The link beams of eccentrically braced steel frames are relied on for energy dissipation through inelastic deformation, but the frames and braces should be elastic enough to resist the strain hardening effect of the link. To reduce steel consumption, high-strength steel with 460 MPa nominal yield stress is used in frames and braces, while normal-strength steel with 345 MPa nominal yield stress is used in vertical links. The seismic behaviour of high-strength eccentrically braced steel frames was investigated in this study. Two half-scale specimens were tested under cyclic loading; the only difference between the two specimens was the link length. The failure modes of the structure, the ductility and energy dissipation capacity were analysed. The experiments indicated that the main failures were concentrated in the short vertical link, while the frames and braces kept their elastic status. The bearing capacity of the shear yielding specimen was higher than the flexural yielding specimen, the ductility of the former was better than the latter and more energy was absorbed in the former than in the latter at the same displacement. The effects of nitrogen (N) forms (ammonium- or nitrate-N) on plant growth under salinity stress [150 mmol sodium chloride (NaCl)] were studied in hydroponically cultured cotton. Net fluxes of sodium (Na+), ammonium (NH4+), and nitrate (NO3−) were also determined using the Non-Invasive Micro-Test Technology. Plant growth was impaired under salinity stress, but nitrate-fed plants were less sensitive to salinity than ammonium-fed plants due mainly to superior root growth by the nitrate-fed plants. The root length, root surface area, root volume, and root viability of seedlings treated with NO3-N were greater than those treated with NH4-N with or without salinity stress. Under salinity stress, the Na+content of seedlings treated with NO3-N was lower than that in seedlings treated with NH4-N owing to higher root Na+efflux. A lower net NO3−efflux was observed in roots of nitrate-fed plants relative to the net NH4+efflux from roots of ammonium-fed plants. This resulted in much more nitrogen accumulation in different tissues, especially in leaves, thereby enhancing photosynthesis in nitrate-fed plants under salinity stress. In order to study the seismic behavior, four one-bay and one-story 1/2 scaled plane specimens were tested under monotonic and cyclic loading respectively. According to the length of link, the specimens were classified into two group; flexural yielding and shear yielding. The failure modes and the main seismic behavior indices of specimens with various link lengths of energy disspation were analyzed. The results indicate that this new structural system is good at energy dissipation and ductility. The bearing capacity of shear yielding specimens is higher than flexural yielding specimens, the ductility of the former is better than the latter, and the energy absorbed in the former is bigger than the latter at the same displacement. In cyclic testing, the main failures of shear yielding specimens were concentrated at links, while the other parts of eccentrically braced frame kept in elastic status. Abstract: Cold is an environmental factor that limits the growing season of cucumber and adversely affects fruit quality and productivity. ICE1 is a positive regulator of CBF3 and has a critical role in cold stress (). To test the function and potential use of cold-induced transcriptome regulator ICE1 (inducer of CBF expression) in improving the chill tolerance of cucumbers, the Arabidopsis ICE1 gene driven by super promoter was introduced into cucumbers by Agrobacterium-mediated transformation. Transgenic plants were obtained, and were confirmed by PCR and southern blot, then the expression of ICE1 was tested by RT-PCR. Transgenic plants exhibited dwarf phenotypes, and shorter internodes, with higher nodes in the first female flowers and a lower female flower rate of every node than wild type plants. Under cold stress, the chilling index of four transgenic lines (IF3, IF6, IF7, and IF10) was decreased significantly, indicating their improved chilling tolerance. Over-expression of ICE1 in cucumbers could induce the expression of cold-responsive genes, further activating the accumulation of soluble sugars and free proline, and inhibiting malondialdehyde (MDA) accumulation. Summary: Coronatine (COR) is a chlorosis-inducing phytotoxin that mimics some biological activities of methyl jasmonate. This study investigated whether COR confers salinity tolerance to cotton and whether such tolerance is correlated with changes in the activity of antioxidant enzymes. COR at 0.01μM was applied hydroponically to cotton seedlings at the two-leaf stage for 24h. A salinity stress of 150mM NaCl was imposed after completion of COR treatment for 15d. Salinity stress reduced biomass of seedlings and increased leaf superoxide radicals, hydrogen peroxide, lipid peroxidation, and electrolyte leakage. Activities of the antioxidant enzymes superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and glutathione reductase (GR), and of the stable free radical, 1,1-diphenyl-2-picrylhydrazyl (DPPH), scavenging activity were altered by salinity to varying degrees. Pretreatment with COR increased the activities of CAT, POD, GR, and DPPH scavenging activity in leaf tissues of salinity-stressed seedlings. Thus, COR might reduce the production of reactive oxygen species by activating antioxidant enzymes and DPPH-radical scavenging, thereby preventing membrane peroxidation and denaturation of bio-molecules. Summary: This study investigated whether uniconazole confers drought tolerance to soybean and if such tolerance is correlated with changes in photosynthesis, hormones and antioxidant system of leaves. Soybean plants were foliar treated with uniconazole at 50mgL−1 at the beginning of bloom and then exposed to water deficit stress at pod initiation for 7d. Uniconazole promoted biomass accumulation and seed yield under both water conditions. Plants treated with uniconazole showed higher leaf water potential only in water-stressed condition. Water stress decreased the chlorophyll content and photosynthetic rate, but those of uniconazole-treated plants were higher than the stressed control. Uniconazole increased the maximum quantum yield of photosystemand ribulose-1,5-bisphosphate carboxylase/oxygenase activity of water-stressed plants. Water stress decreased partitioning of assimilated 14C from labeled leaf to the other parts of the plant. In contrast, uniconazole enhanced translocation of assimilated 14C from labeled leaves to the other parts, except stems, regardless of water treatment. Uniconazole-treated plants contained less GA3, GA4 and ABA under well-watered condition than untreated plants, while the IAA and zeatin levels were increased substantially under both water conditions, and ABA concentration was also increased under water stressed condition. Under water-stressed conditions, uniconazole increased the content of proline and soluble sugars, and the activities of superoxide dismutase and peroxidase in soybean leaves but not the malondialdehyde content or electrical conductivity. These results suggest that uniconazole-induced tolerance to water deficit stress in soybean was related to the changes of photosynthesis, hormones and antioxidant system of leaves. Weed tolerance of UV-B radiation varies with species, and the radiation could affect weed ecology and management. Variations in growth, photosynthesis and defense system among four important agronomic weeds, Abutilon theophrasti Medik, Amaranthus retroflexus L., Digitaria sanguinalis (L.) Scop and Chloris virgata Swartz, under increased UV-B radiation (ambient and increased radiation at 2.7, 5.4 and 10.8 kJ-m−2-d−1) were studied in the greenhouse experiment. After 2 weeks of radiation, the shoots’dry mass decreased with increasing UV-B radiation except for D. sanguinalis. The reduction in biomass was the result of changes in morphology and physiology. Higher levels of UV-B treatment decreased the leaf area, plant height, net photosynthetic rate and chlorophyll contents, while it increased the contents of wax and UV-B absorbing compound in all species, except for A. retroflexus, which did not increase significantly. The activity of superoxide dismutase, catalase, ascorbate