Halogenated organic compounds (HOCs) in marine species collected from the Atlantic Ocean [3 shortfin mako (Isurus oxyrinchus) and 1 porbeagle (Lamna nasus)], and 12 sea turtles collected from the Pacific Ocean [3 loggerhead (Caretta caretta), 3 green (Chelonia mydas), 3 olive ridley (Lepidochelys olivacea), and 3 hawksbill (Eretmochelys imbricata)] were analyzed with a nontargeted analytical method using two-dimensional gas chromatography coupled to high-resolution time-of-flight mass spectrometry. Sharks and sea turtles had distinct HOC profiles. Halogenated methoxyphenols (halo-MeOPs) were the most abundant compound class identified in sea turtle livers, while polychlorinated biphenyls (PCBs) were the most abundant in shark livers. In addition to legacy contaminants and halo-MeOPs, a total of 110 nontargeted/novel HOCs (NHOCs) were observed in the shark livers. Shortfin mako collected from the northern Gulf of Mexico contained the largest number (89) and most diverse structural classes of NHOCs. Among all NHOCs, a group of compounds with the elemental composition C14H12-nCln (n = 5-8) exhibited the highest concentrations, followed by chlorocarbazoles and tris(chlorophenyl) methanes (TCPMs). Using nontargeted workflows, a variety of known and unknown HOCs were observed, which demonstrate the need to develop more complete chemical profiles in the marine environment.

Coastal lagoons and other estuarine habitats are increasingly exposed to the negative effects of growing human populations with associated intensifications in nutrient loading, harmful algal blooms, pollution, and habitat degradation. We examined population dynamics of Gulf pipefish Syngnathus scovelli in estuarine waters of the Indian River Lagoon, on the Atlantic coast of Florida. Substantial declines in abundance of this sentinel species were concurrent with significant losses of seagrass habitat associated with ongoing harmful algal blooms, and other perturbations during the study period spanning from 1998 to 2018. Moderate declines in S. scovelli were observed with early downward trends in seagrasses ahead of the onset of the precipitous seagrass reductions observed. The massive decline of seagrass habitats in the Indian River Lagoon in recent years had negative influences and was directly linked to population declines we observed in the S. scovelli population. Lack of seagrass habitat essential to this and related syngnathid species may reduce optimum seagrass-associated prey, increase predation by lack of appropriate cover, and increase energetic costs which may be realized through reduced growth rates and potential reproductive impairment. Identification and monitoring of population trends of S. scovelli and related sentinel fish species allow for early implementation of management actions that reduce the impact of anthropogenic pressures on the services that estuarine systems provide to the fishes and fisheries they support.

We report for the first time a highly divergent lineage in the Caribbean Sea for the bull shark (Carcharhinus leucas) based on the analysis of 51 mitochondrial DNA genomes of individuals collected in the western North Atlantic. When comparing the mtDNA control region obtained from the mitogenomes to sequences reported previously for Brazil, the Caribbean lineage remained highly divergent. These results support the existence of a discrete population in Central America due to a phylogeographic break separating the Caribbean Sea from the western North Atlantic, Gulf of Mexico and South America.

The Brazilian cownose ray, Rhinoptera brasiliensis has undergone a global population reduction and is currently classified by IUCN as Vulnerable. We sequenced the complete mitochondrial genome of R. brasiliensis, resulting in a 17,759 bp genome size, featuring the typical vertebrate mitochondrial genome structure. The mtDNA genome contains 13 protein-coding genes (PCGs), two ribosomal RNA (rRNA) genes, 22 transfer RNA (tRNA) genes, and a non-coding control region (D-loop). Each PCG was initiated by an authoritative ATG codon, except for COX1 that initiated by a GTG codon. Most of the PCGs were terminated by a complete codon (TAA/TAG), while an incomplete termination codon (TA/T) was found in five out of the 13 PCGs. Both Maximum Likelihood (ML) and Bayesian Inference (BI) analyses were used to reconstruct the phylogenetic relationships which revealed that R. brasiliensis was closely related to R. steindachneri whereas the reported mitogenome from a single specimen identified as R. steindachneri from Sri Lanka (GenBank accession number KM364982) was found to correspond to the sequence for R. javanica. These novel results provide a useful resource for further phylogeographic and population genetics studies of Rhinoptera spp.

Despite indications that sharks have an exceptional capacity to heal from traumatic injuries, no detailed microscopic observations of integumental wounds have been reported for sharks. This study details the histopathological features of such wounds in a free-ranging shark. An adult male Blacktip Shark Carcharhinus limbatus was collected in 2017 during fisheries-independent sampling efforts in the coastal southeastern U.S. Atlantic. The shark had numerous lesions on his head, torso, and left pectoral fin that were compatible with shark bites. Representative samples from two wounds on the head were examined by light microscopy. The epidermal changes included hyperplasia and hypertrophy; intracellular edema; and the absence of goblet cells, denticles, and chromatophores. In the dermis, fibrinocellular exudation, granulation tissue, and marked skeletal muscle necrosis and regeneration were observed. The above features were comparable to wound healing in bony fish, albeit minor differences were found. Although this case documents exceptionally good regeneration of skeletal muscle in the shark, we found no evidence of unique morphological healing patterns. Further studies on wound healing are needed because recent molecular and genetic findings do suggest evolutionary adaptations enhancing healing in sharks.

