacillus. This study identified the structure of the EPS produced by L. bulgaricus 1.0207 and investigated the mitigation of L. bulgaricus 1.0207 EPS on H2O2-induced oxidative stress in IPEC-J2 cells. L. bulgaricus 1.0207 EPS consisted of glucose and galactose and possessed a molecular weight of 4.06 × 104 Da. L. bulgaricus 1.0207 EPS exhibited notable scavenging capacity against DPPH, hydroxyl radicals, superoxide anions, and ABTS radicals. Additionally, L. bulgaricus 1.0207 EPS enhanced cell proliferation, reduced intracellular reactive oxygen species (ROS) accumulation, increased activity of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px), catalase (CAT), and total antioxidant capacity (T-AOC) elevated the relative expression of CAT, SOD, HO-1, NQO1, ZO-1, and Occludin genes. Moreover, L. bulgaricus 1.0207 EPS improved the expression of Nrf2, pNrf2, pNrf2/Nrf2, and Bcl-2 proteins, while decreasing the expression of Keap1, Caspase3, and Bax proteins, with the best effect at a concentration of 100 μg/mL. L. bulgaricus 1.0207 EPS mitigated H2O2-induced oxidative stress injury in IPEC-J2 cells by activating the Keap1/Nrf2 pathway. Meanwhile, L. bulgaricus 1.0207 EPS exhibited the potential to decrease apoptosis and restore the integrity of the gut barrier. The findings establish a theoretical foundation for the development and application of L.bulgaricus 1.0207 and its EPS.

Photoelectrocatalytic approaches show promise for contaminate removal in wastewater through redox reactions. However, the direct treatment of very low concentration heavy metals is a challenging task. Copper bismuth oxide is considered as a potential photocathode material due to its appropriate bandgap width and excellent light absorption properties. In this work, we utilize copper bismuth oxide photoelectrodes with micrometer-scale pores to achieve the efficient and complete reduction of micromolar-level hexavalent chromium(VI) in wastewater. In a continuous 180 min experiment, the reduction rate of 5 µM hexavalent chromium reached 97%, which is an order lower than the drinking standard. Such a process was facilitated by the unique hierarchical microstructure of the oxide thin film and the porous morphology. On the other hand, the structural evolution during the operation was analyzed. A surface passivation was observed, suggesting the possible long-term practical application of this material. This study serves as an important reference for the application of photoelectrocatalysis in addressing Cr(VI) pollution in wastewater, with implications for improving water quality and environmental protection.

Enterohemorrhagic Escherichia coli (EHEC) serotype O157:H7 is a commonly encountered foodborne pathogen that can cause hemorrhagic enteritis and lead to hemolytic uremic syndrome (HUS) in severe cases. Bifidobacterium is a beneficial bacterium that naturally exists in the human gut and plays a vital role in maintaining a healthy balance in the gut microbiota. This study investigated the protective effects of B. longum K5 in a mouse model of EHEC O157:H7 infection. The results indicated that pretreatment with B. longum K5 mitigated the clinical symptoms of EHEC O157:H7 infection and attenuated the increase in myeloperoxidase (MPO) activity in the colon of the mice. In comparison to the model group, elevated serum D-lactic acid concentrations and diamine oxidase (DAO) levels were prevented in the K5-EHEC group of mice. The reduced mRNA expression of tight junction proteins (ZO-1, Occludin, and Claudin-1) and mucin MUC2, as well as the elevated expression of virulence factors Stx1A and Stx2A, was alleviated in the colon of both the K5-PBS and K5-EHEC groups. Additionally, the increase in the inflammatory cytokine levels of TNF-α and IL-1β was inhibited and the production of IL-4 and IL-10 was promoted in the K5-EHEC group compared with the model group. B. longum K5 significantly prevented the reduction in the abundance and diversity of mouse gut microorganisms induced by EHEC O157:H7 infection, including blocking the decrease in the relative abundance of Roseburia, Lactobacillus, and Oscillibacter. Meanwhile, the intervention with B. longum K5 promoted the production of acetic acid and butyric acid in the gut. This study provides insights into the use of B. longum K5 for developing probiotic formulations to prevent intestinal diseases caused by pathogenic bacterial infections.

