The Caribbean spiny lobster Panulirus argus, a long-lived decapod crustacean, is fished throughout its geographical range and generates close to 1 billion US dollars annually. Fisheries across this range are fully- or over-exploited, spurring management strategies such as closed seasons and minimum size limitations. The objective of this study was to determine the minimum legal size and appropriate closed season for the lobster population of Saint Kitts and Nevis by assessing and comparing the functional, physiological and morphological determinants of sexual maturity. Such a comparison has never been attempted for this species. Females did not reach functional maturity until 87.8 mm carapace length (CL) based on the presence of a spermatophore and eggs, but reached morphological maturity based on the presence of ovigerous setae at 72.5 mm CL. Physiological maturity was intermediate at 82.8 mm CL. Determinants of functional maturity overestimate the size at sexual maturity, while the presence of ovigerous setae underestimates it. The gonadosomatic index, a reliable method of assessing physiological maturity in female P. argus, can be used to monitor changes in reproductive activity in both males and females. Gross assessment of the ovary may be used to determine size at physiological maturity, and functional maturity can be used to calculate peak periods of reproduction. The peak physiological and functional maturity for P. argus occurs from March to May. As a result, we propose a closed season from March to June and suggest assessment of reproductive dynamics in order to determine minimum legal size for harvesting P. argus.

Callinectes sapidus, or the 'blue crab', supports an extensive east-coast USA fishery and was one of the first crustacean species in which viruses were observed. Pioneering research by Dr Phyllis Johnson led to these initial discoveries, one of which included the discovery of a virus termed "Baculovirus-A". This virus was considered a potential member of the Baculoviridae, Nimaviridae, or Nudiviridae, in which all viral members are rod-shaped dsDNA viruses found in the nucleus of their host cell. With the availability of genomic and bioinformatic tools, such as Illumina HiSeq and assembly programs, it is now possible to assemble the genomes of viruses and gain additional genomic insight, which can shed light on viral taxonomy. Using these tools, alongside electron micrographs and histology slides, we reveal that the hepatopancreas-infecting 'Baculovirus-A' from Callinectes sapidus is a member of the Nudiviridae, resembling genetic and protein similarity to other crab and lobster infecting nudiviruses from the Gammanudivirus genus. Histologically, the virus causes nuclear hypertrophy as observed for other gammanuriviruses. The genome of the virus is circular, 122,436 bp in length, and encodes a predicted 98 protein coding genes, including all of the nudivirus core genes. The prevalence of virus from across Florida, USA, is provided alongside a genomic comparison of the new viral genome against other Gammanudivirus species, revealing the average prevalence to be 2.2% and that Callinectes sapidus nudivirus is distantly similar to the recently described Carcinus maenas nudivirus from Canada.

The Nudiviridae are a family of large double-stranded DNA viruses that infects the cells of the gut in invertebrates, including insects and crustaceans. The phylogenetic range of the family has recently been enhanced via the description of viruses infecting penaeid shrimp, crangonid shrimp, homarid lobsters and portunid crabs. Here we extend this by presenting the genome of another nudivirus infecting the amphipod Dikerogammarus haemobaphes. The virus, which infects cells of the host hepatopancreas, has a circular genome of 119,754 bp in length, and encodes a predicted 106 open reading frames. This novel virus encodes all the conserved nudiviral genes (sharing 57 gene homologues with other crustacean-infecting nudiviruses) but appears to lack the p6.9 gene. Phylogenetic analysis revealed that this virus branches before the other crustacean-infecting nudiviruses and shares low levels of gene/protein similarity to the Gammanudivirus genus. Comparison of gene synteny from known crustacean-infecting nudiviruses reveals conservation between Homarus gammarus nudivirus and Penaeus monodon nudivirus; however, three genomic rearrangements in this novel amphipod virus appear to break the gene synteny between this and the ones infecting lobsters and penaeid shrimp. We explore the evolutionary history and systematics of this novel virus, suggesting that it be included in the novel Epsilonnudivirus genus (Nudiviridae).

Among the many Callinectes spp. across the western Atlantic, the blue crab C. sapidus has the broadest latitudinal distribution, encompassing both tropical and temperate climates. Its life history varies latitudinally, from extended overwintering at high latitudes to year-round activity in tropical locations. Callinectes sapidus reovirus 1 (CsRV1) is a pathogenic virus first described in North Atlantic C. sapidus and has recently been detected in southern Brazil. Little information exists about CsRV1 prevalence at intervening latitudes or in overwintering blue crabs. Using a quantitative reverse transcription PCR (RT-qPCR) method, this study investigated CsRV1 prevalence in C. sapidus across latitudinal differences in temperature and crab life history, as well as in additional Callinectes spp. and within overwintering C. sapidus. CsRV1 prevalence in C. sapidus was significantly correlated with high water temperature and blue crab winter dormancy. Prevalence of CsRV1 in C. sapidus on the mid-Atlantic coast was significantly lower in winter than in summer. CsRV1 infections were not detected in other Callinectes spp. These findings revealed that CsRV1 is present in C. sapidus across their range, but not in other Callinectes species, with prevalence associated with temperature and host life history. Such information helps us to better understand the underlying mechanisms that drive marine virus dynamics under changing environmental conditions.

