Transient receptor potential vanniloid-1 (TRPV-1) mediates the cough response induced by the pepper extract capsaicin and is expressed in sensory nerves that innervate the airway wall. We determined the expression of TRPV-1 in the airways of patients with chronic persistent cough of diverse causes and with an enhanced capsaicin cough response. We obtained airway mucosal biopsies by fiberoptic bronchoscopy in 29 patients with chronic cough and 16 healthy volunteers without a cough. Immunostaining for nerve profiles with anti--protein gene product (PGP)-9.5 antibody showed no increase in nerve profiles in the airway epithelium of patients with chronic cough; however, with an anti--TRPV-1 antibody, there was a fivefold increase of TRPV-1 staining nerve profiles (p < 0.001). There was a significant correlation between capsaicin tussive response and the number of TRPV-1--positive nerves within the patients with cough. Our findings indicate that TRPV-1 receptors may contribute to an enhanced cough reflex and the cough response in chronic persistent cough of diverse causes. [ABSTRACT FROM AUTHOR]

Biosolids, sewage sludge, and composts are applied to agricultural land for nutrient recovery and soil organic matter replenishment, aligning with sustainable development goals. However, they may contain per- and polyfluoroalkyl substances (PFAS) that can enter the food chain through plant bioaccumulation and leaching into the groundwater. This study analyzed 80 PFAS compounds in sewage sludge, biosolids, commercial composts, and chemical fertilizers in Quebec, Canada, using UHPLC-HRMS (Orbitrap Q-Exactive). PFAS concentrations ranged from 18 to 59 µg/kg in commercial composts, 9.8 to 213 µg/kg in pulp and paper sludge, 15 to 705 µg/kg in sewage sludge, 12 to 1310 µg/kg in biosolids, and 14.6 µg/kg on average in biosolids ash. Dominant PFAS classes included diPAPs, sulfonamides, PFCAs, and PFSAs. High diPAPs concentrations indicated widespread use in domestic, commercial, or industrial applications. This study also observed a negligible correlation between soil organic carbon and PFAS concentration in the biowastes signifying a stronger influence due to different WWTP configurations, the quality of the wastewater inputs and other medium's properties that could affect PFAS partitioning to the biowastes. Environmental assessments showed PFAS loads of up to 30 µg/kg soil from a single application, within some regulatory limits. A decomposition Mueller matrix method is proposed for detection of miRNA and enhanced by using a surface plasmon resonance (SPR). In the proposed approach, a Mueller matrix decomposition method is employed to extract the linear birefringence (LB) and circular dichroism (CD) properties of the miRNA sample. The accuracy of the LB and CD measurements is enhanced through the use of a high-resolution antimonene-based SPR prism coupler with DNA-linked gold nanoparticles (AuNPs). The feasibility of the proposed method is demonstrated by measuring the LB orientation angle (α) and CD property (R) of two miRNA aqueous solutions (hsa-miR-125-5p and hsa-miR-21-5p) over the concentration range of 0∼1000 fM in both cases. The results show that, for both samples, α and R vary linearly with the change in the miRNA concentration. Furthermore, the values of α and R obtained for the two samples are quantifiably different, and hence the selectivity of the proposed SPR sensor is confirmed. Pesticide residues are regularly found in surface water, which could be dangerous for freshwater ecosystems and biodiversity. Pesticides may enter waters through a variety of pathways, but runoff from irrigation or precipitation has the highest quantities. Previous studies analyzing the pesticides pollution or ecological risks of pesticides focused on few regions (e.g., European and United States), whereas analysis of pesticide pollution in Southeast Asia and especially in Vietnam is limited. This study presents an investigation of banned pesticides used across the range of land use in catchments of the Ma river and its tributaries in Thanh Hoa province, Vietnam. Applying principal component analysis (PCA), we investigated the relationship between specific pesticides and land use. Besides, cluster analysis (CA), the method of aggregating monitoring locations, was applied in this study to find spatial pattern of pesticides pollution. Due to their persistence and remobilization during floods and runoff, all ten banned pesticides-eight insecticides (aldrin/dieldrin, BHC, chlordane, endrin, heptachlor, lindane, malathion, and parathion) and two herbicides (paraquat, and 2,4D)-still remain in surface water and are not presumably influenced by the fraction of land use area in the catchments. Clustering results revealed that banned pesticides still occur in some areas. Site TH08 close to Le Mon industrial zone and TH18 in Thanh Hoa city have higher concentrations of banned pesticides than other sites due to their highly toxic and long-time existence in the environment. Overall, our study provides approach to investigate pesticides in surface water for a province in Vietnam that may be used for future ecotoxicological studies to enhance risk assessment for stream ecosystems., (© 2023. Analysis of temporal patterns of high-dimensional time-series water quality data is essential for pollution management worldwide. This study has applied dynamic factor analysis (DFA) and cluster analysis (CA) to analyze time-series water quality data monitored at the five stations installed along the La Buong river in Southern Vietnam. Application of the DFA identified two types of temporal patterns, one of the run-off driven parameters (total suspended solid (TSS), turbidity, and iron) and the other of diffuse source pollution. The association of the variables like BOD 5 and COD at most stations to the run-off-driven parameters revealed their sharing of drivers. On the contrary, separating variables like phosphate (PO 4 3 ) at the three upstream stations from the run-off patterns suggested their local point-source origin. The DFA-derived factors were later used in the time-point CA to explore the seasonality of water quality parameters and their pollution intensities compared to regulatory levels. The result suggested intensification in wet season of Fe, TSS, BOD 5 , and COD concentrations at most sites, which are unobservable in run-off detached parameters like reactive nitrogen, phosphate (PO 4 3- ), and E. coli. Interleukin 23 and the interleukin 23 receptor (IL-23-IL23R) are described as the major enhancing factors for Interleukin 17 (IL-17) in allergic airway inflammation. IL-17 is considered to induce neutrophilic inflammation in the lung, which is often observed in severe, steroid-resistant asthma-phenotypes. For that reason, understanding of IL-23 and IL-17 axis is very important for future therapy strategies, targeting neutrophil pathway of bronchial asthma.This study aimed to investigate the distribution and expression of IL-23R under physiological and inflammatory conditions. Therefore, a house dust mite (HDM) model of allergic airway inflammation was performed by treating mice with HDM intranasally. Immunofluorescence staining with panel of antibodies was performed in lung tissues to examine the macrophage, dendritic cell, and T cell subpopulations. The allergic airway inflammation was quantified by histopathological analysis, ELISA measurements, and airway function.HDM-treated mice exhibited a significant allergic airway inflammation including higher amounts of NE + cells in lung parenchyma. We found only a small amount of IL-23R positives, out of total CD3 + T cells, and no upregulation in HDM-treated animals. In contrast, the populations of F4/80 + macrophages and CD11c + F4/80 - dendritic cells (DCs) with IL-23R expression were found to be higher. But HDM treatment leads to a significant increase of IL-23R + macrophages, only. IL-23R was expressed by every examined macrophage subpopulation, whereas only Mϕ1 and hybrids between Mϕ1 and Mϕ2 phenotype and not Mϕ2 were found to upregulate IL-23R. Co-localization of IL-23R and IL-17 was only observed in F4/80 + macrophages, suggesting F4/80 + macrophages express IL-23R along with IL-17 in lung tissue.The study revealed that macrophages involving the IL-23 and IL-17 pathway may provide a potential interesting therapeutic target in neutrophilic bronchial asthma., (© 2022. The proliferation of harmful cyanobacterial algal blooms is of concern due to the associated release of toxins affecting ecosystems and human health. The paralytic shellfish poison saxitoxin (STX) is a small polar alkaloid that can occur in inland and marine aquatic environments. Here, we optimized a fast and sensitive analytical method for the determination of STX, neosaxitoxin (NeoSTX), and their decarbamoyl analogues in surface waters. The method involves a simple filtration, addition of isotope-labelled internal standard (ILIS), and analysis by on-line solid-phase extraction coupled to hydrophilic