To assess the toxicity of bifenthrin and four mixtures of insecticides to tarnished plant bug, we used an insecticide dip method of green bean to treat adults of a laboratory colony; mortality was assessed after 48 h. LC50s for imidacloprid, bifenthrin, acephate, thiamethoxam, and dicrotophos were 0.12, 0.39, 0.62, 0.67, and 3.96 ppm, respectively. LC75s for imidacloprid, bifenthrin, acephate, thiamethoxam, and dicrotophos were 0.61, 4.22, 5.10, 2.65, and 7.86 ppm, respectively. Based on the LC50s and LC75s, dicrotophos was much less toxic than the other chemicals tested. PoloMix software was used to determine syngerism, antagonism, or addition effects of the mixtures. Three out of four analyses of the joint action of bifenthrin plus imidacloprid or acephate or dicrotophos showed that toxicity was not independent and not correlated. For bifenthrin plus dicrotophos, observed mortality was greater than expected mortality at most concentrations suggesting synergism. Mixtures of bifenthrin plus imidacloprid and bifenthrin plus acephate showed observed mortality significantly less than expected, suggesting antagonism. LC50s for bifenthrin plus dicrotophos, acephate, imidacloprid, and thiamethoxam were 0.38, 1.06, 0.17, and 0.26 ppm, respectively. LC75s for bifenthrin plus dicrotophos, acephate, imidacloprid, and thiamethoxam were 13.61, 13.18, 0.67, and 0.80 ppm, respectively. Based on the LC50s and LC75s, bifenthrin plus acephate was 3- to 10-fold less toxic than the other chemicals tested. Bifenthrin plus acephate is frequently used in tank mixes to control tarnished plant bug and other cotton pests, and the effectiveness of each individual chemical appears to be reduced in one to one ratio mixtures.

The neonicotinoid insecticide acetamiprid (ACT) and seven pesticides [abamectin (ABA), emamectin benzoate (EMB), dicrotophos (DIC), bifenthrin (BIF), cypermethrin (CYP), lambda-cyhalothrin (LCY) and tetraconazole (TET)] are widely applied agrochemicals worldwide. Since most previous studies on these pesticides are performed merely based on toxicity tests with individual active ingredients, only finite knowledge is available on the mixture toxicities of these formulated compounds to crop pollinators. In this study, we examined their toxicities of binary, ternary, quaternary, quinquenary, senary, septenary and octonary mixtures to honey bee (Apis mellifera L.) with feeding toxicity test. Results showed that EMB and ABA had the highest toxicities to A. mellifera with LC50 values of 0.033 (0.028–0.038) and 0.047 (0.039–0.056) μg a. i. mL−1 after exposure for 7 days, respectively, followed by DIC with an LC50 value of 1.22 (1.01–1.41) μg a. i. mL−1. In contrast, relatively low toxicities were found from pyrethroid insecticides, ACT, and TET with their LC50 values ranged from 44.76 (38.75–50.89) to 251.7 (198.4–297.3) μg a. i. mL−1. Most of pesticide mixtures containing ACT and TET elicited synergistic interactions to honey bees. Besides, four pesticide mixtures of ACT + BIF, ACT + BIF + CYP, ACT + BIF + LCY and ACT + CYP + DIC + EMB also displayed synergistic effects. Among 98 tested binary to octonary mixtures of ACT in combination with seven pesticides, 44.90% of combinations exhibited synergistic effects on honey bees. Considering ACT was permitted to use on flowering crops, more attention should be paid to its application in the fields due to the synergistic effects of ACT in combination with other pesticides on A. mellifera under laboratory conditions.