peroxide and the content of ascorbic acid changed differently among the weed species as UV-B radiation increased. D. sanguinalis was the most tolerant and A. retroflexus the most sensitive to increased UV-B radiation. The results also show that the two grass species (D. sanguinalis and C. virgata) were more tolerant to UV-B radiation than the two broad-leafed species (A. theophrasti and A. retroflexus). The UV-B absorbing compound and leaf wax played important roles against UV-B damages in the two grass weeds. A 1050 bp fragment upstream transcription start site of a transcription factor gene NAC1 in Arabidopsis thaliana was amplified and cloned into plasmid pRD420 to construct a green fluorescent protein(GFP) fusion system under the control of NAC1 promoter. Plasmids were introduced into tobacco by Agrobacterium-mediated method to regenerate plains with NAC1-GFP gene, and expression pattern of NAC1-GFP and its responses to auxin and gibberellin (GA) were observed. GFP was found to accumulate specifically in root, and was detected after treatment of auxin, N-1-Naphthylphthalamic acid (NPA, an auxin antagonist) or GA3. It was indicated that the expression of GFP driven by NAC1 promoter was induced not only by auxin but also by Gas, suggesting that NAC1 mediated both the auxin signaling and the GAs signaling involved in lateral roots development. Abstract: Seedling responses of two transgenic Bacillus thuringiensis (Bt) cotton (Gossypium hirsutum L.) cultivars, 99B and SGK9708-41, to salinity stress were studied under 0, 50, 100 and 200mM NaCl treatments. Cotton seedling growth, indicated by height, leaf area, and fresh and dry weights, of both cultivars was reduced by NaCl in a concentration-dependent manner. Total N concentrations of seedling roots, stems and leaves of both cotton cultivars decreased with increasing NaCl concentrations. For both cultivars, soluble protein content increased in leaves and decreased in stems due to salt stress, and decreased in roots by 50 and 100mM NaCl. Bt protein concentrations in leaves of both cultivars were decreased by salt stress, but still were biologically effective to control the neonate bollworm larvae in seedling cotton. Strigolactones (SLs) are plant hormones that regulate several key agronomic traits, including shoot branching, leaf senescence, and stress tolerance. The artificial regulation of SL biosynthesis and signaling has been considered as a potent strategy in regulating plant architecture and combatting the infection of parasitic weeds to help improve crop yield. DL1b is a previously reported SL receptor inhibitor molecule that significantly promotes shoot branching. Here, we synthesized 18 novel compounds based on the structure of DL1b. We performed rice tillering activity assay and selected a novel small molecule, C6, as a candidate SL receptor inhibitor. In vitro bioassays demonstrated that C6 possesses various regulatory functions as an SL inhibitor, including inhibiting germination of the root parasitic seeds Phelipanche aegyptiaca, delaying leaf senescence and promoting hypocotyl elongation of Arabidopsis. ITC analysis and molecular docking experiments further confirmed that C6 can interact with SL receptor proteins, thereby interfering with the binding of SL to its receptor. One of the most destructive diseases, Gibberella stalk rot (GSR), caused by Fusarium graminearum, reduces maize yields significantly. An induced resistance response is a potent and cost-effective plant defense against pathogen attack. The functional counterpart of JAs, coronatine (COR), has attracted a lot of interest recently due to its ability to control plant growth and stimulate secondary metabolism. Although several studies have focused on COR as a plant immune elicitor to improve plant resistance to pathogens, the effectiveness and underlying mechanisms of the suppressive ability against COR to F. graminearum in maize have been limited. We investigated the potential physiological and molecular mechanisms of COR in modulating maize resistance to F. graminearum. COR treatment strongly enhanced disease resistance and promoted stomatal closure with H2O2 accumulation, and 10 μg/mL was confirmed as the best concentration. COR treatment increased defense-related enzyme activity and decreased the malondialdehyde content with enhanced antioxidant enzyme activity. To identify candidate resistance genes and gain insight into the molecular mechanism of GSR resistance associated with COR, we integrated transcriptomic and metabolomic data to systemically explore the defense mechanisms of COR, and multiple hub genes were pinpointed using weighted gene correlation network analysis (WGCNA). We discovered 6 significant modules containing 10 candidate genes: WRKY transcription factor (LOC100279570), calcium-binding protein (LOC100382070), NBR1-like protein (LOC100275089), amino acid permease (LOC100382244), glutathione S-transferase (LOC541830), HXXXD-type acyl-transferase (LOC100191608), prolin-rich extensin-like receptor protein kinase (LOC100501564), AP2-like ethylene-responsive transcription factor (LOC100384380), basic leucine zipper (LOC100275351), and glycosyltransferase (LOC606486), which are highly correlated with the jasmonic acid–ethylene signaling pathway and antioxidants. In addition, a core set of metabolites, including alpha-linolenic acid metabolism and flavonoids biosynthesis linked to the hub genes, were identified. Taken together, our research revealed differentially expressed key genes and metabolites, as well as co-expression networks, associated with COR treatment of maize stems after F. graminearum infection. In addition, COR-treated maize had higher JA (JA-Ile and Me-JA) levels. We postulated that COR plays a positive role in maize resistance to F. graminearum by regulating antioxidant levels and the JA signaling pathway, and the flavonoid biosynthesis pathway is also involved in the resistance response against GSR. The lengths of the basal internodes is an important factor for lodging resistance of maize (Zea mays). In this study, foliar application of coronatine (COR) to 10 cultivars at the V8 growth stage had different suppression effects on the length of the eighth internode, with three being categorized as strong-inhibition cultivars (SC), five as moderate (MC), and two as weak (WC). RNA-sequencing of the eighth internode of the cultivars revealed a total of 7895 internode elongation-regulating genes, including 777 transcription factors (TFs). Genes related to the hormones cytokinin, gibberellin, auxin, and ethylene in the SC group were significantly down-regulated compared to WC, and more cell-cycle regulatory factors and cell wall-related genes showed significant changes, which severely inhibited internode elongation. In addition, we used EMSAs to explore the direct regulatory relationship between two important TFs, ZmABI7 and ZmMYB117, which regulate the cell cycle and cell wall modification by directly binding to the promoters of their target genes ZmCYC1 , ZmCYC3 , ZmCYC7 , and ZmCPP1. The transcriptome reported in this study will provide a useful resource for studying maize internode development, with potential use for targeted genetic control of internode length to improve the lodging resistance of maize. Strigolactones (SLs) are a class of plant hormones and rhizosphere communication signals of great interest. They perform diverse biological functions including the stimulation of parasitic seed germination and phytohormonal activity. However, their practical use is limited by their low abundance and complex structure, which requires simpler SL analogues and mimics with maintained biological function. Here, new, hybrid-type SL mimics were designed, derived from Cinnamic amide, a new potential plant growth regulator with good germination and rooting-promoting activities. Bioassay results indicated that compound 6 not only displayed good germination activity against the parasitic weed O. aegyptiaca with an EC50 value of 2.36 × 10−8 M, but also exhibited significant inhibitory activity against Arabidopsis root growth and lateral root formation, as well as promoting root hair elongation, similar to the action of GR24. Further morphological experiments on Arabidopsis max2-1 mutants revealed that 6 possessed SL-like physiological functions. Furthermore, molecular