Xenotransplantation of human tissues into immunodeficient mice has emerged as an invaluable preclinical model to study human biology and disease progression and predict clinical response. The most common anatomical site for tissue transplantation is the subcutaneous pocket due to simple surgical procedures and accessibility for gross monitoring and advanced imaging modalities. However, subcutaneously implanted tissues initially experience a sharp change in oxygen and nutrient supply and increased mechanical deformation. During this acute phase of tissue integration to the host vasculature, substantial cell death and tissue fibrosis occur limiting engraftment efficiency. Previously, we demonstrated that the implantation of inverted colloidal crystal hydrogel scaffolds triggers proangiogenic and immunomodulatory functions without characteristic foreign body encapsulation. In this study, we examine the use of this unique host response to improve the ectopic transplantation of tissues to the subcutaneous site. Scaffold-assisted tissues preserved morphological features and blood vessel density compared to native tissues, whereas scaffold-free tissues collapsed and were less vascularized. Notably, the supporting biomaterial scaffold modulated the foreign body response to reduce the localization of Ly6G(+) cells within the transplanted tissues. Cotransplantation of patient-derived gastric cancer with a scaffold resulted in a comparable level of engraftment to conventional methods; however, detailed immunohistological characterization revealed significantly better retention of proliferative cells (Ki67(+)) and human immune cells (CD45(+)) by the end of the study. We envision that leveraging the immunomodulatory properties of biomaterial interfaces can be an attractive strategy to improve the functional engraftment of xenotransplants and accelerate individualized diagnostics and the development of novel therapeutic strategies.

Background The expanded scalps are usually used to repair the scalp defects. However, hair growth direction and hair density of the expanded scalps may be inconsistent with those of the defective sites, resulting in unsatisfactory results. Aims The aim of this study is to assess the effect of local flap modification and follicular unit extraction (FUE) on the repair of poor appearance of temporal and sideburn after unsatisfactory tissue expansion. Methods In our study, 19 patients with problems of hair growth direction or hair density after surgery were treated with following four methods: Single local flap modification; Removal of hair follicles by FUE to reduce follicle density; Removal of disordered hair follicles by FUE and ectopic transplantation simultaneously; Removal of hair follicles by FUE and ectopic transplantation after local flap modification. The follow-up periods of 19 patients varied from 6 months to 30 months, and natural, standardized aesthetic sideburn for each patient was achieved. Results All incisions healed by first intention and all flaps survived well. The scars involved in the removal of FUE hair follicles were not obvious. Conclusions Our results revealed that the ideal and cosmetic results can be obtained by flexible using of local flap modification, single FUE method, and FUE ectopic hair transplantation to repair the dissatisfied postoperative outcomes of temporal and sideburn cicatricial alopecia after tissue expansion.

Objectives To understand the differences and similarities between immunocompetent and immunodeficient mice as ectopic transplantation animal models for bone tissue engineering. Materials and Methods Osteogenic cells from mouse leg bones were cultured, seeded on β- TCP granules, and transplanted onto the backs of either immunocompetent or immunodeficient nude mice. At 1, 2, 4, and 8 weeks postoperatively, samples were harvested and evaluated by hematoxylin-eosin staining, tartrate-resistant acid phosphatase ( TRAP) staining, and immunohistochemical staining and quantitative PCR. Results In immunocompetent mice, inflammatory cell infiltration was evident at 1 week postoperatively and relatively higher expression of TNF- α and IL-4 was observed. In immunodeficient mice, new bone area and the number of TRAP-positive cells were larger at 4 weeks than in immunocompetent mice. The volume of new bone area in immunodeficient mice was reduced by 8 weeks. Conclusions Bone regeneration was feasible in immunocompetent mice. However, some differences were observed between immunocompetent and immunodeficient mice in the bone regeneration process possibly due to different cytokine expression, which should be considered when utilizing in vivo animal models., 2015

Objective to investigate whether harmine has a promotive effect on human periodontal ligament cells (hPDLCs)-induced tissue regeneration. Materials and Methods Various concentrations of harmine on hPDLCs proliferation were tested. Osteogenic and cementogenic characteristics were examined in hPDLC/rhBMP-2 and hPDLC/harmine by alizarin red S staining, real-time PCR, and Western blotting assay. The activity of harmine was investigated in an ectopic transplantation nude mouse model. Results We determined that 10 μM of harmine was the threshold concentration. hPDLC/harmine showed similar mineralized nodule formation in alizarin S staining compared to hPDLC/rhBMP-2. In real-time PCR, the highest gene expression level was observed for Runx2 in hPDLC/harmine at all time points. The level of CEMP-1 in hPDLC/harmine was higher at 7 days than hPDLCs alone. Thicker band of Runx2 in hPDLC/harmine was observed than in hPDLC/rhBMP-2 at 7 days by Western blotting. The band for CEMP-1 in hPDLC/harmine was thicker than hPDLCs alone at both 7 and 14 days. In ectopic transplantation, hPDLCs with harmine showed a comparable amount of mineralized tissue formation compared to rhBMP-2. hPDLCs with harmine or rhBMP-2 formed both bone and cementum-like tissue with Sharpey's fiber-like collagen insertion. Conclusion Harmine can be a potential candidate for promoting hPDLCs-induced tissue regeneration.

