The restoration of cultural heritage sites is a complex and challenging process, particularly when the structure holds significant historical and cultural value. This paper refers to the first stages of the restoration study of Pashas Bridge, one of the largest stone bridges in Greece that was destroyed during World War II and further damaged by an earthquake in 1995. The project was assigned to multiple research groups from the Aristotle University of Thessaloniki (AUTh) for a period of 18 months by the Greek Ministry of Culture and Sports. To restore the bridge effectively, it is essential to accurately record, document, and assess its current condition. Through the use of photogrammetry, which creates 3D models of objects or structures using images, and unmanned aerial vehicles (UAVs) which assisted in capturing multiple shots with various angles of the bridge, a detailed 3D model of the bridge’s current condition was generated. In addition to these technical approaches, historical research and documentation were utilized to understand the bridge's cultural heritage value. The process included an examination of historical photographs and records related to the bridge, with a constant effort to discover additional information about its history and importance. Ultimately, the restoration process of Pashas Bridge serves as a valuable case study for the effective restoration of cultural heritage sites. Through utilizing these resources, the team aims to restore the bridge to its former glory, preserving its cultural heritage value and ensuring its place as a valuable and integral part of the community for generations to come.

Preventive actions of cultural heritage could be carried out using photogrammetry and computer vision techniques. Structure from Motion (SfM) techniques have been proved to be one of the best solutions for the cultural heritage field in terms of cost, time, accuracy and quality. The current work uses a commercial SfM – based software to generate orthoimages of the Arch of Galerius in Thessaloniki, Greece. This is part of students' project in the framework of an undergraduate course in the School of Spatial Planning and Development, Thessaloniki, Greece, to investigate the status of the Roman monument.

Preventive actions of cultural heritage continuously emerge in order to preserve the identity of the respective civilizations, retain its cultural significance and ensure its accessibility to present and future generations. 3D geomatics technologies along with UAV systems are widely used for documenting existing structures especially in difficult-to-access areas. In addition, Building Information Modelling (BIM) for cultural heritage gains ground towards the sustainable management, update and maintenance of the information. To this context, the current work generates a Historic Building Information Modelling (HBIM) model of the “Averof’s Museum of Neohellenic art” located in Metsovo, Greece, by using UAV photogrammetry techniques and additional information derived from the architecture designs of the buildings.

This paper presents a Cloud-based open source system for storing and processing data from a 3D survey approach. More specifically, we provide an online service for viewing, storing and analysing BIM. Cloud technologies were used to develop a web interface as a BIM data centre, which can handle large BIM data using a server. The server can be accessed by many users through various electronic devices anytime and anywhere so they can view online 3D models using browsers. Nowadays, the Cloud computing is engaged progressively in facilitating BIM-based collaboration between the multiple stakeholders and disciplinary groups for complicated Architectural, Engineering and Construction (AEC) projects. Besides, the development of Open Source Software (OSS) has been rapidly growing and their use tends to be united. Although BIM and Cloud technologies are extensively known and used, there is a lack of integrated open source Cloud-based platforms able to support all stages of BIM processes. The present research aims to create an open source Cloud-based BIM system that is able to handle geospatial data. In this effort, only open source tools will be used; from the starting point of creating the 3D model with FreeCAD to its online presentation through BIMserver. Python plug-ins will be developed to link the two software which will be distributed and freely available to a large community of professional for their use. The research work will be completed by benchmarking four Cloud-based BIM systems: Autodesk BIM 360, BIMserver, Graphisoft BIMcloud and Onuma System, which present remarkable results.

This paper investigates how the historical and cultural heritage of cities is and can be underpinned by means of smart city tools, solutions and applications. Smart cities stand for a conceptual technology-and-innovation driven urban development model. By becoming ‘smart’, cities seek to achieve prosperity, effectiveness and competitiveness on multiple socio-economic levels. Although cultural heritage is one of the many issues addressed by existing smart city strategies, and despite the documented bilateral benefits, our research about the positioning of urban cultural heritage within three smart city strategies (Barcelona, Amsterdam, and London) reveals fragmented approaches. Our findings suggest that the objective of cultural heritage promotion is not substantially addressed in the investigated smart city strategies. Nevertheless, we observe that cultural heritage management can be incorporated in several different strategic areas of the smart city, reflecting different lines of thinking and serving an array of goals, depending on the case. We conclude that although potential applications and approaches abound, cultural heritage currently stands for a mostly unexploited asset, presenting multiple integration opportunities within smart city contexts. We prompt for further research into bridging the two disciplines and exploiting a variety of use cases with the purpose of enriching the current knowledge base at the intersection of cultural heritage and smart cities.

