Abstract

The transferability and clinical value of genetic risk scores (GRSs) across populations remain limited due to an imbalance in genetic studies across ancestrally diverse populations. Here we conducted a multi-ancestry genome-wide association study of 156,319 prostate cancer cases and 788,443 controls of European, African, Asian and Hispanic men, reflecting a 57% increase in the number of non-European cases over previous prostate cancer genome-wide association studies. We identified 187 novel risk variants for prostate cancer, increasing the total number of risk variants to 451. An externally replicated multi-ancestry GRS was associated with risk that ranged from 1.8 (per standard deviation) in African ancestry men to 2.2 in European ancestry men. The GRS was associated with a greater risk of aggressive versus non-aggressive disease in men of African ancestry (P = 0.03). Our study presents novel prostate cancer susceptibility loci and a GRS with effective risk stratification across ancestry groups.

Abstract

Cet article est largement inspiré de la contribution des auteurs à la table ronde sur la migration des pêcheurs (FAO-DIPA) kokrobité, Ghana 6-9 décembre 1990 Résumé : Pêche de subsistance depuis le 15ème siècle, c’est seulement aux indépendances que l’on note des peuplements littoraux qui s’adonnent à la pêche artisanale en Mauritanie dans le cadre d’une économie de marché. Grâce aux flux migratoires on note une présence de pêcheurs étrangers qui contribuent au développement de ce secteur en introduisant des nouvelles techniques (engins, embarcations) et organisations. Ces pêcheurs (mauritaniens et étrangers), en majorité Wolofs, composent l’essentiel des effectifs présents sur la façade maritime mauritanienne. Résumé : Au mois de mars 1989, 770 embarcations employant 3537 pêcheurs ont été recensées, tandis qu’au mois d’août de la même année seules 540 embarcations ont été recensées. Ces dernières emploient 2236 pêcheurs, soit une diminution du parc d’embarcation de 30% et de l’effectif des pêcheurs de 38%. Les « Imraguen » commencent à diversifier leur technique de pêche en utilisant de nouveaux engins. La pêche artisanale intéresse de plus en plus les mauritaniens qui n’ont pas une tradition de pêche. En effet, des pêcheurs originaires des régions de l’intérieur de la Mauritanie représentaient 10% de l’effectif en février 1989 et 34% en août 1989. # Abstract: In March of 1989, 770 canoes comprising 3537 fishermen were counted in Mauritanian waters. In August of the same year, only 540 canoes comprising 2236 fishermen were observed. The number of canoes and fishermen had reduced by 30% and 38% respectively. The “Imraguen” have begun diversifying their fishing technics by using new gear. The small scale fisheries has become more interesting to those Mauritanians who weren’t traditionally fishermen. 41. Tenofovir-Emtricitabine-Efavirenz in HIV-1-Infected Adults in Senegal: A 96-Week Pilot Trial in Treatment-Naive Patients.

Abstract

We report the results of a pilot open-label trial of a tenofovir (TDF)/emtricitabine (FTC)/efavirenz (EFV) combination conducted in Dakar, Senegal. Forty HIV-1-infected patients, naive of antiretroviral treatment and without active opportunistic disease, were included and followed through 96 weeks. At weeks 48 and 96, respectively, 82.5% and 85% of patients had HIV-1 RNA <400 copies/mL (72.5% and 77.5% with HIV-1 RNA <50 copies/mL). Between baseline and week 96, the mean (SD) CD4 count increased from 126 (102) to 338 (155) cells/mm³. The mean (SD) creatinine clearance decreased from 92 (36) to 73 (19) mL/min (P = .001). Treatment adherence was at least 94% at all scheduled visits. The efficacy and tolerability of a TDF/FTC/EFV combination were high and similar to those observed in Northern countries. This drug combination can be recommended in limited-resource countries, as did the World Health Organization (WHO) and should be made readily available as a fixed-dose combination. 42. Surveillance of births conceived with various infertility therapies in Massachusetts, January-March 2005.

Abstract

OBJECTIVE: Nationally, infertility therapies (IFTs) are increasingly used to overcome fecundity issues. However, it is unclear to what extent noninvasive IFTs are used compared with assisted reproductive technology. To better understand outcomes related to the increasing use of all types of IFTs, we compared self-reported IFT use from a Massachusetts pilot Pregnancy Risk Assessment Monitoring System (MA-PRAMS) to IFT use recorded on birth certificates (BCs). METHODS: In 2005, Massachusetts conducted a three-month pilot study modeled after the Centers for Disease Control and Prevention's PRAMS, a population-based surveillance system that monitors pregnancy experiences. Descriptive and bivariate analyses compared responses to MA-PRAMS survey questions regarding IFT use with data collected on BCs from the same women sampled. RESULTS: According to MA-PRAMS, 6.1% of live births were conceived using IFTs compared with 3.1% reported on BCs. Reported IFT use varied by maternal age and plurality. For women aged 18-34 years, IFT use reported on MA-PRAMS (5.0%) was 2.5 times higher than that reported on BCs (2.0%). For women aged 35 years or older, reported IFT use was comparable in both systems. For women giving birth to singletons, IFT use reported on MA-PRAMS (5.5%) was three times higher than that reported on BCs (1.8%). CONCLUSIONS: Higher use of IFTs was reported by MA-PRAMS than on BCs, particularly among younger women and those having singleton births. These findings suggest that self-reported IFT use might be a more sensitive method for states to use in assessing population-based IFT usage among women and monitoring trends in adverse birth outcomes. Les informations recueillies portent sur les pirogues en état de prendre la mer, les migrations de pêcheurs, l’emploi et l’activité au cours des trois derniers mois, le capital technique, les équipages et l’organisation socio-économique des unités de pêche. La pêche artisanale mauritanienne compte actuellement 735 embarcations réparties sur 22 points de débarquement. Elle génère 3302 emplois directs. Ce document expose la méthodologie d’enquête et les principaux résultats obtenus. Abstract Two complete census of the artisanal canoe fishery are carried up yearly by the « Centre National de Recherches Océanographiques et des Pêches de Nouadhibou (CNROP) ». In July 1987 an economic survey has also been carried up. All canoes are counted; complete statistics are also collected upon fishermen migration and employment in the fishery and fishing activity three months before survey, capital, crews and economic organization of fishing units. 735 canoes are distributed among 22 landing centers. 3302 fishermen are working in the canoe fishery.
