The hrp cluster of Xanthomonas fuscans subsp. fuscans is characterized and the role of hrp genes in the survival and the multiplication of this bacterium in the bean phyllosphere and in the transmission to bean seeds are determined. The study has revealed the involvement of the Xanthomonas fuscans subsp. fuscans T3SS in phyllospheric multiplication and systemic colonization of bean and have suggested a major contribution of hrp regulatory genes in host colonization processes.

Background: Two young men working at a nylon flocking plant in Rhode Island developed interstitial lung disease of unknown cause. Similar clusters at the same company's Canadian plant were reported previously.Objective: To define the extent, clinicopathologic features, and potential causes of the apparent disease outbreak.Design: Case-finding survey and retrospective cohort study.Setting: Academic occupational medicine program.Patients: All workers employed at the Rhode Island plant on or after 15 June 1990.Measurements: Symptomatic employees had chest radiography, pulmonary function tests, high-resolution computed tomography, and serologic testing. Those with unexplained radiographic or pulmonary function abnormalities underwent bronchoalveolar lavage, lung biopsy, or both. The case definition of "flock worker's lung" required histologic evidence of interstitial lung disease (or lavage evidence of lung inflammation) not explained by another condition.Results: Eight cases of flock worker's lung were identified at the Rhode Island plant. Three cases were characterized by a high proportion of eosinophils (25% to 40%) in lavage fluid. Six of the seven patients who had biopsy had histologic findings of nonspecific interstitial pneumonia, and the seventh had bronchiolitis obliterans organizing pneumonia. All seven of these patients had peribronchovascular interstitial lymphoid nodules, usually with germinal centers, and most had lymphocytic bronchiolitis and interstitial fibrosis. All improved after leaving work. Review of the Canadian tissue specimens showed many similar histologic findings. Among the 165-member study cohort, a 48-fold or greater increase was seen in the sex-adjusted incidence rate of all interstitial lung disease.Conclusions: Work in the nylon flocking industry poses substantial risk for a previously unrecognized occupational interstitial lung disease. Abstract Background Bacterial plant pathogens belonging to the Xanthomonas genus are tightly adapted to their host plants and are not known to colonise other environments. The host range of each strain is usually restricted to a few host plant species. Bacterial strains responsible for the same type of symptoms on the same host range cluster in a pathovar. The phyllosphere is a highly stressful environment, but it provides a selective habitat and a source of substrates for these bacteria. Xanthomonads colonise host phylloplane before entering leaf tissues and engaging in an invasive pathogenic phase. Hence, these bacteria are likely to have evolved strategies to adapt to life in this environment. We hypothesised that determinants responsible for bacterial host adaptation are expressed starting from the establishment of chemotactic attraction and adhesion on host tissue. Results We established the distribution of 70 genes coding sensors and adhesins in a large collection of xanthomonad strains. These 173 strains belong to different pathovars of Xanthomonas spp and display different host ranges. Candidate genes are involved in chemotactic attraction (25 genes), chemical environment sensing (35 genes), and adhesion (10 genes). Our study revealed that candidate gene repertoires comprised core and variable gene suites that likely have distinct roles in host adaptation. Most pathovars were characterized by unique repertoires of candidate genes, highlighting a correspondence between pathovar clustering and repertoires of sensors and adhesins. To further challenge our hypothesis, we tested for molecular signatures of selection on candidate genes extracted from sequenced genomes of strains belonging to different pathovars. We found strong evidence of adaptive divergence acting on most candidate genes. Conclusions These data provide insight into the potential role played by sensors and adhesins in the adaptation of xanthomonads to their host plants. The correspondence between repertoires of sensor and adhesin genes and pathovars and the rapid evolution of sensors and adhesins shows that, for plant pathogenic xanthomonads, events leading to host specificity may occur as early as chemotactic attraction by host and adhesion to tissues.