docking studies indicated that the binding mode of 6 was similar to that of GR24 in the active site of OsD14. Aims: In this study, a novel plant growth regulator 2,4-dichloroformamide cyclopropane acid (B2) was used to explore its effects on wheat stalk morphology, chemical composition, anatomical structure, crushing strength and their relationship with lodging resistance. The effects of stem quality change on grain filling and yield were further discussed. Methods: We conducted a series of experiments, including determinations of internode morphology and breaking resistance, analyses of chemical composition and hormone content, observation of internode anatomy, and measurement of grain filling rate, to explore the effects of internode developmental changes induced by B2 treatment on grain filling. Results: Spraying 150 μmol·L−1 B2 at jointing stage significantly decreased plant height, height of center of gravity, basal internode and cell length, while increased stem diameter, wall thickness of basal second internode, cortex thickness, vascular bundle transverse section area and cell cross-sectional area, which resulted in higher breaking strength and culm lodging resistance index. B2 also significantly reduced the content of gibberellin in the second basal internode, and increased the accumulation of lignin and cellulose, thus forming a shorter and stronger stalk. Correlation analysis showed that stem outer diameter, stem wall thickness, cellulose and lignin content were significantly and positively correlated with stem breaking strength. The improvement of stalk structure, especially vascular bundle area, increased bleeding sap, which contributed to grain filling and final yield formation. Conclusions: Lodging resistance and average grain-filling rate can be enhanced by spraying B2 at wheat jointing stage due to the improvement of culm structure, which is conducive to further increase of yield. The use of 1,1-dimethyl-piperidinium chloride (DPC) in Xinjiang is one of the necessary measures for regulating cotton populations and optimizing canopy structure. However, mechanical spraying involves operation and energy consumption, which can alternatively be achieved via drip application. In the present study, to investigate the effect of drip application, two types of hydrophobic nano-released DPCs were used to regulate plant type at different dosages. DPC dripingation reduced plant height by more than 10 cm and plant width by more than 4 cm, and the effect improved with increasing concentration and times. The main effect of height control was at the 6th–10th nodes of the main stem, and the effect of width control was observed at the 1st and 2nd nodes of the fruit branch. The SPAD value was higher than that in the control group during the initial stages. An irregular downward trend was observed in the subsequent stages. The proline content was higher than that of the control and increased with higher concentrations of DPC. There was no significant difference in the soil DPC content at the different sites. The DPC content decreased by more than 30% at the seventh day after dripping, and the content was 2–4 μg·g−1 in each treatment. At 15 days after application, the soil DPC was lower than 1 μg·g−1 in all treatments. The number of harvested plants was more than 150,000 plants·hm−2, and the number of bolls per plant increased at least by one, compared with the control. The final yield of seed cotton increased by at least 300 kg·hm−2, with a maximum increase of 1672.01 kg·hm−2. Considering the different types of DPCs, nano-released types worked better than the conventional type in terms of plant height, plant width, degradation in soil and boll formation. Water uptake is crucial for crop growth and development and drought stress tolerance. The water channel aquaporins (AQP) play important roles in plant water uptake. Here, we discovered that a jasmonic acid analog, coronatine (COR), enhanced maize (Zea mays) root water uptake capacity under artificial water deficiency conditions. COR treatment induced the expression of the AQP gene Plasma membrane intrinsic protein 2;5 (ZmPIP2;5). In vivo and in vitro experiments indicated that COR also directly acts on ZmPIP2;5 to improve water uptake in maize and Xenopus oocytes. The leaf water potential and hydraulic conductivity of roots growing under hyperosmotic conditions were higher in ZmPIP2;5‐overexpression lines and lower in the zmpip2;5 knockout mutant, compared to wild‐type plants. Based on a comparison between ZmPIP2;5 and other PIP2s, we predicted that COR may bind to the functional site in loop E of ZmPIP2;5. We confirmed this prediction by surface plasmon resonance technology and a microscale thermophoresis assay, and showed that deleting the binding motif greatly reduced COR binding. We identified the N241 residue as the COR‐specific binding site, which may activate the channel of the AQP tetramer and increase water transport activity, which may facilitate water uptake under hyperosmotic stress. In recent years, Spodoptera frugiperda (S. frugiperda, Smith) has invaded China, seriously threatening maize production. To explore an effective method to curb the further expansion of the harm of the S. frugiperda, this experiment used maize seedlings of the Zhengdan 958 three-leaf stage (V3) of maize as the material to study the effect of coronatine (COR) on the ability of maize to resist insects (S. frugiperda) at the seedling stage. The results showed that when maize was sprayed with 0.05 μM COR, the newly incubated larvae of S. frugiperda had the least leaf feeding. It was found that 0.05 μM COR significantly increased the contents of abscisic acid (ABA) and jasmonate (JA) in maize leaves through the determination of hormone content. Moreover, transcriptome sequencing revealed that the expression of six genes (ZmBX1, ZmBX2, ZmBX3, ZmBX4, ZmBX5 and ZmBX6), which are associated with the synthesis of benzoxazinoid, were upregulated. Nine genes (ZmZIM3, ZmZIM4, ZmZIM10, ZmZIM13, ZmZIM18, ZmZIM23, ZmZIM27, ZmZIM28 and ZmZIM38) of JA-Zim Domain (JAZ) protein in the JA signal pathway, and seven genes (ZmPRH19, ZmPRH18, Zm00001d024732, Zm00001d034109, Zm00001d026269, Zm00001d028574 and Zm00001d013220) of ABA downstream response protein Group A Type 2C Protein Phosphatase (PP2C) were downregulated. These results demonstrated that COR could induce anti-insect factors and significantly improve insect resistance in seedling maize, which laid a theoretical foundation for further study of the mechanism of COR improving insect resistance in seedling maize, and provided data references for the use of COR as an environmentally friendly pest control method. Botrytis cinerea, the causal agent of gray mold, is one of the most destructive pathogens of cherry tomatoes, causing fruit decay and economic loss. Fludioxonil is an effective fungicide widely used for crop protection and is effective against tomato gray mold. The emergence of fungicide-resistant strains has made the control of B. cinerea more difficult. While the genome of B. cinerea is available, there are few reports regarding the large-scale functional annotation of the genome using expressed genes derived from transcriptomes, and the mechanism(s) underlying such fludioxonil resistance remain unclear. The present study prepared RNA-sequencing (RNA-seq) libraries for three B. cinerea strains (two highly resistant (LR and FR) versus one highly sensitive (S) to fludioxonil), with and without fludioxonil treatment, to identify fludioxonil responsive genes that associated to fungicide resistance. Functional enrichment analysis identified nine resistance related DEGs in the fludioxonil-induced LR and FR transcriptome that were simultaneously up-regulated, and seven resistance related DEGs down-regulated. These included adenosine triphosphate (ATP)-binding cassette (ABC) transporter-encoding genes, major facilitator superfamily (MFS) transporter-encoding genes, and the high-osmolarity glycerol (HOG) pathway homologues or related genes. The expression patterns of twelve out of the sixteen fludioxonil-responsive genes, obtained from the RNA-sequence data sets, were validated using quantitative real-time PCR (qRT-PCR). Based on RNA-sequence analysis, it was found that hybrid histidine kinase, fungal HHKs, such as BOS1, BcHHK2, and BcHHK17, probably