In this contribution, new data concerning the distribution of native vascular flora in Italy are presented. It includes new records, confirmations, exclusions, and status changes to the Italian administrative regions. A new combination in the genus Pilosella is proposed. Nomenclatural and distribution updates, published elsewhere, and corrigenda are provided as Suppl. material 1.

This study aims to increase floristic knowledge of Marche by means of a survey in the Montagna di Torricchio State Nature Reserve (central Italy). The Reserve, located in the central Apennines, covers about 3.2 km2 at altitudes ranging from 820 to 1,491 m a.s.l. It has been owned and managed as a strict reserve by the University of Camerino since 1970: all the anthropic activities ceased about 50 years ago, except for a minimal area where mowing and cattle grazing are still allowed. The floristic list consists of 789 specific and subspecific taxa belonging to 81 families and 352 genera. Two species are new for Italy (Taraxacum calocarpum and T. pulchrifolium) and 14 for Marche regional flora. Compared to previous floristic studies, we found 127 more taxa but we showed a certain stability in the life-form spectrum, suggesting limited effects of dynamic processes related to climate and land-use changes. The negligible number of alien species (11) is probably related to the limitations to anthropic activities in the Reserve. The occurrence of taxa never recorded for Italy and Marche highlights the floristic value of the Reserve for species conservation in the central Apennines.

Extreme weather events, land use, and the presence of invasive species can act as pressures threatening biodiversity, resilience and ecosystem services. Particularly in the open cultural landscape, these pressures can suddenly drive ecosystems across tipping points and beyond thresholds of system integrity. Yet, biodiversity holds features for buffering against change. Potential stabilizing mechanisms include species richness, presence of key species such as legumes, and intraspecific diversity. These potential buffers can be promoted by conservation management and political decisions. On this basis, the results of the SIGNAL project and the related scientific based knowledge, will help to develop recommendations that could be used by International, national and local authorities, EU institutions, NGOs, for agriculture, landscape management and biodiversity conservation. Here we briefly describe the project and the Italian field experimental site.

Stachys annua subsp. annua, well-known in central Italy as 'stregona annuale', is an annual, small, slightly-scented herb, commonly found in fields and uncultivated areas in almost all regions of Italy. In folk medicine, its aerial parts were used as anti-catarrhal, febrifuge, tonic, and vulnerary. In the present work, the chemical composition of the flowering aerial parts was studied. The hydrodistilled volatile oil, analysed by GC/MS, showed sesquiterpenoids as the major fraction (42.5%); phytol (9.8%), germacrene D (9.2%), and spathulenol (8.5%) were the most abundant constituents. The volatile oil was assayed for antioxidant and cytotoxic activity by DPPH, ABTS, FRAP, and MTT methods. The cytotoxicity results against HCT116, A375, and MDA-MB 231 human tumor cell lines were significant, with IC50 values of 23.5, 37.2, and 41.5 μg/ml, respectively, whereas the antioxidant power was negligible. The EtOH extract was composed mainly of three glycosidic flavonoids, namely 7-{[2-O-(6-O-acetyl-β-D-allopyranosyl)-β-D-glucopyranosyl]oxy}-5,8-dihydroxy-2-(4-methoxyphenyl)-4H-1-benzopyran-4-one (1), 7-{[6-O-acetyl-2-O-(6-O-acetyl-β-D-allopyranosyl)-β-D-glucopyranosyl]oxy}-2-(3,4-dihydroxyphenyl)-5,8-dihydroxy-4H-1-benzopyran-4-one (2), and 7-{[6-O-acetyl-2-O-(β-D-allopyranosyl)-β-D-glucopyranosyl]oxy}-2-(3-hydroxy-4-methoxyphenyl)-5,8-dihydroxy-4H-1-benzopyran-4-one (3). On the contrary, iridoids, considered chemotaxonomic markers of the genus Stachys, were absent in this species. Finally, the morphological and histochemical survey showed that glandular trichomes were composed of two main types, i.e. peltate type A and capitate types B and C giving positive response for both lipids and polyphenols.

The aim of this study was to construct a solid basis for developing a crop wild relatives (CWR) and wild harvested plants (WHP) conservation strategy at the European and national levels. To this end we (i) worked out an annotated and synonymized Working Database of the Italian Vascular Plants, (ii) worked out a full CWR-WHP checklist for Italy, (iii) worked out a prioritized CWR and WHP inventory, (iv) identified native taxa with an ascertained utility for breeding, and (v) on the basis of a gap analysis case study on two Brassica taxa, outlined the first steps to be taken immediately to develop their in situ and ex situ conservation strategy. The Working Database of the Italian Vascular Plants includes a total of 11,710 taxa. Of those, 92.0% form the CWR-WHP checklist. On the basis of the importance of the related crops, their status, and their need of protection and/or monitoring, 1118 taxa were prioritized. Of these, 129 taxa deserve the highest priority in planning a conservation strategy, including 16 that currently have a practical use in breeding. The case study showed that little is still known about relic CWR populations and that not all of them are adequately protected in situ and ex situ. Since information on their abundance, location, distribution, ecological conditions, census, and current conservation is lacking, initiatives should be put into action immediately for developing a national and European conservation strategy.

