Abstract

Dopamine is a chemical substance that plays an important role in the transmission of certain nerve signals. It is thought that abnormalities related to dopamine and its actions could be responsible or partially responsible for some neurological and psychiatric disorders. A few studies have reported an association between alcoholism and the presence of a variation of a gene associated with the expression of the dopamine D2 (DRD2) receptor, labelled the A1 allele. Other studies have not found such an association. These conflicting results could be explainable if the A1 allele is a modifying gene related to alcoholism as opposed to being a gene specifically responsible for alcoholism. To test this hypothesis, the prevalence of the A1 allele in various groups of people was measured. A total of 853 subjects were examined, including 314 healthy control subjects, 147 subjects with Tourette's syndrome (TS), 104 with attention deficit hyperactivity disorder (ADHD), 33 with autism, 104 who were alcoholics, 35 who suffered from posttraumatic stress syndrome, 13 who suffered panic attacks, 17 who had Parkinson's disease, and 10 who were obese. The A1 allele was found in 24.5 percent of the control subjects. It was found in 44.9 percent of subjects with TS, 46.2 percent of subjects with ADHD, 54.4 percent of the autistic patients, and 42.3 percent of the chronic alcoholics The prevalence for all of these groups was significantly greater than that in the control group. The differences in prevalence between the control group and the other groups studied were not statistically different. These results indicate that the A1 allele is related to, but not the direct cause of, a number of neuropsychiatric disorders. (Consumer Summary produced by Reliance Medical Information, Inc.)

Abstract

Meta-analysis and meta-regression are statistical methods for synthesizing and modelling the results of different studies, and are critical research synthesis tools in ecology and evolutionary biology (E&E). However, many E&E researchers carry out meta-analyses using software that is limited in its statistical functionality and is not easily updatable. It is likely that these software limitations have slowed the uptake of new methods in E&E and limited the scope and quality of inferences from research syntheses., We developed OpenMEE: Open Meta-analyst for Ecology and Evolution to address the need for advanced, easy-to-use software for meta-analysis and meta-regression. OpenMEE has a cross-platform, easy-to-use graphical user interface (GUI) that gives E&E researchers access to the diverse and advanced statistical functionalities offered in R, without requiring knowledge of R programming., OpenMEE offers a suite of advanced meta-analysis and meta-regression methods for synthesizing continuous and categorical data, including meta-regression with multiple covariates and their interactions, phylogenetic analyses, and simple missing data imputation. OpenMEE also supports data importing and exporting, exploratory data analysis, graphing of data, and summary table generation., As intuitive, open-source, free software for advanced methods in meta-analysis, OpenMEE meets the current and pressing needs of the E&E community for teaching meta-analysis and conducting high-quality syntheses. Because OpenMEE's statistical components are written in R, new methods and packages can be rapidly incorporated into the software. 31. A study of the dopamine D2receptor gene in pathological gambling

Abstract

Pathological gambling has been termed both the 'pure' and the 'hidden' addiction. 'Pure' because it is not associated with the intake of any addicting substance, and 'hidden' because it is an extension of a common, socially accepted behaviour. The Taq Al variant of the human DRD2 gene has been associated with drug addiction, some forms of severe alcoholism, and other impulsive, addictive behaviours. We have sought to determine if there is a similar association with pathological gambling. A total of 222 non-Hispanic Caucasian pathological gamblers from multiple sites across the US participated in the study. Of these 171 donated a sample of blood, 127 filled out several questionnaires, and 102 did both. Of the 171 pathological gamblers 50.9 carried the D2Al allele versus 25.9 of the 714 known non-Hispanic Caucasian controls screened to exclude drug and alcohol abuse, p<0.00000001, odds ratio (OR)2.96. For the 102 gamblers who filled out the questionnaires, 63.8 of those in the upper half of the Pathological Gambling Score (more severe) carried the D2A1 allele (OR versus controls5.03), compared to 40.9 in the lower half (less severe). Of those who had no comorbid substance abuse, 44.1 carried the D2A1 allele, compared to 60.5 of those who had comorbid substance abuse. Forty-eight controls and 102 gamblers completed a shorter version of the Pathological Gambling Score. Of the 45 controls with a score of zero, 17.8 carried the D2A1 allele. Of the 99 gamblers with a score of 5 or more, 52.5 carried the D2A1 allele (X215.36, p0.00009). These results suggest that genetic variants at the DRD2 gene play a role in pathological gambling, and support the concept that variants of this gene are a risk factor for impulsive and addictive behaviours.

32. Dehydrogenase Activity Involved in the Uptake of Glucose 6-Phosphate by a Bacterial Membrane System

Abstract

Membranes prepared from Escherichia coliinduced or constitutive for glucose 6-phosphate transport accumulate this compound from the medium. The transport is independent of the phosphotransferase system, insofar as a mutant deficient in Enzyme I of this system yields membranes fully active in glucose 6-phosphate uptake; and the addition of phosphoenolpyruvate to induced wild type membranes did not stimulate glucose 6-phosphate uptake. Both the rate and extent of uptake are, however, stimulated several-fold in the presence of either d(-)-lactate or succinate. In the case of the d(-)-lactic dehydrogenase activity there is far more pyruvate produced than glucose 6-phosphate transported. Evidence presented indicates that it is the free glucose 6-phosphate which is accumulated, and that this material is freely exchangeable with external compound.
