1. Investigating the Aroma of Syrah Wines from the Northern Rhone Valley Using Supercritical CO2-Dearomatized Wine as a Matrix for Reconstitution Studies

Abstract: This study aimed to investigate the key compounds involved in the aroma of French Syrah wines from the northern Rhone valley from two vintages characterized by distinct climatic conditions. The volatile composition of the wines was assessed through the determination of 76 molecules. After identifying the best matrix and best model for aroma reconstitution studies, omission tests were conducted using the Pivot© profile method. For both vintages, 35 molecules with odor activity values (OAVs) above 0.5 were identified. While remarkably high levels of 2-furfurylthiol (FFT) were reported in both wines, rotundone and 3-sulfanylhexanol (3SH) enabled the strongest discrimination between the two wines. Wine dearomatized using supercritical carbon dioxide (sCO2) was identified as the best matrix. The best models built using this matrix were composed of molecules with OAV > 5 and OAV > 10 highlighting that this dearomatization approach can be valuable to reconstitute the aroma of wine using a small number of molecules. For the cool vintage wine, the omission of rotundone and FFT had the greatest impact on the olfactive profile for non-anosmic and anosmic respondents to rotundone, respectively. 3SH, whose omission decreased the rating of the ‘fruity’ attribute, was identified as the main contributor to the aroma of Syrah wine produced in the warm vintage.

2. Détection précoce de la prise de xénogreffe de leucémie aigue myéloïde (LAM) chez la souris à l'aide d'un prélèvement sanguin non terminal

Abstract: L’efficacité de nouvelles thérapies de la leucémie aiguë myéloïde (LAM) est testée en observant leurs effets chez des souris immunodéficientes (NOD/LTSz-scid IL2Rgamma null ou NSG) greffées avec des lignées de cellules de LAM humaines. Pour s’assurer de la prise de la greffe on a actuellement recours à des ponctions de moelle osseuse, un examen biologique très invasif. Le but était donc de substituer un ou plusieurs marqueurs sanguins, moins invasifs, qui soient aussi, voire plus, précoces que le myélogramme. Une première série de travaux a permis de démontrer que les conditions pré-analytiques permettant d’obtenir un spécimen sanguin de très bonne qualité pour des analyses hématologiques et biochimiques, en respectant la survie de la souris et son bien être étaient les suivantes : anesthésie à l’isoflurane, ponction de la veine jugulaire à l’aiguille 25G, aspiration douce de 100 à 200 μL dans un tube EDTA. Dans ces conditions, on n’observe que peu d’agrégats plaquettaires et l’addition d’agents anti-agrégants est sans effet. Le facteur déterminant de la qualité du spécimen est alors celle de la qualité de l’acte de prélèvement. L’utilisation de marqueurs sanguins nécessite d’en connaître les intervalles de référence. Cela a été fait conformément aux recommandations internationales pour les bilans hématologiques et biochimiques de routine des souris NSG dans des effectifs réduits (20 mâles et 20 femelles) pour des raisons éthiques ainsi que de coût des animaux. Par comparaison à des souris habituellement utilisées, la principale différence est la faible concentration de leucocytes, principalement de lymphocytes. Il en résulte également que l’identification des types de leucocytes par les analyseurs est souvent erronée et doit être faite sur un frottis sanguin par comptage au microscope. L’ensemble des analyses pouvant prendre beaucoup de temps, il est nécessaire de s’assurer de la stabilité des analytes étudiés dans le sang de souris. Après avoir étudié les variations des bilans hématologiques pendant 72 heures à température ambiante et en réfrigération, il apparaît que, comme dans les autres espèces, plus l’analyse est précoce meilleure est la stabilité et qu’au-delà de 24 h à 4°C des altérations notables apparaissent qui doivent être connues pour l’interprétation de l'hémogramme; un autre point essentiel est que le frottis sanguin doit être fait le plus précocement possible pour limiter au maximum les anomalies morphologiques. Chez des souris NSG ayant subi une xénogreffe de cellules de LAM humaine, peu de variations sont observées après 10 jours alors qu’elles sont nombreuses à J17, notamment avec une augmentation des concentrations de neutrophiles et de monocytes. Ces anomalies ne sont pas plus précoces que la chronologie connue de la prise de greffe mais elles sont accompagnées d’images spécifiques des nuages de points générées avec des analyseurs d'hématologie utilisant le principe de la cytométrie de flux, identifiables facilement et immédiatement donnant ainsi un marqueur très peu coûteux accessible à tous les laboratoires sans compétence spécifique en cytologie. Ayant ainsi défini les conditions pré-analytiques d’obtention de spécimens sanguins de qualité chez la souris et montré que la prise de greffe de lignées cellulaires de LAM est facilement identifiable avec une analyse hématologique de routine nécessitant un très faible volume de sang, il devient possible d’envisager leur utilisation en augmentant le nombre d’analyses chez le même sujet en réduisant le volume collecté à 50 μL ce qui pourrait permettre une détection plus précoce de la prise de greffe ; de même il faudra étudier si des variations analogues sont observées lors de greffes de LAM provenant de patients (PDX).

Abstract: Among grapevine diseases affecting European vineyards, Flavescence dorée (FD) and Grapevine Trunk Diseases (GTD) are considered the most relevant challenges for viticulture because of the damage they cause to vineyards. Unmanned Aerial Vehicle (UAV) multispectral imagery could be a powerful tool for the automatic detection of symptomatic vines. However, one major difficulty is to discriminate different kinds of diseases leading to similar leaves discoloration as it is the case with FD and GTD for red vine cultivars. The objective of this paper is to evaluate the potentiality of UAV multispectral imagery to separate: symptomatic vines including FD and GTD (Esca and black dead arm) from asymptomatic vines (Case 1) and FD vines from GTD ones (Case 2). The study sites are localized in the Gaillac and Minervois wine production regions (south of France). A set of seven vineyards covering five different red cultivars was studied. Field work was carried out between August and September 2016. In total, 218 asymptomatic vines, 502 FD vines and 199 GTD vines were located with a centimetric precision GPS. UAV multispectral images were acquired with a MicaSense RedEdge® sensor and were processed to ultimately obtain surface reflectance mosaics at 0.10 m ground spatial resolution. In this study, the potentiality of 24 variables (5 spectral bands, 15 vegetation indices and 4 biophysical parameters) are tested. The vegetation indices are selected for their potentiality to detect abnormal vegetation behavior in relation to stress or diseases. Among the biophysical parameters selected, three are directly linked to the leaf pigments content (chlorophyll, carotenoid and anthocyanin). The first step consisted in evaluating the performance of the 24 variables to separate symptomatic vine vegetation (FD or/and GTD) from asymptomatic vine vegetation using the performance indicators from the Receiver Operator Characteristic (ROC) Curve method (i.e., Area Under Curve or AUC, sensibility and specificity). The second step consisted in mapping the symptomatic vines (FD and/or GTD) at the scale of the field using the optimal threshold resulting from the ROC curve. Ultimately, the error between the level of infection predicted by the selected variables (proportion of symptomatic pixels by vine) and observed in the field (proportion of symptomatic leaves by vine) is calculated. The same methodology is applied to the three levels of analysis: by vineyard, by cultivar (Gamay, Fer Servadou) and by berry color (all red cultivars). At the vineyard and cultivar levels, the best variables selected varies. The AUC of the best vegetation indices and biophysical parameters varies from 0.84 to 0.95 for Case 1 and 0.74 to 0.90 for Case 2. At the berry color level, no variable is efficient in discriminating FD vines from GTD ones (Case 2). For Case 1, the best vegetation indices and biophysical parameter are Red Green Index (RGI)/ Green-Red Vegetation Index (GRVI) (based on the green and red spectral bands) and Car (linked to carotenoid content). These variables are more effective in mapping vines with a level of infection greater than 50%. However, at the scale of the field, we observe misclassified pixels linked to the presence of mixed pixels (shade, bare soil, inter-row vegetation and vine vegetation) and other factors of abnormal coloration (e.g., apoplectic vines).

