1. The Diversity of Root-Associated Endophytic Fungi from Four Epiphytic Orchids in China

Abstract

Root-associated endophytic fungi (RAF) are found asymptomatically in almost all plant groups. However, little is known about the compositions and potential functions of RAF communities associated with most Orchidaceae species. In this study, the diversity of RAF was examined in four wild epiphytic orchids, Acampe rigida, Doritis pulcherrima, Renanthera coccinea, and Robiquetia succisa, that occur in southern China. A culture-independent method involving Illumina amplicon sequencing, and an in vitro culture method, were used to identify culturable fungi. The RAF community diversity differed among the orchid roots, and some fungal taxa were clearly concentrated in a certain orchid species, with more OTUs being detected. By investigating mycorrhizal associations, the results showed that 28 (about 0.8%) of the 3527 operational taxonomic units (OTUs) could be assigned as OMF, while the OTUs of non-mycorrhizal fungal were about 99.2%. Among the OMFs, Ceratobasidiaceae OTUs were the most abundant with different richness, followed by Thelephoraceae. In addition, five Ceratobasidium sp. strains were isolated from D. pulcherrima, R. succisa, and R. coccinea roots with high separation rates. These culturable Ceratobasidium strains will provide materials for host orchid conservation and for studying the mechanisms underlying mycorrhizal symbiosis.

2. Development of intergeneric hybrids between wind orchids ( Sedirea japonica and Neofinetia falcata) and moth orchids ( Phalaenopsis alliances).

Abstract

To develop a new type of orchid with low-temperature tolerance and good fragrance, we generated intergeneric crosses between Korea-native wind orchids and tropical Phalaenopsis. In 2009, 160 cross combinations were made among Sedirea japonica, Phalaenopsis, Doritaenopsis and Neofinetia falcata. High incompatibility was observed from most of crosses as evidenced by traits such as early dropping of seed pods after pod formation or no seed formation after pod maturation. A total of 528 hybrid seedlings were obtained from 12 cross combinations. From these hybrid plants, several lines were selected based on flower type, inflorescence, cold tolerance, and fragrance, and finally the three best lines were selected. The selected lines were generally intermediate in both phenotype and genetic characters of the two parents, but tended to be more similar to Phalaenopsis. [ABSTRACT FROM AUTHOR]

In his previous publication (Buresch 1915a) from about 80 larvae collected (Buresch 1915a: 17) 26 ♂♂ and 24 ♀♀ are mentioned as reared successfully (Buresch 1915a: 26). Abstract

Abstract: Doritis is a wild terrestrial orchid, originated in the Northeastern part of Thailand. With its good floral structure and very diverse color, it has high potential to develop into commercial ornamental plant or to use as a hybridizer with other orchid species. Unfortunately it has not been studied in detail for its potential of development. In this study we developed SSR markers from EST database and examined the transferability of genomic SSR markers from other orchids in Doritis. 8009 ESTs belonged to family Orchidaceae were collected and analyzed for microsatellite. A total of 195 EST-SSR were identified. Twenty-three EST-SSR and seven genomic SSR primers (from Dendrobium and Serapias) were used for genetic diversity assessment of Doritis germplasm. 142 alleles were generated in which, 92 alleles (64.8%) showed polymorphism among 30 Doritis accessions (Doritis regnieriana, Doritis pulcherrima and D. pulcherrima var. buyssoniana.). The polymorphic information content (PIC) average at 0.6076 and ranged from 0.1244 to 0.8439. The phylogenetic tree based on UPGMA showed three major clades based on their taxonomy indicating that SSR markers developed in this study can be used for identification, conservation and selection of appropriate parents for the Doritis hybrid production in future. The genetic distance and relationships of 149 accessions representing 46 species in the genusPhalaenopsisand four species inParaphalaenopsiswere studied using random amplified polymorphic DNA (RAPD) markers. The genusParaphalaenopsiswas used as an outgroup. A total of 20 random primers were screened and out of these, six random primers provided 123 polymorphic bands and zero monomorphic bands. Pairwise genetic distances between accessions were estimated according to Nei and Li (1979). Cluster analysis of data using the UPGMA algorithm placed the species in seven groups that are mostly congruent with those based on morphological characters erected by previous workers. As observed from the banding patterns,Ph. doweryensis, which is suspected to be a hybrid ofPh. giganteaandPh. kunstleriorPh. cochlearis,is not. Abstract

Mature doritis plants (Doritis pulcherrima Lindley, cv. S94-3345) were cultured in plastic pots with 9 h (short-day, SD) and 16 h (long-day, LD) photoperiods, respectively. The main 9 h light period was under field conditions (30 °C day/20 °C night on average). The supplemental 7 h light conditions for the LD were conducted in chambers with 14 μmol m−2 s−1 photosynthetic photon flux. When transferred to SD for 30 days, flower spikes about 2.0–3.0 cm in length were initiated. When plants were transferred to SD for 45 days, spikes grew to 7.0–10.0 cm in length. In contrast, only 10% of the plants produced flowering shoots under LD conditions. Unique 21 and 103 kDa proteins were evident in one-dimensional electrophoresis of proteins from mature leaves under SD conditions. 2D gel electrophoresis confirmed that clear polypeptide spots with a molecular mass of 21 kDa at an isoelectric point of 5.2 and 103 kDa at an isoelectric point of 5.6 accumulated in the leaves when the flowering shoots reached 7.0–10.0 cm (4–5 flower primordia apparent). Possibly, the 21 and 103 kDa proteins play a role during flowering shoot formation in doritis. Polypeptide sequencing of P21 suggested a possible relationship to the cell division-like protein in Arabidopsis thaliana. It is clear that doritis is a facultative SD plant, and photoperiodic induction of its flowering is closely associated with protein synthesis changes in its leaves. [Copyright &y& Elsevier]

Abstract

Root-associated endophytic fungi (RAF) are found asymptomatically in almost all plant groups. However, little is known about the compositions and potential functions of RAF communities associated with most Orchidaceae species. In this study, the diversity of RAF was examined in four wild epiphytic orchids, Acampe rigida, Doritis pulcherrima, Renanthera coccinea, and Robiquetia succisa, that occur in southern China. A culture-independent method involving Illumina amplicon sequencing, and an in vitro culture method, were used to identify culturable fungi. The RAF community diversity differed among the orchid roots, and some fungal taxa were clearly concentrated in a certain orchid species, with more OTUs being detected. By investigating mycorrhizal associations, the results showed that 28 (about 0.8%) of the 3527 operational taxonomic units (OTUs) could be assigned as OMF, while the OTUs of non-mycorrhizal fungal were about 99.2%. Among the OMFs, Ceratobasidiaceae OTUs were the most abundant with different richness, followed by Thelephoraceae. In addition, five Ceratobasidium sp. strains were isolated from D. pulcherrima, R. succisa, and R. coccinea roots with high separation rates. These culturable Ceratobasidium strains will provide materials for host orchid conservation and for studying the mechanisms underlying mycorrhizal symbiosis. [ABSTRACT FROM AUTHOR]
