Scientific advancement in exploration and exploitation of microbial community structure is fundamentally driven by discoveries and development of new technologies. This review summarizes the techno-scientific advancement based exploitation of the bacterial diversity of saline and soda lakes. Advancement in DNA based technologies have become a paradigm shift in microbiome community mapping of extreme environment. Further, we focused on alternative modern techniques that can be used as for the identification of bacteria and classification of bacterial diversity, as well as highlighted general description of hypersaline and soda lakes as microbial habitats based on molecular techniques. However molecular methods are detailed briefly as are the standard techniques for assessing genome content for taxonomic classification of bacterial diversity. This review provides advance microbial techniques insight for exploitation of microbial community landscape from extreme water bodies.

The survival after veno-arterial extracorporeal membrane oxygenation score and its lactate modification predict in-hospital mortality in patients based on pre-extracorporeal membrane oxygenation variables. Cardiac arrest history is a significant variable in these scores; however, patients with ongoing cardiac arrest during cannulation were excluded from these models. The goal of this study is to validate the survival after veno-arterial extracorporeal membrane oxygenation score with a lactate modification among patients with ongoing cardiac arrest. In our study, the survival after veno-arterial extracorporeal membrane oxygenation score predicted mortality in all patients, but did so with higher discrimination among ongoing cardiac arrest patients with a lactate modification.

BACKGROUND Extracorporeal membrane oxygenation (ECMO) use in patients with cardiac arrest is increasing. Utilization remains variable between centers using ECMO as a rescue therapy or early protocolized extracorporeal cardiopulmonary resuscitation. METHODS Single-center, retrospective evaluation of cardiac arrest with cardiopulmonary resuscitation and rescue ECMO support from 2011 through 2019. Study objectives included survival, non-neurologic, and neurologic outcomes; validation of the SAVE and modified SAVE (mSAVE) scores for survival and favorable neurologic outcome; and predictive factor identification in cardiac arrest with ECMO rescue therapy. RESULTS Eighty-nine patients were included. In-hospital survival was 38.2% and median CPC score was 2. Survivors had lower BMI (27.9 ± 4.2 kg/m2 vs. 32.3 ± 7.5 kg/m2, P = 0.003), less obesity (BMI ≥ 30 kg/m2) (26.5% vs. 49.1%, P = 0.035), shorter CPR duration (35.5 ± 31.7 m vs. 58.0 ± 49.5 m, P = 0.019), more tracheostomy (38.2% vs. 7.3%, P

In order to obtain a readily derivatized analog of the ionophore antibiotic valinomycin, [1-lysine] valinomycin (Lys-VAL) was synthesized. The compound was built up on a polystyrene support by stepwise segment condensation and was cyclized in solution. The segments used were didepsipeptides protected by the t-butyloxycarbonyl and p-nitrobenzyloxycarbonyl groups. Derivatives prepared by acylation of the epsilon-amino group of Lys-VAL included [14C]acetyl-Lys-VAL, dansyl-Lys-VAL, palmitoyl-Lys-VAL and dithiodiglycoyl-bis-Lys-VAL. These derivatives had a high potassium binding capacity but were in general much less active than VAL in mediating ion transport in membranes.

SummaryHuman blood platelets have important, regulatory functions in diverse hemostatic and pathological disorders, including vascular remodeling, inflammation, and wound repair. Microarray analysis was used to study the molecular basis of essential thrombocythemia, a myeloproliferative disorder with quantitative and qualitative platelet defects associated with cardiovascular and thrombohemorrhagic symptoms, not infrequently neurological. A platelet-expressed gene (HSD17B3) encoding type 3 17β-hydroxysteroid dehydrogenase (previously characterized as a testis-specific enzyme catalyzing the final step in gonadal synthesis of testosterone) was selectively down-regulated in ET platelets, with reciprocal induction of the type 12 enzyme (HSD17B12). Functional 17β-HSD3 activity corresponding to ∼10% of that found in murine testis was demonstrated in normal platelets. The induction of HSD17B12 in ET platelets was unassociated with a concomitant increase in androgen biosynthesis, suggesting distinct functions and/or substrate specificities of the types 3 and 12 enzymes. Application of a molecular assay distinguished ET from normal platelets in 20 consecutive patients (p < 0.0001). These data provide the first evidence that distinct subtypes of steroidogenic 17β−HSDs are functionally present in human blood platelets, and that the expression patterns of HSD17B3 and HSD17B12 are associated with an uncommon platelet disorder manifest by quantitative and qualitative platelet defects.

