16. Stoichiometric imbalances between soil microorganisms and their resources regulate litter decomposition.

Abstract

Litter decomposition is dependent on the requirements of decomposer communities and their ability to acquire energy and nutrients from their substrates (i.e. litter) and the surrounding environment (i.e. soil). However, knowledge about whether and how stoichiometric imbalance (i.e. the differences in C:N:P ratios between microorganisms and their substrates) regulate litter decomposition rates and whether it can be compensated by soil resources have rarely been evaluated, and even less across different decomposition stages over time.In this study, we conducted a reciprocal litter transplantation experiment using a stoichiometric gradient along the forest‐steppe ecotone to evaluate mechanisms underlying litter‐microbe‐soil interactions at different moments during litter breakdown. We measured the C:N:P stoichiometry of litter, soil, microbes, enzyme ratios and soil microbial community composition (via metabarcoding) after 6 and 12 months of litter decomposition.We found that the stoichiometric imbalances between soil microorganisms and litter substrate controlled decomposition rates directly during the early phase of decomposition. In contrast, the stoichiometric imbalances between soil microorganisms and soil substrate regulated decomposition rates during the later phase of decomposition, but this was an indirect effect mediated via shifts in the saprophytic fungal community composition and enzyme allocation.These findings highlight that the stoichiometric imbalance between soil microorganisms and litter substrates can be partly compensated by the local soil resources over the course of the decomposition process. We conclude that the stoichiometric imbalance between soil microorganisms and their resources is a key mechanism that should not be ignored when predicting soil C and nutrient cycling in terrestrial ecosystems. Abstract

Simultaneous sources acquisition, also referred to as “blended acquisition”, involves recording two or more shots simultaneously. It allows for denser spatial sampling and can greatly speed up the field data acquisition. Thus, it has potential advantage to improve seismic data quality and reduce acquisition cost. In order to achieve the goal of blended acquisition, a deblending procedure is necessary. It attenuates the interference and thus improves the resolution of the pre-stack time migration image. In this paper, we propose an efficient deblending method, which applies frequency-varying median and mean filters to cross-spread azimuth-offset gathers (XSPR-AO). The method can be used with variable window sizes according to the characteristics of the interference. The effectiveness of the method is validated by a field data example.

Abstract

Neoadjuvant chemotherapy (NACT) remains an attractive alternative for controlling locally advanced cervical cancer. However, approximately 15–34% of women do not respond to induction therapy. To develop a risk stratification tool, 56 patients with stage IB‐IIB cervical cancer are included in 2 research centers from the discovery cohort. Patient‐specific somatic mutations led to NACT non‐responsiveness are identified by whole‐exome sequencing. Next, CRISPR/Cas9‐based library screenings are performed based on these genes to confirm their biological contribution to drug resistance. A 15‐gene classifier is developed by generalized linear regression analysis combined with the logistic regression model. In an independent validation cohort of 102 patients, the classifier showed good predictive ability with an area under the curve of 0.80 (95% confidence interval (CI), 0.69–0.91). Furthermore, the 15‐gene classifier is significantly associated with patient responsiveness to NACT in both univariate (odds ratio, 10.8; 95% CI, 3.55–32.86; p = 2.8 × 10−5) and multivariate analysis (odds ratio, 17.34; 95% CI, 4.04–74.40; p = 1.23 × 10−4) in the validation set. In conclusion, the 15‐gene classifier can accurately predict the clinical response to NACT before treatment, representing a promising approach for guiding the selection of appropriate treatment strategies for locally advanced cervical cancer., In a multicenter study of 158 patients with stage IB‐IIB cervical cancer, a 15‐core‐gene classifier is developed by integrated analysis of whole‐exome sequencing and CRISPR library screening. By predicting cervical cancer patients who would benefit from neoadjuvant chemotherapy, the classifier has the potential to guide the proper selection of treatment modalities, implicating a significant advance toward individualizing therapy.

Abstract

Objective: To demonstrate the changes of ovarian aging markers across the Stages of Reproductive Aging Workshop (STRAW) stages and modify it with subclassification of mid reproductive age stage (MR). Design: Healthy females were classified according to the STRAW system. Serum basal FSH, LH, E2, and anti-Müllerian hormone (AMH) were detected, FSH/LH ratio calculated, and antral follicle counts (AFCs) determined in follicular phase. Results: Progression through the whole STRAW stages under MR stage subdivided is associated with elevations in FSH, LH, FSH/LH ratio and decreases in E2, AMH and AFCs ( p < 0.001). Both serum AMH and AFCs decreased early (after 25 years) and significantly ( p < 0.01) with chronological age in MR stage. 0.982 ng/ml AMH and 3 antral follicles (low level of MR 25-30 years) were set as cutoffs to distinguish MR stage into early mid reproductive age (EMR) and late mid reproductive age (LMR) stages. The women in EMR stage compared with LMR could retrieve more oocytes in IVF treatment ( p < 0.05) and has a higher pregnancy chance (57.9%) though not significant. Conclusion(s): The early and marked fall in serum AMH levels and AFCs suggest fine markers to further categorize and define the MR stage, demonstrating disparate reproductive aging period with reduced ovarian reserve in young age across the STRAW stages.