Aquaculture is predicted to supply the majority of aquatic dietary protein by 2050. For aquaculture to deliver significantly enhanced volumes of food in a sustainable manner, appropriate account needs to be taken of its impacts on environmental integrity, farmed organism health and welfare, and human health. Here, we explore increased aquaculture production through the One Health lens and define a set of success metrics — underpinned by evidence, policy and legislation — that must be embedded into aquaculture sustainability. We provide a framework for defining, monitoring and averting potential negative impacts of enhanced production — and consider interactions with land-based food systems. These metrics will inform national and international science and policy strategies to support improved aquatic food system design. By 2050, the majority of aquatic dietary protein will be produced by the aquaculture sector. A set of 15 metrics are presented here to guide the industry sustainably through the rapid growth and development it is experiencing.

Understanding population genetic structure can help us to infer dispersal patterns, predict population resilience and design effective management strategies. For sessile species with limited dispersal, this is especially pertinent because genetic diversity and connectivity are key aspects of their resilience to environmental stressors. Here, we describe the population structure of Ircinia campana, a common Caribbean sponge subject to mass mortalities and disease. Microsatellites were used to genotype 440 individuals from 19 sites throughout the Greater Caribbean. We found strong genetic structure across the region, and significant isolation by distance across the Lesser Antilles, highlighting the influence of limited larval dispersal. We also observed spatial genetic structure patterns congruent with oceanography. This includes evidence of connectivity between sponges in the Florida Keys and the southeast coast of the United States (>700 km away) where the oceanographic environment is dominated by the strong Florida Current. Conversely, the population in southern Belize was strongly differentiated from all other sites, consistent with the presence of dispersal-limiting oceanographic features, including the Gulf of Honduras gyre. Crayfish are common hosts of microsporidian parasites, prominently from the genus

Ovipleistophora (Microsporidia) is a globally distributed genus of obligate parasites that infect fish, Crustacea, and trematodes. We report on two additional crayfish hosts, Procambarus pictus (Simms Creek, Florida) and Procambarus fallax (Santa Fe River, Florida), that exhibited signs of high-intensity microsporidian infection in the musculature. Sequence data (SSU) for the isolates were 99.79% and 99.97% similar to Ovipleistophora diplostomuri parasitizing Procambarus bivittatus. Additional screening of regional fish species (Lepomis macrochirus) revealed additional microsporidian diversity (Potaspora) but did not include Ovipleistophora. Detecting Clade V isolates in multiple host groups provides further evidence for trophic transmission among Clade V microsporidia.

Catch-and-release angling of salmonids is a widely implemented fisheries management strategy intended to provide recreational fishing opportunities while limiting harvest. Previous research has indicated mortality may be low, but the extent of sublethal impacts, including inability to reach spawning grounds or decreased spawning effectiveness, is relatively under-studied. Sockeye salmon (Oncorhynchus nerka) were subjected to simulated catch-and-release events, with a subset also enduring air exposure, to determine how physiological stress influences the ability of sockeye salmon to reach spawning grounds and the viability of eggs produced. Short-term postrelease mortality was highest for fish that were subjected to air exposure, but once recovered, neither simulated catch-and-release nor air exposure had a major impact on their ability to reach spawning grounds. Egg viability was similar between treatment groups, indicating physiological stress associated with the trials did not have a negative impact on reproduction. These results suggest that handling, especially air exposure, is the primary concern and substantially impacts the short-term postrelease mortality of sockeye salmon, which has important implications for management.

The Caribbean spiny lobster Panulirus argus supports a large and valuable fishery in the Caribbean Sea. In 2007-2008, a rare microsporidian parasite with spore characteristics typical of the Ameson genus was detected in 2 spiny lobsters from southeast Florida (FL). However, the parasite species was not confirmed by molecular analyses. To address this deficiency, reported here are structural and molecular data on single lobsters displaying comparable 'cotton-like' abdominal muscle containing ovoid microsporidian spores found at different locations in FL in 2014 and 2018 and in Saint Kitts and Nevis Islands in 2017. In the lobster from 2014, multiple life stages consistent with an Ameson-like monokaryotic microsporidian were detected by transmission electron microscopy. A partial (1228 bp) small subunit (SSU) rRNA gene sequence showed each microsporidia to be identical and positioned it closest phylogenetically to Ameson pulvis in a highly supported clade also containing A. michaelis, A. metacarcini, A. portunus, and Nadelspora canceri. Using ecological, pathological, ultrastructural, and molecular data, the P. argus microsporidian has been assigned to a distinct species: Ameson herrnkindi.

