The novel object recognition (NOR) paradigm is a cognitive test that has been used with many species to detect differences in ability. Various iterations of the paradigm have been implemented, making it difficult to compare results both within and across species. Interpretations of the results are equally diverse, threatening the integrity of the paradigm. These inconsistencies have prompted a deeper dive into the variability of the resultant data. For the purposes of this meta-analysis, data originated from 12 studies involving 367 pigs that were subjected to the same NOR paradigm beginning between postnatal days 21 and 24. The main cognitive measure from the NOR paradigm is recognition index (RI), which was the focus of most of the analyses in this meta-analysis. RI was chosen as the main outcome as it determines a pig's preference for novelty, an innate behavior of cognitively intact pigs. A histogram of RI values (range 0 to 1) showed a bimodal distribution skewed to the right, suggesting that the interpretation of positive performance on the task may need to be stricter. Correlational analyses proved that the number of investigations and investigation time with both the novel and familiar objects were the strongest predictors of resultant RI values. Objective data inclusion criteria were then considered to eliminate non-compliant pigs. Results indicated that requiring at least 5 s of investigation over a minimum of 3 investigations with the novel object reduced overall variability for RI with a concomitant increase in the mean. Further analyses showed that pigs preferred to spend more time with and interact more with the novel object across the entire testing trial, especially in the first minute. Together, these findings suggest that future interpretations of NOR should consider applying stricter statistical analyses as well as additional data processing, such as binning, with emphasis on novel object and familiar object investigation. BackgroundMilk oligosaccharides (OSs) are bioactive components known to influence neonatal development. These compounds have specific physiological functions acting as prebiotics, immune system modulators, and enhancing intestine and brain development.ObjectivesThe pig is a commonly used model for studying human nutrition, and there is interest in quantifying OS composition of porcine milk across lactation compared with human milk. In this study, we hypothesized that OS and sialic acid (SA) composition of porcine milk would be influenced by stage of lactation.MethodsUp to 250 mL of milk were collected from seven sows at each of three time points: day 0 (colostrum), days 7-9 (mature), and days 17-19 (weaning). Colostrum was collected within 6 h of farrowing and 3-day intervals were used for mature and weaning milk to ensure representative sampling. Milk samples were analyzed for OS profiles by Nano-LC Chip-QTOF MS, OS concentrations via HPAEC-PAD, and SA (total and free) was assessed by enzymatic reaction fluorescence detection.ResultsSixty unique OSs were identified in porcine milk. This study investigated the effects of Lacticaseibacillus rhamnosus HN001 supplementation on the architecture and gene expression in small intestinal tissues of piglets used as an animal model for infant humans. Twenty-four 10-d-old entire male piglets (4·3 (sd 0·59) kg body weight) were fed an infant formula (IF) (control) or IF supplemented with 1·3 × 105 (low dose) or 7·9 × 106 (high dose) colony-forming units HN001 per ml of reconstituted formula (n 8 piglets/treatment). After 24 d, piglets were euthanised. Samples were collected to analyse the histology and gene expression (RNAseq and qPCR) in the jejunal and ileal tissues, blood cytokine concentrations, and blood and faecal calprotectin concentrations. HN001 consumption altered (false discovery rate < 0·05) gene expression (RNAseq) in jejunal tissues but not in ileal tissues. The number of ileal goblet cells and crypt surface area increased quadratically (P < 0·05) as dietary HN001 levels increased, but no increase was observed in the jejunal tissues. Similarly, blood plasma concentrations of IL-10 and calprotectin increased linearly (P < 0·05) as dietary HN001 levels increased. In conclusion, supplementation of IF with HN001 affected the architecture and gene expression of small intestine tissue, blood cytokine concentration and frequencies, and blood calprotectin concentrations, indicating that HN001 modulated small intestinal tissue maturation and immunity in the piglet model. Introduction: Human milk contains structurally diverse oligosaccharides (HMO), which are multifunctional modulators of neonatal immune development. Our objective was to investigate formula supplemented with fucosylated (2'FL) + neutral (lacto-N-neotetraose, LNnt) oligosaccharides and/or sialylated bovine milk oligosaccharides (BMOS) on immunological outcomes. Methods: Pigs (n=46) were randomized at 48h of age to four diets: sow milk replacer formula (CON), BMOS (CON + 6.5 g/L BMOS), HMO (CON + 1.0 g/L 2'FL + 0.5 g/L LNnT), or BMOS+HMO (CON + 6.5 g/L BMOS + 1.0 g/L 2'FL + 0.5 g/L LNnT). Blood and tissues were collected on postnatal day 33 for measurement of cytokines and IgG, phenotypic identification of immune cells, and ex vivo lipopolysaccharide (LPS)-stimulation of immune cells. Results: Serum IgG was significantly lower in the HMO group than BMOS +HMO but did not differ from CON or BMOS. The percentage of PBMC Thelper cells was lower in BMOS+HMO than the other groups. Splenocytes from the BMOS group secreted more IL-1β when stimulated ex vivo with LPS compared to CON or HMO groups. For PBMCs, a statistical interaction of BMOS*HMO was observed for IL-10 secretion (p=0.037), with BMOS+HMO and HMO groups differing at p=0.1. Discussion: The addition of amix of fucosylated and sialylated oligosaccharides to infant formula provides specific activities in the immune system that differ from formulations supplemented with one oligosaccharide structure.
