Background: The analysis of the procedural memory is particularly relevant in neurodegenerative disorders like Parkinson’s disease, due to the central role of the basal ganglia in procedural memory. It has been shown that anterograde procedural memory, the ability to learn a new skill, is impaired in Parkinson’s disease. However, retrograde procedural memory, the long-term retention and execution of skills learned in earlier life stages, has not yet been systematically investigated in Parkinson’s disease. Objective: This study aims to investigate retrograde procedural memory in people with Parkinson’s disease. We hypothesized that retrograde procedural memory is impaired in people with Parkinson’s disease compared to an age- and gender-matched control group. Methods: First, we developed the CUPRO evaluation system, an extended evaluation system based on the Cube Copying Test, to distinguish the cube copying procedure, representing functioning of retrograde procedural memory, and the final result, representing the visuo-constructive abilities. Development of the evaluation system included tests of discriminant validity. Results: Comparing people with typical Parkinson’s disease (n = 201) with age- and gender-matched control subjects (n = 201), we identified cube copying performance to be significantly impaired in people with Parkinson’s disease (p = 0.008). No significant correlation was observed between retrograde procedural memory and disease duration. Conclusion: We demonstrated lower cube copying performance in people with Parkinson’s disease compared to control subjects, which suggests an impaired functioning of retrograde procedural memory in Parkinson’s disease.

Surface curvature both emerges from, and influences the behavior of, living objects at length scales ranging from cell membranes to single cells to tissues and organs. The relevance of surface curvature in biology is supported by numerous experimental and theoretical investigations in recent years. In this review, first, a brief introduction to the key ideas of surface curvature in the context of biological systems is given and the challenges that arise when measuring surface curvature are discussed. Giving an overview of the emergence of curvature in biological systems, its significance at different length scales becomes apparent. On the other hand, summarizing current findings also shows that both single cells and entire cell sheets, tissues or organisms respond to curvature by modulating their shape and their migration behavior. Finally, the interplay between the distribution of morphogens or micro-organisms and the emergence of curvature across length scales is addressed with examples demonstrating these key mechanistic principles of morphogenesis. Bicontinuous membranes in cell organelles epitomize nature’s ability to create complex functional nanostructures. Like their synthetic counterparts, these membranes are characterized by continuous membrane sheets draped onto topologically complex saddle-shaped surfaces with a periodic network-like structure. Their structure sizes, (around 50–500 nm), and fluid nature make transmission electron microscopy (TEM) the analysis method of choice to decipher their nanostructural features. Here we present a tool, Surface Projection Image Recognition Environment (SPIRE), to identify bicontinuous structures from TEM sections through interactive identification by comparison to mathematical “nodal surface” models. The prolamellar body (PLB) of plant etioplasts is a bicontinuous membrane structure with a key physiological role in chloroplast biogenesis. However, the determination of its spatial structural features has been held back by the lack of tools enabling the identification and quantitative analysis of symmetric membrane conformations. Using our SPIRE tool, we achieved a robust identification of the bicontinuous diamond surface as the dominant PLB geometry in angiosperm etioplasts in contrast to earlier long-standing assertions in the literature. Our data also provide insights into membrane storage capacities of PLBs with different volume proportions and hint at the limited role of a plastid ribosome localization directly inside the PLB grid for its proper functioning. This represents an important step in understanding their as yet elusive structure–function relationship., The SPIRE tool facilitates recognition of bicontinuous phases on transmission electron micrographs, which enables in-depth spatial prolamellar body structure analysis from 2D micrographs.

This article is a reflection on the themes of the Faraday Discussion meeting on ‘Biological and bio-inspired optics’ held from 20 to 22 July 2020. It is a personal perspective on the nature of this field as a broad and interdisciplinary field that has led to a sound understanding of the material properties of biological nanostructured and optical materials. The article describes how the nature of the field and the themes of the conference are reflected in particular in work on the 3D bicontinuous biophotonic nanostructures known as single gyroids and in bicontinuous structures more broadly. Such single gyroid materials are found for example in the butterfly Thecla opisena, where the questions of biophotonic response, of bio-inspired optics, of the relationship between structure and function, and of the relationship between natural and synthetic realisations are closely interlinked. This multitude of facets of research on single gyroid structures reflects the beauty of the broader field of biophotonics, namely as a field that lives through embracing the serendipitous discovery of the biophotonic marvels that nature offers to us as seeds for in-depth analysis and understanding. The meandering nature of its discoveries, and the need to accept the slowness that comes from exploration of intellectually new or foreign territory, mean that the field shares some traits with biological evolution itself. Looking into the future, I consider that a closer engagement with living tissue and with the biological questions of function and formation, rather than with the materials science of biological materials, will help ensure the continuing great success of this field.

