The McMurdo Dry Valleys (MDVs), Antarctica, represent a cold, desert ecosystem poised on the threshold of melting and freezing water. The MDVs have experienced dramatic signs of climatic change, most notably a warm austral summer in 2001-2002 that caused widespread flooding, partial ice cover loss and lake level rise. To understand the impact of these climatic disturbances on lake microbial communities, we simulated lake level rise and ice-cover loss by transplanting dialysis-bagged communities from selected depths to other locations in the water column or to an open water perimeter moat. Bacteria and eukaryote communities residing in the surface waters (5 m) exhibited shifts in community composition when exposed to either disturbance, while microbial communities from below the surface were largely unaffected by the transplant. We also observed an accumulation of labile dissolved organic carbon in the transplanted surface communities. In addition, there were taxa-specific sensitivities: cryptophytes and Actinobacteria were highly sensitive particularly to the moat transplant, while chlorophytes and several bacterial taxa increased in relative abundance or were unaffected. Our results reveal that future climate-driven disturbances will likely undermine the stability and productivity of MDV lake phytoplankton and bacterial communities in the surface waters of this extreme environment.

Many streams originate in forested watersheds at risk of wildfires. Wildfires can introduce thermally altered organic compounds to terrestrial and aquatic systems. Understanding the degradation of leachates from these burned organic materials, referred to as dissolved pyrogenic organic material (PyDOM), is critical in determining water quality impacts in forested watersheds. This study used fluorescence spectroscopy to examine photochemical alterations of PyDOM generated by leaching organic matter burned at various temperatures. The PyDOM was exposed to natural sunlight for 25 days and the photochemical formation of hydrogen peroxide was monitored. PyDOM was characterized using ultraviolet–visible absorption, excitation–emission matrix (EEM) fluorescence spectroscopy, and fluorescence indices. Throughout the experiment, the emission intensity of the humic peak for all light-exposed leachates decreased while dark leachates exhibited no significant change in their fluorescence spectra. Additionally, hydrogen peroxide concentrations and UV absorbance decreased progressively over time, providing direct evidence that PyDOM concentrations can be significantly reduced by photodegradation. A characteristically low emission peak was observed in the EEMs of the fresh PyDOM, which could help in detecting fresh PyDOM. These results demonstrate that PyDOM derived from burned leachates is susceptible to photodegradation and that fluorescence measurements could be used as proxies for detecting PyDOM immediately post-wildfire.

Many challenges remain before we can fully understand the multifaceted role that natural organic matter (NOM) plays in soil and aquatic systems. These challenges remain despite the considerable progress that has been made in understanding NOM’s properties and reactivity using the latest analytical techniques. For nearly 4 decades, the International Humic Substances Society (IHSS, which is a non-profit scientific society) has distributed standard substances that adhere to strict isolation protocols and reference materials that are collected in bulk and originate from clearly defined sites. These NOM standard and reference samples offer relatively uniform materials for designing experiments and developing new analytical methods. The protocols for isolating NOM, and humic and fulvic acid fractions of NOM utilize well-established preparative scale column chromatography and reverse osmosis methods. These standard and reference NOM samples are used by the international scientific community to study NOM across a range of disciplines from engineered to natural systems, thereby seeding the transfer of knowledge across research fields. Recently, powerful new analytical techniques used to characterize NOM have revealed complexities in its composition that transcend the “microbial” vs. “terrestrial” precursor paradigm. To continue to advance NOM research in the Anthropocene epoch, a workshop was convened to identify potential new sites for NOM samples that would encompass a range of sources and precursor materials and would be relevant for studying NOM’s role in mediating environmental and biogeochemical processes. We anticipate that expanding the portfolio of IHSS reference and standard NOM samples available to the research community will enable this diverse group of scientists and engineers to better understand the role that NOM plays globally under the influence of anthropogenic mediated changes.