Background Reversible cholinesterase inhibitors, when given prophylactically before exposure to organophosphates, are able to decrease organophosphate-induced mortality. However, the efficacy of pyridostigmine, the only pre-treatment substance approved by the US Federal Drug Administration, is unsatisfactory. Methods In search of a better prophylactic compound, we determined in vivo the protection conferred by five cholinesterase inhibitors (ranitidine, physostigmine, tacrine, K-27 and pyridostigmine), which were administered in equitoxic dosage (1/4 of LD01) 30 minutes before exposure to the organophosphate dicrotophos. Efficacy was measured in rats by Cox analysis calculating the relative risk of death (RR), RR being 1 for the reference group which received dicrotophos and no prophylaxis. Results K-27 (RR=0.06), physostigmine (RR=0.15), pyridostigmine (RR=0.22) and tacrine (RR=0.28) significantly (p ≤ 0.05) reduced dicrotophos-induced mortality in comparison to the reference group (dicrotophos without pre-treatment), whereas ranitidine (RR=0.86) had no significant influence. The experimental oxime K-27, when given before dicrotophos exposure, conferred the best in vivo protection. This was significantly (p ≤ 0.05) more efficacious than pre-treatment with any other tested compound. The differences in efficacy between the second best compound, physostigmine, and the less efficacious substances (tacrine and pyridostigmine) were also statistically significant. Conclusion These data indicate that K-27 can be considered a very efficacious prophylactic agent for organophosphate exposure.

Profenofos (PF) is one of the heavily used organophosphorus pesticides (OPPs) of which its contamination is ubiquitous in an agricultural area. This study aims to acquire and characterize PF-degrading bacterial cultures from contaminated soil. OPP degradation by the novel isolates was then investigated. The experiment was performed at the initial PF concentration of 20 mg/L. The result showed that the enriched consortium comprised three predominant PF-degrading strains designated as PF1, PF2, and PF3. The isolates (PF1, PF2, and PF3) were characterized as Pseudomonas plecoglossicida, Pseudomonas aeruginosa, and P. aeruginosa, respectively. A consortium and all isolates could utilize PF as a sole carbon source with PF removal of more than 90% via a hydrolysis process. The bacterial growth and PF degradation rates followed the first-order kinetic reaction with the rates of 0.4 to 2.7/h and 0.15 to 1.96/h, respectively. Additional carbon supplement deteriorated PF biodegradation. The enriched cultures were also capable for degrading chlorpyrifos and dicrotophos pesticides (33-73% removal). The results indicated that the consortium and isolates are efficient for PF and other OPP degradation and have potential for PF remediation.

The southern green stink bug, Nezara viridula (L.) (Heteroptera: Pentatomidae), is cosmopolitan in distribution and feeds on a multitude of food and fiber crops. It induces abscission of bolls of cotton, Gossypium hirsutum L., and reduces seed cotton yield, gin-turnout, and fiber quality. Also, the bug vectors bacterial and fungal pathogens causing necrosis of the locule and rotting the bolls. Control options are mostly dependent upon and limited to the use of insecticides. Data are needed to assess the toxicity of currently available insecticides and develop baseline mortality data to monitor resistance of the insect to insecticides in Central Texas. Application of insecticides for stink bug management through overhead irrigation, also called chemigation, could reduce application costs, soil compaction, and applicator exposure, while enabling growers to treat multiple fields simultaneously. The objective of these laboratory experiments was to compare knockdown, survival, and efficacy of insecticides when appropriately diluted for ground sprayer and chemigation applications. Treatments included water, bifenthrin [0.11 kg (AI)/ha] and dicrotophos [0.56 kg (AI)/ha] diluted for a ground sprayer (93.5 liters/ha), bifenthrin and dicrotophos diluted for chemigation (25,396 liters/ha), and bifenthrin and dicrotophos plus adjuvants diluted for ground sprayer or chemigation. Two- to 14-day-old adults of Nezara viridula (L.), Euschistus servus (Say), and Halyomorpha halys (Stal) were briefly submerged in appropriately diluted insecticides and then introduced into a disposable petri dish with or without food. Dishes were placed in a growth chamber provisioned with digital video cameras to monitor knockdown and feeding after insecticide exposure. Knockdown was visually assessed at 24 h after treatment followed by mortality and recovery from knockdown at 48 h after treatment. All stink bugs were knocked down within 1 h and never recovered when exposed at ground sprayer dilutions. However, many bugs survived chemigation dilutions. Less than half of the stink bugs were knocked down when exposed to dicrotophos (with or without adjuvants) and survival ranged from 17 to 77%, compared to 7-90% survival when exposed to bifenthrin at chemigation dilutions. These results strongly suggest that chemigation applications for stink bug management need to be closely examined.