Our approach, following pioneering work by Deng and Mieczkowski (1998), creates synthetic TEM images of known bicontinuous geometries for interactive structure identification. We apply the method to the inner membrane network in plant cell chloroplast precursors and achieve a robust identification of the bicontinuous diamond surface as the dominant geometry in several plant species. This represents an important step in understanding their as yet elusive structure-function relationship.

Nature’s optical nanomaterials are poised to form the platform for future optical devices with unprecedented functionality. The brilliant colors of many animals arise from the physical interaction of light with nanostructured, multifunctional materials. While their length scale is typically in the 100-nm range, the morphology of these structures can vary strongly. These biological nanostructures are obtained in a controlled manner, using biomaterials under ambient conditions. The formation processes nature employs use elements of both equilibrium self-assembly and far-from-equilibrium and growth processes. This renders not only the colors themselves, but also the formation processes technologically and ecologically highly relevant. Yet, for many biological nanostructured materials, little is known about the formation mechanisms—partially due to a lack of in vivo imaging methods. Here, we present the toolbox of natural multifunctional nanostructures and the current knowledge about the understanding of their far-from-equilibrium assembly processes.

BACKGROUND: After the World Health Organization declared the outbreak of coronavirus disease to be a public health emergency of international concern on January 30, 2020, the first SARS-CoV-2 infection was detected in Luxembourg on February 29, 2020. Representative population-based data, including asymptomatic individuals for assessing the viral spread and immune response were, however, lacking worldwide. METHODS: Using a panel-based method, we implemented a representative sample of the Luxembourgish population based on age, gender and residency for testing for SARS-CoV-2 infection and antibody status in order to define prevalence irrespective of clinical symptoms. Participants were contacted via email to fill an online questionnaire before biosampling at local laboratories. All participants provided information related to clinical symptoms, epidemiology, socioeconomic and psychological assessments and underwent biosampling, rRT-PCR testing and serology for SARS-CoV-2. RESULTS: We included a total of 1862 individuals in our representative sample of the general Luxembourgish population. Of these, 5 individuals had a current positive result for infection with SARS-CoV-2 based on rRT-PCR. Four of these individuals were oligosymptomatic and one was asymptomatic. Overall we found a positive IgG antibody status in 35 individuals (1.97%), of which 11 reported to be tested positive by rRT-PCR for SARS-CoV-2 previously and showed in addition their IgG positive status also a positive status for IgA. Our data indicate a prevalence of 0.3% for active SARS-CoV-2 infection and an infection rate of 2.15% in the Luxembourgish population between 18 and 79 years of age. CONCLUSIONS: Luxembourgish residents show a low rate of acute infections after 7 weeks of confinement and present with an antibody profile indicative of a more recent immune response to SARS-CoV-2. All infected individuals were oligo- or asymptomatic. Confinement or geometric frustration is known to alter the structure of soft matter, including copolymeric melts, and can consequently be used to tune structure and properties. Here we investigate the self-assembly of ABC and ABB 3-miktoarm star copolymers confined to a spherical shell using coarse-grained dissipative particle dynamics simulations. In bulk and flat geometries the ABC stars form hexagonal tilings, but this is topologically prohibited in a spherical geometry which normally is alleviated by forming pentagonal tiles. However, the molecular architecture of the ABC stars implies an additional 'color constraint' which only allows even tilings (where all polygons have an even number of edges) and we study the effect of these simultaneous constraints. We find that both ABC and ABB systems form spherical tiling patterns, the type of which depends on the radius of the spherical substrate. For small spherical substrates, all solutions correspond to patterns solving the Thomson problem of placing mobile repulsive electric charges on a sphere. In ABC systems we find three coexisting, possibly different tilings, one in each color, each of them solving the Thomson problem simultaneously. For all except the smallest substrates, we find competing solutions with seemingly degenerate free energies that occur with different probabilities. Statistically, an observer who is blind to the differences between B and C can tell from the structure of the A domains if the system is an ABC or an ABB star copolymer system.