Marine sponges are hosts to large, diverse communities of microorganisms. These microbiomes are distinct among sponge species and from seawater bacterial communities, indicating a key role of host identity in shaping its resident microbial community. However, the factors governing intraspecific microbiome variability are underexplored and may shed light on the evolutionary and ecological relationships between host and microbiome.Here, we examined the influence of genetic variation and geographic location on the composition of the Ircinia campana microbiome.We developed new microsatellite markers to genotype I. campana from two locations in the Florida Keys, USA, and characterized their microbiomes using V4 16S rRNA amplicon sequencing.We show that microbial community composition and diversity is influenced by host genotype, with more genetically similar sponges hosting more similar microbial communities. We also found that although I. campana was not genetically differentiated between sites, microbiome composition differed by location.Our results demonstrate that both host genetics and geography influence the composition of the sponge microbiome. Host genotypic influence on microbiome composition may be due to stable vertical transmission of the microbial community from parent to offspring, making microbiomes more similar by descent. Alternatively, sponge genotypic variation may reflect variation in functional traits that influence the acquisition of environmental microbes. This study reveals drivers of microbiome variation within and among locations, and shows the importance of intraspecific variability in mediating eco‐evolutionary dynamics of host‐associated microbiomes., For the first time, the authors show that intraspecific genetic variation affects microbiome composition in a marine sponge (Ircinia campana), with positive correlations observed between genetic and microbiome similarity. This has significant implications for our understanding of the ecological and evolutionary relationships between host and microbiome in this important model system.

Carcinus maenas is in the top 100 globally invasive species and harbours a wide diversity of pathogens, including viruses. We provide a detailed description for a novel bunyavirus (Carcinus maenas Portunibunyavirus 1) infecting C. maenas from its native range in the Faroe Islands. The virus genome is tripartite, including large (L) (6766 bp), medium (M) (3244 bp) and small (S) (1608 bp) negative sense, single-stranded RNA segments. Individual genomic segments are flanked by 4 bp regions of similarity (CCUG). The segments encode an RNA-dependent RNA-polymerase, glycoprotein, non-structural protein with a Zinc-Finger domain and a nucleoprotein. Most show highest identity to the ‘Wenling Crustacean Virus 9’ from an unidentified crustacean host. Phylogenomics of crustacean-infecting bunyaviruses place them across multiple bunyavirus families. We discuss the diversity of crustacean bunyaviruses and provide an overview of how these viruses may affect the health and survival of crustacean hosts, including those inhabiting niches outside of their native range.

Crustaceans suffer from diseases that can alter their survival and ecology with additional economic consequences for fisheries and aquaculture. Many parasites have been described from crustaceans and with the advent of novel technologies such as next generation sequencing, the discovery of novel parasites has become increasingly efficient. Molecular techniques are beginning to surpass more conventional tools for parasite discovery, but they typically do not provide information on pathology. Histopathology remains one of the least expensive methods for parasite discovery and allows for both detection of parasites and descriptions of the pathology they cause. When used in concert with modern molecular and electron microscopy techniques, the approach is powerful; however, there are few informational tools for the interpretation of histological slides from crustaceans. Those available do not provide comprehensive images of all organs and early works were limited to lower resolution than currently available. More recent texts provide in-depth details of infection in histological section, but few provide images of healthy material or describe a baseline from which to compare. Here, we provide a series of image plates derived from histologically processed tissues from three palinurid lobsters: Panulirus argus, Palinurus elephas and Panulirus guttatus. Histology from these lobsters shows high visual similarity in all tissue types. We provide a histological atlas of healthy tissue that can be used as a baseline resource for pathobiologists working on these common species (and related crustaceans) and we discuss how disease may result in visual aberrations to these tissues.

