Myoepithelial cells play key roles in normal mammary gland development and in limiting pre-invasive to invasive breast tumor progression, yet their differentiation and perturbation in ductal carcinoma in situ (DCIS) are poorly understood. Here, we investigated myoepithelial cells in normal breast tissues of BRCA1 and BRCA2 germline mutation carriers and in non-carrier controls, and in sporadic DCIS. We found that in the normal breast of non-carriers, myoepithelial cells frequently co-express the p63 and TCF7 transcription factors and that p63 and TCF7 show overlapping chromatin peaks associated with differentiated myoepithelium-specific genes. In contrast, in normal breast tissues of BRCA1 mutation carriers the frequency of p63+TCF7+ myoepithelial cells is significantly decreased and p63 and TCF7 chromatin peaks do not overlap. These myoepithelial perturbations in normal breast tissues of BRCA1 germline mutation carriers may play a role in their higher risk of breast cancer. The fraction of p63+TCF7+ myoepithelial cells is also significantly decreased in DCIS, which may be associated with invasive progression.

In recent years, hepatitis B virus (HBV) core protein allosteric modulators (CpAMs) have become a hot spot to develop new anti‐HBV drugs. Sulfonamide analogues are a new class of CpAMs targeting HBV core protein (Cp), while their biological activity is still needed to be improved and the binding mechanism is unclear still. In this study, we utilized molecular docking and molecular dynamics simulation to explore the binding mode between the novel compounds and HBV Cp. Combining with binding free energy calculation and decomposition, we inferred that TyrE132, ValE124, ThrE128, ThrD109, and IleD105 were key residues during the binding process, and especially, the conformational change of TryE132 was responsible for the stable binding. Besides, reasonable CoMFA (q2 = 0.515, r2ncv = 0.994, and r2pred = 0.628) and CoMSIA (q2 = 0.602, r2ncv = 0.988, and r2pred = 0.681) models were constructed to explore the 3D structure‐activity relationship of the novel compounds. The results suggested that it was favorable to increase the biological activity of sulfonamide analogues when introducing hydrophobic groups to R1 and electronegative groups to R2. In conclusion, we explored the binding mode and structure‐activity relationship between sulfonamides and HBV Cp and provided a theoretical basis for the optimization of this series of compounds. S phase kinase-associated protein 2 (Skp2) has been shown to be required for spontaneous tumor development that occurs in the retinoblastoma protein (pRb) deficient mice. Here we have demonstrated that flavokawain A (FKA), a novel chalcone from the kava plant, selectively inhibited the growth of pRb deficient cell lines and resulted in a proteasome-dependent and ubiquitination-mediated Skp2 degradation. Degradation of Skp2 by FKA was found to be involved in a functional Cullin1, but independent of Cdh1 expression. Further studies have demonstrated that FKA docked into the ATP binding pocket of the precursor cell-expressed developmentally down-regulated 8 (NEDD8)-activating enzyme (NAE) complex, inhibited NEDD8 conjugations to both Cullin1 and Ubc12 in PC3 cells and Ubc12 NEDDylation in an in vitro assay. Finally, dietary feeding of the autochthonous transgenic adenocarcinoma of the mouse prostate (TRAMP) mice with FKA inhibited the formation of high-grade prostatic intra-epithelial neoplasia lesions (HG-PIN) and prostate adenocarcinomas, reduced the tumor burden and completely abolished distant organ metastasis. Immunohistochemistry studies revealed that dietary FKA feeding resulted in marked anti-proliferative and apoptotic effects via down-regulation of Skp2 and NEDD8 and up-regulation of p27/Kip1 in the prostate of TRAMP mice. Our findings therefore provide evidence that FKA is a promising NEDDylation inhibitor for targeting Skp2 degradation in prostate cancer prevention and treatment.

To construct a chaotic system with complex characteristics and to improve the security of image data, a five-dimensional tri-valued memristor chaotic system with high complexity is innovatively constructed. Firstly, a pressure-controlled tri-valued memristor on Liu's pseudo-four-wing chaotic system is introduced. Through analytical methods, such as Lyapunov exponential map, bifurcation map and attractor phase diagram, it is demonstrated that the new system has rich dynamical behaviors with periodic limit rings varying with the coupling parameter of the system, variable airfoil phenomenon as well as transient chaotic phenomenon of chaos-periodic depending on the system parameter and chaos-quasi-periodic depending on the memristor parameter. The system is simulated with dynamic circuits based on Simulink. Secondly, the differently structured synchronous controls of chaotic systems are realized using a nonlinear feedback control method. Finally, based on the newly constructed five-dimensional chaotic system, a variable parameter color image encryption scheme is proposed to iteratively generate varying chaotic pseudo-random sequences by varying the system parameters, which will be used for repetition-free disambiguation, additive modulo left-shift diffusion and DNA encryption for the three components of RGB of the color image after chunking. The simulation results are analyzed by histogram, information entropy, adjacent pixel correlation, etc., and the images are tested using differential attack, noise attack and geometric attack, as well as analyzing the PSNR and SSIM of the decrypted image quality. The results show that the encryption method has a certain degree of security and can be applied to medical, military and financial fields with more complex environmental requirements. Fluorescence lifetime imaging has been a powerful tool for biomedical research. Recently, fluorescence lifetime‐based multiplexing imaging has expanded imaging channels by using probes that harbor the same spectral channels and distinct excited state lifetime. While it is desirable to control the excited state lifetime of any given fluorescent probes, the rational control of fluorescence lifetimes remains a challenge. Herein, we chose boron dipyrromethene (BODIPY) as a model system and provided chemical strategies to regulate the fluorescence lifetime of its derivatives with varying spectral features. We find electronegativity of structural substituents at the 8′ and 5′ positions is important to control the lifetime for the green‐emitting and red‐emitting BODIPY scaffolds. Mechanistically, such influences are exerted via the photo‐induced electron transfer and the intramolecular charge transfer processes for the 8′ and 5′ positions of BODIPY, respectively. Based on these principles, we have generated a group of BODIPY probes that enable imaging experiments to separate multiple targets using fluorescence lifetime as a signal. In addition to BODIPY, we envision modulation of electronegativity of chemical substituents could serve as a feasible strategy to achieve rational control of fluorescence lifetime for a variety of small molecule fluorophores. The single‐functionality of traditional chemodynamic therapy (CDT) reagents usually limits the therapeutic efficacy of cancer treatment. Synergistic nanocomposites that involve cascade reaction provide a promising strategy to achieve satisfactory anticancer effects. Herein, a cuprous‐based nanocomposite (CCS@GOx@HA) is fabricated, which owns the tumor targeting ability and can undergo tumor microenvironment responsive cascade reaction to enhance the tumor therapeutic efficiency significantly. Surface modification of nanocomposite with hyaluronic acid enables the targeted delivery of the nanocomposite to cancer cells. Acid‐triggered decomposition of nanocomposite in cancer cell results in the release of Cu+, Se2− and GOx. The Cu+ improves the Fenton‐like reaction with endogenous H2O2 to generate highly toxic • OH for CDT. While GOx can not only catalyze the in situ generation of endogenous H2O2, but also accelerate the consumption of intratumoral glucose to reduce nutrient supply in tumor site. In addition, Se2− further improves the therapeutic effects of CDT by upregulating the reactive oxygen species (ROS) in tumor cells. Meanwhile, the surface modification endows the nanocomposite the good water dispersibility and biocompatibility. Moreover, in vitro and in vivo experiments demonstrate satisfactory anti‐cancer therapeutic performance by the synergistic cascade function of CCS@GOx@HA than CDT alone. [ABSTRACT FROM AUTHOR]
