ObjectiveTo explore the link between plasma aldosterone concentration (PAC) and the prevalence of metabolic dysfunction-related fatty liver disease (MAFLD) in hypertensive patients.MethodsWe analyzed data from 41,131 hospitalized patients from January 1, 2014, to December 31, 2023. Multivariate logistic regression models tested associations, with threshold, subgroup, and sensitivity analyses conducted to validate findings.ResultsFor each 5-unit increase in PAC, the risk of MAFLD rose by 1.57 times, consistent even in the fully adjusted model. The odds ratios for the Q2, Q3, and Q4 groups compared to Q1 were 1.21, 2.12, and 3.14, respectively. A threshold effect was observed at 14 ng/dL, with subgroup and sensitivity analyses supporting these results.ConclusionsThis study reveals a significant positive association between elevated PAC levels and the prevalence of MAFLD in hypertensive patients. These findings underscore the imperative for further large-scale, prospective studies to validate and expand upon this correlation.

The evasion of carbon dioxide (CO2) from lakes significantly influences the global carbon equilibrium. Amidst global climatic transformations, the role of Qingzang Plateau (QZP) lakes as carbon (C) sources or sinks remains a subject of debate. Furthermore, accurately quantifying their contribution to the global carbon budget presents a formidable challenge. Here, spanning half a century (1970–2020), we utilize a synthesis of literature and empirical field data to assess the CO2 exchange flux of QZP lakes. We find markedly higher CO2 exchange flux in the southeast lakes than that in the northern and western regions from 1970 to 2000. During this time, both freshwater and saltwater lakes served primarily as carbon sources. The annual CO2 exchange flux was estimated at 2.04 ± 0.37 Tg (Tg) C yr−1, mainly influenced by temperature fluctuations. The CO2 exchange flux patterns underwent a geographical inversion between 2000 and 2020, with increased levels in the west and decreased levels in the east. Notably, CO2 emissions from freshwater lakes diminished, and certain saltwater lakes in the QTP transitioned from carbon sources to sinks. From 2000 to 2020, the annual CO2 exchange flux from QZP lakes is estimated at 1.34 ± 0.50 Tg C yr−1, with solar radiation playing a more pronounced role in carbon emissions. Cumulatively, over the past five decades, QZP lakes have generally functioned as carbon sources. Nevertheless, the total annual CO2 emissions have declined since the year 2000, indicating a potential shift trend from being a carbon source to a sink, mirroring broader patterns of global climate change. These findings not only augment our understanding of the carbon cycle in plateau aquatic systems but also provide crucial data for refining China's carbon budget.

Abstract Objective Recently, the stress hyperglycemia ratio (SHR) has been introduced as a metric to signify relative hyperglycemia. This study aimed to investigate the relationship between SHR and in-hospital mortality and length of stay occurring during hospitalization in stroke patients. Methods The retrospective cohort study comprised a total of 4,018 patients diagnosed with acute stroke. The SHR is expressed by the formula: SHR = ABG (mmol/L) / [1.59 × HbA1c (%) − 2.59]. Outcomes included in-hospital mortality and length of stay. Multivariable logistic and linear regression analyses were conducted. Receiver operating characteristic (ROC) analysis was performed to distinguish between the variables, and the area under the ROC curve (AUC) was compared. Results In this analysis, a total of 4,018 individuals participated, including 2,814 male patients, accounting for 70.0%. Overall, in-hospital mortality and length of stay tended to rise as SHR increased. A higher prevalence of in-hospital mortality was observed with each standard deviation (SD) increase of the SHR (odds ratio [OR]: 1.26, 95% confidence interval [CI]: 1.05–1.52). Moreover, after considering the confounders, a significant positive association between SHR levels and length of stay was observed (β = 0.70, 95% CI: 0.40-1.00). ROC analysis showed that among stroke patients, SHR (AUC = 0.693) was more effective than admission blood glucose (ABG) (AUC = 0.646) and glycosylated hemoglobin (HbA1c) (AUC = 0.523), which were more predictive of in-hospital mortality. Conclusions Elevated SHR levels are associated with increased in-hospital mortality and prolonged length of stay in stroke patients.

Abstract The purpose of this study was to determine the long-term pattern of plasma aldosterone concentration (PAC) trajectories and to explore the relationship between PAC trajectory patterns and cardiovascular disease (CVD) risk in patients with hypertension. Participants were surveyed three times between 2010 and 2016, and latent mixed modeling was employed to determine the trajectory of PAC over the exposure period (2010–2016). A Cox regression analysis was used to examine the association between PAC trajectory patterns and the risk of CVD (stroke and myocardial infarction). Hazard ratios (HRs) with corresponding 95% confidence intervals (CIs) were calculated and reported. During a median follow-up of 4.10 (3.37–4.50) years, 82 incident CVD cases (33 myocardial infarction cases and 49 stroke cases) were identified. Among all three PAC models, the high-stability PAC pattern exhibited the highest risk of CVD. After full adjustment for all covariables, HRs were 2.19 (95% CI 1.59–3.01) for the moderate-stable pattern and 2.56 (95% CI 1.68–3.91) for the high-stable pattern in comparison to the low-stable pattern. Subgroup and sensitivity analyses verified this association. The presence of a high-stable PAC trajectory pattern is associated with an elevated risk of CVD in hypertensive patients. Nevertheless, more studies are warranted to confirm these findings.

BackgroundPrior research has highlighted the association between uric acid (UA) and the activation of the renin-angiotensin-aldosterone system (RAAS). However, the specific relationship between aldosterone, the RAAS’s end product, and UA-related diseases remains poorly understood. This study aims to clarify the impact of aldosterone on the development and progression of hyperuricemia and gout in hypertensive patients.MethodsOur study involved 34534 hypertensive participants, assessing plasma aldosterone concentration (PAC)’s role in UA-related diseases, mainly hyperuricemia and gout. We applied multiple logistic regression to investigate the impact of PAC and used restricted cubic splines (RCS) for examining the dose-response relationship between PAC and these diseases. To gain deeper insights, we conducted threshold analyses, further clarifying the nature of this relationship. Finally, we undertook subgroup analyses to evaluate PAC’s effects across diverse conditions and among different subgroups.ResultsMultivariate logistic regression analysis revealed a significant correlation between the occurrence of hyperuricemia and gout and the elevation of PAC levels. Compared to the first quartile (Q1) group, groups Q2, Q3, and Q4 all exhibited a significantly increased risk of occurrence. Moreover, the conducted RCS analysis demonstrated a significant nonlinear dose-response relationship, especially when PAC was greater than 14 ng/dL, with a further increased risk of hyperuricemia and gout. Finally, comprehensive subgroup analyses consistently reinforced these findings.ConclusionThis study demonstrates a close association between elevated PAC levels and the development of UA-related diseases, namely hyperuricemia and gout, in hypertensive patients. Further prospective studies are warranted to confirm and validate this relationship.