The Indian River Lagoon (IRL), located on the east coast of Florida, is a complex estuarine ecosystem that is negatively affected by recurring harmful algal blooms (HABs) from distinct taxonomic/functional groups. Enhanced monitoring was established to facilitate rapid quantification of three recurrent bloom taxa, Aureoumbra lagunensis, Pyrodinium bahamense, and Pseudo-nitzschia spp., and included corroborating techniques to improve the identification of small-celled nanoplankton (A. lagunensis presented here for the first time. An annual bloom index (ABI) was established for each taxon based on occurrence and abundance data. Blooms of A. lagunensis (>2 × 108 cells L–1) were observed in all 6 years sampled and across multiple seasons. In contrast, abundance of P. bahamense, largely driven by the annual temperature cycle that moderates life cycle transitions and growth, displayed a strong seasonal pattern with blooms (105–107 cells L–1) generally developing in early summer and subsiding in autumn. However, P. bahamense bloom development was delayed and abundance was significantly lower in years and locations with sustained A. lagunensis blooms. Pseudo-nitzschia spp. were broadly distributed with sporadic bloom concentrations (reaching 107 cells L–1), but with minimal concentrations of the toxin domoic acid detected (–1). In summer 2020, multiple monitoring tools characterized a novel nano-cyanobacterium bloom (reaching 109 cells L–1) that coincided with a decline in A. lagunensis and persisted into autumn. Statistical and time-series analyses of this spatiotemporally intensive dataset highlight prominent patterns in variability for some taxa, but also identify challenges of characterizing mechanisms underlying more episodic yet persistent events. Nevertheless, the intersect of temperature and salinity as environmental proxies proved to be informative in delineating niche partitioning, not only in the case of taxa with long-standing data sets but also for seemingly unprecedented blooms of novel nanoplanktonic taxa.

Mercury (Hg) deposited into aquatic sediments can be converted into the more toxic methylmercury (MeHg) by microbial activity. Atlantic stingrays (Hypanus sabinus) are an estuarine and nearshore species found in coastal regions of the western North Atlantic, occurring in multiple habitat types, and feeding mainly on benthic invertebrates. Mercury dynamics and speciation in stingrays have not been well examined. This study quantified total Hg and Hg species (Hg (II) and MeHg) in Atlantic stingrays sampled from Florida’s Indian River Lagoon (IRL) from 2012 to 2013. Tissues (muscle and liver) collected from 29 stingrays were lyophilized and homogenized before being analyzed using a direct mercury analyzer. Concentrations of total Hg in muscle were positively related to stingray disk width, but concentrations in liver were not. Mean (±SD) total Hg in muscle (0.56 ± 0.30 mg/kg dw) was significantly higher than mean total Hg in liver (0.23 ± 0.19 mg/kg dw). Nursery areas are critical for the reproductive cycle and biological requirements of Bull Sharks (Carcharhinus leucas) as they increase the survival of populations. Females tend to be philopatric to these areas as documented in estuaries from Australia, and inferred in the northern Gulf of Mexico and western North Atlantic Ocean, but not yet confirmed in these regions. In coastal waters of the southeastern United States, several sites have been proposed as nurseries for the Bull Shark, but little is known about how adult females utilize these areas during parturition. Philopatry for the Bull Shark was evaluated by comparing sequences of the mitochondrial DNA control-region (mtDNA-CR) and 8 microsatellite loci in juveniles and neonates from four previously reported nursery areas in United States (US) coastal waters; three in the northern Gulf of Mexico (Texas, Louisiana and Charlotte Harbor, Florida) and one in the western North Atlantic Ocean (Indian River Lagoon, Florida). A group of adult individuals from the Gulf of Mexico and Atlantic Ocean off the southeastern US were used to test genetic differences owed to limited gene flow between both regions. The analysis of genetic variation with the mtDNA-CR showed no differences among nurseries within the Gulf but significant differences when comparing the nursery areas of the two regions (Gulf vs Atlantic). In contrast, genetic homogeneity was observed among nursery areas within and between regions with the nuclear microsatellites suggesting male biased dispersal among regions. In addition, adult individuals from each of these two broad regions (Gulf of Mexico and Atlantic Ocean) showed no significant differentiation with any of the markers characterized in this study. These patterns of genetic differences support evidence for philopatry, further relaying the importance of protection and effective management of critical nursery habitats for future conservation of the species.

Elasmobranch ecotoxicological investigations are complicated because accessing organs that accumulate organic contaminants is usually lethal. Several metrics among liver, muscle, and brain were evaluated to determine their relative organic contaminant loads and the efficacy of using muscle as a non-lethal proxy for liver. Liver contained the highest concentrations (368–4020 ng/g wet weigth [ww]) and greatest estimated total load of contaminants. Brain had higher toxin concentrations than muscle (4.18–84.2 ng/g ww versus 0.94–4.73 ng/g ww). Liver and brain were similar to each other in terms of contaminant detection occurrence and signature overlap, whereas muscle poorly reflected those of liver and brain. However, the identity of contaminants detected in muscle constituted those that substantially contributed to summed liver and brain concentrations. Thus, studies utilizing muscle as a non-lethal liver alternative to study organic contaminant exposure in elasmobranchs should craft questions with care, considering its limited ability to serve as an accurate proxy.

