Abstract Background Aging is associated with significant structural and functional changes in the spleen, leading to immunosenescence, yet the detailed effects on splenic vascular endothelial cells (ECs) and their immunomodulatory roles are not fully understood. In this study, a single-cell RNA (scRNA) atlas of EC transcriptomes from young and aged mouse spleens was constructed to reveal age-related molecular changes, including increased inflammation and reduced vascular development and also the potential interaction between splenic endothelial cells and immune cells. Results Ten clusters of splenic endothelial cells were identified. DEGs analysis across different EC clusters revealed the molecular changes with aging, showing the increase in the overall inflammatory microenvironment and the loss in vascular development function of aged ECs. Notably, four EC clusters with immunological functions were identified, suggesting an Endothelial-to-Immune-like Cell Transition (EndICLT) potentially driven by aging. Pseudotime analysis of the Immunology4 cluster further indicated a possible aging-induced transitional state, potentially initiated by Ctss gene activation. Finally, the effects of aging on cell signaling communication between different EC clusters and immune cells were analyzed. Conclusions This comprehensive atlas elucidates the complex interplay between ECs and immune cells in the aging spleen, offering new insights into endothelial heterogeneity, reprogramming, and the mechanisms of immunosenescence.

Abstract Exposure to cigarette smoke directly damages the lungs and causes lung inflammation. The anti‐inflammatory properties of edible bird's nest (EBN) have been reported. We aimed to determine the effect of EBN on pneumonia in a mouse model exposed to cigarette smoke. Fifty BALB/c mice were randomly divided into control, model, positive drug, low‐dose EBN, and high‐dose EBN groups (n = 10 each). Except for the control group, the mice in each group were exposed to four cigarettes once a day for 8 days. In addition, we validated the effects of EBN on A549 cells and investigated the mechanism by which EBN alleviates lung inflammation. Edible bird's nest (EBN) could alleviate the structural damage of lung tissue and the smoke‐induced inflammatory response in mice. The best effect was observed at the high dose of EBN (0.019 g). The mice treated with EBN had a stronger ability than those in the model group to resist cigarette smoke stimulation, as indicated by a decrease in serum and lung inflammatory markers (interleukin 6 [IL‐6], tumor necrosis factor‐α [TNF‐α], and interleukin 8 [IL‐8]), an increase in serum interleukin 10 (IL‐10) levels, and a decrease in the expression of inflammasome NOD‐like receptor pyrin 3 (NLRP3). In addition, our cell experiments showed that EBN attenuated cigarette smoke‐induced pulmonary inflammation mainly by inhibiting the tumor necrosis factor receptor 1 (TNFR1)/nuclear factor‐kappa B (NF‐κB)/NLRP3 pathway. These findings provide theoretical evidence for the positive nutritional qualities of EBN for the lung by demonstrating that it inhibits the TNFR1/NF‐κB/NLRP3 signaling pathway, which prevents the development of cigarette smoke‐induced pulmonary inflammation.

Geochemistry of noble gases has been applied to the study of deep Earth materials and celestial meteorites. With advancements in measurement technology, it is also increasingly being used in natural gas research. Using the most advanced noble gas mass spectrometer, a systematic analysis was conducted on the abundance and isotopes of noble gas samples collected from the middle and deep Songliao Basin in eastern China to elucidate the compositional characteristics of noble gases in a faulted basin. The findings reveal that the abundance of noble gases in natural gas decreases from light noble gases to heavy noble gases in a left-to-right reversed L-shaped pattern. Specifically, the isotopic ratios of noble gases are: 3He/4He at 2.64×10-6, 20Ne/22Ne at 9.94, 21Ne/22Ne at 0.029 22, and 40Ar/36Ar at 743.7, all of which are higher than atmospheric values. The isotopes 80Kr, 84Kr, 86Kr, and 131-136Xe show relative excess compared to atmospheric levels, indicating the mixing of mantle-derived gases in the natural gas. Based on the compositional characteristics of noble gases, this study suggests that the natural gas in the middle and deep Songliao Basin originated from inorganic crust and mantle mixing. Additionally, certain differences exist in the contribution of mantle-derived components across different tectonic blocks and categories of natural gas within the basin. Comparison between noble gas compositions and the types of natural gas demonstrates that light noble gases distinguish effectively between different types of natural gas, whereas the composition of heavy noble gases shows no significant differences among various types of natural gas. Noble gas isotopes, in addition to tracing the source of natural gas, can also be used to determine natural gas genesis, distinguish natural gas types and characterize tectonic settings.

Abstract It is a challenging and meaningful task to carry out UAV-based livestock monitoring in high-altitude (more than 4500 m on average) and cold regions (annual average – 4 °C) on the Qinghai Tibet Plateau. The purpose of artificial intelligence (AI) is to execute automated tasks and to solve practical problems in actual applications by combining the software technology with the hardware carrier to create integrated advanced devices. Only in this way, the maximum value of AI could be realized. In this paper, a real-time tracking system with dynamic target tracking ability is proposed. It is developed based on the tracking-by-detection architecture using YOLOv7 and Deep SORT algorithms for target detection and tracking, respectively. In response to the problems encountered in the tracking process of complex and dense scenes, our work (1) Uses optical flow to compensate the Kalman filter, to solve the problem of mismatch between the target bounding box predicted by the Kalman filter (KF) and the input when the target detection in the current frame is complex, thereby improving the prediction accuracy; (2) Using a low confidence trajectory filtering method to reduce false positive trajectories generated by Deep SORT, thereby mitigating the impact of unreliable detection on target tracking. (3) A visual servo controller has been designed for the Unmanned Aerial Vehicle (UAV) to reduce the impact of rapid movement on tracking and ensure that the target is always within the field of view of the UAV camera, thereby achieving automatic tracking tasks. Finally, the system was tested using Tibetan yaks on the Qinghai Tibet Plateau as tracking targets, and the results showed that the system has real-time multi tracking ability and ideal visual servo effect in complex and dense scenes.

This study aimed to investigate the immune regulation effect of edible bird’s nest (EBN) in vivo and elucidate the influence of EBN on immune microbiome, which is the basis of the activity of EBN. The immunosuppression mice model was established by subcutaneous injection of hydrocortisone (HC). The optimal modeling dose of HC was screened by various immune indexes. Studies have found that the mice immunosuppression model can be constructed with a HC dose of 12.5 mg/kg. The effects of EBN in low (0.285 g/kg) and high doses (0.649 g/kg) were investigated on the immunosuppression mice model. EBN low-dose treatment significantly improved macrophage phagocytosis and NK cell activity in immunosuppression mice. EBN high-dose treatment significantly increased the serum TNF-α content phagocytosis ability of macrophages, NK cell activity and serum hemolysin content of immunosuppression mice. It was found that changes in the intestinal flora are usually accompanied by changes in immune cells and inflammatory factors, so it was hypothesized that there is a correlation between changes in intestinal flora and immune regulation, therefore the changes in the intestinal flora of mice were then investigated after EBN administration. The regulation of intestinal flora experiments showed that species richness decreased in the immunosuppression mice, and after EBN treatment the species richness in mice increased. Additionally, after continuous administration of EBN, the abundance of Bacteroides, Akkermansiaceae and other intestinal probiotics increased, while the abundance of Firmicutes decreased, indicating that EBN can regulate intestinal dysfunction, up-regulate beneficial bacteria. It is reported that the increase of Bacteroides and Akkermansiaceae and the decrease of Firmicutes in the intestinal flora caused an increase in the levels of TNF-α and improved immunity, which is consistent with our experimental results. Therefore, it is hypothesized that EBN plays an immune regulation role mainly by affecting the changes in the intestinal flora. The study provides a theoretical basis for the functional investigation of EBN.

