Abstract

Our goal is to study $p$-adic local systems on a rigid-analytic variety with semistable formal model. We prove that such a local system is semistable if and only if so are its restrictions to the points corresponding to the irreducible components of the special fiber. For this, the main body of the paper concerns analytic prismatic $F$-crystals on the absolute logarithmic prismatic site of a semistable $p$-adic log formal scheme. Analyzing Breuil-Kisin log prisms, we obtain a prismatic purity theorem and deduce the above purity theorem for semistable local systems.

Abstract

In this paper, we apply the theory of Bridgeland stability conditions, which was originated from string theory, to study the derived category of coherent sheaves on Fargues--Fontaine curves. This leads us to consider the quasi-coherent sheaves $\mathcal{O}(\theta^{\pm})$ via the convergents of an irrational number $\theta$. We define the continuum envelop $\mathrm{QCoh}_{\mathbb{R}}(X_{FF})$ to be the smallest abelian subcategory in $\mathrm{QCoh}(X_{FF})$ containing $\mathrm{Coh}(X_{FF})$ and $\mathcal{O}(\theta^{\pm})$. We study the homological algebra of $\mathrm{QCoh}_{\mathbb{R}}(X_{FF})$ via Farey diagrams. We show that the homological property of $\mathcal{O}(\theta^{\pm})$ depends heavily on the arithmetic property of $\theta$. Abstract

We introduce the notion of completed $F$-crystals on the absolute prismatic site of a smooth $p$-adic formal scheme. We define a functor from the category of completed prismatic $F$-crystals to that of crystalline \'etale $\mathbf{Z}_p$-local systems on the generic fiber of the formal scheme and show that it gives an equivalence of categories. Abstract

We give a new construction of $(\varphi, \hat G)$-modules using the theory of prisms developed by Bhatt and Scholze. As an application, we give a new proof about the equivalence between the category of prismatic $F$-crystals in finite locally free $\mathcal{O}_\Delta$-modules over $(\mathcal{O}_K)_\Delta$ and the category of lattices in crystalline representations of $G_K$, where $K$ is a complete discretely valued field of mixed characteristic with perfect residue field. Abstract

Let $K$ be a discrete valuation field with perfect residue field, we study the functor from weakly admissible filtered $(\varphi,N,G_K)$-modules over $K$ to the isogeny category of Breuil-Kisin-Fargues $G_K$-modules. This functor is the composition of a functor defined by Fargues-Fontaine from weakly admissible filtered $(\varphi,N,G_K)$-modules to $G_K$-equivariant modifications of vector bundles over the Fargues-Fontaine curve $X_{FF}$, with the functor of Fargues-Scholze that between the category of admissible modifications of vector bundles over $X_{FF}$ and the isogeny category of Breuil-Kisin-Fargues modules. We study those objects appear in the essential image of the above functor and call them arithmetic BKF modules. We show certain rigidity result of arithmetic BKF modules and use it to compare existing $p$-adic Hodge theories at ${A}_{\mathrm{inf}}$ level., Comment: 30 pages, change the title and content by adding the main results in the author's thesis

Abstract

The PI3K/AKT/mTOR pathway is commonly dysregulated in cancer. Rapalogs exhibit modest clinical benefit, likely owing to their lack of effects on 4EBP1. We hypothesized that bi-steric mTORC1-selective inhibitors would have greater potential for clinical benefit than rapalogs in tumors with mTORC1 dysfunction. We assessed this hypothesis in tumor models with high mTORC1 activity both in vitro and in vivo. Bi-steric inhibitors had strong growth inhibition, eliminated phosphorylated 4EBP1, and induced more apoptosis than rapamycin or MLN0128. Multiomics analysis showed extensive effects of the bi-steric inhibitors in comparison with rapamycin. De novo purine synthesis was selectively inhibited by bi-sterics through reduction in JUN and its downstream target PRPS1 and appeared to be the cause of apoptosis. Abstract

Neural stem cells (NSCs) in the prenatal neocortex progressively generate different subtypes of glutamatergic projection neurons. Following that, NSCs have a major switch in their progenitor properties and produce γ-aminobutyric acid (GABAergic) interneurons for the olfactory bulb (OB), cortical oligodendrocytes, and astrocytes. Herein, we provide evidence for the molecular mechanism that underlies this switch in the state of neocortical NSCs. We show that, at around E16.5, mouse neocortical NSCs start to generate GSX2-expressing (GSX2+) intermediate progenitor cells (IPCs). In vivo lineage-tracing study revealed that GSX2+ IPC population gives rise not only to OB interneurons but also to cortical oligodendrocytes and astrocytes, suggesting that they are a tri-potential population. We demonstrated that Sonic hedgehog signaling is both necessary and sufficient for the generation of GSX2+ IPCs by reducing GLI3R protein levels. Using single-cell RNA sequencing, we identify the transcriptional profile of GSX2+ IPCs and the process of the lineage switch of cortical NSCs.

Abstract

Generation of olfactory bulb (OB) interneurons requires neural stem/progenitor cell specification, proliferation, differentiation, and young interneuron migration and maturation. Here, we show that the homeobox transcription factors Dlx1/2 are central and essential components in the transcriptional code for generating OB interneurons. In Dlx1/2 constitutive null mutants, the differentiation of GSX2+ and ASCL1+ neural stem/progenitor cells in the dorsal lateral ganglionic eminence is blocked, resulting in a failure of OB interneuron generation. In Dlx1/2 conditional mutants (hGFAP-Cre; Dlx1/2F/- mice), GSX2+ and ASCL1+ neural stem/progenitor cells in the postnatal subventricular zone also fail to differentiate into OB interneurons. In contrast, overexpression of Dlx1&2 in embryonic mouse cortex led to ectopic production of OB-like interneurons that expressed Gad1, Sp8, Sp9, Arx, Pbx3, Etv1, Tshz1, and Prokr2. Pax6 mutants generate cortical ectopia with OB-like interneurons, but do not do so in compound Pax6; Dlx1/2 mutants. We propose that DLX1/2 promote OB interneuron development mainly through activating the expression of Sp8/9, which further promote Tshz1 and Prokr2 expression. Based on this study, in combination with earlier ones, we propose a transcriptional network for the process of OB interneuron development.
