Respiratory disease is one of the main causes for morbidity and mortality in cetaceans, which highlights the importance of understanding normal lung function and how it may impede homeostasis, and diving capacity. In addition, the use of breathing frequency as a proxy for metabolic rate requires a better estimate of the normal range of tidal volume, respiratory flow, and breath durations. In the current study, we use data on clinically healthy bottlenose dolphins (Tursiops spp.) collected over a decade to define how lung function varies with body mass, age, and sex while in water or during voluntary beaching. The data show that tidal volume consistently varies with body mass both during spontaneous and forced breaths both in water and while beached. Both peak expiratory and inspiratory flow varies with body mass, but also in some circumstances with sex and age. Total and inspiratory breath durations only varied with body mass during forced breaths on land. Expired tidal volume varied with both body mass and either total or expiratory breath duration. These data provide baseline for respiratory function in healthy bottlenose dolphins and suggest that either total or expiratory breath duration provide a useful proxy for tidal volume. Background: Current diving physiology postulates that SCUBA divers' arterial blood gas (ABG) levels vary proportionally to environmental pressure, but, to date, ABGs have only been obtained during simulated dives. Also, recent evidence supports the use of the arterial/alveolar (a:A) partial pressures of oxygen (PO2) ratio to predict the arterial PO2 (PaO2) under hyperbaric conditions from measurements obtained at 1 atmosphere absolute (ATA). This work summarizes ABGs obtained in SCUBA divers in real underwater conditions and aims to validate the a:A ratio in predicting PaO2 in this subset of individuals at depth. Methods: The study was approved by the local ethics committee. After cannulating the radial artery of the non-dominant limb, ABGs were sampled at the surface before the dive (A), at depth (15 meters of freshwater (mfw) or 42 mfw) before (B) and after (C) pedaling on a submersed bicycle for 10 minutes, and back at surface (D). After calculating the surface alveolar PO2 for each subject, the a:A ratio was obtained and used to predict PaO2 at depth. A linear regression between measured and predicted PaO2 was reported, along with the goodness-of-fit F test. Results: Six subjects performed the dive at 15 mfw, and four others at 42 mfw. The PaO2 proportionally increased at both depths, remaining stable before and after pedaling. The a:A calculated from the baseline ABG obtained at rest, out of the water, adequately predicted the PaO2 at depth (R2 = 0.97, p<0.001), better at 15 mfw but losing accuracy at 42 mfw. Conclusions: The ABGs confirmed the proportional rise of PaO2 in SCUBA divers underwater. The a:A ratio could be used to predict the magnitude of PaO2 rise at depth to limit exposure to hyperoxia, especially in repetitive recreational dives and professional divers. [ABSTRACT FROM AUTHOR]

Introduction: Diving in SCUBA modality modifies human physiology in many ways. These modifications have been studied since Paul Bert in a seminal work. This area of research is very sensible to technological development. At now, it is possible to record heart rate (HR) continuously while diving. The study of HR changes in SCUBA diving at different depths in a constant temperature of thermal water is the objective of the present paper. Methods: 18 healthy subjects were enrolled and HR was recorded while SCUBA diving in thermal water at a constant temperature of 33.6∘ C in the deepest Italian pool at Montegrotto (Padova, Italy). Three depths were investigated: −20, −30 and −40 meters. The HR has been recorded with a Galileo SOL diving computer. The dive was subdivided into three phases: descent (DSC), steady on depth (STD), post–dive (RSF), and average HR was evaluated in each phase. Moreover, considering the DSC and STD time duration, a statistical linear regression of HR and relative parameters, intercept and slope, were here assessed. Results: In STD phase, HR slope obtained by regression decreased with depth. A significant difference was found between the slope during STD at −20 vs. −40 m (p ≤ 0.05). Discussion: Present results emphasized different HR physiological adjustments among diving phases. Firstly, during the DSC, a rapid HR decrease is recognized as probably due to a vagal response; secondly, at STD, the inward blood redistribution requires another physiological adjustment. The mammalian dive reflex, characterized by bradycardia and peripheral vasoconstriction, occurs in all mammals, including humans, in response to apnea. However, the dive reflex to a single, maximal, dry, dynamic apnea (DYN) and how it compares to a time-matched exercise control trial (EX) or dry static apnea (SA) has not been studied. We examined the hypotheses that, compared with EX and SA, the magnitude of the 1) cardiovascular response and 2) hematological response to DYN would be greater. Cardiovascular parameters [heart rate (HR), systolic (SBP), diastolic (DBP), and mean arterial (MAP) blood pressure] were continuously collected in 23 (F = 6 females) moderate and elite freedivers, first during a maximal DYN, then during a time-matched SA and EX on a swimming ergometer in randomized order. Venous blood draws were made before and following each trial. The change in calculated oxygen saturation (DYN: −17 ± 13%, EX: −2 ± 1%, ΔSA: −2 ± 1%; P < 0.05, all comparisons) was greater during DYN compared with EX and SA. During DYN, ΔSBP (DYN: 104 ± 31 mmHg; EX: 38 ± 23 mmHg; and SA: 20 ± 11 mmHg), ΔDBP (DYN: 45 ± 12 mmHg; EX: 14 ± 10 mmHg; and SA: 15 ± 8 mmHg), and ΔMAP (DYN: 65 ± 17 mmHg; EX: 22 ± 13 mmHg; and SA: 16 ± 9 mmHg) were increased compared with EX and SA, while ΔHR was greater during EX (DYN: −24 ± 23 beats/min; EX: 33 ± 13 beats/min; and SA: −1 ± 10 beats/min) than either DYN or SA (P < 0.0001, all comparisons). Females had a greater pressor response to EX (ΔSBP: 59 ± 30 mmHg; ΔDBP: 24 ± 14 mmHg; and ΔMAP: 35 ± 8 mmHg) than males (ΔSBP: 31 ± 15 mmHg; ΔDBP: 11 ± 6 mmHg; and ΔMAP: 18 ± 8 mmHg; P < 0.01, all comparisons). Together, these data indicate that DYN elicits a distinct, exaggerated cardiovascular response compared with EX or SA alone. This review examines the relationship between the physiological demands of diving and premature ventricular complexes (PVCs) in divers. In the general population, some individuals have a greater tendency to experience PVCs, often without awareness or a clear understanding of the triggering factors. With the increasing availability and popularity of both scuba and apnoea diving, more people, including those with a predisposition to PVCs, are engaging in these activities. The underwater environment, with its unique stressors, may increase the risk of arrhythmogenic events, particularly PVCs. Here, we review the prevalence, pathophysiology, and aggravating factors of PVCs in divers, emphasising the need for a comprehensive cardiovascular assessment. Evidence suggests a higher prevalence of PVCs in divers compared with the general population, influenced by factors such as age, dive depth, gas bubbles, cold water immersion, pre-existing cardiovascular diseases, and lifestyle factors. The change in environment during diving could potentially trigger an increased frequency of PVCs, especially in individuals with a pre-existing tendency. We discuss diagnostic strategies, management approaches, and preventive measures for divers with PVCs, noting that although guidelines for athletes can be adapted, individual assessment is crucial. Significant knowledge gaps are identified, highlighting the need for future research to develop evidence-based guidelines and understand the long-term significance of PVCs in divers. This work aims to evaluate potential contributing factors to PVCs in divers and identify individuals who may be at higher risk of experiencing major adverse cardiovascular events (MACEs). This work aims to improve diver safety by promoting collaboration between cardiologists and diving medicine specialists and by identifying key areas for future investigation in this field. This work aims to improve the safety and well-being of divers by understanding the cardiovascular challenges they face, including pressure changes, cold water immersion, and hypoxia. We seek to elucidate the relationship between these challenges and the occurrence of PVCs. By synthesising current evidence, identifying knowledge gaps, and proposing preliminary recommendations, we aim to encourage collaboration between cardiologists and diving medicine specialists to optimise the screening, management, and risk stratification of PVCs in the diving population. Although generally thought to be an autonomic reflex, several studies indicate that the cardiovascular changes during diving are anticipatory and can be conditioned. When accounting for this respiratory dependence on cardiac function, several studies have reported an absence of a diving‐related bradycardia except during dives that exceed the duration that is fuelled by aerobic metabolism. This review summarizes what is known about the respiratory physiology in marine mammals, with a special focus on cetaceans. We measured resting metabolic rate (RMR), tidal volume (VT), breathing frequency (fR), respiratory flow, and end‐expired gases in rough‐toothed dolphins (Steno bredanensis) housed in managed care after an overnight fast and 1–2 hr following a meal. The measured average (± standard deviation) VT (4.0 ± 1.3 L) and fR (1.9 ± 1.0 breaths/min) were higher and lower, respectively, as compared with estimated values from both terrestrial and aquatic mammals, and the average VT was 43% of the estimated total lung capacity. The end‐expired gas levels suggested that this species keep alveolar O2 (10.6% or 80 mmHg) and CO2 (7.6% or 57 mmHg), and likely arterial gas tensions, low and high, respectively, to maximize efficiency of gas exchange. We show that following an overnight fast, the RMR (566 ± 158 ml O2/min) was 1.8 times the estimated value predicted by Kleiber for terrestrial mammals of the same size. We also show that between 1 and 2 hr after ingestion of a meal, the metabolic rate increases an average of 29% (709 ± 126 ml O2/min). Both body mass (Mb) and fR significantly altered the measured RMR and we propose that both these variables should be measured when estimating energy use in cetaceans. [ABSTRACT FROM AUTHOR]