Abstract: Ce numéro d'Innovations Agronomiques rassemble des articles issus de communications présentées au colloque national «Les polycultures-élevages: valoriser leurs atouts pour la transition agro-écologique » organisé par le RMT SPyCE et AgroSup Dijon, à Dijon les 10 et 11 octobre 2017.; Recent developments in agriculture in France have been marked by the emergence of collectiveprojects, associating farmers around shared crop areas or bio-gas plants. The potential forsustainability of these projects is discussed. Our objective was to evaluate the impact of two agriculturalprojects involving crops and livestock, with significant exchanges and coordination. One shared croparea and one collective biogas plant were investigated.We aimed to evaluate the main effects of these projects on the sustainability of agricultural systems,and to analyze how they question agricultural systems’ organization and methods for evaluating theirsustainability.Results show an increase of the social sustainability of farms, while a tendency to intensify systems wasobserved. Nevertheless, these projects benefited mainly to a variety of individual projects. They helpedto significantly change the identity of farmers within the group (breeders turned into seed farmers forexample).The study highlights the interrelations between the individual and collective scales, which raisesquestions about the methods of evaluation and action to be implemented for more sustainable systems.; Les systèmes agricoles connaissent aujourd’hui de profondes mutations, notamment l’émergence dedémarches collectives, qui questionnent les modèles organisationnels traditionnels. Des projetsassocient agriculteurs et éleveurs autour de dispositifs collectifs comme des assolements, élevages ouméthaniseurs. Le potentiel de ces dispositifs en termes de durabilité est questionné. Notre objectif estd’évaluer deux projets contrastés associant cultures et élevages autour d’un dispositif collectif: unassolement en commun et un méthaniseur collectif. Nous souhaitions évaluer les principaux effets deces dispositifs sur la durabilité des systèmes agricoles, et analyser en quoi ils questionnentl’organisation des systèmes agricoles et les méthodes d’étude de la durabilité.Les résultats indiquent que les coopérations étudiées participent à augmenter la durabilité sociale desexploitations en augmentant les liens entre membres. L’effet sur la durabilité environnementale estambivalent avec une tendance à l’intensification des systèmes malgré la mise en œuvre de systèmes etde pratiques qui se réfèrent à des démarches d’agroécologie ou d’écologie territoriale. Au-delà del’objectif commun affiché, les coopérations mises en œuvre se font avant tout au service d’une diversitéde projets individuels. Elles ont contribué à modifier fortement l’identité des agriculteurs au sein dugroupe (nouveau métier, voulu ou subi, de semencier pour des éleveurs, par exemple).L’étude met en évidence des complémentarités entre les échelles individuelles et collectives, ce quiquestionne les modes d’évaluation et d’action à mettre en œuvre pour des systèmes plus durables

Abstract: Mountain forests suffer from an increase in frequency and severity of summer droughts and infestations of pathogens and insects. Those factors are causing high mortality of some keystone tree species (forest die-offs). Yet, how tree diebacks and associated changes in forest composition will affect local diversity and ecosystem functions remains unknown. Here, we aim at quantifying the impact of climate-induced forest decline on biodiversity by measuring changes in taxonomic structure of invertebrate communities along gradients of silver fir (Abies alba) dieback in the French Pyrenees. We use DNA metabarcoding to analyse 224 samples of Malaise traps placed on 56 silver fir dominated plots in the French Pyrenees from May to September 2017. Samples were sequenced using Illumina MiSeq and analysed using the DAMe twin-tagging pipeline approach. We conducted taxonomic assignment against reference DNA barcode libraries to streamli ne identification and recover biological information for ecological analysis. We discuss the results of our metabarcoding analysis and the utility of our approach to conduct biomonitoring across a large geographical scale.

Abstract: International audience; Forest edges are often defined as the discontinuity between the forest habitat and an adjacent open habitat, thus they are based on a clear difference in the structure of the dominant vegetation. However, beside this very general definition, in the field we can observe a large diversity of edges, with often different kinds of micro-topography features: bank, ditch, stone wall, path, etc. As these elements are rather common in many temperate forest edges, it seems important to start to characterize them more clearly and with consistency. From a set of observations in south-western France, we build a first typology of the micro-topographic elements associated to forest edges. For each of them we describe the process, natural or human induced, at their origin, and according to the literature available, we identify some of their key ecological roles. Banks, generated by the differential erosion between forest and crops along slopes, are especially analyzed since they are the most common micro-topographic element in our region. It offers many micro-habitat conditions in the soil used by a wide range of species, notably by several bee species. More research is required to study in details the importance of such micro-topographic elements.

Abstract: International audience; The impact of two temperature levels (50 °C and 75 °C) and heating times (30 min and 3 h) on the composition of thermovinified musts and wines from Carignan was investigated at the laboratory scale in 2014 and 2015. The heating temperature had a significant impact on the extraction of amino acids and a probable thermal degradation of anthocyanins was noted at 75 °C. In 2014, musts from grapes that underwent a heat treatment at 50 °C for 3 h had a similar level of phenolic compounds as those treated at 75 °C for 30 min. This indicates that the reduction of the heating temperature in some vintages can be compensated for through an extension of the heating period. Several grape-derived molecules were impacted by the rise in temperature and wines made from grapes treated at 50 °C in most cases contained larger concentrations of geraniol, β-citronellol, β-damascenone and 3-mercaptohexanol.

Abstract: International audience; Polyion complex (PIC) micelles formed from the electrostatic interaction between oppositely charged polymers have been studied for their promising applications in the biomedical field as drug carriers or vectors for gene delivery. In spite of their asset of possible high drug loading, their formation process remains poorly studied. In this work, we investigate the properties of a series of PICs based on poly(ethylene oxide-b-acrylic acid) (PEO–PAA)/dendrigraft poly(L-lysine) (DGL3), using PEO–PAA with different compositions and average molecular weights. For each PEO–PAA/DGL3 pair, the complexes were characterized as a function of the ratios between acid and amine moieties combining different techniques: dynamic light scattering (DLS), flow field-flow fractionation (FlFFF), small-angle X-ray scattering (SAXS), and relaxometry. The coupling of batch techniques, i.e., DLS, SAXS, and relaxometry, together with a soft separation technique like FlFFF enabled a finer analysis to elucidate subtle details of the association process and of the polydispersity of the complexes. We show that the formation of PICs is more complex than previously described. In particular, we demonstrate that PICs with stoichiometry 1:1 may form at low ratios provided that the acidic block is long enough to neutralize the cationic dendrigraft with few polymer chains. Moreover, in such conditions, PICs with stoichiometry 1:1 often coexist with free dendritic polymers and other associated complex species.