Abstract Intercellular communication often relies on exosomes as messengers and is critical for cancer metastasis in hypoxic tumor microenvironment. Some circular RNAs (circRNAs) are enriched in cancer cell‐derived exosomes, but little is known about their ability to regulate intercellular communication and cancer metastasis. Here, by systematically analyzing exosomes secreted by non‐small cell lung cancer (NSCLC) cells, a hypoxia‐induced exosomal circPLEKHM1 is identified that drives NSCLC metastasis through polarizing macrophages toward to M2 type. Mechanistically, exosomal circPLEKHM1 promoted PABPC1‐eIF4G interaction to facilitate the translation of the oncostatin M receptor (OSMR), thereby promoting macrophage polarization for cancer metastasis. Importantly, circPLEKHM1‐targeted therapy significantly reduces NSCLC metastasis in vivo. circPLEKHM1 serves as a prognostic biomarker for metastasis and poor survival in NSCLC patients. This study unveils a new circRNA‐mediated mechanism underlying how cancer cells crosstalk with macrophages within the hypoxic tumor microenvironment to promote metastasis, highlighting the importance of exosomal circPLEKHM1 as a prognostic biomarker and therapeutic target for lung cancer metastasis.

Climate change and human activities have severely impacted Central Asia's mountainous grasslands' health status, making monitoring aboveground biomass (AGB) crucial for grassland protection. However, a high-resolution and low-cost solution for AGB monitoring is lacking for Central Asia's grassland. This research proposes an unmanned aerial vehicle (UAV) based AGB monitoring framework using consumer-level cameras. Texture features from UAV RGB images and vegetation indices from UAV multispectral images are used to predict AGB. As one of the typical mountainous countries in Central Asia, Tajikistan is chosen as the study area. Four different grassland types were chosen to investigate the performance of applying UAV for AGB monitoring. Firstly, correlation analysis was performed to identify important features for AGB estimation. Subsequently, the ground-measured AGB and UAV image features were utilized in multiple linear regression (MLR) and generalized additive model (GAM) to develop an AGB prediction model. The results show that the angular second moment (ASM) of the green band and green normalized difference vegetation index (GNDVI) are the two most important features for AGB estimation in the study area. Furthermore, the GAM-based model demonstrated higher accuracy (R2 = 0.87, RMSE = 127.53 g/m2, rRMSE = 0.17) compared to MLR (R2 = 0.74, RMSE = 181.68 g/m2, rRMSE = 0.24), highlighting the nonlinear relationship between AGB and UAV image features. This research uses multispectral and RGB images to achieve a high accuracy and efficient AGB monitoring framework for Central Asia grasslands and provides a reference to validate satellite images for providing long-term and large-scale monitoring of grassland ecosystems and reasonable utilization of grassland resources.

Through exploring the stress corrosion cracking (SCC) behaviors of the as-cast Mg-8%Li and Mg-8%Li-6%Zn-1.2%Y alloys in a 0.1 M NaCl solution, it revealed that the SCC susceptibility index (ISCC) of the Mg-8%Li alloy was 47%, whilst the ISCC of the Mg-8%Li-6%Zn-1.2%Y alloy was 68%. Surface, cross-sectional and fractography observations indicated that for the Mg-8%Li alloy, the α-Mg/β-Li interfaces acted as the preferential crack initiation sites and propagation paths during the SCC process. With regard to the Mg-8%Li-6%Zn-1.2%Y alloy, the crack initiation sites included the I-phase and the interfaces of I-phase/β-Li and α-Mg/β-Li, and the preferential propagation paths were the I-phase/β-Li and α-Mg/β-Li interfaces. Moreover, the SCC of the two alloys was concerned with hydrogen embrittlement (HE) mechanism.

Maternal diet is indispensable for the proper development of offspring, and the establishment of a well-functioning intestinal barrier is crucial for normal growth and development. Edible bird’s nest (EBN) has positive impacts on intestinal health, but the underlying mechanisms are unknown. Our study showed that maternal EBN supplementation downregulated the serum levels of diamine oxidase and D-lactate, elevated expression of tight junction proteins ZO-1, Occludin, and Claudin-1, and regulated the intestinal microbiota in rat offspring. Spearman’s correlation analysis revealed that Claudin-1, transforming growth factor-β (TGF-β), and transcription factor RUNX3 were positively correlated with changes in intestinal bacteria. EBN may upregulate the expression of TGF-β by modulating the intestinal microbiota composition, enhancing RUNX3 binding to the Claudin-1 promoter region to upregulate its expression. In conclusion, we demonstrated that maternal EBN supplementation during gestation and lactation enhanced intestinal barrier function by upregulating Claudin-1 in rat offspring.

Purpose: In cluster headache, patients often describe a ramp up period with initially milder headache and then build up to a persistent and severe pattern. It is possible that within the early ramp up, patients may be more treatment responsive. We propose to study if response to therapy for prevention of cluster headache is dependent on timing from onset of cluster cycle. Methods: We will conduct a prospective observational cohort study, where we examine the relationship between timing of start of preventive medication and likelihood of response to treatment. We will look at individuals exposed within 10 days of onset of cluster cycle, versus those exposed after 10 days, to preventive medication. Results: We plan to enroll patients into the study and present data on completion. Conclusion: We aim to study the relationship between cluster headache onset and response to therapy. We propose that there is some data to suggest that earlier in cycle cluster headache may be more easily prevented, and we aim to study this relationship systematically. This is an important question that can influence treatment decisions in clinical practice, as well as clinical trial design.

Shiitake mushrooms are renowned for their popularity and robust nutritional value, are susceptible to spoilage due to their inherent biodegradability. Nevertheless, because of their lack of protection, these mushrooms have a short shelf life. Throughout the post-harvest phase, mushrooms experience a persistent decline in quality. This is evidenced by changes such as discoloration, reduced moisture content, texture changes, an increase in microbial count, and the depletion of nutrients and flavor. Ensuring postharvest quality preservation and prolonging mushroom shelf life necessitates the utilization of post-harvest preservation techniques, including physical, chemical, and thermal processes. This review provides a comprehensive overview of the deterioration processes affecting mushroom quality, covering elements such as moisture loss, discoloration, texture alterations, increased microbial count, and the depletion of nutrients and flavor. It also explores the key factors influencing these processes, such as temperature, relative humidity, water activity, and respiration rate. Furthermore, the review delves into recent progress in preserving mushrooms through techniques such as drying, cooling, packaging, irradiation, washing, and coating.

