Abstract

Anthropogenic methane (CH4) emissions increases from the period 1850–1900 until 2019 are responsible for around 65% as much warming as carbon dioxide (CO2) has caused to date, and large reductions in methane emissions are required to limit global warming to 1.5°C or 2°C. However, methane emissions have been increasing rapidly since ~2006. This study shows that emissions are expected to continue to increase over the remainder of the 2020s if no greater action is taken and that increases in atmospheric methane are thus far outpacing projected growth rates. This increase has important implications for reaching net zero CO2 targets: every 50 Mt CH4 of the sustained large cuts envisioned under low-warming scenarios that are not realized would eliminate about 150 Gt of the remaining CO2 budget. Targeted methane reductions are therefore a critical component alongside decarbonization to minimize global warming. We describe additional linkages between methane mitigation options and CO2, especially via land use, as well as their respective climate impacts and associated metrics. We explain why a net zero target specifically for methane is neither necessary nor plausible. Analyses show where reductions are most feasible at the national and sectoral levels given limited resources, for example, to meet the Global Methane Pledge target, but they also reveal large uncertainties. Despite these uncertainties, many mitigation costs are clearly low relative to real-world financial instruments and very low compared with methane damage estimates, but legally binding regulations and methane pricing are needed to meet climate goals.

Abstract

The purpose of this study is to optimize the selection of prophylactic cardioverter defibrillator implantation candidates. Currently, the main criterion for implantation is a low Left Ventricular Ejection Fraction (LVEF) whose specificity is relatively poor. We designed two classifiers aimed to predict, from long term ECG recordings (Holter), whether a low-LVEF patient is likely or not to undergo ventricular arrhythmia in the next six months. One classifier is a single hidden layer neural network whose variables are the most relevant features extracted from Holter recordings, and the other classifier has a structure that capitalizes on the physiological decomposition of the arrhythmogenic factors into three disjoint groups: the myocardial substrate, the triggers and the autonomic nervous system (ANS). In this ad hoc network, the features were assigned to each group; one neural network classifier per group was designed and its complexity was optimized. The outputs of the classifiers were fed to a single neuron that provided the required probability estimate. The latter was thresholded for final discrimination A dataset composed of 186 pre-implantation 30-mn Holter recordings of patients equipped with an implantable cardioverter defibrillator (ICD) in primary prevention was used in order to design and test this classifier. 44 out of 186 patients underwent at least one treated ventricular arrhythmia during the six-month follow-up period. Performances of the designed classifier were evaluated using a cross-test strategy that consists in splitting the database into several combinations of a training set and a test set. The average arrhythmia prediction performances of the ad-hoc classifier are NPV = 77% $\pm$ 13% and PPV = 31% $\pm$ 19% (Negative Predictive Value $\pm$ std, Positive Predictive Value $\pm$ std). According to our study, improving prophylactic ICD-implantation candidate selection by automatic classification from ECG features may be possible, but the availability of a sizable dataset appears to be essential to decrease the number of False Negatives., Comment: Computing in Cardiology, Saragosse : Espagne (2013)

Abstract

Expression of the flagellar genes in Rhodobacter sphaeroides is dependent on one of the four sigma-54 factors present in this bacterium and on the enhancer binding proteins (EBPs) FleQ and FleT. These proteins, in contrast to other well-characterized EBPs, carry out activation as a hetero-oligomeric complex. To further characterize the molecular properties of this complex we mapped the binding sites or upstream activation sequences (UASs) of six different flagellar promoters. In most cases the UASs were identified at approximately 100 bp upstream from the promoter. However, the activity of the divergent promoters flhAp-flgAp, which are separated by only 53 bp, is mainly dependent on a UAS located approximately 200 bp downstream from each promoter. Interestingly, a significant amount of activation mediated by the upstream or contralateral UAS was also detected, suggesting that the architecture of this region is important for the correct regulation of these promoters. Sequence analysis of the regions carrying the potential FleQ/FleT binding sites revealed a conserved motif. In vivo footprinting experiments with the motAp promoter allowed us to identify a protected region that overlaps with this motif. These results allow us to propose a consensus sequence that represents the binding site of the FleQ/ FleT activating complex.

Abstract

The paper first summarizes a general approach to the training of recurrent neural networks by gradient-based algorithms, which leads to the introduction of four families of training algorithms. Because of the variety of possibilities thus available to the 'neural network designer,' the choice of the appropriate algorithm to solve a given problem becomes critical. We show that, in the case of process modeling, this choice depends on how noise interferes with the process to be modeled; this is evidenced by three examples of modeling of dynamical processes, where the detrimental effect of inappropriate training algorithms on the prediction error made by the network is clearly demonstrated.

Abstract

While there are no indications of mixing back to 800 000 years in the EPICA Dome C ice core record, comparison with marine sediment records shows significant differences in the timing and duration of events prior to stage 11 (~430 ka, thousands of years before 1950). A relationship between the isotopic composition of atmospheric oxygen (δ18O of O2, noted δ18Oatm) and daily northern hemisphere summer insolation has been observed for the youngest four climate cycles. Here we use this relationship with new δ18O of O2 measurements to show that anomalous flow in the bottom 500 m of the core distorts the duration of events by up to a factor of 2. By tuning δ18Oatm to orbital precession we derive a corrected thinning function and present a revised age scale for the interval corresponding to Marine Isotope Stages 11–20 in the EPICA Dome C ice core. Uncertainty in the phasing of δ18Oatm with respect to insolation variations in the precession band limits the accuracy of this new agescale to ±6 kyr (thousand of years). The previously reported ~30 kyr duration of interglacial stage 11 is unchanged. In contrast, the duration of stage 15.1 is reduced by a factor of 2, from 31 to 16 kyr.