interaction liquid chromatography high-resolution mass spectrometry (on-line SPE-HILIC-HRMS). Except glass fiber filters, other tested materials (e.g., nylon, nitrocellulose) provided suitable filtration performance. Time-dependent adsorptive losses occurred during the LC-MS batch sequence if glass autosampler vials were used, while no such effect was observed for polypropylene autosampler vials. Matrix effects were evaluated for 4 different quantification scenarios, including external vs. internal curves and neat reagent water vs. matrix-matched curves. Matrix-matched calibration with ILIS correction (NeoSTX- 15 N 7 ) provided the best performance overall. The analytical method was validated in freshwater lake water and estuarine brackish water (30‰ salinity), with suitable determination coefficients (R 2  > 0.9975), matrix spike accuracy (90-107%), and intraday/interday precision (RSD of 0.61-16%). Method limits of detection (LOD in lake water: 0.72-3.9 ng/L) are also improved over most of the recent literature. Extracorporeal membrane oxygenation (ECMO) has become an important therapeutic approach in the COVID-19 pandemic. The development and research in this field strongly relies on animal models; however, efforts are being made to find alternatives. In this work, we present a new mock circuit for ECMO that allows measurements of the oxygen transfer rate of a membrane lung at full ECMO blood flow. The mock utilizes a large reservoir of heparinized porcine blood to measure the oxygen transfer rate of the membrane lung in a single passage. The oxygen transfer rate is calculated from blood flow, hemoglobin value, venous saturation, and post-membrane arterial oxygen pressure. Before the next measuring sequence, the blood is regenerated to a venous condition with a sweep gas of nitrogen and carbon dioxide. The presented mock was applied to investigate the effect of a recirculation loop on the oxygen transfer rate of an ECMO setup. The recirculation loop caused a significant increase in post-membrane arterial oxygen pressure (paO 2 ). The effect was strongest for the highest recirculation flow. This was attributed to a smaller boundary layer on gas fibers due to the increased blood velocity. However, the increase in paO 2 did not translate to significant increases in the oxygen transfer rate because of the minor significance of physically dissolved oxygen for gas transfer. In conclusion, our results regarding a new ECMO mock setup demonstrate that recirculation loops can improve ECMO performance, but not enough to be clinically relevant.

The neurotoxic alkaloid β-N-methyl-amino-l-alanine (BMAA) and related isomers, including N-(2-aminoethyl glycine) (AEG), β-amino-N-methyl alanine (BAMA), and 2,4-diaminobutyric acid (DAB), have been reported previously in cyanobacterial samples. However, there are conflicting reports regarding their occurrence in surface waters. In this study, we evaluated the impact of amending lake water samples with trichloroacetic acid (0.1 M TCA) on the detection of BMAA isomers, compared with pre-existing protocols. A sensitive instrumental method was enlisted for the survey, with limits of detection in the range of 5−10 ng L−1. Higher detection rates and significantly greater levels (paired Wilcoxon’s signed-rank tests, p < 0.001) of BMAA isomers were observed in TCA-amended samples (method B) compared to samples without TCA (method A). The overall range of B/A ratios was 0.67−8.25 for AEG (up to +725%) and 0.69−15.5 for DAB (up to +1450%), with absolute concentration increases in TCA-amended samples of up to +15,000 ng L−1 for AEG and +650 ng L−1 for DAB. We also documented the trends in the occurrence of BMAA isomers for a large breadth of field-collected lakes from Brazil, Canada, France, Mexico, and the United Kingdom. Data gathered during this overarching campaign (overall, n = 390 within 45 lake sampling sites) indicated frequent detections of AEG and DAB isomers, with detection rates of 30% and 43% and maximum levels of 19,000 ng L−1 and 1100 ng L−1, respectively. In contrast, BAMA was found in less than 8% of the water samples, and BMAA was not found in any sample. These results support the analyses of free-living cyanobacteria, wherein BMAA was often reported at concentrations of 2−4 orders of magnitude lower than AEG and DAB. Seasonal measurements conducted at two bloom-impacted lakes indicated limited correlations of BMAA isomers with total microcystins or chlorophyll-a, which deserves further investigation.