For species that do not provide parental care after birth, excess maternal provisioning during development, beyond what is required for embryogenesis, provides offspring with resources to increase their chances of survival. Maternally derived resources are expected to be important for buffering offspring against limited food resources at birth or time needed to learn how to properly feed. Young-of-the-year (YOY) cryptic Scalloped Hammerheads (Sphyrna lewini) and Carolina Hammerheads (Sphyrna gilberti) were sampled from nurseries along the US Atlantic Coast and compared for a number of biological condition metrics across three developmental stages. Large declines in liver lipid content and hepatosomatic indices were found in neonatal sharks, using umbilical scar healing as a proxy for time since birth. Feeding commenced quickly as 96% of sharks had prey remnants in their stomachs. The combination of rapid exhaustion of maternally provided resources and high occurrence of stomachs with prey contents indicate that nursery quality, with respect to prey availability, may be important for YOY hammerhead survivorship. While externally the two species are morphologically similar, longer length-at-birth in S. lewini and higher hepatic condition in neonatal S. gilberti suggest that aspects of reproductive biology, including physiology, may differ between species. While more information is needed to distinguish life history differences between these two species, data collected from YOY may serve as a useful proxy to inform management when adult samples of cryptic species are difficult to collect.

Following the discontinuation of commercial polybrominated diphenyl ether (PBDE) mixtures, a variety of alternative flame retardants (FRs) have been developed and employed. To understand the contamination status of these emerging FRs in marine fish and wildlife, we investigated their bioaccumulation in four shark species, including shortfin mako shark (Isurus oxyrhinchus; n = 26), porbeagle (Lamna nasus; n = 4), sandbar shark (Carcharhinus plumbeus; n = 6), and common thresher (Alopias vulpinus; n = 4), from coastal and offshore waters of the western North Atlantic Ocean. Median concentrations of emerging FRs, including dechlorane analogues (i.e., dechlorane plus, Dec-602, -603, and - 604), tetrabromo-o-chlorotoluene (TBCT), 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE), and hexabromobenzene (HBBZ), ranged from 1.4-7.4, 10.2-22.4, 1.0-16.7, and 4.1-17.7 ng/g lipid weight (lw), respectively. Although concentrations of emerging FRs were generally 1-2 orders of magnitude lower than those of legacy FRs (i.e., PBDEs, 312-800 ng/g lw and hexabromocyclododecane or HBCDD, 17.2-99.3 ng/g lw), they were detected in more than 80% of the shark livers. Analysis of available biological data indicated that fork length significantly correlated with the concentrations of ΣPBDEs, HBCDD, ΣDechloranes or TBCT in shortfin mako livers. This indicates that longer-term exposure likely results in elevated FR concentrations in sharks. Our findings suggest likely exposure of western North Atlantic fish and wildlife to several emerging FRs, including dechloranes, BTBPE, HBBZ, and TBCT. Additional studies are needed to better elucidate their potential risks to fish and wildlife as well as the variety of environmental and biological factors influencing these risks.

In Florida, USA, mercury can bioaccumulate in aquatic food webs to levels that are harmful to human and ecosystem health. Proposed efforts to restore hydrological flow in the Florida Everglades, which encompasses projects throughout central and south Florida, may alter the spatial distribution and temporal trends in mercury bioaccumulation in Florida fish populations. Monitoring of mercury in sentinel fish species permits assessment of changing conditions and can identify natural or anthropogenic impacts on upstream watersheds and predict regional influences on human and wildlife consumers of these fish species. From 2006 through 2008, we assessed total mercury in muscle of 545 gray snapper, Lutjanus griseus (Linnaeus, 1758), and 381 crevalle jack, Caranx hippos (Linnaeus, 1766), from 13 broad estuarine regions in central and south Florida, downstream of proposed Everglades restoration projects. For comparison, we assessed an additional 134 gray snapper collected from adjacent offshore waters in the Gulf of Mexico in 2007–2008 and 2014–2015. Collectively, these results indicate that Florida Bay and Card and Barnes sounds are mercury hot spots for both fish species in Florida estuarine and coastal ecosystems. Mean mercury concentrations in these species from Florida Bay were 1.7–2.5 times those measured in Atlantic coast estuaries (i.e., Loxahatchee estuary, St. Lucie estuary, and Indian River Lagoon). Mercury in snapper from offshore gulf waters did not reveal spatial differences from adjacent coastal estuaries, but did suggest concentrations in offshore snapper have remained stable in recent years. These results provide comprehensive baseline data that can be used to identify mercury hot spots in south Florida and for monitoring temporal and spatial changes in estuarine and coastal mercury concentrations as Everglades restoration plans are implemented.

Background Elasmobranchs are particularly vulnerable to environmental metal contamination, accumulating these contaminants at high rates and excreting them slowly. The blue shark Prionace glauca L. is one of the most heavily fished elasmobranchs, although information regarding metal contamination and detoxification in this species is notably lacking. Methods Blue sharks were sampled in the western North Atlantic Ocean, in offshore waters adjacent to Cape Cod, Massachusetts. Total and metallothionein-bound liver and muscle metal concentrations were determined by inductively coupled plasma mass spectrometry (ICP-MS), metallothionein detoxification and oxidative stress endpoints were determined by UV–vis spectrophotometry. Results Metallothionein detoxification occurred for As, Cd, Cs, Cu, Hg, Pb, Se, Ti and Zn in liver, and for As, Cd, Cs, Pb, Se, and Zn in muscle, while reduced glutathione defenses seem to be related to Co and Zn exposure. Conclusion This is the first report for several metals (Ag, Co, non-radioactive Cs, Sb, Ti and V) for this species, which will aid in establishing baseline elemental data for biomonitoring efforts, health metrics, and conservation measures.

