HCLS1-associated protein X-1 (HAX1) is a newly discovered multifunctional cell regulatory protein that is widely expressed in cells and has a close relationship with multiple cellular proteins. HAX1 plays important roles in various processes, including the regulation of apoptosis, maintenance of mitochondrial membrane potential stability and calcium homeostasis, occurrence and development of diseases, post-transcriptional regulation of gene expression, and host immune response after viral infection. In this article, we have reviewed the research progress on the biological functions of HAX1, thereby laying a theoretical foundation for further exploration of its underlying mechanisms and targeted application. It is critical to quantify latent Plasmopara viticola infections in commercial vineyards rapidly and accurately to estimate the potential for grapevine downy mildew (GDM) epidemics. In this study, a well‐established quantitative PCR (qPCR) assay was employed to quantify the amount of P. viticola in asymptomatic grapevine leaves from various commercial vineyards in the Ningxia Hui Autonomous Region (Ningxia), China. The qPCR revealed efficient detection of P. viticola, with 56.6% and 95.8% of the asymptomatic leaves testing positive to the pathogen in 2018 and 2019, respectively. The molecular disease index (MDI), which offers insights into the progression of latent infection preceding the appearance of visible GDM symptoms, showed relatively modest values in the initial phases. However, it increased variably as grapevines grew. The disease index progress curve and the relative area under the disease progress curve revealed that all assessed vineyards experienced different degrees of epidemics after the initial GDM observation, with most vineyards experiencing more severe GDM epidemics in 2019 than in 2018. Bayesian and non‐linear regression analyses demonstrated that MDI data matched well with occurrence of GDM at early, mid and late stages of disease, and the overall accuracy of predictions exceeded 50%, 64% and 70%, respectively. Furthermore, latent infection levels were positively correlated with subsequent assessments of disease progression (p < 0.001). This study provides evidence for the interrelation between latent P. viticola infections and development of GDM epidemics under field conditions. Parkinson's disease is easy to diagnose when it is advanced, but it is very difficult to diagnose in its early stages. Early diagnosis is essential to be able to treat the symptoms. It impacts on daily activities and reduces the quality of life of both the patients and their families and it is also the second most prevalent neurodegenerative disorder after Alzheimer in people over the age of 60. Most current studies on the prediction of Parkinson's severity are carried out in advanced stages of the disease. In this work, the study analyzes a set of variables that can be easily extracted from voice analysis, making it a very non-intrusive technique. In this paper, a method based on different deep learning techniques is proposed with two purposes. On the one hand, to find out if a person has severe or non-severe Parkinson's disease, and on the other hand, to determine by means of regression techniques the degree of evolution of the disease in a given patient. The UPDRS (Unified Parkinson's Disease Rating Scale) has been used by taking into account both the motor and total labels, and the best results have been obtained using a mixed multi-layer perceptron (MLP) that classifies and regresses at the same time and the most important features of the data obtained are taken as input, using an autoencoder. A success rate of 99.15% has been achieved in the problem of predicting whether a person suffers from severe Parkinson's disease or non-severe Parkinson's disease. In the degree of disease involvement prediction problem case, a MSE (Mean Squared Error) of 0.15 has been obtained. Using a full deep learning pipeline for data preprocessing and classification has proven to be very promising in the field Parkinson's outperforming the state-of-the-art proposals. Abstract Background Bone erosions may appear early or later during rheumatoid arthritis (RA), causing joint damage and functional impairment. However, in some patients erosions do not occur, even after several years of disease. This study evaluates the prevalence, clinical relevance and possible predictors of erosion-free RA. Methods Six hundred and eight patients from an early RA cohort (BARFOT) having radiographs of hands and feet at inclusion and after 1, 2, 5 and 8 years were studied. Clinical and functional assessments were performed on all these time-points. Results In all, 144 patients (24%) did not develop erosions up to 8 years follow-up (Never erosive group), while 464 patients (76%) had erosions on one or more assessments (Ever erosive group). At diagnosis, the patients in the Never erosive group were significantly younger, satisfied fewer ACR criteria, and were less frequently RF- and/or anti-CCP- positive compared with those in the Ever erosive group. The Never erosive patients had consistently more tender joints, lower erythrocyte sedimentation rate (ESR) and, from two years and onwards, fewer swollen joints. Absence of rheumatoid factor (RF) and/or anti-CCP were strong independent predictors for erosion-free disease. The erosion-free patients were less frequently treated with DMARDs and/or prednisolone. Conclusions One-quarter of the patients was erosion-free during eight years in this early RA cohort. Erosion-free patients had a less severe disease course as to disease activity and were more often seronegative compared with those with erosive disease. The results suggest that non-erosive RA represents a milder form of RA.