In the western USA, one legacy of historic mining is drainage of acidic, metal-rich water generated by exposure to oxygen of sulfide minerals in mine workings, referred to as acid mine drainage (AMD). Streams receiving AMD and natural acid rock drainage (ARD) have a low pH, high dissolved metal concentrations, and extensive streambed oxide deposits. Recently, enhanced ARD generation in the Snake River watershed in the Rocky Mountains has been shown to be associated with warmer summer air temperatures, which has been attributed to expanding weathering fronts that promote oxidation due to earlier drying of shallow soils. In mountain watersheds where complex orogeny disseminated minerals throughout the landscape, weathering processes may also mobilize rare earth elements (REEs). We report that in the Snake River REEs are currently distributed in streams at concentrations ranging from 1 to 100 μg/L. Further, analysis of archived sample indicates that REE increases over time are also associated with increased summer air temperatures. In downstream reaches where the Snake River discharges into a water supply reservoir, colloidal and particulate metal oxides are abundant and sorptive processes may influence REE speciation. We also show that REEs accumulate in benthic invertebrates at concentrations comparable to toxic metals associated with ARD.

The McMurdo Dry Valley region is the largest ice-free area of Antarctica. Ephemeral streams flow here during the austral summer, transporting glacial meltwater to perennially ice-covered, closed basin lakes. The chemistry of 24 Taylor Valley streams was examined over the two-decade period of monitoring from 1993 to 2014, and the geochemical behavior of two streams of contrasting physical and biological character was monitored across the seven weeks of the 2010–2011 flow season. Four species dominate stream solute budgets: HCO3–, Ca2+, Na+, and Cl–, with SO42–, Mg2+, and K+ present in significantly lesser proportions. All streams contain dissolved silica at low concentrations. Across Taylor Valley, streams are characterized by their consistent anionic geochemical fingerprint of HCO3 > Cl > SO4, but there is a split in cation composition between 14 streams with Ca > Na > Mg > K and 10 streams with Na > Ca > Mg > K. Diatom metacommunities are structured by environmental, historical, and spatial factors that are often attributed to organism dispersal. In the McMurdo Sound region (MSR) of Antarctica, wind connects aquatic habitats through delivery of inorganic and organic matter. We evaluated the dispersal of diatoms in aeolian material and its relation to the regional diatom metacommunity using light microscopy and 18S rRNA high-throughput sequencing. The concentration of diatoms ranged from 0 to 8.76 * 106 valves · g-1 dry aeolian material. Up to 15% of whole cells contained visible protoplasm, indicating that up to 3.43 * 104 potentially viable individuals could be dispersed in a year to a single 2 cm2 site. Diatom DNA and RNA was detected at each site, reinforcing the likelihood that we observed dispersal of viable diatoms. Of the 50 known morphospecies in the MSR, 72% were identified from aeolian material using microscopy. Aeolian community composition varied primarily by site. Meanwhile, each aeolian community was comprised of morphospecies found in aquatic communities from the same lake basin. These results suggest that aeolian diatom dispersal in the MSR is spatially structured, is predominantly local, and connects local aquatic habitats via a shared species pool. Nonetheless, aeolian community structure was distinct from that of aquatic communities, indicating intra-habitat dispersal and environmental filtering also underlie diatom metacommunity dynamics. The present study confirms that a large number of diatoms are passively dispersed by wind across a landscape characterized by aeolian processes, integrating the regional flora and contributing to metacommunity structure and landscape connectivity.

Residual pit lakes from mining are often dangerous to sample for water quality. Thus, pit lakes may be rarely (or never) sampled. This study developed new technology in which water-sampling devices, mounted on an unmanned aerial vehicle (UAV), were used to sample three pit lakes in Nevada, USA, during 1 week in 2017. Water-quality datasets from two of the three pit lakes on public lands, Dexter and Clipper, are presented here. The current conditions of the Dexter pit lake were assessed by examining cation and anion concentration changes that have occurred over a 17-year period since the pit lake was last sampled in 2000. Data gathered during this sampling campaign assessed 2017 conditions of the Dexter and Clipper pit lakes by comparing constituent concentrations to the Nevada Division of Environmental Protection (NDEP) pit lake water-quality requirements, indicating that selenium concentrations exceeded regulatory standards. We compared our sampling data for Dexter lake to prior water-quality data from the Dexter pit lake collected in 1999 and 2000. This comparison for the Dexter pit lake indicates that evapoconcentration may have caused increasing cation and anion concentrations. This UAV sampling approach can potentially incorporate the use of additional multiparameter probes: pH, oxygen concentration, turbidity, or chlorophyll. Some limitations of this UAV water-sampling methodology are battery duration, weather conditions, and payload capacity.