The scalloped hammerhead (Sphyrna lewini) and its cryptic congener, Carolina hammerhead (S. gilberti), are sympatrically distributed in the western North Atlantic Ocean. Because the species are indistinguishable based on external morphology, little research focused on Carolina hammerheads exists. In this study, the distribution of Carolina hammerheads in waters of the United States off the east coast (U.S. Atlantic) and Gulf of Mexico (Gulf) was examined and their abundance relative to scalloped hammerheads assessed by genetically identifying 1231 individuals using diagnostic single nucleotide polymorphisms. Both species were found in the U.S. Atlantic, where 27 % of individuals were Carolina hammerheads, but only scalloped hammerheads were identified in the Gulf. In Bulls Bay, SC, a well-known hammerhead nursery, assessment of relative abundance from May to September showed scalloped hammerheads were more abundant May-June and Carolina hammerheads more abundant July-September. Results of this study suggest Carolina hammerheads have a spatially limited distribution in the western North Atlantic and highlight the importance of Bulls Bay as a nursery for the species. In addition, the results suggest Carolina hammerheads may comprise a non-trivial proportion of what is considered the U.S. Atlantic scalloped hammerhead stock and should be considered in future decisions regarding management of the hammerhead complex.

Mercury (Hg) exposure poses a threat to both fish and human health. Sharks are known to bioaccumulate Hg, however, little is known regarding how Hg is distributed between different tissue groups (e.g. muscle regions, organs). Here we evaluated total mercury (THg) concentrations from eight muscle regions, four fins (first dorsal, left and right pectorals, caudal-from both the inner core and trailing margin of each fin), and five internal organs (liver, kidney, spleen, heart, epigonal organ) from two different shark species, bonnethead (Sphyrna tiburo) and silky shark (Carcharhinus falciformis) to determine the relationships of THg concentrations between and within tissue groups. Total Hg concentrations were highest in the eight muscle regions with no significant differences in THg concentrations between the different muscle regions and muscle types (red and white). Results from tissue collected from any muscle region would be representative of all muscle sample locations. Total Hg concentrations were lowest in samples taken from the fin inner core of the first dorsal, pectoral, and caudal (lower lobe) fins. Mercury concentrations for samples taken from the trailing margin of the dorsal, pectoral, and caudal fins (upper and lower lobe) were also not significantly different from each other for both species. Significant relationships were found between THg concentrations in dorsal axial muscle tissue and the fin inner core, liver, kidney, spleen and heart for both species as well as the THg concentrations between the dorsal fin trailing margin and the heart for the silky shark and all other sampled tissue types for the bonnethead shark. Our results suggest that biopsy sampling of dorsal muscle can provide data that can effectively estimate THg concentrations in specific organs without using more invasive, or lethal methods.

Although exploited populations of elasmobranchs may be able to recover from fishing pressure, there is little information regarding the Barndoor Skate's ability to cope with other anthropogenic stressors such as organic contaminants (OCs). Legacy OCs were measured in liver, muscle and ova from fourteen Barndoor Skates with mature skates having significantly greater mean concentrations of OCs than immature skates, demonstrating bioaccumulation with age. Using Toxic Equivalency Factors, skates were found to have levels of PCBs that have been shown to elicit negative physiological responses in other fishes and these results highlight the need for future studies to investigate the potential impacts that bioaccumulated organic contaminants have on the recovery and conservation of this vulnerable species.

Despite the widespread use of parabens in a range of consumer products, little is known about bioaccumulation of these chemicals in aquatic environments. In this study, six parabens and four of their common metabolites were measured in abiotic (water, sediment) and biotic (fish including sharks, invertebrates, plants) samples collected from a subtropical marine food web in coastal Florida. Methyl paraben (MeP) was found in all abiotic (100%) and a majority of biotic (87%) samples. 4-Hydroxy benzoic acid (4-HB) was the most abundant metabolite, found in 97% of biotic and all abiotic samples analyzed. The food chain accumulation of MeP and 4-HB was investigated for this food web. The trophic magnification factor (TMF) of MeP was estimated to be 1.83, which suggests considerable bioaccumulation and biomagnification of this compound in the marine food web. In contrast, a low TMF value was found for 4-HB (0.30), indicating that this compound is metabolized and excreted along the food web. This is the first study to document the widespread occurrence of parabens and their metabolites in fish, invertebrates, seagrasses, marine macroalgae, mangroves, seawater, and ocean sediments and to elucidate biomagnification potential of MeP in a marine food web.

During the investigation of dechlorane-related chemicals in North American wildlife, two unknown polychlorinated compounds (referred to as U1 and U2) were discovered. After extensive sample cleanup, structural information on U1 and U2 was characterized by gas chromatography (GC) coupled with single quadrupole mass spectrometer (MS) or GC-quadrupole time-of-flight (QToF) MS. Mass spectral evidence suggests that both U1 and U2 are structurally related to Dechlorane 603 (Dec603; C17H8Cl12), an analogue of the chlorinated flame retardant Dechlorane Plus. From the results we suspect U1 (C17H9Cl11) to be a monohydro analogue of Dec603 (i.e., one chlorine atom in Dec603 is replaced by a hydrogen atom). U1 may be formed via the degradation of Dec603's stereoisomers or present as an impurity in commercial Dec603 products. Mass spectral characterization of U2 (C17H7OCl11) suggests it is a carbonylic derivative of Dec603, likely formed via metabolic transformation of Dec603 or its photoisomer. Semiquantitative measurement revealed that U1 and U2 were present at estimated median concentrations of 49 ng/g lipid weight (lw) and 59 ng/g lw in peregrine falcon ( Falco peregrinus) eggs, from the mid-Atlantic region of the United States, and 4.6 and 3.0 ng/g lw in shortfin mako shark ( Isurus oxyrinchus) livers from the western North Atlantic Ocean, respectively. Our results demonstrate the occurrence of these two novel Dec603-related chemicals in both terrestrial and aquatic ecosystems.