Cadmium (Cd) has strong mobility and could cause toxicity to plants, and selenium (Se) can effectively detoxify Cd stress. However, differences in the detoxification effects of different species and dosages of exogenous Se on Cd and its mechanism are still unclear. In this study, a pot experiment was conducted to determine the effects of different rates of selenite and selenate application on radish growth, the uptake and translocation of Cd, and the fractions of Cd transformation in native Cd-contaminated soil. Results indicated that the decrease in radish biomass in selenate treatment was significantly greater than that in selenite treatment at a high Se application rate (2.5 mg·kg -1 ) (p < 0.05). In contrast to selenite treatments, the application of selenate significantly increased the translocation of Cd from radish roots to shoots (p < 0.05). Cadmium concentration and its bioaccumulation factor in radish decreased gradually with increasing selenite application rates, while these values decreased at low Se rate (1 mg·kg -1 ) and increased at high Se rate for selenate treatment. Different Se application rates resulted in Cd fractions distributions to change in soil. Therefore, the application of selenite treatment had a greater detoxification effect on Cd in soil than that in selenate treatment, and the double toxic effect was observed between Se and Cd in high selenate treatment (2.5 mg·kg -1 ). Combined with human health risk asseeement, the application of 2.5 mg·kg -1 selenite could be a good approach for detoxification in native Cd-contaminated soil used in this study., (© 2021. Feasible countermeasures to mitigate mercury (Hg) accumulation and its deleterious effects on crops are urgently needed worldwide. Selenium (Se) fertilizer application is a cost-effective strategy to reduce Hg concentrations, promote agro-environmental sustainability and food safety, and decrease the public health risk posed by Hg-contaminated soils and its accumulation in food crops. This holistic review focuses on the processes and detoxification mechanisms of Hg in whole soil-plant systems after Se application. The reduction of Hg bioavailability in soil, the formation of inert HgSe or/and HgSe-containing proteinaceous complexes in the rhizosphere and/or roots, and the reduction of plant root uptake and translocation of Hg in plant after Se application are systemically discussed. In addition, the positive responses in plant physiological and biochemical processes to Se application under Hg stress are presented to show the possible mechanisms for protecting the plant. However, application of high levels Se showed synergistic toxic effect with Hg and inhibited plant growth. The effectiveness of Se application methods, rates, and species on Hg detoxification is compared. Cyanotoxins, as secondary metabolites of cyanobacteria, are highly toxic to humans, animals and plants. Cyanobacterial blooms are 'hot spots' for cyanotoxin production, but we hypothesized that cyanotoxins will be present in multiple ecological compartments of agricultural watersheds. We detected cyanotoxins in the vadose zone (soil and drainage water from the soil profile) and in groundwater used as a drinking water source from agricultural watersheds. Cyanotoxins detection was confirmed with enzyme-linked immunosorbent assay kits and ultra-high liquid chromatography with tandem mass spectrometry. This work confirms that cyanotoxins exist outside of freshwater lakes, based on detection of microcystins in the vadose zone and in drinking water sourced from groundwater in agricultural watersheds. This suggests that cyanotoxins may be transferred from cyanobacterial blooms in lakes to groundwater through normal hydrologic processes. Extracorporeal carbon dioxide removal (ECCO 2 R) is an important technique to treat critical lung diseases such as exacerbated chronic obstructive pulmonary disease (COPD) and mild or moderate acute respiratory distress syndrome (ARDS). This study applies our previously presented ECCO 2 R mock circuit to compare the CO 2 removal capacity of circular versus parallel-plated membrane lungs at different sweep gas flow rates (0.5, 2, 4, 6 L/min) and blood flow rates (0.3 L/min, 0.9 L/min). For both designs, two low-flow polypropylene membrane lungs (Medos Hilte 1000, Quadrox-i Neonatal) and two mid-flow polymethylpentene membrane lungs (Novalung Minilung, Quadrox-iD Pediatric) were compared. While the parallel-plated Quadrox-iD Pediatric achieved the overall highest CO 2 removal rates under medium and high sweep gas flow rates, the two circular membrane lungs performed relatively better at the lowest gas flow rate of 0.5 L/min. The low-flow Hilite 1000, although overall better than the Quadrox i-Neonatal, had the most significant advantage at a gas flow of 0.5 L/min. Moreover, the circular Minilung, despite being significantly less efficient than the Quadrox-iD Pediatric at medium and high sweep gas flow rates, did not show a significantly worse CO 2 removal rate at a gas flow of 0.5 L/min but rather a slight advantage. We suggest that circular membrane lungs have an advantage at low sweep gas flow rates due to reduced shunting as a result of their fiber orientation. Efficiency for such low gas flow scenarios might be relevant for possible future portable ECCO 2 R devices.