The non-marine diatom flora of the Antarctic Continent includes several endemic taxa recorded over the past 100 years. One of these taxa, Navicula adminensis D.Roberts & McMinn, was described from the Vestfold Hills, East Antarctica. Detailed light and scanning electron microscopy observations have shown that based on its morphological features, the species does not belong to the genus Navicula sensu stricto. To determine the most closely related genera to N. adminensis, the morphological features of Adlafia, Kobayasiella, Envekadea, Stenoneis, Berkeleya, Climaconeis, and Parlibellus were compared with those of N. adminensis. Although each of these genera shows one or more similar features, none of them accommodates the salient morphological characteristics of N. adminensis. Therefore, a new genus, Sabbea gen. nov., is herein described, and Navicula adminensis is formally transferred to the new genus as Sabbea adminensis comb. nov. The genus Sabbea is characterized by uniseriate striae composed of small, rounded areolae occluded externally by individual hymenes, a rather simple raphe structure with straight, short proximal ends and short terminal raphe fissures, open girdle bands with double perforation and a very shallow mantle.

The period of disrupted human activity caused by the COVID-19 pandemic, coined the "anthropause," altered the nature of interactions between humans and ecosystems. It is uncertain how the anthropause has changed ecosystem states, functions, and feedback to human systems through shifts in ecosystem services. Here, we used an existing disturbance framework to propose new investigation pathways for coordinated studies of distributed, long-term social-ecological research to capture effects of the anthropause. Although it is still too early to comprehensively evaluate effects due to pandemic-related delays in data availability and ecological response lags, we detail three case studies that show how long-term data can be used to document and interpret changes in air and water quality and wildlife populations and behavior coinciding with the anthropause. These early findings may guide interpretations of effects of the anthropause as it interacts with other ongoing environmental changes in the future, particularly highlighting the importance of long-term data in separating disturbance impacts from natural variation and long-term trends. Effects of this global disturbance have local to global effects on ecosystems with feedback to social systems that may be detectable at spatial scales captured by nationally to globally distributed research networks.

