Organosulfur compounds present in garlic and onion have been evaluated as inhibitors of chemical carcinogenesis. Among them diallylsulfide was mainly investigated and studies demonstrated its metabolization to the corresponding sulfoxide and sulfone. In this work, we report the investigation of the interaction between the diallylsulfide and its oxidized derivatives, through cyclic voltammetry, with horse heart cytocrome-c (on a modified electrode with 4-mercaptopyridine). Our results suggest that there is a reversible interaction between cytocrome-c and diallylsulfide and diallylsulfone and an irreversible interaction with the diallylsulfoxide.

The ConBr lectin, extracted from seeds of the Canavalia brasiliensis beans, is a protein capable of specifically and reversibly binding to the glucose and mannose carbohydrates. The immobilization of such lectin on a modified electrode can be an interesting strategy for the development of biosensors with high selectivity for these sugars. Therefore, the objective of this work is to develop and to evaluate an impedimetric biosensor for glucose and mannose detection, which is based on the immobilization of ConBr on self-assembled monolayers (SAM) of the compound 4,4'-biphenyldithiol (HS2BP) on gold electrode. The biosensor was prepared from the immersion of a previously cleaned gold electrode with a known electrochemically active area in a solution containing 10 mmol L–1 HS2BP in acetone for 1 h for SAM Au/HS2BP formation. Then, the Au/HS2BP electrode was immersed in 10 nmol L–1 ConBr solution in tris-HCl buffer for 1 h, thus the Au/HS2BP/ConBr biosensor was obtained. Electrochemical Impedance and Cyclic Voltammetry using the redox [Fe(CN)6]3–/4– (1.0 mmol L–1) as probe molecules were used to characterize the biosensor fabrication and detection of carbohydrates in solution. By Linear Scan Voltammetry it was determined the amount of adsorbed (Γ) HS2BP molecules on surface as 3.03×10–10 mol cm-2 which is consistent with a monolayer. The surface pKa value of the HS2BP species was also determined by voltammetry as 6.0. The charge-transfer resistance (RCT) values between the Au/HS2BP/ConBr electrode and the probe molecules, calculated from electrochemical impedance results, showed a linear relationship with the concentration (from 25 to 125 nmol L–1) of mannose or glucose in the medium. Thus, the detection limit of the biosensor was calculated as 0.73 and 0.07 nmol L–1 for glucose and mannose, respectively. In addition, the biosensor when stored under suitable conditions (4oC in Tris-HCl buffer) showed good stability and reproducibility for more than three weeks. FUNCAP A lectina ConBr, extraída de sementes da espécie Canavalia brasiliensis, é uma proteína capaz de ligar-se de forma específica e reversível aos carboidratos glicose e manose. A imobilização dessa lectina em eletrodo modificado pode ser uma estratégia interessante para o desenvolvimento de biossensores com alta seletividade para esses carboidratos. Explorando esta capacidade da lectina, o objetivo principal deste trabalho é desenvolver e avaliar um biossensor impedimétrico para detecção de glicose e manose baseado na lectina ConBr imobilizada na monocamada automontada (SAM, do inglês Self-Assembled Monolayers) do composto 4,4′- dimercaptobifenil (HS2BP) sobre eletrodo de ouro. O biossensor foi preparado a partir da imersão por 1 h de um eletrodo de ouro limpo em uma solução de HS2BP 10 mmol L–1 em acetona para formação do eletrodo Au/HS2BP. Em seguida, o eletrodo Au/HS2BP foi imerso em solução de ConBr 10 nmol L–1 em tampão Tris-HCl por 1 h, formando o biossensor Au/HS2BP/ConBr. As técnicas de impedância eletroquímica e voltametria cíclica, utilizando o par redox [Fe(CN)6]3–/4– (1,0 mmol L–1) como espécies de prova, foram utilizadas para caracterizar o biossensor e para os estudos de detecção dos carboidratos em solução. Por voltametria de varredura linear, determinou-se a quantidade de moléculas adsorvidas () de HS2BP como 3,03 ×10–10 mol cm2 consistente com uma monocamada. Determinou-se também o valor de pKa de superfície da espécie HS2BP como 6,08. Os valores de resistência a transferência de carga (RCT) entre o eletrodo Au/HS2BP/ConBr e as espécies de provas calculados por meio da técnica de Impedância Eletroquímica, apresentaram uma relação linear com a concentração (de 25 a 125 nmol L–1) de manose ou glicose no meio. O limite de detecção do biossensor foi calculado como 0,73 e 0,07 nmol L–1 para glicose e manose, respectivamente. Além disso, o biossensor quando armazenado em condições adequadas (4 oC em tampão Tris-HCl) apresentou boa estabilidade e reprodutibilidade por mais de três semanas.

Physical and chemical properties inherent of systems in nanometric dimensions make nanoparticles (NPs) highly interesting regarding the production of versatile materials with high potential of applications. This work presents the results obtained for FeCo magnetic NPs covered with polymers of cashew gum (GC) and carboxymethylated cashew gum (CMCG). NPs were synthesized by thermodecomposition of iron and cobalt salts in presence of a reducing agent and the stabilizing agents oleic acid (AO) and oleylamine (OL). Upon transfer to the aqueous medium by using hexadecyltrimethylammonium (CTAB), aliquots were dropped in solution containing the gum of interest. The vibrational spectrum in the infrared region (IR) obtained for the NPs before the polymer immobilization, showed bands typically assigned to the stabilizing and transfer agents indicating the presence of these species on the isolated material (FeCo-AOOL@CTAB). The IR spectra of the NPs covered with the polymers GC (FeCo-AOOL@CTAB@GC) and GCCM (FeCo-AOOL@CTAB@GC) showed bands assigned to the carboxylate groups in the protonated and non-protonated forms. The thermogravimetric analyses allowed to estimate the mass ratio for NP:polymer as 1:3 and 1:1 for FeCo-AOOL@GC and FeCo-AOOL@GCCM, respectively. The stability of the NPs was evaluated in aqueous dispersion and in physiological medium (PBS, pH 7.4) by means of hydrodynamic diameter (DH) and zeta potential (ζ). Although no agglomeration had been observed during 50 h of suspension in these media, the values of DH observed in water for FeCo-AOOL@CTAB@GC and FeCo-AOOL@CTAB@GCCM (459.78 and 231.92 nm) are considerable lower than those observed in PBS (660.47 e 580.9 nm). The more negative values of ζ determined for FeCo-AOOL@CTAB@GCCM (-30 < ζ < -35 mV) in respect to FeCo-OAOL@CTAB@CG (-20 < ζ < -30 mV), are assigned to the higher amount of carboxylate groups in the GCCM polymer. From the values of DH and ζ, it can be concluded that the stability furnished by the GC polymer to the NPs is associated to electrostatic interactions. For the GCCM polymer, besides this interaction, an additional stabilization due to steric hindrance must be taken into account. CAPES As propriedades físicas e químicas inerentes à sistemas em dimensões nanométricas tornam as nanopartículas (NPs) bastante interessantes do ponto de vista de produção de sistemas versáteis e com elevado potencial de aplicação. Esse trabalho apresenta os resultados obtidos para NPs magnéticas de FeCo recobertas com polímeros de goma de cajueiro pura (GC) e carboximetilada (GCCM). As NPs foram sintetizadas por termodecomposição de sais de ferro e cobalto na presença de um agente redutor e dos agentes estabilizantes ácido oleico (AO) e oleilamina (OL). Após transferência para meio aquoso com o auxílio de brometo de hexadeciltrimetilamonio (CTAB), alíquotas das NPs foram gotejadas em solução contendo a goma de interesse. O espectro vibracional na região do infravermelho (IV) obtido para as NPs antes da incorporação dos polímeros mostrou bandas características dos agentes estabilizantes e de transferência indicando a presença dessas espécies sobre o material isolado (FeCo-AOOL@CTAB). Os espectros IV das NPs recobertas com os polímeros GC (FeCo-AOOL@CTAB@GC) e GCCM (FeCo-AOOL@CTAB@GC) apresentaram bandas atribuídas aos grupos carboxilatos nas formas protonada e desprotonada. As análises termogravimétricas permitiram estimar a relação em massa NP:polímero em 1:3 e 1:1 para os materiais FeCo-AOOL@GC e FeCo-AOOL@GCCM, respectivamente. A estabilidade das NPs foi avaliada em dispersão aquosa e em meio fisiológico (PBS pH 7,4) através do diâmetro hidrodinâmico (DH) e potencial zeta (ζ). Apesar de não apresentarem aglomeração por um período de 50 h de suspensão nesses meios, os valores de DH em água para as amostras FeCo-AOOL@CTAB@GC e FeCo-AOOL@CTAB@GCCM (459,78 e 231,92 nm) são consideravelmente inferiores comparados aos observados em PBS (660,47 e 580,9 nm). Os valores mais negativos de ζ determinados para as NPs de FeCo-AOOL@CTAB@GCCM (-30 < ζ < -35 mV) comparativamente às NPs FeCo-OAOL@CTAB@CG (-20 < ζ < -30 mV), devem-se a maior quantidade de grupos carboxílicos presentes no polímero GCCM. A partir dos resultados de DH e ζ, pode-se concluir que a estabilidade conferida às NPs pelo polímero GC deve-se à interações eletrostáticas. Para o polímero GCCM, além dessas interações, deve-se considerar uma estabilização adicional resultante do impedimento estérico entre as cadeias carbônicas.