Abstract: International audience; Mountain areas in Europe are facing important land use changes, mainly land abandonment combined with agriculture intensification. Mountains are known as multifunctional areas, and land use changes imply management trade-offs because some functions are improved to the detriment of others. It has been proved that social learning can contribute to tackling trade-offs in social-ecological systems by increasing the awareness of interdependencies, especially in multi-stakeholder dialogue processes. How can we foster social learning to inform choices about management of mountain farming systems? In this study, we make the hypothesis that using the concept of ecosystem service in multi-stakeholder dialogue processes can improve awareness of interdependencies, and thus social learning. To test this hypothesis, we study the case of Mont Lozère, Cévennes region, France, that is facing important land use change. We studied trade-offs among ecosystem services due to two agricultural practices: rock removal and ploughing of meadows. We designed a role-playing game to reveal hidden interdependencies between stakeholders, and to open dialogue about the governance of trade-offs between ecosystem services. The analysis of the game sessions is still in progress, but we identified three kinds of learning which seem to be fostered by the game: learning on the issue at stake, learning about others, and organizational learning.

Abstract: International audience; Small forest patches, smaller than 10 ha, are very common in many European landscapes and may represent a significant amount of wood available for logging, but also a support of important parts of biodiversity and of many ecosystem services. Few is known about the status of these quantities, because of the difficulty to sample such small habitats. In this presentation, we measured the amount of wood available, according to several classes of quality, in a sample of very small forest patches in southwestern France. In the same places, plant diversity and a set of related ecosystem services were also measured. The results showed a huge variability between patches regarding the amount of available wood, with some of them with a very dense stock of wood, comparatively with national references. Plant diversity was higher in patches with intermediate level of wood stocks, the lowest richness was in the densest patches, but the plant species were then more typical of large forest flora. It seems that allowing older and bigger trees in small patches could reduce the negative impact of the small size of the patch on plant composition. Few of the other ecosystem services were related to the amount of wood but these complex relationships need more detailed analyses. These preliminary results showed that small forest patches represent a significant amount of wood that may be harvested in the future is the demand continue to increase, but this may have consequences on their conservation value for plants, but apparently not for many other ecosystem services. Management practices are also a factor that need to be considered in the future as it may modify strongly the structure of these forests.

But beyond global trends, little is known about small-scale dynamics and factors affecting rural forests. In the Long-Term Social-Ecological Research (LTSER) platform Vallées et Coteaux de Gascogne, we combined GIS monitoring, ethnographic investigations and mental models to understand rural forests dynamics and related anthropogenic factors.

Abstract: International audience; Panarchy theory provides a framework for understanding the dynamics of complex systems with a strong emphasis on levels of organization, disturbance and resilience. However, the spatial dimension of the systems is not fully taken into account despite its crucial roles for many social and ecological processes. Spatial discontinuities are part of the spatial heterogeneity of many systems; it is the place where two different covers (i.e. forest and agriculture habitats) are in contact along a more or less abrupt border. After showing that the interactions between both habitats and their dynamics are not fully acknowledged in the panarchy framework, I propose some new concepts that could be included in the theory in order to cover more easily such cases. I show that the discontinuity between two phases of the cycle of a same system is different from the discontinuity between two systems, and that the speed of the cycle of both systems is a key parameter. Inclusion of adjacent habitats and their interface in a broader system is not sufficient to explain the properties emerging from the interactions. The usefulness of these concepts is shown with some examples dealing with agriculture/forest interface along edges. A better understanding of the theoretical processes governing edge dynamics and associated ecological functions, helps to include them in natural resources management.

Abstract: In recent decades, improvement of prolificacy and body composition has been accompanied by a substantial increase in the mortality of piglets before weaning. The most critical period is the perinatal period, mostly during the first 24-48 hours following birth. The maturity of piglets, defined as the state of full development for survival at birth, is an important determinant of early mortality. The objective of our project is to take advantage of current knowledge about two pig breeds, Large White (LW) pigs selected for prolificacy and body composition and Meishan (MS) pigs being more robust. Maturity of several tissues and metabolite profiles of various fluids are analyzed on the fetuses (LW, MS and reciprocal F1) at day 90 or 110 of gestation (birth at day 114). Here we presented the transcriptomic analysis done on total blood samples (N=63). We did two different statistical analyses, a supervised one to reveal differential pathways for the interaction between gestational stages and genotypes and an unsupervised analysis (hclust and differential analyses) to identify potential predictors of a lesser maturity at birth. All p-values were adjusted with a Bonferroni correction < 1%. The 265 genes differential for the interaction (Bonferroni 1%) in blood samples revealed many genes for mitochondrial ATP synthesis, transcriptional regulation, and response to hypoxia (overexpressed in LW at day 110 of gestation).

Abstract: International audience; The work presented here suggests a new approach in the critical development of polymeric nanovectors for photodynamic therapy (PDT) against cancer. Whereas hundreds of studies quickly jump forward from formation of self-assemblies to biological application without having a thorough examination of the vector solution, we suggest having a parallel assessment of formation/characterization of the nanovectors and biological activity [1]. This is possible by first using a careful physical chemistry characterization of the vectors by both batch techniques (light and neutron scattering, electron microscopy, atomic force microscopy) and Asymmetrical Flow Field-Flow Fractionation (AsFlFFF) coupled to adequate detectors (refractometry, light scattering) [2]. This enables us to fully characterize the vectors regarding purity, size and zeta potential.

Abstract: International audience; Polymer self-assemblies joining oppositely charged chains, known as polyion complexes (PICs), have been formed using poly(ethyleneoxide – b – acrylic acid)/poly(l-lysine), poly(ethyleneoxide-b-acrylic acid)/dendrigraft poly(l-lysine) and poly[(3-acrylamidopropyl) trimethylammonium chloride – b – N – isopropyl acrylamide]/poly(acrylic acid). The self-assemblies have been first characterized in batch by Dynamic Light Scattering. In a second step, their analysis by Flow Field-Flow Fractionation techniques (FlFFF) was examined. They were shown to be very sensitive to shearing, especially during the focus step of the fractionation, and this led to an incompatibility with asymmetrical FlFFF. On the other hand, Frit Inlet FlFFF proved to be very efficient to observe them, either in its symmetrical (FI-FlFFF) or asymmetrical version (FI-AsFlFFF). Conditions of elution were found to optimize the sample recovery in pure water. Spherical self-assemblies were detected, with a size range between 70–400nm depending on the polymers. Compared to batch DLS, FI-AsFlFFF clearly showed the presence of several populations in some cases. The influence of salt on poly(ethyleneoxide-b-acrylic acid) (PEO-PAA) 6000-3000/dendrigraft poly(l-lysine) (DGL 3) was also assessed in parallel in batch DLS and FI-AsFlFFF. Batch DLS revealed a first process of swelling of the self-assembly for low concentrations up to 0.8M followed by the dissociation. FI-AsFlFFF furthermore indicated a possible ejection of DGL3 from the PIC assembly for concentrations as low as 0.2M, which could not be observed in batch DLS.

