The olfactory bulb is a unique site of continuous neurogenesis, primarily generating inhibitory interneurons, a process that begins at birth and extends through infancy and adulthood. This review examines the characteristics of olfactory bulb neurogenesis, focusing on granule cells, the most numerous interneurons, and how their age and maturation affect their function. Adult-born granule cells, while immature, contribute to the experience-dependent plasticity of the olfactory circuit by enabling structural and functional synaptic changes. In contrast, granule cells born early in life form the foundational elements of the olfactory bulb circuit, potentially facilitating innate olfactory information processing. The implications of these neonatal cells on early life olfactory memory and their impact on adult perception, particularly in response to aversive events and susceptibility to emotional disorders, warrant further investigation. Polyamines are involved in the control of the cell cycle and cell growth. In murine kidney, testosterone enhances gene expression of ornithine decarboxylase (ODC), the first enzyme in polyamine biosynthesis. In this study, we document the time course effect of testosterone on 1) gene expression of ODC, antizyme 1 (AZ1), and spermidine/spermine-[N.sup.1]-acetyltransferase ([N.sup.1]-SSAT); 2) ODC activity in proximal convoluted tubules (PCT) and cortical proximal straight tubules (CPST); and 3) renal polyamine levels. Female mice were treated with testosterone for a period of 1, 2, 3, and 5 consecutive days. ODC gene expression was extremely low in kidneys of untreated female mice compared with that of males. Consequently, the renal putrescine level was sevenfold lower in females than in males, whereas spermidine and spermine levels did not differ between sexes. In female kidneys, testosterone treatment sharply increased ODC mRNA and protein levels as well as ODC activity. Testosterone increased the expression of ODC in PCT and CPST over different time courses, which suggests that ODC activity is differentially regulated in distinct tubules. The expression of AZ1 and [N.sup.1]-SSAT mRNA was similar in male and female mouse kidneys. Testosterone treatment enhanced AZ1 and [N.sup.1]-SSAT mRNA levels in a time-dependent manner by unknown molecular mechanisms. Putrescine and spermidine levels increased after testosterone treatment in female kidneys. Surprisingly, although ODC protein and activity were undetectable in female kidneys, the levels of AZ1 mRNA and protein were similar to those in males. - Abstract
Background Little is known about the incidence of severe critical events in children undergoing general anaesthesia in Europe. We aimed to identify the incidence, nature, and outcome of severe critical events in children undergoing anaesthesia, and the associated potential risk factors. Methods The APRICOT study was a prospective observational multicentre cohort study of children from birth to 15 years of age undergoing elective or urgent anaesthesia for diagnostic or surgical procedures. Children were eligible for inclusion during a 2-week period determined prospectively by each centre. There were 261 participating centres across 33 European countries. The primary endpoint was the occurence of perioperative severe critical events requiring immediate intervention. A severe critical event was defined as the occurrence of respiratory, cardiac, allergic, or neurological complications requiring immediate intervention and that led (or could have led) to major disability or death. This study is registered with ClinicalTrials.gov, number NCT01878760. Findings Between April 1, 2014, and Jan 31, 2015, 31 127 anaesthetic procedures in 30 874 children with a mean age of 6.35 years (SD 4.50) were included. The incidence of perioperative severe critical events was 5.2% (95% CI 5.0-5.5) with an incidence of respiratory critical events of 3.1% (2.9-3.3). Cardiovascular instability occurred in 1.9% (1.7-2.1), with an immediate poor outcome in 5.4% (3.7-7.5) of these cases. The all-cause 30-day in-hospital mortality rate was 10 in 10 000. This was independent of type of anaesthesia. Age (relative risk 0.88, 95% CI 0.86-0.90; p<0.0001), medical history, and physical condition (1.60, 1.40-1.82; p<0.0001) were the major risk factors for a serious critical event. Multivariate analysis revealed evidence for the beneficial effect of years of experience of the most senior anaesthesia team member (0.99, 0.981-0.997; p<0.0048 for respiratory critical events, and 0.98, 0.97-0.99; p=0.0039 for

International audience; BackgroundIn the adult brain, structural plasticity allowing gain or loss of synapses remodels circuits to support learning. In Fragile X Syndrome (FXS), the absence of Fragile X Mental Retardation Protein (FMRP) leads to defects in plasticity and learning deficits. FMRP is a master regulator of local translation but its implication in learning-induced structural plasticity is unknown.MethodsUsing an olfactory learning task requiring adult-born olfactory bulb (OB) neurons and cell-specific ablation of FMRP, we investigated whether learning shapes adult-born neuron morphology during their synaptic integration and its dependence on FMRP. We used αCaMKII mutant mice with altered dendritic localization of αCaMKII mRNA as well as a reporter of αCaMKII local translation to investigate the role of this FMRP mRNA target in learning-dependent structural plasticity.ResultsLearning induces profound changes in dendritic architecture and spine morphology of adult-born neurons that are prevented by ablation of FMRP in adult-born neurons and rescued by an mGLUR5 antagonist. Moreover, dendritically translated αCaMKII is necessary for learning and associated structural modifications and learning triggers an FMRP-dependent increase of αCaMKII dendritic translation in adult-born neurons.ConclusionOur results strongly suggest that FMRP mediates structural plasticity of OB adult-born neurons to support olfactory learning through αCaMKII local translation. This reveals a new role for FMRP-regulated dendritic local translation in learning-induced structural plasticity. This might be of clinical relevance for the understanding of critical periods disruption in autism spectrum disorder patients, among which FXS is the primary monogenic cause.