The dynamics of airborne ascospores and disease progress of red leaf blotch (RLB) of almond, caused by Polystigma amygdalinum, and their correlations with weather variables were studied from 2019 to 2021 in two almond orchards located in Lleida, NE Spain. Airborne ascospores were detected and quantified by real‐time qPCR using species‐specific primers for P. amygdalinum. Ascospores were detected mainly from April to June, with a high variability between the yearly cumulative concentrations. Positive significant correlations were found between the weekly proportion of airborne ascospores and the number of wet and mild days—either combined or separated— accumulated rainfall, number of rainy days, accumulated low temperatures on wet days, and mean and maximum relative humidity. In contrast, several thermal variables (maximum temperature, VPD, and number of warm days) were negatively correlated with ascospore catches. Positive significant correlations were found between the cumulative proportion of ascospores and RLB incidence and severity. Weekly variations in RLB incidence and severity showed significant positive correlations with the number of warm days while negative with the number of mild days. Severity was also positively correlated with several thermal variables (mean, maximum, and minimum temperature, and VPD), and negatively correlated with the number of cold days and wet and mild days. Stronger correlations were generally found with ascospore catches or disease progress when using concurrent weekly weather data. Objective：To investigate the risk factors for progression of cervical intraepithelial neoplasia Ⅰ (CIN Ⅰ ) patients, and to provide more appropriate treatment plans for the CIN Ⅰ patients. Methods：The clinical data of 210 patients with CINⅠ confirmed by initial cervical pathological biopsy and conservative treatment were retrospectively analyzed. All the patients were followed up for 24 months to observe the disease progression. Univariate analysis and multivariate Logistic stepwise regression anslysis were used to analyze the influence of patients’age, human papillomavirus (HPV), liquid based thin layer cytology (TCT), menopausal status, and transformation zone (TZ) types and image features under colposcope (abnormal vessels, hyperplasia, lesion area, and lesion boundary) on disease progression of the CINⅠ patient at the time of initial diagnosis of CINI disease. Results：A total of 40 patients had progressive disease, 170 patients had no progressive disease. The univariate analysis results showed that there were statistically significant differences in age (χ² =25. 94, P<0. 01), TCT stuation (χ² =10. 97, P<0. 01), HPV (16/18) infection or not (χ² =4. 10, P=0. 043), hyperplasia (χ² =8. 18, P=0. 004) and menopause status (χ² =25. 56, P<0. 01) of the pateints between two groups. The results demonstrated that a logistic model was the most appropriate describe the temporal progress of NCLB incidence. The initial disease incidence was the key factor affecting disease epidemics under various conditions in the four districts of Beijing during the study. The higher the initial incidence of NCLB, the higher the final incidence. Thus, the earlier in the season NCLB incidence attained 1 %. the higher was the final disease incidence. Greater than 1.0 variance-to-mean ratios suggested that the leaf lesions of NCLB tended to be aggregated on a plant. According to results from join-counts, variance of moving window averages, and semivariogram analysis. diseased corn plants and lesion numbers on each plant were aggregated in the field. The fast adaptation to different growing conditions of a fungus Macrophomina phaseolina, led to its becoming one of the sunflower (Helianthus annuus L.) disease causal agents in regions with a temperate climate. Methods currently used to determine sunflower resistance require laborious manual inoculation and confirmation of pathogen appearance, due to the late stage of testing. The paper proposes a cut-stem method for inoculating sunflower plants in the controlled conditions and the possibility of early-stage disease evaluation. A set of 15 sunflower inbred lines was inoculated using M. phaselolina isolate in the growth chamber and the obtained data were analysed using Cut-stem Disease Severity (CSDS) and compared with disease severity obtained from field experiments using traditional inoculation methods (toothpick, Unwounded Stem Base Inoculation (USBI) and non-inoculated plants). The results showed that, based on CSDS, inbred lines infected with the cut-stem inoculation method significantly differed regarding resistance to M. phaseolina. None of the inbred lines exhibited complete resistance but three lines could be proposed as a source of resistance to this pathogen. Ranking of inbred lines which