Sideritis montana subsp. montana is a small annual herb occurring in countries bordering the Mediterranean and Balkan regions. The secondary metabolism of this plant has not been fully explored so far. The aim of the present study was to understand the complex mixture of secondary metabolites and the type of secretory structures. The polar constituents were isolated by column chromatography from the ethanolic extract, and their structure was elucidated by NMR and MS. The essential oil was isolated by hydrodistillation and analysed by GC/MS. The plant indumentum was studied by light and scanning electron microscopy. To complete the work, the essential oil antioxidant activity and cytotoxicity on tumor cells were evaluated by DPPH, ABTS, FRAP, and MTT methods. Four different classes of secondary metabolites were isolated, namely flavonoids, caffeoylquinic derivatives, glycosidic hydroquinones and iridoids. The essential oil was mainly characterized by sesquiterpenene hydrocarbons. Peltate and long-capitate hairs were the main sites where terpenes and polar constituents are produced. The secondary metabolites found in S. montana subsp. montana are of chemotaxonomic interest, some of them being typical of the genus Sideritis. The trichomes types observed partially differ from those described in other members of the genus Sideritis. The essential oil showed noteworthy inhibition on tumor cells.

Hypericum androsaemum , also known as Tutsan, produces fleshy and berry-like fruits that ripen from red to shiny black. The essential oil and aroma components of the red and black berries were investigated by GC–FID, GC–MS, and HS-SPME–GC–MS. The nutritional profile of the red and black berries in terms of proteins, carbohydrates, crude fiber, ascorbic acid, mineral content and fatty acid composition was also investigated. Finally, the morphological and histochemical study of fruits identified the histolocalization of the complex metabolite mixture produced. Results showed that monoterpene hydrocarbons gave the major contribution to the fruit aroma, with limonene as the most representative compound, even though some differences were found between red and black berries. Nutritional analysis highlighted a relevant content of ascorbic acid (135.7 mg/100 g) in red berries. The morphological and histochemical analysis showed that the fruit secretion is produced in numerous translucent glands that originated from the ovarian mesophyll and arranged in concentric rings.

Sideritis italica (Mill.) Greuter et Burdet belongs to the Lamiaceae family and is endemic to Italy. The glandular trichomes (morphology, distribution, histochemistry, and ultrastructure) of the plant were studied for the first time, along with the chemical composition of the essential oils. Abundant non-glandular hairs and peltate (type A) and capitate (types B, C(1), and C(x)) glandular trichomes were observed both on the vegetative and reproductive organs. The histochemical procedures and the ultrastructural investigation enabled specific location of the main site of essential oil production mainly in type-A peltate hairs. Particular emphasis is given to the release mechanism of the secreted material in all of the types of glands, and the potential taxonomic value of the indumentum in the Lamiaceae family is briefly discussed. Essential oils were hydrodistilled from flowering aerial parts of S. italica, and 136 compounds (112 in flowerheads, 79 in vegetative parts) were identified. The quantitative prevalence of diterpenoids (43.4% in flowerheads and 22.3% in vegetative parts) was the most significant characteristic of the essential oil of S. italica that could be classified as a diterpene-rich essential oil according to the classification of Kirimer.

Octen-3-ol is the most important C8 mushroom aromatic compound produced by many species of edi- ble fungi and is also an aroma component in several food and beverages products. Under this view, the essential oil of flowering aerial parts of Melittis melissophyllum subsp. melissophyllum (Lamiaceae) grow- ing in central Italy, obtained by hydrodistillation was characterised by GC-FID and GC-MS. This oil con- tained extremely high amount of the mushroom-like aroma component 1-octen-3-ol (43.6-54.2%), and could be considered as a new natural product for the use as flavouring agent in the food industry. Further- more, headspace analysis suggested that this aromatic compound is only present in low concentration in the plant part, and is primarily formed in higher amount during hydrodistillation of this material.

In recent years, research in botany was increasingly related with the use of large data-sets and data banks, in order to address emerging issues such as the severe risk of species, habitats and biodiversity loss. In this frame, the anArchive taxonomic Checklist, an online synonymized list of botanical species names, developed to support the botanical data banking and vegetation analysis, is presented and discussed here. The benefits deriving from such a supervised and referenced tool are emphasized. They include the possibility to keep track of old and new species names, pointing out the latest reviewed accepted scientific name and its synonyms, and harmonizing different taxonomic points of view. Furthermore, the list is open access and expert qualified customers can collaborate to its improvement. Methanol-acetone extracts from aerial parts of seven entities of the genus Hypericum [H. perforatum L. subsp. perforatum, H. perforatum L. subsp. veronense (Schrank) Ces., H. montanum L., H. hyssopifolium Chaix, H. hirsutum L., H. hircinum L. subsp. majus (Aiton) N. Robson, H. tetrapterum Fr.] growing in the Appennino Umbro-Marchigiano (central Italy) were tested in vitro for their antimicrobial activity against two Gram-positive bacteria (Staphylococcus aureus and Enterococcus faecalis), two Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) and the yeast Candida albicans, by the Kirby-Bauer agar diffusion method. The two H. perforatum subspecies were particularly active against Gram-positive and Gram-negative bacteria, supporting the use of these plants in the traditional medicine of central Italy to treat wounds, skin and infectious diseases. However significant values of inhibition were given also by H. hirsutum against S. aureus and P. aeruginosa, and by H. hyssopifolium, H. hircinum subsp. majus and H. tetrapterum against C. albicans. These results suggest that hypericin and hyperforin are the main components responsible for antimicrobial activity, but not the only ones, as other extracts, showing a low level of these compounds, possess evident activity. Isolation of pure compounds from the most active extracts is in progress.

