1. Are moorland invertebrates resilient to fire?

Abstract

Fire is an integral part of ecosystem management in many biomes around the world. This is particularly the case for the fire-prone and highly flammable moorlands of the Tasmanian Wilderness World Heritage Area. To maintain ecosystem processes and to conserve biodiversity, fire management in protected areas needs to be based on an understanding of the consequences of fire regimes. Invertebrates are a very diverse group of organisms that are critical for sustaining ecosystem processes. However, world-wide, there are few well-designed, long-term studies involving a broad range of taxa that have investigated the resilience of invertebrates to fire, and none in Tasmanian moorlands. I investigated the resilience of ground- and foliage-active invertebrate assemblages to low-intensity fire in low and moderate productivity moorlands, and identified predictors of assemblage change and its return to the pre-fire state. I used two complementary research designs: (1) a before-after-control impact design that aimed to minimise spatial variation, and (2) a replicated space-for-time design that provided insight into longer-term invertebrate responses to fire, and had a broad spatial scale. Species-level resolution for a broad range of invertebrate groups was used. I found that ground- and foliage-active invertebrate assemblages in moorland were resilient to single fires and to a fire regime with a mean fire interval of at least 24 years in low productivity moorlands and 37 years in moderate productivity moorlands. Low-intensity fire in moorland altered the composition of invertebrate assemblages because many taxa had low abundance in early successional stages. 8. Bedside prediction rule for infections after pediatric cardiac surgery.

Abstract

Purpose: Infections after pediatric cardiac surgery are a common complication, occurring in up to 30% of cases. The purpose of this study was to develop a bedside prediction rule to estimate the risk of a postoperative infection.Methods: All consecutive pediatric cardiac surgery procedures between April 2006 and May 2009 were retrospectively analyzed. The primary outcome variable was any postoperative infection, as defined by the Center of Disease Control (2008). All variables known to the clinician at the bedside at 48 h post cardiac surgery were included in the primary analysis, and multivariable logistic regression was used to construct a prediction rule.Results: A total of 412 procedures were included, of which 102 (25%) were followed by an infection. Most infections were surgical site infections (26% of all infections) and bloodstream infections (25%). Three variables proved to be most predictive of an infection: age less than 6 months, postoperative pediatric intensive care unit (PICU) stay longer than 48 h, and open sternum for longer than 48 h. Translation into prediction rule points yielded 1, 4, and 1 point for each variable, respectively. Patients with a score of 0 had 6.6% risk of an infection, whereas those with a maximal score of 6 had a risk of 57%. The area under the receiver operating characteristic curve was 0.78 (95% confidence interval 0.72-0.83).Conclusions: A simple bedside prediction rule designed for use at 48 h post cardiac surgery can discriminate between children at high and low risk for a subsequent infection. 13. Abundance and population growth estimates for bare-nosed wombats.

Abstract

Wildlife managers often rely on population estimates, but estimates can be challenging to obtain for geographically widespread species. Spotlight surveys provide abundance data for many species and, when conducted over wide spatial scales, have potential to provide population estimates of geographically widespread species. The bare-nosed wombat ( Vombatus ursinus ) has a broad geographical range and is subject to spotlight surveys. We used 19 years (2002-2020) of annual spotlight surveys to provide the first estimates of population abundance for two of the three extant bare-nosed wombat subspecies: V. u. ursinus on Flinders Island; and V. u. tasmaniensis on the Tasmanian mainland. Using distance sampling methods, we estimated annual rates of change and 2020 population sizes for both subspecies. Tasmanian mainland surveys included habitat data, which allowed us to also look for evidence of habitat associations for V. u. tasmaniensis . The average wombat density estimate was higher on Flinders Island (0.42 ha -1 , 95% CI = 0.25-0.79) than on the Tasmanian mainland (0.11 ha -1 , CI = 0.07-0.19) and both wombat subspecies increased over the 19-year survey period with an estimated annual growth rate of 2.90% (CI = -1.7 to 7.3) on Flinders Island and 1.20% (CI = -1.1 to 2.9) on mainland Tasmania. Habitat associations for V. u. tasmaniensis were weak, possibly owing to survey design; however, we detected regional variation in density for this subspecies. We estimated the population size of V. u. ursinus to be 71,826 (CI = 43,913-136,761) on Flinders Island, which when combined with a previously published estimate of 2599 (CI = 2254-2858) from Maria Island, where the subspecies was introduced, provides a total population estimate. We also estimated 840,665 (CI = 531,104-1,201,547) V. u. tasmaniensis on mainland Tasmania. These estimates may be conservative, owing to individual heterogeneity in when wombats emerge from burrows. 14. Density independent decline from an environmentally transmitted parasite.