In order to expand the application of bismuth vanadate (BiVO4) to the field of photoelectrochemistry, researchers have explored the potential of BiVO4 in catalyzing or degrading organic substances, potentially presenting a green and eco-friendly solution. A study was conducted to investigate the impact of electrolytes on the photocatalysis of benzyl alcohol by BiVO4. The research discovered that, in an acetonitrile electrolyte (pH 9) with sodium bicarbonate, BiVO4 catalyzed benzyl alcohol by introducing saturated V5+. This innovation addressed the issue of benzyl alcohol being susceptible to catalysis in an alkaline setting, as V5+ was prone to dissolution in pH 9 on BiVO4. The concern of the photocorrosion of BiVO4 was mitigated through two approaches. Firstly, the incorporation of a non-aqueous medium inhibited the formation of active material intermediates, reducing the susceptibility of the electrode surface to photocorrosion. Secondly, the presence of saturated V5+ further deterred the leaching of V5+. Concurrently, the production of carbonate radicals by bicarbonate played a vital role in catalyzing benzyl alcohol. The results show that, in this system, BiVO4 has the potential to oxidize benzyl alcohol by photocatalysis.

Bifidobacterium longum is a common probiotic; both viable and heat-inactivated Bifidobacterium longum have many probiotic effects, such as anticancer effects. But some mechanisms of anticancer effects are still unclear, especially for heat-inactivated probiotics. In this study, we analyzed the effects of viable and heat-inactivated Bifidobacterium longum D42 on human colon cancer cells (HT-29). Cell proliferation, membrane permeability and apoptosis were detected by using the CCK-8 method, LDH method and Annexin V-FITC/PI kits. The ROS level and mitochondrial membrane potential were examined using the fluorescent probes DCFH-DA and JC-1. Real-time fluorescence quantitative PCR (RT-qPCR) and Western blot were used to detect the expression of mitochondrial apoptosis pathway genes and proteins. The results showed that viable and heat-inactivated Bifidobacterium longum D42 at concentrations of 1 × 106 CFU/mL significantly inhibited the proliferation of and increased the level of LDH release of HT-29 colon cancer cells. We found that they could increase the apoptosis rate of HT-29 cells. Moreover, they could also induce apoptosis by inducing cells to produce ROS and destroying the mitochondrial membrane potential of cells. Further studies found that they could increase the mRNA transcription and protein expression levels of the Caspase-3, Caspase-9 and Bax genes in cells, and reduce the mRNA transcription and protein expression levels of the Bcl-2 gene. In summary, our findings revealed that viable and heat-inactivated Bifidobacterium longum D42 have inhibitory effects on proliferation and promote the apoptosis of human colon cancer cells, and also have certain adjuvant drug therapeutic effects and have potential application value in the adjuvant treatment of colon cancer.

Study region: The Three Gorges Reservoir area (TGR), China Study focus: Soil erosion on purple soil hillslope and its negative effects are deleterious to the ecological environment of the TGR. Sheet erosion is the initial form of the slope erosion, which is notably impacted by land degradation, polyacrylamide (PAM) is commonly applied for soil and water conservation purpose, while the sheet erosion on purple soil hillslope treated with PAM remains unclear. New hydrological insights for the region: The findings suggested that the sheet erosion and runoff rate increased as the rainfall intensity was increased and the slope gradient was decreased with increasing PAM application rate until reaching minimum values at 0.80 g m−2, and these quantities increased afterward. The single and combined impacts of the PAM application rate, rainfall intensity and slope gradient on erosion and runoff were significant. The slope gradient, rainfall intensity and PAM application rate also influenced the hydrodynamic parameters, and the stream power was the optimal parameter for describing the sheet erosion. An equation encompassing the rainfall intensity, slope gradient and PAM application rate was used for sheet erosion rate prediction with high accuracy relative to the stream power and PAM application rate. These results could be helpful to better understand sheet erosion on purple soil slopes treated with PAM and offer fundamental guidelines for soil and water conservation in the TGR.