involved in the fludioxonil resistance of B. cinerea, in addition, a number of ABC and MFS transporter genes that were not reported before, such as BcATRO, BMR1, BMR3, BcNMT1, BcAMF1, BcTOP1, BcVBA2, and BcYHK8, were differentially expressed in the fludioxonil-resistant strains, indicating that overexpression of these efflux transporters located in the plasma membranes may associate with the fludioxonil resistance mechanism of B. cinerea. All together, these lines of evidence allowed us to draw a general portrait of the anti-fludioxonil mechanisms for B. cinerea, and the assembled and annotated transcriptome data provide valuable genomic resources for further study of the molecular mechanisms of B. cinerea resistance to fludioxonil. Rice crab co-culture is a new integrated farming model in China. The application of triazole plant growth regulators (PRGs) is often used as an advantageous option to combat rice lodging. However, there is still a gap regarding the toxicity of these PRGs on the growth and development of the Chinese mitten crab (Eriocheir sinensis, E. sinensis). Here the effect of triazoles (paclobutrazol and uniconazole) on the molting mechanism of E. sinensis was investigated. Monitoring of regulatory genes associated with molting showed that the two PRGs were found to inhibit the expression of ecdysteroid hormone (EH), ecdysteroid receptors gene (EcR), and retinoid X receptors gene (RXR) and induce secretion of molt-inhibiting hormone (MIH) gene. In addition, the activities of chitinase (CHIA) and N-acetyl-β- d -aminoglucosidase (β-NAGase) were also inhibited by exposure to PRGs. Exposure to PRGs also elevated the mRNA expression of the growth-related myostatin gene (MSTN). Low temperature is an important environmental factor limiting the widespread planting of tropical and subtropical crops. The application of plant regulator coronatine, which is an analog of Jasmonic acid (JA), is an effective approach to enhancing crop's resistance to chilling stress and other abiotic stresses. However, the function and mechanism of coronatine in promoting chilling resistance of tomato is unknown. In this study, coronatine treatment was demonstrated to significantly increase tomato chilling tolerance. Coronatine increases H3K4me3 modifications to make greater chromatin accessibility in multiple chilling-activated genes. Corresponding to that, the expression of CBFs, other chilling-responsive transcription factor (TF) genes, and JA-responsive genes is significantly induced by coronatine to trigger an extensive transcriptional reprogramming, thus resulting in a comprehensive chilling adaptation. Soil salinity is one of the most critical factors limiting plant growth and development. Carex leucochlora is an important turfgrass species with a wide distribution in northern China that is highly sensitive to salt stress, which impairs its development. Recently, melatonin has emerged as a nontoxic biomolecule that regulates growth and enhances salt tolerance in plants. In this study, the mechanism of melatonin's regulation of plant growth and anatomical characteristics in C. leucochlora seedlings under continuous salt stress was explored. Our results indicated that salt stress strongly suppressed plant growth and leaf cell activity, inhibited root morphology and root activity, and negatively affected leaf and root anatomic structures in the seedlings. Conversely, melatonin (150 μmol L−1) pretreatment improved the detrimental effect of salt stress by restoring the morphology of the leaf, alleviating damage to the cell membrane, improving root activity, and altering the root architecture and plant growth attributes. Moreover, after 12 days of salt stress, anatomical observations of the leaf showed that the thickness of the leaf blade, vascular bundle area of the leaf main vein, vesicular cell area, thickness of the upper epidermis, and thickness of the lower epidermis were increased by 30.55, 15.63, 12.60, 16.76 and 27.53%, respectively, with melatonin under salinity. Melatonin treatment also showed an increase of 5.91, 7.59, 15.57, and 20.51% in epidermal thickness, vascular cylinder diameter, xylem vessel diameter, and pith cell diameter, respectively, compared with salt stress after 12 days. These results suggest that melatonin alleviated salt stress through augmenting seedling growth, leaf cell activity, and root characteristics, maintained the stability of anatomic traits to maintain chloroplast cell homeostasis, and also protected the vascular tissues to promote the radial transport of water and ions in the C. leucochlora seedlings. Melatonin (MT) can effectively reduce oxidative damage induced by abiotic stresses such as salt in plants. However, the effects of MT on physiological responses and molecular regulation during wheat germination remains largely elusive. In this study, the response of wheat seeds to MT under salt stress during germination was investigated at physiological and transcriptome levels. Our results revealed that application of MT significantly reduced the negative influence of salt stress on wheat seed germination. The oxidative load was reduced by inducing high activities of antioxidant enzymes. In parallel, the content of gibberellin A3 (GA3) and jasmonic acid (JA) increased in MT-treated seedling. RNA-seq analysis demonstrated that MT alters oxidoreductase activity and phytohormone-dependent signal transduction pathways under salt stress. Weighted correlation network analysis (WGCNA) revealed that MT participates in enhanced energy metabolism and protected seeds via maintained cell morphology under salt stress during wheat seed germination. Our findings provide a conceptual basis of the MT-mediated regulatory mechanism in plant adaptation to salt stress, and identify the potential candidate genes for salt-tolerant wheat molecular breeding. Background: Low grain water content (GWC) at harvest of maize (Zea mays L.) is essential for mechanical harvesting, transportation and storage. Grain drying rate (GDR) is a key determinant of GWC. Many quantitative trait locus (QTLs) related to GDR and GWC have been reported, however, the confidence interval (CI) of these QTLs are too large and few QTLs has been fine-mapped or even been cloned. Meta-QTL (MQTL) analysis is an effective method to integrate QTLs information in independent populations, which helps to understand the genetic structure of quantitative traits. Results: In this study, MQTL analysis was performed using 282 QTLs from 25 experiments related GDR and GWC. Totally, 11 and 34 MQTLs were found to be associated with GDR and GWC, respectively. The average CI of GDR and GWC MQTLs was 24.44 and 22.13 cM which reduced the 57 and 65% compared to the average QTL interval for initial GDR and GWC QTL, respectively. Finally, 1494 and 5011 candidate genes related to GDR and GWC were identified in MQTL intervals, respectively. Among these genes, there are 48 genes related to hormone metabolism. Background: MicroRNA (miRNA) plays vital roles in the regulation of both plant architecture and stress resistance through cleavage or translation inhibition of the target messenger RNAs (mRNAs). However, miRNA-induced gene silencing remains a major challenge in vivo due to the low delivery efficiency and instability of miRNA, thus an efficient and simple method is urgently needed for miRNA transformation. Previous researches have constructed a star polycation (SPc)-mediated transdermal double-stranded RNA (dsRNA) delivery system, achieving efficient dsRNA delivery and gene silencing in insect pests. Results: Here, we tested SPc-based platform for direct delivery of double-stranded precursor miRNA (ds-MIRNA) into protoplasts and plants. The results showed that SPc could assemble with ds-MIRNA through electrostatic interaction to form nano-sized ds-MIRNA/SPc complex. The complex could penetrate the root cortex and be systematically transported through the vascular tissue in seedlings of Arabidopsis and maize. Meanwhile, the complex could up-regulate the expression of endocytosis-related genes in both protoplasts and plants to promote the cellular uptake. Furthermore, the SPc-delivered ds-MIRNA could efficiently increase mature miRNA amount to suppress the target gene expression, and the similar phenotypes of Arabidopsis and maize were observed compared to the