Prussian Blue nanoparticles (PBnps) are now popular in nanomedicine thanks to the FDA approval of PB. Despite the numerous papers suggesting or describing the in vivo use of PBnps, no studies have been carried out on the formation of a protein corona on the PBnp surface and its stabilizing role. In this paper, we studied qualitatively and quantitatively the corona formed by the most abundant protein of blood, human serum albumin (HSA). Cubic PBnps (41 nm side), prepared in citric acid solution at PB concentration 5 × 10−4 M, readily form a protein corona by redissolving ultracentrifuged PBnp pellets in HSA solutions, with CHSA ranging from 0.025 to 7.0 mg/mL. The basic decomposition of PBnp@HSA was studied in phosphate buffer at the physiological pH value of 7.4. Increased stability with respect to uncoated PBnps was observed at all concentrations, but a minimum CHSA value of 3.0 mg/mL was determined to obtain stability identical to that observed at serum-like HSA concentrations (35–50 mg/mL). Using a modified Lowry protocol, the quantity of firmly bound HSA in the protein corona (hard corona) was determined for all the CHSA used in the PBnp@HSA synthesis, finding increasing quantities with increasing CHSA. In particular, an HSA/PBnp number in the 1500–2300 range was found for CHSA 3.0–7.0 mg/mL, largely exceeding the 180 HSA/PBnp value calculated for an HSA monolayer on a PBnp. Finally, the stabilization brought by the HSA corona allowed us to carry out pH-spectrophotometric titrations on PBnp@HSA in the 3.5-9-0 pH range, revealing a pKa value of 6.68 for the water molecules bound to the Fe3+ centers on the PBnp surface, whose deprotonation is responsible for the blue-shift of the PBnp band from 706 nm (acidic solution) to 685 nm (basic solution).

This paper addresses a gap in research on lad culture in British Higher Education (HE) by addressing how gender diverse students understand their subjectivities within this context. To do so, we introduce the concept of 'laddish misrecognition'. Drawing on Benjamin and Fraser, we suggest both psychic and social mechanisms are at play in this mis/recognition. We also draw on Muñoz's 'disidentification' to explore what it means to live in this space. We apply this to discussions with 11 LGBTQIA+ students. We identify the complexity of their negotiations regarding: identifications with masculinity versus distancing oneself from 'the lad'; being misrecognised as 'a lad', and wanting to challenge this; and navigating femininity and same-sex relationships while being misrecognised as heterosexual. By exploring accounts that are often positioned at the margins of discussion about lad culture (i.e. not predominantly heterosexual, cisgender), we suggest laddish misrecognition shows how identities are formed in-difference.

En este artículo reflexivo se ofrece una visión panorámica de los estudios realizados sobre la Manosfera y el antifeminismo en el contexto europeo y español en los últimos cinco años. Para ello, se han revisado los estudios más recientes y relevantes realizados en Europa centrados en comprender la misoginia online organizada y las tecnoculturas tóxicas. Además, se han analizado los trabajos incipientes sobre la manosfera española realizados en España con el fin de conectar el conocimiento europeo con los avances locales. Esta revisión crítica ofrece un estado de la cuestión que incluye: una definición de la manosfera, una propugnación de la importancia de su estudio, una revisión de los temas tratados por áreas temáticas, un diagnóstico de lo que queda por hacer y un aviso ético-epistemológico para aquellas investigadoras que se quieran comprometer con el trabajo de desentrañar qué es la manosfera.

Coronary artery segmentation from CT scans is a helpful tool for coronary artery diseases diagnosis, which is frequently characterised by a vessel narrowing (stenosis). This is a highly demanded and high time-consuming process, thus automated procedures are becoming increasingly necessary. In this work, we propose an extremely light computationally 2D UNet that uses transfer learning for the first time in CT images. We compare the results, using different architectures and backbones, of a 2D UNet and a 3D UNet trained from scratch (i.e. weights are randomly initialised) and a 2D EfficientUNet. Both the amount of input data, with a total of 88 patients, and the extension of the structure to be recognised, the aorta and the coronary arteries ( $A + C.A$ ), as well as the coronary arteries only ( $C.A$ ) are analysed. Network outputs in clinically identified stenotic lesion areas are also assessed. The results show the advantage of using transfer learning when data is scarce, improving the $F_{1}$ score by up to 0.6 points for the 2D UNet. On the other hand, when data is sufficient, $F_{1}$ score values are close to 0.9 for all the networks. Besides, the results reveal that the 2D UNet distinguishes the thinnest and most distal vessels, although in the presence of a lesion, there is a clear tendency to overestimate it. The network with the best accuracy is the 3D UNet, with values above 95% and 75% in $A+C.A$ and $C.A$ , respectively. Moreover, the proposed methods show dependence on the amount of training data and dataset structure ( $A + C.A$ or $C.A$ ).