Exopolysaccharides (EPS) are biological macromolecules produced by Bifidobacterium to the outside of the cell wall during growth and metabolism. The EPS of Bifidobacterium have anti-inflammatory and antimicrobial activities and could regulate the immune system. In this study, the Bifidobacterium longum subsp. infantis E4 (B. infantis E4) has 21 specific genes for carbohydrate transport and metabolism compared with other 12 strains of B. infantis and could produce exopolysaccharides. The average molecular weights of EPS-1, EPS-2 and EPS-3 were 2.68 × 104, 3.79 × 104 and 4.72 × 104 Da, respectively. The EPS-1 mainly comprised of 7 kinds of monosaccharide, and EPS-2 and EPS-3 mainly comprised of 8 kinds of monosaccharide. The immunomodulatory and anti-inflammatory properties of EPS-1, EPS-2 and EPS-3 on RAW264.7 cells, spleen lymphocytes and NK cells of mouse were investigated in vitro of this study. EPS-1, EPS-2 and EPS-3 promoted the growth and increased phagocytic index of macrophage RAW264.7 at the concentration of 100 μg/mL. Subsequently, EPS-1, EPS-2 and EPS-3 groups were found to increase splenic lymphocyte proliferation and NK cell activity. Compared with the LPS group, EPS-1, EPS-2 and EPS-3 reduced the production of NO, IL-1β, IL-6, TNF-α and PGE-2 in RAW264.7 cells after in lipopolysaccharide (LPS) induced, with EPS-1 having the best effect. Meanwhile, EPS-1, EPS-2 and EPS-3 down-regulated the expression levels of LPS-induced NF-κβ signaling pathway-related genes p-p65, p-Iκβ, iNOS, COX-2 and reduced the nuclear translocation of p65 protein in RAW264.7 cells. In conclusion, our study suggested that EPS produced by B. infantis E4 has immunostimulatory and anti-inflammatory activities and may serve as a potential prebiotic to maintain future health.

With the rapid development of different nanotechnologies in recent decades, the capture of bioactive ingredients into polymeric matrices for controlled release has become a popular area of research. Incorporation of bioactive components into different encapsulated matrices is a unique approach to protect these valuable components from inactivation in unfavorable in vitro or in vivo environments, maximizing their stability and bioavailability. Compared with conventional encapsulation techniques, electrospinning technology has many advantages, such as high porosity, high surface volume ratio, structural similarity to the extracellular matrix, and high envelope rate for bioactive compounds, and these structural and functional advantages make electrospinning technology a better choice in protecting bioactive components. This paper reviews the basic working principles of electrostatic spinning, the selection of nanofiber polymers, and the parameters affecting the properties of nanofibers. The advantages of electrostatic spinning nanofibers are analyzed, and the application of electrostatic spinning technology in encapsulating different types of bioactive compounds is discussed, and this paper would provide a reference for the further research and application of this technology in food processing and the development of related innovative foods.

With the advent of the era of big data, data mining methods show their powerful information mining ability in various fields, seeking the association information hidden in the data, which is convenient for people to make scientific decisions. This paper analyses the butterfly effect in the agricultural product industry chain from the perspective of producer and consumer by using multidimensional time and space theory and proposes a new price forecasting method. We consider that the price change of agricultural products is not only affected by the balance of market supply and demand but also by the factors of time and space. Taking the pig industry chain of Sichuan Province as an example, this paper explores and excavates the data from 2010 to 2020 in the time dimension. Interestingly, we found that the price changes in pork in the market are generally highly correlated with the prices of slaughtered pigs, piglets a few weeks ago and the prices of multiple feed a few months ago. Based on the precise time-space factors, we improved the price forecasting model, greatly improved the accuracy of price prediction, and proved the effectiveness of multidimensional spatiotemporal association mining. The research in this paper is helpful to establish a brand-new agricultural product price prediction theory, which is of great significance to the development of the agricultural economy and global poverty alleviation.