Purpose of review This review outlines the concept of cell-based therapy to restore tissue function, and addresses four key points to consider in cell transplantation: source, surveillance, safety, and site. Whereas each point is essential, additional attention should be given to transplantation sites if cell therapy is going to be successful in the clinic. Various ectopic locations are discussed, and the strengths and weaknesses of each are compared as suitable candidates for cell therapy. Recent findings Studies in rodents often demonstrate cell transplantation and engraftment in ectopic sites, with little evidence to suggest why it may also work in humans. For example, transplantation to the subcapsular space of the kidney is often performed in rodents, but has not been a good predictor of clinical success. Recent work has shown that the lymph node may be a good site for transplantation of multiple tissue types, and several reasons are highlighted as to why it should be considered for future studies. Summary The use of cell-based therapy in the clinic has been hampered by the lack of appropriate sites for transplantation. The lymph node is a promising alternative for cell transplantation, and offers hope for clinical application.

The anterior visceral endoderm (AVE) is an extra-embryonic tissue required for specifying anterior pattern in the mouse embryo. The AVE is induced at the distal tip of the 5.5 dpc embryo and then migrates to the prospective anterior, where it imparts anterior identity upon the underlying epiblast (the tissue that gives rise to the embryo proper). Little is known about how the AVE is induced and what directs its migration. In this paper, we describe an essential role for another extra-embryonic tissue, the extra-embryonic ectoderm (ExE), in patterning the AVE and epiblast. Removal of the ExE in pre-gastrulation embryos leads to ectopic AVE formation, to a failure of AVE cell migration and to the assumption by the entire epiblast of an anterior identity. Ectopic transplantation of ExE cells inhibits AVE formation and leads to an expansion of the posterior epiblast marker T. These results demonstrate that the ExE restricts the induction of the AVE to the distal tip of the mouse embryo and is required to initiate the migration of these cells to the prospective anterior. Together, these data reveal a novel role for the ExE in the specification of the anteroposterior axis of the mouse embryo.

Creating a model for intravital visualization of femoral bone marrow, a major site of hematopoiesis in adult mammalian organisms, poses a serious challenge, in that it needs to overcome bone opacity and the inaccessibility of marrow. Furthermore, meaningful analysis of bone marrow developmental and differentiation processes requires the repetitive observation of the same site over long periods of time, which we refer to as chronic imaging. To surmount these issues, we developed a chronic intravital imaging model that allows the observation of split femurs, ectopically transplanted into a dorsal skinfold chamber of a host mouse. Repeated, long term observations are facilitated by multiphoton microscopy, an imaging technique that combines superior imaging capacity at greater tissue depth with low phototoxicity. The transplanted, ectopic femur was stabilized by its sterile environment and rapidly connected to the host vasculature, allowing further development and observation of extended processes. After optimizing transplant age and grafting procedure, we observed the development of new woven bone and maturation of secondary ossification centers in the transplanted femurs, preceded by the sprouting of a sinusoidal-like vascular network, which was almost entirely composed of femoral endothelial cells. After two weeks, the transplant was still populated with stromal and haematopoietic cells belonging both to donor and host. Over this time frame, the transplant partially retained myeloid progenitor cells with single and multi-lineage differentiation capacity. In summary, our model allowed repeated intravital imaging of bone marrow angiogenesis and hematopoiesis. It represents a promising starting point for the development of improved chronic optical imaging models for femoral bone marrow.

Aim: Hepatocyte transplantation to host livers by single cell suspension injection from the portal vein has been clinically successful in cases where the host liver architecture is intact. However, further investigation is still needed to achieve regeneration of the liver architecture when the host liver is destroyed, since single cell suspension injection often results in the formation of small hepatocyte colonies or islands. We show a hepatocyte transplantation strategy to ectopic sites. Methods: Primary hepatocytes isolated from green fluorescent protein (GFP)-transgenic rats were cultured on temperature-responsive culture dishes. After harvest as intact cell sheets, triple-layered cell sheets were transplanted over the superficial caudal epigastric artery and vein of athymic rats which had operation of 70% partial hepatectomy. Results: The transplanted hepatocytes were integrated to host tissue with a laminar cell arrangement at transplanted sites within one week after surgery. But the transplanted hepatocytes were hardly detected four weeks after transplantation, when the partially hepatectomized host liver was completely regenerated. GFP-positive bile duct-like tubes and functional blood vessels were observed. Conclusion: These results imply the usefulness of hepatocyte sheets in ectopic transplantation, as well as the need of trophic factors for hepatocyte survival.