BackgroundPrevious studies have suggested that aldosterone may play a major role in calcium-phosphorus homeostasis and bone metabolism. However, the relationship between plasma aldosterone concentrations (PAC) and bone mineral density (BMD) in middle-aged and elderly hypertensive patients remains unclear. Therefore, this study sought to investigate the relationship between PAC levels and BMD and explore PAC’s potential impact on osteoporosis and future fracture risk in hypertensive patients.MethodsOur study included a total of 1430 participants. Associations are tested using multiple linear and logistic regression models. Nonlinearity was investigated using the restricted cubic spline (RCS). We also performed mediating analyses to assess mediating factors mediating the relationship between PAC and osteoporosis.ResultsThe multiple linear regression showed a negative correlation between PAC and BMD and was generally positively associated with FRAX scores. Meanwhile, logistic regression analyses indicated that osteoporosis was highly correlated with PAC levels. In addition, a clear non-linear dose-response relationship was also shown in the constructed RCS model. Finally, mediation analyses showed that serum potassium played an important role in the development of osteoporosis.ConclusionThis study demonstrates that elevated PAC levels are strongly associated with decreased BMD, increased prevalence of osteoporosis, and the risk of future fractures in middle-aged and elderly hypertensive patients. Further studies are needed to confirm this relationship and reveal its underlying mechanisms.

AIM: To observe the epithelial remodeling of femtosecond laser-assisted stromal lenticule addition keratoplasty combined with corneal collagen cross-linking(SLAK-CXL)in patients with progressive keratoconus, investigate the remodeling rules of corneal epithelial and influencing factors, and provide clinical data for further refractive correction.METHODS: Retrospective and observational study. A total of 28 keratoconus patients(29 eyes)who received SLAK-CXL from September 2020 to October 2021 were included. Preoperative and postoperative visual acuity, intraocular pressure(IOP), diopter, keratometry and corneal epithelial thickness(CET)were recorded. The trend of CET change was observed. The factors affecting CET were analyzed according to the thickness and depth of the lenticule.RESULTS: Flattest meridian keratometry(Kf)and steepest meridian keratometry(Ks)at 1mo postoperatively were significantly larger than those preoperatively(P0.05).CONCLUSIONS: It is firstly found that the corneal morphology has changed after SLAK-CXL. CET decreases and then increases and then decreases again. At 1a postoperatively, the CET of the central and paracentral areas is thinner, while the CET of the midperipheral and peripheral areas is thicker. The degree of epithelial remodeling is not correlated with lenticule thickness or depth.

Urban air mobility is an effective solution to address the current issue of ground traffic congestion in future cities. However, as the user scale continues to expand, the current civil aviation flight scheduling and control methods are becoming inadequate to meet the high-volume flight guarantee demands of future urban air transportation. In order to effectively handle and resolve potential issues in this field in the future, this paper proposes a method for planning urban air mobility route networks. The planning process is divided into two stages: construction and optimization. Methods for constructing urban air mobility route networks based on flight routes and global optimization methods based on node movement are proposed in each stage. In the construction stage, a complete construction process is designed to generate routes based on existing flight routes, in line with the trend of urban air transportation development. In the optimization stage, inspired by the ant colony algorithm, node transfer rules and information transfer rules are incorporated to design a global optimization process and algorithm for route networks. Experimental results demonstrate the effectiveness and advancement of the proposed planning method.

Many transcriptomic pathways in the liver show circadian rhythms, which have been reported to be disrupted in aged mice. Here the authors report that the expression of transcription factor Egr-1 decreases and its rhythm is shifted with age in the liver of male mice, and that deletion of Egr-1 results in increased liver fat accumulation.

Abstract Hepatocellular carcinoma (HCC) is one of the most common cancers worldwide. Its invasiveness and ability to metastasize contributes to an extremely high patient mortality. However, the molecular mechanisms that underlie the characteristics of HCC progression are not well understood. BRF2 has been shown to be an oncogene in a number of tumors; however, its role in HCC has not yet been thoroughly examined. In this study, we identified and validated BRF2 as an oncogene in HCC, providing a new insight into HCC pathogenesis and therapeutic possibilities. We showed that BRF2 expression was significantly upregulated in HCC cell lines and tissues, while BRF2 depletion suppressed HCC metastasis and invasion. We then examined the upstream regulation of BRF2 and identified miR-409-3p as being predicted to bind to the 3′ UTR of BRF2. We used a luciferase activity assay and functional verification to show that BRF2 is downregulated by miR-409-3p. Finally, we used bioinformatic analysis to show that BRF2 may be related to early HCC development through the Wnt/β-catenin signaling pathway.

Abstract Background Atherosclerosis (AS) is the leading underlying cause of the majority of clinical cardiovascular events. Retention of foamy macrophages in plaques is the main factor initiating and promoting the atherosclerotic process. Our previous work showed that ox-LDL induced macrophage retention in plaques and that the guidance receptor Uncoordinated-5 homolog B (Unc5b) was involved in this process. However, little is known about the role of Unc5b in regulating macrophage accumulation within plaques. Results In the present study, we found that Unc5b controls macrophage migration and thus promotes plaque progression in ApoE−/− mice. The immunofluorescence colocalization assay results first suggested that fucosyltransferase 8 (Fut8) might participate in the exacerbation of atherosclerosis. Animals with Unc5b overexpression showed elevated levels of Fut8 and numbers of macrophages and an increased lesion size and intimal thickness. However, these effects were reversed in ApoE−/− mice with Unc5b knockdown. Furthermore, Raw264.7 macrophages with siRNA-mediated silencing of Unc5b or overexpression of Unc5b were used to confirm the regulatory mechanisms of Unc5b and Fut8 in vitro. In response to ox-LDL exposure, Unc5b and Fut8 were both upregulated, and macrophages showed reduced pseudopod formation and migratory capacities. However, these capacities were restored by blocking Unc5b or Fut8. Furthermore, the IP assay indicated that Fut8 regulated the level of α-1,6 fucosylation of Unc5b, which mainly occurs in the endoplasmic reticulum (ER), and genetic deletion of the main fucosylation sites or Fut8 resulted in hypofucosylation of Unc5b. Moreover, the macrophage migration mediated by Unc5b depended on inactivation of the p-CDC42/p-PAK pathway. Conversely, macrophages with Unc5b overexpression displayed activation of the p-CDC42/p-PAK pathway and decreased migration both in vivo and in vitro. Conclusion These results demonstrated that hypofucosylation of Unc5b regulated by Fut8 is positively associated with the delay of the atherosclerotic process by promoting the migration of foamy macrophages. These findings identify a promising therapeutic target for atherosclerosis.

As the prevalence of myopia continues to increase, our society shows increasing interest in the visual impairment associated with myopia and methods for myopia control. Current methods for myopia control include atropine, orthokeratology, dual-focus contact lenses, multifocal contact lenses, and functional frame glasses, etc. Low-concentration atropine(0.01%, 0.05%)became the first-line medication which has been administrated and shown to be effective in controlling the occurrence and progression of myopia. Meanwhile, low-concentration atropine shows longer-lasting effects, less frequent adverse effects and higher patients' acceptance than high-concentration atropine. This article reviews the efficacy, safety, time-to-treatment, and effects when combined with other treatments to provide evidence for the clinical application of low-concentration atropine on myopia control.

