Abstract

This review is the result of a survey across the bibliography about the determination of several antibiotics in biological, food and environmental samples by liquid chromatography. Their determination in those samples is usually made in clinical monitoring, food safety and environmental studies. A brief description of the pharmacological activity is provided, to complement the relevance of the topic bioanalytical method. Exposure to antimicrobial agents, which can be evaluated by the analysis of biological fluids, comes mainly from their extensive use as veterinary agents in industrial animal farming, via food chain and residual water poured to the environment. Common columns and mobile phases are C18 ones and mixtures of aqueous buffers, acetonitrile and methanol, respectively. Retention times of these analyses are 20–30 min with column temperatures ranging from 15 to 45 ◦C and injection volumes of 10–20 µL. Detection is mainly performed by UV–Visible absorbance and MS/MS detection in positive mode, excluding glycopeptides where fluorescence was used. These procedures exhibit adequate analytical performances: good selectivity, precision, linearity range, ruggedness and sensitivity, though variable trueness. The future trend in the management and control of antibiotics in the here studied samples should be the development of automatized on-line detection protocols (sample processing coupled to chromatographic separation) for currently studied antibiotics and new methods for unknown metabolites and the transformation products.

21. Micellar Liquid Chromatography for the Determination of Some Less Prescribed Benzodiazepines

Abstract

A simple chromatographic procedure is reported for the determination of some less prescribed but equally important benzodiazepines (Clotiazepam, clozapine and pinazepam) in serum. The optimization studies have been made in CN, C18 and C8 columns, using mobile phase containing sodium dodecyl sulphate (SDS) modified with either propanol, butanol or pentanol. The method proposed for the determination of the three benzodiazepines using a mobile phase of 0.13 M SDS, 2.4% pentanol-0.01 M phosphate buffer- 0.1% triethylamine (pH 7) at 25°C and UV detection (240 nm) in a C8 column. The serum samples was injected directly, without any pretreatment, eluted in less than 8 min, in accordance to their relative polarities, as indicated by their octanol-water partition coefficients. The limits of detection (ng/mL) was in the 1.6 to 5.6 and 7 to 87 range, for aqueous and serum samples, respectively. Repeatability and intermediate precision was tested for three different concentrations of the drugs, resulting in the 0.1 to 2 range. The results obtained here for the separation of the three benzodiazepines in serum were also counter checked at Department of Bio-analytical Chemistry, Universitat Jaume I, Castelló, Spain.

Abstract

A reliable and sensitive high performance liquid chromatographic method for the determination of widely prescribed antidepressant has been developed. Paroxetine is a potent selective serotonin reuptake inhibitor used for the treatment of depression and various mood disorders. The optimum mobile phase was prepared using a combination of 40% acetonitrile and phosphate buffer 0.01 M to pH 3 and running under isocratic mode at a flow rate of 1.0 ml/min. with electrochemical detection at 0.9V. The applicability of the developed method is in the field of quality control as well as for monitoring the level of drug at various concentrations during synthesis of the parent drug. The suggested methodology was validated following the guidelines of the FDA in terms of: sensitivity (LOD and LOQ, 0.005 and 0.01 ng/mL, respectively), linearity between 0.5 - 50 ng/mL (r2 > 0.9999), inter- and intraday precision (< 0.259% and < 0.538%), robustness (less than 5.0%) and recovery (99.7 - 100.7%). The developed method is specific, rapid (less than 10 min), precise, reliable, accurate, cheap and suitable for routine analysis for the determination of paroxetine in pharmaceutical preparations.

Abstract

A micellar liquid chromatographic procedure was developed for the simultaneous determination of threecommonly used stupefacients, lidocaine, ketamine and diazepam, using a C18 reversed-phase column. A micellar mobile phase 0.15 M sodium dodecyl sulfate and 6% (v/v) pentanol, pH 7, and UV detection at 230 nm were used to determine the three stupefacients in food samples. Using the selected mobile phase, the stupefacients were eluted in less than 10 min with linearity (r = 0.998), LOD (range: 0.004–0.03 ppm), LOQ (range: 0.004–0.03 ppm), intraday and interday precision (below 2.84%), and mean recoveries (range: 79.11–110.16%) in the different foodstuffs were in accordance with the internationally established acceptance criteria. Validation of the developed method was performed on the basis of International Conference on Harmonization validation guidelines. The optimized and validated micellar liquid chromatographic method was successfully applied in the determination of lidocaine, diazepam, and ketaminein a real food sample (mango drink) and in spiked food samples (banana, ladoo, soft drink, tea). The developed method could also be easily used by law enforcement laboratories and hospitals for routine analysis.