Abstract: International audience; The casein micelle is a highly aggregated colloid consisting of phosphoproteins and minerals, in particular calcium and phosphate. Its properties are affected by physico-chemical changes which provide possibilities for the development of new casein aggregates (CAs) with novel functionalities. The aim of this study was to investigate the emulsifying and emulsion-stabilizing capacity of gradually demineralized CAs in model dairy emulsions. Tri sodium citrate (TSC) was used to remove calcium and inorganic phosphate from pure casein micelles in order to produce four suspensions of differently demineralized CAs. Two types of milkfat-in-suspension (30:70 v/v) emulsions were then prepared to study the emulsifying and emulsion-stabilizing capacity of these CAs separately. Casein micelles were progressively demineralized (from 24 to 81% calcium reduction) and dissociated with the increase in TSC concentration. Three distinct populations of particles (micelle-like aggregates, sodium caseinate-like aggregates and casein monomers) were present in every suspension in different proportions. The smaller CAs had better emulsifying capacity and similar surface activity according to interfacial studies. The state of aggregation of the CAs was thus the main factor that controlled their emulsifying capacity. However, the emulsions formed with these smaller aggregates were less stable against creaming and flocculation, but still resisted coalescence under our storage conditions (21 days at 50 °C). The properties of the interfacial casein layers did not depend on the aggregation state of the CAs used to form the emulsions. The differences in instability were attributed to the nature of the non-adsorbed CAs and storage conditions.

Abstract: International audience; Raw milk is often described as a major source of lactic acid bacteria for indigenous lactic starter. These indigenous starters contribute to the sensorial quality of cheese. Raw milk, rich in Lactoccocus lactis may therefore be very interesting for the cheese making. Currently, the most commonly used methods to differentiate lactic acid bacteria, and particularly the closely related phenotypes Lactococcus and Enterococcus, are based on DNA sequencing, but the cost and time required for these analytical methods hinder their use for rapid screening of raw material. The present study therefore proposes a simple alternative method to identify and discriminate against Lactococcus and Enterococcus, at the genus, but also at the species level, that is based on collecting near infrared spectra directly from bacterial colonies in Petri dishes. The infrared spectra of 280 strains of Lactococcus and Enterococcus cultured on solid media were collected by using a spectrometer with a wavelength range of 908 to 1684 nm and a remote probe. The best Classification And Regression Trees models for genus and species discrimination gave an excellent classification rate of 87% on an external validation set (30 strains). Loading line plots, with prominent bands at 900–960 and 1270–1390 nm, confirmed that the source of variation was due to changes in the polysaccharides.

Abstract: International audience; This paper deals with the use of tetrabutylammonium fluoride/dimethylsulfoxide (TBAF/DMSO) to characterize the molar mass distribution of non-derivatized cellulosic samples by size exclusion chromatography (SEC). Different cellulose samples with various average degree of polymerization (DP) were first solubilized in this solvent system, with increasing TBAF rates, and then analyzed by SEC coupled to a refractive index detector (RID), using DMSO as mobile phase. The Molar Masses (MM) obtained by conventional calibration were then discussed and compared with suppliers’ data and MM determined by viscosimetry measurements. By this non-classic method, molar mass of low DP samples (Avicel® and cotton fibers) have been determined. For high DP samples (α-cellulose and Vitacel®), dissolution with TBAF concentration of 10 mg/mL involved elution of cellulose aggregates in the exclusion volume, related to an incomplete dissolution or the dilution of TBAF molecules in elution solvent, preventing the correct evaluation of their molar mass.

Abstract: International audience; This work deals with the modifications resulting from the dissolution of four commercial cellulosic samples, with different crystallinity rates and degrees of polymerization (DPs), in four solvent systems, known and used to dissolve cellulose. The dissolution conditions were optimized for the 16 various systems and followed by turbidity measurements. After regeneration, the samples were analyzed by thermal gravimetric analysis (TGA), scanning electron microscopy (SEM), and X-ray diffractometry (XRD) to study their modification. Viscosimetry measurements were used to evaluate the potential decrease of the DP after dissolution. The observed structural modifications established that, for low DP, all the solvent systems were efficient in dissolving the cellulose without altering the DP, except BMIM [Cl], which provoked a decrease of up to 40 % and a decrease of around 20 % of the degradation temperature (onset temperature, T o). For high molecular weight (MW) celluloses, DMSO/TBAF was the only system to allow a complete dissolution without any molar mass loss and degradation temperature modification.

Abstract: Dans un contexte de changement climatique, des dispositifs sont mis en place par les politiques publiques pour permettre le déplacement de la biodiversité et la recolonisation de nouveaux milieux (TVB, trame de vieux bois. . . ). Ces dispositifs semblent adaptés pour les espèces animales les plus mobiles. Or, les espèces ayant une faible capacité de déplacement représentent un enjeu important pour l'efficacité des dispositifs mis en place. Un grand nombre de ces espèces demeurent aujourd'hui mal connues. En forêt, les coléoptères saproxyliques aptères de litière sont des espèces ayant des capacités de déplacement et de dispersion limitées en raison de leurs petites tailles, de leurs aptérismes et de leurs exigences trophiques. Ces espèces constituent le modèle biologique de cette étude pour comprendre comment des espèces à faible capacité de déplacement peuvent évoluer dans les forêts et les paysages anthropisés, dans l'espace et le temps. L'échantillonnage par tamisage de litière de dix arbres par peuplement en utilisant un Winkler et une extraction par Berlese, a montré une puissance d'échantillonnage moyenne de 97.2% pour six forêts. Des relevés mensuels sur deux ans ont permis de caractériser la phénologie des espèces, à savoir qu'elles sont i/ présentes dans la litière toute l'année, ii/ davantage abondantes au printemps et en automne et iii/ ne présentent pas de variation interannuelle. Nous avons comparé la richesse spécique et la répartition des espèces, entre les forêts subnaturelles et les peuplements exploités des montagnes Pyrénéennes. Ceci a montré que les espèces sont peu impactées par l'exploitation sylvicole traditionnelle, mais présentent une grande variabilité de répartition qui semble liée à l'impact anthropique passé. Le paysage fragmenté des Coteaux de Gascogne s'est avéré être très pauvre en coléoptères saproxyliques aptères. Étonnamment, dans ce territoire, les répartitions des espèces sont mieux expliquées par les caractéristiques des paysages que celles des forêts. Pour la moitié des espèces, leurs répartitions sont mieux expliquées par la composition du paysage en 1850 que celle d'aujourd'hui. Un seuil critique de 15% de proportion de forêts dans le paysage en 1850 a pu être mis en évidence pour une espèce (Dienerella clathrata). Nous avons montré que les espèces sont capables (au moins) de se déplacer à travers les haies, quelles que soient les caractéristiques de ces dernières. Si les espèces étaient présentes dans la forêt connectée à la haie, elles ont été échantillonnées dans cette dernière. Les capacités de déplacement de trois espèces ont été étudiées en laboratoire et montrent une vitesse moyenne de 1,6m.h-1. Ces deux éléments conrment le fait que ces espèces sont mobiles dans le paysage. L'ensemble de ces résultats, ainsi que le fait que les territoires les moins anthropisés soient les plus riches en coléoptères saproxyliques aptères, nous amènent à conclure que ce groupe taxonomique supporte bien les impacts humains de faible amplitude spatiale et temporelle, mais qu'ils sont peu résilients à une anthropisation importante dans le temps et dans l'espace.

Abstract: International audience; In most vineyards worldwide, agents of grapevine trunk diseases represent a real threat for viticulture and are responsible for significant economic loss to the wine industry. The conventional microbiological isolation technique used to diagnose this disease is tedious and frequently leads to false negatives. Thus, a dire need exists for an alternative method to detect this disease. One possible way involves infrared spectroscopy, which is a rapid, nondestructive analytical tool that is commonly used for quality control of feed stuffs. In the present work, a midinfrared spectrometer was tested as a fast tool for detecting agents of grapevine trunk disease. Midinfrared spectra were collected from 70 Vitis vinifera L. cv. Cabernet-Sauvignon one year old trunk-wood samples that were infected naturally in one viticulture nursery of the south of France. The samples underwent polymerase chain reaction and morphological identification, and the results were correlated to the midinfrared spectra by using multivariate analysis to discriminate between noninfected and infected samples. Based on comparison with some control samples, the highest percentage of correct identification of fungal contamination when using the midinfrared spectroscopy method is 80%.