Characteristic example of the Balkan architecture of the 19th century, consists the "Tower house" which is found in the region of Epirus and Western Macedonia, Greece. Nowadays, the only information about these heritage buildings could be abstracted by the architectural designs on hand and the model – Tower that is being displayed in the Folklore Museum of the Municipality of Kozani, Greece, as a maquette. The current work generates a scaled 3D digital model of the "Tower house", by using photogrammetry techniques applied on the model-maquette that is being displayed in the Museum exhibits.

Recent research on the field of Building Information Modelling (BIM) technology, revealed that except of a few, accessible and free BIM viewers there is a lack of Free & Open Source Software (FOSS) BIM software for the complete BIM process. With this in mind and considering BIM as the technological advancement of Computer-Aided Design (CAD) systems, the current work proposes the use of a FOSS CAD software in order to extend its capabilities and transform it gradually into a FOSS BIM platform. Towards this undertaking, a first approach on developing a spatial Database Management System (DBMS) able to store, organize and manage the overall amount of information within a single application, is presented.

The paper illustrates the use of open source Computer-aided design (CAD) environments in order to develop Building Information Modelling (BIM) tools able to manage 3D models in the field of cultural heritage. Nowadays, the development of Free and Open Source Software (FOSS) has been rapidly growing and their use tends to be consolidated. Although BIM technology is widely known and used, there is a lack of integrated open source platforms able to support all stages of Historic Building Information Modelling (HBIM) processes. The present research aims to use a FOSS CAD environment in order to develop BIM plug-ins which will be able to import and edit digital representations of cultural heritage models derived by photogrammetric methods.

Three new triterpene glycosides were isolated from Deutzia x hybrida "Strawberry Fields" cultivar via aqueous-ethanolic extraction of the roots, including one derivative of sumaresinolic acid and two of echinocystic acid: 3- O -β-D-glucuronopyranosylsumaresinolic acid 28- O -β-D-xylopyranosyl-(1→4)-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl ester, 3- O -β-D-glucuronopyranosylechinocystic acid 28- O -β-D-xylopyranosyl-(1→4)-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl ester, and 3- O -α-L-arabinopyranosyl-(1→3)-β-D-glucuronopyranosylechinocystic acid 28-O-β-D-xylopyranosyl-(1→4)-α-L-rhamnopyranosyl-(1→2)-α-L-arabinopyranosyl ester. As none of the isolated saponins were previously documented in the literature, their structural elucidation required extensive 1D and homo- and heteronuclear 2D NMR spectroscopy, as well as mass spectrometry analysis. All three glycosides were tested for their stimulatory activity of the sweet taste receptor TAS1R2/TAS1R3. It is the first chemical and biological investigation of Deutzia x hybrida "Strawberry Fields" as well as the first report of sumaresinolic acid glycosides in Deutzia genus.

Telepsychiatry is an effective tool to support and provide mental health services to prison inmates. In Greece, telepsychiatry was formally applied in two correctional facilities in 2018. The objective of this study was to compare inmates' perceptions and satisfaction with telepsychiatry versus face-to-face consultation. The sample consisted of 100 male inmates with a multicultural background and prior experience with both methods of services provision. The data were obtained through a Demographic Data Questionnaire, a Participant Satisfaction Questionnaire to assess satisfaction with face-to-face psychiatric services, and another Participant Satisfaction Questionnaire to assess satisfaction with telepsychiatric services. The results have shown a higher level of satisfaction with telepsychiatry compared to face-to-face care. Statistically significant improvements (all p-values <0.001) were noted in: waiting times, support for relapse prevention, follow up, quality of mental health care, quality of care in the management of psychiatric problems and related medication, behaviour of psychiatrists, duration of the assessment, sense of comfort, and confidentiality. Telepsychiatry has proved to be an acceptable way of approaching and supporting inmates in Greece, with most of the participants expressing high acceptance, satisfaction, and preference rates. Implications for additional research and further development of telepsychiatry are discussed.