Study of the segmental mobility of polymer chains is important when the polymer is used as drag reduction agents of crude oil. Electron spin resonance (ESR) spectroscopy can provide important information about segmental dynamics of polymer chains, which is related to their microenvironment. In this article, we employed an amphiphilic polymer to study the effect of hydrophilic/hydrophobic balance of the polymers on the segmental motion of polymer chains. Poly(styrene-co-maleic acid) (PSMA) was spin labeled with 4-amino TEMPO radicals by increasing the concentration of radical moiety on the polymer chains. The PSMA and spin labeled-PSMAs (SL-PSMAs) were characterized by nuclear magnetic resonance (NMR), Fourier transform infrared (FT-IR) spectroscopy, Cyclic voltammetry (CV) and ESR techniques. Inter-polymer complexes (IPCs) of SL-PSMA-2 were prepared by employing polyethylene glycols (PEGs) of varying molecular weights. The results showed that the increased hydrophobic interactions of nitroxide radicals on the SL-PSMAs’ chains reduced the rotational mobility of spin labels and the random coil-to-globular transition of polymer chains occurred at higher pH value for SL-PSMAs, which showed a slow motion component in the ESR spectra of SL-PSMAs. Further, by increasing the molecular weight of PEGs in IPCs the complexation was increased, which also reduced the rotational motion of spin labels due to interpolymer hydrogen bonding causing a slow motion component in the ESR spectra.

With the rapid advancement of unmanned aerial vehicle (UAV) technology, the widespread utilization of UAVs poses significant challenges to urban low-altitude safety and airspace management. In the coming future, the quantity of drones is expected to experience a substantial surge. Effectively regulating the flight behavior of UAVs has become an urgent and imperative issue that needs to be addressed. Hence, this paper proposes a standardized approach to UAV flight through the design of an air channel network. The air channel network comprises numerous single air channels, and this study focuses on investigating the characteristics of a single air channel. To achieve optimal outcomes, the concept of the artificial potential field algorithm is integrated into the deep Q-learning algorithm during the establishment of a single air channel. By improving the action space and reward mechanism, the resulting single air channel enables efficient avoidance of various buildings and obstacles. Finally, the algorithm is assessed through comprehensive simulation experiments, demonstrating its effective fulfillment of the aforementioned requirements.

In order to further improve the computing power of the information aggregation operator in the q-rung orthopair probabilistic hesitant fuzzy environment, this paper proposes a multi-attribute decision-making method based on the q-rung orthopair probabilistic hesitant fuzzy Schweizer-Sklar power weighted Hamy mean operator. Firstly, the algorithm of q-rung orthopair probabilistic hesitant fuzzy set is improved based on the Schweizer-Sklar T-norm. In order to better reflect the degree of hesitation of decision-making experts, a new q-rung orthopair probabilistic hesitant fuzzy distance measure is proposed, which provides a basis for subsequent power weighted calculations. Furthermore, considering the correlation between attributes and the influence of data extremes, some information aggregation operators and their power weighted forms are proposed. Finally, a multi-attribute decision-making model based on the q-rung orthopair probabilistic hesitant fuzzy Schweizer-Sklar power weighted Hamy mean operator is established, and the reliability and validity of the research content in this paper are verified through decision-making examples and comparative analysis.

AIM: To compare the change of refractive power and corneal high-order aberrations after femtosecond laser-assisted excimer laser in situ keratomileusis(FS-LASIK)and FS-LASIK combined with accelerated corneal cross-linking(FS-LASIK Xtra)for high myopia correction, and to evaluate the early effects of two surgical methods to correct high myopia. METHODS:Retrospective case-control study. Totally 42 patients(84 eyes)with high myopia underwent FS-LASIK Xtra and FS-LASIK in our hospital from April 2019 to April 2020 were followed up for 3mo, including 21 patients in each group(42 eyes). The uncorrected visual acuity(UCVA), spherical equivalent(SE), astigmatism and corneal high-order aberration of the two groups were analyzed and compared.RESULTS: The UCVA of the FS-LASIK Xtra group was significantly lower than that of the FS-LASIK group at 1d after surgery(P0.05). The postoperative SE of the two groups was significantly lower than that before surgery, at 3mo after surgery, 38 eyes(90%)in the FS-LASIK Xtra group and 41 eyes(98%)in the FS-LASIK group had SE within ±1.00D. In both groups, 35 eyes(83%)had residual astigmatism within 0.50D after surgery. The total corneal high-order aberrations, spherical aberrations, coma and clover aberrations in the two groups were increased at 3mo after surgery, the total high-order aberration and clover aberration in the FS-LASIK Xtra group were greater than those in the FS-LASIK group(all P

Abstract Carbon trading mechanism is an effective means to control greenhouse gas emissions. This paper focuses on the low‐carbon economic operation of the integrated energy system under carbon trading mechanism in China. The integrated energy system includes the energy storage, ground source heat pump, and other equipment. The objective of this paper was to minimize the annual total cost of the system considering the carbon trading cost and study the operation modes under different carbon trading prices by commercial optimization software. The simulation results show the operation modes in summer are changed obviously with the increase of the carbon trading prices, while the operation modes in winter basically are not changed with the fluctuation of the carbon trading prices; under the carbon trading mechanism, the integrated energy system can not only reduce the carbon emissions, but also reduce the annual total cost through carbon trading, which shows its advantages and good development prospect. In addition, the setting of the carbon emission quota has a direct impact on the economy of the enterprises. Furthermore, the ground source heat pump and the electric refrigeration units can stabilize the fluctuation of the equipment output caused by the change of natural gas prices in this integrated energy system.

AIM:To analyze satisfaction on night vision and its related factors after FS-LASIK in patients with high and low-to-moderate myopia.METHODS:This was a retrospective study. Totally 75 patients(150 eyes)who underwent FS-LASIK in Xi'an No.1 Hospital from June 2018 to September 2019 were included. All subjects were divided into two groups based on spherical equivalent(SE): high myopia group(SE≥-6.00D)and low-to-moderate myopia group(0.05)between the two groups. Overall satisfaction was 95.3% and 100.0% in high and low-to-moderate myopia group. 18.8% and 9.3% experienced worsen night vision, 51.2% and 40.6% experienced halo, 41.9% and 43.8% experienced starburst, and 18.6% and 15.6% experienced double vision, respectively. Except that there was significant differences in night vision between the two groups(P=0.010), the occurrence of other night vision symptoms were similar and there were no significant differences in any of them(all P>0.05). It is of great significance to identify the hot spots and driving forces of geological hazard gathering for regional disaster early warning. Dingxi region of Gansu Province is taken as a the study area. Seven factors such as slope, aspect, land use, engineering rock group and so on are selected to analyze the spatial agglomeration characteristics and driving factors of geological hazards by using coupling model of determining coefficient and logical regression and spatial autocorrelation. The results show that the sensitivity of geological hazards in Dingxi region is high in the middle region and low in the north and south region. The spatial relationship between geological hazards and precipitation, aspect and engineering rock group is high accumulation. Geological hazard sensitivity has strong spatial autocorrelation, which is significantly positive correlation in 3 km scale, and decreases with the increase of distance threshold. When the distance threshold is 5 km, the high and low aggregation areas are clustered and distributed in the form of clumps, while the high and low aggregation areas and the low and high aggregation areas are mostly scattered. The research can provide reference for regional geological hazard monitoring and control.

