Nitric oxide (NO) is a multifunctional gaseous signal that modulates the growth, development and stress tolerance of higher plants. NO donors have been used to boost plant endogenous NO levels and to activate NO-related responses, but this strategy is often hindered by the relative instability of donors. Alternatively, nanoscience offers a new, promising way to enhance NO delivery to plants, as NO-releasing nanomaterials (e.g. S-nitrosothiol-containing chitosan nanoparticles) have many beneficial physicochemical and biochemical properties compared to non-encapsulated NO donors. Nano NO donors are effective in increasing tissue NO levels and enhancing NO effects both in animal and human systems. The authors believe, and would like to emphasize, that new trends and technologies are essential for advancing plant NO research and nanotechnology may represent a breakthrough in traditional agriculture and environmental science. Herein, we aim to draw the attention of the scientific community to the potential of NO-releasing nanomaterials in both basic and applied plant research as alternatives to conventional NO donors, providing a brief overview of the current knowledge and identifying future research directions. We also express our opinion about the challenges for the application of nano NO donors, such as the environmental footprint and stakeholder's acceptance of these materials.

Nitric oxide (NO) is perfectly suited for the role of a redox signalling molecule. A key route for NO bioactivity occurs via protein S-nitrosation, and involves the addition of a NO moiety to a protein cysteine (Cys) thiol (–SH) to form an S-nitrosothiol (SNO). This process is thought to underpin a myriad of cellular processes in plants that are linked to development, environmental responses and immune function. Here we collate emerging evidence showing that NO bioactivity regulates a growing number of diverse post-translational modifications including SUMOylation, phosphorylation, persulfidation and acetylation. We provide examples of how NO orchestrates these processes to mediate plant adaptation to a variety of cellular cues.

Nitric oxide (NO) emerged as a key signal molecule in plants. During the last two decades impressive progress has been made in plant NO research. This small, redox-active molecule is now known to play an important role in plant immunity, stress responses, environmental interactions, plant growth and development. To more accurately and robustly establish the full spectrum of NO bioactivity in plants, it will be essential to apply methodological best practice. In addition, there are some instances of conflicting nomenclature within the field, which would benefit from standardization. In this context, we attempt to provide some helpful guidance for best practice associated with NO research and also suggestions for the cognate terminology.

To study the NO-dependent gene induction, NO-burst was simulated in cell culture as well as in plants (Arabidopsis thaliana) and the induced transcription was analysed by microarray analyses. About 10 percent of 330 tested stress genes were regulated by NO-treatment. Further biochemical and physiological studies were based on the microarray data. The induction of alternative oxidase (AOX1a) was shown to be an important factor against oxidative damage caused by NO treatment. Another interesting result was that NO correlated with the biosynthesis and signalling of the plant hormone jasmonic acid (JA). As in mammal, NO shows its influence on different cellular processes in plants., Auch in Pflanzen spielt das für Abwehrreaktionen in Säugern wichtige Stickoxid (NO) eine zentrale Rolle. Um die NO-abhängige Geninduktion zu untersuchen, wurde ein NO-Burst in Zell-Kultur sowie in Pflanzen (Arabidopsis thaliana) simuliert, und die induzierten Transkripte mittels eines Mikroarrays analysiert. Ungefähr 10% von 330 getesteten Stress-Genen wurden durch NO-Behandlung reguliert. Die Mikroarray-Experimente waren die Basis für weitere biochemische und physiologische Studien. Es konnte gezeigt werden, dass die Induktion von Alternativer Oxidase (AOX1a) ein wichtiger Faktor gegen oxidative Schädigung durch NO ist. Ein weiteres, überraschendes Ergebnis war, dass NO in die Biosynthese des Pflanzenhormons Jasmonsäure und dessen Signalwege eingreift. NO scheint, wie auch in Säugern, verschiedenste zelluläre Prozesse in Pflanzen zu beeinflussen.