We investigate the equilibrium properties of a single area-minimising bubble trapped between two narrowly-separated parallel curved plates. We begin with the simple case of a bubble trapped between concentric spherical plates. We develop a model that shows that the surface tension energy of the bubble is lower when confined between spherical plates as compared to a bubble trapped between flat plates. We confirm our findings by comparing against Surface Evolver simulations. Next, we derive a simple model for a bubble between arbitrarily curved parallel plates. The energy is found to be higher when the local Gaussian curvature of the plates is negative and lower when the curvature is positive. To check the validity of the model we consider a bubble trapped between concentric tori. In the toroidal case we find that the sensitivity of the bubble's energy to the local curvature acts as a geometric potential capable of driving bubbles from regions with negative to positive curvature.

Purpose Structure-property relations, which relate the shape of the microstructure to physical properties such as transport or mechanical properties, need sensitive measures of structure. What are suitable fabric tensors that quantify the shape of anisotropic heterogeneous materials? The mean intercept length is among the most commonly used characteristics of anisotropy in porous media, for example, of trabecular bone in medical physics. Methods We analyze the orientation-biased Boolean model, a versatile stochastic model that represents microstructures as overlapping grains with an orientation bias towards a preferred direction. This model is an extension of the isotropic Boolean model, which has been shown to truthfully reproduce multi-functional properties of isotropic porous media. We explain the close relationship between the concept of intersections with test lines to the elaborate mathematical theory of queues, and how explicit results from the latter can be directly applied to characterize microstructures. Results In this series of two papers, we provide analytic formulas for the anisotropic Boolean model and demonstrate often overlooked conceptual shortcomings of this approach. Queuing theory is used to derive simple and illustrative formulas for the mean intercept length. It separates into an intensity-dependent and an orientation-dependent factor. The global average of the mean intercept length can be expressed by local characteristics of a single grain alone. Conclusions We thus identify which shape information about the random process the mean intercept length contains. The connection between global and local quantities helps to interpret observations and provides insights into the possibilities and limitations of the analysis. In the second paper of this series, we discuss, based on the findings in this paper, short-comings of the mean intercept analysis for (bone-)microstructure characterization. We will suggest alternative and better defined sensitive anisotropy measures from integral geometry.

Self-perceived unmet needs in people with typical and atypical parkinsonism (PwP) and their caregivers, support network, personalized ways to address self-perceived unmet needs during confinement, as well as the prevalence of self-reported COVID-19 related symptoms, confirmed SARS-CoV-2 infection, and self-reported COVID-19 related hospitalization in Luxembourg and the Greater Region were assessed. From 18th March to 10th April 2020, 679 PwP were contacted by phone. Data was collected in the form of a semi-structured interview. The thematic synthesis identified 25 themes where PwP need to be supported in order to cope with consequences of the pandemic, and to adapt their daily and health-related activities. The present work highlights that in the context of personalized medicine, depending on the individual needs of support of the patient the identified self-perceived unmet needs were addressed in various ways ranging from one-directed information over interaction up to proactive counseling and monitoring. Family and health professionals, but also other support systems were taking care of the unmet needs of PwP (e.g., shopping, picking-up medication, etc.) during the pandemic. 7/606 PwP (1.15%) reported COVID-19 related symptoms, 4/606 (0.66%) underwent a rRT-PCR-based diagnostic test and 2/606 (0.33%) were confirmed as SARS-CoV-2 positive. None of these PwP reported being hospitalized due to COVID-19. Our results will allow health professionals to expand their services in a meaningful way i.e., personalize their support in the identified themes and thus improve the healthcare of PwP in times of crisis.