The subcapsular region of the kidney has frequently served as the site of choice for transplantation studies owing to a number of compelling reasons. High levels of vascularization provide a ready blood supply, whereas the subcapsular region itself can accommodate tissues of a range of size and sources. Historically, transplantation to this site has proven important for studies of both central and peripheral tolerance. Here, the transplantation technique and its major variations are described for broad application.

Previous studies from our laboratory have demonstrated that invasive trophoblast cells of the equine chorionic girdle have the capacity to differentiate and survive after transplantation to ectopic sites in fully allogeneic, non-pregnant mares. The lifespan of the transplanted cells was not determined, and no functional outcome was described.

Background Human tumour xenografts in immune compromised mice are widely used as cancer models because they are easy to reproduce and simple to use in a variety of pre-clinical assessments. Developments in nanomedicine have led to the use of tumour xenografts in testing nanoscale delivery devices, such as nanoparticles and polymer-drug conjugates, for targeting and efficacy via the enhanced permeability and retention (EPR) effect. For these results to be meaningful, the hyperpermeable vasculature and reduced lymphatic drainage associated with tumour pathophysiology must be replicated in the model. In pre-clinical breast cancer xenograft models, cells are commonly introduced via injection either orthotopically (mammary fat pad, MFP) or ectopically (subcutaneous, SC), and the organ environment experienced by the tumour cells has been shown to influence their behaviour. Methods To evaluate xenograft models of breast cancer in the context of EPR, both orthotopic MFP and ectopic SC injections of MDA-MB-231-H2N cells were given to NOD scid gamma (NSG) mice. Animals with matched tumours in two size categories were tested by injection of a high molecular weight dextran as a model nanocarrier. Tumours were collected and sectioned to assess dextran accumulation compared to liver tissue as a positive control. To understand the cellular basis of these observations, tumour sections were also immunostained for endothelial cells, basement membranes, pericytes, and lymphatic vessels. Results SC tumours required longer development times to become size matched to MFP tumours, and also presented wide size variability and ulcerated skin lesions 6 weeks after cell injection. The 3 week MFP tumour model demonstrated greater dextran accumulation than the size matched 5 week SC tumour model (for P Conclusions Dextran accumulation and immunostaining results suggest that small MFP tumours best replicate the vascular permeability required to observe the EPR effect in vivo. A more predictable growth profile and the absence of ulcerated skin lesions further point to the MFP model as a strong choice for long term treatment studies that initiate after a target tumour size has been reached.

The hormonal control of evagination of the imaginal leg discs of the fleshflySarcophaga crassipalpis was studied by means of ectopic transplantation of discs from mature larvae or prepupae onto prepupal hosts. Evagination was influenced by the age of the donor and the developmental commitment of the host. Discs from non-diapuse as well as diapause-committed donors did not differ in their capacity to evaginate and differentiate. Mature larval discs from either type of donor could evaginate only when transplanted onto non-diapause hosts; they failed to evaginate on diapause-committed hosts even though host discs evaginated in situ about 40 h after transplantation. Discs from white prepupae evaginated whether transplanted onto non-diapause or onto diapause-committed hosts.

Isolated adult rat hepatocytes were cryopreserved by cooling, in the presence of dimethyl sulfoxide, to low subzero temperatures and they were stored for 14 days at -196 C. The cells were then recovered and transplanted into the splenic pulp of the original donor animal. A similar transplant procedure was performed with hepatocytes which had not been cryopreserved. Ultrastructural assessment was made 1 month after transplantation. In general fewer hepatocytes survived ectopic transplantation after cryopreservation, but the surviving cells were ultrastructurally similar to ectopically transplanted cells not subjected to cryopreservation. Thus, the reduced metabolic activity measured previously in transplants of cryopreserved hepatocytes is likely to result from a smaller number of competent cells rather than from hepatocytes which have survived in an extensively damaged or abnormal state.

The production of a luteotrophic hormone by ectopically transplanted rat blastocysts was demonstrated by transfer of blastocysts to the kidneys of cyclic and pseudopregnant mice and isogeneic rats. Pseudopregnancy was induced in nearly all cyclic mice bearing trophoblastic outgrowths but not in cyclic rats. In both mice and rats prolongation of pseudopregnancy was observed following ectopic transplantation of rat blastocysts at the time of normal egg implantation. It is concluded that ectopically developing rat trophoblastic tissue produces a luteotrophic hormone.