Marine mammals such as northern elephant seals (NES) routinely experience hypoxemia and ischemia-reperfusion events to many tissues during deep dives with no apparent adverse effects. Adaptations to diving include increased antioxidants and elevated oxygen storage capacity associated with high hemoprotein content in blood and muscle. The natural turnover of heme by heme oxygenase enzymes (encoded by HMOX1 and HMOX2) produces endogenous carbon monoxide (CO), which is present at high levels in NES blood and has been shown to have cytoprotective effects in laboratory systems exposed to hypoxia. To understand how pathways associated with endogenous CO production and signaling change across ontogeny in diving mammals, we measured muscle CO and baseline expression of 17 CO-related genes in skeletal muscle and whole blood of three age classes of NES. Muscle CO levels approached those of animals exposed to high exogenous CO, increased with age, and were significantly correlated with gene expression levels. Muscle expression of genes associated with CO production and antioxidant defenses (HMOX1, BVR, GPX3, PRDX1) increased with age and was highest in adult females, while that of genes associated with protection from lipid peroxidation (GPX4, PRDX6, PRDX1, SIRT1) was highest in adult males. In contrast, muscle expression of mitochondrial biogenesis regulators (PGC1A, ESRRA, ESRRG) was highest in pups, while genes associated with inflammation (HMOX2, NRF2, IL1B) did not vary with age or sex. Blood expression of genes involved in regulation of inflammation (IL1B, NRF2, BVR, IL10) was highest in pups, while HMOX1, HMOX2 and pro-inflammatory markers (TLR4, CCL4, PRDX1, TNFA) did not vary with age. We propose that ontogenetic upregulation of baseline HMOX1 expression in skeletal muscle of NES may, in part, underlie increases in CO levels and expression of genes encoding antioxidant enzymes. HMOX2, in turn, may play a role in regulating inflammation related to ischemia and reperfusion in muscle and circulating immune cells. Our data suggest putative ontogenetic mechanisms that may enable phocid pups to transition to a deep-diving lifestyle, including high baseline expression of genes associated with mitochondrial biogenesis and immune system activation during postnatal development and increased expression of genes associated with protection from lipid peroxidation in adulthood.