BACKGROUND : In a previous quality control assessment of the management of STEMI in our centre–a tertiary centre in a rural area–we observed a decrease in intrahospital delays over time (2004→2013) from 98→39 minutes (median) but unchanged extrahospital delays (203–220 minutes). AIM :To elucidate the determinants of these long extrahospital delays, 181 patients, undergoing PCI for STEMI (2013–2017) in our centre, were carefully interviewed. The first medical contact (FMC) allowed to assess 2 periods: onset of symptoms–FMC (period 1) and FMC–diagnosis (period 2). Results are given as mean values (median). RESULTS :In 36%, the FMC took place via the general practitioner (GP). Period 1 was associated with the type of FMC (GP: 210 ± 294, median = 79 min/EM call: 78 ± 114, median = 43 min/direct hospital admission: 222 ± 44, median = 115 min), the moment of symptoms onset (0–4 am: 198 ± 192, median 142 min/8–12 am: 96 ± 126, median 60 min), age (70–80 y: 262 ± 324, median 120 min/50–60 y: 198 ± 438, median 63 min) and the distance to our PCI centre expressed in minutes by car (< 30′: 108 ± 156, median = 60 min/ > 60′: 216 ± 288, median = 116 min). Period 2 was depending on the type of FMC (GP: 234 ± 600, median 56 min/EM call: 31 ± 29, median 22 min/direct hospital admission: 25 ± 55, median 10 min). CONCLUSION :In our local experience, FMC via the GP in case of STEMI is a major determinant for a longer extrahospital delay and should therefore be discouraged.

Studies have been showing a negative impact of pandemic control measures on mental health. However, few studies assessed these effects longitudinally during the peak of the first wave of the COVID-19 pandemic. The goals of this study were to explore whether differential effects of COVID-19 restrictions on mental health could be observed by sex and age in a Luxembourgish nationally representative sample during the initial outbreak of COVID-19. Furthermore, we aimed to assess whether there are differences in risk and protective factors longitudinally at two assessment times. A total of 1,756 respondents aged 18 years and older (50.74% women) reported sociodemographic and socio-economic characteristics, depression, anxiety, stress, and loneliness. Women and younger respondents reported higher rates of severe depression and anxiety symptoms, suggesting higher vulnerability to the pandemic control measures. This study contributes to the investigation of mental health consequences of the pandemic and the pandemic control measures, particularly related to shifts in care task responsibilities, gender and socio-economic inequalities, as well as younger groups' uncertainty about the future.

Control over the deformation behaviour that a cellular structure shows in response to imposed external forces is a requirement for the effective design of mechanical metamaterials, in particular those with negative Poisson’s ratio. This article sheds light on the old question of the relationship between geometric microstructure and mechanical response, by comparison of the deformation properties of bar-and-joint-frameworks with those of their realisation as a cellular solid made from linear-elastic material. For ordered planar tessellation models, we find a classification in terms of the number of degrees of freedom of the framework model: first, in cases where the geometry uniquely prescribes a single deformation mode of the framework model, the mechanical deformation and Poisson’s ratio of the linearly-elastic cellular solid closely follow those of the unique deformation mode; the result is a bending-dominated deformation with negligible dependence of the effective Poisson’s ratio on the underlying material’s Poisson’s ratio and small values of the effective Young’s modulus. Second, in the case of rigid structures or when geometric degeneracy prevents the bending-dominated deformation mode, the effective Poisson’s ratio is material-dependent and the Young’s modulus View the MathML sourceE˜cs large. All analysed structures of this type have positive values of the Poisson’s ratio and large values of View the MathML sourceE˜cs. Third, in the case, where the framework has multiple deformation modes, geometry alone does not suffice to determine the mechanical deformation. These results clarify the relationship between mechanical properties of a linear-elastic cellular solid and its corresponding bar-and-joint framework abstraction. They also raise the question if, in essence, auxetic behaviour is restricted to the geometry-guided class of bending-dominated structures corresponding to unique mechanisms, with inherently low values of the Young’s modulus.