Abstract

Based on a composite of several measurement series performed on ice samples stored at −25 °C or −50 °C, we present and discuss the first δO2/N2 record of trapped air from the EPICA Dome C (EDC) ice core covering the period between 300 and 800 ka (thousands of years before present). The samples stored at −25 °C show clear gas loss affecting the precision and mean level of the δO2/N2 record. Two different gas loss corrections are proposed to account for this effect, without altering the spectral properties of the original datasets. Although processes at play remain to be fully understood, previous studies have proposed a link between surface insolation, ice grain properties at close-off, and δO2/N2 in air bubbles, from which orbitally tuned chronologies of the Vostok and Dome Fuji ice core records have been derived over the last four climatic cycles. Here, we show that limitations caused by data quality and resolution, data filtering, and uncertainties in the orbital tuning target limit the precision of this tuning method for EDC. Moreover, our extended record includes two periods of low eccentricity. During these intervals (around 400 ka and 750 ka), the matching between δO2/N2 and the different insolation curves is ambiguous because some local insolation maxima cannot be identified in the δO2/N2 record (and vice versa). Recognizing these limitations, we restrict the use of our δO2/N2 record to show that the EDC3 age scale is generally correct within its published uncertainty (6 kyr) over the 300–800 ka period.

Abstract

International audience; The Antarctic Vostok ice core provided compelling evidence of the nature of climate, and of climate feedbacks, over the past 420,000 years. Marine records suggest that the amplitude of climate variability was smaller before that time, but such records are often poorly resolved. Moreover, it is not possible to infer the abundance of greenhouse gases in the atmosphere from marine records. Here we report the recovery of a deep ice core from Dome C, Antarctica, that provides a climate record for the past 740,000 years. For the four most recent glacial cycles, the data agree well with the record from Vostok. The earlier period, between 740,000 and 430,000 years ago, was characterized by less pronounced warmth in interglacial periods in Antarctica, but a higher proportion of each cycle was spent in the warm mode. The transition from glacial to interglacial conditions about 430,000 years ago (Termination V) resembles the transition into the present interglacial period in terms of the magnitude of change in temperatures and greenhouse gases, but there are significant differences in the patterns of change. The interglacial stage following Termination V was exceptionally long--28,000 years compared to, for example, the 12,000 years recorded so far in the present interglacial period. Given the similarities between this earlier warm period and today, our results may imply that without human intervention, a climate similar to the present one would extend well into the future.

Abstract

Background Transcatheter aortic valve replacement (TAVR) has revolutionized management of high-risk patients with severe aortic stenosis. However, survival and the incidence of severe complications have been assessed in relatively small populations and/or with limited follow-up. Objectives This report details late clinical outcome and its determinants in the FRANCE-2 (FRench Aortic National CoreValve and Edwards) registry. Methods The FRANCE-2 registry prospectively included all TAVRs performed in France. Follow-up was scheduled at 30 days, at 6 months, and annually from 1 to 5 years. Standardized VARC (Valve Academic Research Consortium) outcome definitions were used. Results A total of 4,201 patients were enrolled between January 2010 and January 2012 in 34 centers. Approaches were transarterial (transfemoral 73%, transapical 18%, subclavian 6%, and transaortic or transcarotid 3%) or, in 18% of patients, transapical. Median follow-up was 3.8 years. Vital status was available for 97.2% of patients at 3 years. The 3-year all-cause mortality was 42.0% and cardiovascular mortality was 17.5%. In a multivariate model, predictors of 3-year all-cause mortality were male sex (p < 0.001), low body mass index, (p < 0.001), atrial fibrillation (p < 0.001), dialysis (p < 0.001), New York Heart Association functional class III or IV (p < 0.001), higher logistic EuroSCORE (p < 0.001), transapical or subclavian approach (p < 0.001 for both vs. transfemoral approach), need for permanent pacemaker implantation (p = 0.02), and post-implant periprosthetic aortic regurgitation grade ≥2 of 4 (p < 0.001). Severe events according to VARC criteria occurred mainly during the first month and subsequently in <2% of patients/year. Mean gradient, valve area, and residual aortic regurgitation were stable during follow-up. Conclusions The FRANCE-2 registry represents the largest database available on late results of TAVR. Abstract

International audience; A portable helmet based system has been developed to capture motor behavior during singing and other oral-motor functions in a non-laboratory experimental environment. The system, based on vocal tract sensing methods developed for speech production and recognition, consists of a lightweight " hyper-helmet " containing an ultrasonic (US) transducer to capture tongue movement, a video camera for the lips, and a microphone, coupled with a further sensor suite including an electroglottograph (EGG), nose-mounted accelerometer, and respiration sensor. The system has been tested on two rare, endangered singing musical styles, Corsican " Cantu in Paghjella " , and Byzantine hymns from Mount Athos, Greece. The versatility of the approach is furthermore demonstrated by capturing a contemporary singing style known as " Human Beat Box. "