Electrocatalytic properties of cobalt phthalocyanine (CoPc), cobalt tetra-carboxy phthalocyanine (CoTCPc) and cobalt octa-carboxy (CoOCPc), towards the detection of dicrotophos have been studied. Catalytic behavior towards the reduction of dicrotophos was found to be dependent on the pH, as well as the substitution on the phthalocyanine ring. Strong electron withdrawing groups on the phthalocyanine ring yielded best catalysis as evidenced by the enhancement of the reduction peak current, (~5 fold) compared to the bare glassy carbon electrode. The analysis gave a good detection limit of 1.25 × 10-7 M, and good linearity for the studied concentration range. A high Tafel slope value was obtained, indicating a strong interaction between dicrotophos and the cobalt phthalocyanine complex.

Common Quail (Coturnix coturnix) were subjected to controlled and replicated experiments in the summer of 2008 to investigate the effects of short-term dehydration on cholinesterase activity in brain and plasma and the interaction between dehydration and exposure to the organophosphorus pesticide dicrotophos in these same tissues. Our objective was to determine if dehydration could confound the diagnosis of anticholinesterase exposure using inhibition of cholinesterase activity in quail tissues. The effect of dehydration was quantified using measures of plasma osmolality and hematocrit. Dicrotophos exposure caused significant inhibition of cholinesterase activity in brain, while the effects of dehydration and interaction were not significant. Dehydration caused significant duration-dependent increases in plasma osmolality and hematocrit. Dehydration also caused a significant increase in plasma cholinesterase activity. Variation in the change in plasma cholinesterase activity in response to dehydration was significantly and positively correlated with dehydration-induced variation in both the change in plasma osmolality and the change in hematocrit. These correlations suggest that plasma cholinesterase activity in quail is not limited to plasma but occupies some larger pool of the extracellular fluid volume, and we suggest lymph is part of that pool. The effects of dehydration on plasma cholinesterase activity masked the inhibitory effects of dicrotophos. Here, the combination of dehydration and dicrotophos exposure produced plasma cholinesterase activity that was not significantly different from reference and pre-exposure values, confounding the diagnosis of anticholinesterase exposure in dehydrated, dicrotophos-exposed quail. A method to adjust plasma cholinesterase activities for the confounding effects of dehydration and enable the diagnosis of anticholinesterase exposure in dehydrated, dicrotophos-exposed quail was developed. Clinicians and practitioners responsible for the diagnosis of anticholinesterase exposure in birds are cautioned that dehydration, commonly observed in sick wildlife, may mask the effect of anticholinesterases on plasma cholinesterase activity.

Brown stink bug, Euschistus servus (Say), has greatly increased in numbers in the Brazos Valley in Central Texas and in many other areas of the Cotton Belt, and has become a major pest of cotton, Gossypium hirsutum L., and other crops including pecans, Carya illinoensis (Wangenh.) K. Koch. Use of insecticides is among the most important of the limited control options available against this pest. Glass-vial bioassays were used to evaluate the toxicity of selected synthetic pyrethroid and organophosphate insecticides to adult brown stink bug captured in blacklight traps. A comparison was also made of toxicity of λ-cyhalothrin to brown stink bug collected directly from the field to those captured in blacklight traps. Lethal concentration value (LC50) (upper and lower 95% confidence limits) for dicrotophos for 24-hour response, 0.30 (0.24–0.37) µg per vial, was significantly less compared to acephate with an LC50 of 1.38 (1.01–1.81) µg per vial and chlorpyrifos with an LC50 of 5.00 (4.27–5.67) µg pe...

Age-related differences in the acute neurotoxicity of cholinesterase (ChE)-inhibiting pesticides have been well-studied for a few organophosphates, but not for many others. In this study, we directly compared dose-responses using brain and red blood cell (RBC) ChE measurements, along with motor activity, for mevinphos, monocrotophos, dicrotophos, and phosphamidon. Long-Evans hooded male rats were tested as adults and at postnatal day (PND) 17; PND11 pups were also tested with dicrotophos only. All chemicals were administered via oral gavage and tests were conducted at times intended to span peak behavioral and ChE effects. All OPs tested produced a rapid onset and recovery from the behavioral effects. There were age-related differences in the inhibition of brain, but not necessarily RBC, ChE. Mevinphos was clearly more toxic, up to 4-fold, to the young rat. On the other hand, monocrotophos, dicrotophos, and phosphamidon were somewhat more toxic to the young rat, but the magnitude of the differences was2-fold lower. Motor activity was consistently decreased in adults for all chemicals tested; however, there was more variability with the pups and clear age-related differences were only observed for mevinphos. These data show that three of these four OPs were only moderately more toxic in young rats, and further support findings that age-related differences in pesticide toxicity are chemical-specific.

