Previously, the RXR agonist UAB126 demonstrated therapeutic potential to treat obese mice by controlling blood glucose levels (BGL) and altering the expression of genes associated with lipid metabolism and inflammatory response. The purpose of the study was to assess the effects of UAB126 on the progression of diabetic retinopathy (DR) in rodent models of type 1 diabetes (T1D), streptozotocin-induced, and type 2 diabetes (T2D), in db/db mice. UAB126 treatment was delivered either by oral gavage for 6 weeks or by topical application of eye drops for 2 weeks. At the end of the treatment, the retinal function of diabetic mice was assessed by electroretinography (ERG), and their retinal tissue was harvested for protein and gene expression analyses. Bone-marrow cells were isolated and differentiated into bone marrow-derived macrophages (BMDMs). The glycolysis stress test and the 2-DG glucose uptake analysis were performed. Our results demonstrated that in the UAB126-treated diabetic BMDMs, the ECAR rate and the 2-DG uptake were improved as compared to untreated diabetic BMDMs. In UAB126-treated diabetic mice, hyperglycemia was reduced and associated with the preservation of ERG amplitudes and enhanced AMPK activity. Retinas from diabetic mice treated with topical UAB126 demonstrated an increase in Rxr and Ppar and the expression of genes associated with lipid metabolism. Altogether, our data indicate that RXR activation is beneficial to preclinical models of DR.

Background: Chronic spontaneous urticaria (CSU) and urticarial vasculitis (UV) share several clinical features including the occurrence of wheals. As of yet, the criteria for differentiating the 2 disorders are not clearly defined., Objective: Here, we aimed to identify differences, similarities, and the likelihood for specific clinical features in patients with UV versus those with CSU., Methods: Across 10 Urticaria Centers of Reference and Excellence, 106 patients with skin biopsy-confirmed UV and 126 patients with CSU were prospectively recruited to complete a questionnaire on the clinical features, course, and response to treatment of their disease., Results: As compared with CSU, patients with UV more often experienced postinflammatory skin hyperpigmentation, wheals of ≥24-hour duration, eye inflammation, and fever (6.9, 4.0, 3.6, and 2.4 times, respectively). Clinical features that increased the risk for UV diagnosis when present at the onset of disease included wheals of ≥24-hour duration (7.3-fold), pain of the skin (7.0-fold), postinflammatory hyperpigmentation (4.1-fold), and fatigue (3.1-fold). The diagnostic delay was markedly longer for normocomplementemic UV as compared with hypocomplementemic UV and CSU (21 vs 5 vs 6 months, respectively). Oral corticosteroids and omalizumab were the most effective treatments in patients with UV and CSU, respectively. Patients with UV showed a higher need for immunosuppressive and anti-inflammatory therapies than patients with CSU., Conclusions: Long wheal duration, skin pain and hyperpigmentation, and systemic symptoms point to UV rather than CSU as the underlying disease and should prompt further diagnostic workup including a skin biopsy., (Copyright © 2023 American Academy of Allergy, Asthma & Immunology. Gap junctions provide humoral and electric communication between the cells, thus contributing to their morpho-functional cooperation. Gap junction is formed by multiple intercellular channels, each of them being made by two closed hemichannels--connexons, that are oligomeric transmembrane proteins built by 6 subunits, belonging to connexin family. Permeability and electric conductivity of gap junction channels is determined by molecular peculiarities of connexins, their capacity for phosphorilation and by some extra- and intracellular factors. According to the current data, gap junctions in both cell cultures and tissues are dynamic structures with a short half-life period. Main mechanisms responsible for gap junction assembly and destruction have been discovered. These mechanisms were shown to depend upon peculiarities of differential genome activity and to be controlled by extra- and intracellular factors. The data on the gap junctions in the nervous system, heart and epidermis are presented.