Surveying birds during the non-breeding season in prairie environments can be difficult because birds are less visible and vocal during this period than they are during the breeding season. We compared the effectiveness of using fixed-radius point counts and rope-dragging transects for surveying non-breeding birds and determining their relative abundances in the Florida Everglades, from November 1997 through January 1998. Effort (person work hours) was compared using species-effort and abundance-effort curves. Relative abundances of total birds, American Bittern (Botaurus lentiginosus), Sedge Wren (Cistothorus platensis), Common Yellowthroat (Geothlypis trichas), and Seaside Sparrow (Ammodramus maritimus mirabilis) were greater using transects. Abundances of other common species were similar using both techniques. More species were detected on transects than on point counts. When considering effort involved, transects detected more total species, but point counts detected a greater total number of birds. Overall, transects took more effort to cover similar amounts of habitat. Differences in detection using these two techniques may be attributed to species-specific behaviors. Research focused on non-breeding season bird communities should consider using rope-dragging transects in appropriate habitat because point counts may underestimate abundances of some species.

The study lights on the health status and treatment seeking, as the older people mostly have incomplete recovering abilities and are more susceptible to disease, syndrome, and sickness as well affected by the several social issues. Therefore, elderly has higher health problems especially most of them suffer from chronic ailments and their treatment seeking is hindered by various socio-economic factors. With this backdrop, it is worth studying the number of hospitalized person and their treatment seeking behaviour as well as nature of ailment they have and which type of care they are providing as per their economic status and perception towards their health conditions. During the 365 days preceding the survey and significant risk factors associated with the prevalence of any chronic ailment are increasing age, literacy status, caste, region and perceived poor health status. The strengths of this study are it provides an update on the status of elderly health and treatment seeking in the country, which is important for policy and program perspective, especially given the fact proportion of elderly in India, is growing considerably.

SummaryThe platelet transcriptome has been extensively characterised using distinct genetic profiling platforms, with evolving evidence for differential expression patterns between healthy individuals and subject cohorts with various haematologic and cardiovascular disorders. Traditional technological platforms for platelet genetic biomarker quantification have limited applicability for clinical molecular diagnostics due to inherent complexities related to RNA isolation and analysis. We have previously established the feasibility of fluorescent microspheres as a simple and reproducible strategy for simultaneous quantification of platelet mRNAs from small volume of blood using intact platelets. We now extend these observations by formally comparing in a 50-member normal cohort the cross-platform behaviour of fluorescent microspheres to the currently accepted Q-PCR standard, using a clinically relevant 15-biomarker gene subset able to discriminate among normal and thrombocytosis cohorts. When compared to Q-PCR, genetic biomarker quantification using fluorescent microspheres demonstrated lower coefficients of variation for low-abundant transcripts, better linearity in serially diluted samples, and good overall between-platform consistency via the geometric mean regression. Neither platform demonstrated age or gender effects for any of the 15 biomarkers studied. Binding site saturation for highly abundant transcripts using fluorescent microspheres can be readily eliminated using an optimal platelet number corresponding to 0.3 ml of peripheral blood, additionally applicable to thrombocytopenic cohorts. These data provide a detailed cross-platform analysis using a relevant biomarker subset, further highlighting the applicability of fluorescent microspheres as potentially superior to Q-PCR for platelet mRNA diagnostics.

Human blood platelets function in wound healing, inflammation, and clot formation. Despite significant progress molecular mechanisms of megakaryocyte maturation remain poorly characterized. We have applied a customized, platelet-specific oligonucleotide gene chip to dissect the molecular events of megakaryopoiesis. CD34 hematopoietic stem cells obtained from 2 distinct adult human donors were differentiated in vitro for 21 days along the megakaryocyte lineage Cell samples were fixed and screened each day for one-color FACS analysis of GPIIB megakaryocyte-specific events (CD41 marker). In parallel, a daily sample was taken for MK-isolation using magnetically labeled CD61 antibodies (MK GPIllA-specific). Osteoblasts and adipocytes are two differentiated cell types which are both derived from mesenchymal stem cells (MSCs) and a reciprocal relationship may exist between osteoblastogenesis and adipogenesis in the bone marrow. Our previous studies in the mouse have indicated that low-magnitude, high-frequency mechanical stimuli can stimulate bone formation. To investigate whether lipid metabolism gene expression is altered during the application of low-level mechanical stimuli, adult female BALB/cByJ mice were subject to either disuse, mechanical vibration, or control conditions. RNA was extracted from the bones upon sacrifice after 4d. By microarray we screened lipid metabolism genes and confirmed them with real-time RT-PCR. Three genes were picked up - PPAR-gamma, Alox15 and Lrp5. PPAR-gamma and Alox15 were significantly down-regulated with mechanical vibrations, while Lrp5 was significantly down-regulated with disuse as indicated by the microarray data. It is speculated that mechanical stimuli down-regulate PPARgamma and Alox15, both of which are involved in promotion of osteoblastogenesis and inhibition of adipogenesis, result in bone formation and fat loss. However, disuse modulated gene expression changes in lipid metabolism were not necessarily opposite from those suffering from mechanical stimuli.