High‐resolution dive depth and acceleration recordings from nearshore (Sarasota Bay, dive depth < 30 m), and offshore (Bermuda) bottlenose dolphins (Tursiops spp.) were used to estimate the diving metabolic rate (DMR) and the locomotor metabolic rate (LMR, L O2/min) during three phases of diving (descent, bottom, and ascent). For shallow dives (depth ≤ 30 m), we found no differences between the two ecotypes in the LMR during diving, nor during the postdive shallow interval between dives. For intermediate (30 m < depth ≤ 100 m) and deep dives (depth > 100 m), the LMR was significantly higher during ascent than during descent and the bottom phase by 59% and 9%, respectively. In addition, the rate of change in depth during descent and ascent (meters/second) increased with maximal dive depth. The dynamic aerobic dive limit (dADL) was calculated from the estimated DMR and the estimated predive O2 stores. For the Bermuda dolphins, the dADL decreased with dive depth, and was 18.7, 15.4, and 11.1 min for shallow, intermediate, and deep dives, respectively. Hypoxia and hyperoxia are both worrisome issues potentially affecting SCUBA divers, but validated methods to monitor these two conditions underwater are still lacking. In this experiment, a volunteer SCUBA diver was equipped with a pulse oximeter to detect peripheral oxygen saturation (SpO2) and a device to monitor the oxygen reserve index (ORi™). ORi™ values were compared with arterial blood oxygen saturation (SaO2) and the partial pressure of oxygen (PaO2) obtained from the cannulated right radial artery at three steps: at rest out of water; at −15 m underwater after pedaling on a submerged bike; after resurfacing. SpO2 and ORi™ mirrored the changes in SaO2 and PaO2, confirming the expected hyperoxia at depth. To confirm the potential usefulness of an integrated SpO2 and ORi™ device, further studies are needed on a broader sample with different underwater conditions and diving techniques. Tissue and blood gas embolism (GE) associated with fisheries bycatch are likely a widespread, yet underestimated, cause of sea turtle mortality. Here, we evaluated risk factors associated with tissue and blood GE in loggerhead turtles caught incidentally by trawl and gillnet fisheries on the Valencian coastline of Spain. Of 413 turtles (303 caught by trawl, 110 by gillnet fisheries), 54% (n  = 222) exhibited GE. For sea turtles caught in trawls, the probability and severity of GE increased with trawl depth and turtle body mass. In addition, trawl depth and the GE score together explained the probability of mortality (P[mortality]) following recompression therapy. Specifically, a turtle with a GE score of 3 caught in a trawl deployed at 110 m had a P[mortality] of ~50%. For turtles caught in gillnets, no risk variables were significantly correlated with either the P[GE] or GE score. However, gillnet depth or GE score, separately, explained P[mortality], and a turtle caught at 45 m or with a GE score between 3 and 4 had a P[mortality] of 50%. Differences in the fishery characteristics precluded direct comparison of GE risk and mortality between these gear types. Although P[mortality] is expected to be significantly higher in untreated turtles released at sea, our findings can improve estimates of sea turtle mortality associated with trawls and gillnets, and help guide associate conservation efforts. The deep sea, which encompasses the largest habitat on Earth, presents a set of extreme and unique environmental conditions, including high hydrostatic pressure, near-freezing temperatures, and perpetual darkness. These conditions pose significant challenges to the survival and energy management of its inhabitants. Deep-sea organisms have evolved a range of bioenergetic adaptations to negotiate these harsh conditions, ensuring efficient energy acquisition and utilization. This review examines the multifaceted strategies employed by deep-sea animals, focusing on three key areas: energy input, digestive and absorptive efficiency, and energy consumption. We examine the physical environment of the deep sea, highlighting vertical profiles of temperature, salinity, and dissolved oxygen, which contrast sharply with surface conditions. Physiological adaptations of deep-sea species, such as specialized digestive systems and enzyme modifications that function optimally under high pressure, are explored in detail. Furthermore, we discuss behavioral adaptations, including diurnal vertical migration, which optimize energy intake and reduce metabolic costs. Comparative analyses with shallow-water species provide insights into the evolutionary pressures that have shaped these adaptations. This review also addresses the concept of "power budgeting", in which energy expenditures for specific dynamic actions (SDAs) must be balanced with other metabolic demands. This comprehensive examination of bioenergetic adaptation in deep-sea organisms enhances our understanding of their resilience and adaptability, offering glimpses into the complex interplay between environmental constraints and biological processes in one of the most challenging habitats on the planet.

Decompression sickness can occur in divers even when recommended decompression procedures are followed. Furthermore, the physiological state of individuals can significantly affect bubbling variability. These informations highlight the need for personalized input to improve decompression in SCUBA diving. The main objective of this study is to propose a fundamental framework for a new approach to inert gas exchanges. A physiological model of oxygen delivery to organs and tissues has been built and adapted to nitrogen. The validation of the model was made by transferring the N 2 to CO 2 . Under normobaric conditions (air breathing, oxygen breathing, and static apnea) and hyperbaric conditions, the O 2 model replicates the reference physiological Po 2 (Spearman correlation tests P < 0.001). The inert gas models can simulate inert gas partial pressures under normobaric and hyperbaric conditions. However, the lack of reference values prevents direct validation of this new model. Therefore, the N 2 model has been transferred to CO 2 . The resulting CO 2 model has been validated by comparing it with physiological reference values (Spearman correlation tests P < 0.01). The validity of the CO 2 model constructed from the N 2 model demonstrates the plausibility of this physiological model of inert gas exchanges. In the context of personalized decompression procedures, the proposed model is of significant interest as it enables the integration of physiological and morphological parameters (blood and respiratory flows, alveolo-capillary diffusion, respiratory and blood volumes, oxygen consumption rate, fat mass, etc.) into a model of nitrogen saturation/desaturation, in which oxygen and CO 2 partial pressures can also be incorporated. NEW & NOTEWORTHY This is the first model of inert gas transport based on the physiology of respiratory gas. It was built for O 2 delivery and validated against literature data; it was then transposed to N 2 exchanges. The transposition procedure was checked by transposing the N 2 model to CO 2 (and validated against literature data). This model opens the possibility to integrate physiological and morphological inputs in a personalized decompression procedure in SCUBA diving.

Introduction: Pulmonary airway dystrophy (PAD) is a rare disease that may be either innate or acquired. Very few publications have been dedicated to the repercussions of the air pressure variations that may be encountered during an airplane journey in a pressurized cabin, an aerial tramway ascent or a stay in a high-altitude setting. Variations may also occur during deep-sea diving with modification of absolute air pressure., State of Knowledge: The published observations show that all PAD cases have been associated with accidents during exposure or with variations in ambient air pressure. The different published reports should be considered with maximum caution and circumspection. It is of the utmost importance to painstakingly study each case in view of proposing a line of conduct in accordance with the etiology and severity of the disease. It also matters to take into full account the activity under consideration (climbing into altitude or deep-sea diving); the risks incurred are by no means identical., Perspectives: It would be of interest to hold a register of barotrauma cases or other accidents resulting from air pressure variations affecting persons with cystic lung disease or with pulmonary bullae in cases of PAD., Conclusion: While expert opinions and recommendations differ from one another, and notwithstanding the assistance provided by thoracic tomodensitometry, a premium should be placed on caution. Accurate collection of biometric data is important for understanding the biology and conservation of marine organisms, including elasmobranch and teleost fish, both in nature and controlled environments where monitoring marine specimens' health is mandatory. Traditional methods involving specimen capture and handling are invasive, stressful, and disruptive. Some techniques like underwater visual census or laser photogrammetry have been used for noninvasive data collection, but they have limitations and biases. The application of stereo-video photogrammetry through the use of diver-operated stereo-video systems (stereo-DOV) is a noninvasive method that overcomes these challenges, providing highly accurate measurements. It has become popular for species monitoring, studying anthropogenic impacts, and assessing length distributions. However, this technique is still uncommon and barely reported in aquarium settings. This study describes an innovative pilot study targeting multiple species carried out in a Public Aquarium, using a low-cost house-made device. The results revealed that measuring more than 100 individuals in approximately 1 day's work is possible. Total and fork lengths were estimated using specific software for 31 teleost and 16 elasmobranch species and compared with real measurements for the available species. Despite technical limitations that must be reviewed for application in future studies that resulted in high root mean square (RMS) values (> 20 mm), differences between methodological approaches revealed a minimal discrepancy (1.37%-5% in large sharks and rays and 1.8%-5.5% in teleost fish). Semiaquatic taxa, including humans, often swim at the air-water interface where they waste energy generating surface waves. For fully marine animals however, theory predicts the most cost-efficient depth-use pattern for migrating, air-breathing species that do not feed in transit is to travel at around 2 to 3 times the depth of their body diameter, to minimize the vertical distance traveled while avoiding wave drag close to the surface. This has rarely been examined, however, due to depth measurement resolution issues at the surface. Here, we present evidence for the use of this strategy in the wild to the nearest centimeter and document the switch to shallow swimming during naturally occurring long-distance migrations. Using high-resolution depth-accelerometry and video data for little penguins ( Eudyptula minor ) and loggerhead turtles ( Caretta caretta ), satellite-relayed data for green turtles ( Chelonia mydas ), and literature data for further sea turtle, penguin, and whale species, we show that near-surface swimming is likely used broadly across nonforaging diving animals to minimize the cost of transport., Competing Interests: Competing interests statement:The authors declare no competing interest.