transgenic plants overexpressing miRNA. Conclusion: To our knowledge, we report the first construction and application of star polycation nanocarrier-based platform for miRNA delivery in plants, which explores a new enable approach of plant biotechnology with efficient transformation for agricultural application. The aim of this study was to investigate the potential of silicon (Si) for alleviating Ultraviolet-B (UV-B) radiation stress based on changes in biomass, physiological attributes and photosynthetic characteristics of two soybean (Glycine maxL.) cultivars, Kenjiandou 43 (‘K 43’) and Zhonghuang 35 (‘ZH 35’). The cultivars were raised with and without Si in the greenhouse, and then subjected to ambient, ambient + 2.7 kJ m−2d−1and ambient + 5.4 kJ m−2d−1of UV-B radiation. Depending on cultivar, plants suffered severe growth limitations under UV-B radiation, but the application of Si alleviated the adverse effects on growth and development by increasing the stem length, net photosynthetic rate (PN) and leaf chlorophyll content. Concurrently, it decreased the stomatal conductance (Sc) and intercellular carbon dioxide (CO2) concentration (Ci). In response to the UV-B radiation stress, the antioxidant enzyme activities of superoxide dismutase (SOD) increased by 41.2–72.7%, peroxidases (POD) by 49.5–85.7%, malodialdehyde (MDA) by 6.7–20.4% and soluble protein by 4.2–7.6%. The overall results indicated that media treatment with Si might improve soybean growth under elevated UV-B radiation through positive changes in biomass and some physiological attributes that were dependent on cultivar. The aim of this study was to investigate the effect of silicon (Si) on soybean seedlings under (combined) drought and ultraviolet-B (UV-B) radiation stresses. Specifically, we determined dry matter accumulation and partitioning of the mineral elements in different organs of soybean seedlings using the inductively coupled plasma mass spectrometry (ICP-MS). The amount of dry matter accumulated in root, stem, and leaf of seedlings treated with (combined) drought and UV-B radiation stresses were lower than that of the control. Also, the content of macroelements in seedlings under the combined stress was lower than in those under control conditions. Changes in the contents of microelements varied according to the intensity of drought, UV-B radiation, and the organs of soybean. The relationship between the contents of mineral elements and the accumulation of dry matter also varied similarly. These results demonstrated that drought and UV-B radiation induced an alteration in the distribution of mineral elements in root, stem and leaf, leading to decreases in dry matter accumulation and inhibition of soybean growth. Drought is one of the most common environmental stressors that severely threatens plant growth, development, and productivity. B2 (2,4-dichloroformamide cyclopropane acid), a novel plant growth regulator, plays an essential role in drought adaptation, significantly enhancing the tolerance of Carex breviculmis seedlings. Its beneficial effects include improved ornamental value, sustained chlorophyll content, increased leaf dry weight, elevated relative water content, and enhanced root activity under drought conditions. B2 also directly scavenges hydrogen peroxide and superoxide anion contents while indirectly enhancing the activities of antioxidant enzymes (superoxide dismutase, peroxidase, catalase, and ascorbate peroxidase) to detoxify reactive oxygen species (ROS) oxidative damage. Transcriptome analysis demonstrated that B2 activates drought-responsive transcription factors (AP2/ERF-ERF, WRKY, and mTERF), leading to significant upregulation of genes associated with phenylpropanoid biosynthesis (HCT, POD , and COMT). Additionally, these transcription factors were found to suppress the degradation of starch. B2 regulates phytohormone signaling related-genes, leading to an increase in abscisic acid contents in drought-stressed plants. Coronatine, an analog of Jasmonic acid (JA), has been shown to enhance crop tolerance to abiotic stresses, including chilling stress. However, the underlying molecular mechanism remains largely unknown. In this study, we investigated the effect of Coronatine on cotton seedlings under low temperature using transcriptomic and metabolomics analysis. Twelve cDNA libraries from cotton seedlings were constructed, and pairwise comparisons revealed a total of 48,322 differentially expressed genes (DEGs). Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis identified the involvement of these unigenes in various metabolic pathways, including Starch and sucrose metabolism, Sesquiterpenoid and triterpenoid biosynthesis, Phenylpropanoid biosynthesis, alpha-Linolenic acid metabolism, ABC transporters, and Plant hormone signal transduction. Additionally, substantial accumulations of jasmonates (JAs), abscisic acid and major cell wall metabolites were observed. Transcriptome analysis revealed differential expression of regulatory genes, and qRT-PCR analysis confirmed the expression patterns of 9 selected genes. Co-expression analysis showed that the JA-responsive genes might form a network module with ABA biosynthesis genes or cell wall biosynthesis genes, suggesting the existence of a COR-JA-cellulose and COR-JA-ABA-cellulose regulatory pathway in cotton seedlings. Plant Growth Regulators (PGRs) are functional compounds known for enhancing plant growth and development. However, their environmental impact is a concern due to poor water solubility and the need for substantial organic solvents. Recently, nano-delivery systems have emerged as a solution, offering a broad range of applications for small molecule compounds. This study introduces a nano-delivery system for Triacontanol (TA), utilizing a star polymer (SPc), aimed at promoting maize growth and improving physiological indicators. The system forms nearly spherical nanoparticles through TA's hydroxyl group and SPc's tertiary amine group. The TA/SPc nano-complex notably outperforms separate TA or SPc treatments in maize, increasing biomass, chlorophyll content, and nutrient absorption. It elevates chlorophyll content by 16.4%, 10.0%, and 6.2% over water, TA, and SPc treatments, respectively, and boosts potassium and nitrate ion uptake by up to 2 and 1.6 times compared to TA alone, leading to enhanced plant height and leaf growth. qRT-PCR analysis further demonstrated that the nano-complex enhanced cellular uptake through the endocytosis pathway by up-regulating endocytosis-related gene expression. The employment of TEM to observe vesicle formation during the internalization of maize leaves furnishes corroborative evidence for the participation of the endocytosis pathway in this process. Abstract: The oriental tobacco budworm, Helicoverpa assulta, is one of the most destructive pests for numerous commercial crops, and these organisms are responsible for enormous economic losses in Chinese agriculture. Insect larvae often feed within host plant fruits, providing protection from many currently used insecticides and making field control of H. assulta very difficult. Owing to its novel mode of action, high insecticidal activity, and low mammalian toxicity, the nonsystemic insecticide indoxacarb has been considered a promising alternative for the control of lepidopterous pests of agricultural significance. Indoxacarb evidences an elevated insecticidal activity against H. assulta. After 13 generations of selection with indoxacarb and bifenthrin insecticides under laboratory conditions, the LC50 of these compounds for H. assulta increased by 4.19-fold and 10.67-fold, respectively. The synergists diethyl maleate (DEM) and triphenyl phosphate (TPP) increased indoxacarb toxicity by 2.76-fold and 4.10-fold in resistant strains and, comparatively, 1.58-fold and 1.75-fold in susceptible strains, suggesting that carboxylesterase (CarE) and glutathione-S-transferases (GSTs) may be involved in the development of indoxacarb resistance in H. assulta. Activity and kinetic parameters observed in detoxification enzymes further demonstrated that the enhanced activity of CarE and GSTs may be critical in development of indoxacarb resistance in H. assulta. The data provides a foundation for further study of the indoxacarb resistance mechanism observed in H. assulta and the rational