This work presents the results obtained for three novel bipyridine derivtives (mbpy-R), named as: 4'-methyl-4-(naphth-2-yl)-2,2'-bipyridine (mbpy-naph), 4'-methyl-4-(anthracen-2-yl)-2,2'-bipyridine (mbpy-anth), 4'-methyl-yl)-2,2'-bipyridine (mbpy-pyr). The compounds were synthesized from the condensation reactions of the precursor 4'-methyl-2,2'-bipyridine-4- carbaldehyde (mbpy-CHO) with the aromatic amines 2-naphthylamine (naph), 2- aminoanthracene (anth) and 1 -aminopyrene (pyr). Vibrational Spectroscopy in the Infrared Region and Nuclear Magnetic Resonance (NMR) data indicates that the aimed compounds were obtained with high purity. Thus, the work started an extensive description of the spectroscopic properties of theese compounds obtained by electronic absorption and emission spectroscopy in the ultraviolet and visible region. Also, additional data calculated using the Density Functional Theory (DFT) and Time Dependent Density Functional Theory (TD-DFT) were used to support the discussion. The UV-Vis absorption spectra shows that the energy bands are dependent of the attached group and the solvent indicating that an intramolecular charge transfer process occur within the molecule. As bigger is the substituent at the pyridines, more intense is the red shift observed for the absorption maximum, which is according to the frontier orbital energy levels energy gap calculated by TD-DFT: mbpy-naph (3.65 eV), mbpy-anth (3,.13 eV) e mbpy-pyr (3.01 eV). Emission quantum yield (Φ) values were calculated in ethanol using anthracene as reference. In comparison to the substituent aromatic, the mbpy-R compounds showed relatively low values of Φ: 0.001, 0.010 and 0.089 for R = naph, anth and pyr, respectively. Life-time values (τ) of the excited state were determined through emission decay curves in different solvents. The mpy-naph mbpy-anth and mbpy-pyr compounds presented monoexponential decay profiles with values of τ of 1.93, 13.55 and 4.15, respectively. Kinetic radioactive and non-radioactive constants for theese compounds suggests that its excited states decays preferencially by non-radioactive paths, that is accordance with the low singlet oxygen quantum yields determined for the series. The photophysical quenching of theese processes may occur predominant by vibrational deactivation, once the molecules has rotation axes which allows this type of energy loss. CNPq O presente trabalho aborda os resultados obtidos para três novos derivados de bipiridina (mbpyR), a saber: 4’-metil-4-(naft-2-il)-2,2’-bipiridina (mbpy-naph), 4’-metil-4-(antracen-2-il)- 2,2’-bipiridina (mbpy-anth), 4’-metil-4-(piren-2-il)-2,2’-bipiridina (mbpy-pyr). Os compostos citados foram sintetizados a partir de reações de condensação do precursor 4’ -metil-2,2’-bipiridina-4-carbaldeído (mbpy-CHO) com as aminas aromáticas 2-naftilamina (naph), 2-aminoantraceno (anth) e 1 -aminopireno (pyr). Dados de Espectroscopia Vibracional na Região do Infravermelho e Ressonância Magnética Nuclear de 1H e 13C indicam que os produtos desejados foram obtidos com alta pureza. Desse modo, o trabalho seguiu com a descrição extensiva das propriedades espectroscópicas desses compostos analisadas por espectroscopia de absorção eletônica e emissão no ultravioleta e visível (UV-Vis), onde informações adicionais obtidas usando cálculos de Teoria da Densidade Funcional independente (DFT) e dependente do tempo (TD-DFT) foram também utilizados para suportar a discussão. Os espectros de absorção no UV-Vis mostraram uma dependência da energia das bandas com a natureza do fragmento aromático substituinte e com o solvente indicando a existência de processos de transferência de carga intrarmoleculares. Aumentando-se o tamanho do grupo substituinte, observou-se um deslocamento batocrômico consistente com os valores de diferença de energia entre os orbitais de fronteira calculados por TD-DFT: mbpy-naph (3,65 eV), mbpy-anth (3,13 eV) e mbpy-pyr (3,01 eV). Valores de rendimento quântico de emissão (Φ) foram calculados em etanol usando o composto antraceno como referência. Comparativamente às aminas aromáticas, os compostos mbpy-R apresentaram valores relativamente baixos de Φ: 0,001, 0,010 e 0,089 para R = naph, anth e pyr, respectivamente. Os valores de tempos de vida (τ) do estado excitado foram determinados através das curvas de decaimento de emissão em diferentes solventes. Os compostos mbpy-naph, mbpy-anth e mbpy-pyr apresentaram perfis de decaimento monoexponenciais com valores de τ de 1,93, 13,55 e 4,15, respectivamente. As contantes de decaimento radioativo (kr) e não radioativo (knr) indicam que o estado excitado desses compostos decai preferencialmente via rotas não radioativas, o que corrobora com o baixo rendimento quântico de geração de oxigênio singleto para a série. O quenching fotofísico desses processos provavelmente se dá por vias de desativação vibracional, uma vez que a molécula possui eixos de rotação que permitem a perda de energia.

Coordination compounds have been widely used as model systems in attempts for elucidating and/or understanding the mechanism of a myriad of processes including in vivo metabolism and oxygen conversion for the generation of energy or reactive oxygen species (ROS). It is well-known that copper ions are frequently found in enzymes that oxidize organic molecules and take part in interconversion processes of oxygen to water. To study the mechanisms involved in such processes, longstanding efforts have been made for elucidating the geometries and electronic structures of intermediates to unravel the relationships among these aspects and the catalytic functions. A successful strategy that have been adopted by inorganic chemists lies on the synthesis, characterization and reactivity study of copper containing molecules that mimic the enzymatic activity in vitro. In this context, this work presents the results obtained for two novel Schiff bases, 1,3-bis(1,10-phenantrolyn-2-yloxy)-N-(4- (methylhtio)benzylidene)propan-2-amine (2CP-Bz-SMe) and (E)-1-(4'-methil-[2,2'- bipyridine]-4-yl)-N-(4(methylthio)phenyl) metanimine (4-mbipy-Bz-SMe), and for the copper complexes (states I and II) produced from the reaction of these bases with Cu(NO3)2. The isolated compounds were characterized by nuclear magnetic resonance (Schiff bases), vibrational and electronic (room temperature and 90 oC)spectroscopies, electrochemical techniques, and electron paramagnetic resonance (EPR). The thioether moiety of the Schiff bases allowed the immobilization of the copper compounds on gold electrodes and on gold-coated Fe3O4 (Fe3O4@Au) nanoparticles (core@shell structures, Fe3O4@Au@Cu) so that the reactivity of these compounds was also evaluated on surface. To reach such goal, surface techniques were used, citing, beside the conventional electrochemical techniques, surface plasmon resonance (SPR), scanning electrochemical microscopy (SECM), surface-enhanced Raman scattering (SERS), and electrochemical impedance. The characterization of the nanomaterials, in turn, was performed by means of transmission electron microscopy (TEM), X-rays diffraction (XRD), and vibrating sample magnetometry (VSM). All the isolated compounds presented nuclease activity towards plasmid DNA, either in solution or adsorbed on electrodes or nanoparticles. According to EPR and SECM data, this activity was assigned to the generation of hydroxyl radicals ( •OH). The measurements performed to elucidate the mechanism involved in the production of this species showed the formation of H2O2 in a Fenton-like reaction, and a bimetallic intermediate containing peroxo as bridge ligand, {[CuII(4-mbpy-Bz-SMe)2]2(µ-O2 2-)}2+ . Compostos de coordenação têm sido amplamente empregados como sistemas modelos na tentativa de elucidação e/ou entendimento do mecanismo de inúmeros processos incluindo metabolismo in vivo e conversão de oxigênio para geração de energia ou espécies reativas de oxigênio (ROS). Sabidamente, íons cobre são frequentemente encontrados em enzimas que oxidam moléculas orgânicas e participam de processos de interconversão de oxigênio molecular à água. Para estudar os mecanismos envolvidos nesses processos, vem de longas datas as proposições de geometrias e estruturas eletrônicas dos intermediários a fim de desvendar as relações entre esses aspectos e as funções catalíticas. Uma estratégia que tem sido usada com relativo sucesso pelos químicos inorgânicos reside na síntese, caracterização e estudos de reatividade de moléculas que contêm cobre e que mimetizam a atividade enzimática in vitro. Nesse contexto, o presente trabalho apresenta os resultados obtidos para duas bases de Schiff inéditas, 1,3-bis(1,10-fenantrolin-2-iloxi)-N-(4-(metiltio)benzilideno)propan-2-amina (2CP-Bz-SMe) e (E)-1-(4'-metil-[2,2'-bipiridina]-4-il)-N-(4(metiltio)fenil)metanimina (4- mbipy-Bz-SMe), e para os complexos de cobre (estados I e II) produzidos a partir das reações destas bases com o sal Cu(NO3)2. Os compostos isolados foram caracterizados por ressonância magnética nuclear (bases de Schiff), espectroscopias vibracionais e eletrônica (em temperatura ambiente e a 90 oC), técnicas eletroquímicas, e ressonância paramagnética eletrônica (EPR). O fragmento tioéter das bases de Schiff permitiu a imobilização dos compostos de cobre sobre eletrodos de ouro e nanopartículas magnéticas de Fe3O4 recobertas com ouro (estruturas do tipo caroçocasca, Fe3O4@Au@Cu) de modo que a reatividade de tais compostos foi avaliada, também, em superfície. Para esse fim, foram utilizadas técnicas de superfície, podendose citar, além das técnicas eletroquímicas convencionais, ressonância de plásmons de superfície (SPR), microscopia de varredura eletroquímica (SECM), espectroscopias Raman intensificada em superfície (SERS) e de impedância eletroquímica. Já a caracterização dos nanomateriais Fe3O4@Au@Cu foi realizada por microscopia eletrônica de transmissão (TEM), difração de raios-X (XRD) e magnetometria de amostra vibrante (VSM). Todos os compostos isolados apresentaram atividade nuclease em relação a DNA plasmidial, tanto em solução como adsorvidos sobre eletrodos ou nanopartículas. Tal atividade, de acordo com os resultados de EPR e SECM, foi atribuída à geração de radicais hidroxil (•OH). Os experimentos para elucidação do mecanismo envolvido na geração dessa espécie mostraram que há formação de H2O2 em uma reação do tipo Fenton, além de um intermediário bimetálico contendo peroxo como ligante ponte, {[CuII(4-mbpy-Bz-SMe)2]2(µ-O2 2- )}2+ .

Heme-based sensors are a family of proteins that has caught great interest of the scientific community due to their role in several important biological processes. Mycobacterium tuberculosis (Mtb) is the pathogen responsible for causing tuberculosis, currently found in one quarter of the world population in its latent form. This bacterium presents some heme-based sensor proteins including the protein DevS which, together with its response regulator (DevR), are responsible for the entrance of this bacterium into a persistent state of non-replication, known as latency, which is difficult to eliminate. Currently, there is a conflict in the literature regarding the physiological function of the hemeprotein DevS, being suggested by certain groups to work as an oxygen sensor while others indicated as a redox sensor. Here, we present the results of heterologous expression and purification of Mycobacterium tuberculosis DevS protein along with the measurements of the redox potential, Em, at different conditions, such as absence and presence of ligands (CN-, imidazole, CO and NO), different salt concentrations (100, 200 and 500 mmol L-1 NaCl), and pH (6 to 10). The calculated redox potentials were of +11, +96, +392, -147 and -327 mV vs Standard Hydrogen Electrode (SHE) for DevS in the absence of ligands, and bound to CO, NO, imidazole and CN-, respectively. In addition to that, it was estimated the range of redox potential for DevS bound to O2, from +10 to +195 mV. Nonetheless, dissociation constants (Kd) for the imidazole bound to DevS were also calculated by chemical titration, where Kd = 112 x 10-3 and 300 x 10-6 were found for the reduced and oxidized forms of the protein, respectively. Our results indicated that the redox potential of DevS redox potentials are increasingly positive as the ligand strength is intensified, as predicted by the Ligand Field Theory. Furthermore, the redox potential for unbound DevS was shown to be significantly higher than those estimated for Mycobacterium tuberculosis cytosol being consistent, therefore, with the assignment of oxygen sensor function and discarding, thermodynamically, the role as a redox sensor. Hemeproteínas sensoras são uma classe de proteínas que vem despertando grande interesse da comunidade científica, estando envolvidas em diversos importantes processos. O microorganismo Mycobacterium tuberculosis (Mtb) é o agente patógeno responsável por causar a doença conhecida como tuberculose, atualmente presente em um terço da população mundial na forma latente. Esta bactéria apresenta algumas hemeproteínas sensoras, dentre as quais a proteína DevS que, juntamente com seu regulador de resposta (DevR), são responsáveis pela entrada desta bactéria em estado persistente de não replicação dentro do hospedeiro, conhecido como estado de latência, difícil de ser eliminada. Atualmente, existe um conflito na literatura com respeito à função fisiológica da hemeproteína DevS, sendo sugerido por certos grupos ser um sensor de oxigênio enquanto outros acreditam ser um sensor redox. Este trabalho tem como objetivo determinar o papel sensorial da proteína DevS por espectroletroquímica associando as técnicas de eletrólise a potencial controlado com espectroscopia de absorção eletrônica permitindo a determinação do potencial de ponto médio, Em. Esse estudo foi realizado em diferentes condições: na ausência e presença de ligantes (íon cianeto, imidazol, monóxido de carbono e óxido nítrico); em diferentes concentrações salinas: 100, 200 e 500 mmol L-1 NaCl; e em soluções de diferentes valores de pH (6 a 10). Os espectros, obtidos na presença de diferentes mediadores redox, permitiram o cálculo dos seguintes valores de Em: +11, +96, +392, ‒147 e ‒327 mV vs Eletrodo Padrão de Hidrogênio para a proteína DevS na ausência de ligantes e ligada a CO, NO, imidazol e CN−, respectivamente. Adicionalmente, pôde-se estimar a faixa de potencial referente à proteína ligada a O2, entre +10 e +195 mV. Utilizando-se titulação química, foi medida a constante de dissociação (Kd) da proteína ligada a imidazol; Kd = 112 x 10-3 e 300 x 10-6 para as formas reduzida e oxidada, respectivamente. Nossos resultados indicaram que o potencial redox da proteína DevS torna-se mais positivo com o aumento da força dos ligante, tal como previsto pela Teoria de Campo Ligante. Adicionalmente, o valor de Em determinado para a proteína DevS não ligada é significativamente mais elevado que aqueles estimados para o citosol do Mycobacterium tuberculosis sendo consistente, portanto, com a função sensora de oxigênio e descartando, termodinamicamente, a possibilidade de atuar como sensor redox.