Abstract

Invasive environmentally transmitted parasites have the potential to cause declines in host populations independent of host density, but this is rarely characterized in naturally occurring populations. We investigated (1) epidemiological features of a declining bare-nosed wombat ( Vombatus ursinus ) population in central Tasmania owing to a sarcoptic mange (agent Sarcoptes scabiei ) outbreak, and (2) reviewed all longitudinal wombat-mange studies to improve our understanding of when host population declines may occur. Over a 7-year period, the wombat population declined 80% (95% CI 77-86%) and experienced a 55% range contraction. The average apparent prevalence of mange was high 27% (95% CI 21-34), increased slightly over our study period, and the population decline continued unabated, independent of declining host abundance. Combined with other longitudinal studies, our research indicated wombat populations may be at risk of decline when apparent prevalence exceeds 25%. This empirical study supports the capacity of environmentally transmitted parasites to cause density independent host population declines and suggests prevalence limits may be an indicator of impending decline-causing epizootics in bare-nosed wombats. This research is the first to test effects of density in mange epizootics where transmission is environmental and may provide a guide for when apparent prevalence indicates a local conservation threat.

15. Environmental suitability of bare-nosed wombat burrows for Sarcoptes scabiei .

Abstract

Some of the most important pathogens affecting wildlife are transmitted indirectly via the environment. Yet the environmental stages of pathogens are often poorly understood, relative to infection in the host, making this an important research frontier. Sarcoptic mange is a globally widespread disease caused by the parasitic mite Sarcoptes scabiei . The bare-nosed wombat ( Vombatus ursinus ) is particularly susceptible, and their solitary nature and overlapping use of burrows strongly indicate the importance of environmental transmission. However, due to the challenge of accessing and monitoring within wombat burrows, there has been limited research into their suitability for off-host mite survival and environmental transmission (i.e., to serve as a fomite). We created a model using published laboratory data to predict mite survival times based on temperature and humidity. We then implemented innovative technologies (ground-penetrating radar and a tele-operated robotic vehicle) to map and access wombat burrows to record temperature and relative humidity. We found that the stable conditions within burrows were conducive for off-host survival of S. scabiei, particularly in winter (estimated mite survival of 16.41 ± 0.34 days) and less so in warmer and drier months (summer estimated survival of 5.96 ± 0.37 days). We also compared two areas with higher and lower average mange prevalence in wombats (13.35% and 4.65%, respectively), finding estimated mite survival was slightly higher in the low prevalence area (10.10 and 12.12 days, respectively), contrary to our expectations, suggesting other factors are also important for population prevalence. Our study is the first to demonstrate the suitability of the bare-nosed wombat burrow for off-host mite survival and environmental transmission. They have recently become the subject of headlines because of observations of dramatic declines in some places. Although there are hundreds of long-term insect monitoring programs, a global database for long-term data on insect assemblages has so far remained unavailable. In order to facilitate synthetic analyses of insect abundance changes, we compiled a database of long-term (≥10 yr) studies of assemblages of insects (many also including arachnids) in the terrestrial and freshwater realms. We searched the scientific literature and public repositories for data on insect and arachnid monitoring using standardized protocols over a time span of 10 yr or longer, with at least two sampling events. We focused on studies that presented or allowed calculation of total community abundance or biomass. We extracted data from tables, figures, and appendices, and, for data sets that provided raw data, we standardized trapping effort over space and time when necessary. For each site, we extracted provenance details (such as country, state, and continent) as well as information on protection status, land use, and climatic details from publicly available GIS sources. In all, the database contains 1,668 plot-level time series sourced from 165 studies with samples collected between 1925 and 2018. Sixteen data sets provided here were previously unpublished. Studies were separated into those collected in the terrestrial realm (103 studies with a total of 1,053 plots) and those collected in the freshwater realm (62 studies with 615 plots). Most studies were from Europe (48%) and North America (29%), with 34% of the plots located in protected areas. The median monitoring time span was 19 yr, with 12 sampling years. The number of individuals was reported in 129 studies, the total biomass was reported in 13 studies, and both abundance and biomass were reported in 23 studies. 17. COVID-19 restrictions provide a brief respite from the wildlife roadkill toll.