Summary: Patients with Graves’ disease (GD) can develop Graves’ ophthalmopathy (GO), but the underlying pathological mechanisms driving this development remain unclear. In our study, which included patients with GD and GO, we utilized single-cell RNA sequencing (scRNA-seq) and multiplatform analyses to investigate CD169+ classical monocytes, which secrete proinflammatory cytokines and are expanded through activated interferon signaling. We found that CD169+ clas_mono was clinically significant in predicting GO progression and prognosis, and differentiated into CD169+ macrophages that promote inflammation, adipogenesis, and fibrosis. Our murine model of early-stage GO showed that CD169+ classical monocytes accumulated in orbital tissue via the Cxcl12-Cxcr4 axis. Further studies are needed to investigate whether targeting circulating monocytes and the Cxcl12-Cxcr4 axis could alleviate GO progression.

House edible bird's nest and cave edible bird's nest are two main types of EBN. However, the key differential components, which can cause the differences of these EBN are not clear. In order to explore the differences between house EBN and cave EBN and effectively discriminate between different EBN samples, protein, amino acid, sialic acid, mineral elements and other nutritional components, as well as the hazard factors such as nitrite and heavy metal elements in four kinds of house EBN and cave EBN were determined, respectively. Moreover, cluster analysis and key differential components screening were carried out using heatmap and partial least squares regression (PLSR), respectively. The results showed that protein and sialic acid were the most important nutritional components, accounting for 63.09%~67.79% and 7.22%~10.41% in the total EBN, respectively. Leucine, valine, threonine and phenylalanine were the main essential amino acids in EBN, while serine, aspartic acid and proline were the main non-essential amino acids. In addition, the proportion of essential amino acids in total amino acids (45.76%~47.60%) was higher than 40% of the standard according to the high-quality protein recommendation from FAO/WHO. Abstract Background Andrographolide (Andro) is a diterpenoid derived from Andrographis paniculate, which has anti-inflammatory, antibacterial, antiviral and hepatoprotective activities. Gram-negative bacterial infections can cause varying degrees of liver injury in chickens, although Andro has been shown to have a protective effect on the liver, its underlying mechanism of action and effects on liver proteins are not known. Methods The toxicity of Andro on the viability of leghorn male hepatoma (LMH) cells at different concentrations and times was analyzed by CCK-8 assays. Alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities in the culture supernatants were measured using an automatic biochemical analyzer to evaluate the protective effect of androscopolide on LPS-induced injury of LMH cells. Subsequently, TMT proteomics analysis were performed on the negative control group (NC group), LPS, and LPS-Andro groups, and bioinformatics analysis was performed on the differentially expressed proteins (DEPs). Results It was found that Andro reduced ALT and AST levels in the cell supernatant and alleviated LPS-induced injury in LMH cells. Proteomic analysis identified 50 and 166 differentially expressed proteins in the LPS vs. NC group and LPS-Andro vs. LPS group, respectively. Andro may be involved in steroid metabolic processes, negative regulation of MAPK cascade, oxidative stress, and other processes to protect against LPS-induced liver injury. Conclusions Andro protects against LPS-induced liver injury, HMGCS1, HMGCR, FDPS, PBK, CAV1, PRDX1, PRDX4, and PRDX6, which were identified by differential proteomics, may be the targets of Andro. Our study may provide new theoretical support for Andro protection against liver injury.

Transmissible gastroenteritis virus (TGEV) is an etiological agent of enteric disease that results in high mortality rates in piglets. The economic impact of the virus is considerable, causing significant losses to the pig industry. The development of an efficacious subunit vaccine to provide promising protection against TGEV is of the utmost importance. The viral antigen, spike glycoprotein (S), is widely regarded as one of the most effective antigenic components for vaccine research. In this study, we employed immunoinformatics and molecular dynamics approaches to develop an ‘ideal’ multi-epitope vaccine. Firstly, the dominant, non-toxic, highly antigenic T (Th, CTL) and B cell epitopes predicted from the TGEV S protein were artificially engineered in tandem to design candidate subunit vaccines. Molecular docking and dynamic simulation results demonstrate that it exhibits robust interactions with toll-like receptor 4 (TLR4). Of particular significance was the finding that the vaccine was capable of triggering an immune response in mammals, as evidenced by the immune simulation results. The humoral aspect is typified by elevated levels of IgG and IgM, whereas the cellular immune aspect is capable of eliciting the robust production of interleukins and cytokines (IFN-γ and IL-2). Furthermore, the adoption of E. coli expression systems for the preparation of vaccines will also result in cost savings. This study offers logical guidelines for the development of a secure and efficacious subunit vaccine against TGEV, in addition to providing a novel theoretical foundation and strategy to prevent associated CoV infections.

Porcine parvovirus 7 (PPV7) was first discovered in swine in 2016, and PPV7 infection has been detected in aborted pig fetuses and in sows that experienced reproductive failure. The objective of this study was to report the prevalence and genetic characterization of PPV7 in Hunan, China. Seventy of the four hundred and twenty-two (16.6%) serum, semen, and tissue samples collected from pigs were positive for PPV7. One complete PPV7 strain and eighteen complete cap gene sequences were obtained; nucleotide and amino acid identity among the nineteen Cap sequences were 88.1–99.4% and 88.1–100%, respectively. They shared identity with previously discovered sequences ranging from 86.6 to 98.9% and 83.7 to 99.8% at the nucleotide- and amino acid-level, respectively. The phylogenetic tree analysis exhibited that PPV7 strains had two major groups based on the presence or absence of five amino acid (181–185) insertions on the Cap protein. Analysis of the Cap protein demonstrated that PPV7 Cap had significant variability, implying that PPV7 evolved at high substitution rates. Substantial variations of that PPV7 Cap may enable the emergence of newly mutated capsid profiles due to its viral adaptation to host responses. Furthermore, antigenic alteration owing to PPV7 Cap protein amino acid mutations at immune epitopes may enable viruses to escape from the host’s immune system. This study determined the prevalence and genetic characteristics of PPV7 circulating in swine in Hunan, China, and provided the impetus and basis to further investigate the pathogenicity and epidemiology of PPV7.

To tackle the complexity and limited applicability of high-precision segmentation models for damaged road markings, this study proposes a Multi-level Adaptive Lightweight Network (MALNet) based on knowledge distillation. By incorporating multi-scale dilated convolution and adaptive spatial channel attention fusion modules, the MALNet model significantly enhances the precision, integrity, and robustness of its segmentation branch. Furthermore, it employs an intricate knowledge distillation strategy, channeling rich, layered insights from a teacher model to a student model, thus elevating the latter’s segmentation ability. Concurrently, it streamlines the student model by markedly reducing its parameter count and computational demands, culminating in a segmentation network that is both high-performing and pragmatic. Rigorous testing on three distinct data sets for damaged road marking detection—CDM_P (Collective Damaged road Marking—Public), CDM_H (Collective Damaged road Marking—Highways), and CDM_C (Collective Damaged road Marking—Cityroad)—underscores the MALNet model’s superior segmentation abilities across all damage types, outperforming competing models in accuracy and completeness. Notably, the MALNet model excels in parameter efficiency, computational economy, and throughput. After distillation, the student model’s parameters and computational load decrease to only 31.78% and 27.40% of the teacher model’s, respectively, while processing speeds increase to 1.9 times, demonstrating a significant improvement in lightweight design.