Harmful algal blooms of cyanobacteria (CyanoHABs) can lead to the release of potent toxins that can seriously affect ecosystem integrity. Some freshwater watersheds are particularly at risk considering the threats to already imperiled wildlife. The consumption of tainted drinking water and contaminated food also raises concerns for human health. In the present study, a pilot survey was conducted in the riverine ecosystem of the Pike River Ecological Reserve (QC, Canada) near Missisquoi Bay, Lake Champlain. We examined the occurrence of multiclass cyanotoxins including 12 microcystins, anatoxins, cylindrospermopsin (CYN), anabaenopeptins (AP-A, AP-B), and cyanopeptolin-A in surface waters and wild-caught fish during the summer 2018. Out of the 18 targeted cyanotoxins, 14 were detected in bloom-impacted surface water samples; toxins peaked during early-mid September with the highest concentrations for MC-LR (3.8 μg L -1 ) and MC-RR (2.9 μg L -1 ). Among the 71 field-collected fish from 10 species, 30% had positive detections to at least one cyanotoxin. In positive samples, concentration ranges in fish muscle were as follows for summed microcystins (∑MCs: 0.16-9.2 μg kg -1 ), CYN (46-75 μg kg -1 ), AP-A (1.1-5.4 μg kg -1 ), and AP-B (0.12-5.0 μg kg -1 ). To the best of our knowledge, this is one the first reports of anabaenopeptins occurrence in wildlife. The maximum ∑MCs in fish was 1.15-fold higher than the World Health Organization (WHO) daily intake recommendation for adults and nearly equated the derived value for young children. Knowledge of the Se fractionation and the role of dissolved organic matter (DOM) in soil is the key to understanding Se mobility and its bioavailability in the soil-plant system. In this study, single extractions using phosphate-buffer (PBS), sequential extraction procedures (SEP), and diffusive gradients in thin-films (DGT) were used to measure Se bioavailability in soil supplemented with selenite and organic amendment (cow and chicken manures). Selenium fraction was isolated into DOM-Se fractions, such as hydrophilic acid-bound Se (HY-Se), fulvic acid-bound Se (FA-Se), humic acid-bound Se (HA-Se), and hydrophobic organic neutral-bound Se (HON-Se), by a rapid batch technique using XAD-8 resin (AMBERLITE XAD™, USA). Simultaneous application of either cow or chicken manure with selenite could result in the decrease of Se availability in the soil. Isolating Se available fraction into DOM-Se fractions showed that low-molecular-weight DOM-Se as an available fraction and even HY-Se as a less available fraction (OM-Se) were likely the major sources for Brassica juncea (L.) Czern. et Coss uptake in soil. Moreover, knowledge of the DOM-Se composition, especially the low-molecular-weight DOM-Se fractions, is important for assessing the bioavailability of Se in soil, the results of which are more accurate than the chemical extraction method. The high value of Pearson correlation coefficients between CDGT-Se and Se concentrations in shoots, tubers and roots of Brassica juncea (L.) Understanding the effects of processing on the Se content and bioaccessibility in food is critical in guiding the development of Se-enriched products. In this study, Se-enriched Pleurotus eryngii was obtained by applying different Se supplements to the substrate. Selenium content and its bioaccessibility among raw and processed fruit bodies were compared. The application of exogenous Se had no significant effect on the yield of P. eryngii, while amendment Se yeast could slightly promote the growth of P. eryngii. The enrichment ability of P. eryngii among different Se supplements declined in the order of Na 2 SeO 3  > Se yeast > Na 2 SeO 4 . However, the processing treatments resulted in 6.6%-45.9% Se loss. The Se bioaccessibility of P. eryngii was 78.4%-89.7%. Frying treatment reduced Se bioaccessibility in samples, whereas boiling treatment enhanced it. Reproducible analytical procedures and rigorous quality control are imperative for an accurate monitoring of cyanobacterial toxins in environmental water samples. In this study, the short-term and long-term storage stability of diverse cyanotoxins (anatoxins, cylindrospermopsin, anabaenopeptins, and 