In this work, theoretical, spectroscopic and electrochemical studies are shown aiming to understand the structural and electronic changes of the metallic surfaces induced by the formation of Self-Assembled Monolayers (SAMs) produced with 1’-biphenyl-4-thiol (HSBPH, μ ⃗= 1.13), 4′-mercapto-[1,1′- biphenyl]-4-carbonitrile (HSBPCN, μ ⃗=5,3 Debye) and 4,4′-dimercaptobiphenyl (HS2BP, μ ⃗= 0,46) on copper, silver and gold. Such surfaces, spontaneously modified upon immersion in the modifier solutions, showed maxima values of surface coverage fraction () of 0.96 (HSBPH), 0.99 (HSBPCN) and 0.98 (HS2BP) after 1h of modification at 20 oC. The values of of the SAMs as well as those of the heterogeneous charge transfer rate constants (kapp) of the redox probe molecule [Fe(CN)6]3/4, were determined by impedance and shown to be sensitive to the modification time. Considering the maximum of surface coverage fraction, the values of kapp were calculated as 6.8 x105, 1.3 x105 and 9.4 x106 cm s1 for the SAMs of HSBPH, HSBPCN and HS2BP on gold, respectively. The impedimetric responses in KF solution without probe molecules, indicated an increase of the surface coverage up to 5.3x1011 mol cm2 (HSBPH), 1.8x1011 mol cm2 (HSBPCN) and 1.4x1010 mol cm-2 (HS2BP). For the SAM of HSBPCN on gold, the voltammetric results within the double layer showed a meaningful decrease of the differential capacitance (8.9 F cm2) in comparison to the bare gold (35.3 F cm2). The SERS in situ spectra obtained for the SAM of HSBPCN on Cu, Ag and Au indicated the existence of a chemical mechanism of charge transfer between the frontier orbitals HOMO and LUMO of the interfacial complexes, with the transition going from the metal to the molecule for Cu and Ag, and in the opposite direction, from the molecule to the metal, for the Au substrate. According to the computational calculations, the LUMO orbitals of the interfacial complexes are composed by 44, ~3 and 60% of the ns atomic orbitals of Cu, Ag and Au, respectively. In comparison to Cu and Ag, the largest contribution of the ns atomic orbital of Au for the LUMO orbital of the complex is assigned to the relativistic effect that decrease the energy of the atomic orbital making it closer to the resulting molecular orbitals. The charge transfer transition HSBPCN  Au is, thus, justified since this process occurs between the frontier orbitals. With no exciting radiation, however, the polarity of the molecules within the SAM, established by the terminal group (electron withdrawing in the case of HSBPCN SAM), determines the interfacial properties. Indeed, based on the measurements of scanning electrochemical microscopy, the values of the rate constants of the heterogeneous charge transfer reactions (k0) indicate faster processes for the neutral (FcMeOH, k0 = 1.3 cm s1) and cationic ([Ru(NH3)6]3+, k0 > 3 cm s1) species, comparatively to the negatively charged mediators [Fe(CN)6]4- and [Fe(CN)6]3- (k0 ~1,2x104 cm s1). This result indicates that the interfacial polarity seems to be preponderant in the studied systems, taking apart from the interface the negatively charged complexes and approaching the cationic complex. This behavior suggests that the heterogeneous electron transfer reaction at the interface of HSBPCN SAM follows a nonadiabatic pathway. The applicability of the SAMs of HSBPCN, HSBPH e HS2BP on gold was evaluated in relation to the recognition/detection of D-mannose sugar after immobilization of the lectin ConBr. The voltammetric and impedimetric data, obtained in electrolytic medium containing [Fe(CN)6]3-/4-, showed an increase of the faradaic current assigned to the FeIII/II redox pair of the iron complex with the increase of the sugar concentration. Two hypotheses were considered to explain the voltammetric profiles: (1) existence of micropores and (2) interfacial dipole. The existence of micropores resulting from the possible leaching of the immobilized ConBr molecules on the SAMs implies lower resistance to the heterogeneous charge transfer reaction due to the easy access to the underlying gold surface. The role of the interfacial dipole generated by the polar species adsorbed on the substrate, in turn, justifies, more effectively, the current response associated with the heterogeneous charge transfer reaction due to the repulsive effects at the interface. Neste trabalho, são apresentados estudos teóricos, espectroscópicos e eletroquímicos com o objetivo de entender as mudanças estruturais e eletrônicas de superfícies metálicas induzidas pela formação das SAMs (Self-Assembled Monolayers) formadas com os compostos – 1,1’-bifenil-4-tiol (HSBPH, μ ⃗= 1,13), 4′-mercapto-[1,1′-bifenil]-4-carbonitrila (HSBPCN, μ ⃗=5,3 Debye) e 4,4′-dimercaptobifenil (HS2BP, μ ⃗= 0,46) – sobre cobre, prata e ouro. Tais superfícies, espontaneamente modificadas em decorrência da imersão em solução dos modificadores, apresentaram valores máximos de fração de recobrimento () de 0,96 (HSBPH), 0,99 (HSBPCN) e 0,98 (HS2BP) após 1h de imersão a 20 oC. Os valores de das SAMs, assim como das constantes de velocidade heterogênea de transferência de carga (kapp) da molécula de prova redox [Fe(CN)6]3/4, foram determinados por impedância e mostraram-se sensíveis ao tempo de modificação. Considerando o máximo de fração de recobrimento, os valores de kapp foram calculadas em 6,8 x105, 1,3 x105 e 9,4 x106 cm s1 para as SAMs dos compostos HSBPH, HSBPCN e HS2BP sobre ouro, respectivamente. As respostas impedimétricas em solução de KF na ausência de moléculas de prova, indicaram um aumento do recobrimento superficial das SAMs até o valor limite de 5,3x1011 mol cm2 (HSBPH), 1,8x1011 mol cm2 (HSBPCN) e 1,4x1010 mol cm-2 (HS2BP). Para a SAM de HSBPCN sobre ouro, os resultados voltamétricos na região de dupla camada, apresentaram uma significativa diminuição da capacitância diferencial (8,9 F cm2), comparativamente ao eletrodo de ouro limpo (35,3 F cm2). Os espectros SERS in situ obtidos para a SAM de HSBPCN sobre os substratos de Cu, Ag e Au indicaram a existência de um mecanismo químico de transferência de carga entre os orbitais de fronteira HOMO e LUMO dos complexos interfaciais, com a transição ocorrendo do metal para a molécula no caso dos eletrodos de Cu e Ag, e no sentido inverso, da molécula para o metal, no caso do substrato de Au. De acordo com os cálculos computacionais, os orbitais LUMO dos complexos interfaciais têm contribuição de 44, ~3 e 60% dos orbitais ns dos átomos de Cu, Ag e Au, respectivamente. Comparativamente aos átomos de Cu e Ag, a maior contribuição dos orbitais atômicos do ouro na formação do orbital LUMO do complexo se deve ao efeito relativístico que diminui a energia dos orbitais atômicos tornando-a mais próxima da energia dos orbitais moleculares resultantes. Justifica-se, assim, a transição de transferência de carga HSBPCN  Au, uma vez que esta transição ocorre entre os orbitais de fronteira. Na ausência de radiação incidente, todavia, a polaridade das moléculas constituintes da SAM, definida pelo grupo terminal (retirador de elétrons no caso da SAM de HSBPCN), determina as propriedades interfaciais. De fato, baseando-se nos experimentos de microscopia de varredura eletroquímica, os valores das constantes de velocidade das reações heterogêneas de transferência de carga (k0) indicam processos mais rápidos para a espécie neutra (FcMeOH, k0 = 1,3 cm s1) e catiônica ([Ru(NH3)6]3+, k0 > 3 cm s1), comparativamente aos mediadores [Fe(CN)6]4- e [Fe(CN)6]3- (k0 ~1,2x104 cm s1), negativamente carregados. Esse resultado indica que a polaridade interfacial parece ser preponderante nos sistemas estudados, isto é, afasta da interface os complexos negativamente carregados e aproxima aquele de carga positiva ou neutra. Esse comportamento sugere que a reação de transferência de elétrons na interface da SAM de HSBPCN sobre ouro segue um mecanismo não-adiabático. A aplicação das SAMs das moléculas de HSBPCN, HSBPH e HS2BP sobre ouro foi avaliada frente à capacidade de reconhecimento/detecção do açúcar D-manose após imobilização da lectina ConBr. Os resultados voltamétricos e impedimétricos, obtidos em meio eletrolítico contendo o íon [Fe(CN)6]3-/4-, mostraram aumento de corrente faradáica atribuída ao par redox FeIII/II do complexo de ferro com o aumento da concentração de açúcar no meio. Duas hipóteses foram consideradas para a explicação dos perfis voltamétricos: (1) existência de microporos e (2) dipolo interfacial. A existência de microporos resultante da possível lixiviação das moléculas de ConBr imobilizadas sobre as SAMs implica em menor resistência à reação heterogênea de transferência de carga devido à facilidade de acesso à superfície de ouro. O papel do dipolo interfacial gerado por uma espécie polar adsorvida no substrato, por sua vez, justifica, de forma mais efetiva, a resposta de corrente associada à reação heterogênea de transferência de carga devido aos efeitos repulsivos na interface.

