Objective To investigate the effects of miR-655-3p targeting kinesin family member 20A (KIF20A) on cell proliferation, migration and invasion of lung adenocarcinoma cells. Methods RT-qPCR was used to detect the expression of miR-655-3p in human lung adenocarcinoma cell lines H460, A549, HCC-2935, and H1299. A549 cells at logarithmic proliferation stage were divided into: blank group, miR-NC group, miR-655-3p mimics group, si-NC group, si-KIF20A group, miR-655-3p mimics+OE-NC group, miR-655-3p mimics+OE-KIF20A group. RT-qPCR was used to detect the miR-655-3p expression. Western blot was used to detect the KIF20A, matrix metalloproteinase 2 (MMP-2), MMP-9,RAS/mitogen-activated protein kinas (MAPK) pathway related protein expression; CCK-8 was used to detect cell proliferation; transwell experiment was used to detect cell migration and inva-sion; dual luciferase reporter gene detection experiment was used to verify the targeting relationship between miR-655-3p and KIF20A. Results miR-655-3p was less expressed in human lung adenocarcinoma cell lines, and has the lowest expression in A549 cells. Over-expression of miR-655-3p or silencing KIF20A inhibited A549 cell proliferation, migration, invasion and MMP-2, MMP-9, phosphorylation extracellular regulatory protein kinase 1/2(p-ERK1/2), phosphoinositide 3-kinase (p-PI3K) and phosphorylated protein kinase B (p-AKT) protein expression (P＜0.05). The miR-655-3p targeted at and negatively regulated the expression of KIF20A. Up-regulation of KIF20A inhibited the effects of over-expression of miR-655-3p on the proliferation, migration, invasion and RAS/MAPK pathway related proteins of A549 cells. Conclusions Over-expression of miR-655-3p may down-regulate KIF20A and inhibit the proliferation, migration and invasion of lung adenocarcinoma cells. This mechanism may be related to the inhibition of the RAS/MAPK pathway.

To establish a Bayesian network (BN) model to predict the survival of patients with malignant peritoneal mesothelioma (MPM) treated with cytoreductive surgery (CRS) plus hyperthermic intraperitoneal chemotherapy (HIPEC). The clinicopathological data of 154 MPM patients treated with CRS + HIPEC at our hospital from April 2015 to November 2022 were retrospectively analyzed. They were randomly divided into two groups in a 7:3 ratio. Survival analysis was conducted on the training set and a BN model was established. The accuracy of the model was validated using a confusion matrix of the testing set. The receiver operating characteristic (ROC) curve and area under the curve were used to evaluate the overall performance of the BN model. Survival analysis of 107 patients (69.5%) in the training set found ten factors affecting patient prognosis: age, Karnofsky performance score, surgical history, ascites volume, peritoneal cancer index, organ resections, red blood cell transfusion, pathological types, lymphatic metastasis, and Ki-67 index (all p < 0.05). The BN model was successfully established after the above factors were included, and the BN model structure was adjusted according to previous research and clinical experience. The results of confusion matrix obtained by internal validation of 47 cases in the testing set showed that the accuracy of BN model was 72.7%, and the area under ROC was 0.74. A study of the photoinduced electron transfer from 7-methyleneorbornadiene (MN) and 7-methylenequadricyclane (MQ) to an excited sensitizer acceptor in the presence of methanol reveals the importance of nucleophilic capture of the radical cations by methanol in achieving the reaction products. This nucleophilic capture is succeeded by free radical rearrangement to C. as the chief intermediate. The primary capture products have a short lifetime due to the allylic nature of the C4-C5 bonds. MQ radical cation is attacked from the 'exo' face by the nucleophile, which limitedly attacks MN radical cation from the 'endo' face.

Use of the CIDNP method helps analyze the photoinduced electron-transfer reactions of norbornadiene (MN(.+)), 7-methylene quadricyclane (MQ(.+)), 7-spiro(cyclopropanenorbornadiene) (SpN(.+)) and 7-spiro(cyclopropanequadricyclane (SpQ(.+))). Homoconjugative interactions between M and N fragments are responsible for high positive spin density of the exo-methylene carbon C(8) of MN(.+). Exchange interaction between Q SOMO and M FMO causes the negative spin density for exo-methylene carbon C(8) of MQ(.+). The interactions are weak in spirocyclopropane groups of SpN and SpQ.