12 microcystins) was evaluated in water samples, under different scenarios. Transport controls were performed at three monitoring sites in spiked ultrapure water and lake water to investigate short-term stability issues. Medium-term storage stability was evaluated for up to 14-28 days in ultrapure water, chlorine-treated drinking water (amended with reductant), and surface water (filtered and unfiltered) stored at different temperatures (20 °C, 4 °C, and -20 °C). Substantial decreases of cylindrospermopsin and anabaenopeptins were observed in tap water (20 °C) and unfiltered surface water (20 °C or 4 °C). Regardless of matrix type, cyanotoxin recoveries generally remained within an 80-120% range when the water samples were kept frozen. After a prolonged storage duration of 365 days at -20 °C, most cyanotoxins experienced decreases in the range of 10-20%. The notable exception was for the tryptophan-containing MC-LW and MC-WR, with more substantial variations (30% to 50% decrease) and conversion to N-formylkynurenine analogs. Reanalysis of field-collected surface waters after long-term storage at -20 °C also indicated significantly decreasing trends of cyanotoxins (between 6% and 23% decrease). Selenium (Se)-enriched wheat can be improved by altering Se sources and selecting wheat cultivars. Such improvement can affect subcellular distribution and speciation of Se in wheat. Thus, a pot experiment was conducted to investigate Se uptake and distribution when Se was applied as selenite or selenate at low and high rates (1 and 10 mg kg -1 , respectively). Moreover, Se's impact on the grain and biomass yield of eight wheat cultivars was also investigated. The subcellular distribution and speciation of Se were also explored to elucidate Se metabolism and micro-distribution pattern in wheat. Results showed that biomass and grain yield were decreased with the application of both selenite and selenate in almost all the cultivars, regardless of the Se rate. Application high Se rate resulted in a significant (p < 0.05) decrease in grain yield and biomass compared with low rate of Se. Compared with the low rate of selenite application, the grain and the biomass yield of ZM-9023 significantly (p < 0.05) increased by about 15% for low rate of selenate application. In addition, both selenite and selenate treatment increased the uptake of Se in each part of wheat, compared with the control. Selenium was mostly accumulated in the grain and root of wheat under selenite treatment, while more Se accumulation was found in leaves and straw for selenate application. Further investigation on the subcellular distribution of Se showed that the proportion of Se in soluble fraction was significantly (p < 0.05) higher in wheat leaves than that in organelle fraction and cell walls (46%-66%). Meanwhile, Se 6+ was the main species found in soluble fraction, whereas SeMet and MeSeCys were the species predominantly stored in organelle fraction. In conclusion, wheat cultivar ZM-9023 is the most Se-rich potential cultivar, and the isolation of Se in the soluble fraction plays an important role in Se tolerance and accumulation., (Copyright © 2020 The Authors. Background: Extracorporeal carbon dioxide removal (ECCO 2 R) is a promising yet limited researched therapy for hypercapnic respiratory failure in acute respiratory distress syndrome and exacerbated chronic obstructive pulmonary disease. Herein, we describe a new mock circuit that enables experimental ECCO 2 R research without animal models. In a second step, we use this model to investigate three experimental scenarios of ECCO 2 R: (I) the influence of hemoglobin concentration on CO 2 removal. (II) a potentially portable ECCO 2 R that uses air instead of oxygen, (III) a low-flow ECCO 2 R that achieves effective CO 2 clearance by recirculation and acidification of the limited blood volume of a small dual lumen cannula (such as a dialysis catheter)., Results: With the presented ECCO 2 R mock, CO 2 removal rates comparable to previous studies were obtained. The mock works with either fresh porcine blood or diluted expired human packed red blood cells. However, fresh porcine blood was preferred because of better