The Caribbean spiny lobster Panulirus argus is susceptible to infection by Panulirus argus Virus 1 (PaV1), the only virus known to naturally infect any lobster species. However, P. argus is able to mitigate PaV1 transmission risk by avoiding infected individuals. White Spot Syndrome Virus (WSSV) has a particularly wide host range. WSSV has not been documented in wild populations of spiny lobsters, but has been experimentally transmitted to six other lobster species from the genus Panulirus spp. While WSSV has been detected intermittently in wild populations of shrimp in the Caribbean region, the risk to P. argus has not been evaluated. Potential emergence of the disease could result in fisheries losses and ecological disruption. To assess the risk to P. argus, we tested its susceptibility to WSSV via injection and waterborne transmission. We also tested whether healthy lobsters can detect and avoid conspecifics with qPCR-quantifiable WSSV infections. We found P. argus to be highly susceptible to WSSV via intramuscular injection, with mortality reaching 88% four weeks post inoculation. Panulirus argus was also susceptible to WSSV via waterborne transmission, but WSSV burden was low after four weeks via qPCR. Behavioral assays indicated that P. argus can detect and avoid conspecifics infected with WSSV and the avoidance response was strongest for the most heavily infected individuals – a response comparable to PaV1-infected conspecifics. Panulirus argus is the first spiny lobster found to be susceptible to WSSV in the Americas, but it is possible that a generalized avoidance response by healthy lobsters against infected conspecifics provides a behavioral defense and may reduce WSSV infection potential and prevalence. Such avoidance may extend to other directly transmitted pathogens in spiny lobster populations preventing them from becoming common in their population.Author SummaryErica P. The COVID-19 pandemic imposed new norms on human interactions, perhaps best reflected in the widespread application of social distancing. But social distancing is not a human invention and has evolved independently in species as dissimilar as apes and lobsters. Epidemics are common in the wild, where their spread is enhanced by animal movement and sociality while curtailed by population fragmentation, host behavior, and the immune systems of hosts. In the present article, we explore the phenomenon of behavioral immunity in wild animals as compared with humans and its relevance to the control of disease in nature. We start by explaining the evolutionary benefits and risks of sociality, look at how pathogens have shaped animal evolution, and provide examples of pandemics in wild animal populations. Then we review the known occurrences of social distancing in wild animals, the cues used to enforce it, and its efficacy in controlling the spread of diseases in nature.

Abstract This study provides a broad phylogenetic analysis for the Eubrachyura, with the inclusion of three new Panopeidae mitochondrial genomes: Eurypanopeus depressus (flatback mud crab) (15,854bp), Panopeus herbstii (Atlantic mud crab) (15,812bp) and Rhithropanopeus harrisii (Harris, or ‘white-fingered’ mud crab) (15,892bp). These new mitogenomes were analyzed alongside all available brachyuran mitochondrial genomes (n = 113), comprising 80 genera from 29 families, to provide an updated phylogenetic analysis of the infra-order Brachyura (“true crabs”). Our analyses support the subsection Potamoida within the Eubrachyura as the sister group to Thoracotremata. The family Panopeidae aligns with the family Xanthidae to form the Xanthoidea branch, which is supported by current morphological and genetic taxonomy. A unique gene arrangement termed ‘XanGO’ was identified for the panopeids and varies relative to other members of the subsection Heterotremata (within the Eubrachyura) via a transposition of the trnV gene. This gene arrangement is novel and is shared between several Xanthoidea species, including Etisus anaglyptus (hairy spooner crab), Atergatis floridus (brown egg crab), and Atergatis integerrimus (red egg crab), suggesting that it is a conserved gene arrangement within the Xanthoidea superfamily. Our study further reveals a need for taxonomic revision of some brachyuran groups, particularly the Sesarmidae. The inclusion of panopeid mitogenomes into the greater brachyuran phylogeny increases our understanding of crab evolution and higher level Eubrachyuran systematics.

The blue crab Callinectes sapidus is one of the most widely studied marine crustaceans due to its high economic value and ecological significance. Despite extensive research on the blue crab in North America, many questions remain about the distribution and abundance of the species in the subtropics and tropics. In many places, C. sapidus is sympatric with morphologically similar Callinectes spp., which has implications for seafood mislabeling. To enable rapid identification of the species, we designed and tested two PCR-based assays targeting the 12S rRNA mitochondrial gene. The first assay discriminates C. sapidus from other Callinectes spp. via post-PCR restriction digestion (PCR-RFLP) and the second assay discriminates among multiple Callinectes spp. through High Resolution Melting (HRM) analysis and supervised machine learning analyses. A total of 58 DNA samples from five Callinectes spp. (validated via 12S gene sequencing) were used for assay testing. The PCR RFLP assay was 100% accurate identifying C. sapidus from other Callinectes spp. HRM analysis of amplicons showed good discrimination among species, with distinct clusters formed between species with higher sequence homology. Linear discriminant analysis (LDA) classification of HRM curves was quite successful given the small dataset available, producing ~90-91% mean accuracy in classification over all species with 100-fold cross validation. Much of the error came from misclassifications between C. similis and C. danae, which are ~99% similar in sequence for the amplicon; collapsing them into a single class increased overall classification success to 94%. Error also arose from C. bocourti classifications, which had a reference set containing only three samples. Classification accuracy of C. sapidus alone via HRM was 97.5%. Overall, these assays show great promise as rapid and inexpensive methods to identify Callinectes spp. and have application for both ecological research and seafood identification or labeling.