Er stellt Moglichkeiten zur Beobachtung der Veranderung von Denk- und Handlungsmustern sowie interaktive Krisenkategorisierungen bei der Bewaltigung einer Krise dar. Individuelle Denk- und Handlungsmuster werden aufgebrochen, aber auch soziale Deutungsmuster werden modifiziert. Wahrend des Bewaltigungsprozesses werden viele kleine und grosere Entscheidungen getroffen, die schlieslich zu einer veranderten Denk- und Lebensweise fuhren. Solche Vorgange sind im Alltagsgeschehen kaum zu beobachten, deshalb wurde der Zugang zum Thema uber ethnographische Beobachtungen und die Analyse psychiatrischer Therapiegesprache gewahlt.

Background Numerous reports relate continuous improvement of inhospital outcome after aortic valve replacement (AVR). Less data are available on the changes of preoperative risk profile and on the long-term outcome. Purpose To assess the temporal changes on preoperative risk, short and long-term outcomes, the data of a consecutive series of 2218 procedures of AVR analyzed in a single center during a 20-year period (1993–2013) were performed. Methods Bioprotheses (B) were used in 1326 cases, mechanical (M) valves in 892 cases. Mean age was 71 ± 10 y (B: 76 ± 6 y; M: 63 ± 10 y.). Inhospital mortality was 6.4% (B: 7.8%; M: 4.4%). The inclusion period was divided in 4 periods of 5 y. Both Euroscores were strictly calculated according to the published criteria. Data are presented here according to early period versus the latest period. Results Preoperative risk was 8.1 ± 7.2 vs. 9.1 ± 8.4 (NS) according to the log Euroscore and 4.9 ± 5.5 vs. 4.2 ± 5.0 (P Conclusion Despite a trend of improved of inhospital and 1-y outcome after aortic valve replacement, the long-term outcome has not improved over time. In our experience, AVR by a mechanical prosthesis was associated with better long-term outcome.

We investigate collective motions of points in 2D systems, orchestrated by Lloyd algorithm. The algorithm iteratively updates a system by minimising the total quantizer energy of the Voronoi landscape of the system. As a result of a tradeoff between energy minimisation and geometric frustration, we find that optimised systems exhibit a defective landscape along the process, where strands of 5- and 7-coordinated dislocations are embedded in the hexatic phase. In particular, dipole defects, each of which is the simplest possible pair of a pentagon and a heptagon, come into the picture of dynamical arrest, as the system freezes down to a disordered hyperuniform state. Moreover, we explore the packing fractions of 2D disk packings associated to the obtained hyperuniform systems by considering the maximum inscribed disks in their Voronoi cells.

The Quantizer problem is a tessellation optimisation problem where point configurations are identified such that the Voronoi cells minimise the second moment of the volume distribution. While the ground state (optimal state) in 3D is almost certainly the body-centered cubic lattice, disordered and effectively hyperuniform states with energies very close to the ground state exist that result as stable states in an evolution through the geometric Lloyd's algorithm [Klatt et al. Nat. Commun., 10, 811 (2019)]. When considered as a statistical mechanics problem at finite temperature, the same system has been termed the 'Voronoi Liquid' by [Ruscher et al. EPL 112, 66003 (2015)]. Here we investigate the cooling behaviour of the Voronoi liquid with a particular view to the stability of the effectively hyperuniform disordered state. As a confirmation of the results by Ruscher et al., we observe, by both molecular dynamics and Monte Carlo simulations, that upon slow quasi-static equilibrium cooling, the Voronoi liquid crystallises from a disordered configuration into the body-centered cubic configuration. By contrast, upon sufficiently fast non-equilibrium cooling (and not just in the limit of a maximally fast quench) the Voronoi liquid adopts similar states as the effectively hyperuniform inherent structures identified by Klatt et al. and prevents the ordering transition into a BCC ordered structure. Background. Mid-March 2020, with exponentially increasing COVID-19 infections, Luxembourg closed schools and businesses, and declared a crisis (état de crise) to implement confinement measures, including orders to not leave the home unless to fulfill essential needs. The psychological consequences of these policy responses to the pandemic on older people, considered a high-risk group, were unknown at the time. The aim of this study was to use the nationally representative CON-VINCE study that assessed mental health at the height of the confinement measures mid-April 2020, to assess the psychological impact of quarantine on older adults. Method. A total of 451 participants aged 60+ years (55.0% male) filled in the CES-D, the GAD-7 and the 3-item loneliness scale that measured depressive symptoms, level of anxiety, and feelings of social isolation. Results. Mean age was 67.7 years (SD 5.5), average number of school years were 13.1 (SD 3.6). The participants were mainly of Luxembourgish nationality (87.8%), and a majority (69.8%) was married. Clinically relevant depression scores were present in 13.1%, generalized anxiety in 1.8%, and self-perceived isolation in 16.9% of participants. The role of particle shape in self-assembly processes is a double-edged sword. On the one hand, particle shape and particle elongation are often considered the most fundamental determinants of soft matter structure formation. On the other hand, structure formation is often highly sensitive to details of shape. Here we address the question of particle shape sensitivity for the self-assembly of hard pear-shaped particles, by studying two models for this system: a) the pear hard Gaussian overlap (PHGO) and b) hard pears of revolution (HPR) model. Hard pear-shaped particles, given by the PHGO model, are known to form a bicontinuous gyroid phase spontaneously. However, this model does not replicate an additive object perfectly and, hence, varies slightly in shape from a "true" pear-shape. Therefore, we investigate in the first part of this series the stability of the gyroid phase in pear-shaped particle systems. We show based on the HPR phase diagram that the gyroid phase does not form in pears with such "true" hard pear-shaped potential. Moreover, we acquire first indications from the HPR and PHGO pair-correlation functions that the formation of the gyroid is probably attributed to the small non-additive properties of the PHGO potential., Comment: 25 pages, 8 figures