AIM: To investigate and evaluate healing patterns around flaps made with different side-cut angulations after femtosecond laser in situ keratomileusis (FS-LASIK). METHODS: Thirty-four patients (68 eyes) received a 90° side-cut (n=34) or a 120° side-cut flaps (n=34) made with a femtosecond laser. One day, 1wk, 1 and 3mo postoperatively, side-cut scar was evaluated under slit-lamp photography according to a new grading system (Grade 0=transparent scar, 1=faint healing opacity, and 2=evident healing opacity). In vivo corneal confocal microscopy and anterior segment optical coherence tomography (AS-OCT) were used to observe wound-healing patterns around flap margin in the two groups. Sirius Scheimpflug Analyzer was also used to analyze higher order aberrations 3mo after surgery. RESULTS: There were no significant differences in flap wound-healing patterns at each follow up between the two groups (P>0.05). Three months after surgery, the flap edge scar classified as Grade 0 had excellent apposition and rapid nerve regeneration. At 3 mm and 5 mm pupil diameters, there were significant differences in trefoil aberrations between the two groups (P0.05). CONCLUSION: Flap edge scars classified as Grade 0 have excellent apposition and rapid nerve regeneration, and 120° side-cut angle flaps induce less trefoil aberrations after FS-LASIK.

BG45 is a class Ⅰ histone deacetylase inhibitor (HDACI) with selectivity for HDAC3. Our previous study demonstrated that BG45 can upregulate the expression of synaptic proteins and reduce the loss of neurons in the hippocampus of APPswe/PS1dE9 (APP/PS1) transgenic mice (Tg). The entorhinal cortex is a pivotal region that, along with the hippocampus, plays a critical role in memory in the Alzheimer’s disease (AD) pathology process. In this study, we focused on the inflammatory changes in the entorhinal cortex of APP/PS1 mice and further explored the therapeutic effects of BG45 on the pathologies. The APP/PS1 mice were randomly divided into the transgenic group without BG45 (Tg group) and the BG45-treated groups. The BG45-treated groups were treated with BG45 at 2 months (2 m group), at 6 months (6 m group), or twice at 2 and 6 months (2 and 6 m group). The wild-type mice group (Wt group) served as the control. All mice were killed within 24 h after the last injection at 6 months. The results showed that amyloid-β (Aβ) deposition and IBA1-positive microglia and GFAP-positive astrocytes in the entorhinal cortex of the APP/PS1 mice progressively increased over time from 3 to 8 months of age. When the APP/PS1 mice were treated with BG45, the level of H3K9K14/H3 acetylation was improved and the expression of histonedeacetylase1, histonedeacetylase2, and histonedeacetylase3 was inhibited, especially in the 2 and 6 m group. BG45 alleviated Aβ deposition and reduced the phosphorylation level of tau protein. The number of IBA1-positive microglia and GFAP-positive astrocytes decreased with BG45 treatment, and the effect was more significant in the 2 and 6 m group. Meanwhile, the expression of synaptic proteins synaptophysin, postsynaptic density protein 95, and spinophilin was upregulated and the degeneration of neurons was alleviated. Moreover, BG45 reduced the gene expression of inflammatory cytokines interleukin-1β and tumor necrosis factor-α. Closely related to the CREB/BDNF/NF-kB pathway, the expression of p-CREB/CREB, BDNF, and TrkB was increased in all BG45 administered groups compared with the Tg group. However, the levels of p-NF-kB/NF-kB in the BG45 treatment groups were reduced. Therefore, we deduced that BG45 is a potential drug for AD by alleviating inflammation and regulating the CREB/BDNF/NF-kB pathway, and the early, repeated administration of BG45 can play a more effective role.

Here the authors demonstrate that during lipid overload, the liver secretes miRNA-containing EVs through a Ggpps-Rab27 dependent mechanism, which controls adipose tissue lipid storage capacity.

White celery (Apium graveolens L.), a variety of common celery, is an important leafy vegetable in the Apiaceae family and is famous for its nutritional value. However, limited work has been devoted to quality formation and regulation in white celery. In this study, four white celery varieties, ‘Xuebaiqincai’, ‘Saixue’, ‘Baiganshiqin’ and ‘Ruixue’, were selected and analyzed for the comparison of ascorbic acid (AsA) and lignin levels, which are two important quality evaluation indicators in celery. The expression levels of the genes involved in AsA and lignin metabolic pathways were also detected. In the leaf blades, the AsA content was highest in ‘Xuebaiqincai’ compared with other varieties, whereas the most abundant AsA levels in the petioles were observed in ‘Baiganshiqin’ and ‘Ruixue’. The expression levels of AsA-related genes varied among the studied varieties. The highest level was detected in ‘Xuebaiqincai’, whereas other varieties exhibited relatively lower levels. The lignin content in the leaf blades was lower than that in the petioles. Correspondingly, the transcript profiles of genes involved in lignin biosynthesis were in accordance with the different levels in the petioles and blades. The results of this study provide potentially useful information for white celery breeding aimed at quality improvement and regulation.

Endoplasmic reticulum stress (ER stress) is a condition that is defined by abnormal accumulation of unfolded proteins. It plays an important role in maintaining cellular protein, lipid, and ion homeostasis. By triggering the unfolded protein response (UPR) under ER stress, cells restore homeostasis or undergo apoptosis. Chronic ER stress is implicated in many human diseases. Despite extensive studies on related signaling mechanisms, reliable image biomarkers for ER stress remain lacking. To address this deficiency, we have validated a morphological image biomarker for ER stress and have developed a deep learning-based assay to enable automated detection and analysis of this marker for screening studies. Specifically, ER under stress exhibits abnormal morphological patterns that feature ring-shaped structures called whorls (WHs). Using a highly specific chemical probe for unfolded and aggregated proteins, we find that formation of ER whorls is specifically associated with the accumulation of the unfolded and aggregated proteins. This confirms that ER whorls can be used as an image biomarker for ER stress. To this end, we have developed ER-WHs-Analyzer, a deep learning-based image analysis assay that automatically recognizes and localizes ER whorls similarly as human experts. It does not require laborious manual annotation of ER whorls for training of deep learning models. Importantly, it reliably classifies different patterns of ER whorls induced by different ER stress drugs. Overall, our study provides mechanistic insights into morphological patterns of ER under stress as well as an image biomarker assay for screening studies to dissect related disease mechanisms and to accelerate related drug discoveries. It demonstrates the effectiveness of deep learning in recognizing and understanding complex morphological phenotypes of ER.

The production processes for the gypsum- and cement-based materials, which are widely used in civil engineering projects, require the consumption of large amounts of energy and natural resources. At the present time, the main development directions of civil engineering material include realizing solid waste recycling and the applications of green building materials, which can be then used in the fields of disaster treatment and ecological restoration, such as grouting materials. In addition, the grouting quality is always difficult to detect. This study examined the gypsum- and cement-based green grouting materials made from mineral solid waste used in disaster and cloud detection by means of electromagnetic characteristics. According to the requirements of electrical testing of grouting quality, first we designed a material made with solid waste. Then, we studied the resistivity, strength, porosity, and other physical and mechanical properties of the material. The test results of the material and its soaking solution indicate that this material is a green material meeting the environmental safety standards. The experimental results show that the rapid detection of grouting quality can be realized. The following effects on electromagnetic characteristics and mechanical properties of the green materials of gypsum- and cement-based were analyzed. The results showed that the resistivity and strength were related to the ratios of the water content to the modified phosphogypsum; the percentages of cement content in the total amount of modified phosphogypsum and marble powder; the percentages of marble powder in the modified phosphogypsum; the percentages of steel fiber content in the phosphogypsum; and the fineness of the marble powder. The field test results also confirm that the green grouting material can be effectively applied in engineering repair imaging monitoring.