Specific morphological and functional changes in the liver in acute obturative cholecystitis have been experimentally studied in 30 dogs and clinically examined in 21 patients. No morphological substrate of liver insufficiency were found in early period of acute obturative cholecystitis. Early changes in the liver are of reactive nature and have the features of active nonspecific hepatitis. The reactions of compensation and decompensation are changing each other periodically. The intensive reactions of compensation take place in the first 2 or 3 days of disease. The reactions of compensation weaken gradually. There is no correlation between changes in the liver and in blood serum. The early surgery and laparoscopic procedures in acute obturative cholecystitis are advocated.

The importance of inactivated influenza virus vaccines (IIVV) as a risk factor leading to a change of immunologic reactivity in persons earlier revaccinated with BCG once, twice or thrice was studied. The condition of T system was investigated by blast-transformation reaction with PPD-L and PHA in 1 and 3 months after administration of different types of IIVV. If all types of IIVV are administered, there is an inhibition of a specific activity of T cells with a certain stimulation of their nonspecific functions. The intensity of IIVV impact depends on the type, number, and duration of prior BCG vaccinations, as well as the initial condition of T-link.

Reactions between the mineral limonite and hydrogen peroxide were studied and gases produced thereupon were analysed by gas chromatography. Oxygen did not evolve if limonite was added to hydrogen peroxide frozen at a temperature of dry ice. However, at room temperature, a vigorous chemical reaction occurred and a large amount of oxygen evolved. Apparently, the ground of Mars contains not only hydrated iron oxides but also frozen hydrogen peroxide whose thawing in the incubation chamber of Viking resulted in its catalytic degradation under the action of iron ions. The evidence thus obtained and its comparison with the data of American scientists account for considerable evolution of oxygen detected by Viking upon analysis of the Mars ground.

The paper describes a technique of concentrating microorganisms from filtrates obtained from mixtures of the Martian ground analogs and desert soil using electroretention on polarizable carriers (sterilized cotton wool or gauze). The degree of retention can reach 99.9% at a field intensity of 50 V/cm and a flow rate of 0.5 ml/min. A high concentration of microorganisms in a suspension can be reached when the current is switched off and a small volume of the fresh nutrient medium is passed through the suspension.

The effect of hydrogen peroxide and the mineral limonite on the rate of microbial processes was studied in poor and rich soils. The dynamics of CO2 evolution can be registered upon addition of hydrogen peroxide to chernozem samples, which confirms the existence of metabolism of soil microorganisms. In experiments with desert soil, the evolution of O2 increases rather than that of CO2, which is probably due to an increase in the number of microorganisms producing catalase. Limonite stimulates the metabolic activity of microrganisms. The cultural and morphological properties of microflora are described, which are typical of soils incubated in the presence of limonite and hydrogen peroxide. This work supports the conclusion that, theoretically, the ground of Mars may contain microorganisms which have adapted, in the course of evolution, to high concentrations of hydrogen peroxide and hydrated iron oxides (of the limonite type) in the surrounding medium.

The survival of bacteria was studied in the "Artificial Mars" apparatus reproducing a complex of physical extreme factors. Bacteria isolated from microbiocenoses subjected to the action of chemical extreme factors (hydrogen peroxide, catalytically active minerals) were shown to be most resistant among soil heterotrophic bacteria. Cells in the resting state caused by dehydration survived better than vegetative cells. It has been concluded that microorganisms quite different in their physiological requirements and ecological properties can exist in the habitats extreme for life conditions.

A mixture of carbon dioxide and carbon monoxide was irradiated in the presence of limonite and hydrogen peroxide at 340 nm. As has been demonstrated by gas chromatography, chemical reactions occurring in this case yield organic compounds which could be synthesized in the experiments conducted by the Viking stations. Consequently, this method cannot discriminate between the chemical and biological processes which possibly accounted for positive signals registered in the experiments of the Viking stations.

The experience with non-surgical treatment of retained stones in 13 patients is described. The treatment was successful in 8 cases. The method of dissolution of the stones and their mechanical removal with the help of a lithotrap of own design was used. The conclusion is drawn, that the treatment of the patients with gallstones in presence of external biliary fistulas should start from conservative methods.