The time that elapsed between the initial introduction and the proliferation of an invasive species is referred to as the lag phase. The identification of the lag phase is critical for generating plans for pest management and for the prevention of biosecurity failure. However, lag phases have been identified mostly through retrospective searches of historical records. The agricultural pest fall armyworm (FAW; Spodoptera frugiperda) is native to the New World. FAW invasion was first reported from West Africa in 2016, then it spread quickly through Africa, Asia, and Oceania. Here, using population genomics approaches, we demonstrate that the FAW invasion involved an undocumented lag phase. Invasive FAW populations have negative signs of genomic Tajima's D, and invasive population-specific genetic variations have particularly decreased Tajima's D, supporting a substantial amount of time for the generation of new mutations in introduced FAW populations. Model-based diffusion approximations support the existence of a period with a cessation of gene flow between native and invasive FAW populations. Taken together, these results provide strong support for the presence of a lag phase during the FAW invasion. During infancy, intersensory facilitation declines gradually as unisensory perception develops. However, this trade-off was mainly investigated using audiovisual stimulations. Here, fifty 4- to 12-month-old infants (26 females, predominately White) were tested in 2017-2020 to determine whether the facilitating effect of their mother's body odor on neural face categorization, as previously observed at 4 months, decreases with age. In a baseline odor context, the results revealed a face-selective electroencephalographic response that increases and changes qualitatively between 4 and 12 months, marking improved face categorization. At the same time, the benefit of adding maternal odor fades with age (R 2  = .31), indicating an inverse relation with the amplitude of the visual response, and generalizing to olfactory-visual interactions previous evidence from the audiovisual domain., (© 2024 The Author(s). Background: Donors possess heterogeneous red cell concentrates (RCCs) in terms of the biological age of their red blood cells (RBCs) as a direct result of various donor-dependent factors influencing rates of erythropoiesis. This study aimed to estimate the median biological age of RBCs in RCCs based on donor age and sex to investigate inherent differences in blood products' biological ages over hypothermic storage using estimated median densities (EMDs)., Study Design: Sixty RCCs were collected from four donor groups; male and female teenagers (17-19 years old) and seniors (75+ years old). A Percoll density-based separation approach was used to quantify the EMDs indicative of biological age. EMD and mean corpuscular hemoglobin (MCHC) were compared by correlation analyses., Results: Differences in the median biological age of RCC units were observed with male donors having significantly higher EMDs compared to females (p < .001). Teen male donors possessed the highest EMDs with significantly elevated levels of biologically aged RBCs compared to both female donor groups, regardless of storage duration (p < .05). Throughout most of the 42-day storage period, senior donors, particularly senior females, demonstrated the strongest correlation between EMD and MCHC (R 2  > 0.5)., Conclusions: This study provides further evidence that there are inherent differences between the biological age profiles of RBCs between blood donors of different sex and age. Human milk odor is attractive and appetitive for human newborns. Here, we studied behavioral and heart-rate (HR) responses of 2-day-old neonates to the odor of human colostrum. To evaluate detection in two conditions of stimulus delivery, we first presented the odor of total colostrum against water. Second, the hedonic specificity of total colostrum odor was tested against vanilla odor. Third, we delivered only the fresh effluvium of colostrum separated from the colostrum matrix; the stability of this colostrum effluvium was then tested after deep congelation; finally, after sorptive extraction of fresh colostrum headspace, we assessed the activity of colostrum volatiles eluting from the gas chromatograph (GC). Regardless of the stimulus-delivery method, neonates displayed attraction reactions (HR decrease) as well as appetitive oral responses to the odor of total colostrum but not to vanilla odor. The effluvium separated from the fresh colostrum matrix remained appetitive but appeared labile under deep freezing. Finally, volatiles from fresh colostrum effluvium remained behaviorally active after GC elution, although at lower magnitude. In sum, fresh colostrum effluvium and its eluate elicited a consistent increase in newborns' oral activity (relative to water or vanilla), and they induced shallow HR decrease. Newborns' appetitive oral behavior was the most reproducible response criterion to the effluvium of colostrum. In conclusion, a set of unidentified volatile compounds from human colostrum is robust enough after extraction from the original matrix and chromatographic processing to continue eliciting appetitive responses in neonates, thus opening new directions to isolate and assay specific volatile molecules of colostrum., (© 2024 Wiley Periodicals LLC.)