This work presents the expression, purification and spectroelectrochemistry results obtained for the sensor hemeprotein FixL from Rhizobium etli (ReFixL). In addition, the spectroelectrochemistry data allowed the correlation between the potential values of the redox reaction of the Fe metal center presented in the heme group with the kinase activity of the ReFixL protein. The ReFixL protein, expressed and purified in its met-ReFixL(FeIII) form, showed molecular mass value of about 70 kDa (ReFixL). The values of the redox potentials of the studied protein was determined by spectroelectrochemical titration in buffer solution (pH 8 or 9.5) containing redox mediators that allowed to cover a potential range from 500 to + 500 mV vs ENH. Such measurements were performed for the proteins free of coordination and bonded (coordination to the Fe atom of the heme group) to oxygen (O2), carbon monoxide (CO), cyanide (CN) and imidazole molecules; all of them relevant in the activation/inactivation kinase activity in the case of ReFixL. The spectroscopic profiles of the ReFixL protein supported these species were bound to CO and O2 in the reduced state and to imidazole and CN in the oxidized state, being observed, respectively, the following potential values: +21, 51, 57 and 156 mV vs ENH. The inactivation of the kinase activity of the ReFixL protein is observed upon binding to O2, imidazole and CN− molecules, whose redox potentials show negative values. The negative potential shift in relation to the reduced non-bonded state of the protein (deoxy-ReFixL; +19 mV vs ENH) indicates the reduction potential is more affected by conformational changes induced by the ligands than the electron density donor/acceptor character of the ligands do. The potential values observed for Fe-O2 do not follow the expected based on its thermodynamics, assuming the expected potential shift would have been positive for ligands that bind to reduced state FeII. Our results suggest that conformational changes that follow due to kinase activity switching off upon O2 binding have some knock-on effect on the heme environment, that destabilize the FeII state and conterbalances the FeII stabilization normally caused by a ligand to the reduced form. Este trabalho apresenta os resultados de expressão, purificação e espectroeletroquímica obtidos para a hemeproteína sensora FixL do organismo Rhizobium etli (ReFixL). Além disso, os dados de espectroeletroquímica permitiram a correlação entre os valores de potenciais da reação redox do centro metálico de Fe presente no grupo heme com a atividade quinase da proteína ReFixL. A proteína ReFixL, expressa e purificada em sua forma met-ReFixL(FeIII), apresenta valor de massa aproximada de 70 kDa (ReFixL). Os valores dos potenciais redox das proteínas em estudo foram determinados por titulação espectroeletroquímica em solução tampão (pH 8 ou 9,5) contendo mediadores redox que permitiram uma variação de potencial de −500 a + 500 mV vs ENH. Tais experimentos foram realizados para a proteína livres de coordenação e quando ligada (coordenação ao átomo de Fe do grupo heme) às moléculas de oxigênio (O2), monóxido de carbono (CO), cianeto (CN−) e imidazol; todas relevantes no processo de ativação/inativação da atividade quinase da proteína ReFixL. Os perfis espectroscópicos da proteína ReFixL indicam que esta se liga a CO e O2 em seu estado reduzido e à imidazol e CN− no seu estado oxidado, sendo observados, respectivamente, os seguintes valores de potencial: +21, −51, −57 e −156 mV vs ENH. A desativação da atividade quinase da proteína ReFixL é observada quando da ligação com as moléculas de O2, imidazol e CN−, cujos processos redox apresentam valores negativos de potencial. O deslocamento negativo de potencial em relação à proteína no estado reduzido e não ligado (deoxi-ReFixL; +19 mV vs ENH) indica que as mudanças conformacionais induzidas pelos ligantes afeta o potencial de redução de forma mais significativa que o caráter doador ou receptor de densidade eletrônica dos ligantes. Os valores de potencial observados para a proteína ligada ao ligante O2 não corresponde ao esperado baseado na termodinâmica da reação, uma vez que se esperava um deslocamento positivo de potencial para ligantes que se ligam ao estado reduzido da proteína. Os resultados sugerem que as mudanças conformacionais que seguem devido a inativação da atividade quinase quando a proteína se liga a O2, tem um efeito no ambiente do grupo heme que de alguma forma desestabiliza o estado reduzido da proteína, contrabaleanceando a estabilização normalmente causada por um ligante ao estado reduzido.

The effect of the venom of the Crotalus basiliscus snake on the electron transfer (ET) reaction of the myoglobin (Mb) protein was studied using a gold electrode modified with cysteine (Au/Cys) followed by the immobilization of the protein (Au/Cys/Mb). For that, the following techniques were used: cyclic voltammetry, surface plasmon resonance (SPR), absorption spectroscopy in the ultraviolet and visible regions (UV-Vis), circular dicroism (CD), and gel electrophoresis. The electrode reaction assigned to the FeII/III redox pair of Mb was observed at 0.09V vs Ag/AgCl being consistent with the native form of the protein. Upon immersion of the Au/Cys/Mb electrode in solution containing the venom of the Crotalus basiliscus snake, it was observed a decrease in the faradaic current. The deleterious effect of the venom on the electrode reaction of Mb was corroborated by the observation of the loss of the catalytic activity of the Au/Cys/Mb electrode towards the oxidation reaction of the ascorbic acid (AA). The successive mass increments observed by SPR upon the injections of cysteine (0.172 µg cm-2), myoglobin (0.410 µg cm-2) and venom (0.151 µg cm-2) indicated that the adsorbed molecules of cysteine and myoglobin were not desorbed from the gold surface upon exposure to the solution of the venom of the Crotalus basiliscus snake. Rather, the mass increase indicated the molecules of the venom were immobilized on the the Au/Cys/Mb electrode. Such proposition was reinforced by the electrophoresis results which showed a decrease in the mobility of Mb after mixing to the solution containing the snake venom. The electronic UV-Vis spectra did not show meaningful changes in the Soret band of Mb; a band sensitive to the level of protein denaturation. Likewise, the CD spectra obtained in the far-UV, near-UV and Soret regions did not present spectral profile changes indicating that the physical integrity of the protein was maintained after the exposure to the snake venom, i.e. the protein was not denaturated. O efeito do veneno da cobra Crotalus basiliscus sobre a reação de transferência de elétrons (TE) da proteína mioglobina (Mb) foi estudado utilizando-se um eletrodo de ouro modificado com cisteína (Au/Cys) seguido da imobilização da proteína (Au/Cys/Mb). Para tanto, foram utilizadas as técnicas de voltametria cíclica, ressonância de plásmons de superfície (SPR-Surface Plasmon Resonance), espectroscopia de absorção na região do ultravioleta e visível (UV-Vis), espectroscopia de dicroísmo circular (CD-Circular Dichroism) e eletroforese em gel desnaturante. A reação de eletrodo atribuída ao par redox FeII/III da Mb foi observada em 0,09V vs Ag/AgCl sendo consistente com a forma nativa da proteína. Após imersão do eletrodo Au/Cys/Mb em solução contendo o veneno da cobra Crotalus basiliscus, observou-se a diminuição da corrente. O efeito nocivo do veneno sobre a reação de eletrodo da Mb foi corroborado com a observação da perda de atividade catalítica do eletrodo Au/Cys/Mb frente a reação de oxidação do ácido ascórbico (AA). Os aumentos sucessivos de massa observados por SPR quando das injeções de cisteína (0,172 µg cm-2), mioglobina (0,410 µg cm-2) e veneno (0,151 µg cm-2) indicaram que as moléculas adsorvidas de cisteína e mioglobina não experimentam desorção após exposição ao veneno da cobra Crotalus basiliscus. Ao contrário, o aumento de massa indicou que moléculas do veneno foram imobilizadas sobre o eletrodo Au/Cys/Mb. Tal proposição foi reforçada pelos resultados de eletroforese que apresentaram uma diminuição na mobilidade da Mb no gel após a mistura com a solução contendo veneno da cobra Crotalus basiliscus. Os espectros eletrônicos UV-Vis não apresentaram mudança significativa na banda Soret da Mb; banda esta sensível ao grau de desnaturação da proteína. Da mesma forma, os espectros de Dicroísmo Circular obtidos nas regiões do UV-distante, UV-próximo e Soret não apresentaram mudanças no perfil espectral da proteína, indicando que a integridade física da Mb é mantida após exposição ao veneno, ou seja, a proteína não foi desnaturada.