Based on light and scanning electron microscopical studies, two new species of Philometra Costa, 1845 (Nematoda: Philometridae) are described from two species of Mycteroperca Gill (Serranidae), marine perciform fishes, in coastal waters off Florida, USA: Philometra deburonae n. sp. from the inner side of operculum of the yellowfin grouper Mycteroperca venenosa (L.) and P. incognita n. sp. from the ovary of the gag M. microlepis (Goode & Bean). Philometra deburonae n. sp. is mainly characterised by the body length of males (1.90–2.38 mm), the length of the spicules (78–84 µm) and gubernaculum (54 µm) and the presence of small outer cephalic papillae, a pair of fairly large caudal projections and the oesophageal gland extending anteriorly beyond the nerve-ring in subgravid females. Philometra incognita n. sp. is distinguished by the caudal mound consisting of two lateral reniform parts widely separated dorsally from each other, the absence of a pair of large papillae situated posteriorly to the cloaca, the shape and structure of the distal end of the gubernaculum plus the lengths of the spicules (117–141 µm) and gubernaculum (60–81 µm) in the male, the absence of caudal projections and the comparatively large larvae in the uterus (660–675 µm long) of the gravid female, as well as, the body length of both males (2.45–3.11 mm) and gravid females (120–180 mm). The present descriptions of an additional two new philometrids increases the number of recorded nominal species of Philometra parasitising groupers (Serranidae) in the Gulf of Mexico to nine.

Two of the world most endangered marine and terrestrial species are at the brink of extinction. The vaquita (Phocoena sinus) is the smallest existing cetacean and the population has declined to barely 22 individuals now remaining in Mexico's Gulf of California. With the ongoing decline, it is likely to go extinct within few years. The primary threat to this species has been mortality as a result of by-catch from gillnet fishing as well as environmental toxic chemicals and disturbance. This has called for the need to establish a National Park within the Gulf of California to expand essential habitat and provide the critical ecosystem protection for vaquita to thrive and multiply, given that proper conservation enforcement and management of the park are accomplished. In the terrestrial environment, the cheetah (Acinonyx jubatus) is reduced to a low number worldwide with the Iran subpopulation currently listed as Critically Endangered and the Indian subpopulation already extinct. There is a need for conservation efforts due to habitat loss, but also an indication of the conspicuous threat of illegal trade and trafficking from Africa and Arab countries in the Middle East. Funds have also been set up to provide refuges for the cheetah by working directly with farmers and landowners, which is a critical movement in adaptive management. These are the potential options for the preservation and possibly the expansion of the overall vaquita and cheetah populations.

The present work represents a comprehensive study of in utero maternal transfer of legacy and emerging flame retardants (FRs) in marine predators. We analyzed liver tissues from pregnant sharks of five viviparous species, including blacknose shark ( Carcharhinus acronotus; n = 12), blacktip shark ( Carcharhinus limbatus; n = 2), bonnethead ( Sphyrna tiburo; n = 2), Atlantic sharpnose shark ( Rhizoprionodon terraenovae; n = 2), and spinner shark ( Carcharhinus brevipinna; n = 2), as well as their embryos ( n = 84 in total from five species), collected from the western North Atlantic Ocean. Concentrations of frequently detected emerging FRs in adult female blacknose sharks were determined to be 6.1-83.3 ng/g lipid weight (lw) for dechlorane analogues, 2.5-29.8 ng/g lw for tetrabromo- o-chlorotoluene, and nondetection -32.6 ng/g lw for hexabromobenzene. These concentrations were 1-2 orders of magnitude lower than those of legacy polybrominated diphenyl ether flame retardants (85.7-398 ng/g lw). Similar contamination profiles were also found in the other four species, although FR concentrations varied in different species. A total of 21 FRs were commonly found in developing embryos of female sharks from five species, demonstrating maternal transfer in utero. The maternal transfer ratio (i.e., ratio of the mean litter concentration to their mother's concentration) determined in blacknose shark mother/embryo groups for each FR chemical was negatively associated with its octanol-water partition coefficient. Our work lays a solid foundation for future investigation of the underlying mechanisms of in utero transfer and additional physical or chemical factors that affect maternal transfer.

Elasmobranchs can bioaccumulate considerable amounts of persistent organic pollutants (POPs) and utilize several reproductive strategies thereby influencing maternal transfer of contaminants. This study provides preliminary data on the POP transfer from pregnant females to offspring of three species (Atlantic stingrays, bonnethead, blacktip sharks) with different reproduction modes (aplacental, placental viviparity). Polychlorinated biphenyl (PCB) levels were generally higher than any other POPs. Stingrays and blacktip shark embryos contained the lowest POP concentrations while bonnetheads and the blacktip adult female had the highest concentrations. Results suggest that POPs are more readily transferred from the mother to the embryo compared to what is transferred to ova in stingrays. Statistically significant differences in levels of selected POPs were found between embryos from the left and right uterus within the same litter as well as between female and male embryos within the same litter for bonnetheads, but not for the blacktip sharks. The population genetic structure of 251 bonnethead sharks, Sphyrna tiburo, from estuarine and nearshore ocean waters of the Western North Atlantic Ocean (WNA), was assessed using sequences of the mitochondrial DNA-control region. Highly significant genetic differences were observed among bonnetheads from 3 WNA regions; Atlantic coast of Florida, Gulf coast of Florida, and southwestern Gulf of Mexico (analysis of molecular variance, ΦCT = 0.137; P=0.001). Within the Gulf coast of Florida region, small but significant genetic differences were observed between bonnetheads from neighboring estuaries. These overall patterns were consistent with known latitudinal and inshore-offshore movements that occur seasonally for this species within US waters, and with the residency patterns and high site fidelity to feeding/nursery grounds reported in estuaries along the Atlantic coast of Florida and South Carolina. Historical demography also supported the occurrence of past population expansions occurring during Pleistocene glacial-interglacial cycles that caused drastic reductions in bonnethead population size, as a consequence of the eustatic processes that affected the Florida peninsula. This is the first population genetics study for bonnetheads to report genetic divergence among core abundance areas in US and Mexican waters of the WNA. These results, coupled with recent advances in knowledge regarding regional differences in life-history parameters of this species, are critical for defining management units to guide future management strategies for bonnetheads within US waters and across international boundaries into Mexico.