Stachys alopecuros subsp. divulsa (Lamiaceae), a perennial herb endemic to central Italy growing on mountain pastures, was investigated for the first time for the content of secondary metabolites, for the micromorphology and histochemistry of glandular trichomes, and for the biological activity of the volatile oil, namely cytotoxic, antioxidant and antimicrobial. The plant showed the molecular pattern of iridoids, among which a new iridoid diglycoside (4′- O -β- d -galactopyranosyl-teuhircoside) was detected, together with a sterol glucoside and a phenylethanoid glycoside. The essential oil from the flowering aerial parts was characterized by a high proportion of sesquiterpene hydrocarbons (65.1%), with ( E )-caryophyllene (33.2%) as the most abundant, while other main components were germacrene D (7.6%), α-humulene (6.4%) and the oxygenated cis -sesquisabinene hydrate (10.2%). Taken together, polar and apolar chemical profiles support the classification of the species within the section Betonica of the genus Stachys . Micromorphological study revealed three types of glandular hairs secreting different classes of compounds, with type A peltate hairs producing the bulk of the essential oil. MTT assay revealed the potential of the volatile oil in inhibiting A375, HCT116 and MDA-MB 231 tumor cells (IC 50 values below 20 μg/ml).

Two years after its official start, the national vegetation database VegItaly, a collaborative project supported by the Italian scientific community and developed by a large group of scientists, is presented. This article offers a concise overview of the content of the database, currently consisting of 31,100 vegetation plot, including published and unpublished data. Some basic statistics are analysed; for example, data distribution in space and time, represented vegetation types expressed as physiognomic categories. Although rather young and still in progress, VegItaly already contains data from all the Italian regions and stands as an optimal candidate for the development of an Italian national vegetation database. Its main goals,theoretical basis, technical features, functionalities and recent progresses are outlined, showing glimpses of future prospects.

Ferula glauca L. (Apiaceae), formerly believed a subspecies of Ferula communis L., but at the present considered a distinguishable species, was studied for the first time for volatiles from leaves, flowers, fruits and roots. The chemical analysis of the essential oil obtained from different populations growing in Marche (central Italy) was performed by GC-FID and GC–MS. The differences in composition detected between F. glauca and F. communis made the volatile fraction a reliable marker to distinguish between them, and confirmed the botanical data at the base of their discrimination. In particular, the oils obtained from leaves and roots, contained as major compounds (E)-caryophyllene, caryophyllene oxide, myristicin and elemicin, that can be useful as marker components. Finally, the oils contained some daucane derivatives, that were detected also in F. communis and responsible for important biological properties.

The essential oil of Ephedra nebrodensis Tineo ex Guss. subsp. nebrodensis growing in central Italy was analyzed for the first time by GC and GC/MS. Fifty-nine components were identified, representing 89.3% of the oil under study. The oxygenated monoterpenes gave the highest contribution (33.0 %), and citronellol (29.7%) was the main component of this fraction. Esters were the second most abundant group of components (18.8%) with ethyl hexadecanoate (9.5%) as the main representative in this fraction. Qualitative and quantitative composition of the oil was discussed in order to support the actual botanical discrimination from E. major Host, so far exclusively morphologically based.

VegItaly is at present the largest Italian vegetation database. It is the result of a collaborative project aspiring to represent a major reference for the Italian vegetation scientists. The paper emphasizes its benefits for phytosociological data management and describes the solutions adopted to solve several technical problems, like the treatment of different vegetation stratification systems, the conversion of vegetation cover values, taxonomic and syntaxonomic issues, data import and access. The structure of the taxonomic list produced to support the storing of data is described. It allows an easy management of synonymic relationships and is constantly updated according to new publications and revisions. Issues related to data import from different formats have been solved by developing assistant software VegImport and TabImport, which are based on the most used formats in vegetation plot archiving. Bibliographic sources are managed according to the LISY standard and include descriptive geographic information, bibliographic and syntaxonomic reference. Distinct data access regimes can be selected by VegItaly's users: visible, partly visible, invisible. Compared with the original project outline, many fundamental parts of the database structure have been fully built up, although several utilities still have to be developed or improved. © Italian Society for Vegetation Science.