Background: Breast cancer is the most common malignancy and the second leading cause of cancer-related mortality in Spanish women. Ribociclib in combination with endocrine therapy (ET) has shown superiority in prolonging survival in patients with hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2−) advanced breast cancer (ABC) vs. ET alone. Methods: CompLEEment-1 is a single-arm, open-label phase 3b trial evaluating ribociclib plus letrozole in a broad population of patients with HR+, HER2– ABC. The primary endpoints were safety and tolerability. Here we report data for Spanish patients enrolled in CompLEEment-1. Results: A total of 526 patients were evaluated (median follow-up: 26.97 months). Baseline characteristics showed a diverse population with a median age of 54 years. At study entry, 56.5% of patients had visceral metastases and 8.7% had received prior chemotherapy for advanced disease. Rates of all-grade and Grade ≥3 adverse events (AEs) were 99.0% and 76.2%, respectively; 21.3% of patients experienced a serious AE, and 15.8% of AEs led to treatment discontinuation. AEs of special interest of neutropenia, increased alanine aminotransferase, increased aspartate aminotransferase and QTcF prolongation occurred in 77.8%, 14.8%, 11.4% and 4.0% of patients, respectively. Patients aged >70 years experienced increased rates of all-grade and Grade ≥3 neutropenia and anemia. Efficacy results were consistent with the global study. Conclusions: Results from Spanish patients enrolled in CompLEEment-1 are consistent with global data showing efficacy and a manageable safety profile for ribociclib plus letrozole treatment in patients with HR+, HER2− ABC, including populations of interest (NCT02941926). Abstract In this work, we introduce a one-step strategy that is suitable for continuous flow manufacturing of antimicrobial PDMS materials. The process is based on the intrinsic capacity of PDMS to react to certain organic solvents, which enables the incorporation of antimicrobial actives such as salicylic acid (SA), which has been approved for use in humans within pharmaceutical products. By combining different spectroscopic and imaging techniques, we show that the surface properties of PDMS remain unaffected while high doses of the SA are loaded inside the PDMS matrix. The SA can be subsequently released under physiological conditions, delivering a strong antibacterial activity. Furthermore, encapsulation of SA inside the PDMS matrix ensured a diffusion-controlled release that was tracked by spatially resolved Raman spectroscopy, Attenuated Total Reflectance IR (ATR-IR), and UV-Vis spectroscopy. The biological activity of the new material was evaluated directly at the surface and in the planktonic state against model pathogenic bacteria, combining confocal laser scanning microscopy, electron microscopy, and cell viability assays. The results showed complete planktonic inhibition for clinically relevant strains of Staphylococcus aureus and Escherichia coli, and a reduction of up to 4 orders of magnitude for viable sessile cells, demonstrating the efficacy of these surfaces in preventing the initial stages of biofilm formation. Our approach adds a new option to existing strategies for the antimicrobial functionalisation of a wide range of products such as catheters, wound dressings and in-dwelling medical devices based on PDMS.

Abstract Background Heart failure (HF) is a major and growing medical and economic problem, with high prevalence and incidence rates worldwide. Cardiac Biomarker is emerging as a novel tool for improving management of patients with HF with a reduced left ventricular ejection fraction (HFrEF). Methods This is a before and after interventional study, that assesses the impact of a personalized follow-up procedure for HF on patient’s outcomes and care associated cost, based on a clinical model of risk stratification and personalized management according to that risk. A total of 192 patients were enrolled and studied before the intervention and again after the intervention. The primary objective was the rate of readmissions, due to a HF. Secondary outcome compared the rate of ED visits and quality of life improvement assessed by the number of patients who had reduced NYHA score. A cost-analysis was also performed on these data. Results Admission rates significantly decreased by 19.8% after the intervention (from 30.2 to 10.4), the total hospital admissions were reduced by 32 (from 78 to 46) and the total length of stay was reduced by 7 days (from 15 to 9 days). The rate of ED visits was reduced by 44% (from 64 to 20). Thirty-one percent of patients had an improved functional class score after the intervention, whereas only 7.8% got worse. The overall cost saving associated with the intervention was € 72,769 per patient (from € 201,189 to € 128,420) and €139,717.65 for the whole group over 1 year. Conclusions A personalized follow-up of HF patients led to important outcome benefits and resulted in cost savings, mainly due to the reduction of patient hospitalization readmissions and a significant reduction of care-associated costs, suggesting that greater attention should be given to this high-risk cohort to minimize the risk of hospitalization readmissions.