Coordination compounds of copper have been invoked as major actors in processes involving the reduction of molecular oxygen, mostly with the generation of radical species whose assignment have so far not been unanimously addressed. In the present work we have joined results acquired on surface and solution to get insights on the radical oxygen species (ROS) generated by a copper(II) coordination compound containing a thioether clip-phen derivative, 1,3-bis(1,10-phenanthrolin-2-yloxy)-N-(4-(methylthio) benzyli-dene)propan-2-amine (2CP-Bz-SMe). The adsorption of the copper(II) complex on gold surface was spontaneously by simple immersion of the metallic substrate in aqueous solution of the complex. The characterization of the modified gold electrode, which was performed by electrochemistry and surface Raman spectroscopy (SERS – Surface Enhanced Raman Scattering), indicated that the adsorption occurs through the sulfur atom of the thioether fragment of the molecule. While surface plasmon resonance (SPR) and electrochemistry of the adsorbed complex indicated the formation of a dimeric Cu(I) intermediate containing molecular oxygen as bridging ligand, scanning electrochemical microscopy images (SECM) pointed for OH• radical generation. Spin trapping measurements acquired by electron paramagnetic resonance (EPR) and nuclease assays run in the presence of radical scavengers, reinforced such conclusions showing that the radical production is dependent on the amount of oxygen and H2O2. Therefore, it is reasonable to assume a catalytic mechanism involving Fenton-like reaction in which the copper(II) complex is, at first, reduced to copper(I). Accordingly, in the presence of oxygen and in acid medium, the reduced compound is oxidized by H2O2 resulting in the recovering of the parent complex and in the generation of OH• that, in turn, must be the ROS responsible for the DNA cleavage. The results obtained during the developing of this proposal were presented at the XVII Brazilian Meeting on Inorganic Chemistry and are in the final compilation stage for publication in peer-reviewed journal Compostos de coordenação de cobre têm sido considerados peças fundamentais em processos que envolvem a redução de oxigênio molecular, particularmente com a geração de espécies radicalares cujas atribuições, até a atualidade, não são unanimidade. Neste trabalho, foram reunidos resultados obtidos em superfície e em solução a fim de obter informações sobre a espécie reativa de oxigênio (ROS – Reactive Oxygen Species) gerada por um composto de coordenação de cobre(II) contendo um derivado tioéter do ligante clip-phen, 1,3-bis(1,10-fenantrolin-2-iloxi)-N-(4-(metiltio)benzilideno)propan-2-amina (2CP-Bz-SMe). A adsorção do complexo de cobre(II) sobre superfície de ouro foi realizada de forma espontânea por simples imersão do substrato metálico em solução aquosa do complexo. A caracterização do eletrodo de ouro modificado foi feita por eletroquímica e espectroscopia Raman de superfície (SERS – Surface Enhanced Raman Scattering) onde se concluiu que a adsorção ocorre através do átomo de enxofre do fragmento tioéter da molécula. Enquanto os resultados de ressonância de plásmons de superfície (SPR – Surface Plasmon Resonance) e eletroquímica sugeriram a formação de um intermediário dimérico de Cu(I) contendo oxigênio molecular como ligante ponte, imagens de microscopia de varredura eletroquímica (SECM – Scanning Electrochemical Microscopy) indicaram a geração do radical hidroxil, OH•. Os experimentos de captura de spin por ressonância paramagnética de elétrons (EPR – Electron Paramagnetic Resonance) e ensaios de nuclease de DNA, os quais envolveram a utilização de inibidores de ROS, mostram que a geração do radical depende da quantidade de oxigênio e H2O2. Assim, é razoável admitir um mecanismo catalítico envolvendo reações do tipo Fenton onde o complexo de cobre(II) é, inicialmente, reduzido a cobre(I). Na presença de oxigênio e em meio ácido, o composto reduzido é oxidado por H2O2 resultando na regeneração do complexo de partida e na formação do radical OH• que, por sua vez, deve ser a espécie ROS responsável pela clivagem do DNA. FRANCISCO, Thiago dos Santos. Complexos Fosfínicos de Rutênio: Síntese, Caracterização, Imobilização e Atividade Catalítica. 2015. 146 f. The results presented in this thesis involve the synthesis of coordination compounds containing phosphine ligands and redox active (non-innocent ligands) in the ruthenium coordination sphere. This work is part of a research line of Bioinorganic Group research aimed, as the ultimate goal, the application of the isolated compounds in catalysis of hydrogenation reactions. During the development of PhD activities, from March 2010 to January 2015, six compounds (06) unpublished in the literature were synthesized; one of which being classified as a precursor complex, [RuIICl2(binap)(PPh3)], where binap = 2,2′-bis(diphenylphosphine)-1,1′-dinaphthalene. All of these complexes were characterized by means of phosphorous nuclear resonance magnetic (RMN 31P{1H}), elemental analysis, infrared spectroscopy and electronic spectroscopy in the ultraviolet and visible regions. Despite several attempts to produce crystals of the synthesized compounds, only the trans-[RuIICl2(dppb)(opda-Clcat)] complex, where dppb = 1,4-bis(diphenylphosphine)butane and opda-Clcat = 4-cloro-1,2-phenylenediamine, yielded suitable crystals for determination of X-ray structure. From the results obtained during the cited time, two papers were published in indexed journals ((Polyhedron 2010, 29, 3349–3354 and Polyhedron 2012, 31, 104–109) and five works were shown in national and international meetings. The RMN 31P{1H} spectra of the isolated compounds presented only one signal indicating that the phosphorous atoms lie in equivalent positions and trans in relation to the donor atoms of the non-innocent ligands. For the trans-[RuIICl2(dppb)(opda-Clcat)] complex, however, a dublet of dublet was observed and was assigned to the assimetry of the opda-Cl moiety, which was confirmed by X ray diffraction. The electronic spectroscopic data indicated the oxidized state of the o-phenylene ligands upon coordination to the metal atom thus making possible the back-bonding interaction. The catalytic activity assays were monitored by gas chromatography coupled to mass spectrometry using the standard hydrogenation reaction of the alcohol molecule acetophenone to 1-phenyl ethanol. In addition to the complex originally presented in this thesis, [RuIICl2(binap)(PPh3)](1), trans-[RuIICl2(binap)(opda-Clcat)](2), trans-[RuIICl2(binap)(fenantq)](3), trans-[RuIICl2(PPh3)2(opdaq)](5) and trans-[RuIICl2(PPh3)2(fenantq)](6), where fenantq and opdaq are, respectively, 9,10-phenanthraquinone and 1,2-phenylenediamine, such assays were also carried out for compounds which were previously synthesized during the master course and by other researchers of the Bioinorganic Group resulting resulting in a total of twenty (20) studied catalysts. Among these complexes, two were studied in heterogeneous condition upon immobilization on hybrid spheres of silica chitosan. By accounting for all the studied conditions, the highest values of conversion (> 90%) were observed for trans-[RuIICl2(dppb)(Quinone)](10), cis-[RuIICl2(dppb)(opda-Clq)](13), trans-[RuIICl2(dppb)(tabq,cat)](15) and cis-[RuIICl2(dppb)(tabq,cat)](16), where tabq,cat = 3,3’,4,4’-tetraaminobiphenyls. The values of conversion observed for the heterogeneous systems, c.a. 60%, although lower than those calculated for the homogeneous systems under similar conditions, do not constitute a disadvantage, since separation step is no longer needed. The compounds that showed the higher conversion values in homogeneous medium feature in common the presence of non-innocent ligands in the oxidized state. This observation indicates, very probably, two synergistic effects which influence the affinity of the substrate: (i) the increase of the Lewis acidity of the metal by the oxidation of the ligand and (ii) the function of electron density reservoir of the redox active ligand that allows the metal to store (donate) electron density from (to) the ligand during the elementary steps of the catalytic cycle by preventing excessive charge density on the metal. Os resultados apresentados neste trabalho envolvem a síntese de compostos de coordenação contendo fosfinas e ligantes redox ativos (ligantes não-inocentes) na esfera de coordenação do rutênio. Este trabalho está inserido em uma linha de pesquisa do Grupo de Bioinorgânica que visa, como objetivo final, a aplicação dos compostos isolados na catálise de reações de hidrogenação. Durante o desenvolvimento das atividades de doutorado, período de março de 2010 a janeiro de 2015, foram sintetizados seis (06) compostos inéditos na literatura, um dos quais sendo classificado como complexo precursor, [RuIICl2(binap)(PPh3)], onde binap = 2,2′-bis(difenilfosfina)-1,1′-dinaftaleno. Todos estes compostos foram caracterizados por ressonância magnética nuclear de fósforo (RMN 31P{1H}), análise elementar e espectroscopias vibracional na região do infravermelho e eletrônica nas regiões do ultravioleta e visível. A despeito das diversas tentativas de formação de cristais dos complexos isolados, apenas o composto trans-[RuIICl2(dppb)(opda-Clcat)], onde dppb = 1,4-bis(difenilfosfina)butano e opda-Clcat = 4-cloro-1,2-fenilenodiamina, forneceu cristais passíveis de análise por difração de raios X. A partir dos resultados obtidos durante o período citado, dois trabalhos foram publicados em periódicos indexados (Polyhedron 2010, 29, 3349–3354 e Polyhedron 2012, 31, 104–109) e cinco trabalhos foram apresentados em congressos nacionais e internacionais. Os espectros de RMN 31P{1H} dos compostos sintetizados apresentaram um único sinal indicando que os átomos de fósforo ocupam posições equivalentes e trans aos átomos doadores dos ligantes não-inocentes. Apenas o composto trans-[RuIICl2(dppb)(opda-Clcat)] apresentou um dubleto de dubleto que foi atribuído à assimetria do ligante opda-Cl, sendo confirmado pelos dados de difração de raios X. Os dados de espectroscopia eletrônica indicaram o estado oxidado dos ligantes o-fenilênicos após coordenação ao centro metálico sendo possível, portanto, a observação de interações de retrodoação. Os ensaios de atividade catalítica foram monitorados por cromatografia gasosa acoplada à espectrometria de massa utilizando a reação padrão de hidrogenação da molécula de acetofenona ao álcool 1-feniletanol. Além dos complexos originalmente apresentados nesta tese, [RuIICl2(binap)(PPh3)](1), trans-[RuIICl2(binap)(opda-Clcat)](2), trans-[RuIICl2(binap)(fenantq)](3), trans-[RuIICl2(PPh3)2(opdaq)](5) e trans-[RuIICl2(PPh3)2(fenantq)](6), onde fenantq e opdaq são, respectivamente, 9,10-fenantrequinona e 1,2-fenilenodiamina, tais ensaios foram, também, realizados com compostos previamente sintetizados durante a dissertação de mestrado e por outros membros do Grupo de Bioinorgânica, resultando em um total de vinte (20) catalisadores estudados. Destes complexos, dois foram estudados em condições heterogêneas após imobilização sobre esferas híbridas de sílica/quitosana. Dentre os sistemas estudados, os complexos trans-[RuIICl2(dppb)(Quinone)](10), cis-[RuIICl2(dppb)(opda-Clq)](13), trans-[RuIICl2(dppb)(tabq,cat)](15) e cis-[RuIICl2(dppb)(tabq,cat)](16), onde tabq,cat = 3,3’,4,4’-tetraaminobifenil, apresentaram os mais altos valores de conversão (> 90%) em todas as condições analisadas. Os valores de conversão observados para os sistemas heterogêneos, ~ 60%, embora menores que aqueles calculados para os sistemas homogêneos em condições similares, não representam desvantagem, uma vez que implicam na redução da etapa de separação. Os compostos que apresentaram maiores valores de conversão em meio homogêneo apresentam, em comum, a presença de ligantes não-inocentes no estado oxidado. Essa observação indica a presença de efeitos sinérgicos que influenciam na afinidade do substrato, a citar: (i) aumento da acidez de Lewis do metal pela oxidação do ligante e (ii) função de reservatório de densidade eletrônica do ligante redox ativo que permitir o metal estocar (doar) densidade eletrônica no (para o) ligante durante as etapas elementares do ciclo catalítico, evitando a localização excessiva de carga sobre o metal. The results presented in this thesis involve the synthesis of coordination compounds containing phosphine ligands and redox active (non-innocent ligands) in the ruthenium coordination sphere. This work is part of a research line of Bioinorganic Group research aimed, as the ultimate goal, the application of the isolated compounds in catalysis of hydrogenation reactions. During the development of PhD activities, from March 2010 to January 2015, six compounds (06) unpublished in the literature were synthesized; one of which being classified as a precursor complex, [RuIICl2(binap)(PPh3)], where binap = 2,2′-bis(diphenylphosphine)-1,1′-dinaphthalene. All of these complexes were characterized by means of phosphorous nuclear resonance magnetic (RMN 31P{1H}), elemental analysis, infrared spectroscopy and electronic spectroscopy in the ultraviolet and visible regions. Despite several attempts to produce crystals of the synthesized compounds, only the trans-[RuIICl2(dppb)(opda-Clcat)] complex, where dppb = 1,4-bis(diphenylphosphine)butane and opda-Clcat = 4-cloro-1,2-phenylenediamine, yielded suitable crystals for determination of X-ray structure. From the results obtained during the cited time, two papers were published in indexed journals ((Polyhedron 2010, 29, 3349–3354 and Polyhedron 2012, 31, 104–109) and five works were shown in national and international meetings. The RMN 31P{1H} spectra of the isolated compounds presented only one signal indicating that the phosphorous atoms lie in equivalent positions and trans in relation to the donor atoms of the non-innocent ligands. For the trans-[RuIICl2(dppb)(opda-Clcat)] complex, however, a dublet of dublet was observed and was assigned to the assimetry of the opda-Cl moiety, which was confirmed by X ray diffraction. The electronic spectroscopic data indicated the oxidized state of the o-phenylene ligands upon coordination to the metal atom thus making possible the back-bonding interaction. The catalytic activity assays were monitored by gas chromatography coupled to mass spectrometry using the standard hydrogenation reaction of the alcohol molecule acetophenone to 1-phenyl ethanol. In addition to the complex originally presented in this thesis, [RuIICl2(binap)(PPh3)](1), trans-[RuIICl2(binap)(opda-Clcat)](2), trans-[RuIICl2(binap)(fenantq)](3), trans-[RuIICl2(PPh3)2(opdaq)](5) and trans-[RuIICl2(PPh3)2(fenantq)](6), where fenantq and opdaq are, respectively, 9,10-phenanthraquinone and 1,2-phenylenediamine, such assays were also carried out for compounds which were previously synthesized during the master course and by other researchers of the Bioinorganic Group resulting resulting in a total of twenty (20) studied catalysts. Among these complexes, two were studied in heterogeneous condition upon immobilization on hybrid spheres of silica chitosan. By accounting for all the studied conditions, the highest values of conversion (> 90%) were observed for trans-[RuIICl2(dppb)(Quinone)](10), cis-[RuIICl2(dppb)(opda-Clq)](13), trans-[RuIICl2(dppb)(tabq,cat)](15) and cis-[RuIICl2(dppb)(tabq,cat)](16), where tabq,cat = 3,3’,4,4’-tetraaminobiphenyls. The values of conversion observed for the heterogeneous systems, c.a. 60%, although lower than those calculated for the homogeneous systems under similar conditions, do not constitute a disadvantage, since separation step is no longer needed. The compounds that showed the higher conversion values in homogeneous medium feature in common the presence of non-innocent ligands in the oxidized state. This observation indicates, very probably, two synergistic effects which influence the affinity of the substrate: (i) the increase of the Lewis acidity of the metal by the oxidation of the ligand and (ii) the function of electron density reservoir of the redox active ligand that allows the metal to store (donate) electron density from (to) the ligand during the elementary steps of the catalytic cycle by preventing excessive charge density on the metal.