We examined data from Naval Sea Systems Command grant project N0463A-12-C-001, "Hypercapnia: cognitive effects and monitoring", with the objective of validating or repudiating heart rate variability (HRV) as a warning sign of cognitive impairment from diving gas narcosis or oxygen toxicity. We compared HRV feature scores to their temporally corresponding cognitive outcomes under normal and narcotizing conditions to identify specific HRV features associated with cognitive changes. N0463A-12-C-001 was conducted between 17 September 2013 and 29 January 2016 and employed NASA's multi-attribute task battery (MATB-II) flight simulator to examine the independent effects of CO 2 , N 2 , and O 2 partial pressure on diver performance at simulated depths up to 61 msw (200 fsw). We assessed the association of 23 distinct HRV features scores from 432 of the study's analyzable exposure stages in relation to MATB-II's four performance subclasses (motor, memory, attention, strategy) while controlling for exercise and CO 2 , N 2 , and O 2 gas partial pressure. Performance decrements were associated with normalized high-frequency HRVfeatures (HFnu, p = 0.0016) and the number of pairs of successive R-R intervals that differed by more than 50 ms (NN50count1, p = 0.04). Secondary analysis with stratification restricted to non-exercise stages showed that several HRV parameters, including root mean square of the successive difference (RMSSD, p = 0.0015), width of Poincaré plot ( p = 0.0017), NN50count1 ( p = 0.0019), and standard deviation of normal-to-normal R peaks ( p = 0.0082), were associated with performance impairment. The RMSSD association retained statistical significance after Bonferroni correction for multiple tests. HRV features collected from divers tested under narcotizing conditions of breathing gas partial pressure and exercise were associated with performance impairment.

Energy governs species' life histories and pace of living, requiring individuals to make trade-offs. However, measuring energetic parameters in the wild is challenging, often resulting in data collected from heterogeneous sources. This complicates comprehensive analysis and hampers transferability within and across case studies. We present a novel framework, combining information obtained from eco-physiology and biologging techniques, to estimate both energy expenditure and intake in 48 Adélie penguins (Pygoscelis adeliae) during the chick-rearing stage. We employed the machine learning algorithm random forest (RF) to predict accelerometry-derived metrics for feeding behaviour using depth data (our proxy for energy acquisition). We also built a time-activity model calibrated with doubly labelled water data to estimate energy expenditure. Using depth-derived time spent diving and amount of vertical movement in the sub-surface phase, we accurately predicted energy expenditure. Movement metrics derived from the RF algorithm deployed on depth data were able to accurately detect the same feeding behaviour predicted from accelerometry. The RF predicted accelerometry-estimated time spent feeding more accurately compared with historical proxies such as number of undulations or dive bottom duration. The proposed framework is accurate, reliable and simple to implement on data from biologging technology widely used on marine species. It enables coupling energy intake and expenditure, which is crucial to further assess individual trade-offs. Objectives: To determine the influence of a patent foramen ovale and fibroblast growth factor-21 on core temperature (Tc) responses in SCUBA divers. Additionally, we aimed to quantify the individual and combined influences of wetsuit thickness and anthropometric data on Tc changes during the dives., Design: An experimental study comparing the Tc responses between divers with (n = 17) and without a patent foramen ovale (n = 14)., Methods: A total of 31 divers participated in the study. Tc was measured pre- and post-dive in 17-18 °C sea water using a telemetric pill. Additionally, blood was drawn pre-dive and ~1-2 h post-dive for measurement of fibroblast growth factor-21., Results: There was no influence of a patent foramen ovale on the Tc responses during SCUBA diving in either dive profile (p > 0.05). Additionally, there was no influence of SCUBA diving on fibroblast growth factor-21 concentrations (p > 0.05). The strongest positive and significant associations with the ∆Tc/min were found when multiplying wetsuit thickness in millimeters by body mass (r 2  = 0.3147, p = 0.0010), BMI (r 2  = 0.3123, p = 0.0011), and body surface area (r 2  = 0.2877, p = 0.0019). Behavior is fundamental to neuroscience research, providing insights into the mechanisms underlying thoughts, actions and responses. Various model organisms, including mice, flies, and fish, are employed to understand these mechanisms. Zebrafish, in particular, serve as a valuable model for studying anxiety-like behavior, typically measured through the novel tank diving (NTD) assay. Traditional methods for analyzing NTD assays are either manually intensive or costly when using specialized software. To address these limitations, it is useful to develop methods for the automated analysis of zebrafish NTD assays using deep-learning models. In this study, we classified zebrafish based on their anxiety levels using DeepLabCut. Subsequently, based on a training dataset of image frames, we compared deep-learning models to identify the model best suited to classify zebrafish as anxious or non anxious and found that specific architectures, such as InceptionV3, are able to effectively perform this classification task. Although existing literature covers significant detail on the physiology of human freediving, the lack of standardized protocols has hindered comparisons due to confounding variables such as exercise and depth. By accounting for these variables, direct depth-dependent impacts on cardiovascular and blood oxygen regulation can be investigated. In this study, depth-dependent effects on 1 ) cerebral hemodynamic and oxygenation changes, 2 ) arterial oxygen saturation ([Formula: see text]), and 3 ) heart rate during breath-hold diving without confounding effects of exercise were investigated. Six freedivers (51.0 ± 12.6 yr; means ± SD), instrumented with continuous-wave near-infrared spectroscopy for monitoring cerebral hemodynamic and oxygenation measurements, heart rate, and [Formula: see text], performed sled-assisted breath-hold dives to 15 m and 42 m. Arterial blood gas tensions were validated through cross-sectional periodic blood sampling. Cerebral hemodynamic changes were characteristic of breath-hold diving, with changes during ascent from both depths likely driven by decreasing [Formula: see text] due to lung expansion. Although [Formula: see text] was significantly lower following 42-m dives [ t (5) = -4.183, P < 0.05], mean cerebral arterial-venous blood oxygen saturation remained at 74% following dives to both depths. Cerebral oxygenation during ascent from 42 m may have been maintained through increased arterial delivery. Heart rate was variable with no significant difference in minimum heart rate between both depths [ t (5) = -1.017, P > 0.05]. This study presents a standardized methodology, which could provide a basis for future research on human freediving physiology and uncover ways in which freedivers can reduce potential risks of the sport. NEW & NOTEWORTHY We present a standardized methodology in which trained breath-hold divers instrumented with wearable near-infrared spectroscopy (NIRS) technology and a cannula for arterial blood sampling completed sled-assisted dives to two different dive depths to account for the confounding factors of exercise and depth during breath-hold diving. In our investigation, we highlight the utility of wearable NIRS systems for continuous hemodynamic and oxygenation monitoring to investigate the impacts of hydrostatic pressure on cardiovascular and blood oxygen regulation.