Abstract: International audience; The use of ozonized vegetable oils as drugs or cosmetics requires having data on their stability over the time. In this study, ozonized sunflower oil was stored under different temperatures for up to 1 year. Peroxide index, acidity value, gas chromatography profile, infrared profile and antibacterial activity (against Streptococcus uberis) of this ozonized oil were followed. The results highlight the fact that the better way to preserve the initial composition of ozonized oil is to keep it at low temperature. However, the antibacterial activity is not diminished by the changes occurring to the composition.

Abstract: International audience; PURPOSE: Evaluation of particle size distribution (PSD) of multimodal dispersion of nanoparticles is a difficult task due to inherent limitations of size measurement methods. The present work reports the evaluation of PSD of a dispersion of poly(isobutylcyanoacrylate) nanoparticles decorated with dextran known as multimodal and developed as nanomedecine.METHODS: The nine methods used were classified as batch particle i.e. Static Light Scattering (SLS) and Dynamic Light Scattering (DLS), single particle i.e. Electron Microscopy (EM), Atomic Force Microscopy (AFM), Tunable Resistive Pulse Sensing (TRPS) and Nanoparticle Tracking Analysis (NTA) and separative particle i.e. Asymmetrical Flow Field-Flow Fractionation coupled with DLS (AsFlFFF) size measurement methods.RESULTS: The multimodal dispersion was identified using AFM, TRPS and NTA and results were consistent with those provided with the method based on a separation step prior to on-line size measurements. None of the light scattering batch methods could reveal the complexity of the PSD of the dispersion.CONCLUSIONS: Difference between PSD obtained from all size measurement methods tested suggested that study of the PSD of multimodal dispersion required to analyze samples by at least one of the single size particle measurement method or a method that uses a separation step prior PSD measurement.

Abstract: International audience; In order to compare the efficiency of crosslinked nano-vectors in the field of photodynamic therapy (PDT) both on 2D and 3D cell cultures, various polymeric crosslinked self-assemblies based on poly-(ethyleneoxide-b-epsilon-caprolactone) have been synthesized by radical polymerization of acrylate end-functionalized polymers. Crosslinked self-assemblies obtained from the reaction of the functionalized polymers with ethyleneglycoldimethacrylate (EGDMA) were compared to chain-end polymerized and to unreacted ones. Polymeric micelles with a size between 10 and 20 nm were obtained, as well as an elongated system with a length close to 100 nm. They all have been characterized by Transmission Electron Microscopy and Dynamic Light Scattering but also by Asymmetrical Flow Field-Flow Fractionation in order to prove that they consisted of pure self-assemblies. Chain-end polymerization or crosslinking did not induce any change in morphology nor strong size modification. After post-encapsulation of a photosensitizer, namely Pheophorbide a, the systems have been examined for their potential use in PDT on HCT-116 and FaDu human tumor cell lines both in 2D and 3D cultures. The crosslinked vectors were observed to be the most efficient on both cell lines cultivated in 3D spheroids, whereas unreacted or chain-end polymerized ones presented a lower activity. This was different from the trend observed in 2D cell cultures where an uncrosslinked micelle was observed to be efficient at a lower concentration compared to its chain-end polymerized or crosslinked analogue. The different synthesized self-assemblies also allowed assessing the influence of polymer chain length and shape on PDT efficiency. The molecular weight of the polymer did not lead in our case to efficiency change, for similar size and surface characteristics. As for the shape effect, the elongated self-assembly was not observed in our case to be more efficient than spherical micelles. Crosslinked polymeric vectors are therefore promising vectors for 3D tumor treatment.

Abstract: International audience; The objective of this work was to assess the relation between the purity of polymeric self-assemblies vectors solution and their photodynamic therapeutic efficiency. For this, several amphiphilic block copolymers of poly(ethyleneoxide-b-ε-caprolactone) have been used to form self-assemblies with different morphologies (micelles, worm-like micelles or vesicles). In a first step, controlled mixtures of preformed micelles and vesicles have been characterized both by dynamic light scattering and asymmetrical flow field flow fractionation (AsFlFFF). For this, a custom-made program, STORMS, was developed to analyze DLS data in a thorough manner by providing a large set of fitting parameters. This showed that DLS only sensed the larger vesicles when the micelles/vesicles ratio was 80/20 w/w. On the other hand, AsFlFFF allowed clear detection of the presence of micelles when this same ratio was as low as 10/90. Subsequently, the photodynamic therapy efficiency of various controlled mixtures was assessed using multicellular spheroids when a photosensitizer, pheophorbide a, was encapsulated in the polymer self-assemblies. Some mixtures were shown to be as efficient as monomorphous systems. In some cases, mixtures were found to exhibit a higher PDT efficiency compared to the individual nano-objects, revealing a synergistic effect for the efficient delivery of the photosensitizer. Polymorphous vectors can therefore be superior in therapeutic applications.

Abstract: International audience; Ozone treatments were applied on conidia aqueous suspensions in order to determine theminimal applied ozone dose to limit conidia germination and to observe the mechanismsinvolved in the spores inactivation. Conidia germination was significantly reduced, bubbling forat least 0.5 min as a gas with a minimal ozone concentration of 1 g.m−3. The applied ozone dosesinduce the membrane phospholipids oxidation, determined by the malondialdehyde quantifica-tion. Membrane phospholipids oxidation and inactivation rate are correlated. So, lipid peroxida-tion and consequently the alteration of the membrane integrity are involved in the antifungalaction of ozone.

Abstract: International audience; Industrial gelatins have different physicochemical properties that mainly depend of the raw materials origin and the extraction conditions. These properties are closely related to the molar mass distribution of these gelatins. Several methods exist to characterize molar mass distribution of polymer, including the Asymmetrical Flow Field Flow Fractionation method. The goal of this study is to analyze the relationship between physicochemical properties and the gelatins molar mass distribution obtained by Asymmetrical Flow Field Flow Fractionation. In this study, 49 gelatins samples extracted from pig skin are characterized in terms of gel strength and viscosity and their molar mass distribution are analyzed by Asymmetrical Flow Field Flow Fractionation coupled to an Ultraviolet and Multi Angle Light Scattering detector. This analytical method is an interesting tool for studying, simultaneously, the primary chains and the high-molar-mass fraction corresponding to the polymer chains. Correlation analysis between molar mass distribution data from the different fractions highlights the importance of high molar mass polymer chains to explain the gel strength and viscosity of gelatins. These results are confirmed by an additional chemometric approach based on the UV absorbance of gelatin fractograms to predict gel strength (r2Cal = 0.85) and viscosity (r2Cal = 0.79).