Diatoms are diverse and widespread freshwater Eukaryotes that make excellent microbial subjects for addressing questions in metacommunity ecology. In the McMurdo Dry Valleys of Antarctica, the simple trophic structure of glacier-fed streams provides an ideal outdoor laboratory where well-described diatom assemblages are found within two cyanobacterial mat types, which occupy different habitats and vary in coverage within and among streams. Specifically, black mats ofNostocspp. occur in marginal wetted habitats, and orange mats (Oscillatoriaspp. andPhormidiumspp.) occur in areas of consistent stream flow. Despite their importance as bioindicators for changing environmental conditions, the role of dispersal in structuring dry valley diatom metacommunities remains unclear. Here, we use MCSim, a spatially explicit metacommunity simulation package for R, to test alternative hypotheses about the roles of dispersal and species sorting in maintaining the biodiversity of diatom assemblages residing in black and orange mats. The spatial distribution and patchiness of cyanobacterial mat habitats was characterized by remote imagery of the Lake Fryxell sub-catchment in Taylor Valley. The available species pool for diatom metacommunity simulation scenarios was informed by the Antarctic Freshwater Diatoms Database, maintained by the McMurdo Dry Valleys Long Term Ecological Research program. We used simulation outcomes to test the plausibility of alternative community assembly hypotheses to explain empirically observed patterns of freshwater diatom biodiversity in the long-term record. The most plausible simulation scenarios suggest species sorting by environmental filters, alone, was not sufficient to maintain biodiversity in the Fryxell Basin diatom metacommunity. The most plausible scenarios included either (1) neutral models with different immigration rates for diatoms in orange and black mats or (2) species sorting by a relatively weak environmental filter, such that dispersal dynamics also influenced diatom community assembly, but there was not such a strong disparity in immigration rates between mat types. The results point to the importance of dispersal for understanding current and future biodiversity patterns for diatoms in this ecosystem, and more generally, provide further evidence that metacommunity theory is a useful framework for testing hypotheses about microbial community assembly. When deposited on snow and ice, light absorbing impurities (LAIs) such as dust and black carbon (BC) reduce surface albedo and enhance melt. BC comes from incomplete combustion of fossil fuels and biomass. Local and regional sources of BC exist in High Mountain Asia, such as dry-dung burning for heat and fuel, which occurs in close proximity to snow and glaciers. Local dust or dust transported from the Indo-Gangetic Plain is also present. In the Ganges River Basin, meltwater is dominated by seasonal snow, yet relatively few observations of impurities in seasonal snow exist. To understand sources of impurities and their concentrations for seasonal snow on land, we evaluated multiple lines of evidence to scale up from ground-based measurements in the Dudh Koshi River Basin, a remote headwater basin within the Khumbu Region of Nepal. We obtained ground-based in-situ observations of refractory black carbon (rBC) measured by Single Particle Soot Photometer (SP2), including size distributions in snow on land. We interpreted these results in the context of concurrent Moderate Imaging Spectroradiometer (MODIS) satellite observations and speciated aerosol optical depth derived from reanalysis products modeled with the Navy Aerosol Analysis Prediction System global aerosol model. We collected snow samples, mostly in the Gokyo Valley, at varying distances from local tea houses along a 2000 meters above sea level (m a.s.l.) elevation transect from 3250 to 5299 m a.s.l. rBC concentrations ranged from 3.9 to 76.8 μg-rBC/L-H2O. Although previous data do not exist at these lower elevations, our findings are higher than previous surface snow results at higher elevations in the nearby Khumbu Valley. In general, rBC concentrations were lower in fresh snow than aged snow; concurrent MODIS satellite observations of snow albedo also show smaller impacts from LAIs in visible wavelengths in fresh snow. In aged snow samples, rBC decreased with elevation, as did concurrent MODIS albedo observations. rBC-particle size distributions also shifted to a larger mode for aged snow samples. Results from the Navy Aerosol Analysis Prediction System model indicate anthropogenic and biogenic fine aerosols from biofuel (dry-dung burning) are the primary aerosol species in the atmosphere for the study period, at ∼thrice the concentration of dust and smoke.

Much work has been performed to investigate controls on nitrogen (N) uptake in streams, yet the fate of assimilated N is comparatively poorly resolved. Here, we use in‐stream fixed N as an isotopic tracer to study the fate of assimilated N in glacial meltwater streams. We characterized δ15N signatures of Oscillatorean, Chlorophyte, and N‐fixing Nostoc mats over the lengths of two streams, and transported particulate organic matter (POM) in one. POM was isotopically most similar to Nostoc, which always had values near the atmospheric standard (δ15N ≅ 0‰), suggesting N‐fixation. Other mat types were depleted upstream, and became progressively enriched downstream (plateauing at δ15N ≅ 0‰), indicating a shift in N source. These results collectively show that Nostoc‐derived N is mobilized, mineralized, and increasingly assimilated downstream as more depleted glacier‐derived N is exhausted, demonstrating the importance of organic matter processing to balancing elemental budgets, and improving our understanding of nutrient cycling in lotic environments.