was based on resistance to M.phaseolina was similar in all inoculation methods and in non-inoculated plants. There were highly significant correlations between the values obtained from growth chamber experiment and disease severity scores from field evaluations. Thus, the obtained results indicate that the cut-stem method could potentially complement field testing methods and be valuable tool in sunflower breeding for resistance to M. phaseolina. Interaction among nitrogen fertilization using bovine manure, poultry manure, Jatropha curcas seed cake and urea, and the diseases Ramularia leaf spot (RLS) and Boll rot (BR), caused by Ramulariopsis pseudoglycines and Diplodia gossypina, respectively, in cotton plants (Gossypium hirsutum L.), was studied under field conditions. Intensity (incidence and severity in percentage) of RLS and incidence (%) of BR were evaluated over time, starting in reproductive stage B1 (first visible flower bud). A randomized complete block design with a 4x4 factorial arrangement was used (fertilizers x doses), totaling 16 treatments with four replications. Disease progress was analyzed with the nonlinear Logistic and Gompertz models, obtaining the epidemiological parameters amount of initial disease (Y0) and progress rate (r). Plants fertilized with 50 kg N ha-1, presented an incidence twice greater than those obtained with other fertilizers. The Logistic model better fits RLS, but no model could represent BR. Only the epidemiological parameters of RLS were affected differently in this experiment compared to BR disease. The possible role of organic and inorganic nitrogen fertilization in the RLS and BR management is discussed.

Stem rust (or black rust) of wheat, caused by Puccinia graminis f. sp. tritici (Pgt), is a re-emerging, major threat to wheat production worldwide. Here, we retrieved, analyzed, and synthetized the available information about Pgt to develop a mechanistic, weather-driven model for predicting stem rust epidemics caused by uredospores. The ability of the model to predict the first infections in a season was evaluated using field data collected in three wheat-growing areas of Italy (Emilia-Romagna, Apulia, and Sardinia) from 2016 to 2021. The model showed good accuracy, with a posterior probability to correctly predict infections of 0.78 and a probability that there was no infection when not predicted of 0.96. The model's ability to predict disease progress during the growing season was also evaluated by using published data obtained from trials in Minnesota, United States, in 1968, 1978, and 1979, and in Pennsylvania, United States, in 1986. Comparison of observed versus predicted data generated a concordance correlation coefficient of 0.96 and an average distance between real data and the fitted line of 0.09. The model could therefore be considered accurate and reliable for predicting epidemics of wheat stem rust and could be tested for its ability to support risk-based control of the disease. Abstract Introduction Alzheimer's disease (AD) is a degenerative brain disease that progresses over time, heavily burdening patients, families, and aging societies worldwide. Memantine and donepezil are frequently used in its treatment, both as monotherapy and in combination. This multiple treatment comparison meta‐analysis assessed the efficacy of these regimens and placebo in the management of AD. Methods We searched PubMed, Embase, the Cochrane Library, and Wanfang Med Online and China National Knowledge Infrastructure for English and Chinese publications from the first records to 17 April 2020. Two investigators scanned articles for placebo‐controlled trials of memantine and donepezil alone and in combination. We extracted data on the following outcomes: cognition, global assessment, daily activities, neuropsychiatric symptoms, adverse events, and the acceptability and cost of these treatment regimens. Results Of 936 records screened, we included 54 trials in this analysis. The combination therapy was more effective in improving cognition (mean difference (MD)‐5.01, 95% credible interval (95% Crl) −10.73 to 0.86 in the Alzheimer's Disease Assessment Scale‐Cognitive Subscale; MD 9.61, 95% Crl 2.29 to 16.97 in the Severe Impairment Battery), global assessment (MD −2.88, 95% Crl −6.04 to 0.40), daily activities (MD 13.06, 95% Crl −34.04 to 58.92), and neuropsychiatric symptoms (MD −6.84, 95% Crl −10.62 to –2.82) compared with placebo. Memantine was more acceptable than placebo (MD 0.93, 95% Crl 0.69 to 1.22). Conclusions Memantine plus donepezil showed superior outcomes for cognition, global assessment, daily activities, and neuropsychiatric symptoms, but lower acceptability than monotherapy and placebo. Combination therapy may be more cost‐effective, because memantine slows the progression of AD.

