Abstract

Simple Summary: Ticks significantly impact livestock by causing diseases that affect animal health and reduce productivity. This study examined ticks infesting cattle across six provinces in South Africa, aiming to identify and understand the diversity of tick species and their potential to transmit diseases. Over 3500 ticks were collected, preserved, and analysed using a microscope and identification DNA sequencing. The findings revealed 15 species of ticks, with the most common being Amblyomma hebraeum, which accounted for more than half of all ticks found. The genetic analysis confirmed their identification and highlighted their diversity. This research is vital for understanding the tick species present, and their distribution, in South Africa. Regular monitoring of tick populations and using targeted control measures can help protect livestock from the harmful effects of ticks and improve cattle health and productivity. These findings are valuable to farmers, veterinarians, and researchers, offering practical knowledge to mitigate economic losses caused by tick infestations. Effective management of ticks will also enhance food security and the sustainability of livestock farming in South Africa. Ticks are a significant threat to livestock globally, with certain species displaying distinct host preferences at various developmental stages. Accurate species-level identification is essential for studying tick populations, implementing control strategies, and understanding disease dynamics. This study evaluated ticks infesting cattle across six provinces in South Africa using morphological and molecular methods. Ticks were preserved, examined morphologically using an Olympus Digital Camera Microscope, and identified using the 16S rRNA gene. The study identified four genera, namely Amblyomma, Hyalomma, Ixodes, and Rhipicephalus, comprising 15 ixodid species. Amblyomma hebraeum was the most prevalent species, with an infestation rate of 54.4%. Molecular analysis revealed genetic relationships among tick species, with genetic distances ranging from 0.00 to 0.13, and phylogenetic analysis clustered species into distinct genera with high bootstrap support. Principal Component Analysis highlighted clear genetic relatedness among species. These findings enhance our understanding of tick diversity, morphology, and distribution in South Africa's cattle populations, emphasizing their economic significance. The study provides critical baseline data for monitoring and developing effective strategies to manage tick-borne diseases, ensuring improved livestock health and productivity. Abstract

Income from goats highly depends on prolificacy, which is difficult to improve by traditional breeding methods. The study aimed to identify SNP markers for prolificacy, using a case–control genome-wide association study (GWAS) on 111 genotyped Cameroon native goat (CNG) does, based on the 50 K single nucleotide polymorphism (SNP) chip panel. None of the top SNPs reached the significant p-value of 5 × 10−8. The highest p-value was 0.0009. Despite the number of cases being about a quarter of the number of controls, the highest allele frequency of some of the top 20 variants in the cases was indicative of their potential role in the trait. These top variants included the following 15: rs268285661, rs268235169, rs268236449, rs268235135, rs268240394 in Sphingosine-1-phosphate phosphatase 2 (SGPP2) gene, rs268283635 in Solute carrier family 24 member 2 (SLC24A2) gene, rs268251678 in Androgen-induced gene 1 (AIG1) gene, rs268267018, rs268239617, rs268281364, rs268273029, rs268286941, rs268236144, rs268233233 in CEP126 gene and rs268278159, respectively. Our findings indicate that GWAS enable the identification of some loci within genes, with known biological functions and pathways in human being and mice animal model but far-ranging to what was previously hypothesized and tested in goat. Abstract

Background and Aim: Climatic conditions significantly impact the life stages and distribution patterns of ticks and tickborne diseases. South Africa's central plateau and various biomes offer a distinct landscape for studying the geography's effects. The study estimated tick species prevalence and the influential factors on their survival. Materials and Methods: Ticks were gathered from communal cattle in South African provinces including Limpopo (LP), Gauteng (GP), Mpumalanga (MP), KwaZulu-Natal (KZN), the Eastern Cape (EC), and the Free State (FS), from September 2020 to November 2022. Using data from South African weathercasts, the annual climate was assessed. Results: A total of 3,409 ticks were collected, with the highest infestation observed in KZN (45%), followed by LP (26%), EC (19%), GP (5%), MP (2%), and the FS (2%). The most prevalent tick species were Amblyomma hebraeum (55.1%), Rhipicephalus evertsi evertsi (13.9%) and Rhipicephalus (Boophilus), and decoloratus (11.9%). Other species included R. (Boophilus) microplus (10.85%), Hyalomma marginatum (4.8%), Rhipicephalus appendiculatus (1.4%), Harpalus rufipes (0.8%), Rhipicephalus exophthalmos (0.2%), Rhipicephalus glabroscutatus (0.2%), Rhipicephalus sanguineus (0.2%), Haemaphysalis silacea (0.5%), Ixodes pilosus (0.1%), and Rhipicephalus simus (0.1%). The infestations were most prevalent on farms in Pongola and KZN. The temperature fluctuated between 12°C and 35°C during data gathering, while humidity varied between 40% and 65%. Conclusion: This study showed that ticks survive optimally in warm temperatures and high humidity conditions. Livestock farms with high tick infestations may be associated with several risk factors. These practices could involve suboptimal grazing, insufficient acaricidal treatment, and detrimental effects resulting from traditional animal husbandry. Future research is needed to longitudinally evaluate the effects of climate change on tick populations, pathogen transmission, hosts, habitats, and human behavior, influencing potential exposure risks. Abstract

