Introduction. A cross-sectional study was undertaken to deter- mine the prevalence of Giardia and Cryptosporidium in calves of Palermo area (Sicily) and to evaluate the occupational risk associated with occurrence of zoonotic genotypes. Methods. A total of 217 faecal samples, from 149 calves (between 2 and 240 days of age) and 68 farmers, were collected in 19 cattle- farms of Palermo area. A questionnaire regarding demographic characteristics and personal hygienic measures was submitted to all farmers. All faecal samples were analyzed by Immunoflu- orescence assay and Polimerase Chain Reaction (PCR); geno- types were determined by DNA sequencing of Triose Phosphate Isomerase gene for Giardia and Small Subunit Ribosomal RNA gene for Cryptosporidium. Results. None farmer tested was positive for Giardia and Cryptosporidium, whereas these protozoa were respectively detected in 53 (including 5 with zoonotic G. duodenalis geno- type A) and 17 (of which 1 with zoonotic C. ubiquitum) of the examined calves. Discussion. The results indicate that the risk of transmitting both protozoa to farmers in Palermo area is negligible although it can- not be considered null because of identification of human geno- types/species in calves., Journal of Preventive Medicine and Hygiene, Vol 54, No 4 (2013)

Introduction. In recent years an increase in the number of sexu- ally transmitted infections (STIs) in men who have sex with men (MSM) has been reported in different industrialised countries. Because few epidemiological data on the STIs/MSM population in Sicily are available, a survey was conducted to assess the preva- lence of STIs/enteric protozoa and risky sexual behaviours among MSM in western Sicily. Methods. In 2010, 74 MSM with median age of 30 years old, were recruited via networks. All participants to the study were interviewed by anonymous self-administered questionnaire in order to collect social/demographic information, clinic data and STI-related risky sexual behaviours. After completing the ques- tionnaire, blood samples were collected to determine HIV, HCV, HHV8 and Treponema pallidum antibodies; presence of Giardia duodenalis and Cryptosporidium parvum was also investigated in faecal samples by immunofluorescence assay. Results. HIV, HHV8, T. pallidum and Giardia prevalence were 8.1%, 16.2%, 21.6% and 16.4% respectively; all patients were negative for HCV and Cryptosporidium infections. The median values of sexual anal intercourse and oral sex per week were 2 and 1, respectively. 7% of participants always had unprotected anal sex, 50.7% sometimes used condom during sexual anal inter- course and 42.3% always had protected anal sex. All MSM-HIV+ and 7 (43.7%) syphilis seropositives were unaware of their own infection. Discussion. MSM in western Sicily are a high risk group for important STIs. It seems necessary that continuous interventions for preventing HIV/AIDS and other STIs and for improving the level of knowledge of symptoms are needed., Journal of Preventive Medicine and Hygiene, Vol 53, No 4 (2012)

Anthracycline has been shown to induce heart failure. To monitor this toxic damage, echocardiographic parameters of left ventricular (LV) systolic function are usually used. Aim of this study was to evaluate in lymphoma’s patients the reliability of echocardiographic data in comparison with a LV systo-diastolic parameter function: the Tei index.

Background: MicroRNAs (miRNAs) are small non-coding RNAs that regulate the expression of different genes, including genes involved in cancer progression, angiogenesis and metastasis. Thrombospondin-1 (TSP-1) has been shown to contrast angiogenesis in vivo. TSP-1 expression levels are inversely correlated with tumor vascularity and metastasis in colon cancer. Bio-informatic statistical analysis indicated that TSP-1 is hypothetical target of miR-21, miR-182, overexpressed in CRC, and let-7i which expression is down-regulated in this tumor. In this work we investigated whether TSP-1 expression could be regulated by miR-21, mir-182 and let-7i in HT29 colon cancer cell line. Methods: To investigated whether miR-21, mir-182 and let-7i directly modulates TSP-1 expression, we transfected HT29 cell line with pre-mir21, pre-mir182 and pre-let7i by using siPortNeo FX tranfection agent and after 48h we evaluated TSP-1 mRNA, using Quantitative Real Time-PCR, and intracellular and secreted protein level performed by Western blotting and ELISA. To confirm the modulation of TSP-1 by miRNAs we transfected HT29 cell line with anti-mir to target the mature form of miR-21, miR182 and let-7i. Results: Using Real-Time PCR we did not find any variation of TSP-1 mRNA expression levels after transfection with pre-mir21 in HT29 cell line, but we observed a down-regulation of cytosolic and secreted protein by Western blot and ELISA. In cells transfected with pre-mir182 we did not observe any down-regulation both TSP-1 mRNA and cytosolic and secreted protein. Finally, we did not find any variation of TSP-1 level in cells transfected with let- 7i. Results were confirmed by transfection with anti-mir21, anti- mir182 and anti-let7i and, using the same method, we evaluated TSP-1 expression. Conclusions: Data suggest that mir-182 induces degradation of TSP-1 mRNA in HT29 cell line, whereas mir-21 affects probably by blockage of TSP-1 translation. Let-7i does not seem involved in regulation of TSP-1 expression in HT29 cells. Understanding the molecular mechanism by which miRNAs regulate TSP-1 expression could be used to restore TSP-1 expression to contrast angiogenic events in colon cancer.