Electrochemical techniques, quartz crystal microbalance (QCM) and surface plásmons resonance (SPR) were used to study the formation of self-assembled monolayers (SAMs) of 1,4-ditiano (1,4-dt), 4-mercaptopyridine (pyS), 5-(4-piridinyl)-1,3,4-oxadiazole-2-thiol (Hpyt), thionicotinamide (TNA) and thioisonicotinamide (iTNA) as a result of the immersion of gold substrates into the respective solutions. STM (Scanning Tunneling Microscopy) images indicate the sulfur atom as the adsorption site of these molecules and a hexagonal conformation on surface. For the iTNA molecule, the images were not conclusive. In addition, the images indicated the existence of defects even after longer immersion times (24 h). Thermodynamic and kinetic aspects of the adsorption process were evaluated by reductive desorption in alkaline media and QCM. The reductive desorption potentials, Edr, were observed at −0.9, −0.8, and −0.5 V vs. Ag/AgCl for the desorption of iTNA, TNA and Hpyt, respectively. In comparison to 1,4-dt (−0.8 V) and pyS (−0.5 V) species, the Edr value of Hpyt indicates a sigma interaction whereas those of iTNA and TNA indicate an additional pi contribution. The values of the concentration of adsorbed material, gama, and fractional coverage (teta ~ 0.9), determined, respectively, by reductive desorption and impedance are consistent with the STM images. In comparison to the desorption data, the higher values of gama calculated by QCM were assigned to the presence of water molecules since a linear relation was observed between the dipole moment and the mass change calculated by QCM. The desorption curves acquired for the pyS SAM indicated the decomposition of the monolayer thus not allowing the determination of the thermodynamic and kinetic parameters of adsorption. In comparison with the results obtained for the electrode modified after immersion in Na2S solution, it was suggested that this process is associated to the cleavage of the C─S bond which results in the formation of an adlayer composed of atomic and/or oligomeric sulfur species. The adsorption isotherms for Hpyt, TNA and iTNA fitted the Langmuir model of adsorption allowing the determination of the free energy of adsorption, ΔGads, as −35.9, −38.5 and −34,9 kJ/mol, respectively. These values are indicative of strong interaction being typical of chemisorption. For the Frumkin model, the best correlation was found when the interaction parameter, g, was established as −0.45, −0.20 and −0.10 for Hpyt, TNA and iTNA, respectively, indicating repulsive interactions between the adjacent molecules. Cyclic voltammetry was used to determinate the pKa of the SAMs of Hpyt (4.2), TNA (5.0 and 8.5) and iTNA (4.5 and 7.9) by using [Fe(CN)6]3− as a probe molecule. For this study, some changes were suggested since in the method proposed in the literature the existence of more than one protonation site was not considered thus not allowing the determination of more than one pKa value as was observed for TNA and iTNA molecules. Electron transfer reactions (TE) of cytochrome c (cyt c) and myoglobin (Mb) metalloproteins were studied by using the SAMs of Hpyt, TNA, iTNA, pyS and 1,4-dt. For the SAMs formed with TNA and iTNA, the positive shift of 0.2V on the half-wave potential of cyt c in relation to that of the native protein, was assigned to the positive charge density on surface in consequence of the protonation of NH2 groups (pKa~8.0) since these measurements were carried out in physiological medium. QCM and SPR data indicated the formation of a monolayer of cyt c on the studied SAMs. This monolayer, although not being electroactive, allows the study of the TE reaction of the cyt c molecules in solution suggesting that this process involves the orbitals of the modifier molecules. For the Mb metalloprotein, a SAM of L-cysteine amino acid was used since none of the studied sulfur molecules was able to access the TE reaction. The redox process was observed at 0.086 V suggesting the native form of Mb. QCM and SPR data indicated, also, the formation of a monolayer of Mb on the cys SAM (Au/cys/Mb). Values of –49.67 kJ mol−1 and −0.15 for ΔGads and g, respectively, were calculated for the formation of the monolayer of Mb on the cys SAM. The electrode Au/cys/Mb presented catalytic activity toward the oxidation reaction of ascorbic acid presenting a decrease of 400 mV in the overpotential and a kinetic controlled with a rate constant, kf, of 2.0 x 104 L mol-1 s-1. O emprego das técnicas de eletroquímica, microbalança de cristal quartzo (QCM – Quartz Crystal Microbalance) e ressonância de plásmons de superfície (SPR – Surface Plasmon Resonance) mostram que as espécies 1,4-ditiano (1,4-dt), 4-mercaptopiridina (pyS), 5-(4-piridil)-1,3,4-oxadiazol-2-tiol (Hpyt), tionicotinamida (TNA) e isotionicotinamida (iTNA) experimentam adsorção espontânea formando SAMs (Self-Assembled Monolayers) como resultado da imersão de substratos de ouro em solução contendo estas espécies. As imagens obtidas por microscopia de varredura por tunelamento (STM – Scanning Tunneling Microscopy) indicam um arranjo próximo do hexagonal com exceção da iTNA cujas imagens não foram conclusivas. Adicionalmente, as imagens indicam a existência de defeitos nas SAMs mesmo após longos períodos de imersão (24 h). Os estudos termodinâmicos e cinéticos dos processos de adsorção foram realizados por desorção redutiva em meio alcalino e QCM. Os valores dos potenciais de desorção redutiva, Edr, foram observados em −0,9, −0,8 e −0,5 V vs. Ag/AgCl/Cl- para a desorção de iTNA, TNA e Hpyt, respectivamente. Comparativamente às espécies 1,4-dt (−0,8 V) e pyS (−0,5 V), o valor de Edr da SAM de Hpyt indica uma interação sigma ao passo que aqueles observados para iTNA e TNA sugerem uma contribuição pi adicional. Os valores de quantidade de material adsorvido (gama) e da taxa de recobrimento da superfície (teta), calculados por desorção redutiva e impedância eletroquímica, respectivamente, foram consistentes com as imagens de STM. Comparativamente aos resultados de desorção, os maiores valores de gama determinados por QCM foram atribuídos à presença de moléculas de água co-adsorvidas visto que foi observada uma relação linear entre o excesso de massa e o momento de dipolo das espécies modificadoras. As curvas de desorção obtidas para pyS indicam a decomposição da monocamada não possibilitando, portanto, a determinação dos parâmetros termodinâmicos e cinéticos de adsorção. A correlação com os resultados obtidos após imersão do eletrodo de ouro em solução de Na2S sugere que este processo está associado à quebra da ligação C─S com formação de uma camada de enxofre atômico e/ou oligomérico. As isotermas de adsorção obtidas para os processos de formação das SAMs de Hpyt, TNA e iTNA, adequaram-se ao modelo de Langmuir permitindo a determinação da variação da energia livre de adsorção, ΔGads, como −35,9, −38,5 e −34,9 kJ mol−1, respectivamente. Estes valores são indicativos de interação forte sendo característicos de processos de quimissorção. Para o modelo de Frumkin, os dados apresentaram melhores correlações quando o parâmetro de interação (g) foi fixado em −0,45, −0,30 e −0,10, respectivamente, para as SAMs de Hpyt, TNA e iTNA indicando interações repulsivas entre as moléculas adjacentes. Os valores de pKa das SAMs de Hpyt (4,2), TNA (5,0 e 8,5) e iTNA (4,5 e 7,9) foi determinado por voltametria utilizando-se o íon complexo [Fe(CN)6]3−. Neste estudo, foram sugeridas modificações, uma vez que o método proposto na literatura dificulta a determinação de mais de um valor de pKa como observado para as moléculas de TNA e iTNA. As reações de transferência de elétrons (TE) das metaloproteínas citocromo c (cyt c) e mioglobina (Mb) foram estudadas utilizando-se as SAMs de Hpyt, TNA, iTNA, pyS e 1,4-dt. Para as SAMs de TNA e iTNA, o deslocamento positivo de 0,2V no valor do potencial de meia-onda do cyt c (pH~7,0) em relação a forma nativa, foi atribuído à densidade de carga positiva resultante da protonação do grupo NH2 (pKa ~ 8,0). Resultados de QCM e SPR indicaram que há a formação de uma monocamada de cyt c sobre as SAMs estudadas. Esta monocamada, embora não sendo redox ativa, permite o estudo da reação de TE das moléculas de cyt c em solução sugerindo que este processo pode envolver os orbitais das moléculas modificadoras. Para a metaloproteína Mb, utilizou-se uma SAM formada pelo aminoácido L-cisteína (cys), uma vez que nenhuma das SAMs estudadas acessou a reação de TE. O processo redox foi observado em 0,086 V o que sugere a forma nativa. Os dados de QCM e SPR indicaram, também, a formação de uma monocamada sobre a SAM de cys (Au/cys/Mb). Valores de –49,67 kJ mol−1 e −0,15 para ΔGads e g, respectivamente, foram calculados para a formação da monocamada de Mb sobre a SAM de cys. O eletrodo Au/cys/Mb apresentou atividade catalítica em relação a reação de oxidação do ácido ascórbico com uma diminuição de 400 mV no sobrepotencial e uma reação cineticamente controlada com uma constante de velocidade, kf, de ~2,0 x 10^4 L mol-1 s-1.