The African livestock sector plays a key role in improving the livelihoods of people through the supply of food, improved nutrition and consequently health. However, its impact on the economy of the people and contribution to national GDP is highly variable and generally below its potential. This study was conducted to assess the current state of livestock phenomics and genetic evaluation methods being used across the continent, the main challenges, and to demonstrate the effects of various genetic models on the accuracy and rate of genetic gain that could be achieved. An online survey of livestock experts, academics, scientists, national focal points for animal genetic resources, policymakers, extension agents and animal breeding industry was conducted in 38 African countries. The results revealed 1) limited national livestock identification and data recording systems, 2) limited data on livestock production and health traits and genomic information, 3) mass selection was the common method used for genetic improvement with very limited application of genetic and genomic-based selection and evaluation, 4) limited human capacity, infrastructure, and funding for livestock genetic improvement programmes, as well as enabling animal breeding policies. A joint genetic evaluation of Holstein-Friesian using pooled data from Kenya and South Africa was piloted. The pilot analysis yielded higher accuracy of prediction of breeding values, pointing to possibility of higher genetic gains that could be achieved and demonstrating the potential power of multi-country evaluations: Kenya benefited on the 305-days milk yield and the age at first calving and South Africa on the age at first calving and the first calving interval. The findings from this study will help in developing harmonized protocols for animal identification, livestock data recording, and genetic evaluations (both national and across-countries) as well as in designing subsequent capacity building and training programmes for animal breeders and livestock farmers in Africa. National governments need to put in place enabling policies, the necessary infrastructure and funding for national and across country collaborations for a joint genetic evaluation which will revolutionize the livestock genetic improvement in Africa.

Abstract

Africa, a continent of 1.3 billion people, had 326 researchers per one million people in 2018 (Schneegans, 2021; UNESCO, 2022), despite the global average for the number of researchers per million people being 1368 (Schneegans, 2021; UNESCO, 2022). Nevertheless, a strong research community is a requirement to advance scientific knowledge and innovation and drive economic growth (Agnew, et al., 2020; Sianes, et al., 2022). This low number of researchers extends to scientific research across Africa and finds resonance with genomic projects such as the African BioGenome Project (Ebenezer, et al., 2022).The African BioGenome project (AfricaBP) plans to sequence 100,000 endemic African species in 10 years (Ebenezer, et al., 2022) with an estimated 203,000 gigabases of DNA sequence. AfricaBP aims to generate these genomes on-the-ground in Africa. However, for AfricaBP to achieve its goals of on-the-ground sequencing and data analysis, there is a need to empower African scientists and institutions to obtain the required skill sets, capacity and infrastructure to generate, analyse, and utilise these sequenced genomes in-country.The Open Institute is the genomics and bioinformatics knowledge exchange programme for the AfricaBP (Figures 1 & 2). It consists of 10 participating institutions including the University of South Africa in South Africa and National Institute of Agricultural Research in Morocco. It aims to: develop biodiversity genomics and bioinformatics curricula targeted at African scientists, promote and develop genomics and bioinformatics tools that will address critical needs relevant to the African terrain such as limited internet access, and advance grassroot knowledge exchange through outreach and public engagement such as quarterly training and workshops.

Abstract

Animal production in low- and middle-income countries (LMICs), predominantly smallholder systems, faces considerable challenges. Low productivity, exposure to multiple pathogens, and climate-related stresses all cause major animal health and welfare issues. Unless addressed, these challenges will continue to negate initiatives aiming for sustainable animal production. To address this, the Royal (Dick) School of Veterinary Studies jointly established the Centre for Tropical Livestock Genetics and Health (CTLGH) with partners in Nairobi and Edinburgh (https://www.ctlgh.org/) to apply genetic improvement for improved animal productivity, health, and welfare in LMICs.

Abstract

Chickens are an important resource for smallholder farmers who raise locally adapted, genetically distinct breeds for eggs and meat. The development of efficient reproductive technologies to conserve and regenerate chicken breeds safeguards existing biodiversity and secures poultry genetic resources for climate resilience, biosecurity, and future food production. The majority of the over 1600 breeds of chicken are raised in low and lower to middle income countries (LMICs) under resource limited, small scale production systems, which necessitates a low tech, cost effective means of conserving diversity is needed. Here, we validate a simple biobanking technique using cryopreserved embryonic chicken gonads. The gonads are quickly isolated, visually sexed, pooled by sex, and cryopreserved. Subsequently, the stored material is thawed and dissociated before injection into sterile host chicken embryos. By using pooled GFP and RFP-labelled donor gonadal cells and Sire Dam Surrogate (SDS) mating, we demonstrate that chicks deriving entirely from male and female donor germ cells are hatched. This technology will enable ongoing efforts to conserve chicken genetic diversity for both commercial and small holder farmers, and to preserve existing genetic resources at poultry research facilities.

Abstract

Horizon scanning is intended to identify the opportunities and threats associated with technological, regulatory and social change. In 2017 some of the present authors conducted a horizon scan for bioengineering (Wintle et al., 2017). Here we report the results of a new horizon scan that is based on inputs from a larger and more international group of 38 participants. The final list of 20 issues includes topics spanning from the political (the regulation of genomic data, increased philanthropic funding and malicious uses of neurochemicals) to the environmental (crops for changing climates and agricultural gene drives). The early identification of such issues is relevant to researchers, policy-makers and the wider public.