Abstract

The COVID-19 pandemic provides a rare opportunity to reveal the impact of reduced human activity on wildlife. I compared traffic volume and wildlife roadkill data along 18 km of highway before, during and after a 3-month period of COVID-19 restrictions with baseline data from the previous four years. Three marsupial herbivores comprised 89% of the 1820 roadkills recorded during the 4.5-year survey period: rufous-bellied pademelon Thylogale billardierii (31.5% of total), common brushtail possum Trichosurus vulpecula (29.8%) and red-necked wallaby Notamacropus rufogriseus (27.9%). During April 2020, when human activity was most restricted in the study area, traffic volume decreased by 36% (i.e. by an average 13,520 vehicle movements per day) and wildlife roadkill decreased by 48% (i.e. from 44 to 23 roadkills). However, when restrictions eased, traffic volume and wildlife roadkill returned to baseline levels indicating that the respite was brief in terms of animal welfare and of limited conservation value for these widespread and abundant species. Background: The arterial switch operation for transposition of the great arteries was initially believed to be an anatomical correction. Recent evidence shows reduced exercise capacity and left ventricular function in varying degrees in the long term after an arterial switch operation., Objective: To perform a meta-analysis on long-term exercise capacity and left ventricular ejection fraction after an arterial switch operation., Methods: A literature search was performed to cover all studies on patients who had undergone a minimum of 6 years of follow-up that reported either left ventricular ejection fraction, peak oxygen uptake, peak workload, and/or peak heart rate. A meta-analysis was performed if more than three studies reported the outcome of interest., Results: A total of 21 studies reported on the outcomes of interest. Oxygen uptake was consistently lower in patients who had undergone an arterial switch operation compared with healthy controls, with a pooled average peak oxygen uptake of 87.5±2.9% of predicted. The peak heart rate was also lower compared with that of controls, at 92±2% of predicted. Peak workload was significantly reduced in two studies. Pooled left ventricular ejection fraction was normal at 60.7±7.2%., Conclusion: Exercise capacity is reduced and left ventricular ejection fraction is preserved in the long term after an arterial switch operation for transposition of the great arteries.

Abstract

Right ventricular (RV) pressure overload has a vastly different clinical course in children with idiopathic pulmonary arterial hypertension (iPAH) than in children with pulmonary stenosis (PS). While RV function is well recognized as a key prognostic factor in iPAH, adverse ventricular-ventricular interactions and LV dysfunction are less well characterized and the pathophysiology is incompletely understood. We compared ventricular-ventricular interactions as hypothesized drivers of biventricular dysfunction in pediatric iPAH versus PS Eighteen iPAH, 16 PS patients and 18 age- and size-matched controls were retrospectively studied. Cardiac cycle events were measured by M-mode and Doppler echocardiography. Measurements were compared between groups using ANOVA with post hoc Dunnet's or ANCOVA including RV systolic pressure (RVSP; iPAH 96.8 ± 25.4 mmHg vs. PS 75.4 ± 18.9 mmHg; P = 0.011) as a covariate. RV-free wall thickening was prolonged in iPAH versus PS, extending beyond pulmonary valve closure (638 ± 76 msec vs. 562 ± 76 msec vs. 473 ± 59 msec controls). LV and RV isovolumetric relaxation were prolonged in iPAH (P < 0.001; LV 102.8 ± 24.1 msec vs. 63.1 ± 13.7 msec; RV 95 [61-165] vs. 28 [0-43]), associated with adverse septal kinetics; characterized by rightward displacement in early systole and leftward displacement in late RV systole (i.e., early LV diastole). Early LV diastolic filling was decreased in iPAH (73 ± 15.9 vs. PS 87.4 ± 14.4 vs. controls 95.8 ± 12.5 cm/sec; P = 0.004). Prolonged RVFW thickening, prolonged RVFW isovolumetric times, and profound septal dyskinesia are associated with interventricular mechanical discoordination and decreased early LV filling in pediatric iPAH much more than PS These adverse mechanics affect systolic and diastolic biventricular efficiency in iPAH and may form the basis for worse clinical outcomes. We used clinically derived data to study the pathophysiology of ventricular-ventricular interactions in right ventricular pressure overload, demonstrating distinct differences between pediatric pulmonary arterial hypertension (iPAH) and pulmonary stenosis (PS). Abstract