use of indoxacarb as a rotation insecticide with other insecticide classes for the control of H. assulta. Abstract: Silicon (Si) may be involved in metabolic, physiological, and/or structural activity in higher plants exposed to abiotic and biotic stresses. This has not yet been determined due to the absence of direct evidence that it is part of the molecule of an essential plant constituent or metabolite. The aim of this study was to investigate the effect of silicon on soybean seedlings under drought and ultraviolet-B (UV-B) radiation stresses. The relative leaf water content (RWC), which was the main factor resulting in reduced growth in response to drought, increased 19.0% and 30.0% with Si application under drought and drought+UV-B stresses, respectively. Under UV-B radiation, the anthocyanin and phenol levels decreased 91.5% and 10.0% in the treatment of Si. Ultraviolet-B radiation and drought stress caused great membrane damage, as assessed by lipid peroxidation and osmolyte leakage, but Si application significantly reduced the membrane damage. Catalase (CAT), peroxidase (POD), superoxide dismutase (SOD) and hydrogen peroxide were observed under stress conditions. Proline increased primarily in drought-stressed seedlings and may be the drought-induced factor with a protective role in response to UV-B and silicon. Photosynthesis (P N) increased following Si application by 21.0%, 18.3% and 21.5% under UV-B radiation, drought and the combination, respectively. The physiological and biochemical parameters measured indicated that the UV-B light had more adverse effects on growth of soybean seedlings than drought, but the data also showed that Si could alleviate seedling damage under these stress conditions. The objective of this study was to determine if treatment with silicon (Si) may improve nutrient uptake by soybean under ultraviolet-B (UV-B) radiation stress. Soybean (Glycine max L.) cultivars, 'Kennong 18' (K 18) and 'Zhonghuang 13' (ZH 13), were grown in hydroponic cultures under ambient and supplemental levels of ultraviolet-B (UV-B, 280-315 nm) with and without Si. Supplemental UV-B radiation simulating 30% stratospheric ozone depletion significantly decreased plant biomass by 74.9 to 135.6%, increased leaf nitrogen (N) and phosphorus (P) by 9% and 16%, respectively, decreased leaf magnesium (Mg) contents by 9%, and calcium (Ca) by 24%. UV-B radiation caused a substantial increase in the allocation of P, potassium (K) and Ca to roots compared with stem and leaves, presumably to ensure sustained nutrient uptake under the stress. Silicon application improved the uptake of P and Mg by 11%, which favored the partitioning of dry mass to shoots under UV-B radiation and the allocation of tissue P and Ca to roots. Bacillus thuringensis (Bt) transgenic (insect-resistant) cotton cultivars senesce prematurely under potassium (K+) deficiency, more often than conventional cultivars, in the North China Plain. To verify if Bt-transgenic cotton was more susceptible to K+ deficit, two transgenic cultivars, 'CCRI 41' and 'DP 99B', and two conventional cultivars, 'CCRI 35' and 'CCRI 36', selected from widely used cultivars in China, were used in a seedling hydroponic study. The culture solution K+ concentration was 0.5 mM for high K+ and 0.02 mM for low K+ conditions. Seedlings of all four cultivars accumulated more dry matter and K+ when grown at high K+ than low K+ conditions. However, under low K+ condition, the dry weight and K+ content of Bt-transgenic cultivars CCRI 41 and DP 99B were lower than those of the conventional cultivars CCRI 36 and CCRI 35. The results indicated that Bt-transgenic cultivars CCRI 41 and DP 99B were more sensitive to K+ deficiency than conventional cultivars CCRI 36 and CCRI 35, which could be the reason for premature senescence symptoms observed from fields of Bt-transgenic cotton under K+ deficiency. Seedlings of all four cultivars had a higher K+ use efficiency (KUE) under low K+ than high K+ conditions, but the KUE did not account for the differential responses between Bt-transgenic and conventional cultivars at the low K+ concentration. The K+ depletion results did not reveal the mechanism for the above differential responses in Vmax and Km of the seedlings either. Conserving irrigation water resources is a most important measure for sustainable wheat production in the North China Plain. In the present study, the effect of phosphorus (P) application for saving irrigation water was evaluated. The application of fertilizer P increased nitrogen and phosphorus uptake, shoot biomass, head number, seed number and, consequently, grain yield, and increased soil water use and seasonal evapo-transpiration. The lower the volume of irrigation water applied, the more obvious were these effects. When winter wheat was basally fertilized with 88.5 kg P 2 O 5 /ha and irrigated with 90 mm at the jointing stage, the highest fertilizer P use efficiency, apparent P recovery and net profit (due to irrigation and/or fertilizer P) were obtained. Potassium deficiency causes severe losses in yield and quality in crops. Mepiquat chloride, a plant growth regulator, can increase K+ uptake in cotton (Gossypium hirsutum), but the underlying physiological mechanisms remain unclear. In this study, we used a non-invasive micro-test technique to measure K+ and H+ fluxes in the root apex with or without inhibitors of K+ channels, K+ transporters, non-selective cation channels, and plasma membrane H+-ATPases. We found that soaking seeds in mepiquat chloride solution increased the K+ influx mediated by K+ channels and reduced the K+ efflux mediated by non-selective cation channels in cotton seedlings. Mepiquat chloride also increased negative membrane potential (E m) and the activity of plasma membrane H+-ATPases in roots, due to higher levels of gene expression and protein accumulation of plasma membrane H+-ATPases as well as phosphorylation of H+-ATPase 11 (GhAHA11). Thus, plasma membrane hyperpolarization mediated by H+-ATPases was able to stimulate the activity of K+ channels in roots treated with mepiquat chloride. In addition, reduced K+ efflux under mepiquat chloride treatment was associated with reduced accumulation of H2O2 in roots. Our results provide important insights into the mechanisms of mepiquat chloride-induced K+ uptake in cotton and hence have the potential to help in improving K nutrition for enhancing cotton production. Background: Lodging is one of the important factors causing maize yield. Plant height is an important factor in determining plant architecture in maize (Zea mays L.), which is closely related to lodging resistance under high planting density. Coronatine (COR), which is a phytotoxin and produced by the pathogen Pseudomonas syringae, is a functional and structural analogue of jasmonic acid (JA). Results: In this study, we found COR, as a new plant growth regulator, could effectively reduce plant height and ear height of both hybrids (ZD958 and XY335) and inbred (B73) maize by inhibiting internode growth during elongation, thus improve maize lodging resistance. To study gene expression changes in internode after COR treatment, we collected spatio-temporal transcriptome of inbred B73 internode under normal condition and COR treatment, including the three different regions of internode (fixed, meristem and elongation regions) at three different developmental stages. The gene expression levels of the three regions at normal condition were described and then compared with that upon COR treatment. In total, 8605 COR-responsive genes (COR-RGs) were found, consist of 802 genes specifically expressed in internode. For these COR-RGs, 614, 870, 2123 of which showed expression changes in only fixed, meristem and elongation region, respectively. Both the number and function were significantly changed for COR-RGs identified in different regions, indicating genes with different functions were regulated at the three regions. Besides, we found more than 80% genes of gibberellin and jasmonic acid were changed under COR treatment. Conclusions: These data provide a gene expression profiling in different regions of internode development and molecular mechanism of COR affecting internode elongation. A putative schematic of the internode response to COR treatment is proposed which shows the basic process of COR