This study investigated wild caught striped mullet (Mugil cephalus) at Merritt Island National Wildlife Refuge (MINWR) for levels of 15 perfluoroalkyl acids (PFAA) in tandem with individual fecundity measurements (Oocyte sub-stage 2 late, n = 42) and oocyte reproductive stages (Stages 1–5, n = 128). PFAA measurements were quantified in striped mullet liver (n = 128), muscle (n = 49), and gonad (n = 10). No significant negative impacts of liver PFAA burden on wild-caught, mullet fecundity endpoints were observed in this study; however, changes in PFAA were observed in the liver as mullet progressed through different sub-stages of oocyte development. Of the PFAA with significant changes by sub-stage of oocyte development, the carboxylic acids (perfluorooctanoic acid, perfluorononanoic acid, and perfluorotridecanoic acid) increased in the liver with increasing sub-stage while the sulfonic acid and its precursor (perfluorooctanesulfonic acid (PFOS) and perfluorooctanesulfonamide, respectively) decreased in the liver with increasing sub-stage of oocyte development. This is a unique find and suggests PFAA change location of compartmentalization as mullet progress towards spawning. Investigations also revealed higher than expected median muscle and gonad levels of PFOS in striped mullet collected at MINWR (9.01 ng/g and 80.2 ng/g, respectively).

Elasmobranchs are a group of animals that typically occupy upper trophic levels in food webs and have a propensity to accumulate high contaminant concentrations. To date, few studies have investigated maternal offloading processes in sharks, despite the fact that this process represents a substantial source of exposure for young sharks and is a significant pathway for contaminant redistribution within marine ecosystems. Comparable to mammalian systems, scalloped hammerhead sharks (Sphyrna lewini) utilize a yolk-sac placental strategy to nourish young in utero, which may allow females to transfer contaminants to young. Organic contaminants (PCBs and chlorinated pesticides) were measured in livers of both females and males from several age classes that were collected from U.S. Atlantic waters, including two near-term pregnant females and their embryos. Adult female hammerheads (n = 3) were found to have lower levels of PCBs compared to the younger, adult male (mean ± SD, 11.1 ± 1.0 vs. 22.8 μg g(-1) lw), but had substantially higher concentrations of pesticides (4.1 ± 0.9 vs. 1.9 μg g(-1) lw). Embryos from the two litters (n = 36) had similar levels of summed organic contaminant concentrations (4.6 ± 0.9 μg g(-1) lw) and pregnant females were estimated to offload approximately 0.03-2.3% of their hepatic contaminant load to offspring. While the potential health impacts of these transferred contaminants is unknown, this is the first study to demonstrate that scalloped hammerheads are exposed to a substantial amount of contaminants prior to birth and document maternal offloading of organochlorines in a pseudo-placental shark species. Therefore, future research should continue to investigate the potential adverse effects these contaminants have on elasmobranch physiology.

To examine the migratory patterns, habitat utilization and residency of bonnethead sharks (Sphyrna tiburo (L. 1758)) in estuarine systems within coastal South Carolina, a tag-recapture experiment was conducted from 1998 to 2012 during which 2300 individuals were tagged. To assess the intra and inter-annual movements of tagged sharks, six estuaries within state waters were monitored using multiple gear types in addition to the cooperative efforts of recreational anglers throughout the southeastern United States. Over the course of the experiment 177 bonnetheads were recaptured after 3 days to 8.9 years at liberty, representing a recapture rate of approximately 8%. All bonnetheads were recaptured within the same estuary where they were originally tagged on intra and/or inter-annual scales, with the exception of six individuals, which were recaptured during migratory periods (i.e. late fall, winter and spring) in coastal waters off Florida, Georgia, North Carolina, and South Carolina. On 23 occasions cohesion was demonstrated by groups ranging in size from 2 to 5 individuals that were tagged together and recaptured together, with times at liberty ranging from 12 days to 3.6 years. Additionally, 13 individuals were recaptured multiple times with times at liberty ranging from 12 days to 8.9 years; all individuals were recaptured in the same estuary where they were initially tagged. We hypothesize that bonnetheads are using South Carolina's estuaries as summer feeding grounds due to the relatively high abundance of blue crabs (Callinectes sapidus), including ovigerous females during spring and summer months, and the location of these ephemeral yet predictable feeding areas is socially transmitted to relatively young, naive sharks by experienced, older individuals. The high degree of intra and inter-annual site fidelity demonstrated by bonnetheads in this region offers unique opportunities for in situ study of various aspects of the biology of this species, including identification of essential habitats, growth, homing mechanisms, mortality rates, movement patterns and social behavior.