Abstract: International audience; Each year, mycotoxins cause economic losses of several billion US dollars worldwide. Consequently, methods must be developed, for producers and cereal manufacturers, to detect these toxins and to comply with regulations. Chromatographic reference methods are time consuming and costly. Thus, alternative methods such as infrared spectroscopy are being increasingly developed to provide simple, rapid, and nondestructive methods to detect mycotoxins. This article reviews research conducted over the last eight years into the use of near-infrared and mid-infrared spectroscopy to monitor mycotoxins in corn, wheat, and barley. More specifically, we focus on the Fusarium species and on the main fusariotoxins of deoxynivalenol, zearalenone, and fumonisin B1 and B2. Quantification models are insufficiently precise to satisfy the legal requirements. Sorting models with cutoff levels are the most promising applications

Abstract: 'The gastrointestinal tract represents the first barrier against food contaminants as well as the first target for these toxicants. Deoxynivalenol (DON) is a mycotoxin that commonly contaminates cereals and causes various toxicological effects. Through consumption of contaminated cereals and cereal products, human and pigs are exposed to this mycotoxin. Using in vitro, ex vivo and in vivo approaches, we investigated the effects of DON on the intestinal epithelium. We demonstrated that, in intestinal epithelial cell lines from porcine (IPEC-1) or human (Caco-2) origin, DON decreases trans-epithelial electrical resistance (TEER) and increases in a time and dose-dependent manner the paracellular permeability to 4 kDa dextran and to pathogenic Escherichia coli across intestinal cell monolayers. In pig explants treated with DON, we also observed an increased permeability of intestinal tissue. These alterations of barrier function were associated with a specific reduction in the expression of claudins, which was also seen in vivo in the jejunum of piglets exposed to DON-contaminated feed. In conclusion, DON alters claudin expression and decreases the barrier function of the intestinal epithelium. Considering that high levels of DON may be present in food or feed, consumption of DON-contaminated food/feed may induce intestinal damage and has consequences for human and animal health.

Abstract: International audience; Lactic acid bacteria, in particular Lactococcus lactis, play a decisive role in the cheese making process and more particularly in lactic cheeses which are primarily produced on goat dairy farms. The objective of this study was therefore to identify the main lactic acid bacteria found in raw goats' milk from three different regions in France and evaluate if certain farming practices have an effect on the distribution of species of lactic acid bacteria in the various milk samples. Identification at genus or species level was carried out using phenotypic tests and genotypic methods including repetitive element REP-PCR, species-specific PCR and 16S rRNA gene sequencing. The distribution of the main bacterial species in the milk samples varied depending on farms and their characteristics. Out of the 146 strains identified, L. lactis was the dominant species (60% of strains), followed by Enterococcus (38%) of which Enterococcus faecalis and Enterococcus faecium. Within the species L. lactis, L. lactis subsp lactis was detected more frequently than L. lactis subsp cremoris (74% vs. 26%). The predominance of L. lactis subsp cremoris was linked to geographical area studied. It appears that the animals' environment plays a role in the balance between the dominance of L. lactis and enterococci in raw goats' milk. The separation between the milking parlor and the goat shed (vs no separation) and only straw in the bedding (vs straw and hay) seems to promote L. lactis in the milk (vs enterococci).

Abstract: Ozone could be used as a sanitary agent in the food and agricultural industries. The present study investigates whether ozonated water could be used to control conidia dispersal of the esca-associated fungus Phaeoacremonium aleophilum. The fungicide properties of ozonated water was firstly assessed in vitro by exposing spores to several concentrations of ozonated water (2.2, 4.5, or 13.5g/m3 of ozone dissolved into water) and observing the germination rate on plate. Secondarily an in planta approach was conducted on grapevine cuttings of cv. Cabernet Sauvignon clone 15. P. aleophilum conidia were inoculated in pruning wounds, which then received ozonated (4.5g/m3) or sterile water. P. aleophilum DNA was quantified by a quantitative polymerase chain reaction (qPCR) 4 and 9 weeks post-inoculation. The effect of ozonated water on plant-defense gene expression was monitored by reverse-transcriptase qPCR (RT-qPCR) 48h post treatment. The results indicate that ozonated water totally suppresses spore germination in vitro. In addition, at 9 weeks post-inoculation, fungal development was significantly reduced by 50% in planta. RT-qPCR analysis shows that ozonated water did not induce plant-defense-related genes 48h post treatment. The fungicide properties of ozonated water and the absence of gene induction in planta make however ozonated water a promising candidate for limiting grapevine infection by P. aleophilum in nurseries.

36. Esca et vigne : compréhension des mécanismes de défense précoces du bois de la vigne Vitis vinifera L. suite à la maladie, colonisation des champignons in planta et proposition de moyens de lutte pour une viticulture durable

Abstract: L’esca est une maladie du bois de la vigne complexe et mal connue, contre laquelle aucun moyen de lutte efficace n’existe à ce jour. Ce travail s’est concentré sur les interactions précoces entre Vitis vinifera L. et les champignons associés au « young esca » P. chlamydospora et P. aleophilum dans deux types de tissus lignifiés : l’entre-nœud et le nœud (modèle plaie de taille). La colonisation 6 et 12 semaines après traitement des souches transformées P. aleophilum::gfp7 et P. chlamydospora::gfp1 a été observée. Les deux espèces coloniseraient différents tissus dans les premières semaines suivant l’infection. Les fibres du xylème constitueraient un tissu essentiel lors de l’interaction précoce entre P. aleophilum et la vigne, tandis que P. chlamydospora::gfp1 a seulement colonisé les vaisseaux du xylème après 12 semaines. Le bois de la vigne présenterait des réponses spécifiques à la présence de P. aleophilum 6 semaines après traitement, puis générales à la blessure 12 semaines après traitement, en microscopie. L’hypothèse de la spécificité de la réponse induite dans le bois de la vigne par ces deux espèces a été confirmée en étudiant l’expression de 11 gènes associés à la défense 10 h, 24 h, 48 h et 120 hpi. La réponse précoce du bois de la vigne serait spécifique suivant l’identité des pathogènes. Les tissus de l’entre-nœud ont été induits différemment par la blessure par rapport aux tissus dans la région nodale. Un modèle pour le criblage d’agents de biocontrôle ou d’éliciteurs contre l’esca en quelques mois en conditions de laboratoire a permis le développement d’un moyen de lutte durable et novateur, l’eau ozonée. L’eau ozonée présente des propriétés sporicides remarquables contre P. aleophilum in vitro. In planta l’application d’eau ozonée sur une blessure infectée en modèle plaie de taille a réduit de moitié la quantité de mycélium capable de se développer dans le bois 9 semaines après inoculation.

Abstract: International audience; Biomimetic calcium phosphate apatites, analogous to bone mineral, may now be produced synthetically. Their intrinsic biocompatibility and the nanometer dimensions of their constitutive crystals not only allow one to envision applications in bone tissue regeneration, but also in other medical fields such as nanomedicine, and in particular in view of cell diagnosis. In this mini-review, we look back at 10 years of our dedicated research, and summarize the main advances made in terms of preparation, physical-chemical characterizations and biological evaluations of colloidal formulations of biomimetic apatite-based nanoparticles, which we illustrate here with the angle of cancer diagnosis. The confirmed exceptional biocompatibility of these engineered nanoparticles, associated to the possibility to confer them luminescence properties by way of controlled lanthanide doping, and their capacity to be internalized by cells, including with cancer cell addressing abilities (shown here as a proof of concept), underline that biomimetic apatite-based colloidal nanoparticles are particularly promising for nanomedicine applications, for example related to diseased cells diagnosis. Multidisciplinary research on these functional nanoparticles, initiated as described here, has now generated emulation in the scientific community where the concept of apatite nanoparticles for nanomedicine is being, gratifyingly, appropriated.