Background: Neuronal nicotinic acetylcholine receptors (nAChRs) are abundant in the central nervous system (CNS), playing critical roles in brain function. Antigenicity of nAChRs has been well demonstrated with antibodies to ganglionic AChR subtypes (i.e., subunit α3 of α3β4-nAChR) and muscle AChR autoantibodies, thus making nAChRs candidate autoantigens in autoimmune CNS disorders. Antibodies to several membrane receptors, like NMDAR, have been identified in autoimmune encephalitis syndromes (AES), but many AES patients have yet to be unidentified for autoantibodies. This study aimed to develop of a cell-based assay (CBA) that selectively detects potentially pathogenic antibodies to subunits of the major nAChR subtypes (α4β2- and α7-nAChRs) and its use for the identification of such antibodies in "orphan" AES cases., Methods: The study involved screening of sera derived from 1752 patients from Greece, Turkey and Italy, who requested testing for AES-associated antibodies, and from 1203 "control" patients with other neuropsychiatric diseases, from the same countries or from Germany. A sensitive live-CBA with α4β2-or α7-nAChR-transfected cells was developed to detect antibodies against extracellular domains of nAChR major subunits. Flow cytometry (FACS) was performed to confirm the CBA findings and indirect immunohistochemistry (IHC) to investigate serum autoantibodies' binding to rat brain tissue., Results: Three patients were found to be positive for serum antibodies against nAChR α4 subunit by CBA and the presence of the specific antibodies was quantitatively confirmed by FACS. We detected specific binding of patient-derived serum anti-nAChR α4 subunit antibodies to rat cerebellum and hippocampus tissue. No serum antibodies bound to the α7-nAChR-transfected or control-transfected cells, and no control serum antibodies bound to the transfected cells. All patients positive for serum anti-nAChRs α4 subunit antibodies were negative for other AES-associated antibodies. All three of the anti-nAChR α4 subunit serum antibody-positive patients fall into the AES spectrum, with one having Rasmussen encephalitis, another autoimmune meningoencephalomyelitis and another being diagnosed with possible autoimmune encephalitis., Conclusion: This study lends credence to the hypothesis that the major nAChR subunits are autoimmune targets in some cases of AES and establishes a sensitive live-CBA for the identification of such patients., Competing Interests: Authors MP, KK, EK, AT, MD, EN and ST were employed by company Tzartos NeuroDiagnostics. Background: Prisoners are often associated with mental health and substance use disorders. Coercive measures are widely used in prison settings. The objective of this study was to compare inmates' perceptions and satisfaction with telepsychiatry versus face-to-face consultation and the effects of telepsychiatry on the use of coercive measures. The sample consisted of 100 male inmates from various backgrounds who had experienced both approaches of services (face to face and telepsychiatry)., Method: The data were obtained through an interview where the individuals completed a Demographic Data Questionnaire, a Participant Satisfaction Questionnaire to assess satisfaction with face-to-face psychiatric services, and a Participant Satisfaction Questionnaire to assess their satisfaction with services offered via telepsychiatry. Additionally, calculations of time spent waiting for a face-to-face psychiatric evaluation and time spent in handcuffs and in confined spaces were made before and after the introduction of telepsychiatry., Results: Statistically significant improvements (all p -values < 0.001) were noted in waiting times, support for relapse prevention, follow up, quality of mental health care, quality of care in the management of psychiatric problems and related medication, behavior of psychiatrists, duration of the assessment, sense of comfort, and confidentiality. Telepsychiatry led to the elimination of time spent in handcuffs and in confined spaces (transport vehicles)., Conclusion: According to the results of this study, telepsychiatry is an acceptable method of service delivery in correctional facilities and was associated with a reduction of coercive practices.