Iron–chalcogenide superconductors continue to captivate researchers due to their diverse crystalline structures and intriguing superconducting properties, positioning them as both a valuable platform for theoretical investigations and promising candidates for practical applications. This review begins with a comprehensive overview of the fabrication techniques employed for various iron–chalcogenide superconductors, accompanied by a summary of their phase diagrams. Subsequently, it delves into the upper critical field, anisotropy, and critical current density. Furthermore, it discusses the successful fabrication of meters-long coated conductors and explores their applications in superconducting radio-frequency cavities and coils. Finally, several prospective avenues for future research are proposed.

Seeking a low-cost, high-efficiency greenhouse mapping technology has immense significance. While greenhouse extraction methods using deep learning have been proposed, the challenge of extracting dense small objects remains an unresolved problem. The inherent downscaling strategy in general-purpose semantic segmentation (SS) models renders them unsuitable for such tasks. In contrast, the dramatically increasing computational complexity associated with this problem may result in an unaffordable cost for consumer-level applications. To address the aforementioned challenges, this study presents a novel greenhouse mapping model based on remote sensing (RS) images, which not only exhibits high precision and robust generalization capabilities but also offers significant lightweight advantages. To meet broader needs, we also provide corresponding customizable and scalable rules that allow for a trade-off between accuracy and speed. To evaluate the performance of our model, we select several representative works to conduct benchmark experiments on a self-annotated dataset. The results demonstrate that our method can provide more powerful visual representations for greenhouse segmentation with minimal cost. Compared to the control group, the proposed method achieves an mIoU improvement of 1.116 %-10.77 % using only 3.282 M parameters, while maintaining a considerable inference speed. Abstract Background Stroke is a common and frequently-occurring disease in older people. It has the characteristics of high morbidity, high mortality, high recurrence rate and high disability rate. Most stroke risk studies are based on pathophysiology, however psychosocial factors such as diet quality are often understudied. The aim of this study was to assess stroke risk in urban community residents in Tianjin and investigate the factors that affect the dietary quality of older stroke high-risk populations. Methods Using a cross-sectional, multicenter study, recruit people aged 60 to 80 in Tianjin. Dietary intake data were obtained through a validated food frequency questionnaire, which were used to calculate Alternate Healthy Eating Index-2010 (AHEI-2010) and to analyze its association with sociodemographic characteristics, stroke risk factors and health marker variables. Results A total of 1068 participants from 4 community health service centers in Tianjin were recruited, including 300 low-risk individuals and 768 high-risk individuals. Compared with the low-risk group (62.75 ± 3.59), the AHEI-2010 mean score of the high-risk group (56.83 ± 6.54) was significantly lower. The top three most common risk factors among participants were dyslipidemia (80.3%), hypertension (60.6%), and physical inactivity (58.2%). Sewage treatment plants (STPs) are significant routes through which microplastics (MPs) are released into the aquatic environment. Constructed wetland is an effective facility for deep treatment of tailwater. At present, research on the removal of MPs in the tailwater of STPs by multi-stage constructed wetlands is limited. This work investigated and analyzed the removal characteristics of MPs in the tailwater treatment system of Cihu wetland park in Huangshi, Hubei Province of China. The abundance/removal of MPs in the Cihu Lake-wetland microcosm system was investigated. The results showed that the multi-stage constructed wetlands achieved a total removal rate of 94.7% for MPs with 2.2 particles/L MPs in the effluent. The removal rates of MPs reached 89 and 37.5%, respectively, in the (horizontal/vertical) subsurface flow constructed wetland and surface flow constructed wetland. The abundance of MPs in receiving water of Cihu Lake substantially decreased due to the dilution of wetland effluents. This study partially bridged the knowledge gap hypothesis on the treatment of MPs in tailwater by multi-stage constructed wetlands. The visual servoing of manipulators is challenged by two main problems: the singularity of the inverse Jacobian and the physical constraints of a manipulator. In order to overcome the singularity issue, this paper presents a novel approach for image-based visual servoing (IBVS), which converts the propagation of errors in the image plane into the conduction of virtual forces using the principle of virtual work. This approach eliminates the need for Jacobian inversion computations and prevents matrix inversion singularity. To tackle physical constraints, reverse thinking is adopted to derive the function of the upper and lower bounds of the joint velocity on the joint angle. This enables the proposed method to configure the physical constraints of the robot in a more intuitive manner. To validate the effectiveness of the proposed method, an eye-in-hand system based on UR5 in VREP, as well as a physical robot, were established.

The microstructural evolution and mechanical properties of twin-roll casting (TRC) and hot rolling (HR) Mg–3Al–1Bi (in wt.%, TRC-AB31 and HR-AB31) alloys were investigated and compared with AZ31 alloy as the benchmark. The addition of Bi in TRCed alloys can not only reduce the central segregation but also introduce thermally stable Mg3Bi2 phases distributed uniformly in the α-Mg matrix. The second phases with a larger size lead to particle stimulated nucleation (PSN), which in turn, promoted the dynamic recrystallization (DRX) in HR-AB31 alloy and weaken its basal texture. The second phases play two different roles in AB31 alloy, on the one hand, the grain refinement and uniform microstructure resulted from enhanced DRX by second particles which retarding dislocation movement as well as inhibition of grain growth. On the other hand, such brittle particles can also act as the cracking source and degrade elongation. The strengthening mechanism of both AZ31 and AB 31 alloys is discussed and investigated.

A single-pass gradient caliber rolling of a conical-shaped AZ31 Mg alloy was performed at 723 K to obtain a transitional microstructure with strain ranging from 0 to 30%. The recrystallization mechanisms involved and the evolution of texture during the deformation were specifically investigated. The results show that the recrystallization extent increases with strain. The isolated fine grains at 5% strain were formed by the particle stimulated nucleation, which tended to randomize the texture. A large number of {10 1¯ 2} extension twins at 10% strain effectively divided and refined the grains, but no twin-induced dynamic recrystallization was found. Some {10 1¯ 1}-{10 1¯ 2} double twins and {10 1¯ 1} compression twins were activated at 15% strain, which to a certain extent facilitated dynamic recrystallization nucleation. Meanwhile, the continuous dynamic recrystallization and discontinuous dynamic recrystallization mechanisms also occurred at various strains. In particular, the discontinuous dynamic recrystallization was more prominent due to the higher deformation temperature, and the recrystallization grains were evidently different from the orientations of their parent grains. In addition, with increasing strain, the overall texture component of the deformed samples gradually transitioned from the initial (0001) basal texture to the [11 2¯ 0]-[10 1¯ 0] non-fiber component, then tended to form the [11 2¯ 0] fiber orientation eventually.

A non-destructive, convenient, and low-cost yield estimation at the field scale is vital for precision farming. Significant progress has been made in using UAV-based canopy features to predict crop yield during the mid-growth stages. However, there has been limited effort to explore yield estimation specifically after crop maturity. Researching the effectiveness of artificial intelligence for estimating wheat yield utilizing phenotypic features extracted from UAV images, this study applied a deep learning algorithm (Mask R-CNN) to extract three wheat ear phenotypic features at ripening stage, including ear count, ear size, and ear anomaly index. Subsequently, machine learning algorithms (i.e., multiple linear regression, support vector regression, and random forest regression) driven by ear features were intercompared to obtain the optimal grain yield estimation. Based on the findings, (1) field observed ear count which was linearly associated with grain yield (R2 = 0.93), can be largely detected by UAV images (81 %); (2) Mask R-CNN demonstrated satisfactory performance in ear segmentation, achieving an F1 score of 0.87; (3) random forest regression resulted in the most accurate yield estimation (R2 = 0.86 and rRMSE = 17.53 %), when all three ear phenotypic features were combined. Overall, this study demonstrates that utilizing ear phenotypic features is an alternative approach for estimating wheat grain yield at ripening stage, showing potential as a viable substitute to tedious field sampling methods.