The fall armyworm (Spodoptera frugiperda) is one of the major pest insects in diverse crop plants, including maize, rice, and cotton. While the fall armyworm is native to North and South America, its invasion was first reported in West Africa in 2016. Since then, this species has rapidly spread across Sub-Saharan Africa, Asia, and Oceania, as well as Egypt and Cyprus. The fall armyworm is composed of two sympatric strains, the corn and rice strains, designated to their preferred host plants, in native areas. It remains surprisingly unclear whether invasive fall armyworms belong to the corn strain, rice strain, or hybrids of the two, despite a large number of population genetics studies. In this study, we performed population genomics analyses using globally collected 116 samples to identify the strains of invasive fall armyworms. We observed that invasive fall armyworms are genomically most similar to the corn strain. The reconstructed phylogenetic tree supports the hypothesis that invasive fall armyworms originated from the corn strain. All genomic loci of invasive populations exhibit higher genetic similarity to the corn strains compared to the rice strains. Furthermore, we found no evidence of gene flow from rice strains to invasive populations at any genomic locus. These results demonstrate that invasive fall armyworms belong to the corn strain. These results suggest that invasive fall armyworms likely have very limited potential to infest rice. The development of new processing methods is required in order to meet the continuous demand for thinner films with excellent barrier properties for food packaging and other applications. In this study, rice husk organosolv lignin nanoparticles were prepared using the electrospray method, which were applied to produce polylactic acid (PLA)-based films for food packaging. The effect of the following electrospray parameters has been investigated: lignin concentration (LC) ranging from 5-50 mg/mL, flow rate (FR) from 0.5-1 mL/min, applied voltage from 10-30 kV, and tip-to-collector distance (TCD) from 10-25 cm, on the morphology, size, polydispersity index (PDI), and Zeta potential (ZP) of lignin nanoparticles (LNPs). The response surface methodology with a Box-Behnken design was applied to optimize these parameters, while dynamic light scattering (DLS) and scanning electron microscopy (SEM) analyses were used to characterize the controlled LNPs. The results showed that the LNPs shape and sizes represent a balance between the solvent evaporation, LC, applied voltage, TCD and FR. The application of optimal electrospray conditions resulted in the production of LNPs with a spherical shape and a minimal size of 260 ± 10 nm, a PDI of 0.257 ± 0.02, and a ZP of -35.2 ± 4.1 mV. The optimal conditions were achieved at LC = 49.1 mg/mL and FR = 0.5 mL/h under an applied voltage of 25.4 kV and TCD = 22 cm. Then, the optimized LNPs were used to improve the properties of PLA-based films. Three types of PLA-lignin blend films were casted, namely lignin/PLA, LNPs/PLA and PLA-grafted LNPs. PLA-grafted LNPs exhibited a more uniform dispersion in PLA for lignin contents of up to 10% than other composite samples. Increasing the lignin content from 5% to 10% in PLA-grafted LNPs resulted in a significant increase in elongation at break (up to four times higher than neat PLA). The presence of PLA-grafted lignin led to a substantial reduction in optical transmittance in the UV range, dropping from 58.7 ± 3.0% to 1.10 ± 0.01%, while maintaining excellent transparency to visible light compared to blends containing lignin or LNPs. Although the antioxidant capacity of unmodified lignin is well-known, a substantial increase in antioxidant capacity was observed in LNPs and PLA-grafted LNP films, with values exceeding 10 times and 12 times that of neat PLA, respectively. These results confirm the significant potential of using studied films in food packaging applications.

Objective: To understand which attributes men with benign prostatic hyperplasia (BPH) undergoing water vapor thermal therapy (WVTT) find important while considering treatment options for the condition. Methods: Men ( n  = 170) with lower urinary tract symptoms due to BPH who underwent WVTT between April 2019 and November 2020 in a Toronto urologic clinic were invited to participate in an online survey. The survey included eight attributes of BPH surgical procedures and five attributes of WVTT. Patients were asked how important each attribute was to them before they selected a BPH procedure and decided to undergo WVTT. Results: In total, 128 respondents (75%) completed the survey. A majority of the respondents were White (88%), married (83%), and aged 60-69 years old (45%). Approximately 97% of respondents rated the ability to avoid further BPH treatments as "very important" or "extremely important," followed by duration to return to normal activities (79%), and wait times to receive the procedure (57%). Only 47% of patients reported that postprocedural catheterization was important. For WVTT, 98% of the respondents rated avoiding more invasive surgical treatments and 88% rated a quick recovery as important attributes. Conclusions: Among men with moderate-to-severe BPH undergoing WVTT, the most important attributes for selecting a BPH surgical procedure were avoiding further BPH treatments, returning quickly to normal activities, and reducing treatment wait times. Most men chose WVTT to avoid more invasive procedures and have a quick recovery.

Chordomas are rare tumors of the axial skeleton that are refractory to conventional therapy. Few studies have compared the morphological and molecular characteristics of chordomas according to the skull base and sacral locations. Histopathological data and changes revealed by array comparative genomic hybridization (CGH) and next-generation sequencing (NGS) of cell cycle regulation genes were analyzed for 28 skull base (SBCs) and 15 sacral (SC) chordomas. All cases were conventional chordomas. SBCs were significantly more frequent in patients aged <40 years and SCs predominated in patients aged >60 years. Mitotic indices ≥2 mitoses/10 high-power fields were correlated with high degrees of nuclear atypia and Ki67 labeling indices ≥6%. We identified 321 genomic positions, and copy number variation losses were more frequent than gain. Moreover, we report a panel of 85 genetic variants of cell cycle genes and the presence of molecular clusters for chordoma as well in CGH as in NGS. Despite the species' wide distribution, studies of the genetic diversity within Entamoeba coli and Entamoeba hartmanni remain limited. In the present study, we provide further insight into the genetic diversity of both species based on analysis of partial nuclear small subunit ribosomal DNA sequences generated from human fecal DNAs from samples collected in Africa, South America, and Europe. Reinforcing the previous recognition that E. coli is a species complex, our data confirm the existence of the two subtypes, ST1 and ST2, previously identified plus, potentially, a new subtype, ST3. While ST1 appears to be genetically quite homogenous, ST2 shows a substantial degree of intrasubtype diversity. ST2 was more common in samples collected outside Europe, whereas ST1 showed no geographical restriction. The potentially novel subtype is represented to date exclusively by sequences from South American and African samples. In contrast to previous reports, our new data also indicate substantial variation in E. hartmanni that could also support the establishment of subtypes within this species.