Crustaceans permeate every habitat on Earth but are especially impactful in the marine environment. They can be small and extremely abundant like the ubiquitous marine copepods found throughout the world's oceans, or large and highly prized by fishermen like spiny lobsters found in tropical and temperate seas, globally. The latter are among the decapod crustaceans, a group which includes crabs, shrimps, and lobsters - those targeted most commonly by fishery and aquaculture industries. Hence, crustaceans are ecologically important, but they are also directly linked to the economic and nutritional health of human populations. To most effectively manage and conserve crustacean populations in the face of a changing ocean environment, whether they are harvested or not, requires a One Health approach that underscores the linkages between crustacean, human, and environmental health. Here, we give an overview of the need, benefits, and challenges to taking the One Health approach to crustacean health and argue that when viewed through the One Health lens, there is perhaps no other group of marine animals more worthy of that perspective.

Genetic tools can have a key role in informing conservation management of declining populations. Genetic diversity is an important determinant of population fitness and resilience, and can require careful management to ensure sufficient variation is present. In addition, population genetics data reveal patterns of connectivity and gene flow between locations, enabling mangers to predict recovery and resilience, identify areas of local adaptation, and generate restoration plans. Here, we demonstrate a conservation genetics approach to inform restoration and management of the loggerhead sponge (Spheciospongia vesparium) in the Florida Keys, USA. This species is a dominant, habitat-forming component of marine ecosystems in the Caribbean region, but in Florida has suffered numerous mass mortality events. We developed microsatellite markers and used them to genotype sponges from 14 locations in Florida and a site each in The Bahamas, Belize and Barbuda. We found that genetic diversity levels were similar across all sites, but inbreeding and bottleneck signatures were present in Florida. Populations are highly structured at the regional scale, whilst within Florida connectivity is present in a weak isolation by distance pattern, coupled with chaotic genetic patchiness. Evidence of a weak barrier to gene flow was found in Florida among sites situated on opposite sides of the islands in the Middle Keys. Loggerhead sponge populations in Florida are vulnerable in the face of mass mortalities due to low connectivity with other areas in the region, as well as distance-limited and unpredictable local connectivity patterns. However, our discovery of Florida’s high genetic diversity increases hope for resilience to future perturbations. These results provide valuable insight for sponge restoration practice in Florida.

Humans pollute the marine environment biologically, chemically, and physically, which can potentially drive or facilitate the emergence, proliferation, or impact of disease. This chapter synthesizes what is known about the effects of biological (e.g., wastewater), chemical (e.g., pharmaceuticals), and physical (e.g., sound/light) pollution on marine disease dynamics. The presence of these pollutants has been found to alter disease prevalence, increase host susceptibility to infection, and alter the spread and host range of different diseases. Despite the importance of the marine environment as a primary food source for humans, many complexities linking disease ecology and pollution are yet to be explored. Future investigation of these connections would benefit from an integrated approach using experimental, environmental, molecular, and pathological methods.

Infectious marine diseases have profound impacts on fisheries and aquaculture through their effects on growth, fecundity, mortality, and marketability. Economic losses have motivated research to minimize the negative impacts of disease on these industries. However, this relationship is reciprocal, as fishing and aquaculture can shape disease transmission. The effects of fisheries and aquaculture on disease are scale dependent, with different outcomes at the population, metapopulation, community, and ecosystem levels. Management approaches are limited in fisheries, and intense in aquaculture, sometimes with undesirable impacts on wild species. Management needs can be particularly intense in hatcheries, where stocks are sensitive and kept at high densities. Increased interest in microbiome–disease interactions are opening up new opportunities to manage marine diseases in aquaculture. Solutions for marine diseases in fisheries and aquaculture may ultimately improve human health by reducing exposure to pathogens and increasing nutrient quality, but could negatively impact human health through exposure to antibiotics and other chemicals used to treat parasites.

The Caribbean spiny lobster Panulirus argus supports important fisheries throughout the greater Caribbean and is also the only known host for the pathogenic virus Panulirus argus virus 1 (PaV1). While discovered nearly 2 decades ago, gaps still exist in our knowledge of PaV1, such as the dose required to establish infection and its viability outside of the host. To help answer such questions and to enhance diagnostic capabilities, we developed a TaqMan real-time quantitative polymerase chain reaction (qPCR) assay for PaV1. Of the advantages offered by qPCR, one of the most important benefits is its ability to accurately quantify viral DNA copies in a clinical sample. The qPCR assay was found to be efficient (mean ± SD: 99.19 ± 4.67%) and sensitive, detecting as few as 10 copies of PaV1 plasmid DNA. Its diagnostic sensitivity and specificity determined using a set of 165 lobster samples (138 from Florida, USA, and 27 from across the Caribbean) were 100 and 84%, respectively. The qPCR assay should thus prove useful as a research tool and for detecting and quantifying PaV1 infection severity in Caribbean spiny lobsters.