A synthetic approach has been developed to prepare silica gel monoliths that embed well separated silver or gold spherical nanoparticles (NP), with diameters of 8, 18 and 115 nm. Fe3+, O2/cysteine and HNO3 were all successfully used to oxidize and remove silver NP from silica, while aqua regia was necessary for gold NP. In all cases, NP-imprinted silica gel materials were obtained, with spherical voids of the same dimensions of the dissolved particles. By grinding the monoliths, we prepared NP-imprinted silica powders that were able to efficiently reuptake silver ultrafine NP (Ag-ufNP, d = 8 nm) from aqueous solutions. Moreover, the NP-imprinted silica powders showed a remarkable size selectivity, based on the best match between NP radius and the curvature radius of the cavities, driven by the optimization of attractive Van der Waals forces between SiO2 and NP. Ag-ufNP are increasingly used in products, goods, medical devices, disinfectants, and their consequent diffusion in the environment is of rising concern. Although limited here to a proof-of-concept level, the materials and methods described in this paper may be an efficient solution for capturing Ag-ufNP from environmental waters and to safely dispose them.

Celiac disease (CD) is a chronic autoimmune disease characterized by an immune-triggered enteropathy upon gluten intake. The only current treatment available is lifelong Gluten Free Diet (GFD). Several extraintestinal manifestations have been described in CD, some affecting the oral mucosa. Thus, we hypothesized that oral mucosa could potentially be a target for novel biomarkers and an administration route for CD treatment. Six de novo diagnosed and seven CD patients under GFD for at least 1 year were recruited. Non-celiac subjects (n = 8) were recruited as control group. Two biopsies of the cheek lining were taken from each subject for mRNA analysis and immunohistochemical characterization. We observed a significant decrease in the expression of epithelial junction proteins in all CD patients, indicating that oral mucosa barrier integrity is compromised. FoxP3+ population was greatly increased in CD patients, suggesting that Tregs are recruited to the damaged mucosa, even after avoidance of gluten. Amphiregulin mRNA levels from Peripheral Blood Mononuclear Cells (PBMCs) and epithelial damage in the oral mucosa correlated with Treg infiltration in all the experimental groups, suggesting that recruited Tregs might display a “repair” phenotype. Based on these results, we propose that oral mucosa is altered in CD and, as such, might have diagnostic potential. Furthermore, due to its tolerogenic nature, it could be an important target for oral immunotherapy.

Surface engineering is a promising strategy to limit or prevent the formation of biofilms. The use of topographic cues to influence early stages of biofilm formationn has been explored, yet many fundamental questions remain unanswered. In this work, we develop a topological model supported by direct experimental evidence, which is able to explain the effect of local topography on the fate of bacterial micro-colonies of Staphylococcus spp. We demonstrate how topological memory at the single-cell level, characteristic of this genus of Gram-positive bacteria, can be exploited to influence the architecture of micro-colonies and the average number of surface anchoring points over nano-patterned surfaces, formed by vertically aligned silicon nanowire arrays that can be reliably produced on a commercial scale, providing an excellent platform to investigate the effect of topography on the early stages of Staphylococcus spp. colonisation. The surfaces are not intrinsically antimicrobial, yet they delivered a topography-based bacteriostatic effect and a significant disruption of the local morphology of micro-colonies at the surface. The insights from this work could open new avenues towards designed technologies for biofilm engineering and prevention, based on surface topography.

Bacteria infections and related biofilms growth on surfaces of medical devices are a serious threat to human health. Controlled hyperthermia caused by photothermal effects can be used to kill bacteria and counteract biofilms formation. Embedding of plasmonic nano-objects like gold nanostars (GNS), able to give an intense photothermal effect when irradiated in the NIR, can be a smart way to functionalize a transparent and biocompatible material like polydimethylsiloxane (PDMS). This process enables bacteria destruction on surfaces of PDMS-made medical surfaces, an action which, in principle, can also be exploited in subcutaneous devices. We prepared stable and reproducible thin PDMS films containing controllable quantities of GNS, enabling a temperature increase that can reach more than 40 degrees. The hyperthermia exerted by this hybrid material generates an effective thermal microbicidal effect, killing bacteria with a near infrared (NIR) laser source with irradiance values that are safe for skin.