The dependence on the experimental conditions of the production of phosphine ruthenium complexes containing o-phenylene ligands was analyzed in this work. The reaction between mer-[RuIIICl3(dppb)(H2O)] and [RuIICl2(dppb)(PPh3)] starting complexes, where dppb = 1,4-bis(diphenylphosphine)butane and PPh3 = triphenilphosphine, and 4-cloro-1,2-phenylenediamine (opda-Clcat), 3,3’,4,4’-tetraaminebiphenyl (tabcat,cat), benzenedithiol (bzditiolcat), and 9,10-phenantrequinone (fenantq) species resulted in the production of trans-[RuIICl2(dppb)(opda-Clcat)], cis-[RuIICl2(dppb)(opda-Clq)], trans-[RuIICl2(dppb)(tabq,cat)], cis-[RuIICl2(dppb)(tabq,cat)], trans-[RuIICl2(dppb)(Bzditiolcat)], and trans-[RuIICl2(dppb)(fenantq)] complexes. These compounds were characterized by means of electrochemical techniques, electronic and vibrational spectroscopies, and 31P{1H} nuclear magnetic resonance. The 31P{1H} NMR technique allowed the determination of the cis and trans isomers based on correlation with previously reported data and with the isolated compounds from the syntheses made from the [RuIICl2(dppb)(PPh3)] reduced starting complex. The mechanistic scheme for the reaction between the mer-[RuIIICl3(dppb)(H2O)] complex and the opda-Clcat ligand was proposed based on kinetics experiments acquired by using stopped-flow technique. In accordance with the kinetic results, there is the suggestion of the formation of a binuclear intermediate whose metal centers are reduced through an intra-molecular electron transfer process where the ligand is oxidized. This conclusion reinforces the results reported in the literature for the opda ligand. All the synthesized complexes presented catalytic activity toward the hydrogenation reaction of acetophenone molecule. Among these, based on preliminary results, the cis-[RuIICl2(dppb)(opda-Clq)], trans-[RuIICl2(dppb)(tabq,cat)], and cis-[RuIICl2(dppb)(tabq,cat)] complexes presented a conversion of 100%. Although a more careful study should be done, this result suggests that these compounds are able to act as catalysts in reduction reaction of polar double bonding. A influência das condições reacionais na formação de complexos fosfínicos de rutênio com ligantes o-fenilênicos foram estudadas neste trabalho. A reação dos complexos mer-[RuIIICl3(dppb)(H2O)] e [RuIICl2(dppb)(PPh3)], onde dppb = 1,4-bis(difenilfosfina)butano e PPh3 = trifenilfosfina, e os ligantes o-fenilênicos 4-cloro-1,2-fenilenodiamina (opda-Clcat), 3,3’,4,4’-tetraaminobifenil (tabcat,cat), benzenoditiol (bzditiolcat) e 9,10-fenantrequinona (fenantq) resultaram na formação dos complexos trans-[RuIICl2(dppb)(opda-Clcat)], cis-[RuIICl2(dppb)(opda-Clq)], trans-[RuIICl2(dppb)(tabq,cat)], cis-[RuIICl2(dppb)(tabq,cat)], trans-[RuIICl2(dppb)(Bzditiolcat)] e trans-[RuIICl2(dppb)(fenantq)]. A caracterização destas espécies foi realizada por técnicas eletroquímicas, espectroscopias eletrônica e vibracional e ressonância magnética nuclear de 31P{1H}. A técnica de RMN 31P{1H} permitiu a identificação dos isômeros cis e trans dos complexos sintetizados através de comparação com dados da literatura e com espectros obtidos para os produtos isolados a partir do complexo reduzido, [RuIICl2(dppb)(PPh3)]. A proposta mecanística para a reação entre o complexo mer-[RuIIICl3(dppb)(H2O)] e o ligante opda-Clcat foi baseada em experimentos cinéticos obtidos através da técnica de stopped-flow. De acordo com os resultados cinéticos, há a sugestão de formação de um intermediário binuclear cujos centros metálicos são reduzidos através de um processo de transferência de elétrons intramolecular onde o ligante sofre oxidação. Esta conclusão reforça os resultados reportados na literatura para o ligante opda. Todos os complexos sintetizados apresentaram atividade catalítica em relação à reação de hidrogenação da molécula de acetofenona sendo que, de acordo com os ensaios realizados, os complexos cis-[RuIICl2(dppb)(opda-Clq)], trans-[RuIICl2(dppb)(tabq,cat)] e cis-[RuIICl2(dppb)(tabq,cat)] apresentaram uma conversão de 100%. Embora seja necessário um estudo mais criterioso, este resultado sugere que estes compostos podem atuar como catalisadores de reação de redução de duplas ligações polares.

SILVA, M. A. S. Síntese e Caracterização do Composto Heterobimetálico trans-[(SO3)(cyclam)Co-NCS-Ru(NH3)4(NCS)](BF4). 2009. 101 f. Dissertação (Mestrado em Química) - Centro de Ciências, Universidade Federal do Ceará, Fortaleza, 2009. Trans-[Co(cyclam)(SO3)(NCS)]·4H2O, trans-[Ru(NH3)4(NCS)(SO4)], and trans-[(SO3)(cyclam)Co−NCS−Ru(NH3)4(NCS)](BF4) complexes, where cyclam = 1,4,8,11-tetraazacyclotetradecane, were synthesized and characterized by X-ray difraction, vibrational and electronic (ultraviolet, visible and near infrared) spectroscopies, and electrochemical techniques. The electronic communication between Co and Ru metal centers of the binuclear complex was evaluated by electrochemistry and electronic spectrocopy in the near infrared region. Crystals suitable for X-ray studies were only isolated for the trans-[Co(cyclam)(SO3)(NCS)]·4H2O complex. The obtained results indicate a monoclic structure with cyclam ligand at the equatorial plane and SO32− and NCS− moieties occupying the axial positions being coordinated through, respectively, sulfur and nitrogen atoms. This result is reinforced by the observation, in the vibrational spectrum, of bands typically assigned to the cyclam ligand in a trans configuration. The cyclic voltammograms obtained for this compound indicate as Electrochemical-Chemical-Electrochemical mechanism. In fact, the spectroelectrochemical experiments obtained at -0.80 V vs Ag/AgCl show that this compound, upon reduction, suffers a substitution reaction in which the SO32− and NCS− moieties are replaced by solvent molecules (L) thus forming [Co(cyclam)(L)2]2+ type complexes. The observation in the vibrational spectrum of the trans-[Ru(NH3)4(NCS)(SO4)] complex of the 2132, 887 e 478 cm-1 bands assigned, respectively, to the νCN, νCS e δ(NCS) vibrational modes of the NCS− ligand indicates that this moiety is coordinated through the nitrogen atom. Electrochemical and spectroscopic studies of this compound in aqueous medium indicate that the reduction of the metal center induces the replacement of SO42− ligand by a water molecule. The synthesis of the binuclear compound, therefore, was made under reductive conditions aiming to produce the aquo-complex and, then, replace the water molecule by a coordination site of the trans-[Co(cyclam)(SO3)(NCS)]·4H2O complex. The results obtained for the isolated material hints that the binuclear complex is formed with the NCS− fragment as the bridge ligand. The acquired cyclic voltammogram presents two redox process with the half-wave formal potentials (E1/2) observed at −0.27 and 0.13 V vs Ag|AgCl and being assigned to the Co and Ru metal centers, respectively. In comparison to the monomers, the positive potential shift reflects the stabilization of the reduced state of the ruthenium metal atom (RuII) and the destabilization of the cobalt metal center (CoIII). This result is assigned to the coordination to an oxidated metal center, CoIII, whose effective nuclear charge increased the electronic delocalization increasing the withdrawing character of the NCS− bridge ligand. The comproportionation constant, Kc = 5.78 x 106, was calculated from the difference between the E1/2 values. The Kc value indicates a strong electronic communication between the metal atoms and classifies this binuclear complex as a mixed valence system of class II. Os compostos trans-[Co(cyclam)(SO3)(NCS)]·4H2O, trans-[Ru(NH3)4(NCS)(SO4)] e trans-[(SO3)(cyclam)Co−NCS−Ru(NH3)4(NCS)](BF4), onde cyclam = 1,4,8,11-tetraazaciclotetradecano, foram sintetizados e caracterizados por difrações de raios-X, espectroscopias vibracional na região do infravermelho e eletrônica nas regiões do ultravioleta e visível (UV-Vis) e por técnicas eletroquímicas. O grau de comunicação eletrônica entre os átomos de Co e Ru do complexo binuclear foi avaliado por eletroquímica e espectroscopia eletrônica na região do infravermelho próximo. Apenas para o complexo trans-[Co(cyclam)(SO3)(NCS)]·4H2O foi possível a obtenção de cristais que permitiram a determinação estrutural. Os dados obtidos indicam estrutura monoclínica com o ligante cyclam no plano equatorial e os ligantes SO32− e NCS− ocupando posições trans e coordenados ao átomo de Co através, respectivamente, dos átomos de S e N. Este resultado é reforçado através da observação, no espectro vibracional, de bandas tipicamente atribuídas ao ligante cyclam quando este se encontra em uma geometria trans. Os resultados de voltametria cíclica deste composto indicam um mecanismo eletroquímico-químico-eletroquímico. De fato, os experimentos de espectroeletroquímica (potencial controlado em -0,80 V vs Ag/AgCl) indicam que este composto experimenta, após redução, reação de substituição das moléculas SO32− e NCS− por moléculas do solvente (L), formando compostos do tipo [Co(cyclam)(L)2]2+. A observação, no espectro vibracional do complexo trans-[Ru(NH3)4(NCS)(SO4)], das bandas em 2132, 887 e 478 cm-1, atribuídas aos modos de νCN, νCS e δ(NCS), respectivamente, do ligante NCS−, indica a coordenação deste grupo através do átomo de nitrogênio. Estudos eletroquímicos e de espectroscopia eletrônica deste composto em meio aquoso indicam que a redução do centro metálico induz a reação de substituição do ligante SO42− por uma molécula de H2O. A reação para formação do composto binuclear, portanto, foi realizada em condições redutoras a fim de induzir a formação do aquo-complexo de rutênio e, em seguida, a reação de substituição da molécula de H2O por um sítio de coordenação do monômero trans- [Co(cyclam)(SO3)(NCS)]·4H2O. Os resultados obtidos para o material isolado indicam que há a formação do complexo binuclear com o ligante NCS− ocupando a posição ponte. A curva voltamétrica obtida para este composto apresenta dois pares de ondas redox com potenciais formais de meia-onda (E1/2) em −0,27 e 0,13 V vs Ag|AgCl atribuídos, respectivamente, aos centros metálicos de Co e Ru. Comparativamente aos monômeros, há a observação de um deslocamento positivo de potencial o que reflete a estabilização do estado reduzido para o átomo de rutênio, RuII, e desestabilização do estado oxidado para o átomo de cobalto, CoIII. Este resultado é atribuído a coordenação a um centro oxidado, CoIII, cuja carga nuclear efetiva aumenta a deslocalização de densidade eletrônica aumentando o caráter retirador do ligante ponte NCS−. O valor da constante de comproporcionamento, Kc = 5,78 x 106, calculada a partir da diferença entre os valores de E1/2, indica um forte grau de comunicação entre os centros metálicos e classifica este complexo como um sistema de valência mista de classe II.