handling and availability. In the second step of this work, hemoglobin concentration was identified as an important factor for CO 2 removal. In the second scenario, an air-driven ECCO 2 R setup showed only a slightly lower CO 2 wash-out than the same setup with pure oxygen as sweep gas. In the last scenario, the low-flow ECCO 2 R, the blood flow at the test membrane lung was successfully raised with a recirculation channel without the need to increase cannula flow. Low recirculation ratios resulted in increased efficiency, while high recirculation ratios caused slightly reduced CO 2 removal rates. Acidification of the CO 2 depleted blood in the recirculation channel caused an increase in CO 2 removal rate., Conclusions: We demonstrate a simple and cost effective, yet powerful, "in-vitro" ECCO 2 R model that can be used as an alternative to animal experiments for many research scenarios. Moreover, in our approach parameters such as hemoglobin level can be modified more easily than in animal models.

The present 2019 S2k consensus guideline of the German Respiratory Society was written for pneumologists - in contrast to the more general predecessor's S3 guidelines from 2004 to 2010 -, since 2014 the German College of General Practitioners and Family Physicians (DEGAM) published their own cough guidelines. The guidelines contain 48 recommendations agreed upon the consensus and 16 statements, which are explained in the background text in the following nine chapters: epidemiology, physiology, classification, acute, subacute or chronic cough, diagnostics and therapy; an extra chapter was dedicated to chronic idiopathic/refractory cough. Further emphasis of the guidelines is the physiology of cough in anticipation of the introduction of new drugs, as well as detailed treatment for cough triggered by affectations of the upper respiratory tract or gastroesophageal reflux. The guideline should provide the pneumologist with the latest knowledge for neighboring disciplines required for diagnosis and therapy of cough. The clinical chapters also contain a short summary, practical recommendations and a bibliography of their own. Background: Titanium dioxide nanoparticles (TiO 2 NPs) have a wide range of applications in several industrial and biomedical domains. Based on the evidence, the workers exposed to inhaled nanosized TiO 2 powder are more susceptible to the risks of developing respiratory diseases. Accordingly, this issue has increasingly attracted the researchers' interest in understanding the consequences of TiO 2 NPs exposure. Regarding this, the present study was conducted to analyze the local effects of TiO 2 NPs on allergic airway inflammation and their uptake in a mouse model of ovalbumin (OVA)-induced allergic airway inflammation., Methods: For the purpose of the study, female BALB/c mice with or without asthma were intranasally administered with TiO 2 NPs. The mice were subjected to histological assessment, lung function testing, scanning electron microscopy (SEM), inductively coupled plasma mass spectrometry (ICP-MS), and NP uptake measurement. In addition, T helper (Th) 1/Th2 cytokines were evaluated in the lung homogenate using the enzyme-linked immunosorbent assay., Results: According to the results, the mice receiving OVA alone or OVA plus TiO 2 NPs showed eosinophilic infiltrates and mucus overproduction in the lung tissues, compared to the controls. Furthermore, a significant elevation was observed in the circulating Th2 cytokines, including interleukin (IL)-4, IL-5, and IL-13 after NP exposure. The TiO 2 NPs were taken up by alveolar macrophages at different time points. As the results of the SEM and ICP-MS indicated, TiO 2 NPs were present in most of the organs in both asthmatic and non-asthmatic mice., Conclusion: Based on the findings of the current study, intranasally or inhalation exposure to high-dose nanosized TiO 2 particles appears to exacerbate the allergic airway inflammation and lead to systemic uptake in extrapulmonary organs. These results indicate the very important need to investigate the upper limit of intranasally or inhalation exposure to nanosized TiO 2 particles in occupational and environmental health policy.