The generation of hydrogen from water using light is currently one of the most promising alternative energy sources for humankind but faces significant barriers for large-scale applications due to the low efficiency of existing photo-catalysts. In this work we propose a new route to fabricate nano-hybrid materials able to deliver enhanced photo-catalytic hydrogen evolution, combining within the same nanostructure, a plasmonic antenna nanoparticle and semiconductor quantum dots (QDs). For each stage of our fabrication process we probed the chemical composition of the materials with nanometric spatial resolution, allowing us to demonstrate that the final product is composed of a silver nanoparticle (AgNP) plasmonic core, surrounded by satellite Pt decorated CdS QDs (CdS@Pt), separated by a spacer layer of SiO2 with well-controlled thickness. This new type of photoactive nanomaterial is capable of generating hydrogen when irradiated with visible light, displaying efficiencies 300% higher than the constituting photo-active components. This work may open new avenues for the development of cleaner and more efficient energy sources based on photo-activated hydrogen generation.

Abstract Photo-responsive antibacterial surfaces combining both on-demand photo-switchable activity and sustained biocidal release were prepared using sequential chemical grafting of nano-objects with different geometries and functions. The multi-layered coating developed incorporates a monolayer of near-infrared active silica-coated gold nanostars (GNS) decorated by silver nanoparticles (AgNP). This modular approach also enables us to unravel static and photo-activated contributions to the overall antibacterial performance of the surfaces, demonstrating a remarkable synergy between these two mechanisms. Complementary microbiological and imaging evaluations on both planktonic and surface-attached bacteria provided new insights on these distinct but cooperative effects.

PVA films with embedded either silver nanoparticles (AgNP), NIR-absorbing photothermal gold nanostars (GNS), or mixed AgNP+GNS were prepared in this research. The optimal conditions to obtain stable AgNP+GNS films with intact, long lasting photothermal GNS were obtained. These require coating of GNS with a thiolated polyethylene glycol (PEG) terminated with a carboxylic acid function, acting as reticulant in the film formation. In the mixed AgNP+GNS films, the total noble metal content is w/w and in the Ag films < 0.025% w/w. The slow but prolonged Ag+ release from film-embedded AgNP (8–11% of total Ag released after 24 h, in the mixed films) results in a very strong microbicidal effect against planktonic Escherichia coli and Staphylococcus aureus bacterial strains (the release of Au from films is instead negligible). Beside this intrinsic effect, the mixed films also exert an on-demand, fast hyperthermal bactericidal action, switched on by NIR laser irradiation (800 nm, i.e., inside the biotransparent window) of the localized surface plasmon resonance (LSPR) absorption bands of GNS. Temperature increases of 30 °C are obtained using irradiances as low as 0.27 W/cm2. Moreover, 80–90% death on both strains was observed in bacteria in contact with the GNS-containing films, after 30 min of irradiation. Finally, the biocompatibility of all films was verified on human fibroblasts, finding negligible viability decrease in all cases.

Silver nanoparticles were produced with AgF as the starting Ag(I) salt, with pectin as the reductant and protecting agent. While the obtained nanoparticles (pAgNP-F) have the same dimensional and physicochemical properties as those already described by us and obtained from AgNO3 and pectin (pAgNP-N), the silver nanoparticles from AgF display an increased antibacterial activity against E. coli PHL628 and Staphylococcus epidermidis RP62A (S. epidermidis RP62A), both as planktonic strains and as their biofilms with respect to pAgNP-N. In particular, a comparison of the antimicrobial and antibiofilm action of pAgNP-F has been carried out with pAgNP-N, pAgNP-N and added NaF, pure AgNO3, pure AgF, AgNO3 and added NaF and pure NaNO3 and NaF salts. By also measuring the concentration of the Ag+ cation released by pAgNP-F and pAgNP-N, we were able to unravel the separate contributions of each potential antibacterial agent, observing an evident synergy between p-AgNP and the F− anion: the F− anion increases the antibacterial power of the p-AgNP solutions even when F− is just 10 µM, a concentration at which F− alone (i.e., as its Na+ salt) is completely ineffective.