Anthracnose, caused by Elsinoë ampelina, is an economically important grapevine disease in south and southeast Brazil. Control is achieved by lime sulphur application during grapevine dormancy and foliar fungicide sprays until the berries are half‐grown. This study assessed the temporal and spatial progress of grapevine anthracnose under field conditions in order to describe the disease dynamics and its relationship to pathogen survival. The experiment was carried out in a vineyard of table grape Vitis labrusca in Brazil, during the 2014 and 2015 growing seasons. The incidence of vines with diseased leaves, stems and berries and the disease severity on leaves were recorded from bud break to veraison. Monomolecular, logistic and Gompertz models were fitted by non‐linear regression to the incidence and severity data over time to characterize the temporal progress. Ordinary runs, dispersion index, modified Taylor's power law and spatial hierarchy analyses were used to characterize the spatial pattern of diseased plants. The monomolecular model showed the best fit for the incidence progress, with disease progress rates ranging from 0.051 to 0.136 per day. In both seasons, the incidence of diseased plants reached 100% 1 month after bud break. However, the incidence of diseased leaves per plant was around 60% and leaf disease severity was lower than 5% for both years. Ordinary runs and dispersion index analyses revealed that diseased grapevines were distributed randomly on the majority of the assessment dates. Meanwhile, a slight aggregation of diseased vines was observed in the modified Taylor's power law analysis. Our results suggested that the progress of anthracnose incidence and severity over time was governed mainly by the income of the primary inoculum, which survived in the vineyard. Neuroinflammation plays an important role in amyotrophic lateral sclerosis (ALS) pathogenesis. However, it is difficult to evaluate inflammation of the central nervous system (CNS) or the relationship between neuroinflammation and disease progression in ALS patients. Recent advances in the field of exosomes and CNS-derived exosomes extraction technology provide the possibility of measuring the inflammatory status in the CNS without brain biopsy. In this pilot study, we extracted astrocyte-derived exosomes from the plasma of sporadic ALS patients and age-, sex-matched healthy controls and determined Interleukin-6 (IL-6) levels by an enzyme-linked immunosorbent assay (ELISA). The IL-6 levels in astrocyte-derived exosomes were increased in sALS patients and positively associated with the rate of disease progression. However, the association between IL-6 levels and disease progression rate was limited to patients whose disease duration were less than 12 months. These data suggest an increased inflammatory cascade in the CNS of sALS patients. Our pilot study demonstrates that CNS-derived exosomes could be useful to reveal neuroinflammation of the CNS in ALS patients.