BACKGROUND: The commercialization of new insecticides is important for ensuring that multiple effective product choices are available. In particular, new insecticides that exhibit high potency and lack insecticidal cross-resistance are particularly useful in insecticide resistance management (IRM) programs. Sulfoxaflor possesses these characteristics and is the first compound under development from the novel sulfoxamine class of insecticides. RESULTS: In the laboratory, sulfoxaflor demonstrated high levels of insecticidal potency against a broad range of sap-feeding insect species. The potency of sulfoxaflor was comparable with that of commercial products, including neonicotinoids, for the control of a wide range of aphids, whiteflies (Homoptera) and true bugs (Heteroptera). Sulfoxaflor performed equally well in the laboratory against both insecticide-susceptible and insecticide-resistant populations of sweetpotato whitefly, Bemisia tabaci Gennadius, and brown planthopper, Nilaparvata lugens (Stal), including populations resistant to the neonicotinoid insecticide imidacloprid. These laboratory efficacy trends were confirmed in field trials from multiple geographies and crops, and in populations of insects with histories of repeated exposure to insecticides. In particular, a sulfoxaflor use rate of 25 g ha−1 against cotton aphid (Aphis gossypii Glover) outperformed acetamiprid (25 g ha−1) and dicrotophos (560 g ha−1). Sulfoxaflor (50 g ha−1) provided a control of sweetpotato whitefly equivalent to that of acetamiprid (75 g ha−1) and imidacloprid (50 g ha−1) and better than that of thiamethoxam (50 g ha−1). CONCLUSION: The novel chemistry of sulfoxaflor, its unique biological spectrum of activity and its lack of cross-resistance highlight the potential of sulfoxaflor as an important new tool for the control of sap-feeding insect pests. Dicrotophos is a systemic insecticide with a wide range of applications. We investigated the genotoxicity of dicrotophos using the Ames test, the chromosome aberration test in CHO-K1 cells, and the comet assay in the Hep G2 cells, while this chemicals' toxicity to both the cell lines was evaluated with the MTT assay. Results showed that dicrotophos did not show any cytotoxicity to CHO-K1 cells, whereas it was cytotoxic to HepG2 cells incubated for 24 h but not for 2 h. For genotoxicity of dicrotophos, a significant change in the numbers of bacterial reveratnts using Salmomella typhimurium TA97a, TA98, TA100, TA102, and TA1535 as the tester strains, an increase in the frequencies of chromosome aberration in CHO-K1 cells, and an induced DNA damage in HepG2 cells were observed, indicating that dicrotophos was genotoxic in these three performed assays. From this study, we provide further evidence towards of genotoxic effects of dicrotophos.

Stink bugs have become important pests of cotton, Gossypium hirsutum L., in the Lower Gulf Coast region of Texas. Common species, damage potential, and insecticide susceptibility of stink bugs were assessed. A survey of stink bugs in commercial cotton fields in six counties in the Lower Gulf Coastal region of Texas during 2004 and 2005 revealed that brown stink bug, Euschistus servus (Say); E. quadrator Rolston; and E. obscurus (Palisot) were the most common species (>95% both years), while southern green stink bug, Nezara viridula (L.); and green stink bug, Acrosternum hilare (Say), were less common. This stink bug complex differs from that reported for other areas of the Cotton Belt. Also, significantly more stink bugs were collected in the northern counties of Calhoun, Refugio, and Victoria than in the southern counties of Kleberg, Nueces, and San Patricio (approximately 7.5 and 4 times more in 2004 and 2005, respectively). The potential of E. servus and E. quadrator to damage cotton bolls was compared by caging individual adults on bolls in the field. Both E. servus and E. quadrator caused significant reductions in yield (seed cotton weight) and fiber quality (length and fineness) of cotton bolls. Feeding by both species caused abscission of 1.8-cm-diameter bolls from the plant. In general, E. servus caused more damage to bolls than did E. quadrator and damaged a wider size-range of bolls. Foliar-applied insecticides were evaluated against E. servus, E. quadrator, and N. viridula in small plots of cotton in the field. Dicrotophos was most effective against all species. Pyrethroids killed all N. viridula, but the number of E. servus and E. quadrator killed varied significantly. Euschistus quadrator was more susceptible than E. servus to organophosphate, pyrethroid, neonicotinoid, and carbamate insecticides.