Muscle-specific kinase (MuSK) Myasthenia Gravis (MG) represents a prototypical antibody-mediated disease characterized by predominantly focal muscle weakness (neck, facial, and bulbar muscles) and fatigability. The pathogenic antibodies mostly belong to the immunoglobulin subclass (Ig)G4, a feature which attributes them their specific properties and pathogenic profile. On the other hand, acetylcholine receptor (AChR) MG, the most prevalent form of MG, is characterized by immunoglobulin (Ig)G1 and IgG3 antibodies to the AChR. IgG4 class autoantibodies are impotent to fix complement and only weakly bind Fc-receptors expressed on immune cells and exert their pathogenicity via interfering with the interaction between their targets and binding partners (e.g. between MuSK and LRP4). Cardinal differences between AChR and MuSK-MG are the thymus involvement (not prominent in MuSK-MG), the distinct HLA alleles, and core immunopathological patterns of pathology in neuromuscular junction, structure, and function. In MuSK-MG, classical treatment options are usually less effective (e.g. IVIG) with the need for prolonged and high doses of steroids difficult to be tapered to control symptoms. Exceptional clinical response to plasmapheresis and rituximab has been particularly observed in these patients. Reduction of antibody titers follows the clinical efficacy of anti-CD20 therapies, a feature implying the role of short-lived plasma cells (SLPB) in autoantibody production. Novel therapeutic monoclonal against B cells at different stages of their maturation (like plasmablasts), or against molecules involved in B cell activation, represent promising therapeutic targets. A revolution in autoantibody-mediated diseases is pharmacological interference with the neonatal Fc receptor, leading to a rapid reduction of circulating IgGs (including autoantibodies), an approach already suitable for AChR-MG and promising for MuSK-MG. New precision medicine approaches involve Chimeric autoantibody receptor T (CAAR-T) cells that are engineered to target antigen-specific B cells in MuSK-MG and represent a milestone in the development of targeted immunotherapies. Mentally ill offenders constitute a vulnerable population group with unique characteristics, and have endured multiple public stigmatizations, which has not been sufficiently studied. The purpose of this study was to capture attitudes of the public toward mentally ill offenders in relation to their perceptions of mental illness in general, as well as their degree of familiarity with it. Our sample of 2059 people can be overall described as a men preponderance, married, with mean value age of 26 years, higher educational level, and medium or higher socio-economic status. Participants completed the ATMIO, CAMI, and Familiarity scales online. The total familiarity index value with mental illness was found to be 4.88, which counts as moderate to low. It was also concluded that women and those with a high educational level sustained more positive attitudes toward mentally ill offenders. However, negative stereotypes (with a mean value of 26.20), stigmatizing attitudes related to risk in the community (mean 16.10), and reduced responsibility for actions (mean 9.45) were recorded, while some (mean 16.50) showed compassion and emphasized on the mentally ill need of rehabilitation. The youngest people were the ones who recorded the most absolute and harsh attitudes. These findings validate the need of raising awareness and informing, especially, the young public about issues of mental health, including the need to oppose prejudices with everyday measures, which can be accessible to the new generation. Besides, we should extend research to various professional groups that come in contact with mentally ill offenders in order to collect data, which could contribute for intervening policies and formulating different sets of strategies for those people., (© 2023. Background and Objectives: There is accumulating evidence in the literature indicating a strong correlation between Fabry disease (FD) phenotypes and specific sequence variations in the Galactosidase Alpha ( GLA ) gene. Among them, the potential pathogenicity and clinical relevance of D313Y variation in patients with FD remain debated., Methods: We performed a systematic review and meta-analysis of studies reporting D313Y as single occurring variant in the GLA gene and sought to evaluate (1) the prevalence of D313Y variation in different populations with or without clinical manifestations of FD, (2) the clinical FD phenotype in D313Y -positive patients, and (3) the proportion of D313Y -positive patients presenting abnormal laboratory findings (alpha-galactosidase-A deficiency or globotriaosylceramide accumulation)., Results: Forty cohorts comprising 211 individuals with D313Y variation among 42,723 participants with available GLA gene-sequencing data were included. Patients highly suspected for FD had a higher prevalence of D313Y variation (4.9%, 95% CI 1.6%-9.9%; I 2 = 95.5%) compared with the general population (0%, 95% CI 0%-0.1%; I 2 = 1.9%; p = 0.004). The prevalence of D313Y variation was 0.6% (95% CI 0.3%-1%; I 2 = 74.1%), 0.4% (95% CI 0.2%-0.7%; I 2 = 0%), and 0.3% (95% CI 0.2%-0.4%; I 2 = 0%) in patients presenting with neurologic, cardiac, or renal manifestations, respectively. D313Y was associated with a milder, late-onset FD phenotype, as indicated by the mean patient age of 51 years (95% CI 44-59; I 2 = 94%) and the evidence of alpha-galactosidase A deficiency and globotriaosylceramide accumulation in 26.7% (95% CI 15.3%-40%; I 2 = 34%) and 16.2% (95% CI 8%-26.4%; I 2 = 35%) of cases, respectively. D313Y -positive patients displayed predominantly neurologic FD manifestations (58.1%, 95% CI 37.7%-77.1%; I 2 = 78%), with central and peripheral nervous system (CNS/PNS) involvement noted in 28.2% (95% CI 15.4%-43.2%; I 2 = 51%) and 28.5% (95% CI 17.8%-40.5%; I 2 = 61%) of cases, respectively., Discussion: D313Y variation seems to correlate with an atypical, mild late-onset phenotype with predominantly neurologic FD manifestations. Monitoring for CNS/PNS involvement is thus paramount to identify D313Y -positive patients with latent or early-FD pathology, which may qualify for enzyme-replacement therapy or chaperone treatment., (© 2022 American Academy of Neurology.)