The age, growth and maturity of bonnetheads Sphyrna tiburo inhabiting the estuarine and coastal waters of the western North Atlantic Ocean (WNA) from Onslow Bay, North Carolina, south to West Palm Beach, Florida, were examined. Vertebrae were collected and aged from 329 females and 217 males ranging in size from 262 to 1043 mm and 245 to 825 mm fork length, LF , respectively. Sex-specific von Bertalanffy growth curves were fitted to length-at-age data. Female von Bertalanffy parameters were L∞ = 1036 mm LF , k = 0·18, t0 = -1·64 and L0 = 272 mm LF . Males reached a smaller theoretical asymptotic length and had a higher growth coefficient (L∞ = 782 mm LF , k = 0·29, t0 = -1·43 and L0 = 266 mm LF ). Maximum observed age was 17·9 years for females and 16·0 years for males. Annual deposition of growth increments was verified by marginal increment analysis and validated for age classes 2·5+ to 10·5+ years through recapture of 13 oxytetracycline-injected specimens at liberty in the wild for 1-4 years. Length (LF50 ) and age (A50 ) at 50% maturity were 819 mm and 6·7 years for females, and 618 mm and 3·9 years for males. Both female and male S. tiburo in the WNA had a significantly higher maximum observed age, LF50 , A50 and L∞ , and a significantly lower k and estimated L0 than evident in the Gulf of Mexico (GOM). These significant differences in life-history parameters, as well as evidence from tagging and genetic studies, suggest that S. tiburo in the WNA and GOM should be considered separate stocks.

A female longfin mako shark Isurus paucus (Guitart-Manday, 1966) was found moribund on the Atlantic Ocean beach near Canaveral National Seashore, Florida; the shark died shortly after stranding. Macroscopic lesions included a partially healed bite mark on the left pectoral fin, a clefted snout, pericardial effusion and a pericardial mass surrounding a 12/0 circle fishing hook. The heart, pericardial mass, gills, ovary, oviduct, shell gland, epigonal organ, liver, kidney and intrarenal and interrenal glands were processed for histopathology and examined by brightfield microscopy. Microscopic examination revealed chronic proliferative and pyogranulomatous pericarditis and myocarditis with rhabdomyolysis, fibrosis and thrombosis; scant bacteria and multifocal granular deposits of iron were found intralesionally. In addition, acute, multifocal infarcts within the epigonal organ and gill filaments were found in association with emboli formed by necrocellular material. The ovary had high numbers of atretic follicles, and the liver had diffuse, severe hepatocellular degeneration, multifocal spongiosis and moderate numbers of melanomacrophage cells. This report provides evidence of direct mortality due to systemic lesions associated with retained fishing gear in a prohibited shark species. Due to the large numbers of sharks released from both recreational and commercial fisheries worldwide, impact of delayed post-release mortality on shark populations is an important consideration.

Age and growth estimates for the Bull Shark Carcharhinus leucas were derived from 121 vertebral centra collected from Bull Sharks (59.1–223.5 cm FL) between 1966 and 2010 in the western North Atlantic Ocean. Size at birth was confirmed with an additional 20 embryos (44.2–54.4 cm FL). The maximum age based on vertebral band pair counts was 25 (184 cm FL) and 27 (196 cm FL) years for males and females, respectively. The logistic and Gompertz growth models fitted the size-at-age data best for males and females, respectively. Based on previously published estimates of length at maturity, males mature at 15–17 years (176–185 cm FL) and females at 15 years (189 cm FL). Bull Sharks in the western North Atlantic Ocean and the Gulf of Mexico have similar growth rates and reach similar sizes at age.Received December 2, 2013; accepted February 4, 2014

Hybridization between closely related species has been documented across a wide range of taxa but has not been well studied in elasmobranchs. Hammerhead sharks have drawn global conservation concern because they experience some of the highest mortality rates among sharks when interacting with fisheries. Here we report on the detection of hybrids between the globally distributed scalloped hammerhead (Sphyrna lewini) and recently described Carolina hammerhead (S. gilberti) which are only known from the western Atlantic Ocean. Using a genomics approach, 10 first-generation hybrids and 15–17 backcrosses were detected from 554 individuals. The identification of backcrosses demonstrates hybrids are viable, and all backcrosses but one involved a scalloped hammerhead. All hybrids but one possessed Carolina hammerhead mtDNA, indicating sex-biased gene flow between species. Repeated hybridization and backcrossing with scalloped hammerheads could lead to the loss of endemic Carolina hammerheads.

We analyzed total mercury concentration in muscle tissue of 15 serranid species (n = 1,401 fish) collected from the Gulf of Mexico and also developed a comprehensive model relating two commonly used mercury analysis methods. There was considerable interspecific and intraspecific variability in mercury within groupers and sea basses. Mean mercury concentration for individual species ranged from 0.03 to 0.91 mg/kg wet weight across all size ranges, and for legally harvestable grouper and sea bass concentrations were 0.32 mg/kg and 0.09 mg/kg, respectively. Mercury in individual fish ranged from 0.01 to 3.3 mg/kg. Approximately 23% of all grouper samples analyzed contained mercury at concentrations that exceeded the U.S. Environmental Protection Agency's methylmercury consumption guidance criterion for humans (0.3 mg/kg). Mercury in legally harvestable individuals of some species exceeded 0.5 mg/kg, and harvestable black grouper Mycteroperca bonaci had a mean mercury concentration greater than 1.0 m...