Background: Scuba diving is an increasingly popular activity that involves the use of specialized equipment and compressed air to breathe underwater. Scuba divers are subject to the physiological consequences of being immersed in a high-pressure environment, including, but not limited to, increased work of breathing and kinetic energy expenditure, decreased fluid absorption, and alteration of metabolism. Individual response to these environmental stressors may result in a differential risk of decompression sickness, a condition thought to result from excess nitrogen bubbles forming in a diver's tissues. While the mechanisms of decompression sickness are still largely unknown, it has been postulated that this response may further be influenced by the diver's health status. Nutritional intake has direct relevancy to inflammation status and oxidative stress resistance, both of which have been associated with increased decompression stress. While nutritional recommendations have been determined for saturation divers, these recommendations are likely overly robust for recreational divers, considering that the differences in time spent under pressure and the maximum depth could result nonequivalent energetic demands. Specific recommendations for recreational divers remain largely undefined., Methods: This narrative review will summarize existing nutritional recommendations and their justification for recreational divers, as well as identify gaps in research regarding connections between nutritional intake and the health and safety of divers., Results: Following recommendations made by the Institute of Medicine and the Naval Medical Research Institute of Bethesda, recreational divers are advised to consume ~170-210 kJ·kg -1 (40-50 kcal·kg -1 ) body mass, depending on their workload underwater, in a day consisting of 3 hours' worth of diving above 46 msw. Recommendations for macronutrient distribution for divers are to derive 50% of joules from carbohydrates and less than 30% of joules from fat. Protein consumption is recommended to reach a minimum of 1 g of protein·kg -1 of body mass a day to mitigate loss of appetite while meeting energetic requirements. All divers should take special care to hydrate themselves with an absolute minimum of 500 ml of fluid per hour for any dive longer than 3 hours, with more recent studies finding 0.69 liters of water two hours prior to diving is most effective to minimize bubble loads. While there is evidence that specialized diets may have specific applications in commercial or military diving, they are not advisable for the general recreational diving population considering the often extreme nature of these diets, and the lack of research on their effectiveness on a recreational diving population., Conclusions: Established recommendations do not account for changes in temperature, scuba equipment, depth, dive time, work of breathing, breathing gas mix, or individual variation in metabolism. Individual recommendations may be more accurate when accounting for basal metabolic rate and physical activity outside of diving. However, more research is needed to validate these estimates against variation in dive profile and diver demographics.

So called Caisson's disease of bone is well recognized in the literature. The purpose of this paper is to move away from simply considering the orthopaedic aspects of avascular injury to bones and joints and provide some background physiology and physics to help understand the challenges encountered whilst diving. This is relevant to those working with professional divers. Either commercially or within the military setting, and some guidance will be provided for those involved in the acute care of divers injured or affected by the problems covered within this article. Heat shock proteins (HSPs) have protective effects against oxidative stress and decompression sickness. Nitric oxide may reduce bubble formation during decompression and its activity is regulated by HSPs. A simulated dive can cause the HSP response. The aim of this study was to describe the effect of simulated dives on the antioxidant system, HSPs, and nitric oxide synthase response and demonste the relationship between the concentration of HSPs and the intensification of oxidative stress. A total of 20 healthy professional divers took part in training, consisting of simulated dry dives in a hyperbaric chamber and split into experiment I (30 m exposure, 400 kPa) and experiment II (60 m exposure, 700 kPa) over 24 h. The activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPx) and the concentrations of malondialdehyde (MDA), heat shock protein 70 (HSP70), heat shock protein 90 (HSP90), endothelial (eNOS) and inducible (iNOS) nitric oxide synthase were measured. Increases in the activity of SOD and MDA concentration were demonstrated. The activity of GPx depended on the dive profile. The HSP70 serum level in both experiments was significantly lower after the dives. The mean HSP90 level was significantly higher after the simulated dive at 60 m. A significant relationship between HSP concentration and SOD/GPx activity was demonstrated. eNOS concentration increased after 60 m exposure. No change in iNOS concentration was observed. In conclusions, the simulated dive significantly affected the antioxidant system, heat shock protein expression and nitric oxide synthase; however, the changes depend on the diving conditions. Daily energy expenditure (DEE) is the result of decisions on how to allocate time among activities (resting, commuting and foraging) and the energy costs of those activities. Dynamic body acceleration (DBA), which measures acceleration associated with movement, can be used to estimate DEE. Previous studies of DBA-DEE correlations in birds were carried out on species foraging below their thermoneutral zone, potentially decoupling the DBA-DEE relationship. We used doubly labelled water (DLW) to validate the use of DBA on plunge-diving seabirds, Peruvian boobies (Sula variegata), foraging in waters above their thermoneutral zone (>19°C). Mass-specific DEEDLW in boobies was 1.12 kJ day-1 g-1, and higher in males than in females. DBA alone provided the best fitting model to estimate mass-specific DEEDLW compared with models partitioned per activity and time budget models. Nonetheless, the model parametrizing activity at and away from their onshore breeding colony was the most parsimonious (r=0.6). This r value, although high, is lower than that of all other avian studies, implying that temperature is not the main cause of DBA-DEE decoupling in birds. Time at the colony (∼80% of the day) was the largest contributor to DEE as it was the most time-consuming activity and involved nest defence. However, foraging was the most power-consuming activity (4.6 times higher activity-specific metabolic rate than resting at the colony), and commuting flight was higher than in other gliding seabirds. Animal-borne instruments are essential research tools for ecologists and physiologists. An increasing number of studies have shown impacts of carrying a tag on behaviour and energetics, which can have implications for animal welfare and data validity. Such impacts are a result of the additional mass and/or drag loads, with the latter requiring empirical measurements or computational fluid dynamics (CFD) to estimate. To quantify and effectively minimize tag impacts from drag, a novel combined empirical and CFD approach is required. Here, we demonstrate such an approach using captive phocid seals and the widely used Sea Mammal Research Unit (SMRU) Instrumentation Group GPS/GSM tag. We (i) show a significant change in the behaviour of grey seals when carrying a tag (gen 1; associated with 16.4% additional drag); (ii) redesigned the tag (gen 2) resulting in a lower additional drag of 8.6%; (iii) show significant differences in behaviour when carrying a gen 2 compared to gen 1 tag, demonstrating that the redesign successfully reduced impact; and (iv) observed changes in the swim speed of seals that were consistent with predictions from CFD estimates of drag. The gen 2 instrument is now commercially available. This non-trivial case study should pave the way for similar studies in other taxa and species.