For more than 100 years, the Florida Keys (USA) have supported a commercial sponge fishery but there is little information about the small artisanal fishery that now exists, which is nonetheless controversial because of concerns about the ecological consequences of sponge harvest. We estimated the harvest of commercial sponges and bycatch (sublegal or non-commercial species), as well as the mortality, growth, and reproduction of commercial sponge species. In heavily fished areas, ∼33% of the legal-sized sponges, ∼3% of the sublegal sponges, and virtually none of the non-commercial species were harvested. Approximately 40% of our study area was never fished during the four month long peak in the fishing season. Self-reporting of harvest by fishers who participated in our study closely matched our fishery-independent estimates. Natural mortality of sublegal-sized sponges was ∼7% of the population/yr, with little difference among species. Discarded “roller” sponges grew at rates comparable to attached sponges and many reattached to the seafloor within 18 mos, although rates of reattachment varied among species. Growth also differed among species and sampling periods (mean = 3 cm dia/yr), and reproductive effort was positively related to sponge size in some species, but not others. Given the careful targeting of commercial sponge species and sizes by fishers and the small fraction of the sponge community that is commercially valuable, harvest is estimated to have minimal impact on the diverse assemblage of sponges in the region.

Interspecific competition is assumed to have a strong influence on the population dynamics of competing species, but is not easily demonstrated for mobile species in the wild. In the Florida Keys (USA), anecdotal observations have long pointed to an inverse relationship in abundance of two large decapod crustaceans found co-occurring in hard-bottom habitat, the stone crab Menippe mercenaria and the Caribbean spiny lobster Panulirus argus. We used them to explicitly test whether competition for a renewable resource (shelter) can drive the abundance and distribution of the inferior competitor. We first explored this relationship in shelter competition mesocosm experiments to determine the competitively dominant species. Results showed that stone crabs are clearly the dominant competitors regardless of the number of lobsters present, the presence of co-sheltering species such as the spider crab, Damithrax spinosissimus, or the order of introduction of competitors into the mesocosm. We also found that lobsters use chemical cues from stone crabs to detect and avoid them. We then tested the ramifications of this competitive dominance in the field by manipulating stone crab abundance and then tracking the abundance and distribution of spiny lobsters through time. Increased stone crab abundance immediately resulted in decreased lobster abundance and increased aggregation. The opposite occurred on sites where stone crabs were removed. When we stopped removing stone crabs from these sites, they soon returned and lobster abundance decreased. This study explicitly demonstrated that interspecific competition can drive population dynamics between these species, and ultimately, community composition in these shallow water habitats.

Recent genomic and metagenomic studies have led to a dramatic expansion of the known diversity of nucleocytoplasmic large DNA viruses (NCLDVs) of eukaryotes, which include giant viruses of protists and important pathogens of vertebrates, such as poxviruses. However, the characterization of viruses from nonmodel hosts still lags behind. We sequenced the complete genomes of three viruses infecting crustaceans, the Caribbean spiny lobster, demon shrimp, and European shore crab. These viruses have the smallest genomes among the known NCLDVs, with losses of many core genes, some of which are shared with iridoviruses. The deterioration of the transcription apparatus is compatible with microscopic and ultrastructural observations indicating that these viruses replicate in the nucleus of infected cells rather than in the cytoplasm. Phylogenomic analysis indicates that these viruses are sufficiently distinct from all other NCLDVs to justify the creation of a separate family, for which we propose the name “Mininucleoviridae” (i.e., small viruses reproducing in the cell nucleus)., Panulirus argus virus 1 (PaV1) is the only known virus infecting the Caribbean spiny lobster (Panulirus argus) from the Caribbean Sea. Recently, related viruses, Dikerogammarus haemobaphes virus 1 (DhV1) and Carcinus maenas virus 1 (CmV1), have been detected in the demon shrimp (Dikerogammarus haemobaphes) and the European shore crab (Carcinus maenas), respectively, from sites in the United Kingdom. The virion morphology of these crustacean viruses is similar to that of iridoviruses. However, unlike iridoviruses and other nucleocytoplasmic large DNA viruses (NCLDVs), these viruses complete their morphogenesis in the host cell nucleus rather than in the cytoplasm. To date, these crustacean viruses have remained unclassified due to a lack of genomic data. Using an Illumina MiSeq sequencer, we sequenced the complete genomes of PaV1, CmV1, and DhV1. Comparative genome analysis shows that these crustacean virus genomes encode the 10 hallmark proteins previously described for the NCLDVs of eukaryotes, strongly suggesting that they are members of this group. With a size range of 70 to 74 kb, these are the smallest NCLDV genomes identified to date. Extensive gene loss, divergence of gene sequences, and the accumulation of low-complexity sequences reflect the extreme degradation of the genomes of these “minimal” NCLDVs rather than any direct relationship with the NCLDV ancestor. Phylogenomic analysis supports the classification of these crustacean viruses as a distinct family, “Mininucleoviridae,” within the pitho-irido-Marseille branch of the NCLDVs.

A single-stranded DNA (ssDNA) virus is presented from a metagenomic data set derived from Alphaproteobacteria -infected hepatopancreatic tissues of the crab Eurypanopeus depressus . The circular virus genome (4,768 bp) encodes 14 hypothetical proteins, some similar to other bacteriophages ( Microviridae ). Based on its relatedness to other Microviridae , this virus represents a member of a novel genus.