affecting internode elongation. Ethephon is plant growth regulator widely used to control maize lodging at the early stage of jointing. However, the function and efficacy of ethephon due to differences of cultivars and maize growing areas remains elusive, necessitating the evaluation of each cultivar's sensitivities prior to the application of ethylene. In this study, we collected ten maize cultivars from the largest planted area in China and cultivated in three typical growing areas to evaluate their sensitivity to ethephon. The results showed that ethephon reduced maize plant height, ear height, length of base internode, and generally increased the crushing strength of maize base internode. These traits may enhance maize lodging resistance by reducing the center of gravity of the plant structure. The effect of ethephon varied across growing areas, and cultivars within the same region exhibited distinct sensitivity levels to ethephon. The crushing strength of the eighth internode in maize exhibits a wide range of change and small coefficient of variation. Hence, this index's change range could be utilized to assess the sensitivity of maize cultivars to ethephon in various regions. In addition to agronomic traits, we detected shifts in transcriptional levels related to the signaling pathways of phytohormones, particularly ethylene and gibberellin. These changes reveal the association between transcriptional signaling levels of key factors and the sensitivity to ethylene in ten maize cultivars. The results contribute to our understanding of the lodging resistance in different maize cultivars and evaluation the sensitivity of maize cultivar to ethephon in different growing areas, with implications for comparable pedo-climatic conditions elsewhere. Maize yield is limited by water deficit in the arid and semiarid regions of China. Conserving water and enhancing crop water productivity are inevitable requirements for the sustainable development of water efficient agriculture. Plant growth regulators (PGRs) play important roles in maize water stress tolerance. In this study, we investigated the effect of plant growth regulator-brazide, a functional analog of brassinolide (BR), on maize water stress resistance through a two-year (2020–2021) field experiment. The experiment included four water treatments (I100: 4012 m3 ha−1; I80: 3210 m3 ha−1; I60: 2407 m3 ha−1; I40: 1605 m3 ha−1), four PGR concentrations (CK: water; Bra: 0.1 μmol L−1 14-hydroxylated brassinosteroid; B1: 0.1 μmol L−1 brazide; B2: 1 μmol L−1 brazide; B3: 10 μmol L−1 brazide). The results showed that deficit irrigation (I60 and I40) decreased soil water content (SWC), photosynthetic rate (Pn), leaf area index (LAI), aboveground biomass (AB) and yield, but improved crop water productivity (WP c). Under deficit irrigation conditions, 1 μmol L−1 brazide application also increased dry matter accumulation and grain yield, a result attributed to improvement of the photosynthetic rate and increases in the root bleeding saps, water productivity. 1 μmol L−1 Brazide application reduced bare tip length, increasing kernel number per ear, thereby increased the yield of I60 and I40 by 15.1% and 11.4% (p ≤ 0.05; 2020), 16.4% and 20.4% (p ≤ 0.001; 2021), respectively. Path analysis showed brazide improved final yields by increasing thousand grain weight (TGW), photosynthetic rate (Pn) and WP c under deficit irrigation. The increase in TGW with brazide treatment was primarily attributed to the increase of Pn. Waterlogging negatively affects maize growth and yield. In this study, we found that ethylene played a vital role in plant adaptation to waterlogging. ET promotes better growth in seedlings under waterlogging conditions by altering root architecture and increasing lateral root formation by 42.1%. What's more, plants with high endogenous ethylene levels exhibited reduced sensitivity to waterlogging stress. ET also induced the formation of aerenchyma, a specialized tissue that facilitates gas exchange, in a different pattern compared to aerenchyma formed under waterlogging. Aerenchyma induced by ET was mainly located in the medial cortex of the roots and was not prone to decay. ethylene inhibited root elongation under normal conditions, but this inhibition was not alleviated under waterlogging stress. Upon activation of the ET signaling pathway, the transcription factor EREB90 promoted aerenchyma formation by enhancing the programmed cell death process. Overexpression of EREB90 resulted in increased waterlogging tolerance compared to wild type plants. Our findings suggest that pre-treatment of maize seedlings with ET before waterlogging stress can trigger the programmed cell death process and induce aerenchyma formation, thus improving waterlogging resistance. Coronatine (COR) is a phytotoxin produced by Pseudomonas syringae . Its structure is similar to those of jasmonates (JAs), which play diverse roles in multiple plant biotic and abiotic defenses. However, the biological activity of COR is 1000 times greater than the activity of JA. In addition to being involved in the JA pathway, COR affects plant photosynthetic efficiency. In this study, we examined wheat blade pretreatment with COR. Blades treated with COR remained green longer than those of control plants under drought stress conditions, resulting in less yield loss with COR treatment. To investigate the mechanism of COR in drought resistance further, we employed two-dimensional gel electrophoresis technology and matrix-assisted laser desorption/ionization mass spectrometry to sequester and identify key proteins. Six COR-inducible proteins that are located in the chloroplast and involved directly in photosynthesis were found. The wheat homologue of protein gi|326509937 is degradation of periplasmic proteins 1 (DEGP1) in Arabidopsis , which is a response to photosystem II reparation, and was maintained at a low level with COR treatment. Finally, we measured levels of chlorophyll and photosynthetic performance to reveal the phenotypic effect of COR. Roots absorb water and nutrients from the soil, give mechanical support to plants, and supply hormones that affect many physiological and biochemical processes. Roots also exert control over whole-plant growth and development by synthesizing plant hormones and controlling the uptake of mineral nutrients. Coronatine (COR) is an analogue of jasmonic acid and is a chlorosis-inducing non-hostspecific phytotoxin with an alleviative effect on plant growth under stress conditions. A 2-year field study was conducted to determine the effect of COR on the physiological and biochemical attributes of the root and the seed yield of soybean (Glycine maxL. Cv. Zhonghuang 13). COR was foliar-applied at 10 nM during the 7-trifoliolate leafs stage. Root dry weight was significantly increased on average by 37% with COR over the 2 years. The root nitrogen concentration increased by 19.2%, phosphorus concentration by 23.3%, and potassium concentration by 31.2% following COR application. The content of cytokinins was increased by 31.5% and 46.0%, while that of abscisic acid was decreased by 45.0% and 25.5% with COR over the 2 years. Ethylene (ET) is critical importance in the growth, development, and stress responses of plants. Plant hormonal stress responses have been extensively studied, however, the role of ET in plant growth, especially plant height (PH) remains unclear. Understanding the genetic control for PH in response to ET will provide insights into the regulation of maize development. To clarify the genetic basis of PH-related traits of maize in response to ET, we mapped QTLs for PH, ear height (EH), and internode length above the uppermost ear (ILAU) in two recombinant inbred line (RIL) populations of Zea mays after ET treatment and in an untreated control (CK) group. Sixty QTLs for the three traits were identified. Twenty-two QTLs were simultaneously detected under both ET treatment and untreated control, and five QTLs were detected at two geographic locations under ET treatment only. Individual QTL can be explained 3.87–17.71% of the phenotypic variance. One QTL (q2PH9-1, q1PH9, q1EH9/q1ILAU9-1, q2ILAU9, and q2EH9) for the measured traits (PH, EH, ILAU) was consistent across both populations. Two QTLs (q2PH2-5, q2ILAU2-2, q1PH2-2, and q1ILAU2-2; q1PH8-1, q1EH8-1, q2PH8-1) were