The lignocellulose based biorefinery process plays a significant role in the production of value-added products to achieve the sustainable development goals that aim for a bio-circular-green economy. Due to the recalcitrant nature of the biomass components, different types of pretreatment methods are essential to make available carbohydrates for saccharification and fermentation. The mechanism, advantages, challenges, innovations, and technologies of various chemical pretreatment processes are summarized in this review. Based on the literature survey, chemicals such as acids, alkalis, organosolv, oxidative agents, ionic liquids, and deep eutectic solvents are covered in the review. Moreover, combined chemical pretreatment strategies are also discussed in this study, which can be utilized in industries. There are many challenges, such as the efficiency of the process, reaction conditions, formation of inhibitors, techno-economic feasibility, and environmental sustainability, which require further investigations to overcome these limitations to choose the best option for the feasible process. This review could be the guideline for the future direction in the improvement of pretreatment methods for the synthesis of value-added products.

The pretreatment method is one of the challenging steps in the production of biofuel through the biorefinery process that unlocks the recalcitrant nature of lignocellulosic biomass. Ionic liquid pretreatment gained attention for being highly effective to improve the enzymatic saccharification of the biomass, however its high cost hinders its industrial application. In this study, the combined effect of ionic liquid 1- ethyl-3-methylimidazolium acetate (EMIM-Ac) with inorganic salts (NaCl and KCl) was used for the pretreatment of rice straw. Optimization of pretreatment was conducted based on Response Surface Methodology and sugar yields obtained by EMIM-Ac+NaCl (160 °C, 88.7 min, 7.6%wt) and EMIMAc+ KCl (160 °C, 68.2 min, 12.5%wt) were 670.7 and 392.9 mg/g-biomass, respectively. The effect of combined pretreatment on ethanol production was analyzed after 48h fermentation. The results showed that the ethanol yield from pretreated samples with EMIM-Ac+NaCl (0.72%) and EMIM+KCl (0.76%) was increased by 2.18 and 2.25 fold times, respectively, compared to untreated sample (0.33%). This combined effect of inorganic salts and ionic liquid significantly removed the lignin during pretreatment, while maintaining efficient enzymatic saccharification of rice straw. Thus, this cost-effective combined chemical method may be an alternative strategy for increasing cellulosic ethanol production.

Vibrio cholerae, a causative agent of the waterborne disease cholera, still threatens a large proportion of world's population. The role of biofilm formation in V. cholerae pathogenesis is well established, as it provides the bacterium enhanced tolerance to antimicrobial agents and increased transmission. In the present study, four medicinal plants used in traditional medicines with antidiarrheal properties were evaluated for its antibiofilm activity. Methanol extracts of these plants (Centella asiatica, Elephantopus scaber, Camellia sinensis, and Holarrhena antidysenterica) showed promising antibiofilm activity against V. cholerae with crystal violet and air-liquid interface coverslip assays. Results revealed that C. asiatica, E. scaber, C. sinensis, and H. antidysenterica extracts significantly inhibited biofilm formation by approximately 75, 76, 78, and 55% at concentrations of 3, 2, 1, and 0.6 mg/mL, respectively. A promising antibiofilm activity of ∼89% inhibition at 1.5 mg/mL concentration was observed when a combination of E. scaber and C. sinensis was used. The herbal extracts were thermostable at a temperature range of 40 to 100°C. The 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay revealed that the viability of bacteria was not affected by treatment with these plant extracts. Gene expression studies revealed that extracts of H. antidysenterica leaf, H. antidysenterica bark, and the whole plant of E. scaber and C. asiatica down-regulate aphA or aphB, the major regulator genes modulating both virulence and biofilm formation. Hence, we propose that these herbal combinations could serve as a multifaceted approach to combat the pathogen and also, in turn, reduce antimicrobial resistance development.

Brown spot disease, caused by Cochliobolus miyabeanus, is currently considered to be one of the most important yield reducers of rice (Oryza sativa L.). Despite its agricultural importance, little is known about the virulence mechanisms deployed by the fungus. Therefore, we set out to identify novel virulence factors with a role in disease development. This article reports, for the first time, the production of tentoxin by C. miyabeanus as a virulence factor during brown spot disease and the identification of the non-ribosomal protein synthetase (NRPS) CmNps3, responsible for tentoxin biosynthesis. We compared the chemical compounds produced by C. miyabeanus strains differing in virulence ability using ultra-high-performance liquid chromatography (UHPLC) coupled to high-resolution Orbitrap mass spectrometry (HRMS). The production of tentoxin by a highly virulent strain was revealed by principal component analysis of the detected ions and confirmed by UHPLC coupled to tandem-quadrupole mass spectrometry (MS/MS). The corresponding NRPS was identified by in silico genome analysis and confirmed by gene deletion. Infection tests with wild-type and Cmnps3 mutants showed that tentoxin acts as a virulence factor and is correlated with chlorosis development during the second phase of infection. Although rice has previously been classified as a tentoxin-insensitive plant species, our data demonstrate that tentoxin production by C. miyabeanus affects symptom development.

Maize ear rot caused by several Fusarium species is an important fungal disease. Apart from yield losses, ear rot fungi can produce mycotoxins and masked forms in infected grains. Masked mycotoxins have received increased attention in view of their bioavailability and potential toxicity in animals and humans, but their presence and relevance in the field still remain undisclosed. To get a better insight, the present study assessed the presence of various Fusarium parent and masked mycotoxins, i.e. deoxynivalenol, 3-acetyldeoxynivalenol, 15-acetyldeoxynivalenol, deoxynivalenol-3-glucoside, zearalenone, α-zearalenol, β-zearalenol, zearalenone-14-glucoside, zearalenone-14-sulfate, α-zearalenol-14-glucoside, β-zearalenol-14-glucoside, T-2 and HT-2 toxin, in various commercial maize varieties grown under natural infection conditions in Flanders, Belgium. The results showed that the maize varieties were co-contaminated with both parent and masked mycotoxins. Moreover, a positive correlation between these forms was established. A higher contamination with a particular mycotoxin appeared to be coupled with an elevated load of another (masked) mycotoxin. The results highlight the importance to screen for multiple mycotoxins, both parent and masked, to guarantee food and feed safety. Furthermore, analysis was carried out to elucidate the distribution of the various mycotoxins in the field. The maize variety did not significantly influence mycotoxin accumulation, except for deoxynivalenol. Subdivisions in the field with higher mycotoxin levels for deoxynivalenol and its derivatives, zearalenone and its derivatives, and the sum of T-2 and HT-2 toxin were observed.