ObjectiveOur study aims to clarify the role of estradiol and leptin in breast cancer risk and prognostic assessment in postmenopausal Chinese women.DesignThe serum circulating estradiol and leptin level was detected by ELISA. Then the correlation between estradiol, leptin level, and clinical characteristics was analyzed using Fisher’s exact test. Next, the Kaplan-Meier model was used to analyze the association between estradiol, leptin, and prognosis of postmenopausal breast cancer patients in our cohort and the TCGA dataset.SettingThe study was conducted at the National Cancer Center, Cancer Hospital, Chinese Academy of Medical Sciences, and Peking Union Medical College.PatientsA total of 182 postmenopausal breast cancer patients and 111 healthy subjects from January 2010 to August 2010 were included in the analysis. Another 702 cases with breast cancer were retrieved from The Cancer Genome Atlas (TCGA) database for subsequent analysis.Main Outcome MeasureSerum circulating estradiol and leptin level.ResultsThe level of estradiol was significantly higher (P0.05). Estradiol was significantly correlated with tumor T stage (P = 0.002) and leptin was significantly associated with perineural invasion (P = 0.014). In addition, the disease-free survival of patients with a high level of estradiol was significantly shorter (P = 0.025) but leptin tended to be a protective factor for overall survival in TCGA analysis (P = 0.038).ConclusionCirculating estradiol and leptin played important roles in the risk of postmenopausal breast cancer even in low-estrogen nations with an independent expression of ER status. High circulating estradiol was a poor prognostic factor and leptin may be a protection signal in Chinese postmenopausal patients with breast cancer.

Alzheimer’s disease (AD) seriously endangers the health and life of elderly individuals worldwide. However, despite all scientific efforts, at the moment there are no effective clinical treatment options for AD. In this work, the effect of the class I histone deacetylase inhibitor (HDACI) BG45 on synapse-related proteins was investigated in primary neurons from APP/PS1 transgenic mice. The results showed that BG45 can upregulate the expression of synaptotagmin-1 (SYT-1) and neurofilament light chain (NF-L) in primary neurons. In vivo, the APPswe/PS1dE9 (APP/PS1) transgenic mice were treated with BG45 (30 mg/kg) daily for 12 days. Behavioral testing of BG45-treated APP/PS1 mice showed improvements in learning and memory. BG45 can alleviate damage to the dendritic spine and reduce the deposition of Aβ. Similar to the in vitro results, synapse-related proteins in the prefrontal cortex were increased after BG45 treatment. Proteomic analysis results highlighted the differences in the biological processes of energy metabolism and calmodulin regulation in APP/PS1 mice with or without BG45 treatment. Further verification demonstrated that the effect of BG45 on synapses and learning and memory may involve the CaMKII/ITPKA/Ca2+ pathway. These results suggest that class I HDACI BG45 might be a promising drug for the early clinical treatment of AD.

Aggressive behavior is one of congenital social behaviors in many species, which could be promoted by social neglect or isolation in the early stages of life. Many brain regions including the medial prefrontal cortex (mPFC), medial amygdala (MeA) and ventromedial hypothalamus (VMH) are demonstrated to relate to aggressive behavior; however, the dynamic patterns of neural activities during the occurrence of this behavior remain unclear. In this study, 21-day-old male CD-1 mice were reared in social isolation conditions and cohousing conditions for two weeks. Aggressive behaviors of each subject were estimated by the resident–intruder test. Simultaneously, the local field potentials of mPFC, MeA and VMH were recorded for exploring differences in the relative power spectra of different oscillations when aggressive behaviors occurred. The results showed that the following: (1) Compared with the cohousing mice, the socially isolated mice exhibited more aggression. (2) Regardless of “time condition” (pre-, during- and post- attack), the relative power spectra of beta band in the cohousing mice were significantly greater than those in the socially isolated mice, and inversely, the relative power spectra of gamma band in the cohousing mice were significantly smaller than those in the socially isolated mice. (3) The bilateral mPFC exhibited significantly smaller beta power spectra but greater gamma power spectra compared with other brain areas regardless of rearing patterns. (4) For the right VMH of the socially isolated mice, the relative power spectra of the gamma band during attacks were significantly greater than those before attack. These results suggest that aggressive behaviors in mice could be shaped by rearing patterns and that high-frequency oscillations (beta and gamma bands) may engage in mediating aggressive behaviors in mice.

AIM: To investigate differences between 6.0mm and 6.5mm optical zones in visual quality after FS-LASIK. METHODS: This was a prospective study. Totally 25 patients(50 eyes)treated with FS-LASIK were included and they were classified into 6.0mm group and 6.5mm group according to optical zone. Objective visual quality was evaluated by OQASⅡoptical quality analysis system(including OSI, Strel Ratio, MTF cut off, OV100%, OV20%, OV9%)and by Sirius to obtain the higher order aberrations parameters(including total HOAs, spherical aberrations, trefoil and coma)at 3mm, 5mm and 7mm pupil sizes. OSDI questionnaire was applied to evaluate patients' subjective visual quality. RESULTS: At 3mo postoperatively, there were no significant differences in total HOAs, spherical aberrations, trefoil and coma under 3mm pupil size(all P>0.05). Significant differences were only observed in spherical aberrations at 5mm pupil size(PPP>0.05).CONCLUSION: HOAs under scotopic conditions could be less with 6.5mm optic zone after FS-LASIK, however, both 6.0mm and 6.5mm optical zones could achieve good subjective and objective visual quality after FS-LASIK.

AIM: To investigate the related factors of orthokeraology(ortho-K)decentration.METHODS: Within a prospective study, 88 eyes of 88 patients who visited to our center between July 2014 and December 2016 were enrolled. To verify the possible role of the spherical equivalent(SE), the corneal astigmatism(CA), e value, steep K, flat K, front curvature asymmetry index(SIf)and age in the ortho-K location prediction.RESULTS: This study included 24 eyes of grade Ⅰ decentration, 26 eyes with grade Ⅱ, 25 eyes with grade Ⅲ, and 13 eyes with grade Ⅳ, by which based on the decentration system that published by our team. One-way ANOVA showed that there was statically significant difference of SE, Ks and Kf in different grading groups(PPCONCLUSION: The SE might be the main factor which lead to the ortho-K decentration.

Abstract Background Animal vocal signals encode very important information for communication during which the importance of temporal and spectral characteristics of vocalizations is always asymmetrical and species-specific. However, it is still unknown how auditory system represents this asymmetrical and species-specific patterns. In this study, auditory event related potential (ERP) changes were evaluated in the Emei music frog (Babina daunchina) to assess the differences in eliciting neural responses of both temporal and spectral features for the telencephalon, diencephalon and mesencephalon respectively. To do this, an acoustic playback experiment using an oddball paradigm design was conducted, in which an original advertisement call (OC), its spectral feature preserved version (SC) and temporal feature preserved version (TC) were used as deviant stimuli with synthesized white noise as standard stimulus. Results The present results show that 1) compared with TC, more similar ERP components were evoked by OC and SC; and 2) the P3a amplitudes in the forebrain evoked by OC were significantly higher in males than in females. Conclusions Together, the results provide evidence for suggesting neural processing for conspecific vocalization may prefer to the spectral features in the music frog, prompting speculation that the spectral features may play more important roles in auditory object perception or vocal communication in this species. In addition, the neural processing for auditory perception is sexually dimorphic.

