10. Liver function alterations among workers in the shoe industry due to combined low-level exposure to organic solvents.

Abstract

This study aimed to investigate the potential hepatotoxicity, nephrotoxic, and hematotoxic effects of simultaneous occupational low-level exposure of shoe workers to a mixture of organic solvents. The study included 16 male and 55 female workers and non-exposed subjects (n = 60) in the control group. Along with a standard sets of hematological, liver enzymes (aspartate aminotransferase (AST), alanine aminotransferase (ALT) and gamma-glutamyl transpeptidase (GGT), bilirubin total, bilirubin direct, blood glucose, urea, and creatinine were analyzed in all participants. Indoor air quality was monitored using a Gasmet Dx − 4000 multi-component analyzer. Despite the concentration levels of individual chemicals in shoe production units were below the permissible limits, the equivalent exposure (Em) values calculated based on the American Conference of Governmental Industrial Hygienists (ACGIH) and National Institute of Occupational Safety and Health (NIOSH) occupational exposure limits were higher than 1. Statistically significant increase of biochemical parameters (aspartate aminotransferase (AST), gamma-glutamyl transpeptidase (GGT), total bilirubin, and direct bilirubin) was obtained in exposed workers of both genders compared with controls (p < 0.001). Calculated liver damage risk scores were significantly higher in both females and males compared with controls (p < 0.001). The multivariate logistic regression analysis showed that direct bilirubin was the most important predictor of organic solvent mixture exposure in the studied group of workers. These results suggest that combined exposure to organic solvents even at low concentrations may lead to hepatotoxicity. [ABSTRACT FROM AUTHOR]

Abstract

Considering the urgent need for novel therapeutics in ongoing COVID‐19 pandemic, drug repurposing approach might offer rapid solutions comparing to de novodrug design. In this study, we designed an integrative in silicodrug repurposing approach for rapid selection of potential candidates against SARS‐CoV‐2 Main Protease (Mpro). To screen FDA‐approved drugs, we implemented structure‐based molecular modelling techniques, physiologically‐based pharmacokinetic (PBPK) modelling of drugs disposition and data mining analysis of drug‐gene‐COVID‐19 association. Through presented approach, we selected the most promising FDA approved drugs for further COVID‐19 drug development campaigns and analysed them in context of available experimental data. To the best of our knowledge, this is unique in silicostudy which integrates structure‐based molecular modeling of Mproinhibitors with predictions of their tissue disposition, drug‐gene‐COVID‐19 associations and prediction of pleiotropic effects of selected candidates.

Abstract

In efficacy testing of experimental oximes, traditionally reactivation of OP-inhibited acethylcholinesterase (AChE) has been analysed by comparing the obtained effects of the single dose with the control [1]. However, quantitative analysis of in vivo dose-response data by benchmark dose (BMD) approach would improve both identification and quantification of the effect and it will allow more rigorous comparison of different oximes efficacies [2]. Thus, we have evaluated in vivo dose-response relationship for two promising experimental oximes, K203 and K027, concerning reactivation of diaphragmal AChE inhibited by dichlorvos (DDVP). To compare the oximes effects, BMD-covariate method was used to estimate oxime dose (with 90% confidence intervals) that elicits a pre-specified effect size of 50% (1.5-fold increase in AChE activity compared to DDVP-treated group). Wistar rats (5/group) were treated with oxime (0/1.25/2.5/5/25/50% LD50 im) immediately after DDVP challenge (75% LD50 sc). Activity of AChE was measured in rat diaphragm homogenates by modified Ellman's method 60 min after the treatment. Dose-response modeling was done by PROAST software (version 65.5, RIVM, Nederlands). Exponential model m5-ab (y=a[c-(c-1)exp(-bxd)]) was selected as best estimate with parameters: aK203=0.1525, aK027=0.1498, bK203=0.008472, bK027=0.03941, c=2.117 and d=0.8916. Derived BMD50 were K203=117 (56, 209) and K027=21 (10, 37) µmol/kg bw, indicating that oxime K027 induces the same effect size with 5.5-times lower dose compared to oxime K203. Moreover, obtained confidence intervals of BMDs did not overlap allowing the conclusion that more potent dose-response relationship belongs to experimental oxime K027. [ABSTRACT FROM AUTHOR]
