Abstract

Productively infected cells are generally thought to arise from HIV infection of activated [CD4.sup.+] T cells, and these infected activated cells are thought to be a recurring source of latently infected cells when a portion of the population transitions to a resting state. We discovered and report here that productively and latently infected cells can instead originate from direct infection of resting [CD4.sup.+] T cell populations in lymphoid tissues in Fiebig I, the earliest stage of detectable HIV infection. We found that direct infection of resting [CD4.sup.+] T cells was correlated with the availability of susceptible target cells in lymphoid tissues largely restricted to resting [CD4.sup.+] T cells in which expression of pTEFb enabled productive infection, and we documented persistence of HIV-producing resting T cells during antiretroviral therapy (ART). Thus, we provide evidence of a mechanism by which direct infection of resting T cells in lymphoid tissues to generate productively and latently infected cells creates a mechanism by which the productively infected cells can replenish both populations and maintain two sources of virus from which HIV infection can rebound, even if ART is instituted at the earliest stage of detectable infection., Introduction From the beginning of HIV research, HIV has been mainly propagated in vitro in tissue cultures of activated [CD4.sup.+] T cells (1), leading to the prevailing view that activated [...]

22. Curcumin-loaded polyvinyl butyral film with antibacterial activity

Abstract

Antibacterial materials have found widespread interest in different fields nowadays. In this study, curcumin (Cur) was incorporated into the polyvinyl butyral (PVB) matrix by dissolving in ethanol for improving the functional properties of a pure PVB film. We found that Cur was uniformly dispersed in the PVB matrix, which showed good compatibility. Moreover, the incorporation of Cur could also improve thermal stability, hydrophilicity, and mechanical property. The UV-vis spectra of the PVB–Cur film demonstrated that the film could block ultraviolet radiation. Subsequently, the antibacterial activity of the PVB–Cur film was measured by the colony-counting method against S. aureus and E. coli. The results showed that the PVB–Cur film exhibited good antibacterial activity. Therefore, the PVB–Cur film was considered as a promising material for food and medical packaging applications.

Abstract

Despite the significant advancements in hydrogel materials, contemporary hydrogel contact lenses still need to protect against ultraviolet rays. This study explores the impact of silane-modified silica nanoparticles (SMSi nanoparticles) and N-vinylpyrrolidone (NVP) on the properties of hydroxyethyl methacrylate (HEMA) corneal contact lenses. The average diameter of SMSi nanoparticles was 300 ± 100 nm, and in situ copolymerization was employed to create poly-(HEMA-NVP-SMSi) hydrogel contact lenses. The enhanced ultraviolet–visible (UV)-absorbing hydrogel materials were analyzed for their structural, optical, and performance properties, including water content, oxygen permeability, and stability. The findings indicated that NVP increased the hydrophilicity of the material while the silica nanoparticles effectively absorbed ultraviolet rays and underwent modifications to reduce aggregation. Moreover, the oxygen permeability of the hydrogel lenses is more than double that of pure pHEMA lenses, demonstrating excellent stability and safety. [ABSTRACT FROM AUTHOR]