Since its discovery in Escherichia coli, the emergence and rapid spread of the plasmid-mediated colistin resistance gene mcr-1 have become a public health concern. Livestock manure is a potentially important reservoir of mcr-1 because colistin has been widely used in livestock production. Efforts made to accurately quantify the prevalence of mcr-1 in livestock manure and the dynamic changes therein during thermophilic composting have been few and far between. In this study, mcr-1 in 51 collected samples from four kinds of livestock manures was detected and quantified. In total, 16 manure samples were found to be mcr-1 positive, with a detection frequency of 31% in 51 samples. The numbers of mcr-1 gene copies in 12 positive manure samples with a high prevalence of mcr-1 were 107–109 copies/g dry weight. During composting, >90% of mcr-1 in the manure was eliminated in 15 days at high temperature (44–65 °C), and mcr-1 was completely undetectable after 22 days. The reduction of mcr-1 following manure composting may be ascribed to the decreased number of potential mcr-1–harboring bacteria, Enterobacteriaceae and Pseudomonas. The results indicated that thermophilic composting effectively eliminated mcr-1 and inhibited its spread from livestock manure to the environment. Attention, referring to selective processing of task-related information, is central to cognition. It has been proposed that voluntary attention (driven by current goals or tasks and under top-down control) and reflexive attention (driven by stimulus salience and under bottom-up control) struggle to control the focus of attention with interaction in a push–pull fashion for everyday perception in higher vertebrates. However, how auditory attention engages in auditory perception in lower vertebrates remains unclear. In this study, each component of auditory event-related potentials (ERP) related to attention was measured for the telencephalon, diencephalon and mesencephalon in the Emei music frog (Nidirana daunchina), during the broadcasting of acoustic stimuli invoking voluntary attention (using binary playback paradigm with silence replacement) and reflexive attention (using equiprobably random playback paradigm), respectively. Results showed that (1) when the sequence of acoustic stimuli could be predicted, the amplitudes of stimulus preceding negativity (SPN) evoked by silence replacement in the forebrain were significantly greater than that in the mesencephalon, suggesting voluntary attention may engage in auditory perception in this species because of the correlation between the SPN component and top-down control such as expectation and/or prediction; (2) alternately, when the sequence of acoustic stimuli could not be predicted, the N1 amplitudes evoked in the mesencephalon were significantly greater than those in other brain areas, implying that reflexive attention may be involved in auditory signal processing because the N1 components relate to selective attention; and (3) both SPN and N1 components could be evoked by the predicted stimuli, suggesting auditory perception of the music frogs might invoke the two kind of attention resources simultaneously. The present results show that human-like ERP components related to voluntary attention and reflexive attention exist in the lower vertebrates also.

The cucumber (Cucumis sativus) is characterized by its diversity and seasonal plasticity in sexual type. A long day length condition significantly decreased the cucumber female flower ratio by 17.7–52.9%, and the effect of photoperiod treatment is more significant under low temperature than under high temperature. Transcriptome analysis indicates that the photoperiod treatment preferentially significantly influenced flower development processes, particularly MADS-box genes in shoot apices. The long-day treatment resulted in predominantly transposable element (TE)- and gene-associated CHH-types of DNA methylation changes. Nevertheless, there was significant enrichment of CG- and CHG-types of DNA methylation changes nearing transcription start sites (TSSs)/transcription end sites (TESs) and gene bodies, respectively. Predominantly negative association between differentially methylated regions (DMRs) and differentially expressed genes (DEGs) were observed which implied epiregulation of DEGs. Two MADS-box genes that were significantly downregulated by long photoperiod showed significant hypermethylation in promoter regions that is essentially TE-rich. This study indicates MADS-box genes which are partially regulated by promoter methylation state may mediate photoperiod-dependent regulation of cucumber sex expression.

Abstract Background Cucumbers (Cucumis sativus) are known for their plasticity in sex expression. DNA methylation status determines gene activity but is susceptible to environmental condition changes. Thus, DNA methylation-based epigenetic regulation may at least partially account for the instability of cucumber sex expression. Do temperature and photoperiod that are the two most important environmental factors have equal effect on cucumber sex expression by similar epigenetic regulation mechanism? To answer this question, we did a two-factor experiment of temperature and photoperiod and generated methylome and transcriptome data from cucumber shoot apices. Results The seasonal change in the femaleness of a cucumber core germplasm collection was investigated over five consecutive years. As a result, 71.3% of the 359 cucumber accessions significantly decreased their femaleness in early autumn when compared with spring. High temperature and long-day photoperiod treatments, which mimic early autumn conditions, are both unfavorable for female flower formation, and temperature is the predominant factor. High temperatures and long-day treatments both predominantly resulted in hypermethylation compared to demethylation, and temperature effect was decisive. The targeted cytosines shared in high-temperature and long-day photoperiod treatment showed the same change in DNA methylation level. Moreover, differentially expressed TEs (DETs) and the predicted epiregulation sites were clustered across chromosomes, and importantly, these sites were reproducible among different treatments. Essentially, the photoperiod treatment preferentially and significantly influenced flower development processes, while temperature treatment produced stronger responses from phytohormone-pathway-related genes. Cucumber AGAMOUS was likely epicontrolled exclusively by photoperiod while CAULIFLOWER A and CsACO 3 were likely epicontrolled by both photoperiod and temperature. Conclusions Seasonal change of sex expression is a germplasm-wide phenomenon in cucumbers. High temperature and long-day photoperiod might have the same effect on the methylome via the same mechanism of gene-TE interaction but resulted in different epicontrol sites that account for different mechanisms between temperature- and photoperiod-dependent sex expression changes.

Background Anthracycline-induced liver injury (AILI) is one of the serious complications of anthracycline-based adjuvant chemotherapy for postoperative breast cancer patients. Exosomal miRNAs, as signaling molecules in intercellular communication, play the essential roles in drug-induced liver injury (DILI). However, the expression profiles of them in patients with AILI remains unknown. Methods Seven post-chemotherapy patients were recruited in this study. After isolated plasma-derived exosomes, small RNA sequencing revealed exosomal miRNA profiles and differentially expressed miRNAs (DE-miRNAs) were identified between the liver injury group and non-liver injury group. miRTarBase and miRDB were used to predict the potential target genes of DE-miRNAs. DILI-related genes were downloaded from the CTD Database. The intersection of predicted genes and DILI-related genes were identified as the AILI-related target genes of the DE-miRNAs. GO annotation and KEGG pathway enrichment analysis were performed by the DAVID database. Furthermore, the protein-protein interaction (PPI) network was established by the STRING database and essential exosomal miRNAs were identified via Cytoscape software. Results A total of 30 DE-miRNAs and 79 AILI-related target genes were identified. AILI-related target genes of the DE-miRNAs are significantly enriched in NOD-like receptor signaling pathway, the HIF-1 signaling pathway, and the FoxO signaling pathway. Then, the hub genes were screened and we discovered that IL-6 and SOD2 are the most critical genes that may be involved in the development of AILI through the activation of immune response and the occurrence of oxidative stress, respectively. In addition, we found that miR-1-3p could potentially regulate most of the hub genes in the miRNA-hub gene network. Conclusion We explored the potential functions of DE-miRNAs and suggested exosomal miR-1-3p might be the essential exosomal miRNA in the pathogenesis of AILI. Moreover, our study provided an experimental basis for experimental verification to reveal the actual function and mechanism of miRNAs in AILI.