Objectives: Despite excellent survival in patients after the arterial switch operation, reintervention is frequently required and exercise capacity is decreased in a substantial number of patients. This study relates right-sided imaging features in patients long-term after the arterial switch operation to exercise capacity and ventilatory efficiency to investigate which lesions are functionally important., Methods: Patients operated in the UMC Utrecht, the Netherlands (1976-2001) and healthy controls underwent cardiac magnetic resonance imaging and cardiopulmonary exercise testing within 1 week. We measured main, left, and right pulmonary artery cross-sectional areas, pulmonary blood flow distribution, peak oxygen uptake, and minute ventilation relative to carbon dioxide elimination., Results: A total of 71 patients (median age, 20 [12-35] years, 73% were male) and 21 healthy controls (median age, 26 [21-35] years, 48% were male) were included. Main, left, and right pulmonary artery areas were decreased compared with controls (190 vs 269 mm(2)/m(2), 59 vs 157 mm(2)/m(2), 98 vs 139 mm(2)/m(2), respectively, all P < .001); however, pulmonary blood flow distribution was comparable (P = .722). Peak oxygen uptake and minute ventilation relative to carbon dioxide elimination were 88% ± 20% and 23.7 ± 3.8, respectively, with 42% and 1% of patients demonstrating abnormal results (≤ 84% and ≥ 34, respectively). The main pulmonary artery area significantly correlated with peak oxygen uptake (r = 0.401, P = .001) and pulmonary blood flow distribution with minute ventilation relative to carbon dioxide elimination (r = -0.329, P = .008). Subanalysis (<18, 18-25, >25 years) showed that the main pulmonary artery area was smaller in older age groups. In multivariable analysis, the main pulmonary artery area was independently associated with peak oxygen uptake (P = .032)., Conclusions: In adult patients after the arterial switch operation, narrowing of the main pulmonary artery is a common finding and is the main determinant of limitation in functional capacity, rather than pulmonary branch stenosis., (Copyright © 2015 The American Association for Thoracic Surgery. Abstract

Due to advances in cardiac surgery, survival of patients with congenital heart disease has increased considerably during the past decades. Many of these patients require repeated cardiovascular magnetic resonance imaging to assess cardiac anatomy and function. In the past decade, technological advances have enabled faster and more robust cardiovascular magnetic resonance with improved image quality and spatial as well as temporal resolution. This review aims to provide an overview of advances in cardiovascular magnetic resonance hardware and acquisition techniques relevant to both pediatric and adult patients with congenital heart disease and discusses the techniques used to assess function, anatomy, flow and tissue characterization.

22. Assessment of LV ejection fraction using real-time 3D echocardiography in daily practice: direct comparison of the volumetric and speckle tracking methodologies to CMR.

Abstract

Aims: This study is the first to directly compare two widely used real-time 3D echocardiography (RT3DE) methods of cardiac magnetic resonance imaging (CMR) and assess their reproducibility in experienced and less experienced observers., Methods: Consecutive patients planned for CMR underwent RT3DE within 8 h of CMR with Philips (volumetric method) and Toshiba Artida (speckle tracking method). Left ventricular ejection fraction (LVEF), left ventricular end-diastolic volume (LVEDV) and end-systolic volume (LVESV) were measured using RT3DE, by four trained observers, and compared with CMR values., Results: Thirty-five patients were included (49.7 ± 15.7 years; 55 % male), 30 (85.7 %) volumetric and 27 (77.1 %) speckle tracking datasets could be analysed. CMR derived LVEDV, LVESV and LVEF were 198 ± 58 ml, 106 ± 53 ml and 49 ± 15 %, respectively. LVEF derived from speckle tracking was accurate and reproducible in all observers (all intra-class correlation coefficients (ICC) > 0.86). LVEF derived from the volumetric method correlated well to CMR in experienced observers (ICC 0.85 and 0.86) but only moderately in less experienced observers (ICC 0.58 and 0.77) and was less reproducible in these observers (ICC = 0.55). Volumes were significantly underestimated compared with CMR (p < 0.001)., Conclusion: This study demonstrates that both RT3DE methodologies are sufficiently accurate and reproducible for use in daily practice. However, experience importantly influences the accuracy and reproducibility of the volumetric method, which should be considered when introducing this technique into clinical practice.