Crayfish are a vital ecological asset in their native range but can be highly damaging as invasive species. Knowledge of their diseases, including high levels of research on Aphanomyces astaci (crayfish plague), show that disease plays a vital role during crayfish invasions. Microsporidian diseases in crayfish are less studied but are thought to have important links to crayfish health and invasion dynamics. In this study we provide a systematic description of a novel microsporidian parasite from the Floridian crayfish, Procambarus paeninsulanus, with additional genetic identification from related Microsporidia from Procambarus fallax, Cambarellus shufeldtii and Cambarellus blacki. This novel microsporidium from P. paeninsulanus is described in a new genus, Cambaraspora, and species, Cambaraspora floridanus, and represents a novel member of the Clade V Microsporidia within the Glugeidae. The parasite develops in the muscle tissue of P. paeninsulanus, within a sporophorous vesicle, and produces a spore with 19–21 turns of the polar filament measuring 6.136 ± 0.84 µm in length and 2.12 ± 0.23 µm in width. The muscle-infecting nature of the parasite suggests that it is horizontally transmitted. Genetic data for the 18S of the parasite from all hosts confirms its assignment to Clade V and reveal it to be a relative of multiple fish-infecting parasites. It shows closest genetic relationship to Glugea plecoglossi, but branches alongside multiple microsporidia from fish, crustaceans and eDNA isolates. The information presented here suggests that this novel parasite may have the potential to infect piscine hosts and is a likely mortality driver in the P. paeninsulanus population. Its potential as a control agent or wildlife disease invasion threat is explored, as well as the placement of this novel microsporidium within the Glugeidae.

Florida Bay is home to a network of shallow mud-banks which act as barriers to circulation creating small basins that are often subject to extremes in temperature and salinity. Florida bay is also important juvenile habitat for the Caribbean spiny lobster Panulirus argus. While our understanding of the effect of environmental changes on the survival, growth, and movement of spiny lobsters is growing, the effect on their chemosensory abilities has not yet been investigated. Lobsters rely heavily on chemical cues for many biological and ecological activities, and here we report on the effect of extreme environmental events in temperature (32 °C), salinity (45ppt), and pH (7.65 pH) on social behavior and sheltering preference in P. argus. Under normal conditions, chemical cues from conspecifics are used by spiny lobsters to identify suitable shelter and cues from stone crabs and diseased individuals are used to determine shelters to be avoided. In all altered conditions, lobsters lost the ability to aggregate with conspecifics and avoid stone crabs and diseased conspecifics. Thus, seasonal extreme events, and potentially future climate change conditions, alter the chemosensory-driven behavior of P. argus and may result in decreased survivorship due to impaired shelter selection or other behaviors.

For most marine organisms, dispersal occurs primarily during a pelagic larval phase that connects populations. We paired population structure from comprehensive genetic sampling and biophysical larval transport modeling to describe how spiny lobster (Panulirus argus) population differentiation is related to biological oceanography. A total of 581 lobsters were genotyped with 11 microsatellites from ten locations around the greater Caribbean. The overall FST of 0.0016 (P = 0.005) suggested low yet significant levels of structuring among sites. An isolation by geographic distance model did not explain spatial patterns of genetic differentiation in P. argus (P = 0.19; Mantel r = 0.18), whereas a biophysical connectivity model provided a significant explanation of population differentiation (P = 0.04; Mantel r = 0.47). Thus, even for a widely dispersing species, dispersal occurs over a continuum where basin-wide larval retention creates genetic structure. Our study provides a framework for future explorations of wide-scale larval dispersal and marine connectivity by integrating empirical genetic research and probabilistic modeling.

A positive, single-stranded RNA virus is identified from the transcriptome of Probopyrinella latreuticola Gissler, 1882; a bopyrid isopod parasite of the Sargassum shrimp, Latreutes fucorum Fabricius, 1789. The viral sequence is 13,098 bp in length (including polyA), encoding four open reading frames (ORF). ORF-1 encodes a polyprotein, with three computationally discernible functional domains: viral methyltransferase; viral helicase; and RNA-directed RNA polymerase. The remaining ORFs encode a transmembrane protein, a capsid protein and a protein of undetermined function. The raw transcriptomic data reveal a low level of background single nucleotide mutations within the data. Comparison of the protein sequence data and synteny with other viral isolates reveals that the greatest protein similarity (39%) is shared with the Negevirus group, a group that exclusively infects insects. Phylogenetic assessment of the individual polyprotein domains revealed a mixed prediction of phylogenetic origins, suggesting with low confidence that the novel +ssRNA virus could be present in multiple places throughout the individual gene trees. A concatenated approach strongly suggested that this new virus is an early diverging isolate, branching before the Negevirus and Cilevirus groups. Alongside the new isolate are other marine viruses, also present toward the base of the tree. The isopod virosphere, with the addition of this novel virus, is discussed relative to viral genomics/systematics. A great diversity of nege-like viruses appears to be present in marine invertebrate hosts, which require greater efforts for discovery and identification.