Dolphinfish, Coryphaena hippurus Linnaeus, 1758, reproductive biology is well known, but this study, which examined gonad histology, substantially increases estimates of dolphinfish spawning fraction and annual egg production. Ovaries were collected from 621 females in three Florida regions: the Keys, Cape Canaveral, and the Panhandle. Maturation and spawning were evident in all three regions. Median size at maturity, 419 mm fork length (FL; 16.5 in, ?3 mo), was not regionally specific around Florida. Recruitment of primary oocytes to vitellogenesis occurred asynchronously throughout the spawning season, and mature oocytes developed group-synchronously as batches. Thus, an indeterminate method was required to estimate egg production. Once mature, females spawned 70-180 d yr-1. Some females spawned in all months of the year, but spawning fraction was highest in winter and spring. Batch fecundity (BF) ranged from 20,000 to 620,000 eggs and was significantly related to FL: BF = 0.000005 × FL3.62. An egg production model estimated a range from 15 to 174 million eggs yr-1, two orders of magnitude higher than the previous estimate (0.24-3.0 million eggs yr-1) in the Florida Straits. This new, higher estimate arises because our large sample size of gonad histology permitted estimation of spawning fraction throughout the year. Spawning early and often should make C. hippurus resilient to overfishing, but other data gaps�particularly regarding bycatch mortality�confound our ability to evaluate the effectiveness of size regulations in the fishery.

Stable-isotope ratios (δ 13 C and δ 15 N) and mercury in a model predator, and associated prey community assessments were used to make inferences regarding food web relationships and how these relationships are influenced by habitat variability and anthropogenic factors. Although interconnected, the three major basins of the Indian River Lagoon system on the Atlantic coast of Florida comprise noticeably different available habitat types with spatially distinct faunal communities and available prey for spotted seatrout, Cynoscion nebulosus , a model predatory fish species. Water quality, degree of urbanization, human population density, and levels of nitrogen enrichment clearly differ between these representative estuarine basins. The differences can influence feeding ecology and therefore result in different mercury concentrations and different stable-isotope signatures of spotted seatrout between basins. Mercury concentrations in spotted seatrout were greatest in Mosquito Lagoon (ML) and least in the Indian River Lagoon proper (IRL), although concentrations were low for all basins. Spotted seatrout from IRL were carbon-depleted and nitrogen-enriched compared with those from the other basins; this suggests either that the fish's primary source of carbon in IRL is an algae- or phytoplankton-based food web or that the pathway through the food web is shorter there. The δ 15 N values of IRL spotted seatrout were greater than those in the Banana River Lagoon or ML, suggesting slightly different trophic positioning of fish in these basins. The greater δ 15 N values in IRL spotted seatrout may also reflect the greater human population density and resultant anthropogenic inputs (e.g., observed higher total nitrogen levels) in IRL compared with the other more pristine basins examined. Understanding species' responses to broad-scale habitat heterogeneity in estuaries and knowing basin-specific differences in stable isotopes, mercury, prey communities, and comprehensive food web relationships will be useful in the future for long-term monitoring of impacts of anthropogenic disturbances and of recovery from restoration efforts.

Many coastal shark species use shallow estuarine regions as nursery habitat, but there are considerable gaps in our understanding of the seasonal distribution and habitat use patterns of sharks within these systems. We compiled all available sampling data from the Indian River Lagoon (IRL) along Florida's central Atlantic coast to examine the distribution of bull sharks Carcharhinus leucas. The data synthesized in this study spanned the 30-year period 1975–2005 and included information on the seasonal distribution, size structure, and habitat associations of 449 bull sharks. For comparison, data from an additional 106 bull sharks captured in shelf waters adjacent to the IRL were also examined. The IRL is dominated by young-of-the-year (age-0) and juvenile bull sharks, which were most abundant during spring, summer, and autumn. Eighteen microsatellite loci were developed for the silky shark Carcharhinus falciformis and screened across a total of 53 individuals from the western Atlantic Ocean, Eastern Tropical Pacific Ocean, and Red Sea. The number of alleles per locus ranged from 3 to 19, observed heterozygosity ranged from 0.158 to 0.917, and the probability of identity values ranged from 0.010 to 0.460. Though believed to be one of the most abundant species of large sharks, C. falciformis were recently listed as “near threatened” globally and “vulnerable” in the Eastern Tropical Pacific by the IUCN, due to reductions in catch rates from both target and non-target fisheries (Dulvy et al. in Aquat Conserv 18:459–482, 2008). Very little information exists about the population structure and genetic diversity of C. falciformis around the world. These new loci will provide effective tools for examining the sustainability of this declining species.

Acoustic telemetry was employed to resolve seasonal and daily movement patterns of adult red drum (Sciaenops ocellatus) in the northern Indian River Lagoon system, Florida. From May 2006 to September 2008, 44 tagged fish were tracked within an array of 34 autonomous receivers with individuals detected for up to 654 days. Most red drum exhibited strong site fidelity from winter through early summer with movement rates increasing significantly during fall spawning months. While some fish migrated to the nearest ocean inlet at this time, the majority remained within the lagoon year-round suggesting that true estuarine reproduction, a behavior uncommon or poorly documented elsewhere is the dominant life history strategy locally. Diel movement patterns were also pronounced and indicated changing depth preferences over a 24 h period. Despite a harvest prohibition on large red drum, long-term estuarine residency coupled with high angling pressure (41% recapture rate in 50 months) suggest that post-release angling mortality and sub-lethal effects to growth and reproduction may strongly influence the strength of adult size classes.