Microbial biofilms are biological hotspots in many alpine and polar ecosystems, but the controls on and functional significance of their fauna are little known. We studied cyanobacterial mats and the underlying sediment in a glacial meltwater stream in the McMurdo Dry valleys, Antarctica. We investigated mat biomass (total and phototrophic), diatoms, and micro-meiofauna (nematodes, rotifers, and tardigrades) at nine sites along a 1670 m stream reach in a cold, low-flow growing season, and in a warmer growing season in which peak flows (above 100 L s−1) scoured the mats. Diatom and invertebrate communities were not related, but mat biomass in the low-flow year was negatively related to nematode abundance, including that of the omnivore Eudorylaimus. In the high flow year that followed, invertebrate abundance was reduced in the mats, diatom community structure was altered, and mat biomass was higher. The difference in invertebrate abundance between years was greater in mats in upstream reaches, where the greatest increases in flow velocity may have occurred, and was negligible in mats in downstream reaches as well as in the sediment beneath the mats. Integrating our results with previous findings, we generate two predictive hypotheses to be tested in glacial meltwater streams: (1) under peak flows invertebrates decline in the microbial mats, while (2) the sediment beneath the mats is a refuge from the flow disturbance. Our results also suggest that, under stable flow conditions, microinvertebrate grazers could exert top-down control on microbial mat biomass.

It was demonstrated more than two decades ago that microorganisms use humic substances, including fulvic acid (FA), as electron shuttles during iron (Fe) reduction in anaerobic soils and sediments. The relevance of this mechanism for the acceleration of Fe(III) reduction in arsenic-laden groundwater environments is gaining wider attention. Here we provide new evidence that dissolved FAs isolated from sediment-influenced surface water and groundwater in the Bengal Basin were capable of electron shuttling between Geobacter metallireducens and Fe(III). Moreover, all four Bangladesh sediment-derived dissolved FAs investigated in this study had higher electron accepting capacity (176 to 245μmol/g) compared to aquatic FAs, such as Suwanee River Fulvic Acid (67μmol/g). Our direct evidence that Bangladesh FAs are capable of intermediate electron transfer to Fe(III) supports other studies that implicate electron shuttling by sediment-derived aqueous humics to enhance Fe reduction and, in turn, As mobility. Overall, the finding of greater electron accepting capacity by dissolved FAs from groundwater and other sediment-influenced environments advances our understanding of mechanisms that control Fe reduction under conditions where electron transfer is the rate limiting step.

Northern ecosystems are experiencing some of the most dramatic impacts of global change on Earth. Rising temperatures, hydrological intensification, changes in atmospheric acid deposition and associated acidification recovery, and changes in vegetative cover are resulting in fundamental changes in terrestrial-aquatic biogeochemical linkages. The effects of global change are readily observed in alterations in the supply of dissolved organic matter (DOM)-the messenger between terrestrial and lake ecosystems-with potentially profound effects on the structure and function of lakes. Northern terrestrial ecosystems contain substantial stores of organic matter and filter or funnel DOM, affecting the timing and magnitude of DOM delivery to surface waters. This terrestrial DOM is processed in streams, rivers, and lakes, ultimately shifting its composition, stoichiometry, and bioavailability. Here, we explore the potential consequences of these global change-driven effects for lake food webs at northern latitudes. Notably, we provide evidence that increased allochthonous DOM supply to lakes is overwhelming increased autochthonous DOM supply that potentially results from earlier ice-out and a longer growing season. Furthermore, we assess the potential implications of this shift for the nutritional quality of autotrophs in terms of their stoichiometry, fatty acid composition, toxin production, and methylmercury concentration, and therefore, contaminant transfer through the food web. We conclude that global change in northern regions leads not only to reduced primary productivity but also to nutritionally poorer lake food webs, with discernible consequences for the trophic web to fish and humans.