Knowledge of marine host–parasite relationships has been increasing, but little is known about the reproductive strategies of the parasites. Theoretically, parasitic crustaceans inhabiting relatively small and scarce hosts should be monogamous. Parasites are also expected to exhibit higher reproductive performance (RP) than their free-living relatives, a strategy thought to have evolved to increase host-to-host transmission, or to be the outcome of living in a ‘stable, nutrient-rich environment’. Here, the parasitic isopod Parabopyrella lata and its shrimp host Lysmata boggessi were used to test for monogamy and augmented RP, and to examine whether or not P. lata castrates its host. Prevalence, population distribution, and reproductive performance were examined in P. lata over 1 year (2012–2013) off Homosassa Springs, Florida USA (28.8037° N, 82.5761° W). Prevalence of P. lata on L. boggessi was greater during the warmer months of the year and infested shrimps were almost exclusively males. P. lata lives in male–female pairs within the gill chamber of infested shrimps more frequently than expected by chance alone. The sizes of paired males and females were tightly and positively correlated with each other and with host size. Average fecundity (3660 ± 1146 eggs female−1) and reproductive output (61 ± 19%) in P. lata, as with other bopyrid parasites, was much greater than those reported for free-living isopods. This evidence supports the hypotheses that parasites such as P. lata are adapted to be monogamous, display augmented RP, and reproductively castrate their host.

Casitas are artificial shelters used by fishers to aggregate Caribbean spiny lobsters (Panulirus argus) for ease of capture. However, casitas may function as an ecological trap for juvenile lobsters if they are attracted to casitas and their growth or mortality is poorer compared with natural shelters. We hypothesized that juvenile lobsters may be at particular risk if attracted to casitas because they are less able than larger individuals to defend themselves, and do not forage far from shelter. We compared the nutritional condition, relative mortality, and activity of lobsters of various sizes in casitas and natural shelters in adult and juvenile lobster-dominated habitats in the Florida Keys (United States). We found that the ecological effects of casitas are complex and location-dependent. Lobsters collected from casitas and natural shelters did not differ in nutritional condition. However, juvenile lobsters in casitas experienced higher rates of mortality than did individuals in natural shelters; the mortality of large lobsters did not differ between casitas and natural shelters. Thus, casitas only function as ecological traps when deployed in nursery habitats where juvenile lobsters are lured by conspecifics to casitas where their risk of predation is higher. These results highlight the importance of accounting for animal size and location-dependent effects when considering the consequences of habitat modification for fisheries enhancement.

Ovipleistophora diplostomuri (Microsporidia) is an obligate parasite of fish and trematodes in the US. In April 2019, an individual crayfish, Procambarus bivittatus (Escambia River, Florida), with a high-intensity microsporidian infection was delivered to the Emerging Pathogens Institute. Histological analysis determined that infection was restricted to the muscle tissue. Molecular diagnostics (PCR) provided 952 bp of the parasite SSU (18S) sequence. The isolate was 99.16% similar to O. diplostomuri identified from blue gill and their trematode parasites in Washington, USA. This discovery increases our understanding of Microsporidia within aquatic trophic networks, supporting the theory that the Ovipleistophora share complex relationships with vertebrates, invertebrates and helminth parasites.

The PaV1 virus infects spiny lobsters (Panulirus argus) throughout most of the Caribbean, where its prevalence in adult lobsters can reach 17% and where it poses a significant risk of mortality for juveniles. Recent studies indicate that vertical transmission of the virus is unlikely and PaV1 has not been identified in the phyllosoma larval stages. Yet, the pathogen appears subclinically in post-larvae collected near the coast, suggesting that lobster post-larvae may harbour the virus and perhaps have aided in the dispersal of the pathogen. Laboratory and field experiments also confirm the waterborne transmission of the virus to post-larval and early benthic juvenile stages, but its viability in the water column may be limited to a few days. Here, we coupled Lagrangian modelling with a flexible matrix model of waterborne and post-larval-based pathogen dispersal in the Caribbean to investigate how a large area with complex hydrology influences the theoretical spread of disease. Our results indicate that if the virus is waterborne and only viable for a few days, then it is unlikely to impact both the Eastern and Northwestern Caribbean, which are separated by dispersal barriers. However, if PaV1 can be transported between locations by infected post-larvae, then the entire Caribbean becomes linked by pathogen dispersal with higher viral prevalence in the North. We identify possible regions from which pathogens are most likely to spread, and highlight Caribbean locations that function as dispersal “gateways” that could facilitate the rapid spread of pathogens into otherwise isolated areas.