In a previous study we evaluated the microbiological quality of water of seven pools in the city of Palermo through evaluation of bacterial indicators of faecal contamination and of protozoa (Giardia and Cryptosporidium). In this study we also searched for the presence of fungi in two swimming pools of a public swimming facility in the same city. Samples were collected from both pools, their filtration systems and floor surfaces of the facility. Chemical-physical and microbiological examination of water of the two pools have shown that quality of water depends on the concentration of residual free chlorine and on the number of bathers in the pool. The values of four microbiological parameters (bacterial load at 22 degrees C and 36 degrees C, presence of coagulase-negative Staphylococci and Pseudomonas spp.) increased with diminishing chlorine concentrations and with increasing number of pool users. Faecal bacteria, Giardia and Cryptosporidium were not found. On the other hand, various fungi were isolated from floor surfaces and pool water even in the presence of optimal chlorine concentrations. This study confirms the importance of regular maintenance of pool disinfection systems and suggests the need to search for other micro-organisms not included in the current legislation (Giardia, Cryptosporidium and fungi).

The aim of this study was to identify whether the origin of Oreto river Giardia contamination was derived from one of the influents, which flows near Monreale city and receives its treated wastewater, or from livestock living in farms along the river. Genotypes were characterized by DNA sequencing analysis of PCR products of TPI gene. 17 water samples coming from Oreto basin, 20 fecal animal samples and 7 human fecal samples from 3 patients with giardiasis were processed. Cysts were typed as assemblage A (n=11), B (n=2) and A+B (n=4) in water samples, as assemblage A (n=4) and B (n=2) in human samples, while in livestock was found only assemblage E. Subtype AII was prevalent in both water and human samples. This work suggests the need to increase the efficacy of wastewater treatment in order to avoid any further river contamination.

The knowledge, attitude, and practice toward the use of ready-to-eat vegetables were evaluated in a sample of 701 (388 women and 313 men) potential consumers in Palermo, by answering to an anonymous questionnaire handed out. Food safety perceptions and behaviors were analysed; in general, interviewed consumers who perceived higher risks exhibited safer food behaviors. There weren't considerable differences in the use of these salads between two sexes; 81.4% of respondents uses these products to save a lot of time, particularly the graduates attach importance to saving in time (p = 0.001), and only 34.4% is worrying about high price. Statistical analysis showed a significant correlation between better food safety knowledge and female sex; for example, compared with women, men reported to buy ready-to-eat salads on sale at room temperature and at temperature of refrigerator without distinction (p = 0.015). This study confirms the IV range salads meet modern consumer's requirements and also suggests the need of food safety educational strategies for targeted groups.

This paper reports the investigation carried out on the occurrence of Cryptosporidium and Giardia (oo)cysts in water samples of two municipal treatment plants, and in surface water and ground water wells of the province of Palermo. The wastewater samples taken before and after treatment process were assayed over the course of one year Giardia cysts were detected in all samples throught the year at higher concentration levels than Cryptosporidium oocysts, with a peak observed in spring. The overall removal efficiency of (oo)cysts in the treatment plants was about of 90%. Their presence were also searched in surface waters (three artificial lakes and one river); (oo)cysts were detected in one lake at very low concentration; on the contrary, both parasites were found at high concentration levels in all the samples collected throught one year from the water of the river. The pattern of occurrence of both parasites appears temporally related to the level of rainfall trend. Cryptosporidium and Giardia were also found in ground water wells; their presence occurred only in waters taken from wells at a depth lower than 31 meters with concomitant presence of faecal bacteria. These results may provide further insight into the possible source of Cryptosporidium and Giardia in natural environmental and stress the potential risk associated with the use of waters for recreational and agricultural purposes.