Abstract

Cardiac magnetic resonance (CMR) imaging is the preferred method to measure right ventricular (RV) volumes and ejection fraction (RVEF). This study aimed to determine the impact of excluding trabeculae and papillary muscles on RV volumes and function in patients with RV pressure and/or volume overload and healthy controls and its reproducibility using semi-automatic software. Eighty patients (pulmonary hypertension, transposition of the great arteries after arterial switch operation and after atrial switch procedure and repaired Tetralogy of Fallot) and 20 controls underwent short-axis multislice cine CMR. End diastolic volume (EDV), end systolic volume (ESV), RV mass and RVEF were measured using 2 methods. First, manual contour tracing of RV endo- and epi-cardial borders was performed. Thereafter, trabeculae were excluded from the RV blood volume using semi-automatic pixel-intensity based software. Both methods were compared using a Student T test and 25 datasets were re-analyzed for reproducibility. Exclusion of trabeculae resulted in significantly decreased EDV; ranging from -5.7 ± 1.7 ml/m(2) in controls to -29.2 ± 6.6 ml/m(2) in patients after atrial switch procedure. RVEF significantly increased in all groups, ranging from an absolute increase of 3.4 ± 0.8 % in healthy controls to 10.1 ± 2.3 % in patients after atrial switch procedure. Interobserver agreement of method 2 was equal to method 1 for RVEDV, RVESV and RVEF and superior for RV mass. In patients with overloaded RVs exclusion of trabeculae from the blood volume results in a significant change in RV volumes, RVEF and RV mass. Exclusion of trabeculae is highly reproducible when semi-automatic pixel-intensity based software is used.

Abstract

Understanding seasonal changes in invertebrate populations is important for understanding ecosystem processes and for conservation of invertebrate communities. Few studies have investigated variation in seasonal responses of multiorder and multispecies invertebrate assemblages. To determine whether temporal patterns in invertebrate assemblages and taxa vary between locations and vegetation age since burning, patterns of invertebrate occurrence were investigated monthly for 12 mo in cool temperate buttongrass moorlands at two locations (lowland and montane) containing paired plots with different fire history (young and old regrowth). For both locations and fire-ages, invertebrate taxon richness and abundance were generally higher during the warmer months than during the winter months. At the lowland location, foliage dwelling invertebrates were caught in greater numbers during winter than during summer owing to large numbers of Collembola. Each season had a distinct invertebrate assemblage. The invertebrate assemblages did not differ between young and old regrowth. The shifts in composition of monthly invertebrate assemblages between winter and summer differed between locations with assemblages in cooler months more dissimilar from warmer months at the montane location than the lowland location. Most taxa common to both locations had similar patterns of monthly occurrence but some taxa showed markedly different patterns. Mid- to late summer is the optimum time to conduct short-term surveys in buttongrass moorland to maximize species richness and abundance but short-term studies will miss significant components of the invertebrate community.

Abstract

Transcripts of the myotonic dystrophy protein kinase (DMPK) gene, a member of the Rho kinase family, are subject to cell-type specific alternative splicing. An imbalance in the splice isoform profile of DMPK may play a role in the pathogenesis of DM1, a severe multisystemic disorder. Here, we report how structural subdomains determine biochemical properties and subcellular distribution of DMPK isoforms. A newly developed kinase assay revealed that DMPK is a Lys/Arg-directed kinase. Individual DMPK isoforms displayed comparable transphosphorylation activity and sequence preference for peptide substrates. However, DMPK autophosphorylation and phosphorylation of MYPT1 (as putative in vivo target of DMPK), were dependent on presence of an alternatively spliced VSGGG motif and the nature of the C terminus. In-gel effects of the VSGGG motif on the migration behavior of full-length kinase provide evidence for a model in which this motif mediates 3-D-conformational changes in DMPK isoforms. Finally, different C termini endow DMPK with the ability to bind to either endoplasmic reticulum or mitochondria or to adopt a cytosolic location. Our results suggest that DMPK isoforms have cell-type and location dependent substrate specificities with a role in organellar and cytoarchitectural dynamics.