Nutrient supplementation during gestation and lactation is the basis for neurodevelopment, improving brain development including the learning and memory function of the offspring. Maternal supplementation with edible bird’s nest (EBN) has beneficial roles in the brain development of the offspring, but the mechanism has not been elucidated. In the current study, pregnant rats were given EBN (4.5 g/kg, 9 g/kg) and sialic acid (45 mg/kg) until the offspring were weaned. EBN was more effective at increasing learning and memory and hippocampal development than sialic acid of the offspring. In detail, EBN increased the synaptic plasticity of existing neurons and enhanced hippocampal neurogenesis of offspring, as well as upregulated the ERK-CREB-BDNF signaling pathway. It was concluded that maternal administration of EBN improved the learning and memory function of the offspring by accelerating hippocampus development by regulating the ERK-CREB-BDNF signaling pathway. Overall, EBN might be a potential maternal supplement in promoting neurodevelopment in offspring.

Mediastinal yolk sac tumors (YSTs) are highly aggressive germ cell tumors with an extremely poor prognosis. Radiotherapy plays an important role in the treatment of mediastinal YSTs. To maximize benefit from radiotherapy in patients with mediastinal YSTs, exploring functionally relevant biomarkers is essential. Previous studies have demonstrated that mutations in DNA-damage repair (DDR) genes, including BRCA1/2, potentially enhance sensitivity to radiotherapy in solid tumors. However, DDR-gene mutations, as possible predictive biomarkers for radiotherapy in primary mediastinal YSTs, have not yet been reported. Herein, we report a 29-year-old male patient with a refractory metastatic primary YST involving a germline frameshift mutation in the BRCA2 gene (NM_000059.3: exon11: c.4563_4564delAT: L1522fs). During treatment alternation, the patient was found to respond poorly to chemotherapy with or without an immune checkpoint inhibitor but well to radiotherapy. Finally, the patient achieved approximately 17 months of overall survival. To the best of our knowledge, this case report is the first to describe a remarkable response to local radiotherapy in a patient with a refractory metastatic mediastinal YST involving a DDR-gene mutation (germline BRCA2 frameshift variation). This case report provides insightful clues for precision radiotherapy in clinical practice.

This article proposes a hierarchical control strategy to address semi-ABS control as well as the precise clamping force control problems for an integrated electric parking brake (iEPB) system. To this end, a detailed system model, including modeling of the motor, transmission mechanism, friction and braking torque, is constructed for controller and observer design, and a sliding-mode-based observer (SMO) is proposed to estimate the load torque by using the motor rotational speed without installing a force sensor. In addition, a stable and reliable tire–road friction coefficient (TRFC) estimation method is adopted, and the desired slip ratio (DSR) is observed as the target that the rear wheels cycle around. At the upper level of the hierarchical control structure, the desired clamping forces of the rear wheels are generated using a sliding mode control (SMC) technique, and the control objective is to track the DSR to make full use of the road condition. At the lower level, the motor is controlled to track the desired clamping force generated from the upper controller. The hardware-in-the-loop (HIL) experimental results demonstrate the effectiveness and high tracking precision of the proposed strategy under different road conditions, and the estimation parameters based on the proposed observers are timely and accurate to satisfy the control requirements.

Healthcare provider vaccine knowledge and attitudes influence delivery of a strong vaccine recommendation. We aim to describe HPV vaccine knowledge, attitudes, and recommendation or discussion practices (KAP) among New York State medical providers, dentists, and pharmacists. A survey to assess providers’ KAP was distributed electronically to NYS members of medical organizations. Descriptive and inferential statistical methods were used to characterize provider KAP. Responses from 1637 surveys were included, from 864 (53%) medical providers, 737 (45%) dentists, and 36 (2%) pharmacists. 59% (509/864) of medical providers responded that they recommend HPV vaccine to patients, with 390/509 (77%) strongly recommending vaccine at 11–12 years. Medical providers were more likely to report recommending HPV vaccine for children ages 11–12 years if they strongly agreed that HPV vaccine prevents cancer 326/391 (83%) vs 64/117 (55%) and responded that HPV vaccination does not increase the risk of unprotected sex (386/494 (78%) vs 4/15 (25%)) (p

Aging is the strongest risk factor for cardiovascular disease, with progressive decline in the function of vascular endothelial cells (ECs) with age. Systematic analyses of the effects of aging on different cardiac EC types remain limited. Here, we constructed a scRNA atlas of EC transcriptomes in young and old mouse hearts. We identified 10 EC subclusters. The multidimensionally differential genes (DEGs) analysis across different EC clusters shows molecular changes with aging, showing the increase in the overall inflammatory microenvironment and the decrease in angiogenesis and cytoskeletal support capacity of aged ECs. And we performed an in-depth analysis of 3 special ECs, Immunology, Proliferating and Angiogenic. The Immunology EC seems highly associated with some immune regulatory functions, which decline with aging at different degrees. Analysis of two types of neovascular ECs, Proliferating, Angiogenic, implied that Angiogenic ECs can differentiate into multiple EC directions after initially originating from proliferating ECs. And aging leads to a decrease in the ability of vascular angiogenesis and differentiation. Finally, we summarized the effects of aging on cell signaling communication between different EC clusters. This cardiac EC atlas offers comprehensive insights into the molecular regulations of cardiovascular aging, and provides new directions for the prevention and treatment of age-related cardiovascular disease.

Abstract To analyse the microlesion effect (MLE) in the globus pallidus interna (GPi) of deep brain stimulation (DBS) in patients with Meige syndrome. Thirty-two patients with primary Meige syndrome who underwent GPi-DBS in this study. Burke–Fahn–Marsden Dystonia Rating Scale scores (BFMDRS-M) were obtained for the evaluation of clinical symptoms at 3 days before DBS (baseline), 24 h after DBS surgery, once weekly for 1 month until electrical stimulation, 6 months postoperatively and 12 months after surgery. Twenty-seven patients had MLE after GPi-DBS. The mean time of BFMDRS-M scores maximal improvement from MLE was 35.9 h postoperatively (range, 24–48 h), and the mean scores improved by 49.35 ± 18.16%. At 12 months after surgery, the mean BFMDRS-M scores improved by 50.28 ± 29.70%. Abstract The corrosion behavior of TiZrHfBeCu(Ni) high-entropy bulk metallic glasses (HE-BMGs) has been investigated. The TiZrHfBeCu(Ni) HE-BMGs exhibited high corrosion resistance in 3.5 wt. % NaCl solution because of accumulation of ZrO2 and TiO2 in the passive film. Ni promoted increases of the ZrO2, TiO2, and HfO2 contents and a decrease of the BeO content, which improved the HE-BMG corrosion behavior. Compared with Zr41.2Ti13.8Ni10Cu12.5Be22.5 BMG, the high-entropy effect of HE-BMGs can significantly reduce the atomic mobility, which inhibits outward migration of Cu, reduces the kinetics of the dissolution reaction, and inhibits inward erosion by Cl−, thereby improving the corrosion performance.

