Abstract

Z-nucleic acid structures play vital roles in cellular processes and have implications in innate immunity due to their recognition by Zα domains containing proteins (Z-DNA/Z-RNA binding proteins, ZBPs). Although Zα domains have been identified in six proteins, including viral E3L, ORF112, and I73R, as well as, cellular ADAR1, ZBP1, and PKZ, their prevalence across living organisms remains largely unexplored. In this study, we introduce a computational approach to predict Zα domains, leading to the revelation of previously unidentified Zα domain-containing proteins in eukaryotic organisms, including non-metazoan species. Our findings encompass the discovery of new ZBPs in previously unexplored giant viruses, members of the Nucleocytoviricota phylum. Through experimental validation, we confirm the Zα functionality of select proteins, establishing their capability to induce the B-to-Z conversion. Additionally, we identify Zα-like domains within bacterial proteins. While these domains share certain features with Zα domains, they lack the ability to bind to Z-nucleic acids or facilitate the B-to-Z DNA conversion. Our findings significantly expand the ZBP family across a wide spectrum of organisms and raise intriguing questions about the evolutionary origins of Zα-containing proteins. Moreover, our study offers fresh perspectives on the functional significance of Zα domains in virus sensing and innate immunity and opens avenues for exploring hitherto undiscovered functions of ZBPs.

Abstract

BackgroundAs we move toward a polio-free world, the challenge for the polio program is to create an unrelenting focus on smaller areas where the virus is still present, where children are being repeatedly missed, where immunity levels are low, and where surveillance is weak. ObjectiveThis article aimed to describe a possible solution to address weak surveillance systems and document the outcomes of the deployment of the Auto-Visual Acute Flaccid Paralysis Detection and Reporting (AVADAR) project. MethodsThis intervention was implemented in 99 targeted high-risk districts with concerns for silent polio circulation from eight countries in Africa between August 1, 2017, and July 31, 2018. A total of 6954 persons (5390 community informants and 1564 health workers) were trained and equipped with a smartphone on which the AVADAR app was configured to allow community informants to send alerts on suspected acute flaccid paralysis (AFP) and allow health worker to use electronic checklists for investigation of such alerts. The AVADAR and Open Data Kit ONA servers were at the center of the entire process. A dashboard system and coordination teams for monitoring and supervision were put in place at all levels. ResultsOverall, 96.44% (24,142/25,032) of potential AFP case alerts were investigated by surveillance personnel, yielding 1414 true AFP cases. This number (n=1414) reported through AVADAR was higher than the 238 AFP cases expected during the study period in the AVADAR districts and the 491 true AFP cases reported by the traditional surveillance system. A total of 203 out of the 1414 true AFP cases reported were from special population settings, such as refugee camps and insecure areas. There was an improvement in reporting in silent health areas in all the countries using the AVADAR system. Finally, there were 23,473 reports for other diseases, such as measles, diarrhea, and cerebrospinal meningitis, using the AVADAR platform. ConclusionsThis article demonstrates the added value of AVADAR to rapidly improve surveillance sensitivity. AVADAR is capable of supporting countries to improve surveillance sensitivity within a short interval before and beyond polio-free certification.

5. The Role and Impact of Technology on Students' Reading

Abstract

Technology is gaining more and more place in our classrooms today, making its investigation more relevant than ever. Therefore, the author of this paper aimed to explore the role and impact of technology on students reading. To do so, the author has examined several aspects related to the use of technology for teaching and learning, including how technology can promote independent learning, communication, collaboration, assist students with learning needs, and certain limits of technology, before conducting research using interviews and observations with 86 participants (students, teachers, and parents). The research focused particularly on what role technologies such as text-to-speech apps, translators, electronic dictionaries, online reading apps and websites, etc., can play on students' reading, how they can impact students reading, and how they are perceived by students, teachers, and parents. At the end, the author provides some suggestions to consider for an effective use of these technologies.

Abstract

Knowledge of population distribution is critical for building infrastructure, distributing resources, and monitoring the progress of sustainable development goals. Although censuses can provide this information, they are typically conducted every ten years with some countries having forgone the process for several decades. Population can change in the intercensal period due to rapid migration, development, urbanisation, natural disasters, and conflicts. Census-independent population estimation approaches using alternative data sources, such as satellite imagery, have shown promise in providing frequent and reliable population estimates locally. Existing approaches, however, require significant human supervision, for example annotating buildings and accessing various public datasets, and therefore, are not easily reproducible. We explore recent representation learning approaches, and assess the transferability of representations to population estimation in Mozambique. Using representation learning reduces required human supervision, since features are extracted automatically, making the process of population estimation more sustainable and likely to be transferable to other regions or countries. We compare the resulting population estimates to existing population products from GRID3, Facebook (HRSL) and WorldPop. We observe that our approach matches the most accurate of these maps, and is interpretable in the sense that it recognises built-up areas to be an informative indicator of population., Comment: 14 pages, 5 figures