FAM172A, as a newly discovered gene, is little known in cancer development, especially in pancreatic cancer (PC). We investigated the potential role and molecular mechanism of FAM172A in epithelial to mesenchymal transition (EMT) in both human clinical samples and PC cells. FAM172A was downregulated in human PC tissues compared with that in non-cancerous pancreas cells by immunohistochemistry and qRT-PCR. FAM172A expression was negatively associated with tumor size (P=0.015), T stage (P=0.006), lymph node metastasis (P=0.028) and the worst prognosis of PC patients (P=0.004). Meanwhile, a positive relationship between FAM172A and E-cadherin (E-cad) (r=0.381, P=0.002) was observed in clinical samples, which contributed to the better prognosis of PC patients (P=0.014). FAM172A silencing induced EMT in both AsPC-1 and BxPC-3 cells, including inducing the increase of Vimentin, MMP9 and pERK and the decrease of E-cad and β-catenin expression, stimulating EMT-like cell morphology and enhancing cell invasion and migration in PC cells. However, MEK1 inhibitor PD98059 reversed FAM172A silencing-enhanced EMT in PC cells. We conclude that FAM172A inhibits EMT of PC cells via ERK-MAPK signaling.

In order to improve the ability of airspace management, a multi-attribute decision-making tool based on q-rung orthopair probability hesitant fuzzy GRA-TOPSIS is proposed to solve the problem of airspace operation effectiveness evaluation; this is in view of the fact that there are few airspace operation effectiveness evaluation methods in general aviation airports. Firstly, taking general aviation airports as the research object, a complete airspace operation effectiveness evaluation system is newly established, its evaluation indicators are introduced, and its multi-attribute decision-making ideas are explained. Then, based on the q-rung orthopair probability hesitant fuzzy set, a new distance measure and information aggregation operator are defined, which can better deal with symmetry information. Secondly, we build a deviation maximization model to calculate the attribute weights of indicator elements in the decision-making process. Then, we combine the GRA method and TOPSIS method to rank the airspace operation effectiveness evaluation schemes. Finally, combined with calculation examples and comparative analysis, the reliability and rationality of the method proposed in this paper are verified, and the symmetry relationship between the evaluation results and the actual situation is better reflected. Experiments show that the method proposed in this paper can obtain more accurate airspace operation effectiveness evaluation results, and can provide reference for related research.

Background and objective Small cell lung cancer (SCLC) is highly malignant and prone to bone marrow metastasis in early stage, but its related reports are limited. This study analyzed the clinical feature, laboratory examination, treatment and prognosis of SCLC patients with bone marrow metastasis. Methods The clinical data of 26 SCLC patients with bone marrow metastasis were analyzed retrospectively. Prognostic factors were evaluated. Results The median age of 26 patients was 57 years and the median time from diagnosis of SCLC to confirmed bone marrow metastases was 8 d. Most patients (96.2%) were accompanied by other organ metastases. The most common laboratory abnormalities were elevated lactate dehydrogenase in 19 cases (73.1%), thrombocytopenia and elevated alkaline phosphatase respectively in 11 cases (42.3%) and anemia in 7 cases (26.9%). Twenty patients had received chemotherapy and the remaining 6 patients had not. Of this group, 16 patients received at least 2 cycles of chemotherapy after the diagnosis of bone marrow metastasis. The median survival time was 15.7 wk (0.1 wk-82.9 wk) after diagnosis of bone marrow metastasis. The survival of patients with chemotherapy was significantly better than that of those without chemotherapy (χ2=33.768, P

Raphanus has undergone a lengthy evolutionary process and has rich diversity. However, the inter- and intraspecific phylogenetic relationships and genetic diversity of this genus are not well understood. Through SSR-sequencing and multi-analysis of 939 wild, semi-wild and cultivated accessions, we discovered that the European wild radish (EWR) population is separated from cultivated radishes and has a higher genetic diversity. Frequent intraspecific genetic exchanges occurred in the whole cultivated radish (WCR) population; there was considerable genetic differentiation within the European cultivated radish (ECR) population, which could drive radish diversity formation. Among the ECR subpopulations, European primitive cultivated radishes (EPCRs) with higher genetic diversity are most closely related to the EWR population and exhibit a gene flow with rat-tail radishes (RTRs) and black radishes (BRs)/oil radishes (ORs). Among Asian cultivated radishes (ACRs), Chinese big radishes (CBRs) with a relatively high diversity are furthest from the EWR population, and most Japanese/Korean big radishes (JKBRs) are close to CBR accessions, except for a few old Japanese landraces that are closer to the EPCR. The CBR and JKBR accessions are independent of RTR accessions; however, phylogenetic analysis indicates that the RTR is sister to the clade of CBR (including JWR), which suggests that the RTR may share the most recent common ancestry with CBRs and JWRs. In addition, Japanese wild radishes (JWRs), (namely, R. sativus forma raphanistroides) are mainly scattered between CBRs and EPCRs in PCoA analysis. Moreover, JWRs have a strong gene exchange with the JKBR, OR and RTR subpopulations. American wild radishes (AWRs) are closely related to European wild and cultivated radishes, and have a gene flow with European small radishes (ESRs), suggesting that the AWR developed from natural hybridization between the EWR and the ESR. Overall, this demonstrates that Europe was the origin center of the radish, and that Europe, South Asia and East Asia appear to have been three independent domestication centers. The EPCR, AWR and JWR, as semi-wild populations, might have played indispensable transitional roles in radish evolution. Our study provides new perspectives into the origin, evolution and genetic diversity of Raphanus and facilitates the conservation and exploitation of radish germplasm resources.