Abstract Background Cholangiocarcinoma (CHOL) is a malignant tumor that originates in the extrahepatic bile duct and can extend from the hilar region to the lower end of the common bile duct. The prognosis of CHOL patients is particularly poor; therefore, in this study, we screened mRNAs correlated with N6-methyladenosine (m6A) to construct a risk model for prognosis in CHOL. Methods The TCGA-CHOL dataset was applied to obtain and analyze the coexpression of 1281 m6A-related mRNAs, from which 14 were selected for further analysis through univariate proportional hazards (cox) regression analysis. Aryl hydrocarbon receptor interacting protein (AIP), CCAAT/enhancer binding protein beta (CEBPB), syndecan1 (SDC1), vacuolar protein sorting 25 homolog (VPS25) and syntaxin binding protein 2 (STXBP2) were then screened out through the least absolute shrinkage and selection operator (LASSO) and multivariate Cox regression analysis to develop a precise m6A-related mRNA prognosis risk model (MRMRPM) with an area under curve (AUC) of 0.908 and 0.923 after 1 and 2 years, respectively. We divided the samples into high-risk and low-risk groups using the m6A-related mRNA prognosis risk model. Results Kaplan–Meier analysis indicated poor overall survival (OS) for the high-risk group. Two Gene Expression Omnibus (GEO) datasets (GSE89748 and GSE107943) were used to validate the risk model. The results of drug sensitivity and immune cell infiltration analysis showed that the risk model could serve as a prognosis index of potential immunotherapeutic characteristics and drug sensitivity. Furthermore, the proportion of resting dendritic cells and regulatory T cells was positively associated with an increased expression of four m6A-related mRNAs — AIP, CEBPB, SDC1, and VPS25 — in the high-risk CHOL group. Conclusions Our findings suggest that this model can be a prognostic indicator for CHOL patients.

Endothelial cells (ECs) with senescence-associated secretory phenotypes (SASP) have been identified as a key mechanism of aging that contributes to various age-related kidney diseases. In this study, we used single-cell RNA sequencing (scRNA-seq) to create a transcriptome atlas of murine renal ECs and identify transcriptomic changes that occur during aging. We identified seven different subtypes of renal ECs, with glomerular ECs and angiogenic ECs being the most affected by senescence. We confirmed our scRNA-seq findings by using double immunostaining for an EC marker (CD31) and markers of specialized EC phenotypes. Our analysis of the dynamics of capillary lineage development revealed a chronic state of inflammation and compromised glomerular function as prominent aging features. Additionally, we observed an elevated pro-inflammatory and pro-coagulant microenvironment in aged glomerular ECs, which may contribute to age-related glomerulosclerosis and renal fibrosis. Through intercellular communication analysis, we also identified changes in signaling involved in immune regulation that may contribute to a hostile microenvironment for renal homeostasis and function. Overall, our findings provide new insights into the mechanisms of aging in the renal endothelium and may pave the way for the discovery of diagnostic biomarkers and therapeutic interventions against age-related kidney diseases.

Ecosystem conversion processes in different regions of China have different impacts on carbon and water cycle changes. Water use efficiency (WUE) is a key factor in coupling ecosystem carbon and water cycles, and it is important to quantify the effects of ecosystem conversional processes on the WUE in different regions of China to improve our understanding of the effects of ecological restoration projects and ecosystem conversional processes on carbon and water cycles in different regions. In this study, we proposed a dual–type extraction algorithm for ecosystem conversion processes (DTEA–ECP) to study the impact of ecosystem conversion processes on the WUE in different regions of China from 2001 to 2020. Our results showed that in the 2001–2020 period, the area of barren ecosystem conversion to grassland ecosystem increased at the fastest rate (4.29*103 km2 y-1). In the snow climate zone with dry winters and warm summers, projects returning cropland to forest were beneficial for improving WUE. In the northwestern part of the semiarid climate zone, returning cropland to grassland was beneficial for increasing the WUE of the ecosystem, while in northeastern China, the change in WUE after the conversion of grassland ecosystem to cropland ecosystem was not significant. WUE decreased after the conversion of forest ecosystems to grassland ecosystems, while WUE increased after the conversion of grassland ecosystems to forest ecosystems. This study identified where it was most suitable to plant each vegetation type and can help with the implementation of ecological projects such as afforestation, returning farmland to forest and grass, grassland protection and wind and sand management. The results provide information support for the in-depth development of national greening and low-carbon land restoration, improve vegetation carbon sink capacity, enhance carbon emission reduction and carbon sequestration, and help to achieve the goal of peak carbon and carbon neutrality.

The CRISPR-Cas system in Klebsiella pneumoniae can prevent the entry of blaKPC-IncF plasmids. However, some clinical isolates bear the KPC-2 plasmids despite carrying the CRISPR-Cas system. The purpose of this study was to characterize the molecular features of these isolates. A total of 697 clinical K. pneumoniae isolates were collected from 11 hospitals in China, and tested for the presence of CRISPR-Cas systems using polymerase chain reaction. Overall, 164 (23.5%) of 697 K. pneumoniae isolates had type I-E* (15.9%) or type I-E (7.7%) CRISPR-Cas systems. The most prevalent sequence type among isolates carrying type I-E* CRISPR was ST23 (45.9%), followed by ST15 (18.9%). Isolates with CRISPR-Cas system were more susceptible to ten antimicrobials tested, including carbapenems, compared with the CRISPR-negative isolates. However, there were still 21 CRISPR-Cas-carrying isolates that showed resistance to carbapenems, and these isolates were subjected to whole-genome sequencing. Thirteen of these 21 isolates carried blaKPC-2-bearing plasmids, of which nine had a new plasmid type, IncFIIK34, and two had IncFII(PHN7A8) plasmids. In addition, 12 of these 13 isolates belonged to ST15, while only eight (5.6%, 8/143) isolates belonged to ST15 in carbapenem-susceptible K. pneumoniae carrying CRISPR-Cas systems. In conclusion, we found that blaKPC-2-bearing IncFII plasmids could co-exist with the type I-E* CRISPR-Cas systems in ST15 K. pneumoniae.

The phase separation commonly exists in Fe(Se, Te) and damages their superconducting performance. In this paper, we successfully tuned the phase separation in Fe(Se, Te) via adding FeF2 to the raw materials. A macroscopically ordered phase separation which exhibited submillimetre scale striations in width arranged along the c-axis was formed in the FeF2-added Fe(Se, Te), different from the commonly observed flower shape phase separation in FeF2-free Fe(Se, Te). Moreover, the two macroscopical separated phases were identified as FeSe0.6Te0.4 and FeSe0.4Te0.6. The regular phase separation morphology and the fixed phase composition enhanced the flux pinning behavior which induced the appearance of Δκ pinning and deferred the changing from point pinning to surface pinning with the raising temperature. As a result, Fe(Se, Te) with the macroscopically ordered phase separation overwhelms the common phase separation Fe(Se, Te) in Hc2, U0, and Jc. Notably, at 4.2 K, the critical current density Jc reaches 9.3 × 104 A/cm2 in self-field and 1.3 × 104 A/cm2 at 7 T which outclassed the reported values. Besides, the well-defined stoichiometry of the separated phase also lead to a higher Tc of 15.42 K with a narrower transition width of only 0.98 K. Overall, our work reveals the tuning of phase separation in Fe(Se, Te) in terms of phase morphology and stoichiometry, which is a new strategy for improving the superconducting performance in Fe(Se, Te).