Abstract: Fumonisins are major mycotoxins found worldwide in maize and maize products. Because of their toxicity for both human and animals, European Union regulations were created to fix the maximal fumonisin B1 and B2 content allowed in foods and feeds. Unfortunately, directly measuring these mycotoxins by current analytical techniques is tedious and expensive and most measurement methods do not lend themselves to online control. Alternative approaches to chemical analysis have been developed and involve models that allow the mycotoxin contamination to be predicted based on environmental conditions and analysis by near-infrared (NIR) spectroscopy. In the present work, we use NIR spectroscopy to determine the fumonisin and fungal contents of 117 samples of maize. The determination coefficient between fumonisin and fungal-biomass content was 0.44. We establish herein a threshold for the number of CFUs for fungal biomass beyond which the fumonisin content is likely to exceed the European regulatory level of 4000 μg/kg. In addition, we determine the fungal content by using a NIR-spectroscopy model that allows us to sort samples of maize. Upon calibration, the percentage of well-classified samples was 96%, which compares favorably to the 82% obtained by independent verification.

Abstract: International audience; Deoxynivalenol and fumonisins pose a health concern and have economic consequences, so the European regulation CE 1126/2007 dictates the maximal content allowed in cereals. The direct measurement of mycotoxin content using the established method is not only time-consuming and tedious, but also destructive and cannot be used in a silo. Alternative tools such as infrared spectroscopy are therefore being studied. For the present investigation, spectral data collected from maize kernels contaminated naturally by mycotoxins were studied to predict the risk of deoxynivalenol and fumonisins. Discriminant models were used to create and identify batches that satisfy regulations for animal or human consumption.

Abstract: International audience; Sunflower oil is naturally rich in both unsaturated fatty acids and numerous minor components, with high levels of &-tocopherol and phytosterols that could play an important role in plant protection. These micronutriments, tocopherols and phytosterols, also exhibit health benefits for humans including reducing both cardiovascular diseases and aging disorders. &−Tocopherol represents more than 95% of total sunflower tocopherol and has the greatest vitaminic activity. Furthermore, this oil contains high amounts of β-sitosterol which is a promising health beneficial phytosterol. The objective of this study was to compare the content and composition of fatty acids, tocopherols and phytosterols in sunflower oil between different cultivars and locations in a three-year experiment, and to estimate the relationships between these minor components, fatty acids and climatic conditions. Results revealed that content and composition of these molecules are mainly determined by plant genotype. Nevertheless, the environmental conditions seemed to strongly influence the tocopherol content. The results suggest possible improvement in sunflower breeding for minor components.; El aceite de girasol es naturalmente rico en ácidos grasos no saturados y en numerosos componentes menores. Posee altos niveles de α-tocoferol y fitoesteroles que podrían desempeñar un papel importante en la protección de las plantas. Estos micronutrientes, tocoferoles y fitoesteroles, también presentan beneficios para la salud de los seres humanos, reduciendo el riesgo de padecer enfermedades cardiovasculares y trastornos del envejecimiento. El α-tocoferol representa más del 95% de los tocoferoles del girasol y tiene la mayor actividad vitamínica. Además, este aceite contiene altas cantidades de β-sitoesterol que es un prometedor fitoesterol benéfico para la salud. El objetivo de este estudio fue comparar el contenido y la composición de ácidos grasos, tocoferoles y fitoesteroles en el aceite de girasol entre los diferentes cultivares y sitios en un experimento de tres años, y evaluar las relaciones entre estos componentes minoritarios, los ácidos grasos y las condiciones climáticas. Los resultados revelaron que el contenido y la composición de estas moléculas principalmente están determinados por el genotipo de la planta. Sin embargo, las condiciones ambientales parecen influir mucho en el contenido de tocoferoles. Los resultados sugieren una posible mejora genética del girasol con respecto a sus componentes menores.

Abstract: International audience; The use of soy proteins (SoyP) and sunflower proteins (SunP) in the microencapsulation by spray-drying technique of α-tocopherol (T) with a core/wall ratio of 2/1 was studied. SoyP and SunP were used as wall material in an unmodified and modified state. The enzymatic (hydrolysis and cross-linking) and chemical (acylation and cationization) modifications were carried out on vegetable proteins in order to improve their encapsulating properties. The results obtained demonstrated that in the native state, SunP showed higher retention efficiency for T microencapsulation (92.6%) compared to SoyP (79.7%), which could be connected to the different composition of protein extracts. Hydrolysis, acylation and cationization of protein resulted in reduced emulsion viscosity. The retention efficiency of T was improved up to 94.8–99.5% after protein acylation, which was attributed to improved affinity between core and wall material.

Abstract: A field experiment was conducted on Alfisols in South-West France to assess the agronomic and environmental impacts of a single application of heat-dried sludge pellets at 11.1 Mg dry matter ha-1. The sludge pellets, with a moisture level of 9.5%, were spread on an irrigated crop of maize (Zea mays L.). This treatment was compared with inorganic fertilization (urea and diammonium phosphate mixed with KCl). Soil properties, yield and the composition of maize and the quality of drained water were monitored over 1 year to detect any changes resulting from sludge application. Amongst several determined soil properties, only two were significantly modified by the sludge application: The nitric nitrogen stock of the soil was higher in the inorganic fertilized plot, whereas Olsen-P soil content was higher in the sludge-amended plot. Agronomic recovery rates of N and P added by sludge were high: For the first crop following application, total amounts of N and P supplied by the sludge had the same efficiency as approximately 45% of the N and P amounts supplied by inorganic fertilizer. This ratio was 7% for the N uptake by the second maize crop. The quality and quantity of maize were equally good with both types of fertilization. During the 2 years following sludge spreading, N leaching remained as low in the sludged plot as in the inorganically fertilized one. The Cu, Zn, Cr, Cd, Pb and Ni composition of the drainage water was affected by neither of the types of amendment. From the heavy-metal contents of the soil, water and maize monitored over 1 year in the field experiment and from literature data for cow manure and atmospheric emissions, a theoretical balance between crop soil heavy-metal input and output over one century was drawn up. The long-term impact of cow manure on Zn, Ni and Cr in soil is higher than that of the studied heat-dried sludge. Obviously, sludge tended to cause a strong increase in soil Cu storage, valued for these soils, which are otherwise very Cu deficient.

Abstract: A major challenge for agroecology is to identify alternative cropping systems that can both ensure sufficient levels of food production and reduction in pesticide inputs while maintaining ecosystem integrity and simultaneously delivering multiple ecosystem services. Among ecological services, biocontrol of pests by natural enemies could be improved by effective management of semi-natural elements in landscape such as hedgerows (Landis et al., 2000). Another way to reduce pest populations in fields consists in increasing diversity of crop species both in time and space. A practice such as intercropping that creates structures of mixed plant populations with "barrier" or "dilution of resources" effects (Médiène et al., 2011) can reduce the pressure of pathogens, weeds and pests compared with that exerted on sole crops (Letourneau et al., 2011). While the combination of these two kinds of practices designed to strengthen auxiliary communities often appears efficient and could result in a synergy, many cases of study show opposite results (e.g. Straub and Snyder, 2006). Our study aimed at quantifying and comparing the efficiency of both hedgerow and intercropped pea and wheat on pest biocontrol in a field. We examined i) the potential role of different types of field edges (with and without an hedgerow) on biological control of weed seeds, aphids, pea weavils and eggs of Sitona lineatus, ii) the effect of intercropping on the biological control of these pests compared with sole crops and iii) the interaction between the two factors. We conducted both field surveys of pest abundance and pest biocontrol effectiveness by estimating predation rates by using seeds and prey sticked on cards. We found that aphid density was slightly lower in pea-wheat intercrops than in sole crops, and was influenced by the distance from the field edge (whatever the kind of edges). However, neither the hedgerow nor the intercrop situation favored aphid predation. The density of weeds did not vary according to the cropping system and distance from the field edge. We showed that the predation rate of weed seeds or of Sitona lineatus eggs and the parasitism of Bruchus pisorum decreased according to the distance to the hedge in sole crops but increased in the intercrop. This study points out the absence of a global synergy between intercrops and hedges in supporting pest and weed regulation: the regulation efficiency varied according to pest species. This study calls for a deepened multi-species approach in order to find trade-offs between agricultural practices and field edge management to optimize regulating ecosystem services such as pest biocontrol.