Abstract: This review article explores the multifaceted aspects of early specialization in diving, focusing on its implications for injury prevalence, performance, and mental health. It highlights the increasing trend of early specialization driven by ambitions for collegiate and Olympic success, despite recommendations advocating for diversification. Divers are at risk for acute and overuse injuries, particularly during high-velocity water entries; however, data on injury patterns, especially among elite divers and early specializers, remains scarce. Recent data suggest a correlation between early specialization and reduced rates of certain injuries among youth divers, but distinguishing between acute and overuse injuries is crucial. Additionally, early specialization may pose mental health risks, with divers facing pressures related to performance, body image, and parental expectations. This article underscores the need for tailored guidelines to mitigate injury risks and promote holistic well-being among youth divers, recognizing the complex interplay between physical and psychological factors in sports specialization., (Copyright © 2024 by the American College of Sports Medicine.)

Scuba diving and other modes of device-supported diving are popular activities that can be especially demanding and hazardous for people with preexisting physical conditions. Due to the high ambient pressure, the temperature differences, and potential unpredictable events, which have manifold effects on the organism, diving carries a high risk of life-threatening disease. A special risk is present if the body does not readily equalize air pressure changes. Therefore, prior to diving, all divers should undergo detailed education regarding the physical principles of the sport as well as specific physical examination. Consultation of an otolaryngologist is of exceptional relevance because many otorhinolaryngologic diseases can lead to (usually temporary) unfitness to dive. Rendezvous with sperm whales for biological observations is made challenging by their prolonged dive patterns. Here, we propose an algorithmic framework that codevelops multiagent reinforcement learning-based routing (autonomy module) and synthetic aperture radar-based very high frequency (VHF) signal-based bearing estimation (sensing module) for maximizing rendezvous opportunities of autonomous robots with sperm whales. The sensing module is compatible with low-energy VHF tags commonly used for tracking wildlife. The autonomy module leverages in situ noisy bearing measurements of whale vocalizations, VHF tags, and whale dive behaviors to enable time-critical rendezvous of a robot team with multiple whales in simulation. We conducted experiments at sea in the native habitat of sperm whales using an "engineered whale"-a speedboat equipped with a VHF-emitting tag, emulating five distinct whale tracks, with different whale motions. The sensing module shows a median bearing error of 10.55° to the tag. Using bearing measurements to the engineered whale from an acoustic sensor and our sensing module, our autonomy module gives an aggregate rendezvous success rate of 81.31% for a 500-meter rendezvous distance using three robots in postprocessing. A second class of fielded experiments that used acoustic-only bearing measurements to three untagged sperm whales showed an aggregate rendezvous success rate of 68.68% for a 1000-meter rendezvous distance using two robots in postprocessing. We further validated these algorithms with several ablation studies using a sperm whale visual encounter dataset collected by marine biologists.

Air-breathing vertebrates must balance their response to diel shifts in prey accessibility with physiological thresholds and the need to surface after each dive. Weddell seal (Leptonychotes weddellii) dive behaviors were tracked across the year under rapidly-changing light regimes in the Ross Sea, Antarctica ( ~ 75-77°S). This provides a 'natural experiment' with free-living seals experiencing 24-hrs of light (Polar Day), light/dark cycling, and continuous darkness (Polar Night). The Weddell seal's temporal niche switches from nocturnal diving in the summer to diurnality for the remainder of the year. Rhythmicity in dive efforts (depth, duration, post-dive surface recuperation, bottom time, and exceeding physiologic thresholds) is stronger and more closely circadian during times of the year with light/dark cycling compared with Polar Day or Night. With light/dark cycling, animals also make the most extreme dives (those that far exceed the calculated aerobic dive limit, cADL) significantly earlier than solar noon. Offsetting the longest dives that require longer surface recuperation times from mid-day allows animals to maximize total dive time under high-light conditions conducive for visual hunting. SCUBA diving poses risks due to pressure changes during descent (compression) and ascent (decompression). Decompression sickness (DCS) occurs due to gas bubble formation as the pressure decreases, causing joint pain, numbness, dizziness, or even paralysis and death. Immediate treatment involves 100% oxygen to help eliminate inert gases and hyperbaric oxygen therapy (HBOT), which is essential to reduce gas emboli formation and inflammation, thus improving symptoms. We evaluated oxy-inflammation biomarkers in the saliva and urine of nine subjects pre- and post-technical dive on the Haven wreck (GE, Italy). A case of DCS occurred during the dive. The injured diver was treated immediately with O 2 and transported to the hyperbaric center of "ASST Ospedale Ca Granda" in Milan. He was treated following the U.S. Navy Treatment Table 5 at 2.8 ATA and the day after with Table 15 at 2.4 ATA. Venous blood and urine samples were collected before and after each HBO treatment. Our study shows that dive increased oxy-inflammation biomarkers (ROS +126%; lipid peroxidation +23%; interleukins-6 +81%, -1β +19%, and TNFα +84%) and nitric oxide metabolites levels (+36%). HBOT after a DCS episode reduced oxidative stress, lowering the very high marker of lipid peroxidation (8-iso-PGF2α), and inhibited inflammatory interleukins. Overall, HBOT improved physiological responses in the diver affected by DCS.