identified for up to two traits in both locations and populations under both ET treatment and untreated control. These consistent and stable regions are important QTLs of potential hot spots for PH, ear height (EH), and internode length above the uppermost ear (ILAU) response to ET in maize; therefore, QTL fine-mapping and putative candidate genes validation should enable the cloning of PH, EH, and ILAU related genes to ET response. These results will be valuable for further fine-mapping and quantitative trait nucleotides (QTNs) determination, and elucidate the underlying molecular mechanisms of ET responses in maize. Increasing planting density is important to raise maize yield, however, high density often leads to an increase risk of lodging due to dense canopy and weak stem. Maize yield and optimal plant density are increased by applying plant growth regulator compound of ethephon and DA-6, however, we do not know if this compound would interact with location and genotype. In this study, a novel plant growth regulator, as the synthesis of N , N - diethyl − 2 − hexanoyl oxygen radicals − ethyl amine (2-ethyl chloride) phosphonic acid salt (DHEAP), combining the effects of ethephon and DA-6 in one chemical, was developed and tested at three locations, five plant densities (6.75, 8.25, 9.75, 11.25 and 12.75 plants m −2 ) and three cultivars in 2014–2015. This study aimed to quantify the interactions between environment, genotype and management (Appling DHEAP and plant density) on lodging-related optimal plant density and yield. DHEAP significantly increased grain yield by 10.7% due to the increases of kernel weight by 3.2% and kernel number per ear by 4.4%. On average across genotypes and environments, applying DHEAP increased optimum plant density by 6%. The optimal plant density interacted with cultivar, DHEAP and environment. Applying DHEAP reduced lodging percentage by lowering ear height. The yield-lodging relationship was affected by genotype and location. We concluded that maize yield could be enhanced by optimizing plant density, applying DHEAP and cultivar selection, but climatic and environmental differences of locations should be considered. Drought is a key limiting factor for cotton ( Gossypium spp.) production, as more than half of the global cotton supply is grown in regions with high water shortage. However, the underlying mechanism of the response of cotton to drought stress remains elusive., By combining genome-wide transcriptome profiling and a loss-of-function screen using virus-induced gene silencing, we identified Gossypium hirsutum Gh WRKY59 as an important transcription factor that regulates the drought stress response in cotton., Biochemical and genetic analyses revealed a drought stress-activated mitogen-activated protein ( MAP) kinase cascade consisting of Gh MAP3K15-Mitogen-activated Protein Kinase Kinase 4 (GhMKK4)-Mitogen-activated Protein Kinase 6 (GhMPK6) that directly phosphorylates Gh WRKY59 at residue serine 221. Interestingly, Gh WRKY59 is required for dehydration-induced expression of Gh MAPK3K15, constituting a positive feedback loop of Gh WRKY59-regulated MAP kinase activation in response to drought stress. Plant growth regulators (PGRs) play crucial roles in maize drought tolerance. However, most of the studies are focused on seedling stage under greenhouse conditions, little is known on the effects of grain yields at later growth stage under field environment. It is hypothesized that exogenous application of PGRs can increase maize drought tolerance at different growth stages under field conditions. Here we used alginate oligosaccharide (AOS) and coronatine (COR) to investigate the effects of these two PGRs on maize drought tolerance in a two-year field experiment. The results showed that both COR and AOS treatments reduced leaf water loss and significantly increased the relative leaf water content by 12.6% and 13.0%, thus significantly decreasing superoxide anion accumulation by 49% and 42% in maize seedlings. There was no big difference between these two treatments in the seedling stage. In the subsequent reproductive growth period, although both COR and AOS treatments significantly increased leaf photosynthesis and above-ground growth, including photosynthetic rate by 41% and 33%, relative chlorophyll content (SPAD value) by 8.0% and 6.5%, leaf area by 22% and 15%, plant height by 8.3% and 5.9%, dry matter accumulation by 23% and 17%, respectively, under drought conditions, the effect of COR was superior to the AOS, as evidenced by membership function values of COR treatment were 0.12 and 0.08 larger than that of AOS treatment in 2020 and 2021, respectively. Ultimately, COR and AOS treatments significantly increased the grain yields by 21.7% and 16.2% in 2020, 22.3% and 15.7% in 2021, respectively, under drought conditions. Path analysis showed both PGRs increased final grain yields by increasing thousand-kernel weight (TKW). While the TKW increase with COR treatment was mainly due to the increase of relative chlorophyll content (SPAD value), AOS treatment mainly increased dry matter accumulation under drought conditions. Maize (Zea mays L.) is a major global crop, but lodging leads to yield loss. Ethephon can regulate maize growth, reduce plant height, and improve lodging resistance. The application of small crop protection unmanned aerial vehicles (UAV) to replace electric knapsack sprayers (EKS) for the mechanization of agriculture in China has been widely developed. However, there are no reports on the application of ethephon to control maize growth by UAV. In the present study, the effects of spraying by UAV (15 L ha−1 and 30 L ha−1) and EKS (450 L ha−1) on the droplet deposition distribution and dynamic degradation process of ethephon (60 g a.i. ha−1) were studied. The control of maize growth by different ethephon dosages was also investigated, i.e., 48, 60, and 90 g a.i. ha−1 for UAV and 60 g a.i. ha−1 for EKS. The droplet deposition rates of 15 L ha−1 and 30 L ha−1 by UAV were 11.2% and 15.2%, significantly higher than those of EKS. Furthermore, the degradation half-life under UAV application was longer than that under EKS. The 20% dosage reduction of ethephon applied by UAV (48 g a.i. ha−1) had the same maize growth control effect as EKS (60 g a.i. ha−1), and the UAV treatment was better than EKS when the dosage was the same (60 g a.i. ha−1). Considering the input cost and yield, we recommend that 48 a.i. g ha−1 ethephon is applied by UAV when the lodging risk is low and 90 a.i. g ha−1 when the lodging risk is high, at a spray volume of 15 L ha−1. Drought is an important factor limiting plant growth, development, and productivity. Recently, coronatine (COR) has emerged as a mimic of jasmonates, which play diverse roles in plant tolerance to multiple stressors. Although COR affects the response to drought stress in Carex leucochlora seedlings, the mechanism by which it acts is unclear. We characterized the acclimation of C. leucochlora to drought stress using plant growth, osmotic adjustment, photosynthetic parameters, antioxidant enzyme system and comparative transcriptomics analyses during stress treatments, COR (0.1 nmol L−1) was applied under soil water stress (20 ± 5%) in the growth chamber. Drought stress strongly suppressed biomass accumulation and loss of cellular water content, inhibited photosynthesis, and damaged cellular membranes in the seedlings. Conversely, COR treatment improved the detrimental effects of drought stress by restoring leaf morphology, alleviating damage to the cell membrane, and improving photosynthetic efficiency and water use efficiency. In addition, COR enhanced the activities of superoxide dismutase, peroxidase activity, catalase, and ascorbate peroxidase. There were 1205 transcripts (36.7%) that were common with COR application uniquely induced by drought stress. COR regulated transcription factors from 48 families such as AP2/ERF-ERFs, bHLHs, bZIPs, and C2H2s, and the genes were mainly involved in starch and sucrose metabolism, phenylpropanoid biosynthesis, and plant hormone signal transduction. Taken together, these results provide the physiological mechanism underlying improved drought resistance in C. leucochlora in the presence of COR and provide a new perspective for gene function analysis in COR-mediated signaling pathways.