Abstract: International audience; Background: Processed meat intake has been associated with increased colorectal cancer risk. We have shown that cured meat promotes carcinogen-induced preneoplastic lesions and increases specific biomarkers in the colon of rats. Objectives: We investigated whether cured meat modulates biomarkers of cancer risk in human volunteers and whether specific agents can suppress cured meat-induced preneoplastic lesions in rats and associated biomarkers in rats and humans. Design: Six additives (calcium carbonate, inulin, rutin, carnosol, alpha-tocopherol, and trisodium pyrophosphate) were added to cured meat given to groups of rats for 14 d, and fecal biomarkers were measured. On the basis of these results, calcium and tocopherol were kept for the following additional experiments: cured meat, with or without calcium or tocopherol, was given to dimethylhydrazine-initiated rats (47% meat diet for 100 d) and to human volunteers in a crossover study (180 g/d for 4 d). Rat colons were scored for mucin-depleted foci, putative precancer lesions. Biomarkers of nitrosation, lipoperoxidation, and cytotoxicity were measured in the urine and feces of rats and volunteers. Results: Cured meat increased nitroso compounds and lipoperoxidation in human stools (both P < 0.05). Calcium normalized both biomarkers in rats and human feces, whereas tocopherol only decreased nitro compounds in rats and lipoperoxidation in feces of volunteers (all P < 0.05). Last, calcium and tocopherol reduced the number of mucin-depleted foci per colon in rats compared with nonsupplemented cured meat (P = 0.01). Conclusion: Data suggest that the addition of calcium carbonate to the diet or alpha-tocopherol to cured meat may reduce colorectal cancer risk associated with cured-meat intake. Abstract: Chantier qualité GA; International audience; OBJECTIVE: The gut microbiota, which is considered a causal factor in metabolic diseases as shown best in animals, is under the dual influence of the host genome and nutritional environment. This study investigated whether the gut microbiota per se, aside from changes in genetic background and diet, could sign different metabolic phenotypes in mice. METHODS: The unique animal model of metabolic adaptation was used, whereby C57Bl/6 male mice fed a high-fat carbohydrate-free diet (HFD) became either diabetic (HFD diabetic, HFD-D) or resisted diabetes (HFD diabetes-resistant, HFD-DR). Pyrosequencing of the gut microbiota was carried out to profile the gut microbial community of different metabolic phenotypes. Inflammation, gut permeability, features of white adipose tissue, liver and skeletal muscle were studied. Furthermore, to modify the gut microbiota directly, an additional group of mice was given a gluco-oligosaccharide (GOS)-supplemented HFD (HFD+GOS). RESULTS: Despite the mice having the same genetic background and nutritional status, a gut microbial profile specific to each metabolic phenotype was identified. The HFD-D gut microbial profile was associated with increased gut permeability linked to increased endotoxaemia and to a dramatic increase in cell number in the stroma vascular fraction from visceral white adipose tissue. Most of the physiological characteristics of the HFD-fed mice were modulated when gut microbiota was intentionally modified by GOS dietary fibres. CONCLUSIONS: The gut microbiota is a signature of the metabolic phenotypes independent of differences in host genetic background and diet.

Période 2000-2012. http://prodinra.inra.fr/record/256699; he genotoxicity of quinolone and fluroquinolones was assessed using the micronucleus (MN) test on Vicia faba roots by direct contact exposure to a solid matrix. Plants were exposed to quinolones (nalidixic acid) and fluoroquinolones (ciprofloxacin and enrofloxacin) alone or mixed with artificially contaminated soils. Four different concentrations of each of these antibiotics were tested (0.01, 0.1, 1 and 10 mg/Kg) for nalidixic acid and (0.005, 0.05, 0.5 and 5 mg/Kg) for ciprofloxacin and enrofloxacin. These antibiotics were also used in mixture. Exposure of Vicia faba plants to each antibiotic at the highest two concentrations showed significant MN induction. The lowest two concentrations had no significant genotoxic effect. The mixture of the three compounds induced a significant MN induction whatever the mixture tested, from 0.02 to 20 mg/Kg. The results indicated that a similar genotoxic effect was obtained with the mixture at 0.2 mg/Kg in comparison with each molecule alone at 5-10 mg/Kg. Abstract: Ce rapport est le résultat de l’étude exécutée par les élèves de l’École d’Ingénieurs de Purpan sur le Pays de la Haute Vallée de l’Aude. Le diagnostic a révélé que ce territoire d’arrière-pays est composé d’entités territoriales multiples avec une démographie, des paysages et des activités économiques variés. Sur cet ancien bassin industriel, le tourisme et l’agriculture ont progressivement pris le pas sur l’industrie en tant qu’activité économique mais ne sont cependant pas exploités à leur optimum. Le principal enjeu de développement du territoire révélé est d’organiser conjointement les filières du tourisme, du bois et de production/distribution des produits locaux. Aussi, le maintien de la qualité de vie avec en autre le développement des transports et des services à la population a été identifié comme enjeux de développement sur le territoire. Afin de répondre à ces enjeux, le pays pourra mettre en action les préconisations formulées. Elles ont comme objectif d’exploiter les potentialités du territoire de manière cohérente. Parmi ces préconisations, la structuration et la coordination des activités et des acteurs est primordiale. Pour cela, il est important de fédérer la population autour d’une identité commune et de la mobiliser autour d’objectifs communs.

Abstract: National audience; L’amélioration de la connaissance des cortèges de Coléoptères saproxyliques présents dans les forêts gérées des Pyrénées est un préalable indispensable à leur conservation. Dans le cadre d’une étude menée sur la propriété du Groupement Forestier de Hèches, un inventaire des Coléoptères saproxyliques a été conduit en 2003 et 2004, au moyen de pièges et de chasse à vue. Une liste de 374 taxons (espèces non-saproxyliques incluses) a ainsi été établie. Quelques espèces rares, voire inattendues font l’objet de commentaires faunistiques. Cet inventaire offre par ailleurs l’opportunité de situer le massif forestier étudié dans une échelle de valeur biologique des forêts pyrénéennes en 5 classes.

Abstract: National audience; La canicule de 2003 a provoqué dans le Sud Massif Central des dépérissements résineux brutaux et massifs. Après la crise est venu le temps de l’analyse, en évaluant d’abord l’étendue des impacts, ne serait-ce que pour organiser les reconstitutions à l’échelle régionale. La méthode de télédétection imaginée et mise en oeuvre avec succès est bien adaptée à ce type de phénomène, avec ceci d’original qu’elle intègre les coupes sanitaires d’urgence comme indicateur de l’intensité du dépérissement. Mais gardons-nous de généraliser, car de ce fait même elle ne se prêterait pas à l’évaluation d’autres types de dépérissement, plus diffus ou aux symptômes plus « subtils ».