Abstract The aim of this study was to evaluate phage display technology for mapping Haemonchus contortus mimotopes. We screened the PhD-7 Phage Display Peptide Library Kit with a sheep polyclonal antibody against H. contortus. After four rounds of selection, 50 phage peptide clones were selected by biopanning and sequenced. Two clones displaying peptide mimotopes of H. contortus proteins were chosen for sheep immunization: clone 6 - mimotope of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) and clone 17 - mimotope of a disorganized muscle family member (Dim 1). Twelve sheep were allocated into 3 groups of 4 animals as follow: G1: control group; G2/GAPDH: immunized with clone 6; and G3/Dim1: immunized with clone 17. Four immunizations were performed at intervals of seven days (0, 7, 14, and 21 days). On day 28 post initial vaccination, all groups were orally challenged with 2500 H. contortus infective larvae. The mimotope peptides selected by phage display were recognized by IgG from sheep naturaly infected with H. contortus. The immunization protocol showed an increasein IgG anti-M13 phage titers, but no effect was observed in IgG-specific for the anti-mimotope peptides. This is the first report of successful use of a phage display library for the identification of mimotopes of H. contortus proteins.

Total ankle arthroplasty has become popular in the last few years. The lateral transfibular approach is an alternative to the traditional anterior approach. The purpose of this study was to evaluate our 50 first and consecutive clinical and radiological outcomes of transfibular total ankle replacements (Trabecular Metal Total AnkleR Zimmer Biomet, Warsaw, IN) with a follow-up of at least 3 years. This retrospective study included 50 patients. The main indication was post-traumatic osteoarthritis (n = 41). The mean age was 59 (range = 39-81). All patients were followed for at least 36 months postoperatively. Patients were assessed with the American Orthopaedic Foot & Ankle Society (AOFAS) Ankle Hindfoot Score and Visual analog scale (VAS) preoperatively and postoperatively. Range of motion and radiological measures were assessed as well. Postoperatively, patients demonstrated statistically significant improvement in the AOFAS score from 32 (range = 14-46) to 80 (range = 60-100) (p <.01) and VAS from 7.8 (range = 6.1-9.7) to 1.3 (range = 0-6) (p <.01). The average total range of motion increased significantly from 19.8° to 29.2° of plantarflexion and 6.8° to 13.5° of dorsiflexion. Alignment measured by alpha, beta, and gamma angles was satisfactorily achieved. No patient demonstrated any radiographic evidence of tibial or talar lucency at the final follow-up. Five patients (10%) experienced delayed wound healing. One patient (2%) developed a postoperative prosthetic infection. One patient (2%) developed fibular pseudoarthrosis and 2 patients (4%) suffered impingement. Two patients (4%) needed surgery for symptomatic fibular hardware. This study found excellent clinical and radiological results of transfibular total ankle replacement. This is a safe and effective option that allows the correction of sagittal and coronal malalignment. [ABSTRACT FROM AUTHOR]

Las universidades espanolas son un reflejo de las desigualdades de genera en la esfera publica. Diversos trabajos e informes publicados hasta la fecha muestran la falta de equidad en la participaci6n de mujeres y hombres en las instituciones consideradas como democraticas. EI objetivo principal de este trabajo es diagnosticar la composici6n de genera en el ambito de la gerencia universitaria espanola. Asf, se analiza la presencia de la mujer en la universidad espanola, su representaci6n en los 6rganos de gobierno y la evoluci6n de estas cifras en los ultimos anos. Para ello, en primer lugar se revisa la normativa en materia de igualdad de genera, tanto a nivel nacional como europeo y, en segundo lugar, se realiza un analisis longitudinal para abordar la situaci6n de las mujeres y los hombres en las universidades espanolas, tanto publicas como privadas. La investigaci6n se cataloga como descriptiva. Los resultados reflejan una aparente tendencia a la igualdad formal en el acceso discente y docente a la universidad, pera esconde una segregaci6n que sigue manteniendo a las mujeres en situaciones de desigualdad ante los hombres.

This article highlights recent advances in the controlled self-assembly of nanoparticles to produce dimeric nanoparticle structures. The relevance of this emergent field is discussed in terms of recent applications in plasmonics and chemical catalysis. The concept of bond-valence applied to nanoparticles will be discussed, emphasizing some general approaches that have been successfully used to build these structures. Further, the asymmetric functionalization of nanoparticles surfaces as a path to drive selective aggregation, the use of biomolecules to self-assemble nanoparticles into dimers in solution, and the confinement of aggregates in small cavities are discussed.