GRIN2B mutations are rare but often associated with patients having severe neurodevelopmental disorders with varying range of symptoms such as intellectual disability, developmental delay and epilepsy. Patient symptoms likely arise from mutations disturbing the role that the encoded NMDA receptor subunit, GluN2B, plays at neuronal connections in the developing nervous system. In this study, we investigated the cell-autonomous effects of putative gain- (GoF) and loss-of-function (LoF) missense GRIN2B mutations on excitatory synapses onto CA1 pyramidal neurons in organotypic hippocampal slices. In the absence of both native GluN2A and GluN2B subunits, functional incorporation into synaptic NMDA receptors was attenuated for GoF mutants, or almost eliminated for LoF GluN2B mutants. NMDA-receptor-mediated excitatory postsynaptic currents (NMDA-EPSCs) from synaptic GoF GluN1/2B receptors had prolonged decays consistent with their functional classification. Nonetheless, in the presence of native GluN2A, molecular replacement of native GluN2B with GoF and LoF GluN2B mutants all led to similar functional incorporation into synaptic receptors, more rapidly decaying NMDA-EPSCs and greater inhibition by TCN-201, a selective antagonist for GluN2A-containing NMDA receptors. Mechanistic insight was gained from experiments in HEK293T cells, which revealed that GluN2B GoF mutants slowed deactivation in diheteromeric GluN1/2B, but not triheteromeric GluN1/2A/2B receptors. We also show that a disease-associated missense mutation, which severely affects surface expression, causes opposing effects on NMDA-EPSC decay and charge transfer when introduced into GluN2A or GluN2B. Finally, we show that having a single null Grin2b allele has only a modest effect on NMDA-EPSC decay kinetics. Our results demonstrate that functional incorporation of GoF and LoF GluN2B mutants into synaptic receptors and the effects on EPSC decay times are highly dependent on the presence of triheteromeric GluN1/2A/2B NMDA receptors, thereby influencing the functional classification of NMDA receptor variants as GoF or LoF mutations. These findings highlight the complexity of interpreting effects of disease-causing NMDA receptor missense mutations in the context of neuronal function.

Dominant mutations in the human gene GRIN2A, encoding NMDA receptor (NMDAR) subunit GluN2A, make a significant and growing contribution to the catalogue of published single-gene epilepsies. Understanding the disease mechanism in these epilepsy patients is complicated by the surprising diversity of effects that the mutations have on NMDARs. Here we have examined the cell-autonomous effect of five GluN2A mutations, 3 loss-of-function and 2 gain-of-function, on evoked NMDAR-mediated synaptic currents (NMDA-EPSCs) in CA1 pyramidal neurons in cultured hippocampal slices. Despite the mutants differing in their functional incorporation at synapses, prolonged NMDA-EPSC current decays (with only marginal changes in charge transfer) were a common effect for both gain- and loss-of-function mutants. Modelling NMDA-EPSCs with mutant properties in a CA1 neuron revealed that the effect of GRIN2A mutations can lead to abnormal temporal integration and spine calcium dynamics during trains of concerted synaptic activity. White adipose tissue (WAT) thermogenic activity may play a role in whole-body energy balance and two of its main regulators are thought to be environmental temperature (T env ) and exercise. Low T env may increase uncoupling protein one (UCP1; the main biomarker of thermogenic activity) in WAT to regulate body temperature. On the other hand, exercise may stimulate UCP1 in WAT, which is thought to alter body weight regulation. However, our understanding of the roles (if any) of T env and exercise in WAT thermogenic activity remains incomplete. Our aim was to examine the impacts of low T env and exercise on WAT thermogenic activity, which may alter energy homeostasis and body weight regulation. We conducted a series of four experimental studies, supported by two systematic reviews and meta-analyses. We found increased UCP1 mRNA (p = 0.03; but not protein level) in human WAT biopsy samples collected during the cold part of the year, a finding supported by a systematic review and meta-analysis (PROSPERO review protocol: CRD42019120116). Additional clinical trials (NCT04037371; NCT04037410) using Positron Emission Tomography/Computed Tomography (PET/CT) revealed no impact of low T env on human WAT thermogenic activity (p > 0.05). Furthermore, we found no effects of exercise on UCP1 mRNA or protein levels (p > 0.05) in WAT biopsy samples from a human randomized controlled trial (Clinical trial: NCT04039685), a finding supported by systematic review and meta-analytic data (PROSPERO review protocol: CRD42019120213). Taken together, the present experimental and meta-analytic findings of UCP1 and SUV max , demonstrate that cold and exercise may play insignificant roles in human WAT thermogenic activity.