Abstract

Contemporary IoT environments, such as smart buildings, require end-users to trust data-capturing rules published by the systems. There are several reasons why such a trust is misplaced -- IoT systems may violate the rules deliberately or IoT devices may transfer user data to a malicious third-party due to cyberattacks, leading to the loss of individuals' privacy or service integrity. To address such concerns, we propose IoT Notary, a framework to ensure trust in IoT systems and applications. IoT Notary provides secure log sealing on live sensor data to produce a verifiable `proof-of-integrity,' based on which a verifier can attest that captured sensor data adheres to the published data-capturing rules. IoT Notary is an integral part of TIPPERS, a smart space system that has been deployed at the University of California Irvine to provide various real-time location-based services on the campus. We present extensive experiments over realtime WiFi connectivity data to evaluate IoT Notary, and the results show that IoT Notary imposes nominal overheads. Abstract

The clinical manifestations of neurofibromatosis type-1 have in common the presence of neurofibromas, schwannomas and cafe`-au-lait macules, which can potentially appear within any organ system of the body, involving primarily the skeleton, skin and soft tissues. The spinal thoracic tuberculoma in neurofibromatosis is exceptional. We report here, the case of a 31-year-old male with neurofibromatosis type-1 who presented a year agoa spinal thoracic pain. The evolution was marked by the appearance of a dorsal para-vertebral mass, progressively increasing in volume for 03 months. Neurological Examination revealed normal muscle bulk and tone, and power was grade 5/5. Deep tendon reflexes were brisk and the Babinski sign was present. The sensory deficit was present below the D9 level. The remainder of the examination found a voluminous dorsal para vertebral mass ovoid, renitent, and adhering to the superficial and the deep planes. There were also "café-au-lait" spots disseminated on his body. The imagery showed a formation with respect to D11-D12, isointense on T1 and T2, heterogeneously enhanced after contrast with central necrosis, with a foraminal starting point measuring 85x78x66 mm, with external canal extension. Laminectomy D11-D12 was performed with incomplete excision of the lesion. The histological examination showed casein-follicular vertebral tuberculosis, with secondary abdominal changes. Then the patient was put on anti-bacillary drugs. The evolution was marked two weeks later by the reappearance of the back pain and the back mass. On the 42nd day of treatment, the patient had a febrile consciousness disorder with a GCS of 8, right mydriasis and meningeal stiffness. Paraclinic investigations revealed tuberculosis meningoencephalitis, responsible for an active ventricular hydrocephalus associated with a left temporoparietal extrudal hematoma. An external ventricular shunt was performed as well as a left temporo-parietal extrudal hematoma evacuation. The patient died 7 days later in a poly visceral failure chart.

26. Towards Sustainable Census Independent Population Estimation in Mozambique

Abstract

Reliable and frequent population estimation is key for making policies around vaccination and planning infrastructure delivery. Since censuses lack the spatio-temporal resolution required for these tasks, census-independent approaches, using remote sensing and microcensus data, have become popular. We estimate intercensal population count in two pilot districts in Mozambique. To encourage sustainability, we assess the feasibility of using publicly available datasets to estimate population. We also explore transfer learning with existing annotated datasets for predicting building footprints, and training with additional `dot' annotations from regions of interest to enhance these estimations. Abstract

Xanthogranulomatous pyelonephritis is a rare and aggressive form of chronic pyelonephritis, it can occur at all age groups but is more common in women than in men, supposedly relating to the increased incidence of urinary tract infections and chronic nephrolithiasis in woman. Computed tomography (CT) findings are very helpful in making the correct diagnosis, but the definitive diagnosis is still based on histology, as there are many differential diagnoses such as renal cell carcinoma and renal tuberculosis. The complications of this type of pyelonephritis are due to the involvement of adjacent organs. The most frequent ones are Psoas abscess, perinephric abscess, and sepsis. Nephrocutaneous and renocolic fistulas are less common. We report a case of a 61-year-old male, who presented to emergency for left-sided lumbar pain for whom radiological investigations confirmed a renocolic fistula complicating xanthogranulomatous pyelonephritis. The diagnosis of XGP was proven by histopathological examination of the nephrectomy specimen slides, but there was also association with an underlying malignant squamous differentiation consistent with urothelial carcinoma, which was not evident on CT. XGP is a rare variant of chronic pyelonephritis with known imaging features. The treatment of choice is nephrectomy and histopathological examination is required for final diagnosis, as there may be associated renal malignancy.

Abstract

Real-time localization of prostate gland in trans-rectal ultrasound images is a key technology that is required to automate the ultrasound guided prostate biopsy procedures. In this paper, we propose a new deep learning based approach which is aimed at localizing several prostate landmarks efficiently and robustly. We propose a multitask learning approach primarily to make the overall algorithm more contextually aware. In this approach, we not only consider the explicit learning of landmark locations, but also build-in a mechanism to learn the contour of the prostate. This multitask learning is further coupled with an adversarial arm to promote the generation of feasible structures. We have trained this network using ~4000 labeled trans-rectal ultrasound images and tested on an independent set of images with ground truth landmark locations. We have achieved an overall Dice score of 92.6% for the adversarially trained multitask approach, which is significantly better than the Dice score of 88.3% obtained by only learning of landmark locations. The overall mean distance error using the adversarial multitask approach has also improved by 20% while reducing the standard deviation of the error compared to learning landmark locations only. In terms of computational complexity both approaches can process the images in real-time using standard computer with a standard CUDA enabled GPU.
