Proteine des Haupthistokompatibilitätskomplex (MHC) stellen eines der Schlüsselelemente der adaptiven Immunabwehr dar. Als eine Familie hochpolymorpher Peptidrezeptoren präsentieren sie auf der Zelloberfläche Antigene an T-Zellen. Während MHC Moleküle normalerweise in einem intrazellulären Kompartiment beladen werden, können zumindest prinzipiell Antigene auch direkt auf der Zelloberfläche an dem Peptidrezeptor binden. Dieser Vorgang spielt insbesondere bei Peptidvakzinierungen eine Rolle, ist jedoch auf Grund der Belegung der MHC Bindungsgrube durch Peptidliganden sowie eines Inaktivierungsmechanismus des „leeren“ MHC Moleküls erschwert. In dieser Studie werden zwei Klassen organischer MHC Beladungskatalysatoren („MHC loading enhancer“; MLE) vorgestellt, welche die Peptidbeladung von humanen MHC Molekülen (HLA-DR) an der Zelloberfläche katalysieren und damit die antigenspezifische Immunantwort verstärken. Es konnte u.a. gezeigt werden, dass Adamantanethanol (AdEtOH) als einfache niedermolekulare Verbindung ebenso wie eine Klasse von kurzen Dipeptiden in der Lage sind, die Beladung sowie den Austausch von Antigenen auf HLA-DR Molekülen in vitro höchst effizient zu verstärken. Die katalytische Aktivität beruht hierbei auf der direkten Interaktion der MLE Verbindungen mit einer konservierten Tasche (P1) innerhalb der Peptidbindungsgrube von HLA-DR. In einer Reihe von Peptidimmunisierungen im Mausmodell konnte zusätzlich gezeigt werden, dass die hier identifizierten Verbindungen die spezifische CD4+ T-Zellantwort auch in vivo über den hier vorgestellten Mechanismus modulieren. Über die Erhöhung der Zahl von Peptid/MHC Komplexen auf antigenpräsentierenden Zellen konnte dabei die T-Zellantwort gegen einzelne Peptide, aber auch das komplette tumorassoziierte Antigen NY-ESO-1 in vivo signifikant erhöht werden. Vor diesem Hintergrund stellen die hier beschriebenen MLE somit eine neue Klasse potentieller Adjuvant Additiva dar, welche die Effizienz von Peptidvakzinierungen erhöhen könnten., Class II MHC molecules are a class of highly polymorphic cell surface receptors displaying short protein fragments to CD4+ T cells. As antigens are mainly loaded onto MHC class II molecules in specialized intracellular vesicles, direct binding of peptides on cell surface bound molecules is in principle also possible. On the cell surface most MHC II molecules are occupied by ligands or their binding cleft has been blocked by the acquisition of a non-receptive state. Direct loading with antigens, as required for peptide vaccinations, is therefore hindered. In this study we present two classes of organic ‘MHC-loading enhancer’ (MLE) which can amplify immune responses by catalyzing the peptide loading of human MHC class II molecules. Adamantane ethanol (AdEtOH), a small organic molecule, as well as certain dipeptides could be shown to be powerful enhancers of peptide loading and –exchange reaction in vitro. Additionally we could demonstrate that this effect is based on the direct interaction of MLE with the conserved P1 pocket located in the binding groove of MHC II molecules. Moreover, performing peptide immunizations in mice we could show that the in vivo CD4+ T cell response can be improved when antigens are administered together with MLE. By increasing the number of peptide-MHC complexes on the cell surface of antigen presenting cells (APC) the T cell response against several peptides as well as the tumor-associated antigen NY-ESO-1 protein could be significantly enhanced in vivo. MLE may therefore open a new way to improve vaccination efficacy and tumor-immunotherapy.