The toxicity of 10 organophophorus (OP) insecticides-acephate, dimethoate, dichlorvos, dicrotophos, monocrotophos, methamidophos, phosphamidon, omethoate, phosdrin, and trichlorfon-was evaluated in Caenorhabditis elegans using lethality, movement, and acetylcholinesterase (AChE) activity as the endpoints after a 4-hr- exposure period. The OP insecticides tested showed LC50 values ranging from 0.039 mM (for dichlorovs) to 472.8 mM (for methamidophos). The order of toxicity for lethality and movement was not significantly different when tested using the rank order correlation coefficient. AChE activity was markedly affected by all the OP insecticide exposures that caused significant inhibition in movement, indicating that the mechanism of toxicity of OP insecticides in C. elegans is the same as in higher animals. All OP insecticides induced greater than 50% inhibition of AChE at the lowest tested OP insecticide concentration resulting in inhibition in movement. While a significant correlation was evident between LC50 values in C. elegans and the LD50 values in rats for the 10 OP insecticides studied, a correlation was not evident between EC50 values in C. elegans and LD50 values in rats. Overall, the two endpoints, LC50 and movement, were more reliable and easier to perform than measurement of AChE activity in C. elegans for determining the toxicity of OP insecticides. Further, ranking of these endpoints with respect to the OP insecticides studied indicates that these parameters in C. elegans are predictive of OP insecticides mammalian neurotoxicity.

Glass vials coated with several technical insecticides were used to determine the contact toxicity of insecticides on adult laboratory-reared and field-collected cotton fleahopper, Pseudatomoscelis seriatus (Reuter). For the 17 insecticides evaluated for laboratory-reared cotton fleahoppers, bifenthrin (pyrethroid), dicrotophos (organophosphate), thiamethoxam (neonicotinoid), and methomyl (carbamate) were the most toxic insecticides in their respective insecticidal classes based on LC50 values. There were significant differences between the LC50 values for the insecticides tested within each of the four insecticidal classes. There were 13-, 46-, 58-, and 31-fold differences between LC50 values for the insecticides within the pyrethroid, organophosphate, neonicotinoid, and carbarmate classes, respectively. Among fleahoppers collected from horsemint in May/June, adult vial testing showed increased susceptibility in males versus females. This difference can be attributed, at least in part, to differences in insect weights between the males and females since the females weighed significantly more than the males. Data presented herein provide a measure of acute potency of various insecticides against P. seriatus and serve as a measure of inherent relative differences between the insecticides. Baseline data will be useful for future comparison should suspicion of tolerance to these insecticides develop in field populations. These data are also important in comparing results from laboratory and field studies with cotton fleahoppers.

Insecticide resistance is usually associated with pests, but may also evolve in natural enemies. In this study, adult beetles of three distinct North American populations of Hippodamia convergens Guerin-Meneville, and the progeny of reciprocal crosses between the resistant and most susceptible population, were treated topically with varying concentrations of lambda-cyhalothrin and dicrotophos. In addition, the LD50s of both insecticides were applied in combination to resistant individuals. The developmental and reproductive performance of each population was assessed in the absence of insecticide exposure to compare baseline fitness. California and Kansas populations were susceptible to both materials, whereas Georgia (GA) beetles exhibited a resistance ratio (RR50) of 158 to lambda-cyhalothrin and 530 to dicrotophos. Inheritance of lambda-cyhalothrin resistance was X-linked, whereas inheritance of dicrotophos resistance was autosomal. Mortality of resistant beetles treated with a mixture of LD50s of both materials was twice that of those treated with lambda-cyhalothrin alone, but not significantly different from those receiving dicrotophos alone. Life history parameters were largely similar among populations, except that Georgia beetles had higher egg fertility relative to susceptible populations. We conclude that the high levels of resistance to lambda-cyhalothrin and dicrotophos in Georgia beetles reflect heavy loads of these insecticides in local environments, most likely the large acreage under intensive cotton cultivation.