The dive response involves three main components - breath holding, reduced heart rate and increased peripheral vasoconstriction - and is ubiquitous during forced dives in air-breathing vertebrates; however, numerous studies in free-diving animals have shown that the heart rate response to diving varies considerably in a manner that suggests cognitive control. Furthermore, studies on free-diving animals and controlled experiments in trained animals both indicate that the dive response can be conditioned, such that the reduction in heart rate begins before submergence and the extent of the reduction is set early in the dive. In addition, numerous species also experience an increase in heart rate and blood flow during ascent at the end of a dive, a phenomenon commonly called 'ascent tachycardia'. Collectively, these data suggest that although the dive response is under autonomic control, many species can vary its magnitude depending on the length and type of the planned dive - an indication of a role for cognition in the overall physiological responses associated with diving. Here, we provide examples of the conditioned cardiac responses - including anticipatory changes in heart rate - in several diving species and propose potential underlying mechanisms. Background: Saturation diving (SD) is useful and safe in deep diving for long durations. Japan Maritime Self-Defense Force (JMSDF) Undersea Medical Center (UMC) maintained safely deep 45 ATA SDHowever, cognitive performance was reportedly impaired by hyperbaric exposure in over 31 atmosphere absolute (ATA) SD. This study investigated the effects of hyperbaric exposure during 45 ATA deep SD on expert divers' cognitive function using Stroop tasks, a useful method to examine cognitive function, especially in narrow spaces such as SD chambers., Methods: Two numerical Stroop tasks were utilized to create two magnitude comparisons of a pair of single-digit numerical and physical tasks. Both numerical Stroop tasks were examined twice, at 1 and 45 ATAs, during a simulated 440 m of sea water depth for SD. Participants were 18 male expert JMSDF SD divers (age 36.58 ± 4.89 years)., Results: In the numerical task, reaction time (RT) was significantly delayed at 45 ATA compared with 1 ATA in the incongruent condition. In the physical task, RT at 45 ATA was significantly delayed under all the conditions (congruent, incongruent, and neutral). The correct rates (CR) in both numerical Stroop tasks significantly decreased at 45 ATA compared with 1 ATA in the incongruent condition., Conclusions: Our findings suggest that divers' cognition is impaired during 45 ATA deep SD. Skilled activity is a complex mix of automatized action, changed attention patterns, cognitive strategies and physiological adaptations developed within a community of practice. Drawing on physiological and ethnographic research on freediving, this article argues that skill acquisition demonstrates the variety of mechanisms that link biological and cultural processes to produce culturally shaped forms of embodiment. In particular, apneists alter phenotypic expression through patterned practices that canalize development, exaggerating the dive response, developing resistance to elevated carbon dioxide levels (hypercapnia) and accommodating hydrostatic pressure at depth. The community of divers provides technical advice and helps to orient individuals' motivations. Some biological processes are phenomenologically accessible, but others are sub-aware and must be accessed indirectly through behaviour or altered interactions with the environment. The close analysis of embodied skills like freediving illustrates how phenotypic plasticity is inflected by culturally patterned behaviours. Divers do developmental work on bodily traits like the dive response to achieve more dramatic performance, even if they cannot directly control all elements of the neurological and physiological responses. The example of expert freediving illustrates the imbrication of biology and culture in embodiment. This study aimed to create a training load index to measure physiological stress during breath-hold (BH) training and examine its relationship with memory performance. Eighteen well-trained BH divers (Age: 35.8±6.6 years, BH training practice: 5.3±4.5 years) participated in this study. During a standard 1.5-hour BH training in the pool, perceived exertion, heart rate, distance, and duration were measured. The training load index was modelled on the basis of a TRIMP (TRaining IMPulse) with four different equations and was used to measure the stress induced by this BH training. A reference value, based on the ratio between the average heart rate during all BHs and the lowest heart rate during BH training, was used for comparing training load index. Memory assessment was conducted both before and after this training. Of the four equations proposed, equation no. 4, named aTRIMP for "apnoea," showed the strongest correlation with our reference value (r=0.652, p<0.01). No difference was found between any of the memory tests before and after the BH training. The aTRIMP was a new representative index for monitoring habitual training of well-trained BH divers. Introduction: Aortic dissections and dissections of cervical, cerebral, and coronary arteries have been previously reported in scuba divers. These incidents may be the consequence of a variety of physiological effects. We review the reported cases of arterial dissection in scuba divers and discuss potential contributing factors related to immersion and diving., Methods: Medline, CINAHL Plus, and SPORTDiscus were searched for published reports of arterial dissection and the Australasian Diving Safety Foundation fatality database was searched for additional cases from Australia. Identified cases were recorded and scrutinised for possible contributing factors., Results: Nineteen cases of arterial dissection, both fatal and non-fatal, were identified. These included cervical or intracranial artery dissection (n = 14), aortic dissection (n = 4), and coronary artery dissection (n = 1). There were 14 male and five female victims; mean age 44 years (SD 14, range 18-65). Contributing factors may include a combination of vasoconstriction and blood redistribution, untreated hypertension, increased pulse pressure, abnormal neck movement or positioning, constrictive and burdensome equipment, exercise, increased gas density and circuit resistance with concomitant elevated work of breathing, atheroma, and possibly the mammalian dive response., Conclusions: Dissecting aneurysms of the aorta or cervical, cerebral, and coronary arteries should be considered as a potential complication of scuba diving. The development of aneurysms associated with scuba diving is likely multifactorial in pathogenesis. Detailed reporting is important in the evaluation of cases. Seal lice, unique among insects, show remarkable adaptability to the extreme conditions of the deep sea. Evolving with their seal and sea lion hosts, they have managed to tolerate hypoxia, high salinity, low temperature, and elevated hydrostatic pressure. Given the diving capabilities of their mammalian hosts, which can reach depths of hundreds to thousands of meters, our study examines the morphological variation among closely related seal lice species infesting hosts with different maximum diving depths. In particular, our research reveals a significant morphological difference between lice associated with regular and deep-diving hosts, where lice from deep-diving hosts tend to be rounder. This could be an adaptation to withstand the high hydrostatic pressures found in the deep ocean. The rounded shape optimizes the louse's ability to withstand external pressure by redistributing it over a larger ventral/dorsal plane. Diving marine mammals are a diverse group of semi- to completely aquatic species. Some species are targets of conservation and rehabilitation efforts; other populations are permanently housed under human care and may contribute to clinical and biomedical investigations. Veterinary medical care for species under human care, at times, may necessitate the use of general anesthesia for diagnostic and surgical indications. However, the unique physiologic and anatomic adaptations of one representative diving marine mammal, the bottlenose dolphin, present several challenges in providing ventilatory and cardiovascular support to maintain adequate organ perfusion under general anesthesia. The goal of this review is to highlight the unique cardiopulmonary adaptations of the completely aquatic bottlenose dolphin (Tursiops truncatus), and to identify knowledge gaps in our understanding of how those adaptations influence their physiology and pose potential challenges for sedation and anesthesia of these mammals., (© 2024 The Author(s). Background: Immersion pulmonary edema (IPE) is a form of hemodynamic edema likely involving individual susceptibility., Research Question: Can assessing right ventricle (RV) systolic adaptation during immersion be a marker for IPE susceptibility?, Study Design and Methods: Twenty-eight divers participated: 15 study participants with a history of IPE (IPE group; mean ± SD age, 40.2 ± 8.2 years; two women) and 13 control participants (no IPE group; mean ± SD age, 43.1 ± 8.5 years; two women) underwent three transthoracic echocardiography studies under three different conditions: dry (participants were in the supine position on an examination table without immersion), surface immersion (participants were floating prone on the water's surface and breathing through a snorkel), and immersion and negative static lung load (divers were submerged 20 cm below the water's surface in the prone position using a specific snorkel connected to the surface for breathing). Echocardiographic measurements included tricuspid annular plane systolic excursion (TAPSE), tissue S' wave, and right ventricle global strain (RVGLS)., Results: For all divers, immersion increased RV preload. In the no IPE group, the increase in RV preload induced by immersion was accompanied by an improvement in the contractility of the RV, as evidenced by increases in TAPSE (17.08 ± 1.15 mm vs 20.89 ± 1.32 mm), S' wave (14.58 ± 2.91 cm/s vs. 16.26 ± 2.77 cm/s), and RVGLS (25.37 ± 2.79 % vs. 27.09 ± 2.89 %). Negative SLL amplified these RV adaptations. In contrast, among divers with IPE, the increase in RV preload did not coincide with an improvement in RV contractility, indicating altered adaptive responses. In the IPE group, the TAPSE values changed from 17.19 ± 1.28 mm to 21.69 ± 1.67 mm and then to 23.55 ± 0.78 mm, respectively, in the dry, surface immersion, and immersion and negative SLL conditions. The S' wave values changed from 13.42 ± 2.94 cm/s to 13.26 ± 2.96 cm/s and then to 12.49 ± 0.77 cm/s, respectively, and the RVGLS values changed from -24.09% ± 2.91% to -23.99% ± 3.38% and then to -21.96% ± 0.55%, respectively., Interpretation: Changes in RV systolic function induced by immersion (especially with the addition of negative static lung load) vary among divers based on the history of IPE. Analyzing ventricular contractility during immersion, particularly RVGLS, could help to identify individual susceptibility in divers. Bubble use evolved in many small invertebrates to enable underwater respiration, but, until recently, there has been no evidence that vertebrate animals use bubbles in a similar manner. Only one group of vertebrates, semi-aquatic Anolis lizards, may be an exception: these lizards dive underwater when threatened and, while underwater, rebreathe a bubble of air over their nostrils. Although it seems that rebreathing should be adaptive, possibly functioning to extend the time that lizards remain in underwater refugia, this has not been empirically tested. Here, I demonstrate that rebreathing serves to extend dive time in a semi-aquatic anole, Anolis aquaticus . I prevented the formation of normal rebreathing bubbles by applying a commercial emollient on the skin surface where bubbles form to assess the impact of bubbles on rebreathing cycles, gular pumps, and dive times. Lizards that were allowed to rebreathe normally remained underwater an average of 32% longer than those with impaired rebreathing, suggesting a functional role of rebreathing in underwater respiration. Unlike rebreathing, gular pumping was unaffected by treatment and may warrant further research regarding its role in supplementing underwater respiration. This study provides evidence that vertebrates can use bubbles to respire underwater and raises questions about adaptive mechanisms and potential bio-inspired applications.