Background: Inositol polyphosphate 4-phosphatase type II (INPP4B) has been identified as a tumor repressor in several human cancers while its role in endometrial cancer has not been investigated yet. Therefore, the current study was designed to determine whether INPP4B participates in the progression of endometrial cancer by utilizing clinical data and experimental determination., Materials and Methods: We first include six chemotherapy-treated patients with recurrent and metastatic endometrioid carcinoma to determine the relationship between INPP4B mutation and relative tumor burden. By using siRNA-mediated gene silencing and vector-mediated gene overexpression, we further determined the effect of manipulating INPP4B expression on the proliferation, invasion, and survival of endometrial cancer cells. Furthermore, the repressing effect of INPP4B together with its role in chemotherapy was further validated by xenograft tumor-bearing mice models. Western blot analysis was used to explore further downstream signaling modulated by INPP4B expression manipulation., Results: Two of the patients were found to have INPP4B mutations and the mutation frequency of INPP4B increased during the progression of chemotherapy resistance. Endometrial cancer cells with silenced INPP4B expression were found to have promoted tumor cell proliferation, invasion, and survival. Endometrial cancer cells overexpressing INPP4B were found to have decreased tumor cell proliferation, invasion, and survival. An in vivo study using six xenograft tumor-bearing mice in each group revealed that INPP4B overexpression could suppress tumor progression and enhance chemosensitivity. Furthermore, INPP4B overexpression was found to modulate the activation of Wnt3a signaling., Conclusion: The current study suggested that INPP4B could be a suppressor in endometrial cancer progression and might be a target for endometrial cancer treatment. Objective: To explore the inflammatory cascade mechanism through Toll like receptor 2 (TLR2) pathway after cerebral ischemia/reperfusion, and to study molecular mechanisms of Guanmaitong (GMT) Tablet for protecting brain damage., Methods: We used bolt-line method to block/release the middle cerebral artery, causing cerebral ischemia/reperfusion (I/R) injury model. GMT Tablet was given by gastrogavage. Rats were then divided into the high dose GMT group (1200 mg/kg), the middle dose GMT group (600 mg/kg), the low dose GMT group (300 mg/kg), the positive control group (Tanakan, 20 mg/kg). Their right brain tissues were fixed in 10% neutral formalin. TLR2 expressions were detected by immunofluorescence staining. The total protein was extracted from right brain tissues by ultrasonica- tion. Expression levels of extracellular regulated protein kinases (ERK), phospho-extracellular regulated protein kinases (p-ERK), p38-mitogen activated protein kinases (p-ERK), phospho-p38-mitogen activated protein kinases [p-p38-MAPKs(p-p38)] were assessed by Western blot. Abdominal aortic blood was withdrawn. IL-6 and IL-1β levels were detected by ELISA in brain tissues and serum., Results: Compared with the sham-oepration group, expression levels of TLR2, ERK, p-ERK, p38, p-p38 protein were up-regulated (P < 0.05, P < 0.01), and contents of IL-6 and IL-1β in brain tissues and serum were increased in the model group (P < 0.01). Expression levels of TLR2, ERK, p-ERK, p38, p-p38 were down-regulated (P < 0.05, P < 0.01), and contents of IL-6 and IL-1β were reduced in brain tissues and serum in middle and high dose GMT groups (P < 0.05, P < 0.01)., Conclusions: TLR2 pathway was involved in cerebral I/R injury. GMT protected neurons by down-regulating protein expressions of TLR2, ERK, p-ERK, p38, p-p38 and contents of IL-1β and IL-6.

Aim: To investigate the expression and clinical significance of calcium-binding protein S100A9 in the patients of breast cancer., Methods: The serum S100A9 level from 39 cases of preoperative breast cancer, 15 postoperative patients, and 10 healthy women was detected by ELISA. Immunohistochemistry was used to detect the S100A9 expression in 12 specimens of breast cancer tissues., Results: The serum S100A9 level was significantly higher in the preoperative breast cancer patients than that in the healthy women (P<0.01) and the postoperative breast cancer patients(P<0.01). The serum S100A9 level of the breast cancer patients with lymph node metastasis was significantly higher than those without lymph node metastasis (P<0.01). S100A9 level in the serum was related to patient age, tumour size, histological grade, and pathological type (P<0.01). The expression of S100A9 protein in breast cancer tissues was significantly higher than that in paracancerous benign breast tissues., Conclusion: The expression of S100A9 significantly increases in the serum and tissues of patients with breast cancer, which indicates that S100A9 may function as a marker of diagnosis and treatment effect in breast cancer.

Aim: To prepare and characterize monoclonal antibody (mAb) against human LSECtin (liver and lymph node sinusoidal endothelial cell C-type lectin) protein., Methods: BALB/c mice were immunized with prokaryotically expressed human LSECtin protein. The splenocytes from the immunized mice were fused with murine myeloma cells (Sp2/0) and then the mAb-positive hybridoma cells were screened by indirect ELISA. Reaction of mAb to LSECtin antigen was characterized by Western blot, indirect immunofluorescent staining, immunohistochemical staining and FCM., Results: Eight hybridoma cells secreting mAbs were established. The isotypes of the mAbs were IgG. Ascites titers were between 1:10(6) - 1:10(7). All the mAbs recognized human LSECtin protein on LSECtin-transfected 3T3 cells and six of the mAbs specifically recognized liver sinusoidal endothelial cells., Conclusion: Eight anti-LSECtin mAbs have been obtained. The characterization of the mAbs indicate that they show fine specificity by Western blot, indirect immunofluorescent staining, immunohistochemical staining and FCM, which can provide a powerful reagent for the functional study of LSECtin.