Dissertação (Mestrado em Química Inorgânica) - Centro de Ciências, Universidade Federal do Ceará, Fortaleza, 2007. -(4-pyridinyl)-1,3,4-oxadiazole-2-thiol (Hpyt) and [Fe(CN)5Hpyt]3- (FeHpyt) species spontaneously adsorbed on gold forming SAM (Self-Assembled Monolayers) that, based on electrochemical data, contain pinholes through which [Fe(CN)6]4– and [Ru(NH3)6]3+ probe molecules access the underlying gold electrode. From the former molecule and the dependence of the faradaic current on the pH value of the electrolyte solution, the surface pKa of the Hpyt SAM was evaluated as 4.2. Taking into account the STM (Scanning tunneling Microscopy) results obtained for the Hpyt SAM on Au(111), it was possible to estimate that the Hpyt molecules are adsorbed in an hexagonal arrangement with an average distance between the sulfur atoms of 5.5 Å. These images also showed the presence of pinholes, which indicate that a strong lateral interaction does not effectively happen. The thermodynamics parameters ΔHads, ΔGads and ΔSads of the Hpyt and FeHpyt adsorption process were calculated from the correlation between the concentration of the adsorbates on the surface (Γ) and in solution (C) at different temperatures. The surface coverage data were obtained from the reductive desorption process RS-Au + e- �� RS- + Au°, where R = Hpyt or FeHpyt, observed at -555 and -566 mV vs Ag|AgCl|Cl- for Hpyt and FeHpyt SAM, respectively, indicating an enhancement of the Au–S bond as consequence of the π-backbonding interaction from iron metal center toward the Hpyt moiety. The heterogeneous electron transfer reaction of cytochrome c metalloprotein was successfully assessed by Hpyt and FeHpyt and SAM, presenting a quasi-reversible voltammograms with potentials consistent with the native form of the protein. As espécies 5-(4-piridil)-1,3,4-oxadiazol-2-tiol (Hpyt) e [Fe(CN)5Hpyt]3- (FeHpyt) adsorvem espontaneamente sobre ouro formando monocamadas automontadas (SAM - "Self-Assembled Monolayers") das quais, de acordo com os dados eletroquímicos, apresentam defeitos ou nanoporos por onde as moléculas de prova [Fe(CN)6]4– e [Ru(NH3)6]3+ acessam a superfície do eletrodo de ouro policristalino. A partir dessas moléculas de prova e da dependência entre a corrente faradáica e o pH do eletrólito, o pKa da monocamada de Hpyt foi calculada como 4,2. A partir dos resultados de microscopia de tunelamento STM (Scanning Tunneling Microscopy) obtidos para a monocamada automontada de Hpyt sobre Au(111) foi possível estimar que as moléculas de Hpyt são adsorvidas em um arranjo hexagonal com um espaçamento médio entre os átomo de enxofre de 5,5 Å. Essas imagens também mostraram a presença de nanoporos, do qual indica que fortes interações laterais não ocorrem efetivamente. Os parâmetros termodinâmicos ΔHads, ΔGads e ΔSads para o processo de adsorção das moléculas Hpyt e FeHpyt foram calculados a partir da correlação entre a concentração dos adsorbatos na superfície (Γ) e em solução (C) para diferentes temperaturas. A quantidade de moléculas adsorvidas foi calculada a partir do processo de desorção redutiva, onde R = Hpyt ou FeHpyt, observados em -555 e -566 mV versus Ag|AgCl|Cl- para as monocamadas de Hpyt e FeHpyt, respectivamente, indicando um fortalecimento da ligação Au–S como conseqüência do efeito π-backbonding do centro metálico ferro para o ligante Hpyt. A reação heterogênea de transferência de elétrons da metaloproteína citocromo c foi satisfatoriamente acessada pelas monocamadas de Hpyt e FeHpyt, apresentando um voltamograma quasi-reversível com potenciais consistentes com a forma nativa desta proteína.

Trans-[Ru(CNpy)(NH3)4(L)]2+ type complexes, where CNpy (4- Cyanopyridine), L= 1,4-dt (1,4-dithiane), pyS (4-mercaptopyridine), and Tio (thionicotinamide), were synthesized and characterized by HPLC, microanalysis, eletrochemistry and vibrational and eletronic spectroscopies. The eletrochemical and eletronic absorption results are indicative of a π back-bonding interaction, L(nπ*) ← RuII(dπ) for which L is the pyS, CNpy and Tio ligands. The half-wave formal potentials (E1/2) observed for the trans-[Ru(CNpy)(NH3)4(pyS)]2+, trans- [Ru(CNpy)(NH3)4(1,4-dt)]2+ and trans-[Ru(CNpy)(NH3)4(Tio)]2+ complexes: 1.00, 0.67 and 1.03 V vs NHE, respectively, are indicative of the stabilization of the metal in the reduced state comparatively to the start complex, trans-[Ru(CNpy)(NH3)4(OH2)]2+ (E1/2 = 0.35 V vs NHE). This result indicates, also, the strong π withdraw capability of the CNpy ligand. The shift observed for the νC≡N mode from 2239 cm-1 in the vibrational spectra of the CNpy free ligand to ∼2200 cm-1 upon coordination suggests that the coordination occurs through the nitrile fragment. For the pyS and Tio ligands, the observation of the signals assigned to the νC=S and νSH modes, respectively, in the vibrational spectra os the complexes indicates that the coordination occurs through the nitrogen atom os the pyridine ring. The SERS ex situ spectra of the gold surfaces modified with the isolated complexes allow the classification of these species as binfunctional molecules in which the sulfur atom of the pyS, 1,4-dt and Tio ligands is the adsorption site and the nitrogen atom of the CNpy pyridine ring is the functional terminal group. Also, the SERS results point for a trans conformation for the trans- [Ru(CNpy)(NH3)4(pyS)]2+ complex and for a gauche configuration for the trans- [Ru(CNpy)(NH3)4(1,4-dt)]2+ and trans-[Ru(CNpy)(NH3)4(Tio)]2+ complexes on gold. The intensity dependence of some signals in the SERS spectra with the applie potential reflects the π back-bonding effect on surface since this behavior is observed to be strongly related to the E1/2 values of the complexes. The reductive desorption potentials of the SAMs formed with the isolated compounds reflects the π withdraw capability of the CNpy ligand. This effect is much more intense for the trans-[Ru(CNpy)(NH3)4(1,4-dt)]2+ complex since the π back-bonding interaction of the 1,4-dt ligand is lower than the CNpy, pyS and Tio ligands. The heterogeneous electron transfer reaction of the cytochrome c metalloprotein was satisfactorily assessed with the SAMs formed by the isolated complexes (E1/2 = 0.05 V vs Ag⏐AgCl⏐Cl-). The Best voltammetric response was observed with the SAM of the trans- [Ru(CNpy)(NH3)4(pyS)]2+ complex due to the trans conformation on surface. Os complexos trans-[Ru(CNpy)(NH3)4(1,4-dt)]2+ (CNpy = 4-Cianopiridina, 1,4- dt = 1,4-ditiano), trans-[Ru(CNpy)(NH3)4(pyS)]2+ (pyS = 4-mercaptopiridina) e trans- [Ru(CNpy)(NH3)4(Tio)]2+ (Tio = tionicotinamida) foram sintetizados e caracterizados por Cromatografia Líquida de Alta Eficiência, análise elementar, eletroquímica e espectroscopias eletrônica e vibracional. Os espectros de absorção eletrônica e os resultados de eletroquímica evidenciaram uma interação do tipo π back-bonding do centro metálico RuII para os ligantes piridínicos. Os valores dos potenciais formais de meia onda (E1/2) observados para os complexos trans-[Ru(CNpy)(NH3)4(1,4-dt)]2+, trans-[Ru(CNpy)(NH3)4(pyS)]2+ e trans-[Ru(CNpy)(NH3)4(Tio)]2+: 1,00, 0,67 e 1,03 V vs ENH, respectivamente, indicam a estabilização do estado reduzido do centro metálico em relação ao complexo de partida trans-[Ru(CNpy)(NH3)4(OH2)]2+ (E1/2 = 0,35 V vs ENH). Esse resultado indica, também, a forte capacidade π retiradora do ligante CNpy que, no caso do complexo trans-[Ru(CNpy)(NH3)4(1,4-dt)]2+, é tão intensa que facilita comparativamente o processo de oxidação do centro metálico. Podese concluir, portanto, que a capacidade π retiradora dos ligantes CNpy, pyS e Tio é similar. O deslocamento na freqüência de estiramento da ligação C≡N do ligante Cnpy livre de 2239 para ∼2200 cm-1 após coordenação ao centro metálico [Ru(NH3)4]2+ nos espectros vibracionais indica que a coordenação do ligante CNpy ocorre pelo grupamento nitrila. Para os ligantes pyS e Tio, a observação das bandas atribuídas aos νC=S e νSH, respectivamente, indica que a coordenação ocorre pelo átomo de nitrogênio do anel piridínico. Os espectros SERS ex situ das superfícies modificadas com os compostos isolados permitem a classificação destes como moléculas modificadoras bifuncionais, onde o átomo de enxofre dos ligantes pyS, 1,4-dt e Tio atuam como sítio de adsorção e o átomo de nitrogênio piridínico do ligante CNpy atua como grupo funcional terminal. Além disso, os resultados SERS sugerem uma conformação trans para o complexo trans-[Ru(CNpy)(NH3)4(pyS)]2+ e gauche para os complexos trans-[Ru(CNpy)(NH3)4(1,4-dt)]2+ e trans-[Ru(CNpy)(NH3)4(Tio)]2+ sobre ouro. A dependência nas intensidades das bandas com o potencial aplicado aos eletrodos modificados ilustra o efeito π back-bonding dos íons metálicos dos adsorbatos inorgânicos, uma vez que observa-se uma relação direta com os valores dos potenciais formais de meia onda dos complexos estudados. Os valores dos potenciais de dessorção redutiva observados para as SAMs dos complexos isolados refletem a capacidade π retiradora do ligante CNpy que se encontra na posição para em relação ao sítio de adsorção. Esse efeito é bem mais intenso no complexo trans- [Ru(CNpy)(NH3)4(1,4-dt)]2+, uma vez que a capacidade de interação π back-bonding do ligante 1,4-dt é significativamente menor que as dos ligantes pyS e Tio. Sugere-se, para esse complexo, uma oxidação parcial do centro metálico o que implica em uma intensificação da interação σ com o ligante 1,4-dt (RuII-S) e, conseqüentemente, um enfraquecimento da ligação Au-S. A reação hTE da metaloproteína cyt c foi observada em valor consistente com seu estado in natura indicando que as SAMs estudadas acessam satisfatoriamente o processo hTE do cyt c além de evitar a desnaturação desta proteína. A forma das curvas voltamétricas, entretanto, apresentou uma dependência com a configuração dos complexos sobre a superfície. Curvas de melhor definição foram obtidas com a SAM do complexo trans-[Ru(CNpy)(NH3)4(pyS)]2+ que apresenta uma configuração trans em relação à superfície.