During the week of September 9-15, 2013, about 44 cm of rain fell across Boulder County, Colorado, USA, representing a very rare precipitation event. The resultant stream flows corresponded to an extreme event not seen since the historical flood of 1894. For the Boulder Creek Critical Zone Observatory (BcCZO), this event provided an opportunity to study the effect of extreme rainfall on solute concentration-discharge relationships and biogeochemical processes. We measured weathering-derived lithologic solutes (Ca, Mg, Na, K, and Si) and dissolved organic carbon (DOC) concentrations at two sites on Boulder Creek during the recession of peak flow. We also isolated four distinct fractions of dissolved organic matter (DOM) for chemical characterization. At the upper and lower sites, all solutes had their highest concentration at peak flow. At the upper site, which represented a mostly forested catchment, the concentrations of lithologic solutes decreased slightly during flood recession. In contrast, DOC and K concentrations decreased by a factor of three. At the lower site within the urban corridor, concentration of lithologic solutes decreased substantially for a few days before rebounding, whereas the DOC and K concentrations continued to decrease. Additionally, we found spatiotemporal trends in the chemical quality of DOM that were consistent with a limited reservoir of soluble organic matter in surficial soils becoming depleted and deeper layers of the Critical Zone contributing DOM during the flood recession. Overall, these results suggest that despite the extreme flood event, concentration-discharge relationships were similar to typical snowmelt periods in this Rocky Mountain region.

Benthic microbial mats in the glacial-fed meltwater streams are hotspots of productivity in the McMurdo Dry Valleys (MDV), Antarctica. Benthic diatoms are common in these mats and the >45 primarily endemic taxa represent the most diverse group of eukaryotes in the MDV. In this harsh polar desert, streams are thermally dynamic with daily water temperatures varying 6–9 °C and daily maximum temperatures as high as 15 °C. Stream temperature may play a role in determining growth rates and survival strategies. To understand taxon-specific adaptations to their environment, we measured the growth rates of unialgal cultures of four diatom taxa (Psammothidium papilio, Hantzschia abundans, Hantzschia amphioxys, and Hantzschia amphioxys f. muelleri) under three temperature conditions (7.6, 10, and 15 °C) that were representative of maximum daily stream temperatures. We found that P. papilio exhibited a constant growth rate across the full temperature range; this species is most common in streams that begin to flow early in the summer and with less variable thermal regimes. Growth rates for H. abundans were greatest at 15 °C, but showed a non-linear relationship with temperature. H. amphioxys f. muelleri grew faster than the other taxa studied and thrived at 10 °C. Hantzschia amphioxys grew only at the two lower temperatures. These results aligned with the observed relationships between each taxon’s relative abundance and stream temperatures in the long-term record maintained by the MDV Long-Term Ecological Research program. Overall, our observations suggest that differences in thermal optima may be one factor contributing to and maintaining the diversity of benthic diatom flora in the MDV.

Black carbon (BC) is derived from the incomplete combustion of biomass and fossil fuels and can enhance glacial recession when deposited on snow and ice surfaces. Here we explore the influence of environmental conditions and the proximity to anthropogenic sources on the concentration and composition of dissolved black carbon (DBC), as measured by benzenepolycaroxylic acid (BPCA) markers, across snow, lakes, and streams from the global cryosphere. Data are presented from Antarctica, the Arctic, and high alpine regions of the Himalayas, Rockies, Andes, and Alps. DBC concentrations spanned from 0.62 µg/L to 170 µg/L. The median and (2.5, 97.5) quantiles in the pristine samples were 1.8 µg/L (0.62, 12), and non-pristine samples were 21 µg/L (1.6, 170). DBC is susceptible to photodegradation when exposed to solar radiation. This process leads to a less condensed BPCA signature. In general, DBC across the dataset was comprised of less-polycondensed DBC. However, DBC from the Greenland Ice Sheet (GRIS) had a highly-condensed BPCA molecular signature. This could be due to recent deposition of BC from Canadian wildfires. Variation in DBC appears to be driven by a combination of photochemical processing and the source combustion conditions under which the DBC was formed. Overall, DBC was found to persist across the global cryosphere in both pristine and non-pristine snow and surface waters. The high concentration of DBC measured in supra-glacial melt on the GRIS suggests DBC can be mobilized across ice surfaces. This is significant because these processes may jointly exacerbate surface albedo reduction in the cryosphere.

Bioavailable glacial carbon has been thought to be largely ancient or anthropogenic. Analyses of carbon dynamics in an Antarctic supraglacial stream reveal that non-photosynthetic production relies on organic carbon from photosynthetic microbes.