Here, the authors show a molecular design concept that simultaneously achieves all these requirements by covalently linking an in-situ formed rubber matrix with polymer electronic materials.

The intestine is the largest digestive and immune organ in the human body, with an intact intestinal mucosal barrier. Bifidobacterium longum is the specific gut commensals colonized in the human gut for boosting intestinal immunity to defend against intestinal mucosal immune injury. In the LPS-induced intestinal injury model, the Bifidobacterium longum BL-10 was suggested to boost the intestinal immune. Detailly, compared with the LPS-induced mice, the BL10 group significantly reduced intestine (jejunum, ileum, and colon) tissue injury, pro-inflammatory cytokines (TNF-α, IFN-γ, IL-2, IL-6, IL-17, IL-22, and IL-12) levels and myeloperoxidase activities. Moreover, the B. longum BL-10 significantly increased the number of immunocytes (CD4+ T cells, IgA plasma cells) and the expression of tight junction protein (Claudin1 and Occludin). B. longum BL-10 regulated the body’s immune function by regulating the Th1/Th2 and Th17/Treg balance, which showed a greater impact on the Th1/Th2 balance. Moreover, the results also showed that B. longum BL-10 significantly down-regulated the intestinal protein expression of TLR4, p-IκB, and NF-κB p65. The B. longum BL-10 increased the relative abundance of the genera, including Lachnospiraceae_NK4A136_group and Clostridia_UCG-014, which were related to declining the levels of intestinal injury. Overall, these results indicated that the B. longum BL-10 had great functionality in reducing LPS-induced intestinal mucosal immune injury.

Abstract Echinococcosis is endemic in pastoral regions of south, west and mid-Gansu province, China. The present study aimed to determine the prevalence of echinococcosis in east Gansu, and analyze its associated risk factors. A cross-sectional survey was conducted in 2011 in 12 villages of Xiaonangou township, Huan County in east Gansu province by ultrasound abdominal scan and auxiliary serotest, and a prevalence surveillance study from 2008 to 2014 was performed in one villages by ultrasonography screening. Questionnaire information analysis indicates that the risk factors are in association with the gender, age, and education level. The cross-sectional survey found a cystic echinococcosis prevalence of 2.21% (107/4837). Higher prevalence was seen in females (χ 2 = 4.198, P 60 years) (χ 2 trend = 96.30, P

The photocathodes are essential in photoelectrochemical systems for harvesting solar energy as green fuels. However, the light-absorbing p-type semiconductor in them usually suffers from carrier recombination issues. An effective strategy to address it is fabricating the p-n heterojunction to create an interfacial electric field. However, plenty of deposition process of the n-type layer for this purpose requires either sophisticated instruments or subsequent treatments, which may damage the vulnerable p-type structure. Herein, we report a mild approach for a ready-to-use n-type layer with full functionality. Structural analyses proved the successful coating of a uniform titania layer (up to 40 nm) over Cu2O without damaging its structure. Owing to the high Ti3+ content, the layer possesses excellent charge transport ability and requires no additional annealing. The heterojunction effectively facilitates the carrier separation and positively shifts the photocurrent onset potential for 0.2 V. The Mott–Schottky plot and the impedance study reveal an enhanced carrier collection with reduced charge transfer resistances. Such a nano-heterojunction can be further loaded with the hydrogen evolution catalyst, which almost doubles the photocurrent with an extended lifetime than that of the pristine Cu2O nanoarray. This approach puts forward a potentially scalable and efficient choice for fabricating photoelectrochemical devices.