Cysteine-rich receptor-like kinases (CRKs) are transmembrane proteins characterized by the presence of two domains of unknown function 26 (DUF26) in their ectodomain. The CRKs form one of the largest groups of receptor-like protein kinases in plants, but their biological functions have so far remained largely uncharacterized. We conducted a large-scale phenotyping approach of a nearly complete crk T-DNA insertion line collection showing that CRKs control important aspects of plant development and stress adaptation in response to biotic and abiotic stimuli in a non-redundant fashion. In particular, the analysis of reactive oxygen species (ROS)-related stress responses, such as regulation of the stomatal aperture, suggests that CRKs participate in ROS/redox signalling and sensing. CRKs play general and fine-tuning roles in the regulation of stomatal closure induced by microbial and abiotic cues. Despite their great number and high similarity, large-scale phenotyping identified specific functions in diverse processes for many CRKs and indicated that CRK2 and CRK5 play predominant roles in growth regulation and stress adaptation, respectively. As a whole, the CRKs contribute to specificity in ROS signalling. Individual CRKs control distinct responses in an antagonistic fashion suggesting future potential for using CRKs in genetic approaches to improve plant performance and stress tolerance.

Toll-like Rezeptoren (TLRs, TLR1-TLR10) erkennen ein breites Spektrum von bakteriellen und viralen konservierten Strukturen (PAMPs, Pathogen assoziierten molekularen Mustern), unter anderem modifizierte Lipide (LPS und bakterielle Lipoproteine), Proteine (Flagellin) und Nukleinsäuren (DNA, ss und ds RNA). Im Rahmen dieser Arbeit sollten zur Charakterisierung von TLR9 monoklonale Antikörper gegen diesen Rezeptor generiert und geklärt werden, ob eine direkte Interaktion zwischen TLR9 und seinem Liganden CpG-DNA besteht. In TLR9 exprimierenden HEK 293 Zellen konnte mit Hilfe dieser Antikörper TLR9 in verschiedenen Testsystemen detektiert werden. TLR9 konnte im Gegensatz zu anderen Mitgliedern der TLR-Familie nicht auf der Zelloberfläche, sondern innerhalb der Zelle lokalisiert werden. Mit Hilfe der Surface Plasmon Resonance Technologie konnte darüberhinaus gezeigt werden, dass TLR9 direkt und sequenzspezifisch seinen Liganden einzelsträngige, unmethylierte CpG-DNA binden kann. Anhand des Transkriptionsmusters von zahlreichen Stressgenen wurde die Wirkungsweise von Cäsium und ionisierender Strahlung auf den Stoffwechsel von Arabidopsis thaliana charakterisiert Die Kombination der Subtraktiven-Suppressions-Hybridisierung und der Makroarray-Technologie erwies sich als eine sehr sensitive Methode, um eine Vielzahl von differentiell exprimierten Genen mit hoher Reproduzierbarkeit und hohem Durchsatz zu analysieren; mit der quantitativen und semiquantitativen PCR konnten schwach induzierte oder reprimierte Gene effektiv verifiziert werden. Cäsium- und Kalium-Ionen besitzen große chemische Gemeinsamkeiten. Während Kalium jedoch für die Pflanze essentiell ist, besitzt Cäsium keine bekannten physiologischen Funktionen. Folgende Reaktionen der Pflanze auf erhöhte Cäsium-133 Konzentrationen (150µM) in der Umgebung konnten auf Transkriptebene nachgewiesen werden: Eine verstärkte Transkription von Redoxenzymen (Superoxid-Dismutase, Thioredoxin, Peroxidase) und die Induktion der Schwefelaufnahme bzw. die Synthese schwefelhaltiger Verbindungen. Eine Induktion und Repression von Enzymen auf Genebene, die sowohl bei der Stabilisierung (Chaperone, Heat-shock Proteine) als auch beim Abbau von Proteinen (Peptidasen, Protease-Inhibitoren) eine Rolle spielen. Eine erhöhte Transkriptionsrate von Genen, deren Produkte als Schutzstoffe für Makromoleküle oder als osmotisch wirksame Substanzen fungieren können (Prolin, Osmotin, LEA-Proteine, etc.); parallel wurde die Expressionsrate verschiedener Aquaporin-Gene verringert. Radiocäsium (Cäsium-134) wurde nur in sehr geringen Konzentrationen und Aktivitäten (30Bq/cm3) in den Versuchen eingesetzt, da vor allem die Langzeitwirkung schwacher ionisierender Strahlung auf Genebene untersucht werden sollte.