Developments in wearable human medical and sports health trackers has offered new solutions to challenges encountered by eco-physiologists attempting to measure physiological attributes in freely moving animals. Near-infrared spectroscopy (NIRS) is one such solution that has potential as a powerful physio-logging tool to assess physiology in freely moving animals. NIRS is a non-invasive optics-based technology, that uses non-ionizing radiation to illuminate biological tissue and measures changes in oxygenated and deoxygenated hemoglobin concentrations inside tissues such as skin, muscle, and the brain. The overall footprint of the device is small enough to be deployed in wearable physio-logging devices. We show that changes in hemoglobin concentration can be recorded from bottlenose dolphins and gray seals with signal quality comparable to that achieved in human recordings. We further discuss functionality, benefits, and limitations of NIRS as a standard tool for animal care and wildlife tracking for the marine mammal research community.

Adult Weddell seals (Leptonychotes weddellii) haul-out on the ice in October/November (austral spring) for the breeding season and reduce foraging activities for ~4 months until their molt in the austral fall (January/February). After these periods, animals are at their leanest and resume actively foraging for the austral winter. In mammals, decreased exercise and hypoxia exposure typically lead to decreased production of O2-carrying proteins and muscle wasting, while endurance training increases aerobic potential. To test whether similar effects were present in marine mammals, this study compared the physiology of 53 post-molt female Weddell seals in the austral fall to 47 pre-breeding females during the spring in McMurdo Sound, Antarctica. Once body mass and condition (lipid) were controlled for, there were no seasonal changes in total body oxygen (TBO2) stores. Within each season, hematocrit and hemoglobin values were negatively correlated with animal size, and larger animals had lower mass-specific TBO2 stores. But because larger seals had lower mass-specific metabolic rates, their calculated aerobic dive limit was similar to smaller seals. Indicators of muscular efficiency, myosin heavy chain composition, myoglobin concentrations, and aerobic enzyme activities (citrate synthase and β-hydroxyacyl CoA dehydrogenase) were likewise maintained across the year. The preservation of aerobic capacity is likely critical to foraging capabilities, so that following the molt Weddell seals can rapidly regain body mass at the start of winter foraging. In contrast, muscle lactate dehydrogenase activity, a marker of anaerobic metabolism, exhibited seasonal plasticity in this diving top predator and was lowest after the summer period of reduced activity.

Marine mammals such as northern elephant seals (NES) routinely experience hypoxemia and ischemia-reperfusion events to many tissues during deep dives with no apparent adverse effects. Adaptations to diving include increased antioxidants and elevated oxygen storage capacity associated with high hemoprotein content in blood and muscle. The natural turnover of heme by heme oxygenase enzymes (encoded by HMOX1 and HMOX2) produces endogenous carbon monoxide (CO), which is present at high levels in NES blood and has been shown to have cytoprotective effects in laboratory systems exposed to hypoxia. To understand how pathways associated with endogenous CO production and signaling change across ontogeny in diving mammals, we measured muscle CO and baseline expression of 17 CO-related genes in skeletal muscle and whole blood of three age classes of NES. Muscle CO levels approached those of animals exposed to high exogenous CO, increased with age, and were significantly correlated with gene expression levels. Muscle expression of genes associated with CO production and antioxidant defenses (HMOX1, BVR, GPX3, PRDX1) increased with age and was highest in adult females, while that of genes associated with protection from lipid peroxidation (GPX4, PRDX6, PRDX1, SIRT1) was highest in adult males. In contrast, muscle expression of mitochondrial biogenesis regulators (PGC1A, ESRRA, ESRRG) was highest in pups, while genes associated with inflammation (HMOX2, NRF2, IL1B) did not vary with age or sex. Blood expression of genes involved in regulation of inflammation (IL1B, NRF2, BVR, IL10) was highest in pups, while HMOX1, HMOX2 and pro-inflammatory markers (TLR4, CCL4, PRDX1, TNFA) did not vary with age. We propose that ontogenetic upregulation of baseline HMOX1 expression in skeletal muscle of NES may, in part, underlie increases in CO levels and expression of genes encoding antioxidant enzymes. HMOX2, in turn, may play a role in regulating inflammation related to ischemia and reperfusion in muscle and circulating immune cells. Our data suggest putative ontogenetic mechanisms that may enable phocid pups to transition to a deep-diving lifestyle, including high baseline expression of genes associated with mitochondrial biogenesis and immune system activation during postnatal development and increased expression of genes associated with protection from lipid peroxidation in adulthood.