Abstract Background Increasing studies have demonstrated that long non-coding RNAs (lncRNAs) play an important role in tumor progression. However, the potential biological functions and clinical importance of Linc01234 in oral squamous cell carcinoma (OSCC) remain unclear. Methods We evaluated the expression profile and prognostic value of Linc01234 in OSCC tissues by RT-qPCR. Then, functional in vitro experiments were performed to investigate the effects of Linc01234 on tumor growth, migration and invasion in OSCC. Mechanistically, RT-qPCR, bioinformatic analysis and dual luciferase reporter assays were performed to identify a competitive endogenous RNA (ceRNA) mechanism involving Linc01234, miR-433-3p and PAK4. Results We found that Linc01234 was clearly upregulated in OSCC tissues and cell lines, and its level was positively associated with T stage, lymph node metastasis, differentiation and poor prognosis of patients with OSCC. Our results shown that Linc01234 inhibited cell proliferation and metastatic abilities in CAL27 and SCC25 cells following its knockdown. Mechanistic analysis indicated that Linc01234 may act as a ceRNA (competing endogenous RNA) of miR-433-3p to relieve the repressive effect of miR-433-3p on its target PAK4. Conclusions Our results indicated that Linc01234 promotes OSCC progression through the Linc01234/miR-433/PAK4 axis and might be a potential therapeutic target for OSCC.

Increasing evidence has indicated that oxidative stress is associated with the health of infants. Bifidobacterium, especially B. longum subsp. longum strains, are abundant in the gut microbiota of infants, which may have the potential to ameliorate oxidative damage. Thus, this study aimed to isolate and screen B. longum subsp. longum strains with probiotic characters and antioxidant properties as infants’ dietary supplements. In this study, 24 B. longum subsp. longum strains were isolated from 15 healthy infants identified via 16S rRNA and heat shock protein 60 (hsp60) sequences. B. longum subsp. longum B13, F2, K4, K5, K10, K13, and K15 strains were selected based on high values obtained from autoaggregation, hydrophobicity, and adhesion assays to HT-29 cells. Among these seven strains, B. longum subsp. longum F2, K5, K10, and K15 were selected according to the high tolerance of gastrointestinal tract conditions compared to Bifidobacterium animalis subsp. lactis BB-12. Among these four strains, B. longum subsp. longum K5 was susceptible to common antibiotics and showed the highest intestinal epithelial cell proliferation of CCD 841 CoN. Additionally, B. longum subsp. longum K5 showed a strong antioxidant capacity, and its supernatant exhibited better activity of reducing power, hydroxyl radical scavenging, and DPPH radical scavenging than that of the intact cells with cell-free extracts. The findings indicated that B. longum subsp. longum K5 could be used as a probiotic candidate in infant nutrition.

Asymmetric molecule strategy is considered an effective method to achieve high power conversion efficiency (PCE) of polymer solar cells (PSCs). In this paper, nine oligomers are designed by combining three new electron-deficient units (unitA)—n1, n2, and n3—and three electron-donating units (unitD)—D, E, and F—with their π-conjugation area extended. The relationships between symmetric/asymmetric molecule structure and the performance of the oligomers are investigated using the density functional theory (DFT) and time-dependent density functional theory (TD–DFT) calculations. The results indicate that asymmetry molecule PEn2 has the minimum dihedral angle in the angle between two planes of unitD and unitA among all the molecules, which exhibited the advantages of asymmetric structures in molecular stacking. The relationship of the values of ionization potentials (IP) and electron affinities (EA) along with the unitD/unitA π-extend are revealed. The calculated reorganization energy results also demonstrate that the asymmetric molecules PDn2 and PEn2 could better charge the extraction of the PSCs than other molecules for their lower reorganization energy of 0.180 eV and 0.181 eV, respectively.