Abstract Porcine circovirus type 4 (PCV4) is a newly emerging virus, with both PCV4 genomic DNA and specific antibodies detected in swine herds in several provinces in China and South Korea. Although the virus was first identified in 2019 in Hunan, China, retrospective research suggests that serum samples collected as early as 2008 were positive for PCV4 antibody. Infections with only PCV4 or co-infections with other pathogens have been associated with several clinical manifestations, but its pathogenesis remains to be determined. The purpose of this review was the following: (1) to characterize PCV4 epidemiology by assessing evolutionary dynamics and genetic diversity of PCV4 strains circulating in swine herds; (2) to reconstruct a computerized 3D model to analyze PCV4 Cap properties; (3) and to summarize the current evidence of PCV4-associated clinical-pathological manifestations. The origin of PCV4 is apparently distinct from other PCV, based on analysis of phylogenetic trees. Of note, PCV4 shares an ancient common ancestor with mink circoviruses. Furthermore, the amino acid residue at position 27 of the PCV4 Cap is a key benchmark to distinguish PCV4a (27S) from PCV4b (27 N), based on PCV4 strains currently available, and variation of this residue may alter Cap antigenicity. In addition, the capsid surface of PCV4 has characteristics of increased polar residues, compared to PCV2, which raises the possibility that PCV4 may target negatively charged host receptors to promote virus infection. Further studies are required, including virus isolation and culture, and more detailed characterization of molecular epidemiology and genetic diversity of PCV4 in swine herds.

BackgroundLynch syndrome (LS) is caused by a germline mutation in one of the mismatch repair genes (MLH1, MSH2, MSH6, and PMS2) or in the EPCAM gene. The definition of Lynch syndrome is based on clinical, pathological, and genetic findings. Therefore, the identification of susceptibility genes is essential for accurate risk assessment and tailored screening programs in LS monitoring.Patients and methodsIn this study, LS was diagnosed clinically in a Chinese family using Amsterdam II criteria. To further explore the molecular characteristics of this LS family, we performed whole genome sequencing (WGS) to 16 members in this family and summarized the unique mutational profiles within this family. We also used Sanger sequencing technology and immunohistochemistry (IHC) to verify some of the mutations identified in the WGS analysis.ResultsWe showed that mutations in mismatch repair (MMR) related genes, as well as pathways including DNA replication, base excision repair, nucleotide excision repair, and homologous recombination were enhanced in this family. Two specific variants, MSH2 (p.S860X) and FSHR (p.I265V) were identified in all five members with LS phenotypes in this family. The MSH2 (p.S860X) variant is the first reported variant in a Chinese LS family. This mutation would result in a truncated protein. Theoretically, these patients might benefit from PD-1 (Programmed death 1) immune checkpoint blockade therapy. The patients who received nivolumab in combination with docetaxel treatments are currently in good health.ConclusionOur findings extend the mutation spectrum of genes associated with LS in MLH2 and FSHR, which is essential for future screening and genetic diagnosis of LS.

Endothelial cell (EC) function declines with age and contributes to the development of many vascular-related disease processes. Currently, the effects of aging on the molecular regulatory mechanisms of liver ECs have not been fully elucidated. Here, we employed single-cell RNA sequencing to map the transcriptome of ECs and analyzed their relationship with aging. We identified 8 different EC subtypes, interestingly, 2 of which were specially expressed in aged mice ECs namely aged capillary ECs (Aged ECs) and pro-inflammation capillary ECs (Proinfla.ECs). Double immunostaining for an EC marker (Cd31) and a marker of these specialized EC phenotypes confirmed the single-cell RNA sequencing data. Gene ontology analysis revealed that Aged ECs and Proinfla.ECs were associated with inflammatory response. Then we found that liver proliferating capillary ECs (Prolife.ECs) were most affected by senescence. Single-cell transcript analysis suggests that Prolife.ECs and angiogenic capillary ECs may form a poor microenvironment that promotes angiogenesis and tumorigenesis. Pseudo-temporal trajectories revealed that Prolife.ECs have different differentiation pathways in young and aged mice. In aged mice, Prolife.ECs could specifically differentiate into an unstable state, which was mainly composed of angiogenic capillary ECs. Intercellular communication revealed inflammatory activation in old group. Overall, this work compared the single-cell RNA profiles of liver ECs in young and aged mice. These findings provide a new insight into liver aging and its molecular mechanisms, and further exploration of Aged ECs and Proinfla.ECs may help to elucidate the molecular mechanisms associated with senescence.

Background It is unclear whether the long non-coding RNA (lncRNA) OTX2 antisense RNA 1 (OTX2-AS1) plays a pivotal role in gastric cancer (GC). An analysis of The Cancer Genome Atlas (TCGA) database data and bioinformatics was used to explore the relationship between OTX2-AS1 and GC in the current study. Methods We evaluated the relationship between clinical features and OTX2-AS1 expression, prognostic factors, and the significant involvement of OTX2-AS1 in function using various statistical methods, such as Kaplan–Meier method, Cox regression analysis, Gene Set Enrichment Analysis (GSEA), and immune infiltration analysis. GC cell lines were tested for OTX2-AS1 expression using qRT-PCR. Results A high level of OTX2-AS1 expression was significantly and negatively associated with Helicobacter pylori ( H pylori ) infection in GC patients ( P = .006) and predicted a poorer overall survival (OS) (HR: 1.54; 95% CI: 1.10-2.14; P = .011), progression-free interval (PFI) (HR: 1.75; 95% CI: 1.22-2.51; P = .002) and disease-specific survival (DSS) (HR: 1.85; 95% CI: 1.21-2.85; P = .005) in GC patients. There was an independent correlation between OTX2-AS1 expression (HR: 1.771; 95% CI: 1.164-2.696; P = .008) and OS in patients with GC. There were differential enrichments for the OTX2-AS1 high expression phenotype in the olfactory transduction, G alpha (s) signaling events, keratinization, olfactory signaling pathway, and preimplantation embryo. OTX2-AS1 expression may be related to certain immune-infiltrating cells. Compared to gastric epithelial cells (GES-1), GC cell lines showed a significant increase in OTX2-AS1 expression. Conclusion There was a significant association between OTX2-AS1 expression in GC patients and poor survival, suggesting that it may be a useful biomarker for prognosis and immunotherapy outcome of stomach adenocarcinoma (STAD) in GC.