Background: Alteration in cytokine levels are known to be involved in the pathogenesis of schizophrenia. Objectives: To estimate the serum levels of interleukin-17 (IL-17) and interleukin-10 (IL-10) and their association with disease progression in schizophrenia. Methods: A total of 67 schizophrenia cases were enrolled in the present study. IL-17 and IL-10 were estimated by enzyme-linked immunosorbent assay. Positive and Negative Syndrome Scale (PANSS) was used to evaluate disease severity. Results: IL-17 was positively correlated with positive symptom score (r = 0.256, p =.036), general psychopathology score (r = 0.255, p =.038) and total score (r = 0.273, p =.025) in schizophrenia. IL-17 and IL-10 were significantly increased in schizophrenia cases with PANSS more than 85 compared to those with 71–85. Conclusion: IL-17 and IL-10 are associated with disease severity in schizophrenia but are not good markers for predicting the disease progression. [ABSTRACT FROM AUTHOR]

Coronavirus disease 2019 (COVID-19) patients were classified into four clinical stages (uncomplicated illness, mild, severe and critical pneumonia) depending on disease severity. We aim to investigate the corresponding clinical, radiological and laboratory characteristics between different clinical stages. A retrospective, single-centre study of 101 confirmed patients with COVID-19 at Renmin Hospital of Wuhan University from 2 January to 28 January 2020 was enrolled; follow-up endpoint was on 8 February 2020. Clinical data were collected and compared during the course of illness. The median age of the 101 patients was 51.0 years and 33.6% were medical staff. Fever (68%), cough (50%) and fatigue (23%) are the most common symptoms. About 26% patients underwent the mechanical ventilation and 98% patients were treated with antibiotics. Thirty-seven per cent patients were cured and 11 died. On admission, the number of patients with uncomplicated illness, mild, severe and critical pneumonia were 2 [2%], 86 [85%], 11 [11%] and 2 [2%]. Forty-four of the 86 mild pneumonia progressed to severe illness within 4 days, with nine patients worsened due to critical pneumonia within 4 days. Two of the 11 severe patients improved to mild condition while three others deteriorated. Significant differences were observed among groups of different clinical stages in numbers of influenced pulmonary segments (6 vs. 12 vs. 17, P < 0.001). A significantly upward trend was witnessed in ground-glass opacities overlapped with striped shadows (33% vs. 42% vs. 55% vs. 80%, P < 0.001), while pure ground-glass opacities gradually decreased as disease progressed (45% vs. 35% vs. 24% vs. 13%, P < 0.001) within 12 days. Lymphocytes, prealbumin and albumin showed a downtrend as disease progressed from mild to severe or critical condition, an uptrend was found in white blood cells, C-reactive protein, neutrophils and lactate dehydrogenase. Xanthomonas campestris pv. musacearum (Xcm), a bacterium causing banana xanthomonas wilt (BXW) disease, is widely spread in the East and Central Africa and infects all cultivated bananas. The first objective of this study was to identify plant species produced in banana cropping systems of Rwanda that may act as host of Xcm, and to evaluate the susceptibility of different banana cultivars in Rwanda. Eighteen plant species including banana, banana-intercrop species and plant species closely related to banana were used to study the host range of Xcm. Similarly, five banana cultivars (Fhia-17, Fhia-25, Injagi, Mpologoma and Nkazikamwa) were used to assess their susceptibility level to Xcm. Tested plant species and banana cultivars were inoculated with Xcm isolate in a controlled environment. Only banana and its relatives (enset, blood banana, achira, African arrowroot, and Indian shot) developed xanthomonas wilt symptoms. Time to first symptoms expression and to complete wilting varied significantly (P < 0.0001) between susceptible plant species. Longer survival times (P < 0.0001) were observed in wild (blood) banana, Canna species and enset compared to cultivated banana. Since these susceptible plant species could host the bacteria, they should be avoided in farms or gardens neighbouring banana fields. All the five banana cultivars tested were susceptible to Xcm. Among the cultivars, Mpologoma was first to express disease symptoms and to wilt completely; while the longest incubation period and time to complete wilting were recorded in cultivar Fhia-17. Due to the broad host range and high susceptibility of banana cultivars to Xcm, information about how to limit the spread of the bacteria is crucial for disease control. [ABSTRACT FROM AUTHOR]