Objective: A large portion of non-metastatic colorectal cancers (non-mCRCs) recur after curative surgery. In addition to the traditional tumor-related factors, host-related factors are also required to accurately predict prognosis. A few studies have shown an association between the serum lipid profile and the survival and treatment response of patients with colorectal cancer. Methods: We retrospectively evaluated the prognostic significance of the preoperative serum lipid profile [total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C)] in patients with non-mCRC treated with curative surgery. The Spearman rank correlation test was used to analyze associations between lipid levels and categorical variables. Lipid levels were modeled as four equal-sized quartiles based on the distribution among the whole cohort. Kaplan-Meier curves were used to estimate survival probabilities, and the log-rank test was used to detect differences between them. Multivariate fractional polynomial (MFP) analysis was used to model any non-linear effects and avoid categorization. To evaluate the added prognostic value of lipids, the predictive power of two models (with and without lipids as covariates) was compared by using Harrell's C-statistic and the Akaike information criterion (AIC). Results: A total of 266 patients with non-mCRC were enrolled in the present study. Spearman rank correlation test showed that TGÂ levels inversely correlated with N stage (r = â0.20, PÂ =Â 0.00) and Tumor-Node-Metastasis (TNM) stage (r = â0.19, P = 0.00). HDL-C levels positively correlated with perineural invasion (PNI) (r = 0.15, PÂ = 0.02), and LDL-C levels inversely correlated with lymphovascular invasion (LVI) (r = â0.12, PÂ =Â 0.04). None of the four lipids predicted overall survival (OS) in univariate or multivariate analyses adjusted for age, gender, T stage, N stage, TNM stage, histological grade, tumor deposits, LVI, PNI, and adjuvant treatment (all P > 0.05). In agreement, the Kaplan-Meier curves for OS according to the lipid quartiles were not significantly different, as confirmed by the log-rank test (all P > 0.05). MFPÂ analysis also found no significant associations between lipid levels and OS (all P > 0.05). A prognostic model that included lipids had a higher Harrell's C-statistic and a lower AIC value than did a model that did not include lipids (for Harrell's C-statistic: 0.82 vs. 0.77; for AIC: 398 vs. 432). Conclusion: Measuring preoperative serum lipid levels may be a simple and cost-effective way of increasing prognostic accuracy in patients with non-mCRC treated with curative surgery. AIM:To investigate the changes in anterior segment parameters by Sirius anterior system after orthokeratology and the possible mechanisms. METHODS: Twenty-eight patients(56 eyes)with myopia in our hospital from August 2014 to October 2014 were included. Spherical equivalent were -3.83±1.19D. Data of central corneal thickness(CCT), anterior chamber depth(ACD), anterior corneal curvature(a-K)and posterior corneal curvature(p-K)were collected and analyzed at before, 1d, 1wk and 3mo after orthokeratology.RESULTS: After orthokeratology, spherical equivalent of all eyes decreased to predicted level. There were no significant differences in CCT and p-K between different times(P>0.05). There were significant differences in a-K between different times except between 1wk and 3mo after orthokeratology(PPPP=0.016), a-Kc-1wk(P=0.047)were correlated with a-Kpre and axial, respectively. And there were correlations between a-Kpre and a-Kc-3mo(P=0.024), between ACDpre and ACDc-1d(P=0.036), between CCTpre and CCTc-3mo(P=0.012), respectively.CONCLUSION: CCT, ACD and p-K has no significant changes after orthokeratology, while there is significant difference in a-K, which is considered one of the most key factor in orthokeratology. Sirius anterior system for examination before orthokeratology and postoperative assessment, non-invasive, reliable and has a good clinical value.

AIM:To observe the effect of posterior chamber phakic intraocular lens implantation for high myopia on the stereopsis. To evaluate the implantation of implantable contact lens from the highest form of visual angle. METHODS:Fourty-six patients(92 eyes)with high myopia were included from January to December 2014. The average age of patients was 26.13±3.37(19～32)years, and 20 males(43%)and 26 females(57%)were observed. Analysis the stereopsis of these patients before and after the implantation of posterior chamber phakic intraocular lens. RESULTS:The mean spherical equivalent of all the eyes was 10.02±2.71D before and -0.02±0.25D after surgeries. There was significant difference on the spherical equivalent(t=14.062, P=0.000).There were 6 patients with the blind of near stereopsis, the near stereopsis of the rest patients were 637.50±462.08arcsec before the surgeries and there were none with the blind of near stereopsis after surgeries. The near stereopsis of all the patients were 126.09±165.78arcsec after surgeries. There were 12 patients with the blind of far stereopsis, the far stereopsis of the rest patients were 400.59±257.00arcsec before the surgeries. After surgeries there were 2 patients with the blind of far stereopsis, the far stereopsis of the rest patients were 152.73±111.65arcsec. There were both significant differences on the near stereopsis(Z=-5.725, P=0.000)and the far stereopsis(Z=-4.976,P=0.000)before and after surgeries.CONCLUSION:Posterior chamber phakic intraocular lens implantation not only correct the ametropic of the high myopia, but also improve the visual acuity and the stereopsis.

BACKGROUND: The diamondback moth (DBM, Plutella xylostella) is a crucifer-specific pest that causes significant crop losses worldwide. Barbarea vulgaris (Brassicaceae) can resist DBM and other herbivorous insects by producing feeding-deterrent triterpenoid saponins. Plant breeders have long aimed to transfer this insect resistance to other crops. However, a lack of knowledge on the biosynthetic pathways and regulatory networks of these insecticidal saponins has hindered their practical application. A pyrosequencing-based transcriptome analysis of B. vulgaris during DBM larval feeding was performed to identify genes and gene networks responsible for saponin biosynthesis and its regulation at the genome level. PRINCIPAL FINDINGS: Approximately 1.22, 1.19, 1.16, 1.23, 1.16, 1.20, and 2.39 giga base pairs of clean nucleotides were generated from B. vulgaris transcriptomes sampled 1, 4, 8, 12, 24, and 48 h after onset of P. xylostella feeding and from non-inoculated controls, respectively. De novo assembly using all data of the seven transcriptomes generated 39,531 unigenes. A total of 37,780 (95.57%) unigenes were annotated, 14,399 of which were assigned to one or more gene ontology terms and 19,620 of which were assigned to 126 known pathways. Expression profiles revealed 2,016-4,685 up-regulated and 557-5188 down-regulated transcripts. Secondary metabolic pathways, such as those of terpenoids, glucosinolates, and phenylpropanoids, and its related regulators were elevated. Candidate genes for the triterpene saponin pathway were found in the transcriptome. Orthological analysis of the transcriptome with four other crucifer transcriptomes identified 592 B. vulgaris-specific gene families with a P-value cutoff of 1e(-5). CONCLUSION: This study presents the first comprehensive transcriptome analysis of B. vulgaris subjected to a series of DBM feedings. The biosynthetic and regulatory pathways of triterpenoid saponins and other DBM deterrent metabolites in this plant were classified. The results of this study will provide useful data for future investigations on pest-resistance phytochemistry and plant breeding.

Knowing the extent and structure of genetic variation in germplasm collections is essential for the conservation and utilization of biodiversity in cultivated plants. Cucumber is the fourth most important vegetable crop worldwide and is a model system for other Cucurbitaceae, a family that also includes melon, watermelon, pumpkin and squash. Previous isozyme studies revealed a low genetic diversity in cucumber, but detailed insights into the crop's genetic structure and diversity are largely missing. We have fingerprinted 3,342 accessions from the Chinese, Dutch and U.S. cucumber collections with 23 highly polymorphic Simple Sequence Repeat (SSR) markers evenly distributed in the genome. The data reveal three distinct populations, largely corresponding to three geographic regions. Population 1 corresponds to germplasm from China, except for the unique semi-wild landraces found in Xishuangbanna in Southwest China and East Asia; population 2 to Europe, America, and Central and West Asia; and population 3 to India and Xishuangbanna. Admixtures were also detected, reflecting hybridization and migration events between the populations. The genetic background of the Indian germplasm is heterogeneous, indicating that the Indian cucumbers maintain a large proportion of the genetic diversity and that only a small fraction was introduced to other parts of the world. Subsequently, we defined a core collection consisting of 115 accessions and capturing over 77% of the SSR alleles. Insight into the genetic structure of cucumber will help developing appropriate conservation strategies and provides a basis for population-level genome sequencing in cucumber.