Net ecosystem exchange of CO2 (NEE) is a useful tool to analyze the carbon balance of different areas. Since it is impossible to measure NEE in-situ at a regional or global scale, this study has conducted a NEE estimation model to map the NEE of global terrestrial systems. By training the model with 2774 series of data collected from the database of AmeriFlux and Fluxnet, and relevant studies, the model has achieved a substantial global NEE estimation with R2 = 0.60, 0.68, and 0.67 for arable land, forest, and grassland systems. According to the estimation, the average annual global NEE was −18.41 billion tons C. Asia was the continent who had the highest NEE (33.93% of global NEE), and forests were the ecosystems who contributed most to global CO2 absorption (51.75% of global NEE). When comparing with the global CO2 emissions, the model results also showed that Asia, North America, and Europe have run out of their ecosystem potential to absorb the CO2 emitted. The countries with top 10 greatest C emission had smaller effective NEEs to deal with their own emitted C except Russia and Canada. Considering the economic contribution of different countries, NEE estimation model could be a helpful tool to achieve a better fairness of controlling C emission tasks by considering both C emission generation and natural C emission handling ability.

Community-wide vaccine uptake remains sub-optimal. Healthcare provider (HCP) vaccine recommendations influence patient vaccination; however, provider vaccine recommendation behavior is highly influenced by one’s own vaccine attitudes and/or knowledge. We aim to describe vaccine knowledge, attitudes, and recommendation practices (KAP) among New York State HCPs. A survey to assess HCP KAP was developed and electronically distributed to NYS members of national medical organizations via their local chapter administrators. Descriptive statistical methods were used to define provider KAP. A total of 864 surveys were included, 500 (60%) and 336 (40%) primary and specialty care providers, respectively. Eighty-one percent (402/499) of primary care providers (PCPs) report encountering vaccine hesitant patients daily or weekly. Of the 500 PCPs who responded, only 204 (41%) stated strong agreement with confidence in their communications with vaccine hesitant patients. HCPs who correctly answered all four knowledge questions were more likely to self-report routine recommendations of standard vaccines to all patients when compared to those who correctly answered fewer questions (489/588 (83%) vs 135/241 (56%), p

Multipactor is a common discharge phenomenon occurring in space microwave systems. The surface microstructure has been verified to be effective in mitigating multipactor. In this work, we design a square coaxial low-pass filter (SCLPF) with dielectric sheets loaded to check the multipactor dependence on the structure parameters of the loaded dielectric sheets, and further model groove structures on the sensitive area surface to lower the risk of multipactor. Simulation results indicate that the SCLPF loaded with alumina and PTFE exhibits favorable operational characteristics, and the multipactor threshold is significantly improved after introducing the surface grooves. Then, we investigate the effects of three typical groove parameters, groove depth, groove number, and aspect ratio, on the multipactor threshold of the SCLPF device. The results show that the multipactor threshold rises at first as the groove number and groove depth increase, and then the threshold reaches a saturation status. For a deeper analysis of multipactor, we discuss how the grooves shelter the secondary electrons, and further mitigate the electron avalanche. Furthermore, the mechanisms of threshold saturation under the effect of groove parameters are analyzed in detail, and a contour map for the multipactor threshold of PTFE-loaded SCLPFs is given, which makes significant sense for predicting the multipactor threshold of the devices with its sensitive surface being grooved. The regularity of modulating the multipactor threshold by the groove structures obtained in this study is of engineering significance for suppressing multipactor in microwave devices in practical applications.

Abstract Background Non-small cell lung cancer (NSCLC) is a malignancy with considerable morbidity and mortality. Abnormal metabolism is a hallmark of cancer; however, the mechanism of glycolysis regulation in NSCLC progression is not completely understood. Recent studies suggest that some dysregulated long non-coding RNAs (lncRNAs) play important roles in tumor metabolic reprogramming. Methods To identify glycolysis-associated-lncRNAs in NSCLC, we compared RNA-sequencing results between high 18F-fluorodeoxyglucose (FDG)-uptake NSCLC tissues and paired paratumor tissues. The transcript abundance of AL355338 in 80 pairs of clinical samples was evaluated by quantitative real-time PCR assay and fluorescence in situ hybridization. The biological role of AL355338 on NSCLC cells were evaluated by functional experiments in vitro and in vivo. Moreover, RNA pull-down, mass spectrometry and RNA immunoprecipitation (RIP) assays were used to identify the protein interacted with AL355338. Co-immunoprecipitation, in situ proximity ligation assays and western blotting were applied to define the potential downstream pathways of AL355338. Results AL355338 was an upregulated glycolysis-associated lncRNA in NSCLC. Functional assays revealed that AL355338 was critical for promoting aerobic glycolysis and NSCLC progression. Mechanistic investigations showed that AL355338 directly bound with alpha-enolase (ENO1) and enhanced the protein’s stability by modulating its degradation and ubiquitination. A positive correlation was observed between AL355338 and ENO1 in NSCLC, and ENO1 was subsequently confirmed to be responsible for the oncogenic role of AL355338. Furthermore, AL355338 was capable of modulating ENO1/EGFR complex interaction and further activating EGFR-AKT signaling. Conclusions This study indicates that AL355338 confers an aggressive phenotype to NSCLC, and targeting it might be an effective therapeutic strategy.

Resistance to only ertapenem is an unusual phenotype of carbapenem-resistant Klebsiella pneumoniae (CRKP). This study aimed to investigate the molecular epidemiology and underlying mechanism involved in ertapenem resistance of K. pneumoniae strains that are susceptible to meropenem and imipenem. Among the 697 K. pneumoniae strains isolated from 11 grade A hospitals in China, 245 were CRKP strains, of which 18 strains resistant only to ertapenem were isolated. The genotypes, phenotypes, drug resistance homology, and drug sensitivity were analyzed; moreover, the expressions of efflux pump components and outer membrane proteins were assessed. The whole genomes of these 18 strains were sequenced and analyzed for mutations leading to drug resistance. The results revealed that ertapenem resistance may be related to ramR mutation. The function of ramR was confirmed using gene complementation to the original strain to determine the mechanism underlying ertapenem resistance of K. pneumoniae strains. In total, 7.4% of the tested CRKP strains were resistant only to ertapenem. None of these strains contained carbapenemase genes. Of the 18 ertapenem-resistant strains, 17 expressed the efflux pump, and outer membrane protein expression was reduced or absent in 4 strains. Whole-genome sequencing revealed the presence of mutations that introduced premature ramR codons stop in 14 strains (77.78%). When a functional copy of ramR was restored in the 14 strains, the minimum inhibitory concentration of ertapenem decreased, inhibition of efflux pumps was not detected, and the expression of outer membrane protein OmpK35 was either increased or was restored. These findings reveal the existence of ertapenem-resistant K. pneumoniae exhibiting no clonal transmission between strains. Mutations in ramR were demonstrated to cause outer membrane protein OmpK35 inhibition and over-expression of efflux pump in some strains, which is implicated in ertapenem resistance only in K. pneumoniae.

Osteoporotic fractures, also known as fragility fractures, are prevalent in the elderly and bring tremendous social burdens. Poor bone quality, weak repair capacity, instability, and high failure rate of internal fixation are main characteristics of osteoporotic fractures. Osteoporotic bone defects are common and need to be repaired by appropriate materials. Proximal humerus, distal radius, tibia plateau, calcaneus, and spine are common osteoporotic fractures with bone defect. Here, the consensus from the Osteoporosis Group of Chinese Orthopaedic Association concentrates on the epidemiology, characters, and management strategies of common osteoporotic fractures with bone defect to standardize clinical practice in bone repair of osteoporotic fractures.