Neuroinflammation plays an important role in amyotrophic lateral sclerosis (ALS) pathogenesis. However, it is difficult to evaluate inflammation of the central nervous system (CNS) or the relationship between neuroinflammation and disease progression in ALS patients. Recent advances in the field of exosomes and CNS-derived exosomes extraction technology provide the possibility of measuring the inflammatory status in the CNS without brain biopsy. In this pilot study, we extracted astrocyte-derived exosomes from the plasma of sporadic ALS patients and age-, sex-matched healthy controls and determined Interleukin-6 (IL-6) levels by an enzyme-linked immunosorbent assay (ELISA). The IL-6 levels in astrocyte-derived exosomes were increased in sALS patients and positively associated with the rate of disease progression. However, the association between IL-6 levels and disease progression rate was limited to patients whose disease duration were less than 12 months. These data suggest an increased inflammatory cascade in the CNS of sALS patients. Grapevine leaf stripe disease (GLSD) symptom expression was analysed in four vineyards and four cultivars, in Sardinia (Italy), taking into account ten-year annual and five-year monthly surveys. The cumulative incidence of symptomatic plants reached high values on Sauvignon blanc, Cabernet sauvignon and Cannonau (81.9, 79.4 and 66.5% respectively), but low on Merlot (25.1%). Symptoms appeared during or before the 50% flowering stage and maximum increments were assessed in June and partially in July. Annual incidence of foliar symptoms fluctuated in the ten years of the survey. Positive regressions were found between incidence of vines that exhibited foliar symptoms in year n but were symptomless in year n-1 and rain parameters in the 30 days after stabilization of mean temperature around 10°C, when colonization of pruning wounds begins. This relationship could suggest the involvement of new infections or re-infections on symptom expression in the following growth season. Significant regressions between incidence of vines that exhibited foliar symptoms in year n but were symptomless in the year n+1 and climatic parameters were also recorded. High temperatures and low rainfall in the period from pre-flowering to veraìson were conducive to a higher number of asymptomatic plants. Regarding monthly foliar symptoms evolution, an increase in temperature from 50% sprouting until June led to a greater number of new symptomatic plants. On the other hand, a smaller percentage of new symptomatic plants was associated with an increase in temperature from June to July, which may have influenced vine water balance and transport of toxins by the sap flow.

The passion fruit woodiness disease, induced by Cowpea aphid borne mosaic virus (CABMV), is considered the most economically important factor limiting passion fruit production in many countries. This study aimed to assess the progression of symptoms caused by CABMV in different Passiflora species. Eighty genotypes belonging 12 species were evaluated in experiments carried out in a completely randomized block design with treatments arranged in a factorial scheme (80 genotypes x 7 evaluation intervals) considering each inoculated plant as a repetition. The disease severity was estimated by the disease index (DI). The evaluations were performed at 20, 27, 34, 41, 48, 55 and 62 days after inoculation (DAI). The average DI values were used to describe the disease progress at each evaluation interval. The shortest time from evaluation to stabilization of CABMV progress was considered to cluster the genotypes by the Scott-Knott test (p≤0.05) and to classify the genotypes as resistant (R), moderately resistant (MR), moderately susceptible (MS), susceptible (S) or highly susceptible (HS). The mean severity gradually increased in the intervals from 20 to 55 DAI and reached a maximum at 62 DAI. The five classes (R, MR, MS, S and HS) contained about 19, 24, 36, 10 and 11% of the germplasm accessions, respectively. The disease temporal progress was quite variable within and among groups, although no difference in DI was observed in the genotypes after 55 DAI, indicating genotype symptom stabilization at this time, regardless of the genotype resistance level.

In breeding for resistance to black pod, it is important to use an effective and economical method to screen the cocoa genotypes. The detached pod test has been regarded as the most economical and effective screening method as compared to selection of resistant genotypes in the field. This study evaluated the accuracy of the assessment technique used in the detached pod test between the estimation of the area under the disease-progress curve (AUDPC) and the standard assessment of diameter lesion recorded in the sixth day after inoculation of four genotypes at different resistant categories to black pod (KKM4 -- susceptible, KKM5 -- moderate suscetipble, BR25 -- moderate resistant and QH1003 -- resistant) at two development pod stages (young and mature pods). The steps involved in the assessment using AUDPC are measured the diameter of the established lesions for six days after inoculation with Phytophthora palmivora and calculated the disease severity, fitting the disease progress curve with the nonlinear model, calculated the AUDPC and ranked the cocoa genotypes' resistant based on the AUDPC value. The results indicated that the assessment of the mature cocoa genotypes for resistance to black pod using AUDPC gave better accuracy (100%) of resistant level compared to standard assessment (50%) meanwhile both assessment technique on young pods gave similar percentage (50%) of accuracy.