Assemblies of anisotropic particles commonly appear in studies of active many-body systems. However, in two dimensions, the geometric ramifications of the finite density of such objects are not entirely understood. To fully characterize these effects, we perform an in-depth study of random assemblies generated by a slow compression of frictionless elliptical particles. The obtained configurations are then analysed using the Set Voronoi tessellation which takes the particle shape into the account. Not only that we analyse most scalar and vectorial morphological measures, which are commonly discussed in the literature or which have been recently addressed in experiments, but also systematically explore the correlations between them. While in a limited range of parameters similarities with findings in 3D assemblies could be identified, important differences are found when a broad range of aspect ratios and packing fractions are considered. The data discussed in this study should thus provide a unique reference set such that geometric effects and differences from random assemblies could be clearly identified in more complex systems, including ones with soft and active particles that are typically found in biological systems.

The mechanisms behind the formation of bicontinuous nanogeometries, in particular in vivo,\ud remain intriguing. Of particular interest are the many systems where more than one type\ud or symmetry occurs, such as the Schwarz’ diamond surface and Schoen’s gyroid surface;\ud a current example are the butterfly nanostructures often based on the gyroid, and the beetle\ud nanostructures often based on the diamond surface. Here, we present a computational study\ud of self-assembly of the bicontinuous Pn3m diamond phase in an equilibrium ensemble of\ud pear-shaped particles when a small amount of a hard-sphere ‘solvent’ is added. Our results are\ud based on previous work that showed the emergence of the gyroid Ia3d phase in a pure system\ud of pear-shaped particles (Schönhöfer et al 2017 Interface Focus 7 20160161), in which the\ud pear-shaped particles form an interdigitating bilayer reminiscent of a warped smectic structure.\ud We here show that the addition of a small amount of hard spherical particles tends to drive the\ud system towards the bicontinuous Pn3m double diamond phase, based on Schwarz diamond\ud minimal surface. This result is consistent with the higher degree of spatial heterogeneity of\ud the diamond minimal surface as compared to the gyroid minimal surface, with the hard-sphere\ud ‘solvent’ acting as an agent to relieve packing frustration. However, the mechanism by which\ud this relief is achieved is contrary to the corresponding mechanism in copolymeric systems; the\ud spherical solvent tends to aggregate within the matrix phase, near the minimal surface, rather\ud than within the labyrinthine channels. While it may relate to the specific form of the potential\ud used to approximate the particle shape, this mechanism hints at an alternative way for particle\ud systems to both release packing frustration and satisfy geometrical restrictions in double\ud diamond configurations. Interestingly, the lattice parameters of the gyroid and the diamond\ud phase appear to be commensurate with those of the isometric Bonnet transform.
