Abstract

Studies in Benzimidazo[2,1-a]isoquinoline Chemistry by Michael John Donaghy B. Sc. (Hons.) Ellipticine, a member of the pyrido[4,3-b]carbazole alkaloid family first isolated in 1959 from the leaves of the plant Ochrosia Elliptica has been shown to possess anti-cancer activity against various tumours. Ellipticine is thought to undergo biological oxidation to give the more active 9-hydroxyellipticine that is subsequently converted to a highly reactive quinone-imine intermediate. The quinone-imine is then thought to interact with bionucleophiles in the body promoting cell death. The aim of the project was to synthesise a series of benzimidazo[2,1-a]isoquinolines. The benzimidazo[2,1-c]isoquinolines were similar in general structure to ellipticines and should therefore undergo similar biological reactions. The target benzimidazo[2,1-a]isoquinolines have been screened to evaluate their ability to inhibit the enzyme topoisomerase II that is involved in DNA replication and ultimately cell reproduction. The target compounds have also been evaluated by the American National Cancer Institute.

Abstract

1-Deoxysphingolipids (1-deoxySL) are atypical sphingolipids that are formed by the enzyme serine palmitoyltransferase (SPT) due to a promiscuous use of l-alanine over its canonical substrate l-serine. Several mutations in SPT are associated with the hereditary sensory and autonomic neuropathy type I (HSAN1). The current hypothesis is that these mutations induce a permanent shift in the affinity from l-serine toward l-alanine which results in a pathologically increased 1-deoxySL formation in HSAN1 patients. Also, wild-type SPT forms 1-deoxySL under certain conditions, and elevated levels were found in individuals with the metabolic syndrome and diabetes. However, the molecular mechanisms which control the substrate shift of the wild-type enzyme are not understood. Here, we report a novel SPTLC2-S384F variant in two unrelated HSAN1 families. Affected patients showed elevated plasma 1-deoxySL levels and expression of the S384F mutant in HEK293 cells increased 1-deoxySL formation. Previously, S384 has been reported as one of the two (S384 and Y387) putative phosphorylation sites in SPTLC2. The phosphorylation of wild-type SPTLC2 was confirmed by isoelectric focusing. The impact of an S384 phosphorylation on SPT activity was tested by creating mutants mimicking either a constitutively phosphorylated (S384D, S384E) or non-phosphorylated (S384A, Y387F, Y387F+S384A) protein. The S384D but not the S384E variant was associated with increased 1-deoxySL formation. The other mutations had no influence on activity and substrate affinity. In summary, our data show that S384F is a novel mutation in HSAN1 and that the substrate specificity of wild-type SPT might by dynamically regulated by a phosphorylation at this position.

A study to determine the effect on patients following administration of intravenous methcathinone is conducted. Results reveal that patients manifested a parkinsonian syndrome due to the effects of methcathinone raising incidence of acquired immunodeficiency syndrome (AIDS) and human immunodeficiency virus (HIV) besides elevated levels of blood manganese causing a neurological disorder.

Abstract

Many plastic monomers are known to cause serious health effects. However, when these monomers are chemically reacted to form the familiar plastic polymers, they are generally harmless. The plastic monomer acrylamide is known to cause damage to the peripheral nerves. Few reports exist indicating any serious hazard associated with methyl methacrylate monomer. This substance is in much more common use, and the polymer acrylic plastic is a familiar household item to many. Methyl methacrylate is also used in making various prostheses used in dentistry and orthopedic surgery. There have been some reports that some dentists, technicians, and orthopedic surgeons who mold methyl methacrylate monomer products without using gloves may develop sensory abnormalities in their fingers. Now, however, a patient has been seen with generalized peripheral nerve damage that may be the result of exposure to methyl methacrylate monomer. The patient noticed mild leg weakness on walking at the age of 55 which progressed over the next several years. Electrophysiological studies revealed a defect in conduction in the peripheral nerves, and a nerve biopsy revealed a reduced density of myelinated nerve fibers in a peripheral nerve. The observed changes were similar to those which have been observed in factory workers exposed to acrylamide. This patient had been a dental technician for 30 years, and it is estimated that during that time he made 10 pairs of dentures each week and repaired 15 more. During these tasks, he was exposed to methyl methacrylate not only through his fingers, but by breathing the vapors as well. Of course, a single case is insufficient to establish methyl methacrylate as the cause of the peripheral nerve disorder in this patient. Nevertheless, the case strongly suggests that long-term exposure to this plastic monomer carries with it a strong risk of peripheral nerve degeneration.