The French sugar beet industry seeks to increase productivity in order to obtain sugar yields economically satisfactory. Nitrogen fertilization is one of the main factors influencing yield of sugar beet. Many models are designed to simulate the yield of sugar beet but few take into account the effect of nitrogen nutrition conditions. In this context, the objectives of this work were: (i) to reflect leading to the choice of the type of modeling, (ii) to identify the plant characteristics in response to nitrogen nutrition for parameterization of the model and (iii) evaluate our model.The modeling approach has focused on the integration of the nitrogen nutrition effects on the leaf area development, and source-sink relationships within the plant. Two years field experiments have enabled us to acquire data necessary for the study of the sugar beet responses under contrasting nitrogen nutrition conditions and parameterization of the model. A compilation of a data set validation at the ITB makes it possible to take into account parameters variability. We found that sugar beet responded to nitrogen nutrition as the other C3 plants. The model simulates the differences in sugar yield based on nitrogen nutrition status as reported in the literature. The model can find growth stages of sugar beet without force them. Coupled to the supply nitrogen module of the STICS crop model, the model simulates the major trends in yields over the past thirty years.; La filière betteravière française cherche à gagner en productivité afin d'obtenir des rendements en sucre économiquement satisfaisants. La fertilisation azotée est l'un des principaux facteurs influençant le rendement de la betterave sucrière. De nombreux modèles ont pour but de simuler le rendement en sucre de la betterave mais peu prennent en compte l'effet des conditions de nutrition azotée. Neural stem cells (NSC) persist in the adult mammalian brain, within the subventricular zone (SVZ). The endogenous mechanisms underpinning SVZ stem and progenitor cell proliferation are not fully elucidated. Vitamin K-dependent proteins (VKDPs) are mainly secreted factors that were initially discovered as major regulators of blood coagulation. Warfarin ((S(2)-3-acetonylbenzyl)-4- hydroxy-coumarin)), a widespread anticoagulant, is a vitamin K antagonist that inhibits the production of functional VKDP. We demonstrate that the suppression of functional VKDPs production, in vitro, by exposure of SVZ cell cultures to warfarin or, in vivo, by its intracerebroventricular injection to mice, leads to a substantial increase in SVZ cell proliferation. We identify the anticoagulant factors, protein S and its structural homolog Gas6, as the two only VKDPs produced by SVZ cells and describe the expression and activation pattern of their Tyro3, Axl, and Mer tyrosine kinase receptors. Both in vitro and in vivo loss of function studies consisting in either Gas6 gene invalidation or in endogenous protein S neutralization, provided evidence for an important novel regulatory role of these two VKDPs in the SVZ neurogenic niche. Specifically, we show that while a loss of Gas6 leads to a reduction in the numbers of stem-like cells and in olfactory bulb neurogenesis, endogenous protein S inhibits SVZ cell proliferation. Avec plus de 900 000 nouveaux cas chaque année chez les hommes et 330 000 chez les femmes, le cancer du poumon est actuellement l’un des cancers les plus répandus dans le monde. La majorité des patients atteints de ce cancer subiront un traitement par radiothérapie. La radiothérapie ainsi que l’hadronthérapie nécessitent un contrôle précis sur la position du volume tumoral durant le traitement, afin d'éviter l'irradiation des tissus sains. Lorsque la tumeur se trouve sur un organe en mouvement, la difficulté majeure est de cibler la tumeur pendant le traitement. Ce facteur limitant est particulièrement marqué dans le cas des tumeurs pulmonaires, ce qui explique les résultats cliniques encore insatisfaisants. Les stratégies actuelles utilisées en clinique, permettant de prendre en compte le mouvement de la tumeur au cours de la respiration, présentent toutes des inconvénients majeurs. Par exemple, certaines consistent à augmenter les marges d’irradiation ce qui entraine un dépôt de dose important sur les tissus sains avoisinants, d’autres, vont consister à suivre des implants positionnés dans la tumeur ce qui rend la thérapie très invasive et enfin, certaines vont irradier la tumeur à un instant précis du cycle respiratoire, faisant ainsi l’hypothèse contestée d’une reproductibilité du mouvement tumoral au cours du cycle respiratoire. Notre objectif est de mettre en place un modèle biomécanique des poumons et de leur environnement afin de prédire la position de la tumeur à partir de paramètres mesurables directement en externe tels que la quantité d’air inspirée ou encore le mouvement de la surface externe de la peau. Ce modèle s’appuie sur les données anatomiques du patient (géométriques, physiologiques et biomécaniques) et permet de reproduire la variabilité du mouvement respiratoire de façon non-invasif. La variabilité du mouvement pulmonaire s’expliquant, en particulier, par l'action assez indépendante des muscles intercostaux et du diaphragme lors de la respiration, le modèle biomécanique développé au cours de cette thèse comprend la modélisation de ces éléments anatomiques. Des études sur la cinématique des côtes ainsi que sur le mouvement du diaphragme et le comportement des tissus mous ont été menées et ont permis, par la suite, d’établir un modèle de l’environnement pulmonaire précis et personnalisé. Le rôle important de la plèvre lors de la respiration a été démontré et celle-ci a donc été intégrée au modèle biomécanique du système respiratoire. Les simulations ont été réalisées à l’aide d’un logiciel de calcul par éléments finis et une relation entre le mouvement externe de la peau et le mouvement interne des côtes et donc des poumons a pu être établie.