Advances in hydrology are greatly needed and approaches that employ hypotheses to guide research have the potential to contribute to future advances. In this context, hypotheses can serve a range of purposes. Overarching hypotheses can provide a common integrating framework for collaborative research and can be revised as research progresses over time. Hypotheses that attempt to explain unexpected field observations or experimental results can provide a guide for designing further field studies. Focused testable hypotheses can facilitate effective presentation of proposed research, and clarify alternative hypotheses. Finally, the value of employing a hypothesis-based approach depends upon the research environment, which can act as an “environmental filter” in determining successful research outcomes.

Light absorbing particles (LAPs) in snow such as dust and black carbon (BC) influence the radiative forcing at the Earth's surface, which has major implications for global climate models. LAPs also significantly influence the melting of glaciers, sea ice, and seasonal snow. Here, we present an in situ study of surface snow near an active coalmine in the Norwegian Arctic. We couple measurements of spectral hemispherical directional reflectance factor (HDRF) with measurements of LAPs characterized two ways, as refractory black carbon (rBC) using a Single Particle Soot Photometer (SP2) and the total light absorption of LAPs measured with the Light Absorption Heating Method (LAHM). The Snow Ice and Aerosol Radiation model was constrained by LAP measurements. Results were compared to observed spectral albedo measurements. Modeled and observed albedos were similar at the cleaner and more remote sites. However, the modeled spectral albedos do not fully account for the low spectral albedo measured next to the mine. LAP measurements also showed a large variation in particle sizes (tenths-to-tens of microns) related to transport distance of the particles from the mine. Here, we find that LAPs from coal dust reduce the spectral HDRF by up to 84% next to the mine, and 55% 0.5 km downwind of the mine. The coupling of extreme LAP observations, (1 ng g-1 to 4863 ng g-1), with HDRF measurements from 350 – 2500 nm has facilitated the development of spectral band pairs, which could be used in the future to remotely assess LAPs in Arctic snow.

Nearly all microbial communities are dynamic in time. Understanding how temporal dynamics in microbial community structure affect soil biogeochemistry and fertility are key to being able to predict the responses of the soil microbiome to environmental perturbations. Here, we explain the effects of soil spatial structure and relic DNA on the determination of microbial community fluctuations over time. We found that intensive spatial sampling was required to identify temporal effects in microbial communities because of the high degree of spatial heterogeneity in soil and that DNA from nonliving sources masks important temporal patterns. We identified groups of microbes with shared temporal responses and show that these patterns were predictable from changes in soil characteristics. These results provide insight into the environmental preferences and temporal relationships between individual microbial taxa and highlight the importance of considering relic DNA when trying to detect temporal dynamics in belowground communities., Few studies have comprehensively investigated the temporal variability in soil microbial communities despite widespread recognition that the belowground environment is dynamic. In part, this stems from the challenges associated with the high degree of spatial heterogeneity in soil microbial communities and because the presence of relic DNA (DNA from dead cells or secreted extracellular DNA) may dampen temporal signals. Here, we disentangle the relationships among spatial, temporal, and relic DNA effects on prokaryotic and fungal communities in soils collected from contrasting hillslopes in Colorado, USA. We intensively sampled plots on each hillslope over 6 months to discriminate between temporal variability, intraplot spatial heterogeneity, and relic DNA effects on the soil prokaryotic and fungal communities. We show that the intraplot spatial variability in microbial community composition was strong and independent of relic DNA effects and that these spatial patterns persisted throughout the study. When controlling for intraplot spatial variability, we identified significant temporal variability in both plots over the 6-month study. These microbial communities were more dissimilar over time after relic DNA was removed, suggesting that relic DNA hinders the detection of important temporal dynamics in belowground microbial communities. We identified microbial taxa that exhibited shared temporal responses and show that these responses were often predictable from temporal changes in soil conditions. Our findings highlight approaches that can be used to better characterize temporal shifts in soil microbial communities, information that is critical for predicting the environmental preferences of individual soil microbial taxa and identifying linkages between soil microbial community composition and belowground processes.
